Pełny tekst źródłaLentinula edodes is an edible mushroom grown and marketed for centuries due to its nutritional and medicinal properties. Rich in protein, fiber, minerals and low in calories, Shiitake has mainly antimicrobial, antioxidant, anti-inflammatory, hypoglycemic and anticancer activities. Few studies have been done considering the influence of Shiitake growing conditions on biological activities. Our work focused on the comparison of the antioxidant and anti-inflammatory properties of Shiitake cultivated by different producers (from the French region Occitanie), i.e., A and C non-organic and B organic mushroom professionals, using an identical strain of Shiitake (Mycelia-3782). A sequential extraction under sonication (cyclohexane, chloroform, ethanol and water) was carried out on the fungal materials. The antioxidant capacity of the four extracts was evaluated using DPPH and ORAC. The anti-inflammatory activity of the ethanolic and aqueous extracts of all the producers was evaluated in vitro on a model of J774.A1 inflammatory macrophages. The results show that at 1 mg/ml, all the extracts have a quantifiable but moderate antioxidant activity with a more pronounced antioxidant potential in the aqueous extracts, whereas the polyphenol content assayed by the Folin Ciocalteu reagent was greater in cyclohexane extracts. At 50 μg/ml, the ethanolic extracts of the three producers strongly inhibit the production of the NO in a dose-dependent manner (29.66%, 31.30% and 27.56% for the producers A, B and C, respectively) when the cells are pre-treated for 4 hours with a LPS / IFNγ stimulation time of 24 hours without affecting cell viability. On the other hand, the inhibition observed for the aqueous extracts is very low. This activity was not due to a NO scavenging activity of the extracts. The results also showed that TNFα secreted in cell supernatants of macrophages pretreated with ethanolic extracts of Shiitake was inhibited in a concentration-dependent manner, in contrast to aqueous extracts which do not have an inhibitory effect on the molecule. This preliminary study shows that the growing conditions of Shiitake (organic and non-organic) do not influence its antioxidant and anti-inflammatory potentials in vitro

Pełny tekst źródłaAn increase in atmospheric CO2 levels has been observed since the beginning of the industrial era, which had a major impact on crops yield. Increase atmospheric CO2 concentration has been identified as a fertilizing agent that induces an alteration of the nutritional quality of plants. A decrease in proteins and minerals content has been observed. Such variation in nutritional value of crops might conduct to the development of metabolic disorders and the disruption of antioxidant and antiinflammatory defense leading to the increased incidence of non-communicable diseases (NCDs) such as "metabolic syndrome".The objective of this work is to evaluate the effect of atmospheric CO2 enrichment on the nutritional and health quality of two types of tomatoes (Solanum lycopersicum): MicroTom, a dwarf variety that has been extensively studied as model plant and the big tomatoes represented by the parents of the MAGIC population (that resumes the genetic diversity existing in cultivated tomatoes). The determination of macronutrients such as sugars, lipids and proteins followed by the quantification of micronutrients such as carotenoids and minerals allowed us to establish a complete nutritional profile confirming the degradation of the mineral and protein composition accompanied by an accumulation of sugars and a modification of bioactive compounds. A more profound investigation was the evaluation of the antioxidant and anti-inflammatory activity on tomato extracts to better understand the influence of this modification on human nutrition and biological defenses. Our results highlighted that the MicroTom model showed a marked modification of its biochemical profile. The study of antioxidant activity on this model revealed an alteration that can be explained by the interaction between culture conditions and atmospheric CO2 enrichment while the anti-inflammatory activity seems to be unchanged.The second MAGIC model confirmed the results obtained with MicroTom, thus it validated that an alteration of the biochemical composition of the tomato did not show a negative effect on the antioxidant and anti-inflammatory defense. All these observations on MicroTom and MAGIC allowed us to make the link between the atmospheric CO2 enrichment and human health and to give suggestions to better orient future research in this field

Pełny tekst źródłaBlueberry demonstrates therapeutic potential against cancer, cardiovascular and neurodegenerative diseases. These desirable effects are thought to involve abundant phenolic compounds in blueberry such as anthocyanins and flavonoids. Blueberry juice biotransformation by Serratia vaccinii bacteria increases its phenolics content, its antioxidant activity and modifies its physiological properties. The purpose of the present study was to evaluate the neuroprotection activity and the antidiabetic potential of biostranformed blueberry juice (BJ). BJ was studied for: 1) Protection of neurons (N2a) against oxidative stress caused by hydrogen peroxide; 2) Stimulation of glucose uptake by muscle cells (C2C12) and adipocytes (3T3-L1); 3) Anti-hyperglycemic activity in diabetic obese KKAy mice. Indeed, whereas normal blueberry juice did not show any effect, BJ increased antioxidant enzymes activities (catalase, SOD) and protected neurons against modifications in MAPKs pathways and toxicity induced by hydrogen peroxide. BJ also increased glucose uptake by 48% in C2C12 cells and by 142% in 3T3-L1 cells. Enhanced glucose uptake could not be explained by an increase of cytosolic calcium but was related to the stimulation of AMPK phosphorylation. Moreover, BJ inhibited 3T3-L1 adiopogenesis. BJ also reduced hyperglycemia of obese diabetic KKAy mice and protected pre-diabetic young mice against the development of obesity and diabetes mellitus. The anti-hyperglycemic effect of BJ may involve adipokine pathways since BJ increased adiponectin levels in diabetic mice. Thus, BJ represents a promising approach for the prevention and treatment of diabetes mellitus and possibly neurogenerative diseases as well as a source of novel therapeutic agents against these disorders.
