Experiments were conducted to determine the currents involved in the pacemaker activity of aggregates and single cells from the embryonic chick heart. Two microelectrode voltage clamp studies of embryonic chick ventricular heart cell aggregates revealed two time-dependent current components in the pacemaker range of potentials ($-$60 to $-$120 mV). Barium (Ba, 5 mM) blocked the more negatively activated time-dependent component unmasking a component which remained inwardly directed for hyperpolarizing steps beyond the potassium equilibrium potential (E$ sb{ rm K}$). This component, which was blocked by cesium (Cs, 2 mM), is consistent with an inward current which activates upon hyperpolarization (the I, model) as proposed by DiFrancesco (1981a,b), for Purkinje fibers.
In order to minimize the problems associated with the accumulation/depletion of ions in the extracellular space during voltage clamp experiments, studies were carried out on single ventricular cells or small clusters of ventricular cells.

Establishment of the biochemical and molecular nature of cardiac development is essential for us to understand the relationship between genetic and morphological aspects of heart formation. The molecular mechanisms that underly heart development are still not clearly defined. To address this issue we have used two approaches to identify genes involved in early chick cardiac development. Differential display previously conducted in our laboratory led to the identification of two gene fragments differentially expressed in the heart that are further described in this thesis. The full-length cDNA sequence of both eukaryotic translation initiation factor-2b (eIF-2b) and NADH cytochrome b5 reductase (b5R) were isolated using library screening. The upreglation of these genes during heart development is expected given the heart is the first functional organ to form in vertebrates and protein synthesis and cell metabolism at this stage of development is maximal. Limitations in the differential display approach led to the development and optimisation of a subtractive hybridisation approach for use with small amounts of cells or tissue. To focus on cardiac gene expression during the initial phases of heart development, subtractive hybridization was performed between the cardiogenic lateral plate mesoderm of Hamburger and Hamilton stage 4 embryos and the heart primordia of stage 9 embryos. Of the 87 independent clones identified by this procedure, 59 matched known sequences with high homology, 25 matched unknown expressed sequence tag (EST) sequences with high homology, and 3 did not match any known sequence on the database. Known genes isolated included those involved in transcription, translation, cell signalling, RNA processing, and energy production. Two of these genes, high mobility group phosphoprotein A2 (HMGA2) and C1-20C, an unknown gene, were chosen for further characterisation. The role of each gene in early chick heart development and indeed development in general, was addressed using techniques such as in situ hybridisation, transfection analysis, in ovo electroporation and RNAi. HMGA2 is a nuclear phosphoprotein commonly referred to as an architectural transcription factor due to its ability to modulate DNA conformation. In keeping with this function, HMGA2/GFP fusion protein was shown to localise to the nucleus and in particular, the nucleolus. In situ hybridisation analysis suggested a role for HMGA2 in heart and somite development. HMGA2 expression was first detected at HH stage 5 in the lateral plate mesoderm, a region synonymous with cells specified to the cardiac fate. HMGA2 was also strongly expressed in the presomitic segmental plate mesoderm and as somites developed from the segmental plate mesoderm, the expression of HMGA2 showed an increasingly more restricted domain corresponding to the level of maturation of the somite. Restriction of HMGA2 expression was first detected in the dorsal region of the epithelial somite, then the dorsomedial lip of the dermomyotome, and finally the migrating epaxial myotome cells. The novel intronless gene, C1-20C, predicts a protein of 148 amino acids containing a putative zinc finger binding domain and prenyl binding motif. Zinc binding assays showed that the zinc finger domain of C1-20C/MBP fusion protein bound over six times the quantity of zinc compared to MBP alone, although not in a 1:1 stoichiometric molar ratio. C1-20C/GFP fusion protein was shown to localise to as yet unidentified intracellular cytoplasmic vesicular compartments. These compartments did not colocalise with the endosome/lysosome pathway, aparently ruling out a role for C1-20C in protein trafficking, recycling or degradation. Expression of C1-20C in the chick embryo suggests a possible role in heart and notochord development and preliminary results using siRNA suggest that C1-20C is involved in normal heart looping.

Transient Receptor Potential channels (TRP) are a superfamily of cationic specific ionchannels that are regulated by various stimuli such as temperature, pH, mechanical stress, ligandsand ion concentration. The role of TRP channels in disease states such as autosomal dominantpolycystic kidney disease, cancer metastasis, and developmental defects lend credence to thebelief that they play an important part in epithelial morphogenesis events. The development ofsomites, neural tube closure and migration of neural crest cells to form things such as the faceand heart is a good developmental model for the aforementioned cellular processes. We haveshown that TRP channels can be found in the developing ectoderm, hindbrain, and heart and thatthe inhibition of TRP channels in a developing embryo results in phenotypes suggestingperturbation of cellular remodeling processes. This leads to the question of the specific role ofTRP channels in the epithelial mesenchymal transition and remodeling in developing chickembryos.

Clostridium difficile is a pathogen for both humans and animals and is often associated with antibiotic-associated diarrhea. Recently, several human cases of C. difficile-infection with increased mortality and morbidity have been reported. In studies performed in different countries C. difficile has been found in meat. Therefore the question whether C. difficile can be a zoonotic agent has been raised. The aim of this study was to optimize a method for isolation of C. difficile from faeces. When C. difficile is isolated from animals that do not have diarrhea the sample must be cultivated in an enrichment broth. Parameters influencing the enrichment were tested such as enrichment before and after spore selection, enrichment time, alcohol and heat chock for spore selection and if the samples had to be centrifuged or not before cultivation on agar plates. Enrichment in broth before spore selection was better than after. Heat and alcohol chock showed similar results, therefore you can chose which method you want. Cultivation from the pellet after centrifugation of the sample was better than cultivating directly from the inoculated broth. When the sample had low concentration of bacteria long enrichment time, 7 days or more, was best. The next step will be isolation of C. difficile from food-producing animals and humans and the strains will then be compared to se if the same strain is found in humans and in animals, to se if C. difficile-infection can be a zoonoz.

The dextral heart looping is the first morphological manifestation of left-right (L-R) asymmetry during vertebrate development. There is a well-established genetic pathway underlying the rightward heart looping process in the chick. Pitx2, a bicoid-related homeobox gene, plays a critical role in L-R determination and resides downstream of the Shh/Nodal signaling pathway Three isoforms of Pitx2 have been identified in mouse and human. Only two Pitx2 isoforms, Pitx2a and Pitx2c, were isolated from the chick embryo. These two isoforms only differ in the region 5' to the homeodomain. RT-PCR and northern blot analyses verified the existence of Pitx2a and Pitx2c but not Pitx2b in the developing chick embryo. Pitx2c but not Pitx2a is expressed in the left lateral plate mesoderm (LPM). Both isoforms equally encode a transcription activator. Ectopic expression of Pitx2c and Pitx2a to right LPM randomize heart looping. Loss-of-function studies indicate that Pitx2c play a predominant role in the regulation of heart looping in the developing chick embryo Using of gain-of-function and loss-of-function approaches, the extracellular matrix molecule flectin is confirmed to be the downstream effector of Pitx2c. It has been shown that BMP is involved in specification of L-R development. In this study, the role of the BMP receptors in the heart looping was investigated. I found that only BMPIA but not BMPIB was capable of randomizing heart looping when overexpressed on the left LPM. It was also demonstrated that BMPIA might mediate a Pitx2 independent pathway that also regulates heart looping in the chick

Establishment of the biochemical and molecular nature of cardiac development is essential to understand the relationship between genetic and morphological aspects of heart formation. The need to identify genes that are differentially expressed between cell populations within the embryo and within the embryonic heart is critical to elucidate the complex processes of heart development. With this in mind, our laboratory focused on the identification and study of novel genes expressed and involved in the correct development of the vertebrate heart/hemangioblast precursor cell (HPC) lineages. A differential screening using Affymetrix GeneChip Chick system was performed, leading to the identification of several new genes expressed in the haematopoiesis, angiogenesis or cardiogenesis precursor lineages. One of these novel genes, initially denominated CHEP12 (Chick Heart/Hemangioblast Progenitor 12), encodes for a novel secreted protein that contains a Calcium-binding EGF-like domain. Additionally, it was found that besides the canonical CHEP12 transcript, there are several alternative-spliced form variants, indicating that several isoform proteins may be synthesized in response to different signals. The current study intends to accomplish a detailed characterization of the expression pattern of the canonical CHEP12 gene and one of its alternative-spliced form variant (CHEP12-A). WISH and histological sections allowed us to suggest that CHEP12 may be involved in cardiogenic processes due to its expression in the Secondary Heart Field. Moreover, CHEP12 might also be implicated in the development of the venous system as its expression was also detected in the anterior, posterior and common cardinal veins. In addition, comparison between the expression pattern of the full transcript CHEP12 and its isoform CHEP12-A showed no significant differences.

5 Abstract: Aims: The main objective of this work was to analyze in detail the effects of acute temperature changes on the function of isolated chick embryonic heart in vitro in comparison with natural conditions in ovo. Methods: The effects of temperature change (34 řC, 37 řC and 40 řC - hypo-, normo- and hyperthermia, respectively) on calcium dynamics in four days old isolated chick hearts in vitro were investigated by high-speed calcium optical imaging. For comparison and validation of in vitro measurements, experiments were also performed in ovo using videomicroscopy. Artificial electrical stimulation experiments were performed in vitro and in ovo to uncover conduction limits of different heart segments. Results: We observed almost linear dependence of sinus frequency on temperature in our temperature range. Sinus frequency during hypothermia and hyperthermia in vitro and in ovo changed about 20% in comparison with normothermia. We observed no significant changes in amplitude of calcium transients during temperature change to hypothermia but hyperthermia caused a significant decrease in amplitude of calcium transients (atria 35%, ventricles 38%). IC test socket and socket pogo pin are the major cost of consumption parts in IC testing house. Test yield is the key point to determine the profit for IC testing house. When the processing speed of CPU (Central Processing Unit) and GPU (Graphic Processing Unit) are boosting, heat generation and power dissipation became a serious problem for IC testing house. Most package type of CPU and GPU are packed by Flip-Chip BGA type. High temperature will melt the solder ball and cause test socket pogo pin to damage. The excellent cooling capability of impinging jet had been proofed by many literatures in past. In this article, impinging jet applied to IC test handler contact chuck is investigated. The contact chuck had been redesigned with thermal solution and uses a rectangle hot plate to simulate the thermal status of IC testing. A circular air jet impinged on the rectangle hot plate from the topside of contact chuck. Out flow open area, open area on the wall location and the distance between jet nozzle and hot plate are major parameters of this heat transfer problem. Parameter “Z” is the distance between jet nozzle and hot plate; “D” is the diameter of circular air jet. As shown in the result, ratio of Z/D and the location of out flow open area on the wall is obvious on heat transfer capability for redesigned contact chuck. Taguchi method and analysis of variance (ANOVA) method help to clarify the weighting of influence. The optimum Z/D is 0.5 and the optimum location of out flow open area is at dual side corner. Heat transfer capability can be improved approach to 70% after optimization. Width and height of out flow open area only made about 5% impact on heat transfer capability.

The electrostatic chuck for silicon wafers has been developed for used in serial plasma process. The pattern of the electrostatic chuck’s surface and the cooling gas inside have significant influence to the overall heat transfer between the wafer and the susceptor. In this work, the heat transfer model for the wafer and susceptor of a electrostatic wafer holding system is established to investigate the wafer temperature uniformity. The effects of several parameters including input heat flux, the pattern of the chuck surface and the overhang ratio on the wafer temperature distribution are considered for a typical 8 inch silicon wafer. The numerical tool STAR-CD code was employed to simulate the complete details of the heat transfer problem and the computed results are found in good agreement with available data. The grooves on the chuck surface cause undesirable oscillatory temperature distribution along the radial direction. The large overhang ratio also causes temperature increase at the wafer edge. Through the various parameters considered, it is noted that the wafer temperature uniformity can be improved by minimizing the number of grooves and its widths and by matching the size of the chuck closely to that of wafer. That present numerical heat transfer model can be used to optimize the geometry of the electrostatic chuck to provide a uniform distribution of temperature across the surface of the wafer.

The complete heat transfer path on the AlN and Al2O3 electrostatic chuck (ESC), which were utilized under the various operational conditions, is studied for the potential improvement on the temperature uniformity of the 12-inch wafer. In addition, an identical study on the expanded chuck (299mm) is also carried out for a comparison of the original chuck (293mm). An equivalent thermal circuit analogical to an electrical circuit was illustrated and formulated in terms of variables observed to offer a simple calculation toward a potential optimization. In addition, a good agreement with previous work was achieved and examined the reliability in this model system. The content of this study is divided in two parts: electrostatic force and wafer temperature. First, in order to optimize the functionality of the attractive force and in avoiding excessive “trial and error” chuck designs, a set of simulations were obtained under various conditions pertaining to the position of the electrode, material, finish and voltage, individually. Second, the ability of backside cooling plays a critical role in the need to control the wafer temperature and its uniformity. It demonstrates that helium exhibited the best performance among He, Ne and Ar, which shows a controllable function with a linear dependence on the pressure and insensitive to the environmental variation during the process. The histogram with a standard deviation (SD), as an indicator of the temperature uniformity, are used to illustrate a fraction of discrete values of the wafer temperature. It discovered the characteristics of the AlN chuck (293mm) exhibited an excellent ability for the wafer cooling on the edge, but unfavorable to the temperature uniformity which Al2O3 chuck (293mm) is capable without temperature oscillations while the backside pressure increases. In addition, it suggested that the characteristics of the different chucks (AlN and Al2O3 ) become more comparable with the increase of the level of the backside pressure. The AlN chuck (299mm) with a linearly dependent on SD and superior in the mean, is regarded as the best one among other chucks, one of which is the Al2O3 chuck (299mm) which SD becomes independent on the pressure and high mean.

Several embryonic tissues were utilized in the study of creatine analogues. Creatine analogues were taken up and phosphorylated by incubated chick embryo muscle, skin, and heart, in addition to the previously studied chick embryo brain. Accumulated phosphagen was utilized during insult by ischemia or by the addition of various inhibitory compounds to media containing chick embryonic tissues. Resynthesis of the phosphagen, upon cessation of insult and return of the tissues to the incubation media, usually occurred. The enzyme arginine:glycine amidinotransferase was partially purified for the first time from liver. Through the use of several chromatographic techniques a 160-fold purification of the enzyme was achieved from chick liver. Hearts of chicks fed 1-carboxymethyl-2-iminoimidazolidine (cyclocreatine) accumulated 15 (mu)mol/g wet weight of cyclocreatine-P and had glycogen levels elevated three-fold relative to controls. During total ischemia in vitro, cyclocreatine-P was utilized by heart for resynthesis of ATP, glycolysis was greatly prolonged and depletion of both ATP and total adenylate pools was delayed two to five-fold relative to controls. Feeding of a second analogue, 1-carboxyethyl-2-iminoimidazolidine (homocyclocreatine) resulted in the accumulation of homocyclocreatine-P in the heart and a tripling of the glycogen levels. During ischemia, homocyclocreatine-P was only slightly utilized, glycolysis was prolonged, and most importantly, no delay in depletion of ATP levels was observed relative to controls. Feeding of creatine resulted in a doubling of creatine levels in the heart but no delay of ATP depletion during ischemia relative to controls. Maximal conservation of ATP and total adenylates in the heart during ischemia required, in addition to adequate glycogen reserves, a phosphagen which is both kinetically and thermodynamically competent to buffer ATP/ADP during the later stages of ischemia; that is, cyclocreatine-P. Breast muscle of chicks fed cyclocreatine demonstrated a decreased rate of depletion of glycogen upon exposure to the (beta)-adrenergic-agonist, isoproterenol, relative to controls. In addition, during ischemia following exposure to isoproterenol, breast muscle from cyclocreatine-fed chicks was protected against depletion of ATP, with ATP levels remaining 6-fold elevated after 2 hours of ischemia relative to controls. A correlation was demonstrated between total cyclocreatine accumulated and sparing of ATP during ischemia.

When IC has produced, it has to pass thee quality test. In the process of testing, the high processing speed would bring high temperature. The thermal problem will case not only the solder ball of IC package be melted but also the test socket and socket pogo pin those high cost parts be damaged. So, for IC testing department, how to increase the performance of heat transfer, is an important topic in IC testing process.In this article, impinging jet and some geometric characteristics of testing chuck of IC handler are chosen to be the design parameters. The CFD software is used to create the thermal simulation model for analysis the flow field. Taguchi method is applied to conduct the best combination of parameter values for efficiency of heat transfer. Finally, a robust design of IC testing contact chuck is found and results show that the heat transfer capability of new design model can be improved about 50%.
