Cette étude s'insère dans le cadre du projet FAERIE qui a pour but la réalisation d'une fonction d'ergonomie physiologique dans un système C. F. A. O. Cette fonction vise à simuler le comportement de l'Homme dans son environnement. L'un des principaux aspects du comportement est le mouvement. La présente étude analyse les différents problèmes liés au mouvement et propose une approche originale. L'accent est mis sur la réalisation d'un mouvement et particulièrement sur le délicat problème du naturel. Les différents modèles utilisés sont présentés et les conditions de leur généralisation sont étudiés. L'aboutissement de cette approche est une synthèse du mouvement naturel. La génération de mouvement constitue la base de l'outil d'évaluation ergonomique proposé. Il est alors possible d'évaluer les dépenses musculaires, energétiques,. . . Qui sont nécessaires à l'exécution d'un mouvement. Des notions telles que la fatigue et le confort peuvent être appréhendées. Une attention particulière a été portée sur la facilité d'utilisation de la fonction réalisée. Son emploi ne doit pas requérir de connaissances ergonomiques particulières. L'intégration de cet outil au système C. F. A. O constitue alors un atout majeur pour la prise en compte des contraintes ergonomiques pendant la conception.

Les forces et faiblesses des robots différant grandement de celles des humains, les robots chirurgicaux ont plusieurs avantages qui expliquent leur utilisation croissante en salle d'opération. Cependant, la robotique chirurgicale demeure un domaine qui comporte de nombreux défis. Plutôt que de travailler directement au niveau de l'application sur un robot spécifique, il a été décidé de concentrer les efforts sur une des composantes mécaniques qui impose des limitations aux interactions humain-robot : l'actionneur. Dans cette optique, un prototype de bras interactif a été réalisé en utilisant la technologie de l'Actionneur Différentiel Élastique (ADE), un actionneur élastique haute-performance de faible impédance mécanique intrinsèque. Le bras à trois degrés de liberté, dont les actionneurs sont contrôlés en impédance, pourra servir à étudier la façon dont les interactions humain/robot permettent d'augmenter les capacités humaines de manipulation. Le contrôle d'impédance de l'ADE a été utilisé en tant que noyau central lors du développement du contrôle du bras interactif. L'analyse des performances dans la plage de fréquence d'intérêt suggère un bon comportement de l'actionneur lorsque l'impédance mécanique demandée est faible.Les premiers essais d'interaction humain-robot avec le bras interactif suggèrent que l'ADE est tout indiqué pour une telle application. Le contrôle d'impédance développé a pu être utilisé dans le cadre d'une autre application robotique : le contrôle des axes de direction du robot mobile omnidirectionnel AZIMUT[indice inférieur 3].

Depuis quelques années dans le domaine du cyclisme de route, le confort des vélos est devenu un élément au moins aussi recherché que la performance. Le confort est une préoccupation majeure pour 90 % des cyclistes. Les cyclistes sur route passionnés enfourchent leurs vélos pendant plusieurs centaines d’heures chaque année. Ils souhaitent donc utiliser un vélo qui aura la capacité de réduire les vibrations générées par la route. Un sondage réalisé auprès de 320 cyclistes a montré que les vibrations transmises au niveau de la main sont considérées comme la plus grande source d’inconfort. Ce travail a permis de développer une approche permettant d’estimer le niveau de vibrations transmis au niveau de la main du cycliste par sous-structuration. Les méthodes de sous-structuration permettent de prédire le comportement dynamique d’un assemblage de structures par caractérisation de chacune des structures prise séparément. Bien que ces méthodes existent depuis des dizaines d’années, aucune étude n’avait investigué l’assemblage de deux structures par sous-structuration lorsqu’une d’entre elles est l’Humain. L’approche proposée consiste à mesurer la réponse biodynamique du système main-bras en posture cycliste et à prédire la puissance vibratoire au niveau de la main du cycliste à l’aide d’une méthode de sous-structuration. Cette thèse présente les éléments importants issus de ces travaux. Deux méthodes de couplage ont été identifiées et testées pour prédire l’influence du système main-bras sur le comportement dynamique de structures mécaniques simples. La variabilité de réponse du système main-bras de plusieurs sujets a été analysée. Des caractéristiques communes à plusieurs sujets ont été identifiées dans la réponse biodynamique de leurs systèmes main-bras. Finalement, l’importance relative du cycliste par rapport au vélo sur le comportement dynamique de l’assemblage vélo – cycliste au niveau de la main a conduit au développement d’une méthode de caractérisation de la réponse vibratoire des vélos seuls.

Ce mémoire présente le développement d’un algorithme de cinématique d’interaction dans un contexte de coopération humain-robot. Cet algorithme vise à faciliter le contrôle de bras robotiques en évitant les collisions, singularités et limites articulaires du robot lorsqu’il est contrôlé par un humain. La démarche adoptée pour l’ateinte de l’objectif est le prototypage d’un algorithme sur une structure robotique simple, la validation expérimentale de ce prototype, la généralisation de l’algorithme sur une base robotique à 6 degrés de liberté et la validation de l’algorithme final en le comparant expérimentalement avec un algorithme similaire. Premièrement, le prototype d’algorithme est développé sur un bras robotisé Jaco, de l’entreprise Kinova, duquel le poignet a été retiré. Cette architecture permet le déplacement de l’effecteur selon 3 degrés de liberté en translation. L’algorithme développé sur cette base robotique permet : — l’évitement des collisions avec les objets présents dans l’environnement de travail, — l’évitement des limitations articulaires — et l’évitement des singularités propres à l’architecture du robot. Les performances de l’algorithme sont ensuite validées lors d’expérimentations dans lesquelles il a été démontré que l’algorithme a permis une réduction d’approximativement 50% du temps de complétion d’une tâche donnée tout en réduisant l’attention que l’utilisateur doit porter sur le contrôle du robot comparativement à l’attention portée à l’accomplissement de la tâche demandée. Ensuite, des améliorations sont apportées à l’infrastructure : — une méthode de numérisation de l’environnement de travail est ajoutée, — un meilleur algorithme de détection de collisions et de mesure des distances minimales entre les membrures du robot et les obstacles présents dans l’environnement de travail est implémenté. De plus, une méthode de balayage de l’environnement de travail à l’aide d’une caméra Kinect ainsi qu’un algorithme de segmentation de nuage de points en polygones convexes sont présentés. Des tests effectués avec l’algorithme prototype ont été effectués et ont révélé que bien que des imperfections au niveau de la méthode de balayage existent, ces modifications de l’infrastructure peuvent améliorer la facilité avec laquelle l’algorithme de cinématique d’interaction peut être implémenté. Finalement, l’algorithme implémenté sur une architecture robotique à six degrés de liberté est présenté. Les modifications et les adaptations requises pour effectuer la transition avec la version initiale de l’algorithme sont précisées. Les expérimentations ont validé la performance de l’algorithme vis à vis un autre algorithme de contrôle pour l’évitement de collisions. Elles ont démontré une amélioration de 25% en terme de temps requis pour effectuer une tâche donnée comparé aux temps obtenus avec un algorithme de ressorts-amortisseurs virtuels.

Cette these decrit l'etude de deux regions homologues du chromosome 21 humain normal. Elles ont ete detectees grace a un bacteriophage s'hybridant a trois regions du genome: deux distantes de 12 mb en 21q11. 1 et 21q22. 1 et une dans la region pericentrometrique du chromosome 13. Jusqu'a present, les regions homologues les mieux connues du genome normal correspondaient soit a des repetitions de quelques centaines de bases comme les sequences alu, soit a des genes et des pseudogenes apparus au cours de l'evolution. Des cas de duplications plus importantes ont aussi ete decrits mais, en general ils sont associes a des pathologies. Nous avons montre qu'il existe aussi des regions homologues de plusieurs centaines de kilobases dans le genome humain normal, en particulier sur le chromosome 21. Pour ce travail nous avons construit une banque specifique du chromosome 21 dans le chromosome artificiel de la levure (yac) a partir de lignees d'hybrides somatiques. Cette banque represente 1,5 equivalents du chromosome 21 et a ete reunie a la banque de l'icrf. Nous avons crible ces deux banques avec la sonde d21s190 polymorphe dupliquee, et avons obtenu 11 yacs. Les extremites de quelques yacs ont ete localisees par rapport a un assortiment de lignees d'hybrides somatiques contenant des portions differentes du chromosome 21. Certaines lignees contiennent uniquement le locus proximal, d'autres le locus distal et d'autres les deux loci. Cette approche et la recherche de sondes communes a plusieurs yac nous ont permis d'ordonner les clones les uns par rapport aux autres. Nous en avons aussi deduit que les deux regions homologues sont orientees dans le meme sens sur le chromosome. Au locus distal nous avons construit une carte de restriction avec des enzymes coupant dans les regions riches en cg et avons estime la taille de la region d'homologie entre 135 et 500 kb. Cette region ne contient pas de site noti mais deux ilots cg sont presents, indiquant l'existence possible de genes

Le principe de cartographie comparative a ete suivi pour etudier l'evolution de la region de classe i du complexe majeur d'histocompatibilite (cmh). L'auteur a identifie et localise de nouveaux marqueurs conserves entre l'homme et la souris, qui ont permis une comparaison de la moitie distale du cmh entre les deux especes. Cette carte comparative met en evidence une structure conservee de la region de classe i, malgre l'absence d'orthologie entre les genes de classe i. L'auteur postule que les sequences non apparentees aux sequences de classe i sont representatives d'une structure ancestrale, dans laquelle les sequences de classe i auraient evolue independamment. Parmi les sequences conservees, un regroupement de genes de recepteurs olfactifs a ete identifie. Plusieurs hypotheses sont proposees quant a la fonction et l'evolution de ces genes

Le chromosome 22 humain est un petit chromosome acrocentrique associes a diverses pathologies comprenant des anomalies du developpement embryonnaire (le syndrome de digeorge et le syndrome de l'il de chat) et des proliferations tumorales du systeme nerveux (les tumeurs neuroectodermiques peripheriques, la neurofibromatose de type 2, les meningiomes,. . . ) ou du systeme hematopoietique (la leucemie myeloide chronique). Il y a quatre ans les genes responsables de ces pathologies n'etaient pas connus, a l'exception du gene bcr, implique dans la leucemie myeloide chronique. Le travail presente dans cette these, visait a identifier de nouveaux genes dans les regions juxtacentromerique et centrale du bras long du chromosome 22 humain, associees a ces diverses pathologies. L'approche utilisee a repose sur l'isolement de cosmides a partir d'un hybride somatique d'irradiation selectionne pour son contenu en fragments d'adn humain. Dans chacuns des cosmides analyses, des sequences geniques potentielles ont ete recherchees et utilisees pour cribler des banques d'adn complementaire. Cette recherche a, entre autre, abouti a l'isolement de deux nouveaux adnc qui sont: 1/ un adnc chimere isole a partir d'un fragment conserve entre espece. Ce clone comprend d'une part, un adnc de fonction inconnue, code a partir de la region centrale du bras long du chromosome 22, et d'autre part, un adnc partiel localise sur le chromosome 3, correspondant au gene codant pour la glutaminyl-arnt synthetase. 2/ un adnc isole a partir d'un fragment riche en dinucleotide cpg, localise dans la region critique pour le syndrome de digeorge. Cet adnc code pour une proteine presentant des homologies avec deux inhibiteurs de la transcription des histones chez s. Cerevisiae: hir1 et hir2

Les robots sont de plus en plus amenés à interagir avec des humains ou des environnements anthropiques en vue de collaboration. La connaissance des propriétés visco-élastiques cartésiennes humaines durant des interactions physiques avec des environnements procure un éclairage au champ des sciences du mouvement humain, et aussi à la robotique collaborative pour la conception de commandes innovantes bio-inspirées. Dans cette thèse, la focalisation est placée sur une modélisation linéaire très simple en impédance mécanique du membre supérieur, qui fait entrer en jeu les paramètres cartésiens apparents en raideur, amortissement et masse. Cette modélisation permet d'approcher des comportements en rejet de perturbations qui interviennent notamment lors d'interactions physiques.Une expérience a été mise en œuvre avec un robot articulé piloté en admittance cartésienne, pour permettre des estimations d'impédance mécanique du bras de participants pendant une tâche de référence permettant de générer des mouvements rythmiques, avec des retours haptiques. Une méthode permettant l'estimation des paramètres du modèle en impédance, basée sur l'approximation des trajectoires virtuelles en position et force lors de faibles perturbations ne gênant pas la réalisation de la tâche, est proposée. Les trajectoires virtuelles sont approchées par des interpolations de splines ou des optimisations de sinusoïdes.Une trentaine de participants ont pris part aux expériences proposées pour permettre des estimations significatives de variations des paramètres visco-élastiques apparents et mieux comprendre leurs implications dans la réalisation d'une tâche en interaction avec un robot. Le compromis stabilité-transparence du couplage du robot avec un environnement en impédance a finalement été analysé pour proposer une amélioration des réglages du contrôle en admittance cartésienne Robots are more inclined to interact with humans or their environment for collaborative purposes. Knowledge on the human endpoint vis-coelastic properties during physical interactions provides insights for the field of human movement science and also for the design of innovative bio-inspired collaborative robotic control strategies. In this work, the focus is placed on a simplistic linear mechanical model of the human arm, with endpoint apparent parameters like stiffness, damping and mass. Perturbation rejection behaviours occuring remarkably during physical interactions can be met using this modelling.In order to estimate those properties for the human arm, an experimental test-bed was designed using an endpoint admittance controled polyarticulated robot. A benchmark task was used so that rhythmic movements emerged, while haptic feedback were introduced by the robot. A methodology to identify the linear parameters of the chosen impedance model was designed, tackling the issue of the estimation of virtual trajectories of the arm during dynamic movements. The estimations of the arm's virtual trajectories both in position and force relied on spline interpolations and sine optimisations, for small deviations that did not alter the performances of the task.A cohort of participants took part in experiments proposed to observe significant variations of the viscoelastic apparent parameters, and improve the understanding of the implications of such variations during a physical interaction with a robot. The famous trade-off between stability and transparency while the robot is coupled with an environment was then study thanks to the obtained estimations, to enhance the tuning of the endpoint admittance control empirically designed

Le besoin grandissant de flexibilité en milieu industriel conduit à reconsidérer la manière dont les robots sont utilisés dans de tels environnements. Il s'ensuit que la relation entre l'homme et les machines doit évoluer au profit d'une plus grande proximité, en leur permettant de partager un espace de travail commun et d'interagir physiquement.Dans cette optique, cette thèse a pour objectif de contribuer au contrôle de robots bi-bras à des fins collaboratives dans un contexte industriel. Pour ce faire, nous proposons une approche de contrôle cinématique réactif basée sur une loi de contrôle en admittance. Celle-ci permet une manipulation d'objets sécuritaire en collaboration physique avec des opérateurs humains. Le contrôleur résout un problème d’optimisation quadratique (QP) afin de trouver le déplacement articulaire permettant de satisfaire la commande spécifiée dans l'espace de la tâche, ceci tout en respectant un ensemble de contraintes (e.g. limites articulaires, évitement de collision).La résolution cinématique peut être adaptée afin de générer des solutions parcimonieuses au niveau des vitesses articulaires, ce qui signifie qu'un nombre minimal d'actionneurs est activé pour assurer la réalisation de la tâche. Cela induit un comportement potentiellement plus sûr dans un environnement évolutif partagé avec des individus.Les plateformes bi-bras comprennent parfois des extensions (par exemple, une base mobile, un torse articulé, etc.). Dans cette thèse, nous présentons une méthode hiérarchique originale pour le contrôle de systèmes multi-robots.Une implémentation open source du travail, acrfull {rkcl}, a été développée. Cette librairie rassemble tous les composants décrits dans cette thèse et peut être facilement configuré pour inclure de nouveaux robots. Tout au long des développements, des validations expérimentales ont été effectuées sur le cobot mobile à deux bras BAZAR The growing need for flexibility in industrial settings leads to reconsidering the way robotic systems are exploited in such environments. It follows that the relationship between humans and machines has to evolve in favor of more proximity, by letting them share the same workspace and physically interact together.With this in mind, this thesis aims at contributing beyond the state of art in the control of dual-arm robots for collaborative purposes in an industrial context. We propose a generic online kinematic control approach based on an admittance control law which enables safe manipulation of objects in physical collaboration with humans. The controller solves a acrfull{qp} optimization problem to find the joint space motion that satisfies the task space command while respecting a set of constraints (e.g. joint limits, collision avoidance).The kinematic solver can be tuned to generate parsimonious solutions at the joint velocity level, meaning that as few actuators as possible are activated to achieve the tasks. This induces potentially safer behavior in an unstructured environment shared with humans.Dual-arm platforms are sometimes extended to include additional robots (e.g., mobile base, articulated torso, ...). In this thesis, we also present an original hierarchical method for the control of multi-robot systems.An open-source implementation of the work, acrfull{rkcl}, is available. It implements all the components described in this thesis and can be easily configured to work with new robots. Throughout the developments, experimental validations have been performed on the dual-arm mobile cobot BAZAR

Les mouvements humains sont toujours complexes. Même une tâche simple comme prendre un verre d'eau implique de nombreux degrés de liberté i.e., différents groupes de muscles, plusieurs articulations et un nombre infini de trajectoires possible pour le bras. Néanmoins, les mouvements sont facilement disponibles aux sujets sains et semble être naturellement optimisé par le système nerveux central. Cela est souvent modélisé par la minimisation d'un paramètre donné du système, tel que l'énergie ou l'à-coup, qui semblent être des candidats naturels. Malheureusement, ces approches sont souvent limitées dans leur portée et ne peuvent pas décrire les mouvements périodiques dans des environnements changeant dans le temps. Dans de tels systèmes, les invariants adiabatiques sont des observables pertinentes issues de la mécanique hamiltonienne. L'objectif de cette thèse de doctorat est d'étudier le rôle et l'utilisation des invariants adiabatiques dans le contrôle moteur humain. Pour ce faire, nous avons réalisé une série d'expériences. Tout d'abord, nous les avons étudiées en tant que contrainte pour la stabilité globale de la marche, même lorsqu'elle est exposée à une tâche altérant la variabilité, telle que le maintien d'un rythme dicté par un métronome. Ensuite, nous avons utilisé des résultats récents en physique pour évaluer la variabilité inhérente à la marche à longue distance en tant que phénomène de diffusion de la distribution des invariants adiabatiques. Enfin, nous les avons explorés dans des environnements temporels changeants, notamment en modifiant la "gravité" à la fois dans une centrifugeuse et dans un contexte de vol parabolique, où ils semblent être des quantités pertinentes pour montrer les changements dans les stratégies motrices. Les différents résultats de cette thèse indiquent que les invariants adiabatiques révèlent des contraintes génériques cachées affectant les mouvements humains périodiques Human motion is inherently complex. Even an ordinary task like lifting a glass of water involves many degrees of freedom i.e., different muscle groups, multiple joints and an infinite number of trajectories for the arm. Nevertheless, motion is readily available to healthy subjects, and seems to be naturally optimized by the central nervous system. This is often modelized as the minimization of a given parameter of the system e.g., energy or jerk, which appear as natural candidates. Unfortunately, these approaches are often limited in their scopes, and cannot describe periodic motion in time-changing environments. In such systems, adiabatic invariants are relevant observables originating from Hamiltonian mechanics. The aim of this doctoral dissertation is to investigate the role and use of adiabatic invariants in human motor control. This was done in a series of experiments. First, we studied them as a constraint for the global stability of gait, even when exposed to a variability-altering task, such as metronome keeping. Then, we used recent results in physics to assess the inherent variability of long-range walking as a diffusion phenomenon of the distribution of adiabatic invariants. Finally, we explored them in time-changing environments, specifically by altering “gravity” both in a centrifuge and a parabolic flight context, where they seem to be relevant quantities to show changes in motor strategies. The different findings in this dissertation point to adiabatic invariants revealing generic hidden constraints affecting periodic human motion

Cette etude concerne la region pseudo-autosomique humaine, situee a l'extremite des bras courts des chromosomes x et y, region ou se produit un crossing-over unique et obligatoire en meiose male. Nous avons contribue a la cartographie physique de cette region par l'alignement de fragments d'adn clones dans des chromosomes artificiels de levure (yacs). Dans ce but, nous avons tout d'abord crible la banque de yacs du centre d'etude du polymorphisme humain, par hybridation avec un element subtelomerique repete (stir) dont la concentration dans cette region est particulierement importante. Cette premiere approche n'a pas permis de realiser un alignement complet. A partir des yacs identifies, nous avons donc isole des sequences tagged-sites (stss) qui ont servi a identifier de nouveaux yacs afin de completer l'alignement. Toute la region pseudo-autosomique, a l'exception du telomere, a ete couverte par 31 yacs. Nous avons par cette approche genere 25 nouveaux marqueurs sts pseudo-autosomiques, parmi lesquels 4 detectent un polymorphisme de restriction. Nous avons ensuite montre que le gene ant3, qui code pour l'adenine nucleotide translocase, est localise dans la region pseudoautosomique. Ce gene echappe a l'inactivation du chromosome x. Il possede dans sa region promotrice un ilot cpg qui est hypomethyle sur le chromosome x inactif. Sur la carte de yacs et sur la carte de restriction etablie par electrophorese en champ pulse, nous avons determine la position de trois autres genes pseudoautosomiques (il3ra, xe7 et hiomt), ainsi ordonnes du telomere au centromere: csf2ra-il3ra-ant3-xe7/hiomt-mic2. Enfin, nous avons tente de localiser les homologues murins des genes ant3 et hiomt humains chez la souris. Nos resultats confirment la forte divergence des regions pseudo-autosomiques murine et humaine

Bien que plusieurs genes soient connus pour leur role dans la carcinogenese colorectale, et que deux entites genetiques distinctes par leurs alterations aient ete identifiees, il n'existe aucune description exhaustive des alterations genetiques de ces tumeurs. Nous avons realise, par des methodes semi-automatiques de typage genetique, des allelotypes haute-densite (275 locus) sur presque 200 cancers coliques. Cette etude des anomalies quantitatives a l'aide de sondes microsatellites, a guide notre recherche sur une region du genome frequemment perdue dans l'adn somatique, et donc susceptible de porter un gene suppresseur de tumeur : le bras court du chromosome 8. Sur ce segment chromosomique, une etude systematique des pertes alleliques, avec une resolution de l'ordre de 1 centimorgan (soit environ 1 megabase), a ete entreprise dans le but de determiner une region critique de deletion suffisamment petite pour permettre l'identification des genes contenus dans cette region. Le typage dense de presque 200 tumeurs colorectales supplementaires a defini une region critique de deletion s'etendant sur moins de 2 cm, compatible avec certaines regions minimales decrites dans la litterature a propos des cancers du colon et d'autres types tumoraux. De nombreuses etudes de perte d'heterozygotie de ce bras chromosomique ayant ete effectuees dans divers types tumoraux, nous avons elargi l'etude de ces deletions somatique aux cancers bronchiques non a petites cellules et aux hepatocarcinomes. Une region commune de deletion distincte de celle des cancers colorectaux a ete identifiee dans les cancers pulmonaires. La mise en evidence de ces deux regions de taille reduite devrait conduire rapidement a la mise en uvre de techniques d'isolement de genes.

Este projeto de pesquisa de mestrado buscou identificar os impactos das propriedades da superfície urbana sobre as condições meteorológicas da Região Metropolitana de São Paulo (RMSP), buscando um melhor entendimento dos mesmos para que atividades de planejamento urbano possam ser melhor organizadas. O trabalho utilizou como principais ferramentas produtos de sensoriamento remoto, para definir algumas propriedades da superfície urbana, medidas em superfície de variáveis meteorológicas, para determinação de índices de conforto humano, e a modelagem numérica da atmosfera, para identificação de condições atmosféricas e construção de cenários úteis ao planejamento urbano. Com o uso de imagens de satélite, foram criados arquivos de ocupação do solo e da fração vegetada na área urbana da RMSP em altíssima resolução (20 m). Após a definição das propriedades da superfície, valores medidos pela CETESB de temperatura e umidade relativa foram associados a padrões de ocupação urbana. Verificou-se que as regiões de São Caetano do Sul e do Parque D. Pedro II foram as que apresentaram o maior número de casos de desconforto por calor moderado. Regiões como São Miguel Paulista e Ibirapuera mostraram-se mais confortáveis. Na maior parte dos casos, as regiões apresentaram um nível agradável a levemente quente, seguidos pela condição agradável a levemente fresca. Após a análise diagnóstica, foram construídos cenários contemplando atividades de planejamento urbano em busca da melhoria das condições de conforto térmico humano. Os experimentos realizados indicam que os fluxos turbulentos em superfície são consideravelmente modificados e que a temperatura pode sofrer uma diminuição gradativa até cerca de 1,5 ºC com a substituição de 50% da superfície urbana por vegetação, chegando a mais de 2,2 ºC quando esta substituição é completa. O perfil de temperatura potencial na Camada Limite Planetária também foi consideravelmente modificado com o aumento da porção vegetada no domímio modelado. This master\'s research project has the goal of indentifying the impacts of urban surface properties over the meteorological conditions of the Metropolitan Area of São Paulo (MASP), in order to obtain a better understanding of them and to provide means of better organized urban planning activities. The work was made by using as principal tools, remote sensing derived products, in order to define some surface properties, surface measurements of atmospheric variables, in order to obtain human thermal comfort indexes, and numerical modeling of the atmosphere, in order to identify atmospheric conditions and simulate useful scenaries for urban planning. By using sattelite images, land use and vegetated fraction files were created over the MASP with very high resolution (20 m). Afterwards, temperature and relative humidity measured by CETESB were associated to urban land use patterns. It was verified that regions like São Caetano do Sul and Parque D. Pedro II are more likely to have greater discomfort sensation by moderate heat. Regions like São Miguel Paulista and Ibirapuera were identified to be more confortable. In the majority of cases, the regions presented a pleasant to lightly warm, followed by pleasant to lightly fresh conditions. After the definition of the surface properties, scenarios were built comprehending urban planning activities, looking for the improvement of human thermal comfort conditions. The experiments showed that the surface turbulent fluxes are considerably changed and that temperature was gradualy reduced in up to 1,5 ºC with the replacement of 50% of the urban surface by vegetation, reaching more than 2,2 ºC when this replacement was complete. The potential temperature profile inside the Planetary Boundary Layer was also considerably modified following the increase of vegetated area in the modeled domain.

Le cancer du rein représente 2% des cancers de l'adulte et actuellement le seul traitement efficace est l'acte chirurgical. Une meilleure prise en charge thérapeutique de ces pathologies passe par une meilleure caractérisation des tumeurs, aboutissant à un diagnostic plus précoce et à l'identification de nouveaux marqueurs, témoins de la progression tumorale et qui ont probablement une valeur pronostique. La mise en évidence de nouvelles régions génétiques et de gènes impliqués dans les cancers du rein devrait ouvrir la voie à des traitements plus spécifiques et mieux ciblés. Le carcinome rénal à cellules claires, la forme la plus agressive des cancers du rein épithéliaux présente dans la majorité des cas une délétion du bras court du chromosome 3 ou délétion 3p. Dans les tumeurs rénales que nous avons étudiées, par cytogénétique conventionnelle, hybridation génomique comparative (CGH) et hybridation in situ fluorescente ou FISH, des délétions 3p mais aussi des trisomies 5q, des délétions 8p, des monosomies des chromosomes 9 et 14 ont été les altérations les plus fréquemment rencontrées. Notre étude sur la région 3p a mis en évidence dans tous les cas une délétion longue et terminale. Par FISH en utilisant des sondes BACs, 16 gènes candidats suppresseurs de tumeur de cette région ont été testés et aucun ne présente un profil de délétion différent des autres. Nous avons également réalisé l'étude de l'expression par immuno-histochimie de deux gènes d'intérêt dans la région 3p, les gènes VHL et FHIT. Alors que l'expression de la protéine VHL est diminuée dans différents types de tumeurs rénales, celle de la protéine Fhit ne l'est que dans les carcinomes à cellules claires. Ainsi l'altération du gène FHIT, situé dans la région 3p, et plus particulièrement la perte d'expression de la protéine Fhit pourraient plus être spécifiques du carcinome rénal à cellules claires que le gène VHL.

Melanoma is the deadliest type of skin cancer with an increasing incidence for past two decades. Once melanoma is metastasized (cancer cells are spread out through the body) there is no effective treatment available and 84% of the patients die within 5 years. However, the discovery of braf mutation in melanoma increased the hope for developing new treatments. We first evaluated the effect of braf V600E mutation on melanoma patient survival. In a systematic review we revealed that patients with braf V600E mutation have almost two times more risk of death compared with patients with wild type braf. Next we evaluated the correlation of brafV600E mutation with protein expression. We found that compared with nevi samples, BRAF protein expression was remarkably increased in primary melanomas and further increased in metastatic melanoma patients. Higher BRAF protein expression was significantly correlated with other poor prognosis factors and lead to a significant worse five-year survival. However, we did not find a significant correlation between BRAF protein expression and braf V600E mutation. In our attempt to investigate the cause of induced BRAF protein, we found novel expression of BRAF splice variants (BRAFsv) in both melanoma patients and cell lines. We identified new kinase-dead BRAFsv, which have a dominant negative effect on full-length BRAF and are able to suppress downstream signaling and reduce melanoma cell proliferation. These variants were highly expressed in primary melanoma compared to normal samples, while the expression was decreased in metastatic and more aggressive types of melanoma. In addition, kinase-dead BRAFsv showed a protective effect on patient survival, which remained significant at the presence of full-length BRAF. Thus, patients who expressed the kinase-dead variant and had lower levels of full-length BRAF expression showed the best survival rate in 5 years. Our invitro analysis also indicates that over expression of kinase-dead BRAFsv in melanoma cell lines enhances the effect of BRAF inhibitor treatment. All in all, the data presented in this thesis elucidated a new era in the evaluation of melanoma patient prognosis and revealed new possibilities for more effective melanoma treatments. The ability of wheat bran and oat hulls to bind copper and zinc using a new chromatographic technique was investigated and compared to a centrifugation method. Also investigated was the bioavailability of copper which had been exogenously bound to wheat bran then fed to mice and rats. Wheat bran bound 6 mg Cu/g fiber when pH was raised to 7. Less binding occurred at lower pHs. Seven mg of zinc bound to wheat bran at pH 5 while only a trace bound at pH less than 2. Oat hulls bound 3 mg Cu/g fiber at pH 5, and less than 1 mg of zinc bound per g of oat hull at pH 6. Binding for both fibers depended on the level of mineral added to the fiber slurry during incubation. But only wheat bran binding capacity was influenced by pH. The two methods used did not compare favorably, in all cases, to the amount of mineral bound to fiber. Copper when bound to wheat bran, was utilized in both species, with differences occurring between species was noted. Rats fed the copper bound diet compared favorably with rats fed the copper-adequate diet as determined by body weight, weight gain, heart weight, liver copper concentration and heart copper concentration. Mice, on the other hand showed similar liver and heart concentrations of copper for mice fed either the copper-bound diet or the copper-adequate diets. The in vitro results showed that binding of copper or zinc to wheat bran occurred at a pH similar to the intestinal pH and wheat bran binds more copper and zinc than oat hulls. Oat hulls may prove to be a better dietary fiber source for those people who need to increase their dietary fiber, because oat hulls do not appear to bind copper or zinc and would not likely impair their absorption. Although wheat bran had a high binding capacity for copper, this binding did not significantly inhibit copper absorption as determined in animal studies suggesting that fiber-mineral binding (at least for copper bound to wheat bran) does not cause mineral deficiency symptoms as claimed by many researchers.

Chez l'humain, la perte de fonctions motrices causée par l'absence d'une partie du bras affecte l'autonomie et la capacité à réaliser des tâches du quotidien. Pour rétablir certaines des fonctions perdues, la personne handicapée peut utiliser une prothèse qui remplace la partie absente du bras. Aujourd'hui, les prothèses les plus avancées mesurent l'activité des muscles du moignon pour commander leurs articulations. Cependant, plus le handicap est important, plus nombreuses sont les fonctions motrices à restaurer mais moins nombreux sont les muscles à partir desquels recueillir ces mesures. En vue de surmonter cet obstacle, cette thèse explore comment l'emploi de coordinations motrices, c'est-à-dire de régularités dans les rotations des différentes articulations, peut contribuer au pilotage d'une prothèse de bras. À cette fin, deux plateformes expérimentales intervenant comme substituts à une véritable prothèse sont élaborées~: un bras robotique anthropomorphe à taille humaine, et un bras simulé dans un dispositif de réalité virtuelle. Une première expérience met des participants valides aux commandes de ce bras robotique, piloté de façon à ce que son extrémité reproduise les déplacements de la main du participant. Dans une tâche d'atteinte de cible, elle compare la qualité du pilotage selon que le robot adopte des postures plutôt bio-mimétiques ou biologiquement invraisemblables, pour atteindre avec son extrémité le but défini par le participant. Cette expérience montre que la familiarisation au pilotage du robot est meilleure lorsque ses coordinations articulaires sont proches de celles d'un bras humain. Dans une seconde expérience, des participants valides pilotent un bras virtuel dont l'épaule imite les mouvements de leur propre bras, tandis que ses articulations distales (coude et au-delà) sont commandées artificiellement. Dans une tâche de prise et pose d'objet, elle compare la qualité du pilotage selon que ces articulations distales sont commandées uniquement à partir des rotations de l'épaule réelle, ou en intégrant également des informations contextuelles relatives à la cible à atteindre. Cette expérience révèle que l'inclusion d'informations contextuelles améliore notablement la qualité du pilotage. Dans leur ensemble, ces résultats montrent que les coordinations motrices naturelles sont une source d'informations pertinentes pour le pilotage d'une prothèse de bras et peuvent être employées en combinaison avec d'autres signaux de commande pour enrichir ses capacités motrices. En termes d'applications, ils fournissent des pistes pour la conception de techniques de pilotage exploitant les coordinations motrices naturelles pour piloter plusieurs articulations simultanément In humans, the loss of motor functions associated with the absence of part of the arm disrupts autonomy and reduces the ability to carry out tasks of daily life. To restore some of the lost functions, a person with the aforementioned upper limb disability can use a prosthesis which replaces the missing part of the arm. To this day, the most advanced prostheses measure the activity of muscles located in the stump to control their joints. However, a higher level of disability implies that the prosthesis must restore more motor functions with fewer available muscles from which command signals can be measured. In order to overcome this obstacle, this thesis explores how motor coordinations extit{i.e.} regularities in the way the different joints are put in motion, can be used to drive an arm prosthesis. With this aim, two experimental platforms were developed to act as substitutes for an actual prosthesis: a human-like robotic arm, and a simulated arm in a virtual reality setup. In a first experiment, this robotic arm is driven by able-bodied participants so that its endpoint reproduces the motion of their own hand. Based on a target-reaching task, this experiment compares how well participants perform with this control scheme in two distinct conditions. These conditions correspond to two different strategies to choose the robot's postures when placing its endpoint on the goal defined by the participant: rather human-like or biologically implausible. The results show that employing joint coordinations close to those of a human arm elicits better familiarization to the robot's control scheme. In a second experiment, able-bodied participants drive a virtual arm whose shoulder mimics the participant's actual shoulder motion while its distal joints (elbow and lower) are artificially controlled. Based on a pick-and-place task, this experiment compares how efficiently participants manage to drive the virtual arm with two distinct control schemes. One controls these distal joints' rotations solely from the actual shoulder's motion whereas the other uses additional information in the form of contextual, target-related data. The results reveal that including this contextual information notably improves the performance achieved during the task. Overall, these results show that natural joint coordinations provide a relevant source of information for the control of an arm prosthesis and can be combined with other types of command signals to further expand its motor functions. Regarding application to real-life prosthesis use, they provide insight for the design of control schemes employing natural motor coordinations to drive multiple joints simultaneously

La 1ère partie de ce travail a consisté à évaluer l'implication de la fatigabilité des muscles extenseurs du tronc dans l'apparition des maux de dos. Les résultats suggèrent la présence d'un relais d'activité des muscles thoraciques vers les muscles lombaires. Les maux de dos de la femme enceinte ne sont cependant pas liés à une fatigabilité plus importante des muscles étudiés. La 2nde partie concernait l'exploration des caractéristiques biomécaniques du système avant-bras/main. Lors de la grossesse, la raideur musculo-tendineuse des fléchisseurs du poignet est augmentée. La relation entre la grossesse et les facteurs de risque biomécaniques des troubles musculo-squelettiques de l'avant-bras a été analysée lors d'une activité de travail. Selon les caractéristiques du poste, l'astreinte biomécanique est augmentée chez la femme enceinte. La grossesse est un état physiologique caractérisé par une fragilisation du système musculo-squelettique The 1st part of this work assessed the implied of the fatigability of the back extensors in this back pain. The results suggest a load sharing from the thoracic to the lumbar muscles. Low back pain does not seem to result of an increased fatigability of the lumbar muscles. The 2nd part of the study deals with the analysis of the biomechanical characteristics of the forearm-hand system. During pregnancy, the musculotendinous stiffness of the wrist flexors is increased. The influence of pregnancy on biomechanical risk factors of the upper-limb extremity musculoskeletal disorders was also investigated during a video display terminal task. The biomechanical strain is increased by pregnancy, either by a renforcement of the activation of the wrist and fingers flexors, or by an increase of wrist extension related to the workstation design. These results point out that physicians, physiotherapists and ergonomists should consider these approaches to take care of pregnant women

La thématique de la thèse est très vaste. Sont présentées les grandeurs cinématiques locales ou régionales qui agissent sur le corps humain. La deuxième partie de la thèse décrit les méthodes optiques, par lesquelles on peut mesurer les vibrations produites par les machines-outils sur le système humain main-bras au cours de l'activité au travail. The fourth part of this thesis contains the theorical basis of optical techniques used for vibrations measurement. Chapter five presents the experimental methodology of vibrations measurement in comparition betsween the two methods: classical and optical. The results of measurement are given in chapter six of the thesis. The mechanical properties of the lips are of crucial importance to the function of a brass instrument. The natural resonance modes must be able to usefully interact with the instrument air column in order to sustain oscillations. Mechanical frequency responses of human and arti cial lips used to play a brass instrument were measured using a high-speed digital video technique in an attempt to classify the true nature of the lipreed. The results revealed the presence of at least two lip modes that exhibited the characteristic outward-inward striking behaviour seen in many in vitro replica lip-reed measurements. The Q-values of the human lip resonances were considerably lower than those seen for the replica lips. Transverse mechanical response measurements were also performed on an in vitro lip-reed to investigate the coupling between the outward and inward striking modes. The two dimensional motion of the lips during full oscillations was investigated. It is shown that a computational four degree-of-freedom model would be required to fully simulate the observed mechanical motion. The uid behaviour downstream from an in vitro vocal fold model was investigated using particle image velocimetry (PIV). A `free jet' con guration with no downstream acoustical coupling was rst investigated. The measurements revealed an unsteady glottal jet ow, consisting of a high velocity jet core, a transitional region of high jet deceleration and a turbulent mixing region. The jet was consistently skewed at angles to the glottal centreline, and appeared to oscillate back and forth across the centreline during the glottal cycle. The behaviour of the jet core was investigated in detail. A temporal asymmetry was observed in the mean velocity across the jet core such that the highest jet velocities were encountered during the closing phase of the vocal folds. The overall jet behaviour also showed a strong turbulent asymmetry between the opening and closing phases. High levels of vorticity and turbulent motion encountered during the closing phase were associated with the deceleration of the jet. Three vocal fold con gurations that included static replicas of the ventricular bands were nally investigated with the aim of characterising the aerodynamic interaction between the ventricular bands and the vocal folds. A marked e ect on the glottal jet was observed for all con gurations. The most physically realistic con guration appeared to stabilise the glottal jet, leading to a reattachment of the jet to the ventricular bands and a subsequent secondary ow separation from the downstream end. The implications of the aerodynamic interaction is discussed, with particular note to its possible relevance to the lip-reed and mouthpiece interaction in brass playing.

Les robots sont des agents artificiels qui peuvent agir dans le monde des humains grâce aux capacités de perception. Dans un contexte d’interaction homme-robot, les humains et les robots partagent le même espace de communication. En effet, les robots compagnons sont censés communiquer avec les humains d’une manière naturelle et intuitive: l’une des façons les plus naturelles est basée sur les gestes et les mouvements réactifs du corps. Pour rendre cette interaction la plus conviviale possible, un robot compagnon doit, donc, être doté d’une ou plusieurs capacités lui permettant de percevoir, de reconnaître et de réagir aux gestes humains. Cette thèse a été focalisée sur la conception et le développement d’un système de reconnaissance gestuelle dans un contexte d’interaction homme-robot. Ce système comprend un algorithme de suivi permettant de connaître la position du corps lors des mouvements et un module de niveau supérieur qui reconnaît les gestes effectués par des utilisateurs humains. De nouvelles contributions ont été apportées dans les deux sujets. Tout d’abord, une nouvelle approche est proposée pour le suivi visuel des membres du haut du corps. L’analyse du mouvement du corps humain est difficile, en raison du nombre important de degrés de liberté de l’objet articulé qui modélise la partie supérieure du corps. Pour contourner la complexité de calcul, chaque membre est suivi avec un filtre particulaire à recuit simulé et les différents filtres interagissent grâce à la propagation de croyance. Le corps humain en 3D est ainsi qualifié comme un modèle graphique dans lequel les relations entre les parties du corps sont représentées par des distributions de probabilité conditionnelles. Le problème d’estimation de la pose est donc formulé comme une inférence probabiliste sur un modèle graphique, où les variables aléatoires correspondent aux paramètres des membres individuels (position et orientation) et les messages de propagation de croyance assurent la cohérence entre les membres. Deuxièmement, nous proposons un cadre permettant la détection et la reconnais- sance des gestes emblématiques. La question la plus difficile dans la reconnaissance des gestes est de trouver de bonnes caractéristiques avec un pouvoir discriminant (faire la distinction entre différents gestes) et une bonne robustesse à la variabilité intrinsèque des gestes (le contexte dans lequel les gestes sont exprimés, la morpholo- gie de la personne, le point de vue, etc). Dans ce travail, nous proposons un nouveau modèle de normalisation de la cinématique du bras reflétant à la fois l’activité mus- culaire et l’apparence du bras quand un geste est effectué. Les signaux obtenus sont d’abord segmentés et ensuite analysés par deux techniques d’apprentissage : les chaînes de Markov cachées et les Support Vector Machine. Les deux méthodes sont comparées dans une tâche de reconnaissance de 5 classes de gestes emblématiques. Les deux systèmes présentent de bonnes performances avec une base de données de formation minimaliste quels que soient l’anthropométrie, le sexe, l’âge ou la pose de l’acteur par rapport au système de détection. Le travail présenté ici a été réalisé dans le cadre d’une thèse de doctorat en co-tutelle entre l’Université “Pierre et Marie Curie” (ISIR laboratoire, Paris) et l’Université de Gênes (IIT - Tera département) et a été labelisée par l’Université Franco-Italienne Robots are artificial agents that can act in humans’ world thanks to perception, action and reasoning capacities. In particular, robots companion are designed to share with humans the same physical and communication spaces in performing daily life collaborative tasks and aids. In such a context, interactions between humans and robots are expected to be as natural and as intuitive as possible. One of the most natural ways is based on gestures and reactive body motions. To make this friendly interaction possible, a robot companion has to be endowed with one or more capabilities allowing him to perceive, to recognize and to react to human gestures. This PhD thesis has been focused on the design and the development of a gesture recognition system that can be exploited in a human-robot interaction context. This system includes (1) a limbs-tracking algorithm that determines human body position during movements and (2) a higher-level module that recognizes gestures performed by human users. New contributions were made in both topics. First, a new approach is proposed for visual tracking of upper-body limbs. Analysing human body motion is challenging, due to the important number of degrees of freedom of the articulated object modelling the upper body. To circumvent the computational complexity, each limb is tracked with an Annealed Particle Filter and the different filters interact through Belief Propagation. 3D human body is described as a graphical model in which the relationships between the body parts are represented by conditional probability distributions. Pose estimation problem is thus formulated as a probabilistic inference over a graphical model, where the random variables correspond to the individual limb parameters (position and orientation) and Belief Propagation messages ensure coherence between limbs. Secondly, we propose a framework allowing emblematic gestures detection and recognition. The most challenging issue in gesture recognition is to find good features with a discriminant power (to distinguish between different gestures) and a good robustness to intrinsic gestures variability (the context in which gestures are expressed, the morphology of the person, the point of view, etc. ). In this work, we propose a new arm's kinematics normalization scheme reflecting both the muscular activity and arm's appearance when a gesture is performed. The obtained signals are first segmented and then analysed by two machine learning techniques: Hidden Markov Models and Support Vector Machines. The two methods are compared in a 5 classes emblematic gestures recognition task. Both systems show good performances with a minimalistic training database regardless to performer's anthropometry, gender, age or pose with regard to the sensing system. Actuellement, la musicologie de l’improvisation s’appuie pour l’essentiel sur une lecture intentionnelle du projet artistique ; notre thèse propose une approche différente, en regardant l’improvisation comme un processus complexe, comportant une dimension auto-organisée, à l’image des comportements collectifs observés dans les sociétés animales. Ceux-ci résultent de composantes aléatoires, de nombreuses interactions, de logiques d’amplification et de processus non linéaires. Les outils et les concepts mis au point à l’interface physique-biologie pour comprendre ces dynamiques naturelles nous permettent de construire une démarche de modélisation propre à la musicologie, qui décrit les actes de l’improvisateur à partir d’intuitions musicales pour analyser l’interaction entre ce qui participe de savoir pré-construits et de l’intention, d’une part, et ce qui peut être compris comme un réglage de l’aléatoire, d’autre part. Cette démarche de modélisation est mise en oeuvre sur deux transcriptions du pianiste américain Brad Mehldau. Pour la première (Am Zauberberg), la démarche itérative de modélisation est exposée en détail, partant du modèle le plus pauvre jusqu’à la nécessité d’incorporer le geste de la main. Pour la seconde (Bard), cette démarche est étendue à la conception harmonique, chaque voix d’accompagnement étant conçue comme mue d’un mouvement propre au sein de contraintes d’espace donnée par les autres voix. Nous concluons en ouvrant des perspectives de possibles expérimentations inspirées par ces modèles, du côté de l’enseignement de l’improvisation ou de celui du musicien cherchant à incorporer à son jeu une dimension de lâcher-prise qui est au cœur du processus d’improvisation Currently, the musicology of improvisation essentially highlights the intentional part of an artistic project ; our thesis starts with a quite distinct approach, looking at improvisation as a complex process, with a self-organization dimension inspired of the way biologists analyse collective behaviors in animal societies. These behaviors are todays perceived as the result of combined statistical processes at the individual scale, with numerous inter-individual interactions, amplifications, and non linear loops. Such an analysis of observed natural phenomena led biologists and physicists to introduce and set-up concepts and tools that we use here to propose a modeling approach adapted to Musicology. We start with musical intuitions to propose a description of the perceptions and actions of the improviser that puts forward the deep interaction between what is made of pre-builded knowledge and intention, on one side, and of « tuning random behaviors », on the other. This modeling approach is carried out on two transcriptions of the american pianist Brad Mehldau. With the first piece (Am Zauberberg), we fully describe the iterative process leading from the poorest model to the need of incorporating hand-gestures. With the second piece (Bard), we dress the question of improvising harmony, each voice being conceived as animated of its own displacement rules, but spatially constrained by the others. We conclude by opening up prospects of experiments inspired by these models, some concerning the teaching of improvisation, others aiming at better understanding the process by which a musician seeks to incorporate to his play more « letting go », at the heart of what improvisation is about

La premiere partie de cette these concerne l'etude des differents parametres impliques dans la regulation de la mobilite flagellaire des spermatozoides humains demembranes et reactives. Deux types de spermatozoides ont ete etudies : 1 -des spermatozoides humains normaux, et 2 -des spermatozoides humains mutants, presentant une absence specifique des bras externes de dyneine. Les resultats montrent qu'un equilibre entre la concentration en atpmg, en calcium et le ph est necessaire pour la regulation de la mobilite et du mouvement des spermatozoides humains normaux. De facon originale, il est montre que le ph n'agit pas dans cet equilibre dans le cas de spermatozoides humains sont depourvus de bras externes de dyneine. Ceci est en faveur d'une difference fonctionnelle entre les bras externes et internes de dyneine. Dans une deuxieme partie, la phosphorylation des proteines spermatiques a ete etudiee a partir de spermatozoides normaux demembranes et reactives, en relation avec le mouvement. Trois polypeptides, de poids moleculaires de 48, 40 et 24 kd, sont phosphoryles a ph 7,8 mais non a ph 7,1 a meme concentration d'atpmg. L'hypothese d'une dephosphorylation de ces polypeptides en relation avec l'initiation de la mobilite des spermatozoides est proposee. Enfin, il est montre que les proprietes physico-chimiques de la dyneine extraite des bras externes de spermatozoides d'oursins ne sont pas modifiees en relation avec le type de mouvement.

O melanoma é responsável por menos de 5% dos cânceres de pele, porém, 95% das mortes ocorrem devido a ocorrência de metástases. O melanoma metastático é refratário às terapias convencionais e rapidamente adquire resistência às terapias como as oncogene-dirigidas, como o inibidor de BRAF, da via de MAPK. Estudos prévios de screening in silico do nosso grupo, onde se utilizou as bases de dados TCGA e GEO, identificaram o gene adenosina quinase (ADK) como sendo diferencialmente expresso entre o melanoma invasivo e os nevus. A 5-iodotubercidina (5-ITu) é um potente inibidor farmacológico da ADK que dentre os diversos efeitos relatados na literatura destaca-se pelo potencial genotóxico. Os danos no DNA são os principais ativadores de checkpoint do ciclo celular, que levam a parada do ciclo celular transitória ou permanente, além de induzir morte celular, levando a hipótese de que ADK possa ser potencial agente anti-melanoma. Este trabalho objetivou avaliar a expressão do gene ADK em melanomas humanos e quimiorresistentes ao inibidor de BRAF (iBRAF), avaliou os impactos de 5-ITu sobre a proliferação, progressão do ciclo celular e morte celular e por fim avaliamos sua capacidade de aumentar a sensibilidade das células. Foi realizado PCR em tempo real para avaliar os níveis de expressão de mRNA de ADK em linhagens de melanoma e na cultura primária de melanócitos; a fim de avaliar a citotoxicidade de 5-ITu foram realizados os ensaios de exclusão por azul de tripan e de apoptose - Anexina V e PI e em modelo de esferoide, usando live/dead; também foi avaliada a influência de 5-ITu sobre a capacidade clonogênica e seus efeitos sobre a proliferação celular, a partir dos ensaios de ciclo celular e avaliação de marcadores de proliferação por imunofluorescência; as linhagens foram submetidas a diferentes regimes de tratamento com 5-ITu e o iBRAF, a fim de avaliar a curva de crescimento e a sensibilidade ao iBRAF por MTT níveis de expressão de mRNA de ADK maiores nas linhagens tumorais em relação aos melanócitos. 5-ITu mostrou-se capaz de inibir a proliferação (IC50) das linhagens de melanoma em concentrações de 1,9 a 3,5 µM. 5-ITu não foi capaz de induzir inviabilidade celular, apesar de reduzir a quantidade de células viáveis em todas as condições de tratamento, também não foi capaz de induzir aumento significativo de células apoptóticas, nem mesmo necróticas. No entanto, o tratamento com 5-ITu reduziu a capacidade clonogênica de linhagens de melanoma e promoveu parada de ciclo celular nas fases G1 e G2/M, levou ao aumento da população subG1. O tratamento com 5-ITu promoveu a redução da expressão de marcadores de proliferação, como ki67, e a combinação de tratamentos 5-ITu e iBraf foi capaz de aumentar o tempo de dobramento das linhagens de melanoma, embora tenha se mostrado incapaz de sensibilizar as células de melanoma ao tratamento com iBRAF. Desse modo, pode-se concluir que 5-ITu induz o efeito citostático e se mostra um potente agente antiproliferativo para melanoma parental e resistente. Melanoma accounts for less than 5% of skin cancers, but 95% of deaths occur due to metastases. Metastatic melanoma is refractory to conventional therapies and rapidly acquires resistance to therapies such as oncogene-directed, such as the BRAF inhibitor, of the MAPK pathway. Previous studies of screening in silico of our group, using the databases TCGA and GEO, identified the adenosine kinase gene (ADK) as differentially expressed between invasive melanoma and nevus. 5-iodotubercidin (5-ITu) is a potent pharmacological inhibitor of ADK that among the several effects reported in the literature stands out for the genotoxic potential. DNA damage is the main activator of the cell cycle checkpoint, which leads to transient or permanent cell cycle arrest, in addition to inducing cell death, leading to the hypothesis that ADK may be a potential anti-melanoma agent. This work aimed to evaluate the expression of the ADK gene in human melanomas and chemoresistants to the BRAF inhibitor (iBRAF), evaluated the impacts of 5-ITu on proliferation, cell cycle progression and cell death and finally we evaluated its ability to increase the sensitivity of cells. Real-time PCR was performed to assess the levels of ADK mRNA expression in melanoma lines and primary melanocyte culture; in order to evaluate the cytotoxicity of 5-ITu, the trypan blue and apoptosis - Annexin V and PI exclusion and blue spheroid models were performed using live / dead; the influence of 5-ITu on the clonogenic capacity and its effects on cell proliferation, from the cell cycle assays and the evaluation of proliferation markers by immunofluorescence; the cell lines were submitted to different treatment regimens with 5-ITu and iBRAF in order to evaluate the growth curve and the sensitivity to iBRAF by MTT levels of mRNA expression of ADK higher in the tumor lines in relation to the melanocytes. 5-ITu was able to inhibit the proliferation (IC 50) of melanoma lines at concentrations of 1.9 to 3.5 181;M. 5-ITu was not able to induce cell non-viability, although it reduced the amount of viable cells in all treatment conditions, nor was it able to induce a significant increase in apoptotic or even necrotic cells. However, treatment with 5-ITu reduced the clonogenic capacity of melanoma cells and promoted cell cycle arrest in the G1 and G2 / M phases, leading to an increase in the subG1 population. Treatment with 5-ITu promotes the reduction of expression of proliferation markers, such as ki67, and the combination of 5-ITu and iBRAF treatments was able to increase the doubling time of melanoma cells, although it has been shown to be unable to sensitize melanoma cells to treatment with iBRAF. Thus, it can be concluded that 5-ITu induces the cytostatic effect and shows a potent antiproliferative agent for parental and resistant melanoma.

Human noroviruses (HuNoVs) are the leading cause of viral epidemic acute gastroenteritis and responsible for the deaths of over 200,000 children each year worldwide. HuNoV research has been hampered by the long absence of a readily reproducible cell culture system and a suitable small animal model, while gnotobiotic (Gn) pigs have been a unique animal model for understanding HuNoV pathogenesis and immunity, as well as evaluating vaccine and therapeutics. Recent reports of HuNoVs infection and replication in B cells supplemented with commensal bacteria Enterobacter cloacae and in Blab/c mice deficient in RAG/IL2RG have gained extensive attention, and my studies utilized the well-established Gn pig model to investigate the effects of these two interventions on HuNoV infection. Surprisingly, the colonization of E. cloacae inhibited HuNoV infectivity in Gn pigs, evidenced by the significantly reduced HuNoV shedding in feces and HuNoV titers in intestinal tissues and blood compared to control pigs. Moreover, HuNoV infection of enterocytes but not B cells was observed with or without E. cloacae colonization, indicating B cells were not a target cell type for HuNoV in Gn pigs. On the other hand, using RAG2/IL2RG deficient pigs generated by CRISPR/Cas9 system, with confirmed severe combined immunodeficiency, I evaluated the effects of host immune responses on HuNoV infection. Compared to wild-type Gn pigs, longer HuNoV shedding was observed in RAG2/IL2RG deficient pigs (16 versus 27 days), and higher HuNoV titers were detected in intestinal tissues and contents and in blood, indicating increased and prolonged HuNoV infection in RAG2/IL2RG deficient pigs. In addition, I evaluated dietary interventions including probiotics and rice bran using Gn pig model of HuNoV infection and diarrhea. While the colonization of probiotic bacteria Lactobacillus rhamnosus GG (LGG) and Escherichia coli Nissle 1917 (EcN) in Gn pigs completely inhibited HuNoV fecal shedding, the two cocktail regimens, in which rice bran feeding started either 7 days prior to or 1 day after viral inoculation in the LGG+EcN colonized Gn pigs, exhibited dramatic anti-HuNoV effects, including reduced incidence and shorter duration of diarrhea, as well as shorter duration of virus fecal shedding. The anti-HuNoV effects of the cocktail regimens were associated with the enhanced IFN-𝛾⁺ T cell responses, increased production of intestinal IgA and IgG, and longer villus length. Taken together, my dissertation work improves our understanding of HuNoV infection and immunity, and further supports for Gn pigs as a valuable model for future studies of human enteric virus infection, host immunity, and interventions. Cette thèse traite des réseaux de neurones sous leur forme artificielle et biologique et du parallélisme. L'étude des réseaux de neurones biologiques est abordée par le biais de l'analyse du contrôle moteur lors de mouvements du bras dirigé vers un but. Le mouvement de préhension est étudié plus précisément. Les grandes lignes de la neurophysiologie du mouvement sont rappelées ainsi que les problèmes ouverts concernant leur fonctionnement. Un dispositif expérimental novateur est décrit qui a été mis au point pour l'analyse des mouvements de préhension. Il permet l'enregistrement 3d des composantes du mouvement et la génération de perturbations de l'environnement en fonction de paramètres du mouvement évalués en temps réel. Des modélisations neuronales sont présentées pour répondre au problème de la génération des commandes motrices de mouvements diriges vers un but. L'apprentissage, dans les modèles connexionnistes, est un processus itératif long. Les machines parallèles présentent un potentiel de puissance capable de répondre aux exigences des chercheurs. La connexion machine cm-2 a été utilisée pour l'implémentation de réseaux multicouches à retro propagation. L'algorithme proposé s'appuie sur la connaissance matérielle de la machine et sur l'utilisation d'outils logiciels pour permettre un gain de temps optimal pour la parallelisation par les exemples de la phase de reconnaissance. Une application industrielle est présentée concernant la prédiction de pollution. La modélisation utilisée pour résoudre le problème est celle des réseaux multicouches. L'utilisation de méthodes connexionnistes conduit à de meilleurs résultats que des méthodes statistiques classiques sans requérir de connaissance sur le problème. Nous présentons une implémentation des cartes de Kohonen sur une machine simd (maspar mp-1). Un nouvel algorithme est présenté, utilisant une stratégie par blocs, permettant d'augmenter le facteur d'accélération de l'implémentation parallèle. Cet algorithme se comporte mieux que l'algorithme classique sur un problème particulier en séquentiel. L'étude de la charge de calcul induite par une carte de Kohonen permet de définir un placement optimal des neurones sur les processeurs

Colorectal cancer is the second most common cause of cancer death in Western Europe and North America. Current therapies are largely ineffective and are associated with considerable morbidity. Activating mutations in KRAS and BRAF genes are frequent in colorectal cancer, especially at later stages of the disease, and result in constitutive activity of the MAPK pathway, leading to increased proliferation and tumour survival. The MEK1/2 inhibitor AZD6244, that targets the MAPK pathway downstream of these mutations, has been tested as novel therapy for colorectal cancer. However, clinical trials have been disappointing due to an apparent intrinsic and/or acquired resistance to treatment. Mechanisms underlying this resistance have been studied using cell lines and tumour xenografts. However, the relevance of these data to advanced human colorectal cancer is unclear. One of the difficulties in testing and developing novel therapies for colorectal cancer is the lack of representative models of human disease. Thus, the initial aim of my PhD was to develop a method to culture human colorectal cancers ex vivo in order to use this as a platform for investigating response to AZD6244 and other therapies. These studies indicated that regardless of growth conditions, colonic tumour explants suffered extensive apoptosis in the first 24h in culture, which limited their application in drug response assays. Therefore, as an alternative to long term culture of human colorectal explants, I tested the effects of AZD6244 using acute treatments. Twenty three fresh colonic tumours were obtained from patients and treated for 1h with AZD6244 ex vivo in dose response studies. In all samples, MEK1/2 inhibition occurred within 1h of treatment. In one group of particularly sensitive tumours, the drug also had a distinct phenotypic effect. In these tumours, I found that the agent induced a dose-dependent decrease in proliferation and increase in apoptosis within 1h of treatment. Analysis of markers for this sensitivity indicated it was not clearly dependent of the presence of KRAS or BRAF mutations, which have previously been shown to confer sensitivity. Other markers of sensitivity / resistance were also examined. In addition to studies with AZD6244 alone, I examined the combined effects of this agent and aspirin in colon cancer cells lines and in tumour explants, with promising results. Whilst the use of fresh patient tumour tissue has some technical and logistical challenges, these data suggest that such methodologies are worthy of further investigation as a means to examine determinants of sensitivity and resistance to novel therapies, or their likely activity in combination.

Le syndrome de Lynch (SL) est dû à des mutations dans un gène su système de réparation des mesappariements. Les tumeurs de Lynch présentent une instabilité des microsatellites (MSI). Dans le but d'une identification optimale des patients avec un SL, 2 stratégies sont comparées sur une série de 214 patients non sélectionnés avec un cancer colorectal. 1)Test MSI pour toutes tumeurs puis exclusion des tumeurs sporadiques MSI+. 2)Test MSI pour les patients à haut risque seulement, définis selon des critères cliniques et l'histoire familiale (critères de Bethesda ; modèles de prédiction). Nos résultats indiquent que se limiter à rechercher une mutation germinale chez les seuls patients à haut risque avec une tumeur MSI+ conduit à ne pas identifier environ 25°/° des patients avec un SL. Dans une seconde étude, nous avons recherché les altérations génètiques sur le chromosome 10q sur une série de tumeurs colorectales primitives et de métastases. Nos résultats montrent des pertes de la région de PTEN dans 22°/° des cas avec de très rares mutations de gènes The Lynch syndrome (LS) is due to germline DNA mismatch repair gene mutation (MMR). Tumors from LS patients exhibit microsatellites instability at simple DNA repeat sequences. To establish the most effective and efficient way for the detection of LS patients, we compared two strategies in an unselected series of 214 patients with newly diagnosed colorectal cancer. 1) MSI testing for high-risk patients only, selected by clinical criteria and family history (revised Bethesda guidelines ; predictive models). Our results showed that genotyping only hight-risk patients with a MSI+ tumor failed to identify about 25°/° of patients with LS. In a second study, we investigated the possible role of chromosome 10q losses in colorectal cancer metastasis and our results demonstrate a deletion of PTEN region in 22°/° of cases with a low frequency of mutations of this gene

Artifi cial potential fi elds is a technique that use attractive and repelling forces to control e.g. robots, or non player characters in games. We show how this technique may be used in a controller for Super Mario in a way create a human-like playing style. By combining fi elds of progression, opponent avoidance and rewards, we get a controller that tries to collect the rewards and avoid the opponents at the same time as it is progressing towards the goal of the level. We use human test persons to improve the controller further by letting them make pair-wise comparisons with human play recordings, and use the feed-back to calibrate the bot for human-like play. El presente trabajo de investigación tiene como objetivo principal la realización del diseño conceptual de un sistema mecatrónico que ayudará a los restaurantes de pollos a la brasa, a contar con una propuesta automatizada para la cocción y facilitar empaquetamiento de pollos, buscando realizar el empaquetamiento sin la intervención humana. Para lograr el objetivo principal, se debe determinar los factores de mercado, implementación y tecnología en relación a los hornos de pollos a la brasa. Con el diseño del horno, se busca brindar una solución que de mayor resiliencia a las pollerías frente a factores adversos como a las consecuencias producto del COVID-19. Como principal conclusión del trabajo, se logró diseñar preliminarmente un horno de pollos a la brasa, que luego de alcanzar la cocción deseada, serán extraídos automáticamente para ser llevados a una máquina empaquetadora. Rôle de la NADPH oxydase NOX4 dans la régulation de l'expression du symporteur sodium/iode (NIS) dans le cas du cancer papillaire de la thyroïde (PTC). L’activation autocrine de la voie TGF-β induite par BRAFV600E régule négativement l’expression du symporteur sodium/iode (NIS) via une production de ROS dépendante de la NOX4 dans le cancer papillaire de la thyroïde. Résumé : Le cancer papillaire de la thyroïde (PTC) est la pathologie thyroïdienne la plus répandue. Les mutations ponctuelles de BRAF sont retrouvées dans 40 à 60 % des cas de PTC. La transversion BRAFT1799A est la mutation de BRAF la plus fréquente. Les tumeurs porteuses de la mutation BRAFV600E sont souvent associées avec une diminution significative de l’expression du transporteur sodium/iode (NIS). Les résultats cliniques sur les patients atteints d’un cancer de la thyroïde porteur de la mutation BRAFV600E ont montré que l’inhibition de la voie MAPK ne permet pas de rétablir de manière assez importante l’expression du NIS induite par BRAFV600E. L’expression de BRAFV600E induit la sécrétion de TGF-β fonctionnel, qui inhibe l’expression des protéines thyroïdiennes impliquées dans le métabolisme de l’iode, et particulièrement le NIS. La NOX4 est surexprimée dans un nombre croissant de tumeurs, et particulièrement dans les cas de PTC. Dans le cas du cancer du sein, les mécanismes critiques pour le développement du cancer impliquent la régulation par le TGF-β de la NOX4 au niveau transcriptionnel via le facteur de transcription Smad3. Ces données nous mènent à considérer la NOX4 comme un candidat sérieux pour le rôle de système générateur de ROS contrôlé par la boucle autocrine TGF-β induite par BRAFV600E. Dans cette étude, nous avons tout d’abord observé une corrélation entre la présence de l’oncogène BRAFV600E, la surexpression de la NOX4 et l’inhibition de l’expression du NIS dans les cancers papillaires de la thyroïde. Puis, en utilisant la lignée BCPAP comme modèle in vitro de PTC, nous avons démontré BRAFV600E contrôle l’expression de la NOX4 et de la p22phox par l’intermédiaire de la signalisation TGF-β/Smads. La boucle TGF-β induite par BRAFV600E induit l’expression de la NOX4 et de la p22phox au niveau transcriptionnel via phosphorylated SMAD3. L’expression constitutive de la NOX4 et de la p22phox, qui forment ensemble un complexe NADPH oxydase fonctionnel, contribue au stress oxydatif observé dans les cellules BCPAP. Le traitement des cellules BCPAP par des scavengers de ROS comme le N-acetyl cysteine (NAC) et le Tiron permettent d’augmenter l’expression du NIS au niveau transcriptionnel et de rétablir l’expression d’une protéine fonctionnelle permettant la captation d’iode, ce qui indique que les ROS sont impliqués dans l’inhibition de l’expression du NIS. L’inhibition spécifique de la NOX4 par siRNA permet de réinduire l’expression de l’ARN messager et de la protéine NIS. Ces résultats montrent pour la première fois que les ROS produits par la NOX4 jouent un rôle critique dans l’inhibition de l’expression du NIS induite par BRAFV600E via la signalisation TGF-β/SMAD3 BRAFV600E induced-TGF-β secretion down-regulates sodium iodide symporter (NIS) expression via NOX4-dependent ROS generation in papillary thyroid carcinoma. Abstract : Papillary thyroid cancer (PTC) is the most common thyroid pathology and BRAF point mutations account for 40-60% of tumors. BRAFT1799A is the most frequent BRAF mutation and BRAFV600E positive tumors are often associated with a significant loss of sodium/iodide symporter (NIS) expression. Clinical results on patients harboring thyroid cancer with BRAF mutation have recently shown that MAPK pathway inhibition does not fully reverts the BRAF-induced NIS repression. BRAFV600E expression induces secretion of functional TGF-β which is a repressor of thyroid specific genes such as NIS. Importantly, NOX4 has been shown to be prominently expressed in an increasing number of tumors, in particular in PTCs. In breast cancer cells, a critical mechanism for cancer development involves the transcriptional regulation of NOX4 by TGF-β. This result prompted us to test NOX4 as a ROS-producing candidate induced by BRAF-induced TGF-β. In this report, we first show in PTCs a correlation between BRAFV600E status, NOX4 overexpression, and low NIS expression level. Then, using BCPAP cells as an in vitro PTC model, we demonstrate that BRAFV600E controls NOX4 and p22phox expression via TGF-β signalling. The TGF-β autocrine loop activated by BRAFV600E induces NOX4 and p22phox expression at the transcriptional level via phosphorylated SMAD3. Both constitutively expressed proteins form a functional NADPH oxidase which produces high intracellular ROS levels. ROS scavengers increase the NIS expression at both mRNA and protein levels, and rescue a functional NIS, indicating that ROS are involved in the repression of NIS. Knocking down NOX4 with specific siRNAs reinduces NIS expression at both mRNA and protein levels. Altogether, these results show for the first time that NOX4-dependent ROS generation has a critical role in BRAF-induced NIS repression via the TGF-β/SMAD3 oncogenic signalling

When activated by growth factors and mitogens, cellular receptors like EGFR become activated and transmit signals from the cell surface through the MAPK pathway into the nucleus of the cell, in order to elicit a cellular response like growth, apoptosis, proliferation, and survival. Over activation of EGFR even without receptor binding and members of the MAPK pathways like BRAF and MEK1 have been reported in tumorigenesis. We used Gateway Technology to study the recombinant protein expressions from these 3 proto-oncogenes in HEK293T and HeLa cell lines, using pLenti 6.3/V5 as the expression system. BRAF was neither detected in the HeLa nor in the HEK 293T cell lines, by mass spectrometry after transfection. Only in the HeLa cell line was EGFR identified meanwhile MEK1 was identified both in HEK293T and HeLa cell lines. However, the results showed that the proteins identified were endogenous to these cell lines and therefore, no recombinant proteins were expressed using the pLenti 6.3/V5, as the expression system.

Cette thèse est consacrée à l'étude de la manipulation et de la coordination robotique à deux bras ayant pour objectif le développement d'une approche unifiée dont différentes tâches seront décrites dans le même formalisme. Afin de fournir un cadre théorique compact et rigoureux, les techniques présentées utilisent les quaternions duaux afin de représenter les différents aspects de la modélisation cinématique ainsi que de la commande.Une nouvelle représentation de la manipulation à deux bras est proposée - l'espace dual des tâches de coopération - laquelle exploite l'algèbre des quaternions duaux afin d'unifier les précédentes approches présentées dans la littérature. La méthode est étendue pour prendre en compte l'ensemble des chaînes cinématiques couplées incluant la simulation d'un manipulateur mobile.Une application originale de l'espace dual des tâches de coopération est développée afin de représenter de manière intuitive les tâches principales impliquées dans une collaboration homme-robot. Plusieurs expérimentations sont réalisées pour valider les techniques proposées. De plus, cette thèse propose une nouvelle classe de tâches d'interaction homme-robot dans laquelle le robot contrôle tout les aspects de la coordination. Ainsi, au-delà du contrôle de son propre bras, le robot contrôle le bras de l'humain par le biais de la stimulation électrique fonctionnelle (FES) dans le cadre d'applications d'interaction robot / personne handicapée.Grâce à cette approche générique développée tout au long de cette thèse, les outils théoriques qui en résultent sont compacts et capables de décrire et de contrôler un large éventail de tâches de manipulations robotiques complexes This thesis is devoted to the study of robotic two-arm coordination/manipulation from a unified perspective, and conceptually different bimanual tasks are thus described within the same formalism. In order to provide a consistent and compact theory, the techniques presented herein use dual quaternions to represent every single aspect of robot kinematic modeling and control.A novel representation for two-arm manipulation is proposed—the cooperative dual task-space—which exploits the dual quaternion algebra to unify the various approaches found in the literature. The method is further extended to take into account any serially coupled kinematic chain, and a case study is performed using a simulated mobile manipulator. An original application of the cooperative dual task-space is proposed to intuitively represent general human-robot collaboration (HRC) tasks, and several experiments were performed to validate the proposed techniques. Furthermore, the thesis proposes a novel class of HRC taskswherein the robot controls all the coordination aspects; that is, in addition to controlling its own arm, the robot controls the human arm by means of functional electrical stimulation (FES).Thanks to the holistic approach developed throughout the thesis, the resultant theory is compact, uses a small set of mathematical tools, and is capable of describing and controlling a broad range of robot manipulation tasks

Arabinoxylans (AXs) are major components of non-starch polysaccharides (NSPs). Recently, AXs have attracted a great deal of attention, because of their possible antitumor and immunomodulation activities. These activities have been suggested to be related to the content of low molecular weight (Mw) AXs, in particular those with a Mw below 32 kDa. Rice bran and wheat pentosan are rich sources of AXs. However, extraction of AXs is difficult and often gives low yield. Various methods have been used to increase the extraction yield of AXs with varying degrees of success, such as alkaline hydrolysis and enzymatic treatment. However, some of these treatments have been reported to be either expensive or produce hazardous wastes and non-environmentally friendly. Extrusion processing has been used to increase the solubility of cereal dietary fibre, however, these studies used alkaline or enzyme treatments with the extrusion to maximise the extraction yield. The use of extrusion alone as a pre-treatment method to increase the extraction yield and reduce the molecular weight (Mw) of AXs from rice bran or wheat endosperm pentosan has not been investigated. Hence, the current study aimed to determine if extrusion alone could change the extraction yield and Mw of the AXs. Wheat endosperm pentosan and rice bran were extruded with a twin-screw extruder at screw speeds of 80 and 160 revolutions per minute (rpm). It was found that the extraction yield of AXs increased with an increase in screw speed and was accompanied by a decrease in the Mw of the AXs. In vitro studies using immunoassays to measure proinflammatory markers showed that AXs from extruded rice bran and wheat pentosan significantly (P < 0.05) increased nitric oxide (NO) and tumour necrosis factor α (TNFα) production from both U937 monocytes and macrophages in a concentration-dependent manner at (50, 500 and 1000 μg/ml), respectively. Moreover, the immunomodulatory activity of AXs was associated with the very low Mw of AXs. Moreover, inhibition of toll like receptor 4 (TLR4), which is known to be the receptor for bacterial lipopolysaccharides (LPS), significantly inhibited the AXs-induced increase in NO and TNFα production in both U937 monocytes and macrophages (P < 0.05), suggesting the actions of AXs may be mediated at least in part through TLR4. The findings of this study indicate that AXs may compete with LPS for the same receptor TLR4, resulting in decreasing the inflammatory response that LPS produces during the infection. Thus AXs can produce a non-detrimental moderate increase in the inflammatory response.

Inom disciplinen kognitiv hörselvetenskap tillämpas en integrerad ansats där taluppfattning beskrivs ur både ett auditivt bottom-up-perspektiv och ett kognitivt top-down-perspektiv. Studien undersökte hur taluppfattning varierar med, respektive utan top-down-stöd och vad de kognitiva förmågorna uppmärksamhet, minne och inhiberingsförmåga hade för inverkan. Taluppfattningsförmåga utvärderades med hjälp av ett Speech-In-Noise-test (SIN-test) som bestod av tre deltest där enstaviga ord presenterades i en ofördelaktig miljö bestående av röster som talande i mun på varandra. I deltest 1 presenterades ord i uppåtgående förhållande till rösterna utan kognitivt stöd, i deltest 2 presenterades ord i nedåtgående förhållande till rösterna med kognitivt stöd och i deltest 3 presenterades ord i uppåtgående förhållande till rösterna med kognitivt stöd. Detta resulterade i identifiering av tre tröskelnivåer för taluppfattning. Kognitiva förmågor utvärderades med hjälp av Reading span test, Size Comparison span task (SIC-span) och Paced Auditory Serial-Addition Test (PASAT). Envägs variansanalys av taluppfattningstesten visade på signifikanta skillnader mellan alla tre testen. I test 2 identifierades lägst tröskelvärde, därefter test 3 och slutligen test 1. Korrelationsanalys visade på icke signifikanta samband mellan de tre SIN-testen. Av de kognitiva förmågorna och SIN- testen visade endast PASAT A på ett signifikant samband med SIN-test 2. Analys av de kognitiva förmågorna och differenserna mellan SIN-testen gav inga signifikanta samband. Slutligen vid analys av de kognitiva förmågorna hade PASAT A och PASAT B ett starkt positivt samband medan PASAT B och Reading span test hade ett starkt negativt samband. Resultaten i studien talade för att top-down-stöd ger bättre taluppfattningsförmåga och att uppmärksamhetskapacitet är en förmåga som har en signifikant inverkan på taluppfattning i nedåtgående förhållande till ett brus av röster.

As fibras de coprodutos da produção de sucos, néctar e refrescos possuem propriedade funcional, envolvidas no metabolismo fermentativo da microbiota intestinal, com benefícios potenciais á saúde dos cães. O emprego de coprodutos alimentícios humano no desenvolvimento de petfood apresentam vantagens de sustentabilidade ambiental e econômica. O estudo objetivou avaliar os efeitos de dietas extrusadas com inclusão fixa de 3,5% de celulose (CEL), farelo de arroz desengordurado (FAR), polpa cítrica (PCT) e polpa de maçã (PMA) como fontes de fibras dietéticas solúveis e insolúveis nos coeficientes de digestibilidade aparente dos nutrientes (CDA), respostas glicêmicas e insulinêmicas pós- prandiais e produtos de fermentação nas fezes. O experimento foi realizado com oito cães adultos hígidos, peso médio de 15,0±5,0kg e idade média de 5,0±2,0 anos, distribuídos em delineamento em dois quadrados latino 4x4. Após 7 dias de adaptação dos animais, realizou-se coleta de fezes nos dias 5º-10º para determinar CDA das dietas, entre 12º ao 15º dia mensurados pH, produção de ácido lático, AGCC, AGCR e aminas biogênicas (AB) das fezes, seguidos pelas coletas de sangue para determinação das respostas glicêmicas e insulinêmicas pós-prandiais nos 16º e 17º dias. Não houve efeito dos tratamentos (p>0,05) no CDA da matéria orgânica (MO) e matéria mineral (MM), entretanto, houve efeito (p<0,05) nos CDAs da proteína bruta (PB), extrato etéreo em hidrólise ácida (EEHA), fibra bruta (FB) e extrativos não nitrogenados (ENN). O tratamento CEL apresentou maior CDA da PB e menor CDA da FB. O CDA dos ENN não apresentou diferença entre PMA e CEL. Em relação às características fecais, observou-se maior MS no tratamento CEL. Os tratamentos PCT e PMA resultaram em pH das fezes inferior aos tratamentos CEL e FAR e sem diferenças entre si. O tratamento PMA resultou em maior escore fecal e menor produção de fezes por cão/dia na MS, contrário ao tratamento PCT, que resultou em maior produção de fezes cão/dia, na MO e MS. Não foi observado efeito dos tratamentos (p>0,05) nas concentrações fecais de lactato e AB. O tratamento PCT resultou em maior produção de acetato, propionato e butirato que o tratamento CEL (p<0,05), enquanto que os tratamentos PCT e PMA resultaram em produção de AGCR mais elevados que FAR (p<0,05). The coproducts of fiber production of juice, nectar and soft drinks have functional property, involved in fermentative metabolism of intestinal microbiota, with potential benefits to health of dogs. The use of co-products of human food industry in the development of petfood has advantages of environmental and economic sustainability. The study aimed to evaluate the effects of extruded diets with fixed inclusion of 3.5% cellulose (CEL), defatted rice bran (FAR), citrus pulp (PCT) and apple pulp (PMA) as sources of soluble and insoluble dietary fibers on dog total tract apparent digestibility coefficient of nutrients (ADC), glycemic and insulin postprandial responses, and fermentation products in the stool. The experiment was carried out on eight healthy adult dogs of mean weight of 15.0 ± 5,0kg and a mean age of 5,0 ± 2,0 years allotted to a 4 x 4 Latin square two. After 7 days of adjustment of the animals to the diets, there was feces collection in the days 5th-10th to determine CDA diets, between 12th to 15th measured pH, lactic acid production, AGCC, AGCR and biogenic amines (BA) of the stool, followed by blood samples for determination of glucose and insulin responses postprandial in 16 and 17 days. There was no effect of the treatments (p> 0.05) in the ADC of organic matter (OM) and mineral matter (MM), however, was no effect (p <0.05) in CDAs of dry matter (DM), crude protein (CP), ether extract acid hydrolysis (EEHA), crude fiber (CF) and nitrogen free extract (NFE). The CEL treatment obtained higher ADC of CP and lower ADC of FB. The ACD of NFE no difference between PMA and CEL. For fecal characteristics, there was a higher DM in CEL treatment. PCT treatments and PMA resulted in fecal pH lower than the CEL and FAR treatments and without differences. The PMA treatment resulted in greater fecal score and lower production of faeces per dog / day in DM, contrary to the PCT treatment, which resulted in increased production of dog feces / day in MO and DM. There was no effect of the treatments (p> 0.05) in fecal concentrations of lactate and AB. The PCT treatment resulted in increased production of acetate, propionate and butyrate that CEL treatment (p <0.05), while the PCT and PMA treatments resulted in higher production AGCR that FAR (p <0.05). There was no effect (p <0.05) of sources of fiber studied for glycemic and insulin curves, area under the curve (AUC) of glucose and insulin, increasing blood glucose and insulin.
