One of the most celebrated gardens in early modern Rome was built by Cardinal Federico Cesi (d. 1565) near St. Peter’s Basilica. Earlier studies of the site have concentrated on the famous sixteenth-century antiquities collection displayed in the garden. The Afterlife of the Cesi Garden: Family Identity, Politics, and Memory in Early Modern Rome shifts the scholarly focus to also examine the changing appearance, functions, and the broader social, political, and economic significance of the garden for the Cesi family and for the city of Rome over the course of the seventeenth and eighteenth centuries. Through a close analysis of visual evidence, unpublished archival documents, and a plan of the garden by the architect Giovanni Battista Contini (d. 1723), Katherine M. Bentz demonstrates that the long post-Renaissance afterlife of the Cesi Garden reveals the ways in which politics shaped specific urban environments in Rome, how aristocratic Romans considered and used gardens over generations, and the vital and symbolic role that the garden played for centuries.

We define a notion of a splitting tree, by means of which we prove that every analytic countably splitting family contains a closed countably splitting family. An application of this notion solves a problem of Blass. On the other hand we show that there exists an Fσ splitting family that does not contain a closed splitting family.

Twelve clones of monkey DNA obtained by a procedure that enriches 10(3)- to 10(4)-fold for nascent sequences activated early in S phase (G. Kaufmann, M. Zannis-Hadjopoulos, and R. G. Martin, Mol. Cell. Biol. 5:721-727, 1985) have been examined. Only 2 of the 12 ors sequences (origin-enriched sequences) are unique (ors1 and ors8). Three contain the highly reiterated Alu family (ors3, ors9, and ors11). One contains the highly reiterated alpha-satellite family (ors12), but none contain the Kpn family. Those remaining contain middle repetitive sequences. Two examples of the same middle repetitive sequence were found (ors2 and ors6). Three of the middle repetitive sequences (the ors2-ors6 pair, ors5, and ors10) are moderately dispersed; one (ors4) is highly dispersed. The last, ors7, has been mapped to the bona fide replication origin of the D loop of mitochondrial DNA. Of the nine ors sequences tested, half possess snapback (intrachain reannealing) properties.

AbstractLet ℓ be a prime number. It is not known whether every finite ℓ-group of rank n≥1 can be realized as a Galois group over ${\Bbb Q}$ with no more than n ramified primes. We prove that this can be done for the (minimal) family of finite ℓ-groups which contains all the cyclic groups of ℓ-power order and is closed under direct products, (regular) wreath products and rank-preserving homomorphic images. This family contains the Sylow ℓ-subgroups of the symmetric groups and of the classical groups over finite fields of characteristic not ℓ. On the other hand, it does not contain all finite ℓ-groups.

Major intrinsic proteins (MIPs) family include the group of proteins also called channel proteins or water channels. Aquaporins facilitate the transport of water and other uncharged small molecules, forming pores in cellular boundaries which help in plant growth, development, and sometimes in plant adaptation. They usually have six tranSmembrane alpha-helical domains with conserved amino acids residues along with hydrophobic and hydrophilic regions. Though aquaporins were found both in animals and plants, also in bacteria, fungi cells, it comprises the largest and most complex group in plant species. A little is known about the aquaporins family of the plant Momordica charantia. About 25 aquaporins of its are characterized in this study. The aquaporins of M. charantia are divided into five sub-families here, they are PIPs, TIPs, XIPs, SIPs, and NIPs. There are twelve PIPs, three TIPs, eight NIPs, and one AQPs in both XIP and SIP. In the XIP subfamily, it contains SPI other than the NPA motif. Moreover, SIP and NIP subfamily also contain different motifs both in loop B and loop E other than NPA motifs. McSIPs contain NPT in loop B and McNIPs contain NPS in loop B (McNIP1; 1, McNIP1; 3 and McNIP3; 1). Again, McNIPs contain NPV in loop E (McNIP2; 2, McNIP3; 1). The conserved residues and motifs of this plant show its transportation of water and the possibility of other molecules' transportation through those channel proteins.

The family Apiaceae includes 474 genera and 3992–4050 species that are distributed around the globe. Of all the variety of species, our attention was drawn to the raw material of anise – Anisum vulgare Gaertrn.; fennel – Foeniculum vulgare Mill., which contains derivatives of phenylpropane; coriander – Coriandrum sativum L.; dill – Anethum graveolens L.; caraway – Carum carvi L., which is rich in monoterpenoid derivatives. These plants are widely used in medical practice and known to have antimicrobial, antispasmodic, expectorant, antihypertensive, diuretic, choleretic, carminative effects. This versatility is due to the presence of different groups of biologically active substances in the raw materials of plants. The aim of the research is to determine, by means of gas chromatography, a component composition of fruit tinctures of Anisum vulgare Gaertrn., Foeniculum vulgare Mill., Coriandrum sativum L. Anethum graveolens L. and Carum carvi L. Materials and methods. Tinctures were prepared in ratio 1:5 (extractant – 70 % ethyl alcohol) from anise, fennel, coriander, dill, and caraway fruits, which had been harvested in the experimental field of Zaporizhzhia State Medical University in August 2019. Qualitative and quantitative determination of the active compounds was performed using an Agilent 7890B gas chromatograph with a 5977B mass spectrometric detector. The NIST14 mass spectrum library was used to identify the components. Results. It has been revealed that the tinctures of Anisum vulgare Gaertrn., Foeniculum vulgare Mill., Coriandrum sativum L. Anethum graveolens L., and Carum carvi L. differ in qualitative and quantitative composition of components of chromato-mass spectrometry. Tinctures of Anethum fructus contain 28 components, among which 8.028 RT (-)-Carvone (52.63 %) and 4.861 RT D-Limonene (17.74 %) prevail. Tinctures of Coriandrum fructus contain 37 components with the highest content of 11,016 RT Benzene, 1-(1,5-dimethyl-4-hexenyl)-4-methyl (20.7 %) and 19.372 RT 1-(4-hydroxy-3-methoxyphenyl)dec-4-en-3-one (8.57 %). In the tincture of Carum carvi L., which contains 33 components, the following may be considered predominant: 7.988 RT Benzaldehyde, 4-(1-methylethyl) (47.37 %) and 19,311 RT undetermined compound (5.88 %). The tincture of Anisum fructus has 22 components, with the highest content of 8.531 RT Estragole (60.32 %) and 4.852 RT D-Limonene (5.15 %); Foeniculum fructus tincture contains 52 components, among which 5.745 RT Fenchone (15.39 %) and 8.5 RT Anethole (11.37 %) prevail. Conclusions. The results of gas chromatography of fruits of the Apiaceae family significantly expand the information about the chemical composition of raw materials and indicate the prospects for use in pharmaceutical and medical practice. Analyzing the results of gas chromatography, it has been found that tinctures of fruits of the Apiaceae family differ in qualitative and quantitative composition of components. Tinctures of Foeniculum fructus contain 52 components, Coriandrum fructus – 37 components, Carum carvi L. contain 33 components, Anethum fructus – 28 components, Anisum fructus – 22 components. The obtained experimental data can be used to create regulatory documentation for a particular type of medicinal plant raw materials.

Introduction. The problem discussed in the article concerns family pedagogy, which has recently been separated from the pedagogical sciences as an independent scientific field. The object of the research is family development trends and its present state, the topic is the essence of family upbringing, this process mechanisms and their influence on personal development. Family pedagogy does not attempt to create a single rule that is binding on every family and clearly regulates the upbringing of children. It studies only those situations that are controversial and cause the young its difficulty. As all scientific knowledge, its conclusions can never fully replace wisdom and life experience. Preparing youth for family relationships cannot help but affect this aspect of pedagogical activity, more precisely on the preparation of youth for family relationships.

The semaphorin family consists of a set of secreted and transmembrane proteins which contain a domain of approximately 500 amino acids called the semaphorin domain. The investigation of the role of semaphorin proteins in the nervous system has established them as chemorepellents of axons. Recent studies have identified the semaphorin proteins on the surface of cells in the immune system and in the genomes of two lytic viruses. These strongly suggest the possible involvement of the semaphorin proteins in the immune system. This study aims to understand the expression patterns of four semaphorin genes, Sema3B, Sema4A, Sema4F, and Sema6C in the lymphoid tissues of wild type mice. These proteins were chosen to represent the three subfamilies III, IV, and VI. The first family contains secreted proteins which have immunoglobulin domains (III), the second contains transmembrane proteins which have immunoglobulin domains (IV), and the third contains transmembrane proteins which do not have immunoglobulin domains (VI).

The related species Limnodynastes fletcheri contains only one peptide in its glandular extract; this compound, which we have named fletcherin, is unusual amongst amphibian peptides in that the N-terminal group corresponds to Ala. The primary structures of the dynastins and fletcherin are described.

TPX2 (Targeting Protein for Xklp2) is an evolutionary conserved microtubule-associated protein important for microtubule nucleation and mitotic spindle assembly. The protein was described as an activator of the mitotic kinase Aurora A in humans and the Arabidopsis AURORA1 (AUR1) kinase. In contrast to animal genomes that encode only one TPX2 gene, higher plant genomes encode a family with several TPX2-LIKE gene members (TPXL). TPXL genes of Arabidopsis can be divided into two groups. Group A proteins (TPXL2, 3, 4, and 8) contain Aurora binding and TPX2_importin domains, while group B proteins (TPXL1, 5, 6, and 7) harbor an Xklp2 domain. Canonical TPX2 contains all the above-mentioned domains. We confirmed using in vitro kinase assays that the group A proteins contain a functional Aurora kinase binding domain. Transient expression of Arabidopsis TPX2-like proteins in Nicotiana benthamiana revealed preferential localization to microtubules and nuclei. Co-expression of AUR1 together with TPX2-like proteins changed the localization of AUR1, indicating that these proteins serve as targeting factors for Aurora kinases. Taken together, we visualize the various localizations of the TPX2-LIKE family in Arabidopsis as a proxy to their functional divergence and provide evidence of their role in the targeted regulation of AUR1 kinase activity.

It is known that the disconnected Julia set of any polynomial map does not contain buried Julia components. But such Julia components may arise for rational maps. The first example is due to Curtis T. McMullen who provided a family of rational maps for which the Julia sets are Cantor of Jordan curves. However, all known examples of buried Julia components, up to now, are points or Jordan curves and comes from rational maps of degree at least five. This paper introduces a family of hyperbolic rational maps with disconnected Julia set whose exchanging dynamics of postcritically separating Julia components is encoded by a weighted dynamical tree. Each of these Julia sets presents buried Julia components of several types: points, Jordan curves, but also Julia components which are neither points nor Jordan curves. Moreover, this family contains some rational maps of degree three with explicit formula that answers a question McMullen raised.

Banna virus (BAV) particles contain seven structural proteins: VP4 and VP9 form an outer-capsid layer, whilst the virus core contains three major proteins (VP2, VP8 and VP10) and two minor proteins (VP1 and VP3). Sequence analysis showed that VP3 contains motifs [Kx(I/V/L)S] and (Hx n H) that have previously been identified in the guanylyltransferases of other reoviruses. Incubation of purified BAV-Ch core particles with [α-32P]GTP resulted in exclusive covalent labelling of VP3, demonstrating autoguanylation activity (which is considered indicative of guanylyltransferase activity). Recombinant VP3 prepared in a cell-free expression system was also guanylated under similar reaction conditions, and products were synthesized (in the presence of non-radiolabelled GDP) that co-migrated with GMP, GDP and GpppG during TLC. This reaction, which required magnesium ions for optimum activity, demonstrates that VP3 possesses nucleoside triphosphatase (GTPase) activity and is the BAV guanylyltransferase (RNA ‘capping’ enzyme).

Background and Objectives: Direct observation is a critical part of assessing learners’ achievement of the Accreditation Council for Graduate Medical Education (ACGME) Milestones and subcompetencies. Little research exists identifying the content of peer feedback among residents; this study explored the content of residents’ peer assessments as they relate to ACGME Milestone subcompetencies in a family medicine residency program. Methods: Using content from a mobile app-based observation tool (M3App), we examined resident peer observations recorded between June 2014 and November 2017, tabulating frequency of observation for each ACGME subcompetency and calculating the proportion of observations categorized under each subcompetency, as well as for each postgraduate year (PGY) class. We also coded each observation on three separate dimensions: “positive,” “constructive,” and “actionable.” We used the χ2 test for independence, and estimated odds ratios and 95% confidence intervals for two-by-two comparisons to compare numbers of observations within each category. Results: Our data include 886 peer observations made by 54 individual residents. The most frequently observed competencies were in patient care, communication, and professionalism (56%, 47%, and 38% of observations, respectively). Practice-based learning and improvement was observed least frequently (16% of observations). On average, 97.25% of the observations were positive, 85% were actionable, and 6% were constructive. Conclusions: When asked to review their peers, residents provide comments that are primarily positive and actionable. In addition, residents tend to provide more feedback on certain subcompetencies compared to others, suggesting that programs may rely on peer feedback for specific subcompetencies. Peers can provide perspective on the behaviors and skills of fellow residents.

Abstract The class I gene family of the major histocompatibility complex (MHC) of the miniature swine (SLA) contains seven members. Six of these are highly homologous, whereas the seventh, PD6, is only distantly related. A genomic clone containing PD6 has been isolated and characterized. The PD6 SLA gene, although only 55% homologous to the other SLA class I sequences, is still a member of the family; its size and exon/intron organization are similar to other class I genes. When aligned with other SLA genes, exons 1 to 6 of PD6 have open reading frames; exon 7 contains a termination codon. PD6 is transcribed in transfected mouse L cells where its expression is enhanced by interferon. In vivo expression of PD6 is observed in a variety of tissues, with the highest levels in mature lymphoid tissues. Peripheral T cells contain high levels of PD6-specific RNA relative to B cells, whereas no PD6 transcripts are detectable in thymocytes.

AbstractThe multicopper blue protein family, which contains cupredoxin-like domains as a structural unit, is one of the most diverse groups of proteins. This protein family is divided into two functionally different types of enzymes: multicopper oxidase and nitrite reductase. Multicopper oxidase catalyzes the oxidation of the substrate and then reduces dioxygen. The structures of many multicopper oxidases are already known, and until recently they were classified into two main groups: the three- and six-domain types. Both function as monomers and have three spectroscopically different copper sites: Types I (blue), II, and III (tri-nuclear). Nitrite reductase is a closely related protein that contains Types I and II (mono-nuclear) coppers but reduces nitrite instead of dioxygen. Nitrite reductase, which consists of two domains, forms a homotrimer. Multicopper oxidase and nitrite reductase share similar structural architectures and also contain Type I copper. Therefore, it is proposed that they have a common ancestor protein. Recently, some two-domain type multicopper oxidases have been found and their crystal structures have been determined. They have a trimeric quaternary structure and contain an active site at the molecular interface such as nitrite reductase. These results support previous hypotheses and provide an insight into the molecular evolution of multicopper blue proteins.

src family tyrosine kinases contain two noncatalytic domains termed src homology 3 (SH3) and SH2 domains. Although several other signal transduction molecules also contain tandemly occurring SH3 and SH2 domains, the function of these closely spaced domains is not well understood. To identify the role of the SH3 domains of src family tyrosine kinases, we sought to identify proteins that interacted with this domain. By using the yeast two-hybrid system, we identified p62, a tyrosine-phosphorylated protein that associates with p21ras GTPase-activating protein, as a src family kinase SH3-domain-binding protein. Reconstitution of complexes containing p62 and the src family kinase p59fyn in HeLa cells demonstrated that complex formation resulted in tyrosine phosphorylation of p62 and was mediated by both the SH3 and SH2 domains of p59fyn. The phosphorylation of p62 by p59fyn required an intact SH3 domain, demonstrating that one function of the src family kinase SH3 domains is to bind and present certain substrates to the kinase. As p62 contains at least five SH3-domain-binding motifs and multiple tyrosine phosphorylation sites, p62 may interact with other signalling molecules via SH3 and SH2 domain interactions. Here we show that the SH3 and/or SH2 domains of the signalling proteins Grb2 and phospholipase C gamma-1 can interact with p62 both in vitro and in vivo. Thus, we propose that one function of the tandemly occurring SH3 and SH2 domains of src family kinases is to bind p62, a multifunctional SH3 and SH2 domain adapter protein, linking src family kinases to downstream effector and regulatory molecules.

BackgroundGovernment leaders have called for increased use of video-consulting (VC) in clinical practice. Although provided by the private sector, it is rarely used in NHS primary care. It is unclear how VC differs from telephone (TC) or face-to-face consulting (FTFC), or how useful clinicians and patients find it.AimTo assess the feasibility and acceptability to patients and clinicians of running a VC service in general practice and to explore differences in duration and content compared with FTFC and TC.MethodVC equipment was installed in six GP practices and 13 clinicians were asked to identify patients who required follow-up consultations and who had access to a smart-device or laptop. They were offered the choice of a VC, TC, or FTFC. Consultations were audiorecorded and content analysed using the RIAS system. Patient and clinician views were obtained by questionnaires and interviews.ResultsOne hundred and forty-seven patients were recruited (FTFC = 51, TC = 51, VC = 45). VC users were generally younger. VCs and TCs were shorter (mean VC 5.9, TC 5.6, FTFCs 9.6 minutes), addressed fewer problems (mean VC 1.5, TC 1.7, FTFC 2.0) and had less information exchange than FTFCs. Technical problems were common, but VCs offered significant advantages over TC in terms of non-verbal communication. VC was considered particularly useful to working and housebound people and for those with mental health problems.ConclusionVC has distinct advantages over TC. When integrated into current systems VC will provide an alternative to FTFC. Where formal physical examination is not required (discussing test results, medication changes and mental health) it offers considerable time savings to patients.

Virtually every tropical lowland forest contains at least one echimyid species, and many areas contain half a dozen or more species. The commonest terrestrial representatives of the family are within the genus Proechimys, and several species such as Proechimys semispinosus (Central American spiny rat) have been the subject of intensive ecological studies (e.g. Adler 1994, 1996; Adler & Beatty 1997, Alberico & Gonzalez 1993, Fleming 1971, Gliwicz 1984, Gonzalez-M. & Alberico 1993). P. semispinosus is often the most abundant rodent throughout its geographic range in Central America and northwestern South America, and its ecology is now fairly well-known.

We constructed the family tree database (DB) by using a new family code system that can logically express interpersonal family relationships and by comparing and complementing health insurance eligibility data and resident register data of the National Health Information Database (NHID). In the family tree DB, Parents and grandparents are matched for more than 95% of those who were born between 2010 and 2017. Codes for inverse relationships and extended relationships are generated using sequences of the three-digit basic family codes. The family tree DB contains variables such as sex, birth year, family relations, and degree of kinship (maximum of 4) between subjects and family members. Using the family tree DB, we find that prevalence rates of hypertension, diabetes, ischemic heart disease, cerebrovascular disease, and cancer are higher for those with family history. The family tree DB may omit some relationships due to incomplete past data, and some family relations cannot be uniquely determined because the source data only contain relationships between head and members of the household. The family tree DB is a part of the NHID, and researchers can submit requests for data on the website at http://nhiss.nhis.or.kr. Requested data will be provided after approval from the data service review board. However, the family tree DB can be limitedly provided for studies with high public value in order to maximize personal information protection.

Expression of cold-regulated genes needed for protection against freezing stress is mediated, in part, by the CBF transcription factor family. Previous studies with temperate cereals suggested that the CBF gene family in wheat was large, and that CBF genes were at the base of an important low temperature tolerance trait. Therefore, the goal of our study was to identify the CBF repertoire in the freezing-tolerant hexaploid wheat cultivar Norstar, and then to examine if the coding region of CBF genes in two spring cultivars contain polymorphisms that could affect the protein sequence and structure. Our analyses reveal that hexaploid wheat contains a complex CBF family consisting of at least 65 CBF genes of which 60 are known to be expressed in the cultivar Norstar. They represent 27 paralogous genes with 1–3 homeologous copies for the A, B, and D genomes. The cultivar Norstar contains two pseudogenes and at least 24 additional proteins having sequences and (or) structures that deviate from the consensus in the conserved AP2 DNA-binding and (or) C-terminal activation-domains. This suggests that in cultivars such as Norstar, low temperature tolerance may be increased through breeding of additional optimal alleles. The examination of the CBF repertoire present in the two spring cultivars, Chinese Spring and Manitou, reveals that they have additional polymorphisms affecting conserved positions in these domains. Understanding the effects of these polymorphisms will provide additional information for the selection of optimum CBF alleles in Triticeae breeding programs.

The research work is devoted to the study of the biodiversity of medicinal plants in the Kyzylzhar district of the North Kazakhstan region. Compiled a list of medicinal species and taxonomic composition distribution. Based on our research, 162 species from 131 genera and 40 families were identified. A large family containing 12 species (18.75%) of medicinal plants used in official medicine is Rosaceae. In second place is the aster family (Asteraceae), which includes 10 plants from 9 genera. The legume family (Fabaceae) contains 8 plants from 4 genera, the buckwheat family (Polygonaceae) includes 6 medicinal plants from 3 genera. Of these, 22 families are represented by only one species and one genus, for example, these are such families as: Iridaceae, Asparagaceae, Utricaceae, Lamiaceae and others. A large family containing 12 species (12.24%) of medicinal plants used in traditional medicine is the Asteraceae family. 7 species of medicinal plants contain families: ranunculaceae (Ranunculaceae), legumes (Fabaceae), umbellate (Apiaceae). The family Rosaceae (Rosaceae) consists of 6 species of medicinal plants. The Rosaceae family, which is a large family in terms of the number of species of medicinal plants used by official medicine, is considered in more detail. The useful properties of some medicinal plants belonging to traditional and alternative medicine are described.

We analyzed the histone mRNA population found in several adult tissues of the sea urchin Strongylocentrotus purpuratus and in testis of Lytechinus pictus. Unique species of H1 and H2b mRNAs encoding the sperm-specific histone subtypes can be found exclusively in testis RNA. S. purpuratus contains two distinct testis-specific H1 transcripts, while L. pictus contains one such transcript. Each of these mRNAs is larger than either early or late embryonic H1 mRNAs. Other somatic adult tissues contain transcripts derived from members of the late embryonic H1 histone gene family. S. purpuratus contains one H2b transcript found exclusively in testis, while L. pictus contains two such H2b mRNAs. Similarly, in tissues other than testis, late H2b transcripts were found. While there is no sperm-specific H2a protein, a limited set of late histone H2a genes encoding primarily the H2a-beta subtype is expressed in testis. The majority of the H2a protein found in diploid adult tissues is also the H2a-beta subtype; however, the size of the H2a transcripts differs between testis and other tissues. We conclude that different members of the late H2a gene family are differentially expressed in embryos and adult tissues. We prepared and characterized cDNA clones encoding the sperm-specific H2b protein as well as the H2a-beta protein found in testis.
