Streszczenie:
A winter bloom event of Trichodesmium erythraeum was monitored for the first time in the southeastern coastal parts of Bangladesh along the Bay of Bengal. This study presents the brownish to light pinkish bloom that appeared in mid-winter and disappeared abruptly during spring. Heavy blooms of T. erythraeum revealed the highest concentration of 91.47 ± 52.94 × 103 colonies/L in the Bakkhali River Estuary, and 66.93 ± 12.95 × 103 colonies/L in the Maheshkhali Channel of the Bay of Bengal. Three distinct morphological shapes, namely puffs, tufts and asymmetrical colonies, were depicted as major types. Several environmental factors, such as water temperature, salinity, pH, dissolved oxygen, NO3–N and PO4–P, were analyzed to determine their relationship with the occurrence, abundance and bloom formation of T. erythraeum. The abundance of the species showed a positive correlation with salinity and pH while exhibiting a negative correlation with temperature and DO. A cluster analysis revealed a clear indication of T. erythraeum bloom during winter. Thus, the prevalence of the highest density of the bloom in the present study area strongly suggests increased monitoring and research efforts in order to effectively manage or impede harmful algal blooms.

Streszczenie:
Shaker-type K+ channels are critical for plant K+ acquisition and translocation that play key roles during plant growth and development. However, molecular mechanisms towards K+ channels are extremely rare in fruit trees, especially in peach. In this study, we identified 7 putative shaker-type K+ channel genes from peach, which were unevenly distributed on 5 chromosomes. The peach shaker K+ channel proteins were classified into 5 subfamilies, I-V, and were tightly clustered with pear homologs in the phylogenetic tree. Various cis-acting regulatory elements were detected in the promoter region of the shaker-type K+ channel genes, including phytohormone-responsive, abiotic stress-responsive, and development regulatory elements. The peach shaker K+ channel genes were expressed differentially in distinct tissues, and PpSPIK was specifically expressed in the full-bloom flowers; PpKAT1 and PpGORK were predominantly expressed in the leaves, while PpAKT1, PpKC1, and PpSKOR were majorly expressed in the roots. The peach shaker K+ channel genes were differentially regulated by abiotic stresses in that K+ deficiency, and ABA treatment mainly increased the shaker K+ channel gene expression throughout the whole seedling, whereas NaCl and PEG treatment reduced the shaker K+ channel gene expression, especially in the roots. Moreover, electrophysiological analysis demonstrated that PpSKOR is a typical voltage-dependent outwardly rectifying K+ channel in peach. This study lays a molecular basis for further functional studies of the shaker-type K+ channel genes in peach and provides a theoretical foundation for K+ nutrition and balance research in fruit trees.

Streszczenie:
Heavy reduction (HR) is an effective technique to control V segregation in continuous casting bloom, but the effect of segregation improvement is limited by the parameters such as reduction position and reduction amounts. In order to improve the macrosegregation of bloom, numerical simulation and plant experiments are adopted in this research. A heat transfer model and a reduction model with comprehensive thermo-physical parameters were established. The two models were verified by comparing the measured surface temperature and the theoretical strain at the solidification front. It is determined that the position of the HR of the bearing steel bloom is 20.82 m~24.97 m from the meniscus, and the solid fraction in the center of the bloom is 0.6~1. The total reduction of the HR is set to 30 mm, and the reduction of each roller in the reduction range is set to 4 mm, 5 mm, 9 mm, 7 mm, and 5 mm, respectively, to prevent the formation of internal cracks. Plant trials were conducted to verify the effect of the optimized HR. The results show that the carbon segregation degree on the V channel and non-channel of the bloom decreases from 1.2 to 1.16 and increases from 0.93 to 0.95, respectively, and the central carbon segregation degree decreases from 1.17 to 1.15. Meanwhile, the internal crack was not found in the bloom.

Six blooms of Heterosigma akashiwo(Raphidophyceae) were observed from March 2007 through March 2008 in the Rodrigo de Freitas Lagoon, a semi-confined eutrophic system located in Rio de Janeiro state, southeast Brazil. Vegetative cells of H. akashiwo analysed by optical and electron microscopy showed morphology as described in the literature. The blooms (2.8 × 104 to 4 × 108 cell.L–1) were restricted to the middle section of the Piraquê Channel, which is situated in the northeastern part of the lagoon and receives freshwater inflow. The salinity of subsurface water and the channel depth showed significant negative correlations with H. akashiwo abundances, and appeared to restrict the blooms to this compartment of the lagoon. No fish mortality was associated with the H. akashiwo blooms, nor were brevetoxins detected in a cell extract obtained from the bloom observed on 19 March 2007.

The evolution of a Phaeocystis bloom off Plymouth in spring 1990 is described. Data were obtained (1) during weekly visits to station El to measure physical parameters, nutrient concentrations, primary production and grazing pressure and (2) by regular tows of an Undulating Oceanographic Recorder around a circuit of five stations including El to detect stratification, monitor the geographical distribution of chlorophyll and determine vertical profiles at each station. The data show that the Phaeocystis bloom emanated from near-shore, its spread towards the south-east correlating with the wind direction prevailing at the time. Due to bad weather, seasonal stratification had not been established at the time of the bloom so that a major diatom outburst had not occurred and nutrients remained high as the Phaeocystis moved offshore.Zooplankton populations at El, averaged over 50 m, decreased whilst Phaeocystis was present, resulting in very low grazing pressure. Termination of the bloom was not, therefore, due to consumption by herbivores. It was, instead, caused by Phaeocystis colonies becoming nutrient-depleted and releasing single cells which were transported downwards. As a result of this, chlorophyll concentrations near the bottom remained high for some time after the Phaeocystis had disappeared from the surface water.A scenario is proposed to explain the observations in terms of our current understanding of the ecophysiology of Phaeocystis.

Streszczenie:
SynopsisFor the onset of the phytoplankton spring bloom in temperate waters, the irradiance, the concentration of accumulated nutrients and the stability of the water column are of great importance. The “new” production in spring is produced mainly by chain forming diatoms. The dissipation of the spring bloom is due to nutrient depletion in the stabilised surface layer, loss of cells by sinking and grazing by herbivorous zooplankton. After the nutrient concentration is lowered, the rate of production will depend primarily on the rate of replenishment of nutrients. In open ocean environments we find “regenerated production” which is due to exudated and excreted nutrient salts e.g. ammonium. The dominating group of phytoplankters are small flagellates. It is argued that a substantial part of the phytoplankton standing stock in spring is lost from the euphotic zone due to direct sinking of cells or accelerated sinking of cell-aggregates. It is postulated that faecal pellets of micro- and mesozooplankton are retained and recycled in the mixed layer, whereas macrozooplankton faecal strings transport a considerable amount of organic matter to the benthal.

Streszczenie:
Algal bloom is a major concern worldwide. In this study, we characterised the physical and biochemical parameters during an algal bloom event in a coastal lagoon in an attempt to predict local blooms in the future. Results showed that the highest concentrations of dissolved inorganic phosphorus (DIP), chlorophyll a (chl a) and phytoplankton abundance were found in the inner area, whereas the highest dissolved inorganic nitrogen (DIN) concentration occurred near the inlet-outlet channel. Chl a was correlated with DIP, and there was a significant exponential relationship between chl a and the nitrogen to phosphorus ratio (N/P ratio) across all sampling stations and times. A higher proportion of the variation in chl a was explained by the N/P ratio than either DIP or DIN. We found that a N/P ratio <2.38 will likely trigger an algal bloom (chl a ≥ 10 µgL–1) in the lagoon. Our results suggest that the N/P ratio could be used as an expedient and reliable measure of the potential eutrophic status of coastal lagoons.

Streszczenie:
Molecular communication (MC) is a new and promising interdisciplinary bio-inspired communication paradigm, which uses molecules as information carriers. Differing from traditional communication, MC is proposed as a feasible solution for nanoscale communication with the help of biological scenarios to overcome the communication limitations. Meanwhile, it is inspired by intracellular and intercellular communication, which involves exchange of information through the transmission, propagation, and reception of molecules. Blood capillaries, extensively distributed in the human body and mutually connected with tissues, are potentially applied to MC, which is the major motivation of this thesis. The focus of this PhD thesis is on the channel modelling of blood capillaries or blood vessels. The objectives of the research are to provide solutions to the modelling of blood capillary-based MC from a communication engineering and information theory perspective. The relationship of the biological scenario in blood capillaries to a communication system is studied. After demonstrating the mapping from biological phenomenon to emission, propagation and reception processes, system models are established. There are three models of blood capillaries behind different biological scenarios. Firstly, the thesis establishes a basic model of vesicle release, vesicle diffusion through blood capillary and ligand reception processes within the endocrine phenomenon. Moreover, differing from previous research in macroscopic Fick's diffusion, this work involves microscopic Langevin diffusion to describe the propagation process with a frequency domain method being proposed to calculate the information-theoretical performance channel capacity. Secondly, a much more realistic blood capillary model with blood flow drift which matches a laminar flow regime is presented, where a generalised Langevin equation is used to model the drift force exerted by blood flow. Finally, the thesis establishes a single input and multiple output MC model with hierarchical levels of Y-shaped bifurcation of blood capillaries, then BER, SNR, and channel capacity performance are analysed.

Streszczenie:
The epithelial sodium channel (ENaC) is the central component of the sodium absorption pathway in epithelia. It is critical for sodium homeostasis and blood pressure control, which is demonstrated by rare genetic disorders such as Liddles syndrome and pseudohypoaldosteronism type I, that are associated with hyper- and hypotension, respectively. ENaC is mainly regulated by mechanisms that control the expression of active channels at the cell surface. Ubiquitin ligases of the Nedd4-like family, such as Nedd4 and Nedd4-2 decrease epithelial sodium absorption by binding to and targeting ENaC for endocytosis and degradation. This is most likely achieved by catalyzing the ubiquitination of ENaC. Conversely the serum- and glucocorticoid regulated kinase (SGK) increases ENaC activity. This effect is partly mediated by the interaction of SGK with the ubiquitin ligases Nedd4 and Nedd4-2. SGK is able to bind to both Nedd4 and Nedd4-2, however only Nedd4-2 is phosphorylated by SGK. The phosphorylation of Nedd4-2 inhibits its interaction with ENaC, thus reducing ENaC ubiquitination, thereby increasing surface expression and sodium absorption. Nedd4-like proteins interact with ENaC via their WW-domains. These domains bind PY-motifs (PPXY) present in ENaC subunits. Nedd4 and Nedd4-2 both have four highly similar WW-domains. Previous studies have shown that interaction between Nedd4 and ENaC is mainly mediated by WW-domain 3. SGK also has a PY-motif; therefore it was analyzed whether the WW-domains of Nedd4 and Nedd4-2 mediate binding to SGK. Here, it is shown that single or tandem WW-domains of Nedd4 and Nedd4-2 mediate binding to SGK and that, despite their high similarity, different WW-domains of Nedd4 and Nedd4-2 are involved. These data also suggest that WW-domains 2 and 3 of Nedd4-2 mediate the interaction with SGK in a concerted manner, and that in vitro the phosphorylation of SGK at serine residue 422 increases its affinity for the WW-domains of Nedd4-2. The stimulatory effect of SGK on ENaC activity is partly mediated via Nedd4-2 and will decrease if competition between Nedd4 and Nedd4-2 for binding to SGK occurs. Here it is shown that Nedd4 and Nedd4-2 are located in the same subcellular compartment and that they compete for binding to SGK. Besides its function in the proteasomal degradation pathway ubiquitination is involved in the regulation of membrane protein trafficking, including their endocytosis. ENaC was shown previously to be ubiquitinated. Here, we provide evidence that ENaC can be ubiquitinated differentially depending on its cellular location. Channels residing in the plasma membrane are multiubiquitinated and we suggest that this serves as an internalization signal for ENaC and a control for further trafficking. Cytosolic ENaC is mainly polyubiquitinated, and therefore probably targeted for proteasomal degradation. However, mono- and multiubiquitination of ENaC located within the cytosol is very likely to occur as well. In addition, it is shown that both proteasomal and lysosomal pathways are involved in the regulation of ENaC.

Streszczenie:
Erythrocytes are one of the cellular models frequently used to decipher the physiology of membrane transport. However, if pumps, antiporters and cotransporters are now well defined, the molecular identity and the physiological role of the conductive pathways present in the membrane of red blood cell (RBC) is still elusive albeit the growing evidences of their role in physiological and pathological conditions. The present work, using the patch clamp technique and biophysical studies, shows that: 1) The changes in osmotic fragility observed after Gardos channel dependent dehydration is mainly not due to rehydration of the cell via membrane transporters but rather due to change in membrane properties elicited by the Ca2+ loading. It reflects a specific calcium-induced lytic vulnerability of the membrane leading to rupture before the cells attain their maximal spherical volumes. 2) The Gardos channel can be transiently activated when seal formation induces membrane deformation. This phenomenon can result only from activation of a permeability pathway with a finite Ca2+ conductance. This transient activity generates secondary transient anionic channel activity that has been studied further. 3) The diversity of anionic channel activities recorded in normal human RBCs, as well as in P. Falciparum-infected RBCs, corresponds to different kinetic modalities of a unique type of maxi anion channel with multiple conductance levels, gating properties and pharmacology. Finally, the present work contributes to the understanding of the role of ion channels in RBCs and opens questions on the contribution of ion channels in the rheological properties of RBCs. Keywords: erythrocyte, ionic channels, osmotic fragility, patch clamp technique
In this study we propose a new cooling concept using encapsulated phase-change particles flowing in a parallel-plate mini-channel. This novel concept is inspired by the gas exchange process in alveolar capillaries, where red blood cells (RBCs) flow with blood plasma, yielding very high gas transfer efficiency. Another important characteristic of alveolar capillary blood flow, which is related to the high efficiency of the lungs, is the snug fitting of the RBCs into the capillary. Hence, we conjecture that using particles similar to RBCs, i.e. with diameter similar to the spacing between the parallel plates, is likely to lead to high heat transfer efficiency as well. We report here experimental results of preliminary tests performed with octadecane paraffin (C18H38), a phase-change material, encapsulated in a thin melamine shell, flowing with water through a heated parallel-plate channel test module. We measured the temperature distribution along the heated surface of the channel for various water flow rates, with and without particles, and varying the number of particles. Results are reported in terms of the channel heated surface average temperature and the average Nusselt number.

Streszczenie:
As it has been shown previously, the blood flow in the heart and the main vessels is a self-organizing tornado-like flow of a viscous fluid, exhaustively described by the particular solution of nonstationary hydrodynamic equations coined in 1986. This solution is implemented in a local dynamic cylindrical coordinate system, which origin moves with the flow. Streamlines of these flows are characterized with an axial symmetry and determine certain restrictions to the geometry of the flow channels. It has been proved, that the experimentally measured orientations of intraventricular trabeculae and dynamic geometric characteristics of the aortic flow channel during the cardiac cycle satisfy the conditions for the self-organization of tornado-like blood flow. This enables to obtain relative estimates of the flow parameters using the geometric characteristics of the flow channel. For this, a set of experimentally measured quasi-invariant anatomical marks was used, which static and dynamic parameters are utterly determined by the structure of the dominant blood jet. Such marks are the position of the embouchures of pulmonary veins and the left atrium auricle, the directions of the trabecular profile on the left ventricle streamlined surface, the dynamic geometry of the mitral and aortic valves, the length and radial elasticity of the aorta, etc. These marks allow a formal localization of the dynamic coordinate system position of a swirling blood flow in the investigated part of the cardiovascular channel and more accurate estimation of the jet structural parameters evolution. Basing on these parameters, a consistent theory of the initiation and evolution of swirling blood jet in the flow channel beginning from the left atrium to the aorta and its main branches has been proposed. The result of the study is a concept, integrating the measured characteristics of the heart and the aorta flow channel (dynamic dimensions, elasticity) and the structural parameters of swirling blood flow. It was shown that flow channel pathological changes significantly affect the flow structure.

Experimental studies of blood microflows in rectangular biocompatible polymer microchannels measured using micro-particle image velocimetry are reported. The data processing methods, data collection methods, and choice of channel material are demonstrated to impact the velocity profile measurements obtained. Results show that the use of red blood cells as tracer particles creates a large depth of correlation which can approach the size of the vessel itself and decrease the accuracy of the method. It is shown that changing the amount of overlap in the post-processing parameters affects the results by nearly 10%. The velocity profile is studied as a function of the flow rate of the blood, the hematocrit, or percentage of red blood cells, the shape of the channel, and the channel material. The results highlighted here show that the best processing options include pre-processing, the use of fluorescent tracer particles instead of the red blood cells themselves as tracers give a more accurate prediction of the profile, and the use of silicone as the channel material more closely mimics the behavior of physiology. Acrylic biocompatible polymer channels are shown to give a more parabolic profile at lower levels of hematocrit, while silicone biocompatible polymer channels give a velocity profile that looks more like in vivo flow studies.

This study presents numerical simulations of forced convection with parachute-shaped encapsulated phase-change material particles in water, flowing through a square cross-section duct with top and bottom iso-flux surfaces. The system is inspired by the gas exchange process in the alveolar capillaries between the red blood cells (RBC) and the lung tissue. The numerical model was developed for the motion of elongated encapsulated phase change particles along a channel in a particulate flow where particle diameters are comparable with the channel height. Results of the heat transfer enhancement for the parachute-shaped particles are compared with the circular particles. Results reveal that the key role in heat transfer enhancement is the snugness movement of the particles and the parachute-shaped geometry yields small changes in heat transfer coefficient when compared to the circular ones. The effects of various parameters including particle diameter and volume-fraction, as well as fluid speed, on the heat transfer coefficient is investigated and reported in this paper.

Currently, several studies and experiments are being done to create a new generation of ultra-low-power wearable sensors. For instance, our group is currently working towards the development of a high-performance flexible pressure sensor. However, with the creation of new sensors, a need for a standard test method is necessary. Therefore, we opted to create a standardized testbed to evaluate the pressure applied to sensors. A pulse wave is generated when the heart pumps blood causing a change in the volume of the blood vessel. In order to eliminate the need of human subjects when testing pressure sensors, we utilized polymeric material, which mimics human flesh. The goal is to simulate human pulse by pumping air into a polymeric pocket which s deformed. The project is realized by stepper motor and controlled with an Arduino board. Furthermore, this device has the ability to simulate pulse wave form with different frequencies. This in turn allows us to simulate conditions such as bradycardia, tachycardia, systolic pressure, and diastolic pressure.

Streszczenie:
Purpose/Background Direct laryngoscopy and endotracheal intubation after induction of anesthesia can cause a reflex sympathetic surge of catecholamines caused by airway stimulation. This may cause hypertension, tachycardia, and arrhythmias. This reflex can be detrimental in patients with poor cardiac reserve and can be poorly tolerated and lead to adverse events such as myocardial ischemia. Fentanyl, a potent opioid, with a rapid onset and short duration of action is given during induction to block the sympathetic response. With a rise in the opioid crisis and finding ways to change the practice in medicine to use less opioids, dexmedetomidine, an alpha 2 adrenergic agonist, can decrease the release of norepinephrine, has analgesic properties, and can lower the heart rate. Methods In this scoping review, studies published between 2009 and 2021 that compared fentanyl and dexmedetomidine during general anesthesia induction and endotracheal intubation of surgical patients over the age of 18 were included. Full text, peer-reviewed studies in English were included with no limit on country of study. The outcomes included post-operative reviews of decrease in pain medication usage and hemodynamic stability. Studies that were included focused on hemodynamic variables such as systolic blood pressure, diastolic blood pressure, mean arterial pressure, heart rate, and use of opioids post-surgery. Result Of 2,114 results from our search, 10 articles were selected based on multiple eligibility criteria of age greater than 18, patients undergoing endotracheal intubation after induction of general anesthesia, and required either a dose of dexmedetomidine or fentanyl to be given prior to intubation. Dexmedetomidine was shown to effectively attenuate the sympathetic surge during intubation over fentanyl. Dexmedetomidine showed a greater reduction in heart rate, systolic blood pressure, diastolic blood pressure, mean arterial pressure than fentanyl, causing better hemodynamic stability in patients undergoing elective surgery.Implications for Nursing Practice Findings during this scoping review indicate that dexmedetomidine is a safe and effective alternative to fentanyl during induction of general anesthesia and endotracheal intubation in attenuating the hemodynamic response. It is also a safe choice for opioid-free anesthesia.

Harmful algal blooms (HABs) are occurring with increasing frequency and severity across the globe in part due to climate change and anthropogenic pollution (Bullerjahn et al. 2016). HABs produce several classes of toxins; however, microcystins (MCs) are the most commonly studied (Lone et al. 2015) and can be potent toxins with LD50s in the range of 50 μg/kg (Puddick et al. 2014). Sample analysis in laboratories, typically by high-pressure liquid chromatography tandem mass spectrometry (HPLC-MS/MS) or by Enzyme Linked Immunosorbent Assays (ELISAs) (USEPA 2015). These analytical techniques are highly sensitive and selective for the given toxins; however, the time it takes to collect, transfer, prepare, and analyze a sample before the data can be reported is significant; often, multiple days is the most expeditious.
