Physiol. 83(1): 172–178, 1997.—The effectiveness of dobutamine (Dob) in preventing bone loss during 14 days of hindlimb suspension (Sus) was tested in exercise-trained (Ex; n = 25) and sedentary (Sed; n = 22) rats (age 155 days). One-half of each group was given Dob (2 mg ⋅ kg−1 ⋅ day−1) or saline (Sal). Histomorphometric measurements at midfemur revealed a 17% smaller cortical bone area (CBA) and a 32% lower periosteal mineral apposition rate (MAR) in suspended vs. nonsuspended Sed/Sal rats. Dob abolished this decline in CBA in Sed/Sus rats, probably via an attenuation of the decrease in periosteal MAR; similar but nonsignificant effects on cross-sectional moment of inertia were observed. Nonsuspended Ex rats had no change in bone CBA when CBA is indexed to body weight. Sus appeared to uncouple the relationship between soleus weight and CBA. Dob attenuated the 43% decline in soleus weight after Sus in Ex but not in Sed rats. In summary, vigorous Ex before Sus does not affect loss of bone mass due to unloading; Dob effectively maintains CBA in Sed rats subjected to suspension.

Abstract Metabolic reprogramming plays a crucial role in the maintenance and differentiation of various cancers, including multiple myeloma (MM), the second-most common incurable hematological malignancy. Drug resistance poses a significant challenge to conventional proteasome inhibitor (PI) based therapy. A body of literature suggests that several metabolites are significantly deregulated in MM patients, however, their role in driving resistance is unclear. This study aims to identify metabolites modulated during acquired resistance to proteasome inhibitors in relapsed refractory (RR)MM. Two clinically relevant cell line models of PI-resistance (PIR) were developed by dose-escalation of the PI over one year. Cell cytotoxicity assays, immunoblotting, immunofluorescence, and flow cytometric analyses were used to assess the resistance phenotype. Mass spectrometry-based metabolomic profiling was performed to identify altered metabolites in resistant as compared to sensitive cell lines. PIR cells demonstrated pan-resistance to multiple PIs, including a >10-fold increase in IC50 for oprozomib compared to parental cells. PIR cells had higher bioenergetic and glycolytic activity (Seahorse) while gene expression profiling suggested downregulation of oxidative phosphorylation. Mass spectrometry-based metabolomic profiling identified distinct metabolites deregulated in drug-resistant cells. The metabolomic profiles of the drug-resistant cell lines showed that the lipid synthesis and beta-oxidation of fatty acid pathways were highly modulated. Several pathway components demonstrated a negative association with overall survival in multiple myeloma patient datasets (MMRF, GSE31161 & GSE9782). PI resistance led to increased phosphorylation of ACLY involved in fatty acid synthesis and increased expression of CPT1A indicative of activation of the beta-oxidation pathway. Use of ACLY and FASN tool compounds affected proliferation, survival, and response to treatment in PIR cells. Metabolites modulated during acquired resistance to proteasome inhibitor in relapsed refractory (RR)MM were identified and potential mechanisms driving resistance were studied. After his break with Kinsey, Legman aimed to make a place for himself among New York City intellectuals as a full-time writer and editor on topics of sex. Between 1946 and 1948, he was at work drafting Love & Death, a series of essays on the effects of censorship and repression on American culture. Love & Death was an early foray into the criticism of mass culture and an early content analysis of comic books, detective fiction, and stereotypes of women in mass culture. In it Legman argued that the repression of sex in mass culture inevitably led to a sick and violent society. This chapter shows Legman developing and circulating his essays among New York’s writers and editors, unpacks his negative critique of mass culture, and presents the responses of some of his prominent readers. Although the book was rejected by several dozen publishers, Legman got attention for his views. Through Fredric Wertham, one of the principal psychiatrists in New York City, Legman gave a public lecture on psychoanalytic approaches to comic books. Also discussed is Legman’s marriage to his first wife, Beverley Keith, and the salons they held at their tiny house in the Bronx.

Die filling is a process in which particles/granules are deposited into a cavity (i.e. a die) from a feed shoe/hopper and is a critical processing stage in the manufacture of particulate products, such as pharmaceutical and detergent tablets, catalyst pellets, ceramic and powder metallurgical components. In most particulate product manufacturing processes, a relative low filling speed (< 1 m/s) is normally employed. Nevertheless, high speed die filling processes have been developed recently for dosing individual granules, in which granules are deposited into a die when it travels at a speed up to 20 m/s passing under a stationary feed shoe/hopper. For this process, the mass flow rate is an important process parameter that controls the accuracy and consistency of granule dosing. In order to determine how the mass flow rate varies with the filling speed, in this study, the high speed die filling process was modelled using a discrete element method (DEM). In order to validate the DEM model, hopper flow with mono-disperse systems of various particle sizes was simulated and the mass flow rate was examined and compared with the prediction of Beverloo equation. Excellent agreements were obtained, demonstrating that the DEM can well predict the flow behaviour. DEM modelling of high speed die filling processes at various filling speeds were then performed. It was found that the mass flow rate in the high speed die filling process is proportional to the filling speed.

Abstract In reaction to the sacking of First African Baptist Church by the police and white Tuscaloosans, the Black church community armed themselves, and formed an armed self-defense group called the Defenders. In addition to these protective measures, Reverend Martin Luther King, Jr. sent three of his most reliable SCLC workers—James Bevel, Harold Middlebrook, and Richard Boone—to help not only sustain the movement in Tuscaloosa, but also over see the birth of a more aggressive campaign of boycotting and marching and filing a series of lawsuits. A story of a new mass meeting at a different church, Bailey Tabernacle CME Church, and the efforts of police to stop it, are covered. Reverend Rogers has mixed feelings about the Defenders and the increasing purchase and ownership of guns by Blacks, even as he and his family suffered from the constant stress of knowing they were targets for the Klan, despite being guarded by the Defenders. Reverend Rogers comes to see the necessity of ideological flexibility on issues of non-violent social change, and greets the passage of the Civil Rights Act with a mix of joy and caution, as knew that the fight for civil rights was still far from over.

The mass flow of granular matter through orifices can be described by the well-known Beverloo law. It depends on particle and orifice sizes, interparticle and particle/container interaction forces, particles’ surfaces - to name a few influences on the mass flow rate. We present an experimental study of the flow of a set of ready-to-press (RTP) hard metal powders through orifices of varying diameter. The obtained parameters of the Beverloo law are compared with angle of repose measurements. The interplay between attractive interparticle forces and gravitational forces are discussed for both types of experimental measurements and related to the difference between particle and orifice size.

Initial results investigating windage power loss on a rotating shrouded spiral bevel gear using a parametric solid model and Computational Fluid Dynamics (CFD) are presented. The context behind this study is a desire to use CFD as a tool to investigate heat-to-oil within gas turbine bearing chambers and gearboxes in order to reduce costly rig-based experiments. This paper contains the methodology for creation of the parametric model of a spiral bevel gear in Pro/Engineer, formulation of a mesh in ICEM CFD and the subsequent CFD analysis in Fluent 6.2.26 and 12.0.16. A single tooth segment of a 91 teethed spiral bevel gear is produced with periodic boundaries imposed to reduce computational cost. Validation against experimental results for a single control gear is shown with particularly good correlation between static pressure rise across the face of the gear. Mesh verification is also presented. Using the model to change the module of the gear (effectively the number of teeth), investigations show that windage power loss reduces when the number of teeth increases. Analysis of the static pressure variation throughout the domain shows that all gears tested exhibit a linearly increasing relationship between non-dimensional mass-flow-rate and the pressure drop through the shroud restriction. The control gear was seen to have only a weak increase in static pressure gain across the gear tooth as the mass-flow-rate increases; however, a far larger increase exists for the module cases tested — at comparable mass-flow-rates to the control gear. As the number of teeth increase, the pressure gain across the gear reduces, and vice-versa. It is this difference between the gears that results in dissimilar windage power losses.

This paper presents a computational investigation into the effects of inlet swirl on the fluid flows computed for a spiral bevel gear and shroud pairing. This is with a specific focus on aeroengine internal gearboxes where the power offtake gear is located in a chamber where a highly swirled environment exists. Previous work for a single rotating gear and stationary shroud [1] has been performed in isolation and with idealised boundary conditions at both the inlet and outlet of the system. This effectively decouples the shrouded gear subsystem from the rest of the gearbox. In the present study a parametric investigation has been conducted varying the amount of inlet swirl and the effect this has on the mass flowrate of air “pumped” through the gear. The paper presents data showing that compared to no inlet swirl, a higher mass flowrate is induced when there is swirl, with up to 20% higher mass flowrate occurring when the tangential velocity at shroud inlet is 50% of the axial velocity component. For swirl above 50% the mass flowrate drops back somewhat remaining higher than for the no-swirl case for values investigated. Gear windage power loss is a function of mass flowrate and consequently higher windage losses occur for higher mass flowrates. In an aeroengine flow through the gear exits through slots in the shroud and this paper shows that the swirl velocity component at the shroud exit holes is relatively insensitive to the velocity components entering the gear/shroud system for a given geometry. Further to this, the effect of the shroud outlet geometry on the flow leaving the back of the gear has been investigated and quantified. It is shown that accurate geometric representation is required as the outlet geometry has a significant effect on the computed mass flow through the gear-shroud system and consequently on the computed windage power loss.

A computational methodology is proposed for prediction of power losses due to pocketing (pumping or squeezing) of oil at the mesh interface of spiral bevel gears. The model employs an existing cutting simulation procedure to define surface geometries of the gears through face-milling and face-hobbing processes. A novel hypoidal discretization method is proposed to define pocket volumes between meshing gear teeth along circumferential and face width directions. An existing fluid mechanics formulation, utilizing principles of conservation of mass, momentum and energy, is used to compute the load-independent (spin) power losses due to pocketing of the medium in the gear mesh interface. A simulation of aerospace applications is presented to highlight the effects of lubrication conditions, on pocketing power losses.

Abstract Experimental tests were performed on the OH-58A helicopter main-rotor transmission in the NASA Lewis 500-hp helicopter transmission test stand. The testing was part of a joint Navy/NASA/Army lubrication program. The objectives of the joint program are to develop and demonstrate a separate lubricant for gearboxes with improved performance in life and load-carrying capacity. The goal of these experiments was to develop a testing procedure to fail certain transmission components using a MIL-L-23699 based reference oil and then to run identical tests with improved lubricants and demonstrate improved performance. The tests were directed at components that failed due to marginal lubrication from Navy field experience. These failures included mast shaft bearing micropitting, sun gear and planet bearing fatigue, and spiral bevel gear scoring. A variety of tests were performed and over 900 hr of total run time accumulated for these tests. Some success was achieved in developing a testing procedure to produce sun gear and planet bearing fatigue failures. Only marginal success was achieved in producing mast shaft bearing micropitting and spiral bevel gear scoring.

The use of robotics to enhance visualization and tissue manipulation capabilities is contributing to the advancement of minimally invasive surgery (MIS). For the development of surgical robot manipulators, the use of advanced dynamic control is an important aspect at the design stage to determine the driving forces and/or torques which must be exerted by the actuators in order to produce a desirable trajectory of the end effector. Therefore, this study focuses on the generation of inverse dynamic models for a spherical bevel-geared mechanism called CoBRASurge (Compact Bevel-geared Robot for Advanced Surgery), which is used as a surgical tool manipulator. For given typical trajectories of end effectors in clinical experiments, the motion of each element in the mechanism can be derived using the inverse kinematic equations. The driving torques exerted by actuators can be determined according to the presented inverse dynamic formulations. The simulation results of CoBRASurge reveal the nature of the driving torques in spherical bevel-geared mechanisms. Such models can be used for the design of advanced dynamic control systems, including gravity compensation and haptic interfaces for enhanced surgical functionality. In addition, sensitivity analysis of mass contribution has been performed to evaluate the effect of individual elements on the peak driving torques, which provides a solid guideline for the design of the next-generation CoBRASurge prototype. The present dynamic modeling methodology also presents a general dynamic analysis approach for other spherical articulated linkage mechanisms.

In some aero-engine applications a spiral bevel gear is mounted in a bearing chamber. The windage power losses (WPL) associated with the gear make a significant contribution to the overall heat generation within the bearing chamber and a potential method of WPL reduction and lubrication/cooling oil management is to shroud the gear. At the University of Nottingham Technology Centre in Gas Turbine Transmission Systems experimental and computational techniques are being applied to enhance understanding of shroud performance and design. This paper presents results from the first stage of the investigation in which the windage losses associated with rotating an unmeshed spiral bevel gear in air have been studied. The potential to reduce gear WPL by shrouding is clearly demonstrated with the WPL for the shrouded gear reduced on average by 75% for clockwise rotation and by 70% for anticlockwise rotation. Given the physical similarity between a shrouded gear and a centrifugal fan an attempt was made to characterize the performance of the shrouded gear in a similar manner to that used for a centrifugal fan i.e. as a function of the mass flow rate of air flowing through the shroud. It is demonstrated that in many aspects the gear performance was analogous to centrifugal fan behavior. It is further demonstrated that fan design approaches could be adopted to inform the detail of the shroud design and to translate test rig performance to different operating conditions.

Although bevel-gear wrists are widely used in industrial manipulators due to their simple kinematics and low manufacturing cost, their gear trains function under rolling and sliding, the latter bringing about noise and vibration. Sliding is inherent to the straight teeth of the bevel gears of these trains. Moreover, un-avoidable backlash introduces unmodeled dynamics, which mars robot performance. To alleviate these drawbacks, a gearless pitch-roll wrist is currently under design for low backlash and high stiffness. The wrist consists of spherical cam-rollers and spherical Stephenson linkages; two roller-carrying disks drive a combination of cams and Stephenson mechanisms rotating as a differential mechanism. In this paper, the design of the chain of spherical Stephenson mechanisms (SSMs) is introduced. The problem of the dimensional synthesis is addressed and interference avoidance is discussed. An embodiment of the concept is also included.

In many aeroengines the accessory power offtake is achieved using a spiral bevel gear set running off one of the main shafts. The crown and bevel gears are housed in an internal gearbox and there is significant heat generation within this chamber, some of which is attributed to windage power loss (WPL) generated by the gear. Over the past few years the University of Nottingham Technology Centre (UTC) in Gas Turbine Transmissions has been researching spiral bevel gear windage power loss both computationally and experimentally using a purpose-built test rig at the UTC. In this study the test rig has been adapted such that chamber pressures up to 8 bar can be generated. Test data has been obtained that shows the effect on WPL of chamber pressure, advancing understanding of the relationship between data obtained at ambient and pressurised conditions. Three configurations have been studied: unshrouded gear, gear with 360° shroud and shrouded, crown and pinion meshing pair. Further, the effect of oil mist within the chamber on WPL has been studied. An oil mist generation system was developed for introducing a fine mist into the chamber and results are presented for varying mist flowrates. A mist measurement system was developed to sample mist mass fraction within the chamber and the data obtained is used to calculate an effective (oil/gas mixture) chamber density. Increasing chamber pressure increases Reynolds number, moving the system behavior further along the Moment coefficient-Reynolds number correlation. The Cm-Re correlation is similar in form to that for a shrouded cone, showing transitional behavior around Re = 2×106. Beyond transition Cm decreases with increasing Re. Introducing an oil spray has two effects: reduction in chamber temperature and increase in effective density of chamber fluid. Both effects can be accounted for by calculating Re and Cm based on mixture properties but it seems highly likely that the properties of the fluid under the shroud differ from those of the fluid in the external chamber.

Recently there has been a growing interest to develop innovative surgical needles for percutaneous interventional procedures. Needles are commonly used to reach target locations inside of the body for various medical interventions. The effectiveness of these procedures depends on the accuracy with which the needle tips reach the targets, such as a biopsy procedure to assess cancerous cells and tumors. One of the major issues in needle steering is the force during insertion, also known as the insertion (penetration) force. The insertion force causes tissue damage as well as tissue deformation. It has been well studied that tissue deformation causes the needle to deviate from its target thus causing an ineffective procedure. Simulation of surgical procedures provides an effective method for a robot-assisted surgery for pre- and intra-operative planning. Accurate modeling of the mechanical behavior on the interface of surgical needles and organs, specifically the insertion force, has been well recognized as a major challenge. Overcoming such obstacle by development of robust numerical models will enable realistic force feedback to the user during surgical simulation. This study investigates feasibility of predicting the insertion force of bevel-tip needles based on experimental data using neural network modeling. Simulation of the proposed neural network model is performed using Kera’s Python Deep Learning Library with TensorFlow as a backend. The insertion forces of needles with different bevel-tip angles in gel tissue phantom are measured using a specially designed automated needle insertion test setup. Input-output datasets are generated where the inputs are defined as bevel-tip angles and gel tissue phantom stiffness, and the output is defined as the insertion force. A properly trained neural network then maps the input data to the output data and the input-output dataset is supplied to train a neural network. Its performance is then evaluated using different and unseen input-output dataset. This paper shows that the proposed neural network model accurately predicts the insertion force.
