Streszczenie:
The present study compares the performance of bubbling-bed updraft and a fixed-bed downdraft gasification systems for producing hydrogen-rich (H2) syngas from olive pomace on a semi-industrial scale. The focus is on examining the effects of temperature and efficiency ratio (ER) on the composition, low heat value (LHV), carbon conversion efficiency (CCE), and cold gas efficiency (CGE) of the produced syngas. The results presented for the fixed bed show the concentration of H2 (15.6–16.52%), CGE (58.99–66.80%), CCE (69.07–71.86%), and LHV (4.82–5.70 MJ/Nm3). The CGE reaches a maximum of 66.80% at a temperature of 700 °C and an ER of 0.20, while the syngas yield (2.35 Nm3/kg) presents a maximum at a temperature 800 °C and an ER of 0.21, with a tendency to decrease with the increase in the temperature. For the bubbling fluidized bed, results were shown for the concentration of H2 (12.54–12.97%), CGE (70.48–89.51%), CCE (75.83–78.49%), and LHV (6.10–6.93 MJ/Nm3), where, at a temperature of 700 °C and an ER of 0.23, the CGE is 89.51% and the LHV is 6.93 MJ/Nm3, with a tendency to decrease with the increase in the temperature, while the maximum syngas yield (2.52 Nm3/kg) occurs at a temperature of 800 °C and an ER of 0.23. Comparing the two gasification processes, the fixed bed has a higher concentration of H2 at all the temperatures and ERs of the experiments; however, the bubbling fluidized bed has a higher CGE. These findings have implications for applications involving syngas, such as energy production and chemical synthesis, and can guide process optimization and enhance energy efficiency. The information obtained can also contribute to emission mitigation strategies and improvements in syngas-based synthesis reactors.

To investigate the CGE on hair growth and to explore the mechanism that is involved in the acceleration of anagen induction, we investigated the effects of CGE studied on cell proliferation and molecular mechanism in human hair dermal papilla cells (hDPCs) and keratinocytes (HaCaT cells). Additionally, hair growth evaluation was carried out following topical treatment of the dorsal skin of telogen C57BL/6 mice with CGE for 14 days. As result, CGE increased cell viability and ALP activity in hDPCs. Moreover, CGE increased the expression of catenin beta 1 (CTNNB1), ALP, sex-determining region Y-box 2 (SOX2), insulin-like growth factor 1 (IGF1), and vascular endothelial growth factor A (VEGFA) genes in hDPCs. CGE increased the expression of proteins such as ALP, β-catenin, and phosphorylation of glycogen synthase kinase 3β (pGSK3β), and protein kinase B (pAKT) in hDPCs. Furthermore, CGE induced the proliferation of HaCaT cells and up-regulated AKT-ERK-GSKβ-β-catenin signaling in HaCaT cells. Additionally, the anagen induction effects of CGE were confirmed on the telogen-anagen transition mice model. these findings demonstrated that CGE promoted the entering the growth phase of hair follicle via activation of β-catenin signaling pathways in vivo. Thus, this study suggests that CGE might be a potential therapeutic reagent for hair growth.

Sperm–egg fusion induces cortical granules exocytosis (CGE), a process that ensures the block to polyspermy. CGE can be induced independently by either a rise in intracellular calcium concentration or protein kinase C (PKC) activation. We have previously shown that myristoylated alanine-rich C kinase substrate (MARCKS) cross-links filamentous actin (F-actin) and regulates its reorganization. This activity is reduced either by PKC-induced MARCKS phosphorylation (PKC pathway) or by its direct binding to calmodulin (CaM; CaM pathway), both inducing MARCKS translocation, F-actin reorganization, and CGE. Currently, we examine the involvement of Ca2+/CaM-dependent protein kinase II (CaMKII) and MARCKS in promoting CGE and show that PKC pathway can compensate for lack of Ca2+/CaM pathway. Microinjecting eggs with either overexpressed protein or complementary RNA of constitutively active αCaMKII triggered resumption of second meiotic division, but induced CGE of an insignificant magnitude compared with CGE induced by wt αCaMKII. Microinjecting eggs with mutant-unphosphorylatable MARCKS reduced the intensity of 12-O-tetradecanoylphorbol 13-acetate or ionomycin-induced CGE by 50%, indicating that phosphorylation of MARCKS by novel and/or conventional PKCs (n/cPKCs) is a pivotal event associated with CGE. Moreover, we were able to demonstrate cPKCs involvement in ionomycin-induced MARCKS translocation and CGE. These results led us to propose that MARCKS, rather than CaMKII, as a key mediator of CGE.

Calotropis gigantea (L.) R. Br (Apocynaceae) (commonly known as milkweed or crown flower) is a large shrub native to temperate regions of Asia, including China, Bangladesh and India and has a long history of use in traditional medicines. In this study, we investigated the neuromodulatory effects of the ethanol extracts of C. gigantea leaves (CGE) during synaptogenesis in the late stage of neuronal development and during early stage neuritogenesis in cultured rat hippocampal neurons. Maximum neuritogenic activity was achieved at a CGE concentration of 7.5[Formula: see text][Formula: see text]g/ml. At this concentration, CGE facilitated the early development of cytoarchitecture, as evidenced by increases in morphometric parameters, such as, the numbers, lengths, and number of branches of initial neurites, axon and dendrites. During the synaptogenic stage (DIV 14), immunocytochemistry (ICC) showed that CGE upregulated synaptic vesicle 2 (SV2, a marker of axon terminals) and postsynaptic density-95 (PSD-95, a postsynaptic marker) and their colocalization. CGE upregulated nerve growth factor (NGF) and activated extracellular signal-regulated kinase 1/2 (Erk1/2), which is blocked by a TrkA-specific inhibitor suggesting the neuritogenic and synaptogenic potential of CGE was due to the activation of NGF-TrkA-Erk1/2 signaling. Moreover, UPLC of CGE did not detect stigmasterol, an active component of C. gigantea. However, the chloroform-methanol and ethyl acetate subfractions of CGE exhibited initial neuritogenic activity, suggesting that multiple active components were responsible for the neurotrophic-mimetic properties of CGE. Our data prove the neuromodulatory ability of CGE and provide a means of identifying new active phytochemicals with potential nootropic, preventative or therapeutic effects on the human brain.

This paper develops a general equilibrium model for the Republic of Croatia to evaluate CGE model use in macroeconomic management and forecasting. Since Croatia is a small open country subject to large external shock and growth constraints efficient macroeconomic management framework is fundamental. The lack of data prevents to follow historic economic variables over a longer period of time, which is why CGE models look as possible solution. CGE models use calibration to solve the problem of missing most macroeconomic variables. After developing the model, we use it to run different scenarios for economic policies using a CGE model for Croatia. Results show that CGE model can be an important instrument for policy makers in running macroeconomic policies for small open countries.

Cladophora glomerataextract (CGE) has been shown to exhibit antigastric ulcer, anti-inflammatory, analgesic, hypotensive, and antioxidant activities. The present study investigated antidiabetic and renoprotective effects of CGE in type 2 diabetes mellitus (T2DM) rats. The rats were induced by high-fat diet and streptozotocin and supplemented daily with 1 g/kg BW of CGE for 12 weeks. The renal transport function was assessed by the uptake ofpara-aminohippurate mediated organic anion transporters 1 (Oat1) and 3 (Oat3), using renal cortical slices. These two transporters were known to be upregulated by insulin and PKCζwhile they were downregulated by PKCαactivation. Compared to T2DM, CGE supplemented rats had significantly improved hyperglycaemia, hypertriglyceridemia, insulin resistance, and renal morphology. The baseline uptake ofpara-aminohippurate was not different among experimental groups and was correlated with Oat1 and 3 mRNA expressions. Nevertheless, while insulin-stimulated Oat1 and 3 functions in renal slices were blunted in T2DM rats, they were improved by CGE supplementation. The mechanism of CGE-restored insulin-stimulated Oat1 and 3 functions was clearly shown to be associated with upregulated PKCζand downregulated PKCαexpressions and activations. These findings indicate that CGE has antidiabetic effect and suggest it may prevent diabetic nephropathy through PKCs in a T2DM rat model.

Economic impacts of forestry developments in Alberta are estimated using two interindustry approaches. The results suggest that estimates derived from input-output (I-O) models differ from those of computable general equilibrium (CGE) models. Employment and GDP estimates derived from CGE models are much smaller than those of I-O models. Unlike I-O estimates, estimates derived from CGE models are not unidirectional because of general equilibrium effects. The results also indicate that CGE models provide greater flexibility and have more potential for forest policy analysis when compared with I-O models, but they should be used with caution.

Cortical granule exocytosis (CGE), following egg activation, is a secretory process that blocks polyspermy and enables successful embryonic development. CGE can be triggered independently by either a rise in intracellular calcium concentration ([Ca2+]i) or activation of protein kinase C (PKC). The present study investigates the signal transduction pathways leading to CGE through activation of PKC or stimulation of a rise in [Ca2+]i. Using Western blot analysis, co-immunoprecipitation and immunohistochemistry, combined with various inhibitors or activators, we investigated the link between myristoylated alanin-rich C kinase substrate (MARCKS) translocation and CGE. We were able to demonstrate translocation of MARCKS from the plasma membrane to the cortex, in fertilized as well as in parthenogenetically activated eggs. MARCKS phosphorylation was demonstrated upon PKC activation, whereas a PKC inhibitor (myrPKCψ) prevented both MARCKS translocation and CGE in 12-O-tetradecanoyl phorbol-13-acetate (TPA)-activated eggs. We have further shown that upon egg activation the amount of phosphorylated MARCKS (p-MARCKS) and the amount of calmodulin bound to MARCKS were increased. MARCKS translocation in ionomycin activated eggs was also inhibited by the calmodulin inhibitor N-(6-aminohexyl)-5-chloro-1-napthalenesulfonamide hydrochloride (W7). These results complement other studies showing MARCKS requirement for exocytosis and imply that upon fertilization, MARCKS translocation is followed by CGE. These findings present a significant contribution to our understanding of CGE in mammalian eggs in particular, as well as cellular exocytosis in general.

Purpose: This study aimed to examine the effects of hair dyeing employing used-up coffee grounds extract (CGE).Methods: The total polyphenol concentration of CGE was analyzed using the Folin─Ciocalteu’s method, and a shampoo formulation was prepared by adding different CGE concentrations. The hair dyeing effectiveness was observed according to changes in these two variables: dyeing concentration and dyeing time. To investigate the dyeing effects, L* (Lightness), a* (Red─Green), and b* (Yellow─Blue) values were measured using a colorimeter. The optical density value was measured to examine the hair’s fading degree after bleaching. Results: The total polyphenol concentration in CGE was 122─133 mg GAE/g. The samples showed significant dyeing effects before and after application in terms of L*, a*, and b* values. Hair dyeing ability increased with both amplified CGE concentration and dyeing time. Regarding the bleaching process, no decrease in hair colour intensity or change in hair’s fading degree was observed for 21 days. Conclusion: The possibility of dyeing using CGE has been demonstrated, and further commercialization as a shampoo formulation is considered possible.

Abstract OBJECTIVE To explore the histological, electrophysiological, radiological, and behavioral effects of radiosurgery using a new model of proton beam radiosurgery (PBR) of the rodent hippocampus. METHODS Forty-one rats underwent PBR of the right hippocampus with nominal doses of 5 to 130 cobalt Gray equivalents (CGE). Three control animals were untreated. Three months after PBR, 41 animals were evaluated with the Morris water maze, 23 with T2-weighted magnetic resonance imaging, and 22 with intrahippocampal microelectrode recordings. Animals that were studied physiologically were killed, and their brains were examined with Nissl staining and immunocytochemical staining for glutamic acid decarboxylase, heat shock protein 72 (HSP-72), parvalbumin, calmodulin, calretinin, calbindin, and somatostatin. RESULTS Ninety and 130 CGE resulted in decreased performance in the Morris water maze, increased signal on T2-weighted magnetic resonance imaging, diminished granule cell field potentials, and tissue necrosis, which was restricted to the irradiated side. These doses also resulted in ipsilateral up-regulation of calbindin and HSP-72. Parvalbumin was down-regulated at 130 CGE. The 30 and 60 CGE animals displayed a marked increase in HSP-72 staining on the irradiated side but no demonstrable cell loss. No asymmetries were noted in somatostatin, calretinin, and glutamic acid decarboxylase staining. Normal physiology was found in rats receiving up to 60 CGE. CONCLUSION This study expands our understanding of the effects of radiosurgery on the mammalian brain. Three months after PBR, the irradiated rat hippocampus demonstrates necrosis at 90 CGE, but not at 60 CGE, with associated abnormalities in magnetic resonance imaging, physiology, and memory testing. HSP-72 was up-regulated at nonnecrotic doses.

Lipid metabolism dysregulation can lead to dyslipidemia and obesity, which are major causes of cardiovascular disease and associated mortality worldwide. The purpose of the study was to obtain and characterize six plant extracts (ACE—Allii cepae extractum; RSE—Rosmarini extractum; CHE—Cichorii extractum; CE—Cynarae extractum; AGE—Apii graveolentis extractum; CGE—Crataegi extractum) as promising adjuvant therapies for the prevention and treatment of dyslipidemia and its related metabolic diseases. Phytochemical screening revealed that RSE was the richest extract in total polyphenols (39.62 ± 13.16 g tannic acid/100 g dry extract) and phenolcarboxylic acids (22.05 ± 1.31 g chlorogenic acid/100 g dry extract). Moreover, the spectrophotometric chemical profile highlighted a significant concentration of flavones for CGE (5.32 ± 0.26 g rutoside/100 g dry extract), in contrast to the other extracts. UHPLC-MS quantification detected considerable amounts of phenolic constituents, especially chlorogenic acid in CGE (187.435 ± 1.96 mg/g extract) and rosmarinic acid in RSE (317.100 ± 2.70 mg/g extract). Rosemary and hawthorn extracts showed significantly stronger free radical scavenging activity compared to the other plant extracts (p < 0.05). Pearson correlation analysis and the heatmap correlation matrix indicated significant correlations between phytochemical contents and in vitro antioxidant activities. Computational studies were performed to investigate the potential anti-obesity mechanism of the studied extracts using target prediction, homology modeling, molecular docking, and molecular dynamics approaches. Our study revealed that rosmarinic acid (RA) and chlorogenic acid (CGA) can form stable complexes with the active site of carbonic anhydrase 5A by either interacting with the zinc-bound catalytic water molecule or by directly binding Zn2+. Further studies are warranted to experimentally validate the predicted CA5A inhibitory activities of RA and CGA and to investigate the hypolipidemic and antioxidant activities of the proposed plant extracts in animal models of dyslipidemia and obesity.

Mammalian sperm–egg fusion results in cortical granule exocytosis (CGE) and resumption of meiosis. Studies of various exocytotic cells suggest that filamentous actin (F-actin) blocks exocytosis by excluding secretory vesicles from the plasma membrane. However, the exact function of these microfilaments, in mammalian egg CGE, is still elusive. In the present study we investigated the role of actin in the process of CGE, and the possible interaction between actin and protein kinase C (PKC), by using coimmunoprecipitation, immunohistochemistry and confocal microscopy. We identified an interaction between actin and the PKC alpha isoenzyme in non-activated metaphase II (MII) eggs and in eggs activated by phorbol ester 12-O-tetradecanoyl phorbol-13-acetate (TPA). F-actin was evenly distributed throughout the egg’s cytosol with a marked concentration at the cortex and at the plasma membrane. A decrease in the fluorescence signal of F-actin, which represents its depolymerization/reorganization, was detected upon fertilization and upon parthenogenetic activation. Exposing the eggs to drugs that cause either polymerization or depolymerization of actin (jasplakinolide (JAS) and cytochalasin D (CD) respectively) did not induce or prevent CGE. However, CD, but not JAS, followed by a low dose of TPA doubled the percentage of eggs undergoing complete CGE, as compared with TPA alone. We further demonstrated that myristoylated alanin-rich C kinase substrate (MARCKS), a protein known to cross-link F-actin in other cell types, is expressed in rat eggs and is colocalized with actin. In view of our results, we suggest that the cytoskeletal cortex is not a mere physical barrier that blocks CGE, but rather a dynamic network that can be maneuvered towards allowing CGE by activated actin-associated proteins and/or by activated PKC.

Streszczenie:
Few studies have directly compared passive scattering (PS) to intensity-modulated proton therapy (IMPT) in the delivery of ultra-hypofractionated proton beams to the localized prostate cancer (PCa). In this preliminary study involving five patients previously treated with CyberKnife, treatment plans were created for PS and IMPT (36.25 CGE in five fractions with two opposing fields) to compare the dosimetric parameters to the planning target volume (PTV) and organs-at-risk (OAR: rectum, bladder, femoral heads). Both plans met the acceptance criteria. Significant differences were observed in the minimum and maximum doses to the PTV. The mean dose to the PTV was lower for PS (35.62 ± 0.26 vs. 37.18 ± 0.14; p = 0.002). Target coverage (D98%) was better for IMPT (96.79% vs. 99.10%; p = 0.004). IMPT resulted in significantly lower mean doses to the rectum (16.75 CGE vs. 6.88 CGE; p = 0.004) and bladder (17.69 CGE vs. 5.98 CGE p = 0.002). High dose to the rectum (V36.25 CGE) were lower with PS, but not significantly opposite to high dose to the bladder. No significant differences were observed in mean conformity index values, with a non-significant trend towards higher mean homogeneity index values for PS. Non-significant differences in the gamma index for both fields were observed. These findings suggest that both PS and IMPT ultra-hypofractionated proton therapy for PCa are highly precise, offering good target coverage and sparing of normal tissues and OARs.

Climate change impacts on forests are largely expected to intensify over the next few decades. Computable general equilibrium (CGE) modeling is increasingly becoming a popular tool for assessing these impacts. Previous analyses in this area have been based on either single-region or multiregional CGE model specifications, each with their own advantages and disadvantages. To date, however, there has been no systematic comparison of the potential differences in economic impact estimates between the two CGE model specifications. To examine the extent of these potential differences, we conducted a comparative economic impact analysis of climate change in the forest sector across Canadian provinces, the United States, and the rest of the world using dynamic, single-region and multiregional CGE models over the 2006–2051 period. Results revealed that, within each region, different model specifications produced unique economic impact estimates, differing by as much as 18% under each climate change scenario considered. Overall, a majority of Canadian regions recorded smaller (in absolute value terms) and more positive economic impacts using single-region models compared with the multiregional model. Differences in international trade specifications between models, together with unique climate change impact considerations across regions, played key roles in the findings. While few general conclusions emerge from this analysis, it is clear that CGE model specifications can have a significant effect on regional economic impact estimates of climate change in the forest sector. Thus, caution is advised when using the estimates of any one CGE model for policy purposes.

AbstractThe problem of how to evaluate investments in airports has now been studied for over 50 years. This paper analyzes the use of different methods like cost–benefit analysis (CBA), economic impact analysis (EIA), and computable general equilibrium (CGE) models to address the question. It assesses the strength and weaknesses of each method, and it discusses which methods have been used in different countries. The paper argues that the CBA approach and the newer CGE modeling approach address the policy issue well and that both methods are appropriate, although improvements are possible, especially in the newer aspects of evaluation. Furthermore, more data intensive CGE models are able to analyze broader aspects of the evaluation question for which CBA has had difficulty. EIA does not address the problem satisfactorily, and it misleads air transport policy. But this evaluation contrasts sharply with practice. EIA has been extensively used to decide on airport investment. CGE approaches are very promising, though further work is needed for them to reach their full potential. This paper pays particular attention to the relationship between CBA and CGE in airport investment evaluation and also the possible role of wider economic benefits (WEBs) of aviation in evaluation.

PurposeThe purpose of this paper is to construct an econometric model of the determinants of private investment with a particular focus on the impact of democracy on investment.Design/methodology/approachThe first step was to econometrically derive the long‐run elasticities; then to modify the Fiji computable general equilibrium (CGE) model to incorporate the investment function. Also the econometrically derived long run elasticities in the CGE model were used.FindingsIt was found that democracy has a positive and statistically significant impact on private investment in Fiji. The paper's simulation of Fiji becoming a fully democratic country on investment and other macroeconomic fundamentals, based on a CGE model, reveals that real gross domestic product and real national welfare increase by around 0.01 and 0.05 per cent, respectively; government savings and revenue performance improves; there is a trade balance surplus; and both private consumption and disposable income increase by around 0.05 and 0.12 per cent, respectively.Originality/valueThis is the first study that uses a CGE model to examine the impact of democracy, via investment, on other macroeconomic fundaments. No other study is known to have modelled democracy in a CGE framework.

This comprehensive analysis delves into the landscape of Computable General Equilibrium (CGE) models, examining their historical development, key components, applications, strengths, limitations, and future directions. CGE models have become indispensable tools for understanding the intricate dynamics of modern economic systems, offering a holistic perspective that considers the complex interactions among sectors, households, and governments. The study reveals the strengths of CGE models, including their capacity to provide a comprehensive view of the economy, analyse policy interactions, and incorporate behavioural responses. However, challenges such as data limitations and simplifying assumptions pose inherent limitations. The analysis identifies promising future directions, including the integration of machine learning, leveraging big data, and advancing computational power. By addressing current challenges and embracing these innovations, CGE models can continue to play a pivotal role in economic analysis and policymaking, navigating the complexities of a dynamic and evolving economic landscape.

The purpose of this study was to determine if senior management support (SMSI) in Yemeni commercial banks mediates the association between internal audit function (IAF) quality characteristics and improved corporate governance effectiveness (CGE). Internal auditors, heads of internal audit, chairmen and participants in audit committees, CEOs, and financial management of Yemeni commercial banks were given a list of questions to answer. 158 full lists were obtained to evaluate after distributing the survey. For data analysis and hypothesis testing in this work, Smart PLS 3 was used. The study findings demonstrate a substantial relationship between CGE and IAF competence and due professional care (CPCI), IAF independence and objectivity (INOI), and IAF professional ethics (PEI). The outcomes of the study also demonstrate that there is no relationship between CGE and chief audit executive (CAE) Leadership Style (CLS). In terms of the moderate variable’s influence, the findings revealed that SMSI positively changes the link between CLS, CPCI, and corporate governance effectiveness. SMSI, on the other hand, has no influence on the link between INOI, PEI, and the efficacy of corporate governance. The findings add to the knowledge on IAF factors affecting the efficacy of CG and the role of SMSI in changing this relationship in developing countries such as Yemen. Background: We aimed to investigate an automated semi-quantitative software as an imaging biomarker for the prediction of tissue response (TR) after completion of neoadjuvant chemotherapy (NAC). Methods: Breast magnetic resonance imaging (MRI) (1.5T, protocol according to international recommendations) of 67 patients with biopsy-proven invasive breast cancer were examined before and after NAC. After completion of NAC, histopathologic assessments of TR were classified according to the Chevallier grading system (CG1/4: full/non-responder; CG2/C3: partial responder). A commercially available fully automatic software (CADstream) extracted MRI parameters of tumor extension (tumor diameter/volume: TD/TV). Pre- versus post-NAC values were compared (ΔTV and ΔTD). Additionally, the software performed volumetric analyses of vascularization (VAV) after NAC. Accuracy of MRI parameters to predict TR were identified (cross-tabs, ROC, AUC, Kruskal-Wallis). Results: There were 37 (34.3%) CG1, 7 (6.5%) CG2, 53 (49.1%) CG3, and 11 (10.2%) CG4 lesions. The software reached area under the curve levels of 79.5% (CG1/complete response: ΔTD), 68.6% (CG2, CG3/partial response: VAV), and 88.8% to predict TR (CG4/non-response: ΔTV). Conclusion: Semi-quantitative automated analysis of breast MRI data enabled the prediction of tissue response to NAC.

Abstract Citizen-gathered evidence (CGE) gathered by individuals organized in collectives have the potential to demonstrate environmental and social wrongdoings in court. We identify (collective) agency and resistance in how individuals and communities that have been exposed to socio-environmental stressors turn to gather CGE. We explore the modes through which people gather scientific data, produce CGE, alert authorities to environmental harm, and the methods by which data can be shared with communities, beginning with the case studies of civic environmental monitoring addressing coal ash pollution in North Carolina. We analyze the case studies through a theoretical lens built on (1) the theory of civic monitoring “as resistance;” (2) the argument that CGE can embody “collective intelligence;” and (3) the concept of “crowd science” through a decentralized yet coordinated network. In the first part of the article, we introduce the case studies. Then, we illustrate our theoretical frame as applied to the analysis of the cases. We discuss the cases and in particular the CGE at issue through the prism of the frame. We demonstrate how CGE can be valuable not only for civil society and civic organizations, but also for competent authorities. In our conclusion, we identify critical junctures of our case studies, such as that of “knowledge crowdsourcing” to demand justice that triggers the government to take action and the existence of a “coordinated” yet decentralized crowd to capture the full extension of the problem. Lastly, we outline the limitations of our study, a future research agenda, and still open questions.

Abstract A detection method for genetically modified foods is an essential regulatory requirement for many countries. The present study is aimed at developing a qualitative method for detection of genetically modified organisms by combining PCR methodology with capillary gel electrophoresis (PCR-CGE) in a sequencing platform to detect Bacillus thuringiensis (Bt)-cotton (MON 531)and Roundup Ready (RR) soybean (GTS 40-3-2). A sensitive duplex PCR-CGE method was developed in which target DNA sequences (35S and Nos) were separated both by size and color to detect 0.01% Cry1Ac DNA (w/w) in Bt-cotton. A multiplex PCR-CGE method was developed to simultaneously detect fourtargets such as Sad1, Cry1Ac, 35S, and Nos in Bt-cotton. Four novel PCR primers were designed to customize amplicon size for multiplexing for better visualizationof multiple peaks. The LOD for Cry1Ac DNA specific PCR was 0.01% for Bt-cotton. The LOD for multiplex PCR assay was 0.05% for Bt-cotton. A singleplex PCR-CGE method was developed to detect Lec, 35S and Nos in a trace sample of RR soybean grainpowder (0.1%, w/w). This study demonstrates aPCR-CGE-based method for the qualitative detection of35S, Nos and Cry1Ac targets associated with genetically modifiedproducts.

Minced beef is a popular meat product due to its low price and superior nutritional value. The contamination of minced beef is a significant risk for the worldwide meat market. Both natural and synthetic preservatives are used to expand the shelf life and improve the quality properties of meat. The harmful effects of synthetic preservatives make natural preservatives more appealing. Therefore, this research was performed to study the impact of different concentrations of Cassia glauca leaf extract (CGE) on increasing the shelf life of minced beef. Seventy-two minced beef samples were divided into control, 0.25, 0.5, and 1% w/w CGE treated groups. The control and treated samples were kept at 3 ± 1 °C in the refrigerator for 15 days. Minced beef samples’ sensory, chemical, and microbiological properties were assessed every three days. The gained results showed that the CGE addition effectively decreased the microbial count and maintained the minced beef’s sensory and chemical quality. Additionally, CGE extended the shelf life of minced meat up to 15 days under the proper refrigeration condition compared to the control group, which decomposed after the sixth day of refrigeration. Our study suggested that CGE could be used as a natural preservative for refrigerated minced meat.

The aim of this study was to investigate the effect of Canavalia gladiata extract (CGE) on the regulation of AMP-activated protein kinase (AMPK) in 3T3-L1 preadipocytes and evaluate the adipogenesis and lipogenesis mechanisms. In 3T3-L1 preadipocytes, lipid accumulation and differentiation were suppressed by 1.1, 1.3, and 1.4 times under the CGE treatment at 0.25, 0.5, and 1.0 mg/mL, respectively. The expression of the main genes involved in the inhibition of adipogenesis was evaluated at the mRNA level via a transcription-polymerase chain reaction. The extract at 1.0 mg/mL increased the mRNA expressions of AMPK and carnitine palmitoyl transferase-1 (CPT-1) by 1.9 and 1.2 times, respectively, while it decreased the expression of sterol regulatory element binding proteins-1c (SREBP-1c), peroxisome proliferator activated receptor-γ (PPAR-γ), CCAAT enhancer binding protein-α (C/EBP-α), and fatty acid synthase (FAS) by 1.1, 1.2, 1.8, and 1.5 times, respectively, indicating inhibition of the adipogenesis and lipogenesis potential of CGE. Gallic acid (4.02 mg/g) was identified as the main component of the CGE via LC-MS/MS and HPLC analysis. The results of this study suggested that CGE can be utilized as an anti-obesity food additive or medication by activating the AMPK-induced regulation and suppressing adipogenesis transcription factors.

With the increase in global carbon dioxide emissions, China has put forward the goals of a carbon peak and carbon neutrality (double carbon) and formulated an action plan to consolidate and enhance the carbon sink capacity of the ecosystem. The Chinese grassland ecosystem (CGE) is widely distributed and is the key link for China to achieve the double carbon objectives. However, there is a relative lack of research on carbon sources and sinks in the CGE, so it is urgent to integrate and analyze the carbon sources and sinks in the grassland ecosystem on the national scale. Based on the refined grid data, the net ecosystem productivity (NEP) of the CGE was estimated by coupling the vegetation production model and soil respiration model. The results showed that the cumulative carbon sequestration of the CGE was 14.46 PgC from 2010 to 2020. In terms of spatial distribution, this shows that the differentiation characteristics are high in the northwest of China and low in the southeast of China, which strongly corresponds with the 400 mm isohyet and 0 °C isotherm of China. The results of the correlation analysis showed that the NEP of the CGE was positively correlated with precipitation and negatively correlated with temperature; that is, precipitation mainly promotes the accumulation of NEP, and temperature mainly inhibits it. The coupling effect of temperature and precipitation jointly affects the spatial change of carbon sources and sinks of the CGE. This study can provide a scientific basis for government departments to formulate targeted policies to deal with climate change, which is of great significance for China to improve ecosystem management, ensure ecological security and promote the realization of China’s double carbon goal.

In the concept of CGE management, the environment of the animal is used to control reproduction rather than technological tools. Nutrition is the primary factor but, rather than feeding ruminants with potential human food, we need to focus on forages with occasional ‘smart supplements’. This focus also opens up opportunities – new forages can supply energy and protein whilst improving animal health and welfare, and reducing carbon emissions.Nutritional inputs must be accurately coordinated with reproductive events to ensure that the metabolic signals are appropriate to the stage of the reproductive process. To control the timing of reproduction, we begin with simply managing the presence of the male but then seek more precision through the greater use of ultrasound. Finally, genetic improvement should be part of every industry strategy and it is critical in the long-term development of CGE management. Most aspects of CGE management have a strong genetic component, as evidenced by variation among genotypes, and among individuals within genotypes. For example, a combination of nutritional management with genetic improvement in the rate of muscle accumulation can accelerate sexual maturity, potentially leading to simultaneous improvements in meat production, reproductive efficiency and environmental footprint. For each local situation, we need to introduce the various elements of the CGE package in stages, adapting the process to cover variations in genotype and in geographical and socio-economic environments. Some concepts might need further research and development for local conditions. Ultimately, CGE management is a simple and cost-effective way to improve productivity whilst safeguarding the future of the livestock industries.

The current tourism industry has the problems of low service efficiency, poor coordinated development, and slow economic growth in terms of service volume and economic growth. This paper is based on computable general equilibrium (CGE) model. Firstly, a CGE data analysis model based on bee colony intensive breakthrough algorithm is established to store and analyze the data in the whole chain of tourism. Then, combined with the comparative analysis of tourism economic data over the years, it is fed back to the CGE model for error analysis. Finally, relevant experiments are designed to analyze the relationship between tourism and economic growth. The correlation degree of local economic growth is analyzed. The results show that, compared with the traditional research method of tourism economic growth based on module data analysis, this CGE model can realize the correlation analysis of the data involved in the process of tourism economic growth and analyze the factors affecting the speed of economic growth, which has the advantages of good reliability and strong pertinence.

Streszczenie:
Aim/purpose – This study sought to assess the impact of an increased historical fixed VAT rate of 14% to the current rate of 15% on the South African economy. Design/methodology/approach – The method applied in this study was based on a Dynamic Computable General Equilibrium (CGE) model to evaluate the impact of both the VAT rate of 14% and a new rate of 15% on the South African economy. The CGE model has been proven over the years to be a suitable model when evaluating the impact assessment of any shock within an economy. Enhancements were made by the researcher to the direct and indirect tax section of the model, i.e., the direct tax section was disaggregated, such that for both firm and household revenues, a dividend income stream is separated from other income streams. The main reason is to facilitate a detailed analysis of Corporate Income Tax (CIT) and Personal Income Tax (PIT), as well as the latest implemented Dividend Tax (DT). Findings – When VAT was increased from 14% to 15%, the immediate reaction of the shock from the Dynamic CGE model indicates that the Gross Domestic Product (GDP) declined by 0.0002% in 2018, but increased by 0.0028% in the following year (2019). The trend continued until 2021, hence the 1% increase in the VAT tax rate will increase the expected forecast of VAT collection by approximately R3.2 billion on average. Research implications/limitations – The findings of this study will be implemented by the South African government, which will use a dynamic CGE model to assess South Africa’s VAT contribution to the economy. The database of the CGE model was limited to the Social Accounting Matrix (SAM) for 2015. Originality/value/contribution – The study recommends the use of this method for assessing the impact of tax policy changes to the South African economy. The CGE model seems to be the best model as far as the impact assessment of a shock in the econ- omy is concerned. This will assist the South African authorities with their decision mak- ing regarding future VAT revenue. Keywords: South African Revenue Service (SARS), Value Added tax (VAT), Dynamic computable general equilibrium (CGE) model. JEL Classification: H21, C68, E62.

Background: Microsatellite instability (MSI) testing is important for the classification of Lynch syndrome, as a prognostic marker and as a guide for adjuvant chemotherapy in colorectal cancer (CRC). The gold standard for determining MSI status has traditionally been fluorescent multiplex polymerase chain reaction (PCR) and capillary gel electrophoresis (CGE). However, its use in the clinical setting has diminished and has been replaced by immunohistochemical (IHC) detection of loss of mismatch repair protein expression due to practicability and cost. The aim of this study was to develop a simple, cost-effective and accurate MSI assay based on CGE. Method: After amplification of microsatellites by polymerase chain reaction (PCR) using the National Cancer Institute (NCI) panel (BAT 25, BAT26, D5S346, D2S123, D17S250) of MSI markers, parallel CGE was utilized to classify colorectal cancers as MSI-H, MSI-L and MSS using the 5200 Fragment Analyzer System. Cell lines and patient cancer specimens were tested. DNA from 56 formalin-fixed paraffin-embedded cancer specimens and matched normal tissue were extracted and CGE was performed. An automated computational algorithm for MSI status determination was also developed. Results: Using the fragment analyser, MSI status was found to be 100% concordant with the known MSI status of cell lines and was 86% and 87% concordant with immunohistochemistry (IHC) from patient cancer specimens using traditional assessment and our MSI scoring system, respectively, for MSI determination. The misclassification rate was mainly attributed to IHC, with only one (1.8%) sampling error attributed to CGE testing. CGE was also able to distinguish MSI-L from MSI-H and MSS, which is not possible with IHC. An MSI score based on total allelic variability that can accurately determine MSI status was also successfully developed. A significant reduction in cost compared with traditional fluorescent multiplex PCR and CGE was achieved with this technique. Conclusions: A simple, cost-effective and reliable method of determining MSI status and an MSI scoring system based on an automatic computational algorithm to determine MSI status, as well as degree of allelic instability in colorectal cancer, has been developed using the 5200 Fragment Analyzer System.

Abstract Objectives Ketogenic foods are anti-inflammatory in part via the NLRP3 inflammasome. Companion dogs, susceptible to chronic gastroenteritis (CGE), manifest infiltration of leukocytes (WBC) into bowel tissues. We evaluated circulating WBC in dogs eating a ketogenic food in two studies: 1) healthy dogs; 2) a chronic gastroenteritis (CGE) case/control cohort. Hypotheses- Ketogenic food will reduce WBC and this benefit will extend to dogs with CGE. Methods Dogs housed in pairs, with daily group exercise in outdoor grassy runs. CGE dogs diagnosed by histopathology; criteria included WBC infiltration into intestinal tissue. Healthy controls matched by age, gender, weight and breed. Blood collections after fasting 16 hours. Macronutrients (P/F/C; % energy): Study #1 (St1, healthy) CON1 (21/32/48); KETO1 (26/68/5); Study #2 (St2, chronic colitis & healthy) CON2 (24/19/57); KETO2 (28/68/4). KETO2 differed from KETO1 by protein ingredient replacement. St1: dogs ate CON1 and then consumed KETO1 (each for 5 weeks). St2 a randomized crossover trial (5 weeks feeding each food). Food analyticals measured by AOAC methods. Clinical measures, pancreatic lipase (cPL) by enzyme assay. Statistical testing by dependent t-test. Significance at α = 0.05. Results Ketogenic ratios of foods: CON1 (0.46), KETO1 (1.63), CON2 (0.33), KETO2 (1.69). Relative to CON foods, KETO foods decreased circulating monocytes, neutrophils and total WBC; KETO1 also reduced lymphocytes. KETO foods decreased serum globulin, increased albumin and total protein was unchanged. KETO foods decreased alkaline phosphatase (ALP). KETO foods increased red blood cells, platelets and blood hemoglobin concentration; KETO2 also increased hematocrit and decreased immature reticulocytes. In St2, cPL was not different in healthy or CGE dogs on KETO2 vs CON2 foods. The CGE dog responded in the same manner to the KETO foods as did healthy dogs. Conclusions Ketogenic foods altered circulating inflammatory cell status in healthy dogs and those with enteritis in a manner consistent with reduced inflammatory potential. KETO foods were broadly supportive of health in CGE dogs; the KETO foods increased markers of erythropoiesis and did not increase pancreatic lipase levels. Ketogenic foods may benefit companion animals with inflammatory conditions. Funding Sources Hill's Pet Nutrition, Inc; Topeka, KS 66617, USA

Purpose – The purpose of this paper is to demonstrate the value of computable general equilibrium (CGE) modeling for impact analysis of real estate developments and redevelopments. Design/methodology/approach – Uses a model constructed for Colorado to compare estimates of economic impact of a hypothetical mixed-use development from a CGE model with an input-output (IO) model similar to those commonly used in applied economic impact analysis. Findings – Economic impact estimates of construction activity are demonstrated to be lower when using a CGE approach as compared to an IO approach while impact estimates of continuing operations of a property are demonstrated to be more accurate and potentially higher using a CGE approach. Practical implications – A CGE approach as opposed to an IO approach will be particularly useful for practitioners in particular cases where IO models are ill suited to provide meaningful estimates concerning impact of continuing operations. This is especially likely where commercial tenants are unknown or when the development includes a residential component. Social implications – More complete and accurate assessments of economic impact may positively affect views on property development and redevelopment by the public and government. Originality/value – This paper adds to the existing literature concerning economic impact analysis of real estate and is the first paper in the field, to the authors’ knowledge; to compare estimates from the standard IO approach to those derived using more sophisticated modeling techniques.

In clinical practice, many diseases can lead to changes in serum albumin concentration (HSA) in patients. Accurate detection of HSA concentration is of great significance for disease diagnosis. Based on this, this study designed and synthesized 9-(4-amino-aniline)-acridine (AAA) as a fluorescent probe. By laser induction and capillary gel electrophoresis (CGE), a new rapid and highly specific HSA detection method based on fluorescence-CGE was established. Various experimental control factors were investigated, and the optimal experimental conditions were determined as follows: the running buffer was H3PO4–KH2PO4 (pH = 2.45, 15.0 mmol L−1), the separation voltage was 30 kV, and the experimental temperature was 25 °C. The sample solution injected 10 s with hydrodynamic mode (3.43×103 Pa), HSA could be directly determined by fluorescence-CGE method. The linear range was 0.10–1.0 μg L−1, the detection limit was 0.012 μg L−1, the relative standard deviation (RSD) was less than 0.30%. This method can be used for the determination of real HSA samples. In addition, in the presence of various biological macromolecules, small molecules, ions and ethanol, the accurate detection of HSA by fluorescence-CGE method will not be affected, suggesting that this method has a high specificity for HSA. The rapid and highly specific fluorescence-CGE method of HSA constructed in this study provides a new way to detect HSA, which is of great significance for the diagnosis of clinical diseases.

ABSTRACT Using limited proteolytic analyses, we show that gE present in soluble herpes simplex virus type 1 gE-gI complexes is cleaved into a C-terminal (CgE) and an N-terminal (NgE) domain. The domain boundary is in the vicinity of residue 188 of mature gE. NgE, but not CgE, forms a stable complex with soluble gI.
