The experiment was conducted during rabi season 2019-2020 at Rangpur and Kurigram districts under Agricultural Research Station, On Farm Research Division, Alamnagar, Rangpur to promote and disseminate newly released potato variety, BARI Alu-35, BARI Alu-36, BARI Alu-37, BARI Alu-40 and BARI Alu-41among the potato growers of Rangpur sadar Upzilla in Rangpur and Chilmari Upazilla in Kurigram. The experiment was arranged in a randomized complete block design (RCBD) with four dispersed replications. The treatments included T1: BARI Alu-35, T2: BARI Alu-36, T3: BARI Alu-37, T4: BARI Alu-40 and T5: BARI Alu-41. BARI Alu-41 showed excellent performance and higher yield followed by BARI Alu-40 and BARI Alu-36. Farmers were happy to observe the performance of the varieties and demanded quality seed for next year cultivation. The highest common scab infection was observed in (BARI Alu-35) (1.62%) where the lowest in BARI Alu-41. Maximum virus infected was found in BARI Alu-40 (2.29%), where the lowest infection was observed in BARI Alu-41 (1.22%). Progressive Agriculture 32 (1): 17-21, 2021

An on-farm trial with four orange fleshed sweet potato varieties viz. BARI Mishti Alu-4, BARI Mishti Alu-8, BARI Mishti Alu-12, BARI Mishti Alu-13 including local check (Muli Alu) were evaluated at farming system research and development (FSRD) site, Jalalpur, Sylhet, Bangladesh during rabi season for two consecutive years (2016-2017 & 2017-2018) under Surma‐Kushiyara Floodplain of Bangladesh. The experiment was laid out in a randomized complete block design (RCBD) with six dispersed replications. The results revealed that the genotypes varied considerably in tuber root yields and yield attributing characters. Considering the average tuber root yield performance orange fleshed sweet potato varieties performed better than local variety (Muli Alu). However, among the varieties under studied BARI Mishti Alu-12 produced higher roots yield (40.63 t ha-1) than the other tested varieties. Considering economic return, the highest gross margin (Tk. 5,65,110 ha-1) and benefit cost ratio (5.50:1.00) was also obtained from sweet potato var. BARI Mishti Alu-12 and the lowest returnfrom var. BARI Mishti Alu-4 with benefit cost ratio of 3.54. Though the local cultivar (Muli Alu) produced lower root yields but it provided considerable gross margin (Tk.3,76,240 )and BCR (3.99:1.00 ) which closed to BARI Mishti Alu-12. It is due to higher demand of local cultivar with high price in the local market. Therefore, the genotypes BARI Mishti Alu-12 and local cultivar (Muli Alu) were found suitable for their better growth and yield under the acidic soils Bangladesh Agron. J. 2018, 21(2): 59-65

An experiment was conducted during 20 November 2015 through March 20I6 to assess the effect of planting dates on growth and yield performance of three potential varieties of potato at Plant Physiology research field, Bangladesh Agricultural Research Institute, Gazipur. Three planting dates (November 20, December 5 and December 29) and three varieties (var. BARI Alu-35, BARI Alu-40 and BARI Alu-41) were the treatment variables. The experiment was laid out in a randomized complete block design with three replications. Maximum plant height (42.3 cm) was observed in 5 December sowing in var. BARI Alu-40. Highest number of tuber/plant (13) was recorded from December 5 sowing in var. BARI Alu-41. The highest leaf area was found in December 5 sowing of BARI Alu-40 while the lowest leaf area in November 20 sowing in var. BARI Alu-35. The highest tuber weight/plant (97.25 g) was observed in var. BARI Alu-5 at December 5 sowing and the lowest tuber weight/plant (25.58 g) in var. BARI Alu-35 at November 20 sowing. The maximum potato yield (42.12 t/ha) was obtained at December 5 sowing of BARI Alu-35 followed by same date of var. BARIAlu- 41. From the experiment it was revealed that the first decade of December is the optimum date for planting of potato due to the physiological maturity and tuber yield.Bangladesh Agron. Due to the difficulties associated with scaling of silicon transistors, various technologies beyond binary logic processing are actively being investigated. Ternary logic circuit implementation with carbon nanotube field effect transistors (CNTFETs) and resistive random access memory (RRAM) integration is considered as a possible technology option. CNTFETs are currently being preferred for implementing ternary circuits due to their desirable multiple threshold voltage and geometry-dependent properties, whereas the RRAM is used due to its multilevel cell capability which enables storage of multiple resistance states within a single cell. This article presents the 2-trit arithmetic logic unit (ALU) design using CNTFETs and RRAM as the design elements. The proposed ALU incorporates a transmission gate block, a function select block, and various ternary function processing modules. The ALU design optimization is achieved by introducing a controlled ternary adder–subtractor module instead of separate adder and subtractor circuits. The simulations are analyzed and validated using Synopsis HSPICE simulation software with standard 32 nm CNTFET technology under different operating conditions (supply voltages) to test the robustness of the designs. The simulation results indicate that the proposed CNTFET-RRAM integration enables the compact circuit realization with good robustness. Moreover, due to the addition of RRAM as circuit element, the proposed ALU has the advantage of non-volatility.

This research paper deals with design and implementation of low power 8-bit arithmetic logic units. The main part of power consumption is consumed in ALU in any processor. Therefore, reducing power dissipation in ALU should be requiring. The proposed technique disabled one of the main block of ALU using tri-state logic which is not necessary to use, except the required processes. In this work, the suggested design is realized by using ASIC methodologies. In order to implement the arithmetic and logic architectures, 130 nm standard cell libraries are used for ASIC execution. The architecture of the design has been created using Verilog HDL language. In addition, it is simulated using ModelSim-Altera 10.3c (Quartus II 14.1) tools. Arithmetic and logical unit (ALU) are the core operational programmable logic block in microprocessors, microcontrollers, and real-time-integrated circuits. The conventional ALUs were developed using complementary metal oxide semiconductor (CMOS) technology, which resulted in excessive power consumptions, path delays, and number of transistors. Therefore, this article focuses on the design and development of hybrid delay-controlled reconfigurable ALU (DCR-ALU) using field-effect transistor (FinFET) and graphene nanoribbon field-effect transistor (GnrFET) technologies. Initially, a novel carry output predictable full adder (COPFA) and carry input selectable full adders (CISFA) are developed using multiplexer selection logic; then, delay-controlled hybrid adder (DCHA) and delay-controlled hybrid subtractor (DCHS) are developed using these full adders. In addition, a unified delay-controlled hybrid adder and subtractor (DCHAS) is developed by combining these DCHA and DCHS. Further, a delay controller array multiplier (DCAM) also developed using DCHA modules. Finally, DCR-ALU is developed by adopting the DCHAS, DCAM, and logical operations. The obtained simulation results shows that the proposed nanotechnology-based models outperformed the conventional adders and subtractors in terms of area, power, and delay reduction.

Quantum-dot Cellular Automata (QCA) based reversible logic is the utmost necessity to achieve an architecture at nano-scale, which promises extremely low power consumption with high device density and faster computation. This paper emphasises on the design of an efficient reversible Arithmetic Logical Unit (ALU) block in QCA technology. We have considered [Formula: see text] RUG (Reversible Universal Gate) as the basic unit, and also report a HDLQ model for RUG with 52.2% fault tolerance capability. Further, the reversible ALU has synthesized with reversible logic unit (RLU) and reversible arithmetic unit (RAU). We also demonstrate QCA implementation of RLU and RAU with lesser complexity and cell counts. The proposed ALU needs only 64 MVs (Majority Voters), which demonstrates 40% optimizations in the majority gate counts than the existing result. QCADesigner-2.0.3 is used to verify the proposed designs.

The application of biochar may enhance the yield of potato for different processing categories. A field experiment was conducted at Sher-e-Bangla Agricultural University, Dhaka-1207, during the period from November, 2020 to April, 2021 to find out the response of biochar on yield of potato for different processing categories. The experiment comprised of Potato varieties (3): V1: BARI Alu-29 (Courage), V2: BARI Alu-28 (Lady Rosetta) and V3: BARI Alu-25 (Asterix) and Biochar level (5): B0: 0 t ha-1, B1: 2.50 t ha-1, B2: 5.00 t ha-1 and B3: 7.50 t ha-1 and B4: 10 t ha-1. The study was laid out in a randomized complete block design with 3 replications. The results showed that biochar amendment could enhance the yield of processing category potato. The total yield and marketable yield of potato gradually increased with increasing biochar level. The results also revealed that the processing category potato viz., canned, chips and French fry potato yield progressively increased with advancing biochar level irrespective of varieties except dehydrated category. In case of marketable yield, BARI Alu-25 and BARI Alu-29 with biochar level 5 to 10 t ha-1performed superior than other combinations and produced 19.50 to 21.30 t ha-1which are 18.54 to 36.45% higher than without biochar. The combination of V2B4 produced maximum canned (8.10 t ha-1) and dehydrated potato (10.09t ha-1) but V3B4 made significantly highest chips (9.03 t ha-1) and French fry (5.70 t ha-1) potato, whereas, BARI Alu-29 and BARI Alu-28 did not produce any French fry category potato. However, the level of biochar of 5 to 10 t ha-1 could enhance processing category potato production. It may be concluded that potato growers may apply biochar along with recommended rate of other fertilizers for producing maximum processing category potato. Bangladesh Agron. J. 2021, 24(2): 63-72

This study examines how different initial design decisions affect the area, timing, and power of technology-mapped designs. ASIC design flow, tools used during the flow, and the factors to consider to maximize the performance and power ratio are discussed. The ALU (Arithmetic Logic Unit) is a fundamental part of all processors. In this study, two ALUs were implemented using two different types of adder circuits: a Ripple Carry Adder (RCA) and a Sklansky adder. The Cadence EDA tools were used for the implementation. A comparative analysis was conducted for the two designed ALUs in terms of area, power, and timing analysis. The ALU design was also used as an example to examine the whole workflow front-end wise by constructing a block schematic and back-end wise by floorplanning, placing, and routing the physical design.

L’objective est d’améliorer l’activité photocatalytique de TiO2 sous irradiations UV et Visible. Pour contourner les limites de TiO2 intrinsèque nous envisageons une fabrication de nanocomposite plasmonique à base de nanofils de TiO2 périodiquement organisés et assemblés avec des nanoparticules plasmoniques. Pour la fabrication des nanofils de TiO2 mécaniquement stables, deux approches ont été réalisées. La première approche est basée sur la croissance sélective en phase vapeur, la deuxième approche consiste en l’utilisation d’un moule de membranes AAO et d’un dépôt de films conformes par ALD. En parallèle les films de TiO2déposés par ALD sont assemblés avec les nanoparticules plasmoniques d’or. Les différentes architectures de TiO2 sont valorisées par des tests photocatalytiques (UV et Visible) sur les polluants modèles. Une nouvelle approche de la fabrication des films mesoporeux d’H-TiO2 avec efficacité photocatalytique à la fois sous irradiation UV et Visible est développée
The objective of this thesis is to improve the photo-response of well-known photocatalytic material such as TiO2, which is usually only active in the UV range. The basic idea is to assemble several approaches within one device to improve the photocatalytic properties: fabrication of periodically-organised TiO2 nanostructures and their assembly with plasmonic nanoparticles. Two fabrication strategies were investigated for these purposes. The first approach consists of selective vapour phase growth. The second approach implements the use of an AAO template. In parallel, TiO2 films deposited by ALD and assembled with plasmonic gold nanoparticles are investigated. The photocatalytic measurements on various TiO2 architectures were performed in both irradiation ranges UV and Vis. A new fabrication approach of mesoporous H-TiO2 films was developed giving promising results of photocatalytic efficiency improvement in both UV and Visible ranges

Lucy Addison High School was an all-Black high school located in Roanoke, Virginia. All-black high schools are defined in this study as high schools that were segregated by race and attended only by Black students. Lucy Addison operated as an all-Black high school from 1928 until 1970 in two different buildings. Roanoke's secondary schools were desegregated in 1963. Addison was integrated in the fall of 1970 and closed in 1973. The purpose of the study was twofold. The primary purpose was to document the experiences of the graduates of the classes of 1963-70 of Lucy Addison High School. The secondary purpose was to determine if the supports found in the research literature about all-Black high schools prior to desegregation were present in the Lucy Addison students' experiences during the years between desegregation and integration. The supports are (a) the importance of a spiritual foundation, (b) high expectations from school administrators and teachers, and (c) parent and community support. Six common themes emerged from the interviews with participants. They were: (a) the importance of a spiritual foundation, (b) high expectations from teachers and administrators, (c) parent and community support, (d) school leadership, (e) attitudes on segregation and integration, and (f) school pride. These themes helped answer the four research questions that guided the study. After conducting interviews with the graduates, their accounts confirmed that the supports identified in the literature regarding all-Black high schools were present in their school experiences. The importance of a spiritual foundation, high expectations from teachers and administrators, and parent and community support could easily be seen in the experiences of all 16 students who attended Lucy Addison High School from 1963-1970. Upon reflection on the study, the researcher made certain recommendations for further study. The recommendations for further study revolve around the continued documentation of experiences of Lucy Addison High School students, conducting a study of Lucy Addison High School as an integrated school from 1970 to 1973, and assessing the reason why Lucy Addison High School was allowed to stay open as an integrated high school.

Antiangiogenic drugs are used clinically for treatment of different types of cancers and in the case of renal cell carcinoma (RCC) are now standard first-line treatments (Rini, 2009). Nevertheless, these agents mainly serve to stabilize the disease but are not able to eliminate all tumor cells and resistance eventually develops concomitant with progression (Kerber and Folkman, 2002). Using different orthoxenograft mouse models of RCC we confirmed that inhibitors of the VEGF pathway have a therapeutic window of effectiveness but unfortunately adaptation and tumor relapse always occur. Several different mechanisms of resistance to antiangiogenics have been already described, many concerning the activation of compensatory signals that produce re-vascularization and tumor re-growth. (Bergers and Hanahan, 2008). In our study we determined that even if resistance is characterized by the up-regulation of the pro-angiogenic enzyme ECGF1 (previous data from the lab); re-vascularization does not occur, the vascular trimming is maintained by the treatment pointing to an alternative mode of adaptation. Using two different approaches, we demonstrated that PD-ECGF1 is responsible of the acquisition of resistance to anti-anigiogenics. In fact, its enzymatic inhibition as second-line treatment post resistance resulted in the arrest and stabilization of tumor growth. PD-ECGF1 inhibition did not increase the anti-vascular effect of DC101, confirming that resistance was vessel independent, but dramatically affected tumor cell proliferation and apoptosis. Considering the numerous proprieties ascribed to its final metabolite 2-deoxy-D-ribose (Ikeda et al., 2006; Bjinsdorp et al., 2008), we evaluated its role, finding that recombinant 2-deoxy-D-ribose nullified the effect of AEAC on 786O- and Ren28 tumor growth rescuing tumor cell proliferation and apoptosis, without promoting re-vascularization. Overall these findings, confirmed also in an in vitro setting, demonstrate that tumor stroma plays a minor role in this model of anti-angiogenics resistance, and that PD-ECGF1 acts mainly intracellularly supporting tumor cell proliferation and protecting from apoptosis by its enzymatic activity and final metabolite 2-deoxy-D-ribose. Using a genetic approach in which PD-ECGF1 protein expression was silence as a second line of treatment post DC101-resistance, mimicking what done pharmacologically, we confirmed and improved the anti-tumoral response described, without perceiving any doxycycline toxic effect. In fact, PD-ECGF1 genetic knock down resulted in a complete arrest of tumor progression enhancing the merely partial stabilization produce by AEAC treatment in 786O- tumor bearing mice. Unfortunately VEGFR-blockers can’t be used in vitro on cancer cells, and to verify this hypothesis we decided to mimic the final effects of anti-angiogenic treatments, finding that under nutrient deprivation conditions cancer cells significantly up-regulate PD-ECGF1 expression, and metabolizing thymidine acquired considerable growth advantages. Finally, the analysis of plasma and tissue samples of ccRCC patients from the Bellvitge Hospital confirm in a clinical set that PD-ECGF1 is exclusively found in pathologic conditions, where if highly expressed correlates with poor prognosis, suggesting that might represent a good therapeutic predictor factor. Moreover, the analysis of tissues biopsies before and after anti-angiogenic treatment revealed that whilst PD-ECGF1 treatment-induce up-regulation was a common feature, the patients that unfortunately didn’t respond showed the sharp difference, confirming PD-ECGF1 as a possible therapeutic target.
Los fármacos antiangiogénicos se usan clínicamente para el tratamiento de diferentes tipos de cáncer y en el caso del carcinoma de células renales (CCR) representan la primera diana terapéutica (Rini, 2009). Sin embargo, no son capaces de eliminar todas las células tumorales y muchos pacientes desarrollan resistencia al tratamiento y recrecimiento tumoral (Kerber y Folkman, 2002). Mediante el uso de diferentes modelos de ratón hemos confirmado que los inhibidores de la vía VEGF tienen una limitada ventana terapéutica de efectividad, seguida desafortunadamente de adaptación y recaída tumoral que en nuestro estudio esta` caracterizada por la regulación positiva de la enzima PD-ECGF1. Usando dos enfoques diferentes, demostramos que PD-ECGF1 es responsable de la adquisición de resistencia a anti-anigiogénicos. De hecho, su inhibición enzimática como tratamiento de segunda línea después de la resistencia dio como resultado la detención y la estabilización del crecimiento tumoral. La inhibición de PD-ECGF1 no aumentó el efecto antivascular de DC101, pero afectó de manera dramática la proliferación y apoptosis de las células tumorales. Teniendo en cuenta las numerosas propiedades atribuidas a su metabolito final 2-deoxy-D-ribose (Ikeda et al., 2006; Bjinsdorp et al., 2008), evaluamos su papel, encontrando que la 2-deoxy-D-ribose anuló el efecto de la inhibición de PD-ECGF1 rescatando el crecimiento tumoral. Experimentos en vitro demostraron como las células tumorales incrementaban la expresión de PD-ECGF1 en condiciones de falta de nutrientes haciéndonos especular que esta proteína tenga un papel en la adaptación metabolica. Finalmente, el análisis de muestras de plasma y tejido de pacientes con ccRCC del Hospital de Bellvitge confirma en un conjunto clínico que PD-ECGF1 se encuentra exclusivamente en condiciones patológicas, donde se correlaciona con mal pronóstico, sugiriendo que podría representar un buen factor predictor terapéutico.

Abstract In this paper, we will show that it is highly beneficial to model dual-porosity reservoirs using matrix refinement (similar to the multiple interacting continua, MINC, of Preuss, 1985) for water displacing oil. Two practical situations are considered. The first is the effect of matrix refinement on the unsteady-state pressure solution, and the second situation is modeling water-oil, Buckley-Leverett (BL) displacement in waterflooding a fracture-dominated flow domain. The usefulness of matrix refinement will be illustrated using a three-node refinement of individual matrix blocks. Furthermore, this model was modified to account for matrix block size variability within each grid cell (in other words, statistical distribution of matrix size within each grid cell) using a discrete matrix-block-size distribution function. The paper will include two mathematical models, one unsteady-state pressure solution of the pressure diffusivity equation for use in rate transient analysis, and a second model, the Buckley-Leverett model to track saturation changes both in the reservoir fractures and within individual matrix blocks. To illustrate the effect of matrix heterogeneity on modeling results, we used three matrix bock sizes within each computation grid and one level of grid refinement for the individual matrix blocks. A critical issue in dual-porosity modeling is that much of the fluid interactions occur at the fracture-matrix interface. Therefore, refining the matrix block helps capture a more accurate transport of the fluid in-and-out of the matrix blocks. Our numerical results indicate that the none-refined matrix models provide only a poor approximation to saturation distribution within individual matrices. In other words, the saturation distribution is numerically dispersed; that is, no matrix refinement causes unwarranted large numerical dispersion in saturation distribution. Furthermore, matrix block size-distribution is more representative of fractured reservoirs.

This paper proposes a block hash (BH) method for efficiently storing multipleIndustry Foundation Classes (IFC) models in the database. We find that there are lots of duplicationswhenwe store different models or different versions of a modelinto the databasebecause of the temporal-spatial correlation of data between these models. The main idea of our approach is to divide these model files into appropriate blocks andto calculate the hash valuesof theseblocks to reuse the them in different models. These blocks should not be too small in case too many nodes need to be compared, nor should they be too large in case it is difficult to find identical blocks to share between different models.So the BH method is proposed to efficiently make the database redundancy-free. For the experiments we use multi-versions of multiple models of a same project to validate this method.The experimental results showthat our method is practicable and efficient.

Blockchain-based cryptocurrency has attracted the immersive attention of individuals and businesses. With distributed ledger technology (DLT) consisting of growing list of record blocks and securely linked together using cryptography, each block contains a cryptographic hash of the previous block, a timestamp, and transaction data. The timestamp proves that the transaction data existed when the block was created. Since each block contains information about the block previous to it, they effectively form a chain, with each additional block linking to the ones before it. Consequently, blockchain transactions are irreversible in that, once they are recorded, the data in any given block cannot be altered retroactively without altering all subsequent blocks. The blockchain-based technologies have been emerging with a fleet speed. In this paper, the trustworthy Artificial Intelligence will be explored for blockchain-based cryptocurrency where the prohibitive price leap creates a challenge for financial analysis and prediction.

There are many PE(Pre-Erection) Blocks in open areas of the shipyard, which are supported by the wooden, concrete, or steel supports. Their position and numbers are decided on a basis of the workers’ experiences. Recently, many shipyards have been making PE blocks with various shape and weight distribution because of the variety of kinds of ships and their building technology. Seriously, they are treating blocks which have not been experienced. In order to evaluate the reliability of all support plans of PE Blocks in such situation, many man hours (MH) are consumed, and the special knowledge and technology about structural analysis are required. We proposed how to conveniently and quickly evaluate the structural safety on PE block supports and developed an application system to implement the evaluation process with three dimensional part models of the PE block and their weight information. The evaluation is based on the simplification to a grillage model of a PE block and its grillage analysis. The load distribution on the grillage model is calculated by two approaches. The first is that the load distribution on the grillage nodes are estimated with the real weight and the center of gravity of each part of a PE block, which can be provided from design databases of the shipyard. The other is that the load distribution is optimally estimated only with the weight and the center of gravity of sub blocks of a PE block. The latter is useful in the case that block information of mother ships can be obtained without the detail design of the PE block. The integrated system has been comprehensively implemented in order to make the grillage model from the three dimensional CAD models of the PE block and their weight information, and to perform the grillage analysis for the reaction forces on the block supports. In the application program, the grillage model can be automatically built from CAD models of PE block. Also the grillage can be modified by inserting, splitting, and deleting a beam element.

The report considers structural-tectonic, geochemical and hydrodynamic condi-tions of preservation of the potential oil reservoir BC8 at the Ai-Yaunskoye field. Formation BS8 has a good reservoir in the area. The reservoir deposits are represented by a reservoir of marine genesis with high filtration-volume properties.Because of the faults, the formation has a block structure.The BS8 formation in the Ai-Yaun area is in the zone of potential oil-bearing strata. As of today, one of the blocks has opened an aquifer. The rest of the blocks have not yet been drilled. Therefore, the potential of these sediments is considered unexplored to the end. According to the analysis of the fault model, some of the faults are considered to be through. Hydrocarbons could have migrated along these faults up the section to the PK 1–2 reservoirs. Some of the faults are considered screens, which increases the chances of the BS8 formation discovering oil deposits in it in other tectonic blocks of the reservoir.

This report investigates and develops specifications for using blockchain and distributed organizations to enable decentralized delivery and finance of urban infrastructure. The project explores use cases, including: providing urban greening, street or transit infrastructure; services for street beautification, cleaning and weed or graffiti abatement; potential ways of resource allocation ADU; permitting and land allocation; and homeless housing. It establishes a general process flow for this blockchain architecture, which involves: 1) the creation of blocks (transactions); 2) sending these blocks to nodes (users) on the network for an action (mining) and then validation that that action has taken place; and 3) then adding the block to the blockchain. These processes involve the potential for creating new economic value for cities and neighborhoods through proof-of-work, which can be issued through a token (possibly a graphic non-fungible token), certificate, or possible financial reward. We find that encouraging trading of assets at the local level can enable the creation of value that could be translated into sustainable “mining actions” that could eventually provide the economic backstop and basis for new local investment mechanisms or currencies (e.g., local cryptocurrency). These processes also provide an innovative local, distributed funding mechanism for transportation, housing and other civic infrastructure.

The black carp (bc)GtH IIb cDNA was amplified and isolated, cloned and sequenced. Comparison of the bcGtH IIb deduced a.a. sequence with that of GtH IIb from other teleosts revealed high homology to cyprinid species and a lower homology to salmonid or perciform fish. The gene coding for the GtH IIb was isolated and sequenced. Three bc recombinant phages which hybridized to the goldfish GtH Ib cDNA probe were isolated and are currently being characterized. The region coding for the mature GtH IIb was expressed in a bacterial expression vector resulting in the production of a recombinant protein. In vitro folding resulted in a protein only 1.3% of which displaced the native common carp GtH II in a RIA. Therefore, the common carp GtH RIA was utilized for the physiological studies at the current phase of the project. Two non-functional sites were identified along the brain-pituitary gonadal axis in the immature black carp. The pituitary is refractory to GnRH stimulation due to a block proximal to the activation of PKA and PKC probably at the level of GnRH receptors. The gonads, although capable of producing steroids, are refractory to gonadotropic stimulation but do respond to cAMP antagonists, indicating a block at the GtH receptor level. Attempts to advance puberty in 2 and 3 y old black carp showed that testosterone (T) stimulates GtH synthesis in the pituitary and increases its sensitivity to GnRh. A 2 month treatment combining T+GnRH increased the circulating GFtH level in 3 y old fish. Addition of domperidone to such a treatment facilitated both the accumulation of GtH in the pituitary and its response to GnRH. The cDNA of striped bass GtH a, Ib and IIb subunits were amplified, isolated, cloned and sequenced, and their deduced a.a. sequences were compared with those of other teleosts. A ribonuclease protection assay was developed for a sensitive and simultaneous determination of all GtH subunits, and of b-actin mRNAs of the striped bass. GnRH stimulated dramatically the expression of the a and GtH IIb subunits but the level of GtH Ib mRNA increased only moderately. These findings suggest that GtH-II, considered in salmonids to be involved only in final stages of gametogenesis, can be induced by GnRH to a higher extent than GtH-I in juvenile striped bass. The native GtH II of the striped bass was isolated and purified, and an ELISA for its determination was developed. The production of all recombinant striped bass GtH subunits is in progress using the insect cell (Sf9) culture and the BAC-TO-BAC baculovirus expression system. A recombinant GtH IIb subunit has been produced already, and its similarity to the native subunit was confirmed. The yield of the recombinant glycoprotein can reach 3.5 mg/ml after 3 days culture. All male striped bass reach puberty after 3 y. However, precocious puberty was discovered in 1 and 2 y old males. Females become vitellogenic during their 4th year. In immature 2 y old females, T treatment elevates the pituitary GtH II content while GnRH only potentiates the effect. However, in males GnRH and not T affects GtH accumulation in the pituitary. Neither GnRH, nor T treatment resulted in gonadal growth in 2 y old striped bass, indicating that either the accumulated GtH II was not released, or if released, the gonads were refractory to GtH stimulation, similar to the situation in the immature black carp. In 3 y old female striped bass, 150 day GnRHa treatment resulted in an increase in GSI, while T treatment, with or without GnRHa, resulted in a decrease in oocyte diameter, similar to the effect seen in the black carp. Further attempts to advance puberty in both fish species should take into account the positive effect of T on pituitary GtH and its negative effect of ovarian growth.

The Environmental Laboratory (EL) and the Coastal and Hydraulics Laboratory (CHL) have jointly completed a number of large-scale hydrodynamic, sediment and water quality transport studies. EL and CHL have successfully executed these studies utilizing the Geophysical Scale Transport Modeling System (GSMB). The model framework of GSMB is composed of multiple process models as shown in Figure 1. Figure 1 shows that the United States Army Corps of Engineers (USACE) accepted wave, hydrodynamic, sediment and water quality transport models are directly and indirectly linked within the GSMB framework. The components of GSMB are the two-dimensional (2D) deep-water wave action model (WAM) (Komen et al. 1994, Jensen et al. 2012), data from meteorological model (MET) (e.g., Saha et al. 2010 - http://journals.ametsoc.org/doi/pdf/10.1175/2010BAMS3001.1), shallow water wave models (STWAVE) (Smith et al. 1999), Coastal Modeling System wave (CMS-WAVE) (Lin et al. 2008), the large-scale, unstructured two-dimensional Advanced Circulation (2D ADCIRC) hydrodynamic model (http://www.adcirc.org), and the regional scale models, Curvilinear Hydrodynamics in three dimensions-Multi-Block (CH3D-MB) (Luong and Chapman 2009), which is the multi-block (MB) version of Curvilinear Hydrodynamics in three-dimensions-Waterways Experiments Station (CH3D-WES) (Chapman et al. 1996, Chapman et al. 2009), MB CH3D-SEDZLJ sediment transport model (Hayter et al. 2012), and CE-QUAL Management - ICM water quality model (Bunch et al. 2003, Cerco and Cole 1994). Task 1 of the DOER project, “Modeling Transport in Wetting/Drying and Vegetated Regions,” is to implement and test three-dimensional (3D) wetting and drying (W/D) within GSMB. This technical note describes the methods and results of Task 1. The original W/D routines were restricted to a single vertical layer or depth-averaged simulations. In order to retain the required 3D or multi-layer capability of MB-CH3D, a multi-block version with variable block layers was developed (Chapman and Luong 2009). This approach requires a combination of grid decomposition, MB, and Message Passing Interface (MPI) communication (Snir et al. 1998). The MB single layer W/D has demonstrated itself as an effective tool in hyper-tide environments, such as Cook Inlet, Alaska (Hayter et al. 2012). The code modifications, implementation, and testing of a fully 3D W/D are described in the following sections of this technical note.
