Pełny tekst źródłaSerotonin-activated cell-surface receptor 5-HT₇ is the most recently discovered 5-HT receptor. Intensive studies of structure-activity relationships have been pursued by several research groups, including our laboratory, and have resulted in the publication in the literature of an impressive number of potential ligands 5-HT₇. In this context, the main purpose of this thesis is to design four distinct classes of ligands. First, we were interested in the preparation of 2,4-diaminopyrido[2,3-d]pyrimidinique and 2,4-diaminopyrimidinique ligands. Subsequently, we developed a third family of compounds built on a pyridine scaffold, from one of the most interesting current 5-HT₇ selective agonists, via an effective multistep strategy. The last part of this thesis was devoted to developing an original and rapid method for the synthesis of polysubstituted tetrahydro-1,6-naphthyridines starting from the 3-vinyl-1,2,4-triazines platforms via a domino addition sequence of aza-Michael and intramolecular inverse-electron-demand Diels-Alder cyclization

Pełny tekst źródłaLes récepteurs 5-HT4 de la sérotonine sont des récepteurs couplés aux protéines G (RCPG). Les RCPGs peuvent se dimériser, ce qui influence leurs propriétés pharmacologiques. Les récepteurs 5-HT4 étant impliqués dans d'importants processus physiologiques, nous avons étudié leur processus de dimérisation. Nous avons montré par co-immunoprécipitation et Bioluminescence Resonnance Energy Transfert, que les isoformes 5-HT4 s'homodimérisent et s'hétérodimérisent constitutivement, entre elles et avec le récepteur β2-adrénergique. Le dimère de récepteur 5-HT4 est sensible au dithiothréitol (agent réducteur de ponts disulfures). La mutation de deux cystéines localisées dans les domaines transmembranaires 3 et 4, inhibe la dimérisation du récepteur 5-HT4 et entraîne sa rétention dans le réticulum endoplasmique. Des ligands bivalents spécifiques du récepteur 5-HT4 ne présentent pas d'affinité ou d'efficacité particulière pour le récepteur 5-HT4 mais peuvent stabiliser les dimères de récepteurs

Pełny tekst źródłaSintering is a key step during the fabrication of UOx and MOx nuclear fuels used in the PWR reactors. The first stage of this process that is characterized by the formation of necks between the grains, leading to the mechanical consolidation of the sample, is particularly crucial. It is usually described through mathematical models based on two spherical grains in contact. In this frame, an experimental approach of the first stage of sintering for ceramic materials of interest for the nuclear fuel cycle, mainly based on in situ high-temperature environmental scanning electron microscopy observations, is developed in this PhD work. An original synthesis route towards size-controlled UO2+x and (U,Ce)O2+x microspheres was first developed. Then, experimental observations of the first stage of UO2+x sintering have been carried out under different operating conditions by varying the oxygen partial pressure and temperature. Quantitative data obtained through image processing led to determine prevailing diffusion mechanisms and associated activation energies for different hyper-stoichiometric uranium dioxides as well as for U3O8. Under reducing conditions, complementary ex situ experiments and high-temperature transmission electron microscopy observations has allowed identifying a two-step mechanism based on the growth of crystallites in the grains, then on the formation and growth of a neck between the grains. Finally, a preliminary study on (UO2 – CeO2) heterogeneous systems revealed a drastic modification of the morphological evolution of the two grains, induced by the increasing complexity of the involved diffusion mechanisms (including the preferential diffusion of uranium in CeO2). Conversely, the first sintering tests carried out on homogeneous (U,Ce)O2 solid solutions evidenced the classical behavior described by the models. The stoichiometry of the samples involved in the sintering of actinide dioxides, but also their homogeneity, must be carefully taken into account in the aim to build predictive numerical models

Pełny tekst źródłaSeveral studies have established an association between some cardiac valve injuries and overexpression of the serotonergic system. Valve lesions are observed following carcinoid tumours (with high blood levels of 5-HT) and during the chronic use of 5-HT2 serotonergic agonists (ergot derivatives or fenfluramines). The current dogma is based on a mitogenic effect of serotonin, by activating 5-HT2B receptors, leading to resident valvular cells proliferation, but does not explain the degeneration of acellular cardiac bioprosthesis. Our work identified endothelial progenitor cells (CD34 + / CD309 +), expressing 5-HT2A and 5-HT2B receptors, in human aortic and mitral valve lesions, regardless of the etiology of their degeneration. Our work highlights the dual role of serotonin in valvular degeneration: 1) stimulation of the 5-HT2B receptor contributes to blood mobilization of CD34+ progenitors (recruited from the bone marrow, and migrating in the valve tissue), 2) the role of 5-HT2 receptors in the transdifferentiation of endothelial progenitor cells in activated valvular cells. The results of this work could drive to the development of 1) a predictive biomarker of cardiac valve injuries in high-risk populations, 2) a model to study heart valve disease cellular and molecular mechanisms, and 3) identify therapeutic targets around the serotonergic system, to slower the progression of the lesions and delay surgical replacement, the only current alternative

Pełny tekst źródłaSkin hydrolipidic film is "the final frontier between the organism and the environment." Thus, it provides skin protection and contributes to its properties. However, it may be involved in several skin disturbances. The aim of this study was to obtain an overall profile of skin surface lipids (SSLs), rich in structural information and able to describe the organization of these molecules. This information allows monitoring the physiological evolution of hydrolipidic film at the molecular level. Several analytical approaches have been developed and chemometric and statistical tools were used to improve the interpretation of the results.SSL characterization was performed using high-temperature gas chromatography coupled with mass spectrometry (HT-GC/MS). The analytical protocol has been developed keeping the lipids in their intact structures. Over than 200 compounds were identified in the same run. These compounds have been classified in five lipid classes: free fatty acids, hydrocarbons, waxes, sterols and glycerides. The palmitic acid (C16:0) and sapienic acid (C16:16) are predominant and participate in the structure of many compounds. Several quantitative approaches have been implemented: calculation of chromatographic descriptors, normalization of the peak area for analysis by class and chemometric exploitation of individual compounds data.Three issues were explored by these techniques and lead to have the following results:The SSL distribution on different areas of the body, expressed as a squalene / cholesterol ratio, is higher in body areas rich in sebaceous glands than in poor areas. This descriptor is thus a good criterion for sebum secretion / desquamation balance measurement.The SSL evolution based on the geographical location and / or color of skin has been investigated by studying the variations in the composition and organization using HT-GC/MS and Raman spectroscopy. A difference in composition has been demonstrated between the volunteers according to geographical location: the most involved compound is the sapienic acid (C16: 1Δ6). A difference in conformation (trans / gauche) of at the alkyl chains was detected. Skin barrier adaptation after birth has already been reported. In this study, its evolution at the molecular level has been studied in infants from the first days after birth until the age of 6 months. A decrease in the total amount of sebaceous secretion was observed with, in parallel, an increase of cholesterol esterified with fatty acids of epidermal origin. This indicates an increase in the relative participation of epidermal lipids in the hydrolipidic film as the result of a decrease in sebaceous secretion. The analytical approaches developed in this thesis provide a detailed molecular characterization of SSLs in order to study the physiological evolution of the skin hydrolipidic film. Statistical and chemometric tools have allowed further exploitation of the results to extract information not accessible by conventional approaches. This investigative work offers many opportunities, promising in medicine and in cosmetics

Pełny tekst źródłaThe C60 reaction diagram continues to be a subject of discussion and controversy, despite the vast amount of experimental and theoretical work done over the years. This is mainly due the lack of in situ studies, the highly disordered-states showing poorly resolved diffraction peaks and the coexistence of several low-density polytypes. This manuscript presents a systematic in situ study of high-pressure–high-temperature forms C60 in the range of 1-10 GPa and 300-1200 K. In order to discriminate poly(a)morphs with similar densities in heterogeneous samples, we used a combination of microdiffraction and microtromography. The samples were synthesized in a Paris-Edinburgh cell and characterized using in situ angular dispersive X-ray diffraction. Three-dimensional submicron images were obtained on quenched samples using diffraction/scattering microtomography. This method provides 3D analysis of the scattering intensity reconstructed from sets of 2D microdiffraction projections. Such analysis is non-destructive and provides high sensitivity (0.1% volume), high spatial resolution (µm3) and can be multi-modal providing quantitative information on the morphology, density, elemental composition or structure of materials. Additionally, we describe the development of in situ high-pressure–high-temperature microtomography using a new rotating Paris-Edinburgh cell (RoToPEC) combined with synchrotron radiation. The ability to fully rotate the sample chamber under load, overcomes the limited angular aperture of ordinary high-pressure cells for acquiring tomographic projections in both, full-field imaging or microdiffraction modes. This innovative method enables dynamic studies of materials under extreme pressure-temperature-stress conditions, impacting areas such as physics, chemistry, materials science or geology. The potential of this new experimental technique is demonstrated on the in situ investigation of of high-pressure–high-temperature polymerization of C60 . Pełny tekst źródłaExtensive mechanisms induce interactions between tectonic, sedimentary and metamorphic processes, especially when they are associated with a high temperature metamorphism. Therefore, the study of these mechanisms requires a threefold approach in order to identify these processes and understand the interactions between them. Such study is hardened because of the depth of these mechanisms and the lack of direct observation. To overcome such difficulties we chose to study a paleo passive margin now visible thanks to many outcrops. For this thesis, we focused on the North Pyrenean Zone (NPZ) which happens to be a Mesozoic passive margin which outcroped during Eocene / Oligocene (40-20 Ma) Pyrenean compression. This margin recorded HT / LP metamorphism which makes it a good analogue for our study. We focused on the eastern part of the NPZ, in the Agly Massif area where are located the Saint Paul-de-Fenouillet basin, the bas-Agly basin and the Boucheville basin all of which recorded different intensity of metamorphism. The Boucheville basin is the worst documented but recorded the most metamorphism which is why we decided to study it in detail. We undergone a structural, stratigraphic and sedimentary analysis of these three basins in order to better understand their geometry and geological history. We combined this study with metamorphism analyzes mainly provided by Raman spectrometry on maximum temperatures recorded during Cretaceous extension. Finally, we synthetised these results to produce restored cross sections of our study area. Those cross sections allowed us to suggest a model for Mesozoic sedimentary basin formation in the eastern part of the NPZ. Considering first order analysis, on a N-S cross section, the area made of a shoal,the future Agly Massif. It is bordered on its northern side by a north verging detachment fault and on its southern by a south verging detachment fault. On both sides of this shoal are sedimentary basins much thicker towards these detachment faults. However, we must consider the system is not cylindrical. The Agly Massif acts as a transfer zone between two highly subsident basins, the Bas-Agly basin at its north-east and the Boucheville basin at its south-west

Pełny tekst źródłaTrihalomethanes (THMs) and haloacetic acids (HAAs) constitute the only regulated disinfection by-products (DBPs) in various countries. DBPs are compounds formed during drinking water treatment, from the reaction between natural organic matter and the disinfectant. Most DBPs are not regulated, even if they may have more pronounced toxicological effects than regulated ones. There is currently very little information about the occurrence of non-regulated DBPs, particularly in small water systems (SWS). Paradoxically, in many cases, SWS supplied by surface waters lack adequate treatment processes to remove DBP precursors in water subjected to the disinfection process. Their tap water may be more vulnerable to the presence of DBPs. This thesis is dedicated to improving the knowledge of the occurrence of DBPs (especially non-regulated DBPs) in SWS by studying their spatial and temporal variability. To do that, two sampling programs were carried out in 25 SWS during two years (between 2010 and 2012) in Canada. Small systems in the provinces of Newfoundland & Labrador and Quebec were considered. The following DBPs were measured during the study: THMs, HAAs, haloacetonitriles, haloketones and halonitromethanes. The obtained results contribute to a better understanding of the DBP spatio-temporal variation patterns, to establishing models to evaluate their levels and to developing decision-making schemes for simultaneously monitoring various families of DBPs, including non-regulated DBPs.

Pełny tekst źródła@Aclacinomycin (Aclarubicinâ) is a trisaccharide anthracycline anticancer drug active against a wide variety of solid tumors and haematological malignancies. We have evaluated its antimigrative and antiinvasive properties in a Boyden chamber with or without Matrigel and in wound repair assays. Aclacinomycin was demonstrated to inhibit HT-1080 cell migration and invasion while being more potent than the classical anthracycline doxorubicin. This decrease occurred in a dose-dependent manner and without affecting cell proliferation. Importantly, the antiinvasive effect was not associated to a modification in the production of the matrix-degrading enzymes MMP-2 and MMP-9 but rather to changes in cytoskeletal and focal contact formation. Indeed, the drug reduces cell polarity, impairs the actin-mediated membrane ruffling at the leading edge and decreases b1 integrin expression and activation. Dramatic alterations in the distribution of vinculin and in the expression and phosphorylation state of both FAK and Src kinases were also detected. As a conclusion, these data suggest a novel application for this chemotherapeutic agent due to its ability to reduce tumor cell invasion. Combination of aclacinomycin with MMP inhibitors could have therapeutic potential in preventing tumor metastasis

Pełny tekst źródłaOne of the current public health challenges is to propose new strategies for the treatment of neurodegenerative and neuropsychiatric diseases which, due to their multifactorial causes, remain difficult to treat. Over the last decades, a new and promising therapeutic alternative has been developed, with the aim to obtain not only a preventive action but also a curative action: the polypharmacological approach. Several in vivo assays have demonstrated the neuroprotective and pro-cognitive effects caused by the simultaneous modulation of different biological targets involved in the pathophysiology of these disorders, such as MT1, MT2 and 5-HT2C receptors. This project firstly focused on the determination of the structure-activity relationships of an in-house chemical library, constituted of MT1, MT2 and 5-HT2C receptors MTDLs, with additional activity towards 5-HT7 receptors for some derivatives, and with anti-depressive properties. The use of various chemoinformatics and molecular modeling approaches allow then to design and synthesize, by pharmacomodulation, original, potent and selective ligands, on the one hand of MT2 and 5-HT2C receptors, with potential pro-cognitive and antidepressive actions, and on the other hand of 5-HT7 receptors. Among the synthesized molecules, some compounds are of particular interest for the modulation of these targets

Pełny tekst źródłaMacroautophagy is amajor lysosomal catabolic pathway conserved from yeast to human. This process involves the sequestration and delivery of cytoplasmic contents including organelles like mitichondria, into the lysosome for degradation. Moreover, this process promotes cell survival during cellular stress like starvation. However autophagy has been associated to non-apoptotic programmed cell death. The role of autophagy in cell survival or cell death is unclear. This study focuses, in a first part, on the relationship between autophagy and cancer in HT-29 human colon cancer cells through the regulatory action of tumor suppressor gene PTEN on this process. In a second part, the relationship between autophagy and cell death induced in response to non-steroidal anti-inflammatory drugs were assessed on this model

Pełny tekst źródłaOur laboratory has developed from many years a serotoninergic chemolibrary (ATBI program). This chemolibrary contains more than 1500 compounds tested toward the most recently discovered receptors: 5-HT4R, 5-HT5R, 5-HT6R and 5-HT7R. We report here several works carried out in the context of ATBI datasets analysis. After a brief introduction we develop the most important biological aspect of serotoninergic system (chapter II). In chapter III, we deal with evaluation and determination of optimal clustering protocols in relation with our internal chemolibrary. Because a good clustering classification for two of our ATBI datasets is really an issue (5-HT6 and 5-HT7) and in order to understand the reasons, we have developed a new method to extract 2D topological pharmacophores using emerging patterns (chapter IV) and built homology models to study binding mode of 5-HT6 ligands (chapter V). Finally we show how a single amino acid (F7. 38) can explain interspecies (human/rat) selectivity ligands of 5-HT7Rs using homology modelling and site-directed mutagenesis (chapter V)
