In the first instalment of a seven volume series, The Lion, The Witch and The Wardrobe, C. S. Lewis offers us an extraordinary tale involving four children who enter a magical land, Narnia, a myriad of talking animals, a Christ-like lion named Asian and a satanic creature known simply as the Witch (although she claims the title of Queen of Narnia and Empress of the Lone Islands). At the heart of the story is a drama of salvation, or at least saving deliverance. One of the children, Edmund, is held captive by the Witch and will be released only on the condition of Asian's taking Edmund's place. Asian is the ransom for Edmund. In rough outline, Asian's ransoming Edmund and his subsequent resurrection fits the classic ransom theory of the atonement, a theory which can hardly boast of enormous contemporary appeal. I do not think its unpopularity is altogether deserved. Versions of the theory may be found in many patristic writers including Irenaeus, Gregory of Nyssa, Basil the Great, St Ambrose, St Jerome, and Origen. I wish here to defend a modified ransom theory against six of the classic objections which have contributed to its neglect. As C. S. Lewis, the father of Narnia, has presented a rich story of a ransom drama which is more familiar (and sometimes more fun to read) than the Nicene and Prenicene fathers, I will use The Lion, The Witch and The Wardrobe to set forth a bold, unsophisticated ransom theory.

Despite his significance, however, his correspondence remained mostly unpublished up to these days. This paper analyses Diana Wynne Jones's use of the Arthurian tradition in her novel Hexwood and the links she establishes with the contemporary traditions of the fantasy novel for children and science fiction. By employing a complex non-linear narration and a rich network of intertextual allusions ranging from Thomas Mallory and Edmund Spenser to T. H. White, Wynne Jones creates an unusual and successful genre amalgam. The central concept of the novel, a version of virtual reality where individuals adopt false identities and act accordingly, enables a highly uncommon self aware use of motifs adopted from myth and literature.

Em seguida, apresentamos como a fenomenologia heideggeriana se transformou com o apoio da hermenêutica e como ela se distanciou das teses fenomenológicas de seu professor. Por último, mostramos qual a definição que Heidegger atribui à intencionalidade e como a estrutura ontológica chamada cuidado é um aprofundamento deste conceito, o que lhe atribui uma rica significação diante das teorias modernas que ainda estavam bastante evidentes na filosofia de Husserl.

It was believed in the medieval west that incorruption of the body after death was a sign of especial sanctity. Ælfric remarked that England was rich in the incorrupt bodies of saints (Skeat 1900, 333 lines 250–63). Bede dwells on the incorruption of St Æthelthryth of Ely (Plummer 1896, i, 245–6, Book iv ch. 1), besides that of St Cuthbert (ibid., 1, 276–7 Book iv ch. 30), and also mentions the incorruption of St Æthelthryth's sister, Æthelberga (ibid., i, 144 Book iii ch. 9), and of St Fursey (ibid., 1, 168 Book iii ch. 19). A rich critical literature explores the relation between Edmund Burke's theatrical style and his counterrevolutionary argument. Redirecting this line of inquiry, the essay treats Reflections on the Revolution in France (1790) as a histrionic literary performance, arguing that to appreciate its significance we must recover a neglected subtext: a preoccupation with boredom and restlessness. Burke's loyalties are divided: defending England, he counsels against extremes of torpor and excitement. He works to preserve England in a state of settled “repose,” yet his rhetoric reveals a baseline of boredom. Indulging in fantasies of reform and utopia and deploying strategies of tragic hyperbole and self-parody, he mobilizes conventional associations of boredom and revolution to negotiate a new position from which to exercise cultural authority. Textual histrionics do more than contain a revolutionary threat; they establish an alternative theater of boredom.

Edmund Morgan tells the story memorably inAmerican Slavery, American Freedom. Having narrowly evaded death and devastation at the hands of the former indentured servants who comprised the region's lower classes, and still faced with a chronic shortage of labor, the planters began to import African slaves to work the tobacco crops on which their wealth depended. They weren't the first in the New World to try this approach. West Indian sugar planters had already successfully organized the labor on their large plantations along these “racial” lines, and there was sufficient precedent in Western culture at large for associating “blackness” with evil to defuse moral alarm at the practice.

British Burma has never beenadequately or even systematically studied as both students of modern Burmese history and British empire historians have given it relatively short shrift. Nonetheless, imperial rule lasted for nearly five generations and helped to produce the nation which now identifies itself as Myanmar. By the end of the nineteenth century, Burma was crucial to the wider South Asian economy, supplying oil, minerals, teak, and, above all, rice to destinations around the Indian Ocean. Yet, it took three Anglo-Burmese Wars to make Burma a part of British India. These conflicts are largely forgotten but they determined not only the fate of the country, but helped to shape its future trajectories. Military conflict proved more durable than colonization as independence brought with it a situation in which the “state has been continuously at war with the population mapped into its territorial claim” (Callahan 13). Nonetheless, the intellectual and cultural history of British Burma is rich and fascinating: colonial authors made the country their subject matter and they left behind a diverse corpus which bore the stamp of Victorian civilization. The experience of writing about Burma – particularly by those writers who identified with Burmese culture – produced some forgotten masterpieces. However, the dominant British understanding of the country arose from military conflict and occupation; this paper focuses on four British war narratives (which followed each of the Anglo-Burmese Wars) because they disclose more than their recounting of these conflicts might suggest. By exploring the works of John James Snodgrass, Henry Gouger, William F. B. Laurie, and Major Edmond Charles Browne, it will be possible to trace the beginnings of the colonizing narrative which helped to shape British rule. These writers experienced the Anglo-Burmese Wars directly and their narratives illustrate that they were “involuntary sightseers” recording not only the details of conflict, but their assessments of Burma and the Burmese.

The subject matter of this paper is the language and stylistics of selected fragments of „Serce” (“Heart”), a children’s novel by Edmondo de Amicisa in the 19th-century translation into Polish by Maria Konopnicka. With reference to selected translation theories, the conducted analysis covers the strategy adopted by the translator, that is one consisting in familiarising the Polish reader with foreign realities while preserving their originality and “foreignness” (i.e. the simultaneous “domestication” and “foreignisation”). This determines the faithfulness and up-to-dateness of her translation, re-editions of which have been published to this day as it is appreciated for its rich language and accurate rendition of the original style. Due to the similarity to her own writing technique, the translator appropriately refl ects the nature of the work, enriching it with elements of school jargon and colloquial language. Despite the presence of archaisms, Konopnicka’s language has remained comprehensible to the contemporary reader. Background: Lactoferrin (LF) exhibits multiple beneficial biological activities and thus has been used as a health food and additive. To broaden its application in the food industry, the porcine LF (pLF) gene has been engineered into rice to produce recombinant LF (rpLF) for use as a food additive. The iron-binding and antimicrobial activities of rpLF and its positive effects on early weaned piglets have been previously evaluated, yet several features, such as the signal peptide removal, glycosylation sites and antioxidant activity of rpLF, have not been fully characterized. Objective: In this work, the rice-produced rpLF was purified and its biochemical structure and antioxidant activities characterized. Methods: HPLC, Western blot, PAS/VVL/PNA staining, Edman degradation assay, MALDI-TOF, LC-MS/MS and antioxidant activity assays were performed. Results: The results showed that this purified rpLF is a mature form of LF; its signal peptide was correctly removed, and two N-glycosylation sites located at N365 and N472 were identified. The molecular mass heterogeneity of rpLF could be eliminated by treatment with PNGase glycosidase, suggesting that different degrees of N-glycosylation occur in rpLF. A series of assays including the iron chelating activity, reducing power assay, lipid peroxidase activity and radical-scavenging activity showed that the antioxidant activity of rice-produced rpLF was equivalent to that of bovine LF. Conclusion: Rice-produced rpLF was correctly processed post-translationally and displayed antioxidant activity equivalent to that of bovine LF; thus, rice-produced rpLF can be recognized as a plant-based antioxidant to be used as a functional additive in animal feed and for the food industry.

AbstractImaging spectrophotometry in the main nebular lines has been performed on 65 H ɪɪ regions in the ringed galaxy NGC 4736. O/H abundances were derived using the line ratios [O ɪɪɪ]/Hβ and [N ɪɪ]/[O ɪɪɪ] calibrated by Edmunds & Pagel (1984). We show that the O/H scatter in the resonance ring of star forming regions is small, no greater than normally expected in the well-mixed ISM of disks of gas-rich galaxies. The global O/H gradient (−0.046 dex/kpc) in the disk of NGC 4736 is shallower than gradients of normal spirals but comparable to gradients observed in weakly barred spirals. This last result could indicate that radial mixing is or was present in NGC 4736. The oval distortion in the central regions can be responsible for this homogenization but it is also possible that a strong bar was present in the past.

Although there are many antimicrobial proteins in plants, they are not well-explored. Understanding the mechanism of action of plant antifungal proteins (AFPs) may help combat fungal infections that impact crop yields. In this study, we aimed to address this gap by screening Oryza sativa leaves to isolate novel AFPs. We identified a thioredoxin protein with antioxidant properties. Being ubiquitous, thioredoxins (Trxs) function in the redox balance of all living organisms. Sequencing by Edman degradation method revealed the AFP to be O. sativa Thioredoxin m-type isoform (OsTrxm). We purified the recombinant OsTrxm and its cysteine mutant proteins (OsTrxm C/S) in Escherichia coli. The recombinant OsTrxm proteins inhibited the growth of various pathogenic fungal cells. Interestingly, OsTrxm C/S mutant showed higher antifungal activity than OsTrxm. A growth inhibitory assay against various fungal pathogens and yeasts confirmed the pertinent role of cysteine residues. The OsTrxm protein variants penetrated the fungal cell wall and membrane, accumulated in the cells and generated reactive oxygen species. Although the role of OsTrxm in chloroplast development is known, its biochemical and molecular functions have not been elucidated. These findings suggest that in addition to redox regulation, OsTrxm also functions as an antimicrobial agent.

The abundant hydrophobic, proline-glutamine, and histidine-rich (over 90%) amelogenins constitute the major class of proteins in forming extracellular enamel matrix. These are thought to play a major role in the structural organization and mineralization of developing enamel. The present report describes the successful sequencing of the major human amelogenin protein, by use of both Edman degradation and cDNA sequencing. When Edman degradation was used, over 75% of the primary structure of the protein was determined. This sequence was supplemented with cDNA sequencing studies, which revealed the predicted sequence of this protein. Together, they provide the complete sequence of an important human enamel protein. The information complements recent studies on bovine and human amelogenin genes. A comparison between the present results and the protein sequences predicted from the corresponding human amelogenin genomic coding regions and that of cDNA sequences of other species is described.

Summary SAGD is one successful thermal recovery technique applied in the Athabasca and Peace River reservoirs in central and northern Alberta, Canada. In SAGD, steam is injected into a horizontal injection well and is forced outward, losing its latent heat when it comes into contact with the cold bitumen at the edge of a depletion chamber. As a consequence, the viscosity of the bitumen falls several orders of magnitude, its mobility rises several orders of magnitude, and then it flows under gravity toward a horizontal production well located several meters below and parallel to the injection well. Heat-transfer mechanisms are pivotal to the SAGD process. Though heat energy is transferred from steam to reservoir by conduction and convection, heat transfer by convection is not considered in the classic SAGD mathematical models such as Butler's. Researchers such as Butler and Stephens (1981), Reis (1992), Akin (2005), Liang (2005), Nukhaev et al. (2006), and Azad and Chalaturnyk (2010) considered conduction from steam to cold reservoir to be the only heat-transfer component. However, because the heat capacity of water is typically two to five times that of bitumen, convection caused by the mobile condensate flow in the reservoir may contradict these studies. Farouq-Ali (1997) was the first to criticize the assumption that there is only a thermal conduction mechanism in the SAGD process. He pointed out that with so much condensate flowing, convection would be expected to be the dominant heat-transfer mechanism, which can be plausible at high temperatures. In response, Edmunds (1999a) stated that on the basis of the associated change in enthalpy, the heat transfer into a cold reservoir because of convection is probably less than 5% of that because of conduction. Summary SAGD is one successful thermal recovery technique applied in the Athabasca and Peace River reservoirs in central and northern Alberta, Canada. In SAGD, steam is injected into a horizontal injection well and is forced outward, losing its latent heat when it comes into contact with the cold bitumen at the edge of a depletion chamber. As a consequence, the viscosity of the bitumen falls several orders of magnitude, its mobility rises several orders of magnitude, and then it flows under gravity toward a horizontal production well located several meters below and parallel to the injection well. Heat-transfer mechanisms are pivotal to the SAGD process. Though heat energy is transferred from steam to reservoir by conduction and convection, heat transfer by convection is not considered in the classic SAGD mathematical models such as Butler?s. Researchers such as Butler and Stephens (1981), Reis (1992), Akin (2005), Liang (2005), Nukhaev et al. (2006), and Azad and Chalaturnyk (2010) considered conduction from steam to cold reservoir to be the only heat-transfer component. However, because the heat capacity of water is typically two to five times that of bitumen, convection caused by the mobile condensate flow in the reservoir may contradict these studies. Farouq-Ali (1997) was the first to criticize the assumption that there is only a thermal conduction mechanism in the SAGD process. He pointed out that with so much condensate flowing, convection would be expected to be the dominant heat-transfer mechanism, which can be plausible at high temperatures. In response, Edmunds (1999a) stated that on the basis of the associated change in enthalpy, the heat transfer into a cold reservoir because of convection is probably less than 5% of that because of conduction. Miroculis Edmunds represents one of the most species-rich genera amongst Neotropical leptophlebiids comprising 25 valid species, with 18 of them reported from Brazil. Although several species have been described throughout the years, the taxonomy of Miroculis has focused mainly on male imagos, with other semaphoronts often unknown. Here, Miroculis (Miroculis) niltoi sp. nov. is described based on nymphs and imagos from Chapada Diamantina’s Complex, Northeast Brazil. Male imagos of M. (M.) niltoi sp. nov. can be distinguished by the styliger plate being medially cleft with a well-sclerotized ventrally-directed process at the posteromedial margin, while female imagos can be distinguished by the egg guide being well-developed, reaching sternum IX, and sternum IX about as long as wide, with a shallow apical cleft. Together with Miroculis (Miroculis) samba Costa, Almeida & Salles and Miroculis (Miroculis) wolverine Costa, Almeida & Salles, M. (M.) niltoi sp. nov. represents species within Miroculis with male imago exhibiting both extremely long penes and styliger plate. Now, Miroculis comprises 26 valid species, with 19 of them reported from Brazil.

BChE (butyrylcholinesterase) protects the cholinergic nervous system from organophosphorus nerve agents by scavenging these toxins. Recombinant human BChE produced from transgenic goat to treat nerve agent intoxication is currently under development. The therapeutic potential of BChE relies on its ability to stay in the circulation for a prolonged period, which in turn depends on maintaining tetrameric quaternary configuration. Native human plasma BChE consists of 98% tetramers and has a half-life (t½) of 11–14 days. BChE in the neuromuscular junctions and the central nervous system is anchored to membranes through interactions with ColQ (AChE-associated collagen tail protein) and PRiMA (proline-rich membrane anchor) proteins containing proline-rich domains. BChE prepared in cell culture is primarily monomeric, unless expressed in the presence of proline-rich peptides. We hypothesized that a poly-proline peptide is an intrinsic component of soluble plasma BChE tetramers, just as it is for membrane-bound BChE. We found that a series of proline-rich peptides was released from denatured human and horse plasma BChE. Eight peptides, with masses from 2072 to 2878 Da, were purified by HPLC and sequenced by electrospray ionization tandem MS and Edman degradation. All peptides derived from the same proline-rich core sequence PSPPLPPPPPPPPPPPPPPPPPPPPLP (mass 2663 Da) but varied in length at their N- and C-termini. The source of these peptides was identified through database searching as RAPH1 [Ras-associated and PH domains (pleckstrin homology domains)-containing protein 1; lamellipodin, gi:82581557]. A proline-rich peptide of 17 amino acids derived from lamellipodin drove the assembly of human BChE secreted from CHO (Chinese-hamster ovary) cells into tetramers. We propose that the proline-rich peptides organize the 4 subunits of BChE into a 340 kDa tetramer, by interacting with the C-terminal BChE tetramerization domain.

Before Joseph Lister's landmark Lancet publications on the use of carbolic acid wound dressings in 1867, surgeons Jules Lemaire in France and Enrico Bottini in Italy had already used carbolic acid on hundreds of patients to control suppurative wounds. After Friedlieb Runge isolated it from coal tar in 1834, a number of scientists recognized the efficacy of carbolic acid in preventing decay and neutralizing the stench of dead animals and human cadavers. Frederick Calvert, Alexander McDougall, and Angus Smith in Manchester promoted a powdered form of carbolic acid as a deodorizing agent to treat municipal sewage across the United Kingdom, most notably during London's famous “Great Stink” of 1858. Edmond Corne in France introduced his formulation, which Alfred-Armand-Louis-Marie Velpeau, Ferdinand LeBeuf, and Lemaire adapted for clinical use in 1859. Lemaire wrote extensively on carbolic acid and its surgical application in three publications from 1860 to 1862. In 1866, Bottini published his experience of 600 cases where it was used. In 1865, Lister began to use carbolic acid in open fractures after Thomas Anderson, his colleague in agricultural chemistry at the University of Glasgow, told him about its use in Carlisle sewage works. This article traces the rich history of carbolic acid from an unknown compound in coal to the cornerstone of Listerism in late-19th–century operating rooms.

This work reports the purification and functional characterization of BmooPAi, a platelet-aggregation-inhibiting factor fromBothrops moojenisnake venom. The toxin was purified by a combination of three chromatographic steps (ion-exchange on DEAE-Sephacel, molecular exclusion on Sephadex G-75, and affinity chromatography on HiTrap™ Heparin HP). BmooPAi was found to be a single-chain protein with an apparent molecular mass of 32 kDa on 14% SDS-PAGE, under reducing conditions. Sequencing of BmooPAi by Edman degradation revealed the amino acid sequence LGPDIVPPNELLEVM. The toxin was devoid of proteolytic, haemorrhagic, defibrinating, or coagulant activities and induced no significant oedema or hyperalgesia. BmooPAi showed a rather specific inhibitory effect on ristocetin-induced platelet aggregation in human platelet-rich plasma, whereas it had little or no effect on platelet aggregation induced by collagen and adenosine diphosphate. The results presented in this work suggest that BmooPAi is a toxin comprised of disintegrin-like and cysteine-rich domains, originating from autolysis/proteolysis of PIII SVMPs fromB. moojenisnake venom. This toxin may be of medical interest because it is a platelet aggregation inhibitor, which could potentially be developed as a novel therapeutic agent to prevent and/or treat patients with thrombotic disorders.

The complete amino acid sequence of a structural protein isolated from pharate cuticle of the locust Locusta migratoria was determined. The protein has an unusual amino acid composition: 42% of the residues are alanine and only 14 of the 20 common amino acid residues are present. The primary structure consists of regions enriched in particular amino acid residues. The N-terminal region and a region close to the C-terminus are enriched in glycine. The rest of the protein is dominated by alanine, except for two short regions enriched in hydrophilic residues. Almost all the proline residues are situated in the alanine-rich regions in a conserved sequence ‘A-A-P-A/V’. An internal duplication has taken place covering most of the protein except for the glycine-rich regions. Owing to the unusual features of the protein a combination of automated Edman degradations and plasma-desorption m.s. was used to determine the complete sequence. The protein does not show sequence homology to other proteins, but proteins divided into regions enriched in the same kind of amino acid residues have been isolated from other insect structures.

G-BOIS (“Flora Orientalis Herbarium”), the result of many years (1844–1888) of titanic work by an outstanding botanist from Geneva Pierre-Edmond Boissier, is the most important taxonomic and nomenclature source of data on the rich flora of the territory from Greece to Middle Asia and the borders of India. It was isolated from an extensive Boissier’s private herbarium after the latter was included in the collection of Conservatoire et Jardin Botaniques and kept in a special underground bunker. A material for 366 species and 47 infraspecific taxa of the Umbelliferae is stored in G-BOIS. Among lectotypes, the vast majority relate to the Boissier taxa; a small minority belong to taxa of other authors (their types and lectotypes are in other herbaria). The materials of other herbaria, Geneva and non-Geneva (Geneva General Herbarium, De Candolle Prodromus Herbarium, K, LE, P, JE, W, B and others) were also checked. Among the Umbelliferae material kept in G-BOIS, lectotypes of 255 names were designated here and those of 107 names have been designated before, including lectotypes selected prior 2011 as types and holotypes which were corrected to lectotypes (89 names). Besides the designated lectotypes and isolectotypes, G-BOIS also contains holotypes of 44 Boissierian Umbelliferae names and syntypes of 81 names. Additionally, holotypes, lectotypes and isolectotypes (in total for 168 names) were revealed in Geneva General herbarium (G). One new species name (Prangos iranica nom. nov.) has been proposed here in the genus Prangos.

Ants are a biodiverse group of insects that have evolved toxic venom containing many undiscovered bioactive molecules. In this study, we found that the venom of the ruby ant Myrmica rubra is a rich source of peptides. LC-MS analysis revealed the presence of 142 different peptides varying in molecular weight, sequence length, and hydrophobicity. One of the most abundant peaks was selected for further biochemical and functional characterization. Combined Edman degradation and de novo peptide sequencing revealed the presence of a novel decapeptide (myrmicitoxin) with the amino acid sequence NH2-IDPKLLESLA-CONH2. The decapeptide was named U-MYRTX-MRArub1 and verified against a synthetic standard. The amidated peptide was tested in a synthetic form to determine the antimicrobial activity towards the bacterial pathogens and insecticidal potential against pea aphids (Acyrthosiphon pisum). This peptide did not show antimicrobial activity but it significantly reduced the survival of aphids. It also increased the sensitivity of the aphids to two commonly used chemical insecticides (imidacloprid and methomyl). Since ant venom research is still in its infancy, the findings of this first study on venom peptides derived from M. rubra highlight these insects as an important and rich source for discovery of novel lead structures with potential application in pest control.

RESUMOA pesquisa desenvolvida teve como alvo o fenômeno das relações homem-animal, investigando as diferentes modalidades e manifestações dessa relação, bem como o processo de criação do vínculo homem-animal, dando um enfoque em como essas relações e vínculos permitem um processo terapêutico e quais as consequências e desdobramentos desse processo (TAAs). A pesquisa é qualitativa e consistiu em uma pesquisa bibliográfica, na qual foram evidenciados na literatura diferentes modalidades de vínculos e suas implicações. O modelo metodológico utilizado foi a fenomenologia Husserliana, proposta e fundada pelo filósofo e matemático Edmund Husserl. A fenomenologia consiste em um modelo de pesquisa radical e rigoroso que visa constituir uma ciência universal das essências através da suspensão dos apriores e da crítica à postura ingênua/natural, evidenciando o fenômeno tal qual ele se mostra. Desse modo, a pesquisa teve como resultado a descrição e compreensão de diferentes modalidades da relação homem-animal com finalidades terapêuticas, sendo a primeira uma modalidade que anula o animal enquanto outro e o assume no lugar de um objeto, recurso e instrumento, perdendo qualquer implicação de uma conduta ética sobre eles, o que resulta na manifestação de atos de crueldade, abandono, experimentações cruéis e sem objetivos e etc; enquanto a segunda apresenta um modo de relação na qual o animal é reconhecido enquanto outro, e esse reconhecimento permite a manifestação de empatia, alteridade e respeito. Fora isso, as duas modalidades apresentam diferentes horizontes de possibilidade para o desdobramento e para a compreensão do processo terapêutico. Enquanto a primeira, condizente com o modo de pesquisa das ciências naturais, foca em aspectos mensuráveis, tentando justificar o evidente bem que os animais nos causam, e perdendo de vista uma série de fenômenos; a segunda consiste na apreensão e compreensão desse fenômeno tal como ele se manifesta e evidência na vida, permitindo-nos intuir uma série de fenômenos, outrora encobertos, para um processo terapêutico mais rico, consciente e eficiente. The glycoprotein (GP) Ib-IX complex on the surface of human platelets functions as the von Willebrand factor receptor and mediates von Willebrand factor-dependent platelet adhesion to blood vessels. GPIX is a relatively small (Mr, 17,000) protein that may provide for membrane insertion and orientation of the larger component of the complex, GPIb (Mr, 165,000). Using antibody screening, we cloned a cDNA encoding GPIX from a human erythroleukemia cell cDNA library constructed in phage lambda gt11. Lacking a 5' untranslated region and start codon, the cDNA sequence includes 604 nucleotides, beginning with 495 bases at the 5' end coding for 165 amino acids, followed by a stop codon and 106 noncoding bases at the 3' end. By Northern blot analysis, the GPIX cDNA hybridizes with a single 1.0-kilobase species of platelet poly(A)+ RNA. Translation of the cDNA sequence gives a predicted protein sequence beginning with a truncated putative signal sequence of 5 amino acid followed by a sequence of 17 amino acids matching that determined directly by Edman degradation of intact GPIX. The predicted amino acid sequence of mature GPIX includes an NH2-terminal extracytoplasmic domain of 134 residues, a transmembrane domain of 20 residues, 6 intracytoplasmic residues, and 1 N-linked glycosylation site. GPIX contains a leucine-rich glycoprotein (LRG) sequence of 24 amino acids similar to conserved LRG sequences in GPIb and other proteins from humans, Drosophila, and yeast. "Flanking" sequences of approximately 22 amino acids are present at the NH2 and/or COOH sides of the "central" LRG sequence(s) in GPIX, GPIb, and the other human and Drosophila members of the LRG family. The role of the flank-LRG center-flank structure in the evolution and function of the LRG proteins remains to be defined.

Peptides H-TPPQC(Acm)FTV-NH2 (I) and H-VSVPC(Acm)QSASSAS-NH2 (III) were prepared by the solid phase method. Their oxidation with iodine afforded hexadecapeptide II, tetracosapeptide IV and eicosapeptide VI. The disulfide peptides II, IV and VI are designed according to the sequence of the processing loop in human cathepsin D. The purity of the peptides was determined by analytical HPLC and capillary zone electrophoresis. In addition to the expected [M + H]+ ion, FAB MS of HPLC-pure tetracosapeptide IV exhibited a molecular ion with the same relative molecular mass as the starting dodecapeptide III, in spite of clean HPLC separation of III and IV. Free-flow zone electrophoresis of IV separated peptide V, isomeric with III. Mass spectra, amino acid analysis and Edman sequencing revealed that the peptide V is a product of iodine-mediated S-->O shift of Acm group in the serine-rich peptide III. Daughter-ion spectra of protonated molecules, recorded after collision-induced dissociation, have shown that the Acm moiety is bonded to Ser 9 or Ser 10.

A pageant of curiosities and dynamic images inhabits the margins of manuscripts, sometimes ornamenting, sometimes competing, sometimes commenting on the text they surround. They are a commonplace of codicological study, even more so since the publication of Lilian Randall's Images in the Margins of Gothic Manuscripts in 1966 and of Michael Camille's Image on the Edge in 1992, which has done much to bring us to understand and interpret this panoply in ink and paint. The images these writers treat, of course, are late, and it is the more rich and entertaining margins that command the attention, such as the copulating figures in the top margin of a book of hours (New York, Pierpont Morgan Library, M. 754, f. 65v), or the Christ-like figure showing forth his buttocks to a spear-wielding, monkey-like creature mounted on an ostrich in the Rutland Psalter (London, British Library, Additional 62,925, ff. 66v-67r). The margins of Anglo-Saxon manuscripts seem, by comparison, rather barren fields. There are certainly some notable exceptions, such as the illustrations in the Bury St Edmunds Psalter (Rome, Vatican Library, MS Reg. lat. 12), or perhaps those in the Exeter Book (Exeter Cathedral Library MS 3501, ff. 78r, 87v, 123r). But in truth, such examples are few in number and rather sober occasions in any case, lacking the spirited, immensely entertaining, and often surprising creations that fill the margins of later manuscripts. But if the Anglo-Saxons possessed vastly different conceptions of space, margins, and response to text than later generations, we nevertheless often find in their manuscripts a quiet, typically hidden or overlooked world of text and image entered into manuscripts by Anglo-Saxon and later users of these codices.

It is impossible to describe all of this on just a few pages, which is why the aim of my paper is to describe the most outstanding expeditions, those that completely changed our thinking about Himalayan mountaineering and showed that we can deceive not only our bodies but also our subconscious.The first attempts to ascend an eight-thousander were made as early as in the 1920s; unfortunately, they all failed. Owing to the harsh conditions in the mountains, the first successful ascent of an eight-thousander did not take place until 1950, when Annapurna was ascended. The first Polish expedition was organised in 1939. Unfortunately, the Second World War and the political situation in Poland prevented Polish climbers from making further attempts for many years. The political situation in the country made it impossible for Poles to travel abroad. But Himalayan mountaineering at the time was developing very rapidly. The Poles, hungry for success, wanted to go down in history. Given the fact that all eight-thousanders had already been ascended, the Poles began a new chapter — winter Himalayan mountaineering, challenging Edmund Hillary’s assertion that in winter no form of life had a chance to survive over 7000 metres above sea level. In my paper I focus on presenting the most remarkable achievements. I describe the Golden Age of Polish Himalayan mountaineering, like the first winter ascent of Everest or Krzysztof Wielicki’s extraordinary one-day ascent of Broad Peak. There is also room for expeditions featuring Jerzy Kukuczka, an icon of Himalayan mountaineering. Climbing two eight-thousanders in one winter season and establishing a new route on K2 are still unbeaten feats. A part of my paper is devoted to women. The first all-female expeditions headed by Wanda Rutkiewicz were also a Polish domain. The number of great achievements in the mountains are truly numerous. Each of them in described in the paper, as is the death of the greatest Himalayan climbers, which led to a crisis and revisions. The paper ends with a fragment concerning an attempt to revive Polish Himalayan mountaineering.

In this paper, we describe the purification/characterization of BmooAi, a new toxin fromBothrops moojenithat inhibits platelet aggregation. The purification of BmooAi was carried out through three chromatographic steps (ion-exchange on a DEAE-Sephacel column, molecular exclusion on a Sephadex G-75 column, and reverse-phase HPLC chromatography on a C2/C18 column). BmooAi was homogeneous by SDS-PAGE and shown to be a single-chain protein of 15,000 Da. BmooAi was analysed by MALDI-TOF Spectrometry and revealed two major components with molecular masses 7824.4 and 7409.2 as well as a trace of protein with a molecular mass of 15,237.4 Da. Sequencing of BmooAi by Edman degradation showed two amino acid sequences: IRDFDPLTNAPENTA and ETEEGAEEGTQ, which revealed no homology to any known toxin from snake venom. BmooAi showed a rather specific inhibitory effect on platelet aggregation induced by collagen, adenosine diphosphate, or epinephrine in human platelet-rich plasma in a dose-dependent manner, whereas it had little or no effect on platelet aggregation induced by ristocetin. The effect on platelet aggregation induced by BmooAi remained active even when heated to 100°C. BmooAi could be of medical interest as a new tool for the development of novel therapeutic agents for the prevention and treatment of thrombotic disorders.

ABSTRACT A 30-residue N-terminally acetylated peptide derived from the N-terminal part of histone H1 was identified as the dominant antimicrobial peptide in skin mucus from Atlantic salmon (Salmo salar). The peptide (termed salmon antimicrobial peptide [SAMP H1]) was purified to homogeneity by a combination of reversed-phase and cation-exchange chromatographies. By Edman degradation of the deacetylated peptide and by sequencing of the PCR-amplified DNA that encodes the peptide, the complete amino acid sequence was determined to be AEVAPAPAAAAPAKAPKKKAAAKPKKAGPS. The theoretical molecular weight of N-terminally acetylated SAMP H1 was calculated to be 2,836, which is the same as that determined by matrix-assisted laser desorption ionization mass spectrometry. The peptide was active against both gram-negative and -positive bacteria. The N-terminal acetyl group was not necessary for activity since deacetylation did not reduce the activity. A synthetic peptide whose sequence was identical to that of the isolated fragment was initially inactive but could be activated by binding it to a cation-exchange column. Treatment of the synthetic peptide when it was bound to the exchange column with peptidylproline cis-trans-isomerase increased the amount of active peptide, indicating that isomerization of the proline peptide bond(s) was necessary for activation of the synthetic peptide. Comparison of the active and inactive forms by circular dichroism and chromatographic analyses suggests that the active form, both the natural and the synthetic forms, is more structured, condensed, and rigid than the inactive form, which has a more nonstructured conformation. This work shows for the first time the importance of proline isomers in the activity of an antimicrobial peptide.

Abstract We have sequenced 5-aminolevulinate dehydratase (ALAD; EC 2.4.1.24) of a plant. A fulllength cDNA clone (1727 bp) encoding this enzyme has been identified by immunoscreening a lambda gt 11 cDNA library of spinach. ALAD is not a plant-specific enzyme; however, the plant enzyme differs from the well known ALAD enzymes of bacteria, yeast and animals in structural and biochemical properties and in that it is located in the plastid. Differences and homologies can be traced back to the molecular level. The mature ALAD subunit, whose N-terminus was determined by automatic Edman degradation, is a protein of 367 amino acid residues and has a Mr of 40,132. This figure is in the range of molecular weights of non-plant ALADs. The active centre is highly conserved and the same is true for the ion-binding domain, except that 4 cysteines of the non-plant enzymes (binding Zn2+) have disappeared and a total of 6 aspartic acids meets the demands of Mg2+-binding. However, there are more distinct differences. Apart from a transit sequence of 56 amino acids targeting the plastid, the N-terminal part of the mature plant enzyme differs considerably from non-plant ALAD enzymes. It is rich in prolines and hydroxylated amino acids. The apparent Mr on SDS-PAGE is 45,000 or higher, but up to now posttranslational modifications have not been found.

The myristoylated alanine-rich C-kinase substrate (MARCKS) purified from brain was recently characterized as a proline-directed kinase(s) substrate in vivo [Taniguchi, Manenti, Suzuki and Titani (1994) J. Biol. Chem. 269, 18299-18302]. Here we have investigated the phosphorylation of MARCKS by various cyclin-dependent kinases (Cdks) in vitro. We established that Cdk2, Cdk4 and, to a smaller extent, Cdk1 that have been immunoprecipitated from cellular extracts phosphorylate MARCKS. Comparison of MARCKS phosphorylation by protein kinase C (PKC) and by the purified cyclin E-Cdk2 complex suggested that two residues were phosphorylated by Cdk2 under these conditions. To identify these sites, Cdk2-phosphorylated MARCKS was digested with lysyl endoprotease and analysed by electrospray MS. Comparison with the digests obtained from the unphosphorylated protein demonstrated that two peptides, Gly12-Lys30 and Ala138-Lys152, were phosphorylated by cyclin E-Cdk2. The identity of these peptides was confirmed by automatic Edman degradation. On the basis of the consensus phosphorylation sequence described for Cdk2, and on MS/MS analysis of the Ala138-Lys152 peptide, we concluded that Ser27, one of the phosphorylation sites identified in vivo, and Thr150 were the Cdk2 targets in vitro. None of the other sites described in vivo were phosphorylated in these conditions. Interestingly, a preliminary phosphorylation of MARCKS by PKC improved the initial rate of phosphorylation by Cdk2 without modifying the number of sites concerned. In contrast, phosphorylation of MARCKS by Cdk2 did not significantly affect further phosphorylation by PKC.

Low-molecular-mass Arg-Gly-Asp (RGD)-containing polypeptides were isolated from the venom of Trimeresurus elegans by a simple two-step procedure consisting of membrane filtration and reverse-phase HPLC. A combination of electrospray MS, fast-atom bombardment MS and Edman degradation allowed us to ascertain the presence in the venom of different isoforms and to determine their primary structures. The amino acid sequences resembled the structure of elegantin, the only disintegrin previously reported from the T. elegans venom [Williams, Rucinski, Holt and Niewiarowski (1990) Biochim. Biophys. Acta 1039, 81–89]. MS analyses indicated the occurrence of differential proteolytic processing at both the N-terminus and the C-terminus of the polypeptide chains. The amino acid sequence alignment of the elegantin isoforms with known components of the disintegrin family demonstrated the complete conservation of the 12 cysteine residues involved in disulphide bridges. Molecular modelling of elegantins predicted an overall folding of these molecules quite similar to that reported for the kistrin solution structure. The newly identified polypeptide isoforms strongly inhibited ADP-induced aggregation in both human and canine platelet-rich plasma but showed a different species-dependent specificity. These molecules were also able to inhibit B16-BL6 murine melanoma cell adhesion to immobilized fibronectin. The comparison of the structures and biological activities of elegantin isoforms and kistrin allowed us to highlight some structural features that, in addition to the RGD locus, might be involved in the interaction of these snake-venom polypeptides with the integrin receptors on the platelet and cell surface.

ABSTRACT Salivary proline-rich proteins (PRPs) attach commensal Actinomyces and Streptococcus species to teeth. Here, gel filtration, mass spectrometry and Edman degradation were applied to show the release of a pentapeptide, RGRPQ, from PRP-1 upon proteolysis by Streptococcus gordonii. Moreover, synthetic RGRPQ and derivatives were used to investigate associated innate properties and responsible motifs. The RGRPQ peptide increased 2.5-fold the growth rate of S. gordonii via a Q-dependent sequence motif and selectively stimulated oral colonization of this organism in a rat model in vivo. In contrast, the growth of Streptococcus mutans, implicated in caries, was not affected. While the entire RGRPQ sequence was required to block sucrose-induced pH-decrease by S. gordonii and S. mutans, the N-terminal Arg residue mediated the pH increase (i.e., ammonia production) by S. gordonii alone (which exhibits Arg catabolism to ammonia). Strains of commensal viridans streptococci exhibited PRP degradation and Arg catabolism, whereas cariogenic species did not. The RGRPQ peptide mediated via a differential Q-dependent sequence motif, adhesion inhibition, and desorption of PRP-1-binding strains of A. naeslundii genospecies 2 (5 of 10 strains) but not of S. gordonii (n = 5). The inhibitable A. naeslundii strains alone displayed the same binding profile as S. gordonii to hybrid peptides terminating in RGRPQ or GQSPQ, derived from the middle or C-terminal segments of PRP-1. The present findings indicate the presence of a host-bacterium interaction in which a host peptide released by bacterial proteolysis affects key properties in biofilm formation.

Membrane cofactor protein (MCP), a regulatory molecular of the complement system with cofactor activity for the factor I-mediated inactivation of C3b and C4b, is widely distributed, being present on leukocytes, platelets, endothelial cells, epithelial cells, and fibroblasts. MCP was purified from a human T cell line (HSB2) and the NH2-terminal 24-amino acid sequence obtained by Edman degradation. An oligonucleotide probe based on this sequence was used to identify a clone from a human monocytic (U937) cDNA library. Nucleotide sequencing showed a 43-bp 5'-untranslated region, an open reading frame of 1,152 bp, and a 335-bp 3'-untranslated region followed by a 16-bp poly(A) track. The deduced full-length MCP protein consists of a 34-amino acid signal peptide and a 350-amino acid mature protein. The protein has, beginning at the NH2 terminus, four approximately 60-amino acid repeat units that match the consensus sequence found in a multigene family of complement regulatory proteins (C3b-receptor or CR1, C3d-receptor or CR2, decay-accelerating factor, C4-binding protein, and factor H), as well as several other complement and non-complement proteins. The remainder of the MCP protein consists of 25 amino acids that are rich in serine and threonine (probable site of heavy O-linked glycosylation of MCP), 17 amino acids of unknown significance, and a 23-amino acid transmembrane hydrophobic region followed by a 33-amino acid cytoplasmic tail. The MCP gene was localized to human chromosome 1, bands 1q31-41, by analysis of human x rodent somatic cell hybrid clones and by in situ hybridization. This same genetic region contains the multigene family of complement-regulatory proteins, which is thereby enlarged to include the functionally and structurally related MCP.

ABSTRACT Pseudomonas lemoignei is equipped with at least five polyhydroxyalkanoate (PHA) depolymerase structural genes (phaZ1 to phaZ5) which enable the bacterium to utilize extracellular poly(3-hydroxybutyrate) (PHB), poly(3-hydroxyvalerate) (PHV), and related polyesters consisting of short-chain-length hxdroxyalkanoates (PHASCL) as the sole sources of carbon and energy. Four genes (phaZ1,phaZ2, phaZ3, and phaZ5) encode PHB depolymerases C, B, D, and A, respectively. It was speculated that the remaining gene, phaZ4, encodes the PHV depolymerase (D. Jendrossek, A. Frisse, A. Behrends, M. Andermann, H. D. Kratzin, T. Stanislawski, and H. G. Schlegel, J. Bacteriol. 177:596–607, 1995). However, in this study, we show that phaZ4 codes for another PHB depolymeraes (i) by disagreement of 5 out of 41 amino acids that had been determined by Edman degradation of the PHV depolymerase and of four endoproteinase GluC-generated internal peptides with the DNA-deduced sequence of phaZ4, (ii) by the lack of immunological reaction of purified recombinant PhaZ4 with PHV depolymerase-specific antibodies, and (iii) by the low activity of the PhaZ4 depolymerase with PHV as a substrate. The true PHV depolymerase-encoding structural gene, phaZ6, was identified by screening a genomic library of P. lemoigneiin Escherichia coli for clearing zone formation on PHV agar. The DNA sequence of phaZ6 contained all 41 amino acids of the GluC-generated peptide fragments of the PHV depolymerase. PhaZ6 was expressed and purified from recombinant E. coliand showed immunological identity to the wild-type PHV depolymerase and had high specific activities with PHB and PHV as substrates. To our knowledge, this is the first report on a PHASCLdepolymerase gene that is expressed during growth on PHV or odd-numbered carbon sources and that encodes a protein with high PHV depolymerase activity. Amino acid analysis revealed that PhaZ6 (relative molecular mass [M r], 43,610 Da) resembles precursors of other extracellular PHASCLdepolymerases (28 to 50% identical amino acids). The mature protein (M r, 41,048) is composed of (i) a large catalytic domain including a catalytic triad of S136, D211, and H269 similar to serine hydrolases; (ii) a linker region highly enriched in threonine residues and other amino acids with hydroxylated or small side chains (Thr-rich region); and (iii) a C-terminal domain similar in sequence to the substrate-binding domain of PHASCL depolymerases. Differences in the codon usage of phaZ6 for some codons from the average codon usage of P. lemoignei indicated thatphaZ6 might be derived from other organisms by gene transfer. Multialignment of separate domains of bacterial PHASCL depolymerases suggested that not only complete depolymerase genes but also individual domains might have been exchanged between bacteria during evolution of PHASCLdepolymerases.

A review article devoted to the book of Andrzej Blikle – Doktryna jakości. Rzecz o skutecznym zarządzaniu. As pointed out by the Author, the book is a case of a work rare on the Polish publishing market, written by an outstanding scientist, who successfully runs a business activity. The combination of practical experience with theoretical knowledge gave a result that may be satisfying both for practitioners as well as theorists, and also those who want to get to know the ins and outs of an effective and efficient business management. The Author of the review believes that it is an important voice for shaping an inclusive socio-economic system, which constitutes a value in itself. Although the book is mainly concerned with business management, its message has a much wider dimension and is concerned with real measures of wealth, money and people’s lives. The book was awarded The SGH Collegium of Business Administration Award “For the best scientific work in the field of business administration in the years 2014-2015”. Andrzej Jacek Blikle Doktryna jakości. Rzecz o skutecznym zarządzaniu (The Doctrine of Quality. On Effective Management) Gliwice, Helion Publishing Company, 2014, p. 546 Introduction One of the distinctive features of the contemporary economy and contemporary world is a kind of obsession of quantity which is related to thoughtless consumerism, unfavourable to the care for the quality of the work and the quality of the produced and consumed goods and services. It is accompanied by culture (or rather non-culture) of singleness. Therefore, the book The Doctrine of Quality by Andrzej Blikle is like a breath of fresh air. It is a different perspective on the economy and the model of operation of enterprises, on the model of work and life of people. A. Blikle proves that it can be done otherwise. He proves it on the basis of careful studies of the source literature – as expected from a professor of mathematics and an economist, but also on the basis of his own experience gained during the scientific and educational work, and most of all through the economic practice. In the world governed by the obsession of quantity, characterised by fragility, shortness of human relationships, including the relationship of the entrepreneur – employee, A. Blikle chooses durability of these relations, creativity, responsibility, quality of work and production, and ethics. The Doctrine of Quality is a rare example of the work on the Polish publishing market, whose author is a prominent scientist, successfully conducting a business activity for more than two decades, which has contributed to the development of the family company – a known confectionery brand “A. Blikle”. The combination of practical experience with theoretical knowledge gave a result that may be satisfying both for practitioners as well as theorists, and also those who want to get to know the ins and outs of an effective and efficient business management, or develop the knowledge on this topic. In an attractive, clear narrative form, the author comprehensively presents the complexities of business management, indicating the sources of success, but also the reasons and the foundations of failures. At the same time, he presents these issues with an interdisciplinary approach, which contributes to thoroughness of the arguments and deeper reflections. Holism, typical to this book, is also expressed in the focus of A. Blikle not only on the economic, but also on social and ecological issues. Here, the author points to the possibility and need of reconciliation of the economic interests with social interests, and the care for the public good. Analyses of this subject are presented using the achievements of many areas of studies, in addition to economic sciences, including mathematics, sociology, psychology, medicine, and others. This gives a comprehensive picture of the complexity of business management – taking into account its close and distant environment. There are no longueurs in the book, although extensive (over 500 pages), or lengthy, or even unnecessary reasoning overwhelming the reader, as the text is illustrated with a number of examples from practice, and coloured with anecdotes. At the same time, the author does not avoid using expressions popular in the world of (not only) business. He proves that a motivational system which is not based on the approach of “carrot and stick” and without a devastating competition of a “rat race” is possible. The author supports his arguments with references not only to the interdisciplinary scientific achievements, but also to the economic historical experiences and to a variety of older and newer business models. There is a clear fascination with the reserves of creativity and productivity in the humanization of work. In fact, the author strongly exposes the potential of productivity and creativity in creating the conditions and atmosphere of work fostering elimination of fear of the future. He shows that such fear destroys creativity. It is not a coincidence that A. Blikle refers to the Fordist principles, including the warning that manufacturing and business do not consist of cheap buying and expensive selling. He reminds that Henry Ford, a legendary creator of the development of the automotive industry in the United States, put serving the public before the profit. The Doctrine of Quality is at the same time a book – proof that one of the most dangerous misconceptions or errors in the contemporary understanding of economics is finding that it is a science of making money, chremastics. Edmund Phelps and others warned against this in the year of the outbreak of the financial crisis in the USA in 2008, reminding that economics is not a science of making money but a science of relations between the economy and social life [Phelps, 2008]. Economics is a science of people in the process of management. Therefore, by definition, it applies to social values and ethos. Ethos is a general set of values, standards and models of proceedings adopted by a particular group of people. In this sense, ethos and economics as a science of people in the process of management are inseparable. Detaching economics from morality is in contradiction to the classical Smithian concept of economics, as Adam Smith combined the idea of the free market with morality. He treated his first work, The Theory of Moral Sentiments, as an inseparable basis for deliberations on the nature and causes of the wealth of nations, which was the subject of the subsequent work of this thinker [Smith, 1989; Smith, 2012]. Identifying economics with chremastics would then mean that all actions are acceptable and desired, if their outcome is earnings, profit, money. The book of A. Blikle denies it. It contains a number of case studies, which also stimulate broader reflections. Therefore, and also due to the features indicated above, it can be a very useful teaching aid in teaching entrepreneurship and management. The appearance of a book promoting the doctrine of quality and exposing the meaning of ethos of work is especially important because today the phenomenon of product adulteration becomes increasingly widespread, which is ironically referred to in literature as the “gold-plating” of products [Sennett, 2010, pp. 115-118], and the trend as “antifeatures”, that is intentionally limiting the efficiency and durability of products of daily use to create demand for new products. A model example of antifeature is a sim-lock installed in some telephones which makes it impossible to use SIM cards of foreign operators [Rohwetter, 2011, p. 48; Miszewski, 2013]. These types of negative phenomena are also promoted by the development of systemic solutions aiming at the diffusion of responsibility [Sennett, 2010]. This issue is presented among others by Nassim N.N. Taleb, in the book with a meaningful title Antifragile: How to Live in a World We Don’t Understand? The author proves that the economy and society lose their natural durability as a result of the introduction of numerous tools and methods of insurance against risks, but mostly by shifting the burden of risks on other entities [Taleb, 2012]. N.N. Taleb illustrates his arguments with numerous convincing examples and references to history, recalling, inter alia, that in ancient times there was no building control, but the constructors, e.g. of bridges had to sleep under them for some time after their construction, and the ancient aqueducts are still working well until today. So, he shows that a contemporary world, focused on quantitative effects, does not create a sound base for ethical behaviours and the care for the quality of work and manufacturing. Andrzej Blikle points to the need and possibility of opposing this, and opposing to what the Noble Price Winner for Economics, Joseph Stiglitz described as avarice triumphs over prudence [Stiglitz, 2015, p. 277]. The phrase emphasised in the book “Live and work with a purpose” is the opposition to the dangerous phenomena listed above, such as for example antifeatures. convincing that although the business activity is essentially focused on profits, making money, limited to this, it would be led to the syndrome of King Midas, who wanted to turn everything he touched into gold, but he soon realised that he was at risk of dying of starvation, as even the food turned into gold. What distinguishes this book is that almost every part of it forces in-depth reflections on the social and economic relations and brings to mind the works of other authors, but at the same time, creates a new context for them. So, A. Blikle clearly proves that both the economy and businesses need social rooting. This corresponds to the theses of the Hungarian intellectual Karl Polanyi, who in his renowned work The Great Transformation, already in 1944 argued that the economy is not rooted in the social relations [Polanyi, 2010, p. 70]. He pointed to the risk resulting from commodification of everything, and warned that allowing the market mechanism and competition to control the human life and environment would result in disintegration of society. Although K. Polanyi’s warnings were concerned with the industrial civilization, they are still valid, even now – when the digital revolution brings fundamental changes, among others, on the labour market – they strengthen it. The dynamics of these changes is so high that it seems that the thesis of Jeremy Rifkin on the end of work [Rifkin, 2003] becomes more plausible. It is also confirmed by recent analyses included in the book of this author, concerning the society of zero marginal cost and sharing economy [Rifkin, 2016], and the analyses concerning uberisation [Uberworld, 2016]. The book of Andrzej Blikle also evokes one of the basic asymmetries of the contemporary world, which is the inadequacy of the dynamics and sizes of the supply of products and services to the dynamics and sizes of the demand for them. Insufficient demand collides with the rapidly increasing, as a result of technological changes, possibilities of growth of production and services. This leads to overproduction and related therewith large negative implications, with features of wasteful economy of excess [Kornai, 2014]. It is accompanied by phenomena with features of some kind of market bulimia, sick consumerism, detrimental both to people and the environment [Rist, 2015]. One of the more compromising signs of the economy of excess and wasting of resources is wasting of food by rich countries, when simultaneously, there are areas of hunger in some parts of the world [Stuart, 2009]. At the same time, the economy of excess does not translate to the comfort of the buyers of goods – as in theory attributed to the consumer market. It is indicated in the publication of Janos Kornai concerning a comparative analysis of the features of socio-economic systems. While exposing his deep critical evaluation of socialist non-market systems, as economies of constant deficiency, he does not spare critical opinions on the capitalist economy of excess, with its quest for the growth of the gross domestic product (GDP) and profits. As an example of the economy of excess, he indicates the pharmaceutical industry, with strong monopolistic competition, dynamic innovativeness, wide selection for the buyers, flood of advertisements, manipulation of customers, and often bribing the doctors prescribing products [Kornai 2014, p. 202]. This type of abnormalities is not alien to other industries. Although J. Konrai appreciates that in the economy of excess, including the excess of production capacities, the excess is “grease” calming down and soothing clashes that occur in the mechanisms of adaptation, he also sees that those who claim that in the economy of excess (or more generally in the market economy), sovereignty of consumers dominates, exaggerate [Kornai, 2014, pp. 171-172], as the manufacturers, creating the supply, manipulate the consumers. Thus, there is an excess of supply – both of values as well as junk [Kornai, 2014, p. 176]. Analysing the economy of excess, J. Kornai brings this issue to the question of domination and subordination. It corresponds with the opinion of Jerzy Wilkin, according to whom, the free market can also enslave, so take away individual freedom; on the other hand, the lack of the free market can lead to enslavement as well. Economists willingly talk about the free market, and less about the free man [Wilkin, 2014, p. 4]. The economy of excess is one of the consequences of making a fetish of the economic growth and its measure, which is the gross domestic product (GDP) and treating it as the basis of social and economic activity. In such a system, the pressure of growth is created, so you must grow to avoid death! The system is thus comparable to a cyclist, who has to move forwards to keep his balance [Rist, 2015, p. 181]. It corresponds with the known, unflattering to economists, saying of Kenneth E. Boulding [1956], criticising the focus of economics on the economic growth, while ignoring social implications and consequences to the environment: Anyone who believes in indefinite growth in anything physical, on a physically finite planet, is either mad or an economist. [from: Rist, 2015, p. 268]. GDP is a very much needed or even indispensable measure for evaluation of the material level of the economies of individual countries and for comparing their economic health. However, it is insufficient for evaluation of the real level of welfare and quality of life. It requires supplementation with other measures, as it takes into account only the values created by the market purchase and sale transactions. It reflects only the market results of the activity of enterprises and households. Additionally, the GDP account threats the socially desirable and not desirable activities equally. Thus, the market activity related to social pathologies (e.g. functioning of prisons, prostitution, and drug dealing) also increase the GDP. It was accurately expressed already in 1968 by Robert Kennedy, who concluded the discussion on this issue saying that: the gross national product does not allow for the health of our children, the quality of their education or the joy of their play. It does not include the beauty of our poetry or the strength of our marriages, the intelligence of our public debate or the integrity of our public officials. It measures neither our wit nor our courage, neither our wisdom nor our learning, neither our compassion nor our devotion to our country, it measures everything in short, except that which makes life worthwhile [The Guardian, 2012]. While Grzegorz W. Kołodko even states that it should be surprising how it is possible that despite a number of alternative measures of social and economic progress, we are still in the corset of narrow measure of the gross product, which completely omits many significant aspects of the social process of reproduction [Kołodko, 2013, p. 44]. In this context he points to the necessity of triple sustainable growth – economic, social, and ecological [Kołodko, 2013, p. 377]. Transition from the industrial civilisation model to the new model of economy, to the age of information, causes a kind of cultural regression, a phenomenon of cultural anchoring in the old system. This type of lock-in effect - described in the source literature, that is the effect of locking in the existing frames and systemic solutions, is a barrier to development. The practice more and more often and clearer demonstrates that in the conditions of the new economy, the tools and traditional solutions turn out to be not only ineffective, but they even increase the risk of wrong social and economic decisions, made at different institutional levels. All this proves that new development models must be searched for and implemented, to allow counteraction to dysfunctions of the contemporary economy and wasting the development potential, resulting from a variety of maladjustments generated by the crisis of civilisation. Polish authors who devote much of their work to these issues include G.W. Kołodko, Jerzy Kleer, or Maciej Bałtowski. Studies confirm that there is a need for a new pragmatism, new, proinclusive model of shaping the social and economic reality, a model which is more socially rooted, aiming at reconciling social, economic and ecological objectives, with simultaneous optimisation of the use of the social and economic potential [Kołodko, 2013; Bałtowski, 2016; Kleer, 2015]. There is more and more evidence that the barriers to economic development growing in the global economy are closely related with the rooting of the economy in social relations. The book of A. Blikle becomes a part of this trend in a new and original manner. Although the author concentrates on the analyses of social relations mainly at the level of an enterprise, at the same time, he comments them at a macroeconomic, sociological and ethical level, and interdisciplinary contexts constitute an original value of the book. Conclusion I treat the book of Andrzej Blike as an important voice in favour of shaping an inclusive social and economic system, in favour of shaping inclusive enterprises, that is oriented on an optimal absorption of knowledge, innovation and effective reconciliation of the interests of entrepreneurs with the interests of employees and the interests of society. Inclusiveness is indeed a value in itself. It is understood as a mechanism/system limiting wasting of material resources and human capital, and counteracting environmental degradation. An inclusive social and economic system is a system oriented on optimisation of the production resources and reducing the span between the actual and potential level of economic growth and social development [Reforma, 2015]. And this is the system addressed by Andrzej Blikle in his book. At least this is how I see it. Although the book is mainly concerned with business management, its message has a much wider dimension and is concerned with real measures of wealth, money and people’s lives. null
