Disulfide-bond formation, mediated by the Dsb family of proteins, is important in the correct folding of secreted or extracellular proteins in bacteria. In Gram-negative bacteria, disulfide bonds are introduced into the folding proteins in the periplasm by DsbA. DsbE fromEscherichia colihas been implicated in the reduction of disulfide bonds in the maturation of cytochromec. The Gram-positive bacteriumMycobacterium tuberculosisencodes DsbE and its homologue DsbF, the structures of which have been determined. However, the two mycobacterial proteins are able to oxidatively fold a proteinin vitro, unlike DsbE fromE. coli. In this study, the crystal structure of a DsbE or DsbF homologue protein fromCorynebacterium diphtheriaehas been determined, which revealed a thioredoxin-like domain with a typical CXXC active site. Structural comparison withM. tuberculosisDsbF would help in understanding the function of theC. diphtheriaeprotein.

Disulfide bond formation in periplasmic proteins is catalysed by the DsbA/DsbB system in most Gram-negative bacteria. Salmonella enterica serovar Typhimurium also encodes a paralogous pair of proteins to DsbA and DsbB, DsbL and DsbI, respectively, downstream of a periplasmic arylsulfate sulfotransferase (ASST). We show that DsbL and DsbI function as a redox pair contributing to periplasmic disulfide bond formation and, as such, affect transcription of the Salmonella pathogenicity island 1 (SPI1) type three secretion system genes and activation of the RcsCDB system, as well as ASST activity. In contrast to DsbA/DsbB, however, the DsbL/DsbI system cannot catalyse the disulfide bond formation required for flagellar assembly. Phylogenic analysis suggests that the assT dsbL dsbI genes are ancestral in the Enterobacteriaceae, but have been lost in many lineages. Deletion of assT confers no virulence defect during acute Salmonella infection of mice.

Disulfide-bond-forming (DSB) oxidative folding enzymes are master regulators of virulence that are localized to the periplasm of many Gram-negative bacteria. The archetypal DSB machinery from Escherichia coli K-12 consists of a dithiol-oxidizing redox-relay pair (DsbA/B), a disulfide-isomerizing redox-relay pair (DsbC/D) and the specialist reducing enzymes DsbE and DsbG that also interact with DsbD. By contrast, the Gram-negative bacterium Wolbachia pipientis encodes just three DSB enzymes. Two of these, α-DsbA1 and α-DsbB, form a redox-relay pair analogous to DsbA/B from E. coli. The third enzyme, α-DsbA2, incorporates a DsbA-like sequence but does not interact with α-DsbB. In comparison to other DsbA enzymes, α-DsbA2 has ∼50 extra N-terminal residues (excluding the signal peptide). The crystal structure of α-DsbA2ΔN, an N-terminally truncated form in which these ∼50 residues are removed, confirms the DsbA-like nature of this domain. However, α-DsbA2 does not have DsbA-like activity: it is structurally and functionally different as a consequence of its N-terminal residues. Firstly, α-DsbA2 is a powerful disulfide isomerase and a poor dithiol oxidase: i.e. its role is to shuffle rather than to introduce disulfide bonds. Moreover, small-angle X-ray scattering (SAXS) of α-DsbA2 reveals a homotrimeric arrangement that differs from those of the other characterized bacterial disulfide isomerases DsbC from Escherichia coli (homodimeric) and ScsC from Proteus mirabilis (PmScsC; homotrimeric with a shape-shifter peptide). α-DsbA2 lacks the shape-shifter motif and SAXS data suggest that it is less flexible than PmScsC. These results allow conclusions to be drawn about the factors that are required for functionally equivalent disulfide isomerase enzymatic activity across structurally diverse protein architectures.

The ability of apatite to form on the surface of biomaterials in simulated body fluid (SBF) has been widely used to predict the bone-bonding ability of bioceramic and bioceramic/polymer composites in vivo. Porous β-tricalcium phosphate/poly(L-lactic acid) (β-TCP/PLLA) composite scaffold was synthesized by new method. The ability of inducing calcium phosphate (Ca-P) formation was compared in static simulated body fluid(sSBF) and dynamic simulated body fluid (dSBF). The Ca-P morphology and crystal structures were identified using SEM, X-ray diffraction and Fourier transform infrared (FT-IR) spectroscopy. The results showed that the typical features of bone-like apatite formation on the surface and the inner pore wall of β-TCP/PLLA. Ca-P formation on scaffold surfaces in dSBF occurred slower than in sSBF and was more difficult with increasing flow rate of dSBF. The ability of apatite to form on β-TCP/PLLA was enhanced by effect of each other that has different degradable mechanism. Porous β-TCP/PLLA composite scaffold indicates good ability of Ca-P formation in vitro.

ABSTRACT The Dsb family of enzymes catalyzes disulfide bond formation in the gram-negative periplasm, which is required for folding and assembly of many secreted proteins. Pertussis toxin is arguably the most complex toxin known: it is assembled from six subunits encoded by five genes (for subunits S1 to S5), with 11 intramolecular disulfide bonds. To examine the role of the Dsb enzymes in assembly and secretion of pertussis toxin, we identified and mutated the Bordetella pertussis dsbA, dsbB, and dsbC homologues. Mutations in dsbA or dsbB resulted in decreased levels of S1 (the A subunit) and S2 (a B-subunit protein), demonstrating that DsbA and DsbB are required for toxin assembly. Mutations in dsbC did not impair assembly of periplasmic toxin but resulted in decreased toxin secretion, suggesting a defect in the formation of the Ptl secretion complex.

ABSTRACT Dsb proteins (DsbA, DsbB, DsbC, and DsbD) catalyze formation and isomerization of protein disulfide bonds in the periplasm ofEscherichia coli. By using a set of Dsb coexpression plasmids constructed recently, we analyzed the effects of Dsb overexpression on production of horseradish peroxidase (HRP) isozyme C that contains complex disulfide bonds and tends to aggregate when produced in E. coli. When transported to the periplasm, HRP was unstable but was markedly stabilized upon simultaneous overexpression of the set of Dsb proteins (DsbABCD). Whereas total HRP production increased severalfold upon overexpression of at least disulfide-bonded isomerase DsbC, maximum transport of HRP to the periplasm seemed to require overexpression of all DsbABCD proteins, suggesting that excess Dsb proteins exert synergistic effects in assisting folding and transport of HRP. Periplasmic production of HRP also increased when calcium, thought to play an essential role in folding of nascent HRP polypeptide, was added to the medium with or without Dsb overexpression. These results suggest that Dsb proteins and calcium play distinct roles in periplasmic production of HRP, presumably through facilitating correct folding. The present Dsb expression plasmids should be useful in assessing and dissecting periplasmic production of proteins that contain multiple disulfide bonds in E. coli.

ABSTRACT The cytoplasmic membrane protein CcdA and its homologues in other species, such as DsbD of Escherichia coli, are thought to supply the reducing equivalents required for the biogenesis of c-type cytochromes that occurs in the periplasm of gram-negative bacteria. CcdA-null mutants of the facultative phototroph Rhodobacter capsulatus are unable to grow under photosynthetic conditions (Ps−) and do not produce any active cytochrome c oxidase (Nadi−) due to a pleiotropic cytochrome c deficiency. However, under photosynthetic or respiratory growth conditions, these mutants revert frequently to yield Ps+ Nadi+ colonies that produce c-type cytochromes despite the absence of CcdA. Complementation of a CcdA-null mutant for the Ps+ growth phenotype was attempted by using a genomic library constructed with chromosomal DNA from a revertant. No complementation was observed, but plasmids that rescued a CcdA-null mutant for photosynthetic growth by homologous recombination were recovered. Analysis of one such plasmid revealed that the rescue ability was mediated by open reading frame 3149, encoding the dithiol:disulfide oxidoreductase DsbA. DNA sequence data revealed that the dsbA allele on the rescuing plasmid contained a frameshift mutation expected to produce a truncated, nonfunctional DsbA. Indeed, a dsbA ccdA double mutant was shown to be Ps+ Nadi+, establishing that in R. capsulatus the inactivation of dsbA suppresses the c-type cytochrome deficiency due to the absence of ccdA. Next, the ability of the wild-type dsbA allele to suppress the Ps+ growth phenotype of the dsbA ccdA double mutant was exploited to isolate dsbA-independent ccdA revertants. Sequence analysis revealed that these revertants carried mutations in dsbB and that their Ps+ phenotypes could be suppressed by the wild-type allele of dsbB. As with dsbA, a dsbB ccdA double mutant was also Ps+ Nadi+ and produced c-type cytochromes. Therefore, the absence of either DsbA or DsbB restores c-type cytochrome biogenesis in the absence of CcdA. Finally, it was also found that the DsbA-null and DsbB-null single mutants of R. capsulatus are Ps+ and produce c-type cytochromes, unlike their E. coli counterparts, but are impaired for growth under respiratory conditions. This finding demonstrates that in R. capsulatus the dithiol:disulfide oxidoreductases DsbA and DsbB are not essential for cytochrome c biogenesis even though they are important for respiration under certain conditions.

DsbA is the major oxidase responsible for generation of disulfide bonds in proteins of E. coli envelope. In the present work we provided the first detailed characterization of disulfide exchange between DsbA and its natural substrate, HtrA protease. We demonstrated that HtrA oxidation relies on DsbA, both in vivo and in vitro. We followed the disulfide exchange between these proteins spectrofluorimetrically and found that DsbA oxidizes HtrA with a 1:1 stoichiometry. The calculated second-order apparent rate constant (kapp) of this reaction was 3.3x10(4)+/-0.6x10(4) M-1s-1. This value was significantly higher than the values obtained for nonfunctional disulfide exchanges between DsbA and DsbC or DsbD and it was comparable to the kapp values calculated for in vitro oxidation of certain non-natural DsbA substrates of eukaryotic origin.

This study set out to investigate the impact of inhibiting bacterial enzymes on pathogen growth and virulence as an alternative antimicrobial strategy to antibiotics. Results support the hypothesis that DsbA enzyme inhibition is a robust anti-virulence strategy that can 'disarm but not kill bacteria' using uropathogenic Escherichia coli as the model pathogen. These findings have enhanced the pharmacological importance of DsbA as an anti-virulence drug target and contribute to ongoing research that aims to develop DsbA inhibitors against different bacterial pathogens with diverse DsbA enzymes.

DSB Mobile is a small Peruvian software developing company based in Lima. DSB Mobile specializes in the development of both mobile and web applications and has worked with major companies such as Samsung, Claro & Entel. The company is composed of the General Manager, Zico Herrera, a sales manager, operations manager and both fulltime and contract based software developers that are hired based on the current demand for service. DSB Mobile has established a strong reputation and brand in Peru and is now looking to expand outside of Peru where they can introduce their software products into international markets. In their aspiration to internationalize, DSB Mobile is seeking to find out not only the most profitable markets for their company but also markets that would best align with DSB Mobiles mission. The solution to their question of expansion was to determine the best markets using a variety of both quantitative and qualitative factors. In using an IT Competitiveness report that was done by the British Software Alliance this was used as a reference point to determine the best ranked countries for IT competitiveness and the best countries to conduct business in base off of important IT indicators. Combined with software data statistics in terms of charges per project and cost of IT Consultants, this further narrowed down the scope to the most attractive, profitable and mutually beneficial market for DSB Mobile. The implementation plan proposed involved two market lines namely, the North American market line and the European Market line. The proposed solution takes into consideration different scenarios, one of them being the moderate work balance of 1 project per month, giving a total cost of $391,065 per year with a profit of $180,736. The Gantt chart outlined is intended to guide the company with the step by step implementation of this international expansion and prepare them to execute this plan in the most efficient and effective manner

Klebsiella pneumoniae is a clinically important bacterial pathogen causing urinary tract infections and pneumonia which can progress to more severe disease. With the increase in antimicrobial resistance globally, K. pneumoniae infections have become difficult to treat, with many antibiotics now ineffective. Therefore, it is imperative that we investigate new and effective treatments for these drug resistant infections. This thesis describes the identification and characterization of multiple diverse disulphide bond enzymes in K. pneumoniae that have potential as novel therapeutic targets.

DNA中の二本鎖切断(DSB)に対するアスコルビン酸(AA)およびDMSOの保護効果を、蛍光顕微鏡による巨大DNA(T4 DNA; 166kbp)の単分子観察によって評価した。凍結/解凍の状態に対して3つの異なる形態の放射源、可視光、γ線、および超音波の環境下にさらした。1‐プロパノールと2‐プロパノールの間で異なる効果が表れた。ゲノムDNA分子の高次構造の変化は、1−プロパノールを用いると、長軸長が濃度60%で最小を示し、次にアルコール含有量の増加と共に増加する傾向があることを見出した。一方、2−プロパノールを用いると、長軸長はアルコール含有量の増加と共にほぼ単調な減少を示した。
The protective effect of ascorbic acid (AA) and DMSO against double-strand breaks (DSBs) in DNA was evaluated by single-molecule observation of giant DNA (T4 DNA; 166kbp) through fluorescence microscopy. Samples were exposed to three different forms of radiation: visible light, γ-ray, and ultrasound or freeze/thawing. The change of the higher-order structure of genomic DNA molecules in the presence of alcohols by use of single DNA observation with fluorescence microscopy, by focusing our attention to unveil the different effect between 1-propanol and 2-propanol.

La quasi-totalite des proteines d'interet economique est constituee de proteines extracellulaires dont l'un des elements essentiel de stabilite et de repliement est la presence de ponts disulfure. Les strategies utilisees pour la production recombinante de ces proteines impliquent leur expression dans le periplasme de bacteries hotes. Dans le periplasme d'e. Coli, les enzymes du systeme dsb catalysent la formation et l'isomerisation des ponts disulfure. Dans des conditions d'expression normale, elles permettent le repliement correct des proteines naturelles. En revanche, la surexpression de proteines heterologues dans le periplasme conduit souvent a l'agregation ou a la degradation des proteines, en raison de l'accumulation de formes a ponts disulfure non formes ou non natifs. L'amelioration par ingenierie des proprietes de ces catalyseurs s'avere donc essentielle pour la production en grande quantite de proteines correctement repliees. Dsba, le principal agent de controle du potentiel oxydoreducteur periplasmique, constitue une cible ideale pour ce type d'etude. Afin de mieux comprendre ses proprietes catalytiques, l'etude de ses caracteristiques structurales et dynamiques a fait l'objet de ce travail. Dans la premiere partie, l'etude structurale comparative des formes oxydee et reduite a permis d'identifier les differences d'environnement electronique entre ces deux etats redox. Dans la seconde partie, l'etude d'un complexe covalent entre dsba et un peptide substrat a fourni une description detaillee des interactions entre les deux partenaires. Enfin, la mobilite au sein des trois formes (oxydee, reduite, complexee) a ete etudiee par la technique d'echange proton-deuterium. Au contraire de la structure, qui apparait tres similaire dans les differentes formes, la flexibilite de dsba varie largement entre les trois formes. Les informations obtenues au cours de cette etude fournit les elements de base pour la conception de nouveaux catalyseurs derives de dsba.

Background: Neuropsychological tests are important tools for the diagnosis of mild cognitive impairment or dementia due to Alzheimer’s disease (AD). Objective: To investigate the accuracy of common neuropsychological tests used in the clinical setting for AD diagnosis. Methods: Forty two patients with diagnosis of AD continuum [A+T+/-(N)+/-] and 32 non-AD [A-T+/-(N)+/-]. All participants were submitted to a thorough neuropsychological assessment with the following instruments: Mattis Dementia Rating Scale (DRS), Rey’s Auditory Verbal Learning Test (RAVLT), Boston naming-Consortium to Establish a Registry for Alzheimer’s Disease, a reduced version of the CERAD, Digit Span Forward (DSF), Digit Span Backward (DSB) and Cubes from The Wechsler Adult Intelligence Scale (WAIS), verbal fluency – animals (VF-A), and FAS. Results: Memory (MEM) and Initiation/Perseveration (I/P) subscales of the DRS, FAS, Digit Span Backward (DSB) and Boston naming displayed good discrimination between AD and non-AD patients. The MEM subscale of the DRS, RAVLT A6 and FAS presented high sensitivity (90% or more) for AD diagnosis, while DSF displayed high specificity. Non-AD patients had greater difficulty in FAS, DSB and in Boston naming. Conclusion: Performance of patients with biological diagnosis of AD on MEM and I/P of DRS, and RAVLT A7 was significantly different from that of non-AD subjects.

<p>From a structural engineering point of view, wind effects pose one of the major challenges to tall buildings. From a performance/architectural point of view, climatologic aspects pose a major challenge. Remedies for each challenge separately have been proposed. One of the remedies for wind effects is the Tunes-Mass-Damper (TMD) or multiple TMD's. To mitigate climatological issues, the Double-Skin-Façade (DSF) has been developed. Recently it has been suggested to take advantage of the space between the two skins of the DSF system to allocate TMD's.</p><p>In this work, another step is taken towards a single remedy for both challenges. A modified version of the TMD-DSF system proposed by Moon (2016) is presented. That is, parts of the mass of the DSF envelope itself are used as part of a multiple TMD (MTMD) system. This is obtained by connecting these parts to the building using springs and dampers while allowing the DSF to move parallel to the floor edges. Furthermore, the DSF-MTMD system is optimized using a formal optimization approach. The optimization indicates which parts of the envelope should be connected to the building rigidly and which should be used as TMD's. Furthermore, the properties of the springs and the dampers are determined by minimizing the cost associated with transforming the DSF system to a DSF-MTMD system and limiting wind responses to desired values.</p>

A new switching network architecture, called the Dilated Slipped Banyan Network(DSBN), is proposed for the hub of a photonic active-star Local Area Network (LAN). A lithium niobate implementation of a classical 4-by-4 banyan architecture was proposed[1]: for bit-interleaved time-multiplexing. The DSBN has properties, described herein, that make it more general in size and more amenable to lithium niobate implementation than the classical banyan. Furthermore, since the active-star LAN application could use a word-interleaved data format instead of a bit-interleaved format, the hub switch need not be bit-synchronous with the data.

Incremental Sheet Forming (ISF) is a flexible forming process suitable for low volume production of sheet metal components. Single Point Incremental Forming (SPIF), which has only one tool forming the geometry, is the simplest variant of incremental forming. Bending of sheet between the component opening and the fixed boundary is unavoidable in SPIF due to the absence of support/backup. Double Sided Incremental Forming (DSIF) has two tools which can be used interchangeably for forming and providing local support. The accuracy of parts formed using DSIF is superior to those formed using SPIF as the unwanted bending is substantially reduced by providing local support. In addition DSIF is capable of forming components with features on both sides of the initial plane of sheet and convex and concave features without additional setup. In ISF, as the deformation progresses, the intended geometry slowly develops, this increases the stiffness of the sheet. While forming multiple features, the forming sequence greatly affects the way stiffness builds-up, which further affects the geometry of formed components. In the present work, an experimental investigation is carried out to demonstrate the affect of forming sequence on the geometries and accuracy of formed component. Results presented show that the feature sequencing greatly affects the geometry and accuracy of formed components.

In this work a DSF system (Double Skin Facade) is used to improve the thermal comfort level that the occupants are subjected in an experimental chamber. The DSF system, build with three unities, is equipped in a south facade of the experimental chamber. The study, made in winter conditions, uses the solar radiation to heated the air, injected in the occupied space with 6 people, to improve the thermal comfort conditions. The experimental chamber is build with wood and isolating material. The DSF system is build with two glass façades, equipped with 15 lamellas. This system, subjected to solar radiation, is connected to the interior with a duct system connected with two ventilators. The numerical study consider a software that simulates the building and the DSF thermal response. The numerical model, that considers buildings with complexes topologies, uses energy and mass balance integral equations for the opaque surfaces, transparent surfaces and internal air. The software also considers the solar radiation simulator, the radiative and convective coefficients evaluation, the glass radiative properties and the thermal comfort and internal air quality simulator. In accordance with the obtained results the thermal comfort level, using the PMV (Predicted Mean Vote), during the occupation time, in general, is in accordance with the international standards.

Double skin facades (DSFs) are building envelopes comprised of two glass facades, a ventilated air cavity and shading devices placed within the cavity. In this paper airflow and heat transfer simulation was conducted for a DSF system equipped with a venetian blind using computational fluid dynamics (CFD) with RNG turbulence model. Simulation was done for a three-level combination of slat tilt angle and blind position. The heat transfer coefficient was directly obtained from CFD simulation. The CFD prediction was validated using experimental data collected for a mechanically ventilated DSF (1.6 m wide and 2.5 m high and 0.15 m wide cavity) equipped with venetian blinds. The present study indicates that the presence of venetian blinds influences the surface heat transfer coefficients (SHTCs), the temperature and the air distribution in the DSF system. Specifically, for the cases considered, the position of the blinds is more important than the slat angles.

Single Point Incremental Forming (SPIF) is plagued by an unavoidable and unintended bending in the region of the sheet between the current tool position and the fixture, which leads to significant geometric inaccuracies. Double Sided Incremental Forming (DSIF) uses two tools, one on each side of the sheet to form the sheet into the desired shape. This work explores the capabilities of DSIF in terms of improving the geometric accuracy as compared to SPIF by using a novel toolpath strategy in which the sheet is locally squeezed between the two tools. Experiments and simulations are performed to show that this strategy can improve the geometric accuracy of the component significantly. An examination of the forming forces indicates that after a certain amount of deformation using this strategy a loss of contact occurs between the bottom tool and the sheet. The effects of this on the geometric accuracy are discussed as well.

Abstract Piezoelectric materials have been widely used in modern science and technology due to their electro-mechanical coupling response. Sometimes, because of the stiff and brittle nature of some piezoelectric materials, the piezoelectric devices with defects may face fracture or failure during their service procedures. Therefore, it has become important to investigate the failure behaviors caused by defects, such as cracks and holes. Based on the study of the dynamic anti-plane characteristics for radial crack emanating from a circular cavity in piezoelectric bi-materials, this paper aims to analyzes the dynamic incident anti-plane shearing (SH-wave) in piezoelectric bi-materials, which contains two interfacial cracks, near an eccentric elliptical hole. Green’s function method, the conformal mapping method, the interface conjunction techniques and the crack-deviation techniques are utilized to obtain a series of first kind Fredholm’s equations, based on which the dynamic stress intensity factor (DSIF) at the outer and the inner cracks’ tips are theoretically expressed. Numerical examples were graphically presented to illustrate the effects of the piezoelectric parameter, the effective piezoelectric elastic modulus, the dimensionless incident wave number and geometric parameters on the DSIF at both of the tips. Previous studies are not comprehensive, especially when the center of the hole deviates from the interface. Therefore, the impact of eccentric distance on DSIF is considered in this paper. The solution of this problem provides a more accurate and efficient method for the investigation of dynamic fracture properties of piezoelectric materials and has an important theoretical significance in engineering design.

Gene targeting (GT) is a much needed technology as a tool for plant research and for the precise engineering of crop species. Recent advances in this field have shown that the presence of a DNA double-strand break (DSB) in a genomic locus is critical for the integration of an exogenous DNA molecule introduced into this locus. This integration can occur via either non-homologous end joining (NHEJ) into the break or homologous recombination (HR) between the broken genomic DNA and the introduced vector. A bottleneck for DNA integration via HR is the machinery responsible for homology search and strand invasion. Important proteins in this pathway are Rad51, Rad52 and Rad54. We proposed to combine our respective expertise: on the US side, in the design of zincfinger nucleases (ZFNs) for the induction of DNA DSBs at any desired genomic locus and in the integration of DNA molecules via NHEJ; and on the Israeli side in the HR events, downstream of the DSB, that lead to homology search and strand invasion. We sought to test three major pathways of targeted DNA integration: (i) integration by NHEJ into DSBs induced at desired sites by specially designed ZFNs; (ii) integration into DSBs induced at desired sites combined with the use of Rad51, Rad52 and Rad54 proteins to maximize the chances for efficient and precise HR-mediated vector insertion; (iii) stimulation of HR by Rad51, Rad52 and Rad54 in the absence of DSB induction. We also proposed to study the formation of dsT-DNA molecules during the transformation of plant cells. dsT-DNA molecules are an important substrate for HR and NHEJ-mediatedGT, yet the mode of their formation from single stranded T-DNA molecules is still obscure. In addition we sought to develop a system for assembly of multi-transgene binary vectors by using ZFNs. The latter may facilitate the production of binary vectors that may be ready for genome editing in transgenic plants. ZFNs were proposed for the induction of DSBs in genomic targets, namely, the FtsH2 gene whose loss of function can easily be identified in somatic tissues as white sectors, and the Cruciferin locus whose targeting by a GFP or RFP reporter vectors can give rise to fluorescent seeds. ZFNs were also proposed for the induction of DSBs in artificial targets and for assembly of multi-gene vectors. We finally sought to address two important cell types in terms of relevance to plant transformation, namely GT of germinal (egg) cells by floral dipping, and GT in somatic cells by root and leave transformation. To be successful, we made use of novel optimized expression cassettes that enable coexpression of all of the genes of interest (ZFNs and Rad genes) in the right tissues (egg or root cells) at the right time, namely when the GT vector is delivered into the cells. Methods were proposed for investigating the complementation of T-strands to dsDNA molecules in living plant cells. During the course of this research, we (i) designed, assembled and tested, in vitro, a pair of new ZFNs capable of targeting the Cruciferin gene, (ii) produced transgenic plants which expresses for ZFN monomers for targeting of the FtsH2 gene. Expression of these enzymes is controlled by constitutive or heat shock induced promoters, (iii) produced a large population of transgenic Arabidopsis lines in which mutated mGUS gene was incorporated into different genomic locations, (iv) designed a system for egg-cell-specific expression of ZFNs and RAD genes and initiate GT experiments, (v) demonstrated that we can achieve NHEJ-mediated gene replacement in plant cells (vi) developed a system for ZFN and homing endonuclease-mediated assembly of multigene plant transformation vectors and (vii) explored the mechanism of dsTDNA formation in plant cells. This work has substantially advanced our understanding of the mechanisms of DNA integration into plants and furthered the development of important new tools for GT in plants.
