Purpose The aim of this paper is to characterize how innovation may happen through cross-functional teams (CFT) in an organization of the public sector. Design/methodology/approach A case study helped to characterize several behavior patterns, team structures and respective links with generating innovation in internal processes and public answering contexts. Findings The results highlight that formal-temporary teams present a higher capacity to generate incremental innovation in products, whereas permanent-informal teams have a higher capacity to generate innovation in the internal processes and public answering contexts. Research limitations/implications The limitations of this research relate to the fact that this is a single case study, and although it is an important case to examine innovation and CFTs, by its very nature, it is not possible to extend and generalize the obtained data to other organizations. The evaluation of its propositions was merely qualitative, and future research is needed to validate its characteristics. Practical implications Several settings of CFTs are presented, as well as their ability to generate different types of innovation, such as the computerization of documents, petitions and papers, which decreases the time to answer the taxpayer. Moreover, CFTs can help to create products, such as computer programs that can be used not only locally but also in several public organizations related to tax management. Originality/value The field research provides the perceptions of the respondents regarding CFT characteristics that can lead to specific types of innovation, as well as the types of products or services that can be generated by these processes.

Research limitations/implications While this is a case study, the lessons learned can be applied at any library where cross-functional teams are considered. Originality/value This study provides an account of teams at an academic library spanning a decade. Trials and tribulations are discussed.

Purpose – The purpose of this paper is to present the effect of the introduction of cross-functional team (CFT)-based organization, rather than, on planning and performance of OR teams. Design/methodology/approach – In total, two surgical departments of the Radboud University Nijmegen Medical Center (RUNMC) in the Netherlands were selected to illustrate the effect on performance. Data were available for a total of seven consecutive years from 2005 until 2012 and consisted of 4,046 OR days for surgical Department A and 1,154 OR days for surgical Department B on which, respectively 8,419 and 5,295 surgical cases were performed. The performance indicator “raw utilization” of the two surgical Departments was presented as box-and-whisker plots per year (2005-2011). The relationship between raw utilization (y) and years (x) was analyzed with linear regression analysis, to observe if performance changed over time. Findings – Based on the linear regression analysis, raw utilization of surgical Department A showed a statistically significant increase since 2006. The variation in raw utilization reduced from IQR 33 percent in 2005 to IQR 8 percent in 2011. Surgical Department B showed that raw utilization increased since 2005. The variation in raw utilization reduced from IQR 21 percent in 2005 to IQR 8 percent in 2011. Social implications – Hospitals need to improve their productivity and efficiency in response to higher societal demands and rapidly escalating costs. The RUNMC increased their OR performance significantly by introduction of CFT-based organization in the operative process and abandoning the so called functional silos. Originality/value – The stepwise reduction of variation – a decrease of IQR during the years – indicates an organizational learning effect. This study demonstrates that introducing CFTs improve OR performance by working together as a team.

Purpose Accountability, deemed necessary for organisations and society alike, is linked to increased levels of individual, group and organisational performance. Although there has been a recent rise of academic literature’s attempt to better understand accountability, many gaps remain. This research focuses on one of the under-researched areas of accountability, known as peer accountability, where accountability is necessary on a peer to peer level. This research attempts to understand how this type of accountability is experienced by individuals operating in cross-functional teams, and in so doing, identify the antecedents of accountability of this nature. Methodology Exploratory in its nature, this research takes form of a qualitative study. The research utilises the Interactive Qualitative Analysis process, which is a structured approach to performing qualitative research. This approach is reliant on the voices and experiences of those closest to the phenomenon and prescribes the collection of data through the use of a focus group. The focus group consisted of 12 individuals who operate in cross-functional teams and lasted 4 hours. Results the results of the research highlighted five key elements associated with peer accountability. These are, management within the cross-functional team, team culture and dynamics within the cross-functional team, collaborative communication within the cross-functional team and individual accountability within the cross-functional team. Value Through combination of the results gathered and a review of existing accountability literature, a model highlighting the antecedents of peer accountability within cross-functional teams was proposed.

Traditional engineering and science teaching methodology has been to train like-minded students within the discipline of their respective majors. Curriculum time constraints, however, limit the number and nature of out of discipline elective courses. As a result, students are well trained within their respective fields of study but lack the breadth of experience in interacting with other diverse disciplines. Industry, particularly technology-based companies, has observed that solutions to problems have a greater probability of success when all interested parties (purchasing, innovation, marketing, sales, manufacturing, etc.) have input in developing a plan to achieve a desired corporate outcome. It is through this collective action of diverse disciplines that unique solutions are conceived. Many times breakthroughs in innovation and product development occur not through the actions of companies in direct competition but through new entrant companies by modifying technology currently residing in different markets and applications. The breakthrough occurs because the new entrants are not bound by the technology paradigms constraining innovation in their particular market arena. Our goal is to take the diversity lessons gleaned from industry and incorporate them into coursework that creates diverse cross-functional teams such that students learn the benefits of cross-discipline diversity. The College of Business and Technology at ETSU is itself a diverse blend of disciplines (Engineering Technology, Entrepreneurship, Human Nutrition, Marketing, Digital Media, etc) and several graduate and undergraduate courses residing in different departments within the college have intentional programs that encourage cross-discipline enrollment. This action is further facilitated through dual course listings between departments for the same course. Examples of diverse discipline teams will be discussed with attention to outcomes and challenges. Through this diverse cooperative program, students from the technology, business, applied human sciences and digital media disciplines gain a perspective for each other’s expertise and learn to develop teams with diverse skills to meet the increasing challenges for managing business and technology.

This report evaluates the team-structure of three software maintenance teams in order to decide their level of featureness (a term that defines to what extent a team has the quality (the set of characteristics) of being a feature team). Simulations of changes that are expressed as beneficial in an agile environment and that could increase the teams‘ level of featureness within the team structure are performed. The results show that each team‘s level of featureness is affected differently by each change. Partly, this underlines the importance of understanding the current team-structure before implementing changes that aim to increase the level of featureness. And secondly, within the scope of the study, the change where a user expert is declared a team member is concluded as the change that increases the teams‘ level of featureness the most. Based on the results the report also concludes that it is essential to implement changes that affect different, which in combination can increase the level of featureness.

In today's manufacturing environment, due to the complexity of products, and the progress of technology, organisations are forced to be more responsive to the pressure of the dynamic market by developing, producing and delivering products quickly and before competitors (Bendoly et al., 2012; Gattiker, 2007). As a result, the integration between marketing and operations as core functions of a manufacturing organisation (Slack et al., 2013) has increasingly received attention from many academics and practitioners (e.g., Hausman et al., 2002; O’Leary-Kelly & Flores, 2002; Prabhaker, 2001; Swink & Song, 2007; Tang, 2010). This is because of the importance of the marketing and operations interface to achieve more rapid responsiveness to market demand through the fit between market requirements and operations capabilities (Slack et al., 2009). Despite the importance of this work, empirical research on how to achieve and develop this integration is still limited in comparison with conceptual work (Felekoglu et al., 2013; Paiva, 2010; Sharma, 2013; Song et al., 2010). Therefore, this study is an attempt to narrow this gap by investigating why and how to manage the marketing and operations functional relationship effectively in order to become more market oriented. The framework of this research consists of four phases namely; the needs (reasons for integration), the methods (cross-functional teams), the development (potential problems), and the achievement (delivery priority). This framework represents a strategic imperative for developing the delivery performance of an organisation based on the fit between strategy (time-based strategy), organisational structure (cross-functional integration), and environment (the competitive position) (Lenz, 1980; Miller, 1988). Empirically, due to the need to develop the performance of Iraqi public industry sector, two Iraqi public textile organisations were chosen as case studies to conduct this project by using semi-structured interviews and direct observation to gather data. According to the findings of this research, it can be argued that this study would be an approach to implement market orientation in the Iraqi context albeit one which is difficult to execute.

Cross-functional teams are becoming increasingly common in organisations. However, a large proportion of these teams fail to meet their full potential as social and productive units. The present research was conducted under the assumption that a cross-functional team involves simultaneous intragroup and intergroup contact. The failure of cross-functional teams often involves the neglect of normal social psychological processes that occur in intergroup contexts that may potentially be employed to increase the likelihood of success with cross-functional teams. The social identity theory approach to intergroup relations was used to formulate two identity management strategies intended to improve the functioning of a cross-functional team. One strategy involved social interaction within an intragroup social frame. The intragroup aspect was apparent in that there was no outgroup present, with participants operating at the intergroup level of psychological processing after exposure to (successful) pre-task manipulations intended to facilitate acceptance of the task group social identity. The intragroup identity management process required increasing pre-task salience of the task group social identity, absence of any outgroup, wearing a team uniform, and performance of an intellectually challenging problem solving as a group. A second identity management process was based in an intergroup social frame. The intergroup procedure involved pre-task manipulation of social identity, wearing of a team uniform, and performance of a physically and intellectually involving problem solving task in a competitive intergroup social frame. Two separate studies were performed. In Study one, 110 university students were randomly assigned to 'mono-functional' teams (teams with no obvious basis for internal intergroup differentiation). The relative efficacy of either of the two identity management processes was assessed with regards to changes in social identity, subjective uncertainty, conceptualisation of the aggregate, similarity, heterogeneity, effort, and trust. Pre-post within groups differences were analysed Improving functioning of cross-functional teams by repeated measures ANOVA. Between groups differences were analysed with ANCOVA. The results indicated both identity management strategies resulted in improved team functioning. In general, neither strategy was notably superior to the other, however there was less subjective uncertainty reported by participants from the intragroup condition than those from the intergroup condition due to the effect of losing the competition. Participants from losing teams also became more aware of 'subgroups within the single group' than those from the intragroup condition. Participants from the intergroup condition were less likely to think of themselves as separate individuals than participants from the intragroup condition. The social identity theory approach to intergroup relations was useful for interpreting the results as well as developing the two strategies which suggests SIT/SCT provide a potentially useful conceptual base from which to develop team building processes in mono-functional teams. In study 2, 110 university students were assigned to cross-functional teams (composite task groups containing three academic subgroups). The identity management processes used in Study one were extended by drawing participant attention to the presence of subgroups within the cross-functional teams. The intragroup process involved pre-task manipulation of social identity, wearing uniforms which denoted both task group and subgroup membership, and performance of an intellectually challenging problem solving task which was performed with out an outgroup present. The intergroup identity management strategy involved pre-task manipulation of social identity, performance of a physically and intellectually involving problem solving task requiring integration of subgroup knowledge and inter-functional cooperation within a competitive intergroup context. As in the intragroup condition, participants were made aware of the social complexity of the task group through their uniforms. Participants responded differently to the two identity management processes, with the intergroup strategy proving more beneficial than the intragroup strategy. The intragroup process was marked by non-significant pre-post differences, indicating neither Improving functioning of cross-functional teams a marked improvement nor decline in group functioning. In contrast, participants from the intergroup condition reported results indicating increased post-task self-definition with the cross-functional team, increased trust, and higher effort. Losing the competition did not impact on post-task levels of any dependent variable with the exception of subjective uncertainty, where 'winners' reported less uncertainty than 'losers'. The intergroup condition gave rise to the most potential perceived 'distinctiveness threat'; however there was no sign of any threat across the array of dependent variables. Therefore it can be suggested that the intergroup identity management strategy provided some protection to the cross-functional team from the negative impact of inter-functional distinctiveness threat. All results could be explained with recourse to the concepts of SIT/SCT which suggests social identity theory has utility for interpreting results as well as developing team building processes in cross-functional teams. Future research in cross-functional team settings would benefit from the development of comprehensive measures of uncertainty, status, and heterogeneity with item content drawn from social identity and self-categorisation theories.

Purpose The purpose of the research work is to investigate the importance of the communication in the cross-functional teams and in what which communicational tools provide better convenience of communication in cross-functional team members. Methodology An empirical study including the data collection from the interviews conducted from the participants working in cross-functional teams. Qualitative approach has been adopted to understand the communicational attributes in the cross-functional teams. For the analysis both deductive and abductive approach has been used. With the help of theoretical framework and empirical data, conclusions drown for better understanding of the concept. Findings The author has divided the search in two main segments, communicational effects on cross-functional team performance and effectiveness of particular communicational tools used by team members and team leaders. Empirical findings shows that without proper communication, cross-functional team are less tend to perform well to achieve set goals as well as face to face communications are more effective than other communicational tools.  Research limitations The research work is limited to Scandinavian countries. Organizational communicational structures are not studied due to time factor. Other limitation of this research work is effect of cultural differences on communication. Implications The research work provides deep understanding of the different communicational effects on cross-functional team performance. The choice of communicational tool and proportional importance for the team members will help managers while selecting the tools of communication with the team members. Originality / value During the literature studies, author determined that there is potential of research for the communicational tools used in cross-functional team communications. The social media is taking over the traditional communicational tools which provide value for findings.

The research aims at analyzing the internal communication in a new product development project of Sandvik with expectation to explore possibilities of improvement. Throughout the research, internal communication seems to have a great impact on innovation and project performance. Several critical factors in building effective communication were identified as team size, superordinate goals, centralization of communication, early involvement, physical proximity, and leadership. Meetings were considered as a major and efficient method of communication within the project. Based on these issues, recommendations for improving internal communication within the project were suggested.

This is a study of nonconformances experienced by a laboratory of a pharmaceutical manufacturing facility in East Africa. There has been an increase in nonconformances from 216 nonconformances in 2017 to 229 in 2018 and by September 2019, 306 nonconformances were already logged. Increasing nonconformances result in delayed release of tested materials and many resources are wasted (e.g. chemicals, man hours and equipment). Analysts become frustrated, which may result in inexhaustive investigations. Understanding the reason for the increase in nonconformances will enable the facility to derive effective solutions to the identified causes, hence reducing the number of nonconformances and improving the productivity and morale of employees. This quantitative, nonexperimental, longitudinal survey study was intended to evaluate and understand the reason for increasing nonconformances. Trends of the nonconformances, previous investigations, procedure for investigation and the training given to analysts have been reviewed. Laboratory incidences were the most recurring nonconformances; and these were mainly caused by analyst errors. Corrective and Preventive Actions (CAPAs) were derived by cross functional teams whenever root causes were identified. Procedure for investigation of nonconformances refers to investigative tools. Identification of root causes to nonconformances recently became mandatory. Analysts have limited advanced industrial training on investigation of nonconformances. Another study should be carried out to understand the cause of analyst errors. The study can be rolled out to other departments at the manufacturing facility to create similar improvements. Analysts should enroll into advanced courses of industrial pharmacy to gain advanced industrial skills which they can apply in investigations to find root causes to nonconformances.

Original objectives: Understanding the regulation of abscission competence by exploring the nature and function of auxin-related gene expression changes in the leaf and pedicelAZs of tomato (as a model system), was the main goal of the previously submitted proposal. We proposed to achieve this goal by using microarray GeneChip analysis, to identify potential target genes for functional analysis by virus-induced gene silencing (VIGS). To increase the potential of accomplishing the objectives of the previously submitted proposal, we were asked by BARD to show feasibility for the use of these two modern techniques in our abscission system. Thus, the following new objectives were outlined for the one-year feasibility study: 1.to demonstrate the feasibility of the VIGS system in tomato to perform functional analysis of known abscission-related genes; 2. to demonstrate that by using microarray analysis we can identify target genes for further VIGS functional analysis. Background to the topic: It is a generally accepted model that auxin flux through the abscission zone (AZ) prevents organ abscission by rendering the AZ insensitive to ethylene. However, the molecular mechanisms responsible for acquisition of abscission competence and the way in which the auxin gradient modulates it are still unknown. Understanding this basic stage of the abscission process may provide us with future tools to control abscission for agricultural applications. Based on our previous study, performed to investigate the molecular changes occurring in leaf and stem AZs of MirabillisJalapaL., we have expanded our research to tomato, using genomic approaches that include modern techniques for gene discovery and functional gene characterization. In our one-year feasibility study, the US team has established a useful system for VIGS in tomato, using vectors based on the tobacco rattle virus (TRV), a Lcreporter gene for silencing (involved in regulation of anthocyanin biosynthesis), and the gene of interest. In parallel, the Israeli team has used the newly released Affymetrix Tomato GeneChip to measure gene expression in AZ and non-AZ tissues at various time points after flower removal, when increased sensitivity to ethylene is acquired prior to abscission (at 0-8 h), and during pedicelabscission (at 14 h). In addition, gene expression was measured in the pedicel AZ pretreated with the ethylene action inhibitor, 1-methylcyclopropene (1-MCP) before flower removal, to block any direct effects of ethylene. Major conclusions, solutions and achievements: 1) The feasibility study unequivocally established that VIGS is an ideal tool for testing the function of genes with putative roles in abscission; 2) The newly released Affymetrix Tomato GeneChip was found to be an excellent tool to identify AZ genes possibly involved in regulation and execution of abscission. The VIGS-based study allowed us to show that TAPG, a polygalacturonase specifically associated with the tomato AZ, is a key enzyme in the abscission process. Using the newly released Affymetrix Tomato GeneChip we have identified potential abscission regulatory genes as well as new AZ-specific genes, the expression of which was modified after flower removal. These include: members of the Aux/IAAgene family, ethylene signal transduction-related genes, early and late expressed transcription factors, genes which encode post-translational regulators whose expression was modified specifically in the AZ, and many additional novel AZ-specific genes which were previously not associated with abscission. This microarray analysis allowed us to select an initial set of target genes for further functional analysis by VIGS. Implications: Our success in achieving the two objectives of this feasibility study provides us with a solid basis for further research outlined in the original proposal. This will significantly increase the probability of success of a full 3-year project. Additionally, our feasibility study yielded highly innovative results, as they represent the first direct demonstration of the functional involvement of a TAPG in abscission, and the first microarray analysis of the abscission process. Using these approaches we could identify a large number of genes involved in abscission regulation, initiation and execution, and in auxin-ethylene cross-talk, which are of great importance, and could enable their potential functional analysis by VIGS.

Citrus is one of the major agricultural crops common to Israel and the United States, important in terms of nutrition, foreign exchange, and employment. The economy of both citrus industries have been chronically plagued by diseases caused by Citrus tristeza virus (CTV). The short term solution until virus-resistant plants can be used is the use of mild strain cross-protection. We are custom designing "ideal" protecting viruses to immunize trees against severe isolates of CTV by purposely inoculating existing endangered trees and new plantings to be propagated as infected (protected) citrus budwood. We crossed the substantial technological hurdles necessary to accomplish this task which included developing an infectious cDNA clone which allows in vitro manipulation of the virus and methods to then infect citrus plants. We created a series of hybrids between decline-inducing and mild CTV strains, tested them in protoplasts, and are amplifying them to inoculate citrus trees for evaluation and mapping of disease determinants. We also extended this developed technology to begin engineering transient expression vectors based on CTV as tools for genetic improvement of tree crops, in this case citrus. Because of the long periods between genetic transformation and the ultimate assay of mature tree characteristics, there is a great need for an effective system that allows the expression or suppression of target genes in fruiting plants. Virus-based vectors will greatly expedite progress in citrus genetic improvement. We characterized several components of the virus that provides necessary information for designing virus-based vectors. We characterized the requirements of the 3 ’-nontranslated replication promoter and two 3 ’-ORF subgenomic (sg) mRNA controller elements. We discovered a novel type of 5’-terminal sgRNAs and characterized the cis-acting control element that also functions as a strong promoter of a 3 ’-sgRNA. We showed that the p23 gene controls negative-stranded RNA synthesis and expression of 3 ’ genes. We identified which genes are required for infection of plants, which are host range determinants, and which are not needed for plant infection. We continued the characterization of native dRNA populations and showed the presence of five different classes including class III dRNAs that consists of infectious and self-replicating molecules and class V dRNAs that contain all of the 3 ’ ORFs, along with class IV dRNAs that retain non-contiguous internal sequences. We have constructed and tested in protoplasts a series of expression vectors that will be described in this proposal.

Constraints on water resources and the environment necessitate more efficient use of water. The key to efficient management is an understanding of the physical and physiological processes occurring in the soil-root hydraulic continuum.While both soil and plant leaf water potentials are well understood, modeled and measured, the root-soil interface where actual uptake processes occur has not been sufficiently studied. The water potential at the root-soil interface (yᵣₒₒₜ), determined by environmental conditions and by soil and plant hydraulic properties, serves as a boundary value in soil and plant uptake equations. In this work, we propose to 1) refine and implement a method for measuring yᵣₒₒₜ; 2) measure yᵣₒₒₜ, water uptake and root hydraulic conductivity for wild type tomato and Arabidopsis under varied q, K⁺, Na⁺ and Cl⁻ levels in the root zone; 3) verify the role of MIPs and ion channels response to q, K⁺ and Na⁺ levels in Arabidopsis and tomato; 4) study the relationships between yᵣₒₒₜ and root hydraulic conductivity for various crops representing important botanical and agricultural species, under conditions of varying soil types, water contents and salinity; and 5) integrate the above to water uptake term(s) to be implemented in models. We have made significant progress toward establishing the efficacy of the emittensiometer and on the molecular biology studies. We have added an additional method for measuring ψᵣₒₒₜ. High-frequency water application through the water source while the plant emerges and becomes established encourages roots to develop towards and into the water source itself. The yᵣₒₒₜ and yₛₒᵢₗ values reflected wetting and drying processes in the rhizosphere and in the bulk soil. Thus, yᵣₒₒₜ can be manipulated by changing irrigation level and frequency. An important and surprising finding resulting from the current research is the obtained yᵣₒₒₜ value. The yᵣₒₒₜ measured using the three different methods: emittensiometer, micro-tensiometer and MRI imaging in both sunflower, tomato and corn plants fell in the same range and were higher by one to three orders of magnitude from the values of -600 to -15,000 cm suggested in the literature. We have added additional information on the regulation of aquaporins and transporters at the transcript and protein levels, particularly under stress. Our preliminary results show that overexpression of one aquaporin gene in tomato dramatically increases its transpiration level (unpublished results). Based on this information, we started screening mutants for other aquaporin genes. During the feasibility testing year, we identified homozygous mutants for eight aquaporin genes, including six mutants for five of the PIP2 genes. Including the homozygous mutants directly available at the ABRC seed stock center, we now have mutants for 11 of the 19 aquaporin genes of interest. Currently, we are screening mutants for other aquaporin genes and ion transporter genes. Understanding plant water uptake under stress is essential for the further advancement of molecular plant stress tolerance work as well as for efficient use of water in agriculture. Virtually all of Israel’s agriculture and about 40% of US agriculture is made possible by irrigation. Both countries face increasing risk of water shortages as urban requirements grow. Both countries will have to find methods of protecting the soil resource while conserving water resources—goals that appear to be in direct conflict. The climate-plant-soil-water system is nonlinear with many feedback mechanisms. Conceptual plant uptake and growth models and mechanism-based computer-simulation models will be valuable tools in developing irrigation regimes and methods that maximize the efficiency of agricultural water. This proposal will contribute to the development of these models by providing critical information on water extraction by the plant that will result in improved predictions of both water requirements and crop yields. Plant water use and plant response to environmental conditions cannot possibly be understood by using the tools and language of a single scientific discipline. This proposal links the disciplines of soil physics and soil physical chemistry with plant physiology and molecular biology in order to correctly treat and understand the soil-plant interface in terms of integrated comprehension. Results from the project will contribute to a mechanistic understanding of the SPAC and will inspire continued multidisciplinary research.

The known advantages and disadvantages of each system do not presently allow a clear decision. Whether essentially zero operating costs can offset the less dependable cooling of natural ventilation systems is question this report hopes to answer. Major Conclusions US It was concluded very early on that FV greenhouses without evaporative pad cooling are not competitive with NV greenhouses during hot weather. During the first year, the US team found that average air temperatures were always higher in the FV houses, compared to the NV houses, when evaporative pad cooling was not used, regardless of ventilation rate in the FV houses or the vent configuration in the NV houses. Canopy temperatures were also higher in the FV ventilated houses when three vents were used in the NV houses. A second major conclusion was that the US team found that low pressure fogging (4 atm) in NV houses does not completely offset the advantage of evaporative pad cooling in FV houses. High pressure fog (65 atm) is more effective, but considerably more expensive. Israel Experiments were done with roses in the years 2003-2005 and with tomatoes in 2005. Three modes of natural ventilation (roof, side and side + roof openings) were compared with a fan-ventilated (with evaporative cooling) house. It was shown that under common practice of fan ventilation, during summer, the ventilation rate is usually lower with NV than with FV. The microclimate under both NV and FV was not homogeneous. In both treatments there were strong gradients in temperature and humidity in the vertical direction. In addition, there were gradients that developed in horizontal planes in a direction parallel to the direction of the prevailing air velocity within the greenhouse. The gradients in the horizontal direction appear to be larger with FV than with NV. The ratio between sensible and latent heat fluxes (Bowen ratio) was found to be dependent considerably on whether NV or FV is applied. This ratio was generally negative in the naturally ventilated house (about -0.14) and positive in the fan ventilated one (about 0.19). Theoretical models based on Penman-Monteith equation were used to predict the interior air and crop temperatures and the transpiration rate with NV. Good agreement between the model and experimental results was obtained with regard to the air temperature and transpiration with side and side + roof ventilation. In Israel and in the U.S., Streptococcus iniae is responsible for considerable losses in various fish species. Poor understanding of its virulence factors and limited know-how-to of vaccine formulation and administration are the main reasons for the limited efficacy of vaccines. Our strategy was that in order to Improve control measures, both aspects should be equally addressed. Our proposal included the following objectives: (i) construction of host-pathogen interaction models; (ii) characterization of virulence factors and immunodominant antigens, with assessment of their relative importance in terms of protection and (iii) genetic identification of virulence factors and genes, with evaluation of the protective effect of recombinant proteins. We have shown that two different serotypes are involved. Their capsular polysaccharides (CPS) were characterized, and proved to play an important role in immune evasion and in other consequences of the infection. This is an innovative finding in fish bacteriology and resembles what, in other fields, has become apparent in the recent years: S. iniae alters surface antigens. By so doing, the pathogen escapes immune destruction. Immunological assays (agar-gel immunodiffusion and antibody titers) confirmed that only limited cross recognition between the two types occurs and that capsular polysaccharides are immunodominant. Vaccination with purified CPS (as an acellular vaccine) results in protection. In vitro and ex-vivo models have allowed us to unravel additional insights of the host-pathogen interactions. S. iniae 173 (type II) produced DNA fragmentation of TMB-8 cells characteristic of cellular necrosis; the same isolate also prevented the development of apoptosis in NCC. This was determined by finding reduced expression of phosphotidylserine (PS) on the outer membrane leaflet of NCC. NCC treated with this isolate had very high levels of cellular necrosis compared to all other isolates. This cellular pathology was confirmed by observing reduced DNA laddering in these same treated cells. Transmission EM also showed characteristic necrotic cellular changes in treated cells. To determine if the (in vitro) PCD/apoptosis protective effects of #173 correlated with any in vivo activity, tilapia were injected IV with #173 and #164 (an Israeli type I strain). Following injection, purified NCC were tested (in vitro) for cytotoxicity against HL-60 target cells. Four significant observations were made : (i) fish injected with #173 had 100-400% increased cytotoxicity compared to #164 (ii) in vivo activation occurred within 5 minutes of injection; (iii) activation occurred only within the peripheral blood compartment; and (iv) the isolate that protected NCC from apoptosis in vitro caused in vivo activation of cytotoxicity. The levels of in vivo cytotoxicity responses are associated with certain pathogens (pathogen associated molecular patterns/PAMP) and with the tissue of origin of NCC. NCC from different tissue (i.e. PBL, anterior kidney, spleen) exist in different states of differentiation. Random amplified polymorphic DNA (RAPD) analysis revealed the "adaptation" of the bacterium to the vaccinated environment, suggesting a "Darwinian-like" evolution of any bacterium. Due to the selective pressure which has occurred in the vaccinated environment, type II strains, able to evade the protective response elicited by the vaccine, have evolved from type I strains. The increased virulence through the appropriation of a novel antigenic composition conforms with pathogenic mechanisms described for other streptococci. Vaccine efficacy was improved: water-in-oil formulations were found effective in inducing protection that lasted for a period of (at least) 6 months. Protection was evaluated by functional tests - the protective effect, and immunological parameters - elicitation of T- and B-cells proliferation. Vaccinated fish were found to be resistant to the disease for (at least) six months; protection was accompanied by activation of the cellular and the humoral branches.
