Pełny tekst źródłaDairy cows are very susceptible to negative energy balance, which occurs when their energy needs exceed their intake. Negative energy balance affects milk production as well as the animal health. Its early detection is thus essential and feed restriction experiments are conducted to it. The objective of this thesis was to identify milk molecules affected by feed restrictions of different intensity to propose a panel of non-invasive biomarkers of negative energy balance, usable in dairy farming. The study of milk contents, metabolites, major proteins, as well as proteome and miRNome, allowed the identification of variable molecules reflecting the adaptation of the mammary metabolism to the energy stress. Variations induced by these restrictions were a factor of their intensity and some of the identified variableswere verified on other animals, during the physiological negative energy balance in early lactation. The integrative study of all these data led to 25 candidate biomarkers among which some were selected for their potential to be monitored by a tool commonly used in breeding: the mid infrared spectrometry. Equations will be developed to evaluate the concentrations of glutamate, uric acid and isocitrate via these spectra and will be used with those already existing to evaluate the lactose content and the fatty acids concentrations, to constitute a panel of biomarkers of the negative energy balance. Validation of this tool will have to be done but, if its reliability is confirmed, it will allow a better monitoring of the energy balance in dairy cows and will help to improve their health, their welfare, and their productivity

Pełny tekst źródłaFor a long time, the increase in life expectancy in humans inspires research, especially in biology. Beyond the notion of mortality, those of morbidity and active and independent life expectancy are at the center of public health issues. However, faced with the decrease in physical capacity and the increase in the number of diseases during aging, interest is now focused on improving the quality of life. Indeed, the loss of autonomy in the elderly directs us towards a goal of improving the healthspan. Physical activity is a fundamental factor in the well-being, impacting physical performance as well as different pathologies (diabetes, cardiac pathologies). In the elderly population, the necessity to reach fast results, without suffering or long time training led to our first study. This one focused on a new model of short acceleration-based training protocol in elderly mice. The results showed an improvement in physical performance and in the whole of the energetic metabolisms, while being more efficient than endurance training and more suitable than interval training. Moreover, given the recent interest in energy transfers in the regulation of energetic metabolisms and in the aging process, we were interested in understanding the mechanisms of energy flux and in particular the one involving mitochondrial creatine kinase. Our second study analyzed the physical and physiological capacity of mitochondrial creatine kinase deficient mice from 6 to 18 months. Our results suggest a predominant lipid metabolism and showed a conservation of physical abilities in these mitochondrial creatine kinase deficient mice. Knowledge of the mechanisms of energy flux and their importance to physical capacities during aging would lead to better design future training protocols, such as our acceleration training, which opens new perspectives in improvement of physical capacity and autonomy during aging

Pełny tekst źródłaThe aim of this work is to study mitochondrial dysfunctions related to a defect of protein lipoylation in fibroblasts of 14 patients. These patients carry a point mutation in a gene encoding for a protein involved either in lipoic acid biosynthesis (LIPT1 or LIPT2) or in the mitochondrial pathway devoted to iron-sulfur cluster biogenesis (FDX1L, ISCA1, ISCA2, IBA57, NFU1, BOLA3) essential for maturation of mitochondrial Fe-S proteins such as lipoic acid synthase (LIAS). This work describes the second case of FDX1L deficiency and a patient with a new mutation in ISCA1 gene.We found that mitochondrial [4Fe-4S] proteins (mitochondrial aconitase, complexes I and II of the respiratory chain and LIAS) are mainly affected in fibroblasts of patients with defect in the mitochondrial Fe-S maturation pathway. Secondary, LIAS dysfunction leads to decreased lipoylation of PDHc and KGDHc, complexes involved in energy metabolism. Neither mitochondrial network nor oxidative stress biomarkers was modified in our study. Addition of exogenous lipoic acid did not rescue the mitochondrial deficiency.Protein expression profiles obtained in fibroblasts of patients suggest that NFU1, BOLA3 and IBA57 and also ISCA1, ISCA2 and IBA57 could function and interact together to form protein complexes

Pełny tekst źródłaObesity is a risk factor for metabolic and mood disorders. The increasing abundance of the "Western diet" that is rich in saturated fatty acids (SFA) and low in polyunsaturated fatty acids (PUFA) omega-3 (N-3) can generate a physiological imbalance in the ratio of N-6/N-3 fatty acids. Such an imbalance has also been implicated in the increased prevalence of mood disorders and metabolic diseases. Docosahexaenoic acid (DHA, 22:6n-3), the most abundant fatty acid (FA) in the brain is accumulated not only during the post-natal period but also during the perinatal period. It plays an important role in neuronal development and other brain functions such as learning and memory. Disturbances of the perinatal environment can influence the sustainable future of the offspring, making it more likely to develop obesity in rich nutritional context. Some data suggest that an inadequate maternal intake of N-3 during pregnancy and the perinatal period may cause an increase in appetite signalling in the offspring as well as an increase rate of neurological and cardio-metabolic disease. It is not known if maternal dietary deficiency in N-3 will have an impact on food motivation of the offspring. The main objective of this thesis was to study the potential effect of dietary deficiency of DHA during the perinatal period on energy balance, food motivation and the anxiety-like behavior of adult male mouse offspring placed on HFD. Our data showed, as expected, that the maternal DHA deficient diet during perinatal period encouraged offspring to work harder to get a palatable food, entailed a dysregulation of energy homeostasis by increasing the body weight and reducing locomotor activity and exacerbated the anxiety-like behavior once they were exposed to an obesogenic environment. Free fatty acids (FFA) are essential nutrients that they also function as signalling molecules in the brain and have receptors that play a significant role in the control of energy metabolism. Among them is GPR120, also known as N-3 FA receptor, a g-protein coupled receptor that is reportedly activated by PUFA and shown to mediate the anti-inflammatory and insulin-sensitizing effects of N-3 FA. A mutation in the GPR120 gene that is associated with reduced GPR120 signalling activity is linked to human obesity. The objective of our second study was to test the impact of pharmacological GPR120 stimulation in the CNS on feeding, energy expenditure, anxiety-like behavior and food reward. Our results showed that an acute central injection GPR120 agonist III: reduced ad libitum food intake and the rewarding effect for high fat, high sugar food; produced a significant decrease in the anxiety-like behavior. While mice with the chronic injection (21 days) of GPR120 III agonist pump on HFD didn’t show any modification in food intake or body weight gain, but show a reduction of anxiety-like behavior in Elevated plus maze (EPM) and open-field test (OFT) compare to the vehicle group. DHA is notifies to have an anorectic action. Furthermore, stimulation of GPR120 receptors with synthetic agonist may cause an acute and profound effect on food related behavior in rodents. To find an approach that aims to control neuroinflammation, food reward and emotional disorders would greatly assist the treatment of obesity and type 2 diabetes.

Pełny tekst źródłaMost conditions detected by expanded newborn screening result from deficiency of one of the enzymes that degrade acyl-CoA esters in mitochondria. The role of acyl-CoAs in the pathophysiology of these disorders is poorly understood, in part because CoA esters are intracellular and samples are not generally available from human patients. We created a mouse model of one such condition, deficiency of 3-hydroxy-3-methylglutaryl-CoA lyase (HL), in liver (HLLKO mice). HL catalyses a reaction of ketone body synthesis and of leucine degradation. Chronic HL deficiency and acute crises each produced distinct abnormal liver acyl-CoA patterns, which would not be predictable from levels of urine organic acids and plasma acylcarnitines. In HLLKO hepatocytes, ketogenesis was undetectable. Measures of Krebs cycle flux diminished following incubation of HLLKO mitochondria with the leucine metabolite 2-ketoisocaproate (KIC). HLLKO mice also had suppression of the normal hyperglycemic response to a systemic pyruvate load, a measure of gluconeogenesis. Hyperammonemia and hypoglycemia, cardinal features of many inborn errors of acyl-CoA metabolism, occurred spontaneously in some HLLKO mice and were inducible by administering KIC. KIC loading also increased levels of several leucine-related acyl-CoAs and reduced acetyl-CoA levels. Ultrastructurally, hepatocyte mitochondria of KIC-treated HLLKO mice show marked swelling. KIC-induced hyperammonemia improved following administration of carglumate (N-carbamyl-L-glutamic acid), which bypasses an acetyl-CoA-dependent reaction essential for urea cycle function, thus demonstrating an acyl-CoA-related mechanism for this complication. In a second animal model of an inborn error of acyl-CoA metabolism, short chain acyl-CoA dehydrogenase (SCAD)-deficient mice, the main finding in liver acyl-CoAs is increased butyryl-CoA, particularly during fasting or after enteral loading with medium chain triglyceride precursor of butyryl-CoA.
