Abstract We report on ephemerides for the satellites of Pluto based on the large set of astrometric measurements. Our orbit fit yielded the following masses with 1σ uncertainties: GM Pluto = 869.3 ± 0.4 km3 s−2, GM Charon = 106.1 ± 0.3 km3 s−2, GM Nix = 1.50 ± 0.52 × 10−3 km3 s−2, GM Hydra = 2.01 ± 0.27 × 10−3 km3 s−2, corresponding to the densities of ρ Pluto = 1.853 ± 0.004 g cm−3, ρ Charon = 1.705 ± 0.006 g cm−3, ρ Nix = 0.88 ± 0.31 g cm−3, and ρ Hydra = 1.21 ± 0.19 g cm−3. Masses of Kerberos and Styx remain unconstrained, and it is unlikely that we will be able to measure them even if we obtain abundant 1 mas precision astrometry during the next 20 yr. We summarize the results of orbit integration in terms of osculating and precessing ellipse model mean elements. All satellites reside in near-circular orbits, and Kerberos and Styx have 0.4 deg and 0.3 deg inclinations with respect to Charon’s orbit plane. The nodal regression periods for Kerberos and Hydra are ∼9 and ∼14 yr respectively. We found that Charon’s orbit pole can be approximated as: R.A. = 133.0073 + 0.0036T deg, and decl. = –6.2449 + 2.5 × 10−5 T deg, where T is Julian centuries from the epoch J2000, based on 5000 yr of orbit integration.

Purpose: Previous studies have shown a positive correlation between amount of aortic valve calcification (AVC) and degree of aortic valve stenosis (AVS). We have investigated whether calcium scoring of AVC from contrast-enhanced images is reliable. Material and Methods: Nineteen patients with suspected AVS underwent retrospectively ECG-gated multislice computed tomography (MSCT). Standardized scan protocols were applied prior to (120 KV, 133 mAseff) and after (120 KV, 500 mAseff) the administration of non-ionic contrast material. Image reconstruction was performed at 60% of the RR interval (slice thickness 3 mm, reconstruction increment 2 mm). AVC was quantified using Agatston score and calcium mass. The number of lesions was calculated. All non-enhanced images were scored using thresholds of 130 HU and 350 HU. Contrast-enhanced images were assessed with a threshold of 350 HU exclusively. Results: Fifteen patients with AVCs were included in the statistical analysis. The mean Agatston score (calcium mass) in non-enhanced images was 2888.4±2844.4 (694.2 mg±869.3 mg). Altering the threshold from 130 HU to 350 HU led to a 58.2% (30.5%) decrease in the AVC score ( P values <0.001). Contrast-enhanced images showed an increased Agatston score (calcium mass) of 56.2% (33.5%) compared to non-enhanced images ( P values <0.05) with the same threshold of 350 HU. Conclusion: Quantification of AVC from contrast-enhanced images is not reliable, as contrast material simulates calcification.

7027 Background: Variable responses to imatinib in the treatment of myelogenous leukemia (CML) are incompletely understood. Previous studies focused on cellular mechanisms of resistance to imatinib. Another hypothesis for variable response lies in pharmacokinetic variability that may reduce drug exposure in patients receiving imatinib. Methods: Using high- performance liquid chromatography-tandem mass spectrometry, we assessed trough plasma imatinib concentrations at steady state (Cmin) in more than 150 CML patients. Major molecular response (MMR) was defined as a 3 logarithm reduction of BCR-ABL transcripts, quantified from peripheral blood using Q-RTPCR. In addition, suboptimal response criteria defined by the European Leukemianet were used to increase doses of imatinib. Results: i) we assessed Cmin in 85 CML patients and demonstated a correlation with imatinib doses (median values 813, 1,135, 1,709 ng/ml for respectively 300,400, 600 mg/day). ii) for 68 patients after at least one year's treatment mean Cmin were significantly higher in the group with MMR (34 patients) than in the group without (1,452.1±649.1 ng per milliliter vs. 869.3±427.5 ng per milliliter, P<0.001) whereas there was no difference in the imatinib daily dose. For Cmin and their discrimination potential for MMR, the area under receiver-operating characteristic curve was 0.775, with best sensitivity (76.5 percent) and specificity (70.6 percent) at a plasma threshold of 1,002 ng per milliliter. iii) dose escalation of imatinid, 400mg up to 600mg was proposed in 8 patients in suboptimal response (3 according to cytogenetic criteria and 5 according to molecular criteria). The median Cmin increase from 573 ng/ml [181–1,376] to 1,139 ng/ml [733–2,262] and 5 patients had a cytogenetic (n=3) or a molecular (n=2) response. Conclusions: Monitoring of imatinib plasma levels could be very useful for the management of CML patients, or should at least be checked in the case of treatment failure or suboptimal response or in patients nonadherent with imatinib or with a drug-drug interaction suspicion. Thermoelectric ceramics are those capable of converting thermal into electricity energy by the Seebeck effect. The non-conventional synthesis of ceramic powders and the doping of these powders with rare earth elements are factors that can improve the efficiency in energy conversion. The objective of this work was to synthesize and analyze pure and doped SrTiO3 powders by solid state reaction (SSR) and ultrasonic synthesis (USS). The precursors of SSR were: strontium carbonate (SrCO3), titanium oxide (TiO2) and lanthanum and neodymium oxide (La2O3 and Nd2O3). They were mixed in a ball mill for 3.5h in stoichiometric ratio to obtain SrTiO3 (STO), Sr0.96La0.04TiO3 (STO-04La), Sr0.96Nd0.04TiO3 (STO-04Nd) and Sr0.96La0.02Nd0.02TiO3 (STO-02La02Nd). The powder mixtures were calcined at 1000°C in a conventional oven for 2h. The USS powders were produced by solubilization of Ti (C4H9O)4 and SrCO3 in an HNO3 aqueous solution. The solution was precipitated with NH4OH and, then, this powder was dried at 70°C for 24h, in air. The dried powder was added to a solution of KOH and subjected to ultrasound irradiation. The same procedure was followed for the synthesis of USS doped powders, STO-04La, STO-04Nd, and STO-02La02Nd powders using as dopant lanthanum and neodymium oxides. The powders were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM) and crystallite sizes were calculated by the Scherrer equation to evaluate the differences on structural and microstructural properties.

Biodiesel production from waste palm oil catalyzed by hierarchical ZSM-5 supported calcium oxide was studied. The activity of CaO increased after supported on h-ZSM-5 resulting an increase in conversion from 93.17% to 95.40%. A maximum conversion of 95.40% was achieved at 6 h reaction time, 3 wt.% catalyst amount, 12:1 methanol to oil molar ratio and 65 °C reaction temperature. The waste palm oil showed a high potential as a feedstock in biodiesel production in which there was no significant different in the conversion of fresh and waste palm oil. The properties of the obtained biodiesel required the limits of biodiesel specification according to ASTM D6751-08 and EN 14214 with the methyl ester content of 97.18%, the acid value of 0.24 mg KOH/g, the kinematic viscosity of 4.64 cSt and the density of 869.9 kg/m3.

Abstract Background Individuals with spinocerebellar ataxia type 3 (SCA3) present communication and swallowing disorders, and consequent deterioration in quality of life (QOL). Objective To evaluate the impact of a speech therapy rehabilitation program on the QOL of patients with SCA3. Methods All participants were randomly assigned to two groups, an intervention group receiving speech therapy (STG) and a control group (CG). The International Cooperative Ataxia Rating Scale scores were 32.4 ± 20.2, and the Scale for the Assessment and Rating of Ataxia scores were 11.8 ± 8.0. The intervention consisted of a 12-session speech therapy rehabilitation program with oral, pharyngeal, and laryngeal strengthening exercises—the so-called ATAXIA–Myofunctional Orofacial and Vocal Therapy (A-MOVT). They all were submitted to pre- and postintervention evaluations using the World Health Organization's Quality of Life (WHOQOL-BREF) assessment, as well as the Living with Dysarthria (LwD), Quality of Life in Swallowing Disorders (SWAL-QOL), and Food Assessment Tool (EAT-10). Results The study sample consisted of 48 patients with SCA3 (STG = 25; CG = 23), mean age was 47.1 ± 11.4 years; mean age at symptom onset was 36.9 ± 11.3 years; disease duration was 11.9 ± 13.3 years. After the 3-month intervention, there were significant changes in the QOL in the STG compared with the CG, when assessed by the LwD (179.12 ± 62.55 vs. 129.88 ± 51.42, p < 0.001), SWAL-QOL (869.43 ± 153.63 vs. 911.60 ± 130.90, p = 0.010), and EAT-10 (5.16 ± 7.55 vs. 2.08 ± 3.85, p = 0.018). Conclusions Patients with SCA3 should receive continuous speech therapy as part of the A-MOVT program, because therapy helps to improve difficulty swallowing and dysarthria.

Abstract2,3,4,4-Tetrachloro-1-oxo-1,4-dihydronaphthaline, ß-TKN, crystallizes in the orthorhombic space group D1 6 2 h - Pnma with Z = 4, and the 35Cl NQR spectrum consists of three lines with an intensity ratio of 2:1:1. The crystal structure of 2,2,3,4-tetrachloro-1-oxo-1,2-dihydronaphthaline, α-TKN, is described in literature as belonging to the polar space group C22 - P 21 with Z = 2. In contradiction to the reported space group and Z, we observed a three line 35Cl NQR spectrum with an intensity ratio of 2:1:1. Therefore, the crystal structure of α-TKN was redetermined at 7 = 296 K: space group C22h-P21/m, Z = 2, a = 888.4 (2) pm, b = 692.3 (2) pm, c = 869.6 (2) pm, β = 91.483 (5)°. The nuclear quadrupole coupling tensors have been investigated using a 4 π Zeeman NQR goniometer and FT NQR for tracing the zero splitting cones. According to the crystal structure of β-TKN, two symmetry related zero splitting cones have been found for Cl(2), two for Cl(3), and four for Cl(4) ( T = 297 K). From the single crystal Zeeman split NQR measurements of α-TKN one zero splitting cone has been found for Cl(3), one for Cl(4), and two for CI(2) (T=290 K).The asymmetry parameters η and the direction cosines of the principal axes Φxx, and Φyy, and Φzz of the nuclear quadrupole coupling tensors have been determined. The results are: Cl(j), v (35Cl(j)), e2 Q q h -1 (35Cl(j)), η(35Cl(j)) for β-TKN (at T = 297 K): Cl(2), 37.442 MHz, 74.469 MHz, 0.184; Cl(3), 36.854 MHz, 73.083 MHz, 0.160; Cl(4) , Cl(4'), 38.685 MHz, 77.307 MHz, 0.070 and for α-TKN (at T = 290 K): Cl(2), CI(2'), 38.297 MHz, 76.501 MHz, 0.086; Cl(3), 36.943 MHz, 73.700 MHz, 0.123; Cl(4), 36.311 MHz, 72.314 MHz, 0.160. By comparing the crystal structures and the 35Cl NQR results, the C-Cl bonds in the title compounds are discussed.

Abstract The principal objective of this paper was to assess the physical properties and weight or structural implications of some potential green roof growth media compositions practicable for use in the Nigerian built environment. The study carried out an essential selection of material constituents of growth media blends mixed in a 3:1:1 ratio of natural stone-based gravels, soil and compost respectively. Six substrate blends based on later-ite stones, ory and empirical field evaluation methods. The results revealed that the granite-based blend is the heaviest sample with 1,713.30 kg/m3 in its saturated state, while the lightest in weight is the pumice blend with 869.30 kg/m3 which is 50.7% less than the granite blend. The heaviest and the lightest outlined models were subsequently subjected to a weight analysis on a proposed reinforced concrete flat-roofed structure. The results showed that all the extensive green roof samples fall within the IBC stipulated range. The heaviest granite substrate obtained a design load of 0.951 kN/m2, while the lightest pumice blend recorded a design load of 0.576 kN/m2. Hence, it stands to offer an optimum alternative in green roof retrofitting projects for existing flat-roofed buildings. The study, therefore, submits that all samples evaluated involve readily available materials in the studied area and can be used with respect to their characteristic properties as presented in this study. It also serves as a reference point for all stakeholders in the research and building construction industry in Nigeria and beyond.

Objective.Anticyclic citrullinated peptide antibodies (anti-CCP) are considered specific markers of rheumatoid arthritis (RA) and have been included in the revised classification criteria for RA diagnosis. However, these antibodies have also been detected in patients with other types of chronic inflammatory rheumatism. Our objectives were to identify the prevalence of positive anti-CCP patients in non-RA diseases, to determine the diagnostic value of anti-CCP for the diagnosis of RA, to specify the clinical characteristics of non-RA patients positive for anti-CCP, and to determine the discriminatory value of the levels of anti-CCP in patients among the various diseases.Methods.We carried out an observational and descriptive study. All the determinations of anti-CCP requested by the 2 rheumatology departments at Cochin Hospital over a period of 18 months were analyzed. Such determinations were requested for 1162 patients in total. Anti-CCP levels were determined with the Euro Diagnostica ELISA kit, with values ≥ 25 U for this test being considered positive. The diagnosis of rheumatic conditions was the responsibility of the treating physician.Results.Anti-CCP antibodies were detected in 357 (30.7%) of the 1162 patients. The prevalence of anti-CCP was 292/417 (70.0%) in RA, 13/122 (10.6%) in patients with psoriatic arthritis, 13/62 (20.9%) in patients with unclassified rheumatism, 11/33 (33.3%) in patients with primary Sjögren syndrome, 5/30 (16.6%) in patients with systemic lupus erythematosus, 3/28 (10.7%) in patients with mixed connective tissue disorder, 3/36 (8.3%) in patients with systemic sclerosis, 7/44 (15.9%) in patients with juvenile arthritis, and 6/220 (2.7%) in patients with noninflammatory diseases. In the population of patients positive for anti-CCP, mean anti-CCP levels were 869.4 (± 978.4) U/ml, with no significant difference between RA [854.8 (± 959.8) U/ml] and any of the non-RA conditions [922.7 (± 1070.0) U/ml].Conclusion.Anti-CCP are a hallmark of RA, but may be observed in other inflammatory, systemic, or mechanical diseases. In this large cohort of patients, the presence of second-generation anti-CCP (anti-CCP2) antibodies is useful in diagnosing RA (70% sensitivity, 91.3% specificity), but examining the levels of these antibodies does not appear to offer further discriminatory power among patients who are anti-CCP2-positive.

Stoichiometric hydrates of Li3VO4, the hexahydrate and two polymorphs of the octahydrate, were prepared by evaporation of alkaline aqueous solutions 1 molar in LiOH and 0.5 molar in the metavanadate LiVO3 at r. t. with or without the addition of Lithium sulfide, i. e. at different pH values. Their crystal structures have been determined and refined using single crystal X-ray data; all lithium and hydrogen atom positions were localised and refined without contraints. All three title compounds crystallise in non-centrosymmetric space groups. The water molecules belong to the tetrahedral coordination spheres of the Li cations, i. e. they are embedded as water of coordination exclusively. The tetrahedral orthovanadate(V) anions VO3−4 and the LiO4 tetrahedra are connected via common O corners to form building units which are further held together by strong, nearly linear hydrogen bonds. The hexahydrate Li3VO4 ・ 6H2O (space group R3, a = 962.9(2), c = 869.2(2) pm, Z = 3, R1 = 0.0260) contains isolated orthovanadate(V) anions VO3−4 surrounded by a 3D network of cornersharing Li(H2O)4 tetrahedra forming rings of three, seven and eight units. The water molecules are ‘isolated’ in the sense that no hydrogen bonds are formed between water molecules. The octahydrate is dimorphous: The triclinic polymorph of Li3VO4 ・ 8H2O (space group P1, a = 592.6(2), b = 651.3(2), c = 730.2(4) pm, α = 89.09(2), β = 89.43(2), γ = 88.968(12)°, Z = 1, R1 = 0.0325) contains two types of chains of tetrahedra: One consists of corner-sharing Li(H2O)4 tetrahedra only, the second one is formed by alternating LiO4 and VO4 tetrahedra, also sharing oxygen corners. Only one water molecule is ‘isolated’, the other seven form a branched fragment of a chain with hydrogen bonds between them. In the monoclinic form of Li3VO4・8H2O (space group Pc, a = 732.6(1), b = 653.7(1), c = 1292.9(3) pm, β = 112.21(1)°, Z = 2, R1 = 0.0289) a fragment of a chain of three LiO4 tetrahedra, two of which share a common edge, and one VO4 tetrahedron represent the formular unit. These building blocks are connected via hydrogen bonds formed by three ‘isolated’ water molecules and a chain fragment of five connected water molecules.

Following the observation that the binary dicadmides and diaurides of calcium and strontium (A) both form the KHg2 structure type, the two sections AIICdxAu2−x have been studied systematically by means of synthetic, X-ray structural and theoretical investigations. The binary border compound CaCd2 is dimorphic forming the KHg2 structure at elevated temperatures (orthorhombic, space group Imma, a = 488.63(9), b = 754.1(2), c = 851.3(2) pm, Z = 4, R1 = 0.0498) and the MgZn2-type Laves phase at ambient conditions (hexagonal, space group P63/mmc, a = 599.71(9), c = 962.7(2) pm, Z = 4, R1 = 0.0309). Starting from the known binary calcium auride CaAu2 only a very small amount of Au can be replaced by Cd. Around the 1 : 1 ratio of Au and Cd the TiNiSi structure type (orthorhombic, space group Pnma), an ordered variant of the KHg2 type, has a small homogeneity range (CaCdxAu2−x with x = 1/0.76(2): a = 735.0(1)/731.7(1), b = 433.66(6)/431.43(7), c = 873.7(2)/869.9(2) pm, Z = 4, R1 = 0.0482/0.0539). The analogous structure type is also observed in the Sr compounds with the difference that in this case a continuous transition from the KHg2 type of SrAu2 (i. e. x = 0) towards the distorted TiNiSi structure type (up to x = 0.86) is observed in the series SrCdxAu2−x (for x = 0.86(1)/0.45(1): a = 764.0(1)/758.4(1), b = 458.07(7)/474.6(1), c = 872.16(12)/829.2(2) pm, Z = 4, R1 = 0.0446/0.0410). Attempts to prepare the Ca compounds of intermediate composition around a Cd content of x ≈ 0.5 resulted in the formation of the Aurich phase Ca5Cd2Au10 crystallizing with the Zr7Ni10 structure type (orthorhombic, space group Cmca, a = 1390.6(4), b = 1015.7(3), c = 1025.6(2) pm, Z = 4, R1 = 0.0657). In this compound, Cd and Ca occupy common crystallographic sites, which are occupied by In in the isotypic ternary compound Ca4In3Au10. Similarly, at the Cd-rich parts of the sections AIICdxAu2−x no simple phase width of the KHg2 structure type exists. In the case of the calcium series the new compound Ca11Cd18Au4, which shows only a very small phase width, is formed instead. This compound crystallizes with a new structure type (Ca11Cd18+xAu4−x with x = 0.6/0: tetragonal, space group I41/amd, a = 1030.83(6)/1029.39(6), c = 3062.5(3)/3051.0(3) pm, Z = 4, R1 = 0.0475/0.0379) exhibiting a complicated Cd/Au polyanion with four-, five- and six-bonded Cd/Au atoms. The results of FPLAPWband structure calculations are used to explain the electronic stability of the compounds. The calculated Bader charges of cadmium and gold atoms (and In and Au atoms for comparison) are used to discuss the transition between Cd-rich cadmides (like CaCd2 and Ca11Cd18Au4), auridocadmates (like CaCdAu) and the Cd-poor cadmium aurides (like Ca5Cd2Au10).

Abstract Background There is a significant association between psoriasis and IBD, which share the pathogenic role of IL-23. In the dermatological field, drugs targeting the p19 subunit of IL-23, such as risankizumab, guselkumab, and tildrakizumab, have been approved for the treatment of moderate to severe psoriasis. However, there are still no real-life data on the efficacy and safety profile of IL-23 inhibitors in patients with IBD. Methods A prospective, single center observational study was conducted on patients with IBD and psoriasis from April 2021 to March 2023 at the gastroenterological-dermatological outpatient clinic at “Città della Salute e della Scienza of Turin”, Italy. Patients received a dermatological indication to start drug therapy with IL-23 inhibitors according to the dermatological dosing schedule. They were treated either with guselkumab subcutaneously at a dose of 100 mg every 8 weeks, or with risankizumab subcutaneously at a dose of 150 mg every 12 weeks, or with tildrakizumab 100 mg every 12 weeks subcutaneously. Objectives of this study included evaluating the achievement of clinical remission at 3, 6, and 12 months, defined as disease activity with Harvey-Bradshaw Index (HBI) < of 5 in CD and pMAYO < of 2 in UC. Results At 3 months after the start of drug treatment, clinical remission was achieved by 73.3% of patients (p=0.006). This number also remained almost constant at 6 (62.4%, p=0.008) and 12 months (60.3%, p=0.04). In addition, there was a statistically superior steroid-free clinical remission compared to T0 at 3 (15.3% vs 73.3%, p=0.006) and 6 months (7.1% vs 54.1%, p=0.04). No major side effects were reported. Two cases of arthralgias (11.8%) and one case of fever with chills (5.9%) were reported. IL-23 inhibitors appear to be effective in maintaining the steroid-free clinical remission. In addition, there was a reduction in fecal calprotectin values to a median value at 6 months of 81.0 (95%IC= 28.2-249.8; p=0.03) from a median value at T0 of 343.1 (95%IC= 44.8-869.7). The maintenance rate of IL-23 inhibitors therapy at the end of follow-up was 75.8%. Conclusion In this study risankizumab, guselkumab, and tildrakizumab appear to be effective in inducing clinical remission and steroid-free clinical remission in patients with IBD. In addition, they appear to be effective in maintaining steroid-free clinical remission. IL-23 inhibitors demonstrated a good safety profile in patients with IBD. Despite the limitations of the study with the need to publish further real-world data on a larger scale and with higher dosing of IL-23 inhibitors, the results obtained are promising as they, for the first time, provide real-life support for the clinical potential of these drugs as treatment for IBD in addition to psoriasis.

Abstract Introduction Sleep disturbance is common in military personnel and is often related to stressful conditions and deployment to a warzone. Among soldiers, the achievement of unit goals and performance is often dependent upon how cohesive members of the unit are. The quality of unit experiences in terms of reliance on other unit members, feeling respected, and interpersonal relationships could impact the ability to sleep at night. The primary hypothesis of this analysis was that poorer quality unit experiences would be associated with worse sleep at night. Methods Data were acquired from the All Army Study of the Army Study to Assess Risk and Resilience in Service members (STARRS; N= 21,449; 28.65 {SD=7.45} years old; 88.24% male). Participants completed the Brief Insomnia Questionnaire, and current insomnia status was determined by DSM-5 criteria. They also completed survey items related to how much respect they had for their officers and other members of their unit as well as how much respect they received in return, their overall morale and if they felt they could rely on members of their unit. T-tests and Chi-square tests were used to examine the associations between insomnia and unit experiences. Results More negative feelings about unit cohesiveness was associated with higher rates of insomnia (t(19401)= -41.19, p<.0001). Similarly, greater feelings of not being respected by other unit members was associated with higher rates of insomnia (X2 (3, N=19275) = 869.9, p<.0001). Strongly believing that the job reward was not worth the effort that was put into their work also had a significant increase in disturbed sleep (X2 (4, N=19214) = 1120.5, p<.0001). Conclusion These results demonstrate that, among Army soldiers, poorer quality experiences with other unit members are related to higher rates of insomnia symptoms. Service members who felt they could not rely on other members of their unit, did not feel their officers respected them, and could not speak openly to officers endorsed strong feelings regarding how cohesive their unit was and ultimately reported poorer sleep. Suspension feeding bivalves use a ciliary pump driven by tracts of lateral cilia located on gill filaments to provide water currents for feeding, respiration and waste elimination. These cells in Crassostrea virginica are controlled by serotonergic‐dopaminergic innervation via the branchial nerve. Serotonin is the neurotransmitter that increases beating rates, while dopamine decreases it. Lateral cells of many other bivalves are similarly innervated. The cellular mechanism regulating lateral cell cilia beating is not fully explained. Second messenger systems are involved and gill filaments receive branches from the branchial nerve, but it is not known if each lateral cell is individually innervated. Lateral cells have gap junctions and questions arise if gap junctions are involved in controlling the rate or coordination of beating. We hypothesize gap junctions are involved in the cellular mechanism regulating lateral cell cilia beating rate and/or coordination to maintain the metachronal wave pattern. To study this we made immunohistological preparations using antibodies to visualize the gap junctions, and used the Sihler Whole Mount method to visualize nerves in gill filaments. We also set up preparations in which gill filaments were divided into proximal and distal sections by a petroleum jelly barrier, which prevented diffusion of drugs from one side to the other. We stimulated the branchial nerve entering the proximal end of the filaments with suction electrodes (5 hz, 2 msec, 10 – 20 volts), which causes a release of serotonin and increases beating rates. Stroboscopic microscopy was used to measure cilia beating rates. Drugs were applied to distal ends of preparations and beating rates of proximal and distal ends were compared. Applying procaine, a local anesthetic, to distal ends prevented electrical stimulation from increasing beating rates, while beating rate at the proximal ends increased, showing nerve impulse propagation is necessary for ciliary response. Using the same procedure, we tested 3 gap junction blockers, each with different mechanisms of action: lindane, diphenylborinic anhydride and mefloquine. Mefloquine had no effect on beating, but lindane and diphenylborinic anhydride both produced statistically significant reductions in beating rates at the distal ends. Lindane was stronger, causing a dose‐dependent (10−7 – 10−4M) blockage of beating, while diphenylborinic anhydride only blocked at a high dose of 10−4M. Results show neuronal innervation to filaments in conjunction with functioning gap junctions in the cells are necessary to increase cilia beating rates. Disrupting either one interfered with the normal response. Lindane is reported to reduce gap junction permeability and increase intracellular Ca2+ levels; either one or both of these actions may be responsible for the results we obtained.Support or Funding InformationThis work was supported in part by 2R25GM06003 of the Bridge Program of NIGMS and 0516041071 of NYSDOE.

AbstractThe current study evaluated the effects of parsley essential oil on broiler growth performance, carcass features, liver and kidney functions, immunity and antioxidant activity, and lipid profile. A total of 160 unsexed 7‐day broiler chicks (Cobb500) were distributed into five groups; each group contained five replicates with eight birds each. The treatments were (1) basal diet (no additive, T1), (2) basal diet + 0.5 mL parsley essential oil/kg (T2), (3) basal diet + 1 mL parsley essential oil/kg (T3), (4) basal diet + 1.5 mL parsley essential oil/kg (T4), and (5) basal diet + 2 mL parsley essential oil/kg (T5). According to GC‐MS analysis, parsley oil contains D‐limonene, hexadecanoic acid, α‐cyclocitral, globulol, α‐pinene, myristicin, cryophyllene, bergapten, α‐chamigrene, etc. The current results indicated that the most abundant molecules in parsley oil were D‐limonene (18.82%), oleic acid (14.52%), α‐cyclocitral (11.75%), globulol (11.24%), α‐guaiene (7.34%), apiol (5.45%), and hexadecanoic acid (4.69%). Adding parsley essential oil to the broiler diet quadratically increased body weight (BW) during 1–3 weeks of age. The T5 group recorded the highest value (869.37 g) of BW in comparison to other treatments and the control group. The cholesterol, triglyceride, low‐density lipoprotein (LDL) cholesterol, and total immunoglobulin, including immunoglobulin G (IgG) and immunoglobulin M (IgM) levels in the birds fed parsley essential oil were not affected. The T3 group recorded the highest value (159 ng/mL) of superoxide dismutase (SOD) and the lowest value (2.01 ng/mL) of malondialdehyde (MDA) when compared to the control and other treatment. In conclusion, we recommend using parsley oil at levels of 1 mL/kg diet of broiler chicks.

ObjectiveWe designed a new type of ‘physical activity calorie equivalent’ (PACE) food label in Iran to compare its effect with that of the traffic light food label (TLL) on caloric choices.DesignMixed-method study.ParticipantsMothers of school children between the ages of 6–12 years.SettingIn the qualitative phase, 10 focus group discussions (FGDs) were conducted with various groups of mothers, and two FGDs were conducted with food science and nutrition experts to design a new PACE label. In the quantitative phase, 496 mothers were randomly assigned to five groups: (1) no nutrition label, (2) current TLL, (3) current TLL + educational brochure, (4) PACE label, and (5) PACE label + brochure. Samples of dairy products, beverages, cakes, and biscuits were presented. ANOVA and multiple linear regressions were applied to examine the association between label types and calories of the selected products as our main outcome.ResultsThe mothers’ perspectives were classified into two sub-themes, the PACE label’s facilitators and barriers. The new PACE label’s characteristics were divided into two subcategories: (a) appearance, and (b) nutritional information, including 14 codes. In the quantitative section, mean calories of the selected foods were lowest in the TLL + brochure group (831.77 kcal; 95% CI: 794.23–869.32), and highest in the PACE label group (971.61; 95% CI: 926.37–1016.84).ConclusionThe new PACE label was a combination of PACE, TLL, and warning labels. It did not significantly affect lower caloric choice, however, the TLL + brochure option was effective in choosing foods with fewer calories.Clinical trial registration: The study was registered in the Iranian Registry of Clinical Trials 23 (IRCT20181002041201N1).

AbstractThe Japanese plum (Prunus salicina Lindl.) is one of the most important stone fruit crops in China. High-density linkage map is valuable resources which enhance functional genomics and genetic breeding studies. So far several Japanese plum linkage maps have been reported using different kinds of molecular markers; however, the marker numbers and chromosome coverage are limited. Recently, a newly developed strategy which genome sequencing towards specific-locus amplified fragments (SLAF) markers, has been proven to be powerful for rapid genotyping of genome-wide markers and for high-density genetic map construction. In this study, SLAF was used to genotype markers with 114 F1 seedlings from the ‘09–16’ × ‘Fortune’ cross. Suitable SLAF markers (160,344 out of 343,436,902 pair-end reads) were chosen to conduct genetic map construction, 16.31% of which were polymorphic. The overall integrated map contained 3,341 high quality SLAFs and 720 loci that were grouped in eight genetic linkage groups with a total length of 869.9 cM and an average distance of 1.21 cM, and only five gaps with a genetic distance > 5 cM between adjacent markers occurred in linkage group (LG) 3 and LG6. The number of markers with each LG ranged from 82.3 cM (LG3) to 138.3 cM (LG1). Aligning the map against the peach reference genome sequence (Prunus persica L.) indicated a strictly co-linear relationship between the LGs and peach genome, demonstrating the markers on ours LGs were well ordered. Overall, our studies identified large-scale of genetic markers and constructed high-density linkage maps for Japanese plum, which will obviously provide a solid foundation for marker-assisted selection and sequence assembly of the Japanese plum reference genome.

Background: Moyamoya Disease (MMD) is a steno-occlusive arteriopathy associated with transient ischemic attacks, stroke, and cognitive impairments. In adult MMD, normal-appearing deep gray and white matter structures (NADWM) show diffusion changes that are associated with cognitive impairments. Apparent diffusion coefficient [ADC] measures the mean diffusion and is sensitive to these changes. The association of ADC changes in NADWM in pediatric MMD is not clear. Our objective is to determine whether ADC is elevated in NADWM in pediatric MMD patients. Hypothesis: ADC values in NADWM will be elevated in MMD patients compared to controls. Methods: Twenty-nine MMD patients and no stroke and 7 healthy controls (HC) were included in a single center, retrospective study. Diffusion and T1-weighted images were analyzed. We constructed a script implementing a convolutional neural network (CNN) based approach to segment and binarize cortical regions of interest (ROI) and allow extraction of ADC values (units =[x10 -6 mm 2 /s]). Segmented ROIs included: 1) all white matter (WM), and NADWM including: 2) thalamus, 3) caudate, 4) putamen, 5) globus pallidus, 6) hippocampus. Mean ADC values were compared for: 1) all ROIs between MMD and controls, and 2) all ROIs in the MMD affected and the contralateral hemispheres within the MMD group. Results: Patients were on average 7.7 + 5.4 years at MMD diagnosis. In all WM, MMD patients had significantly higher ADC values than controls in the left (mean= 865.7 + 101.6 versus 762.4 + 21.6, p=0.01) and right hemispheres (mean= 869.9 + 104.9 versus 761.0 + 21.3, p=.01), respectively. In NADGM, there were no significant differences found between the MMD and control groups in any of the NADGM ROIs. In the NADGM of MMD patients, higher ADC values were found in the putamen (mean= 778.2 + 46.0, p=0.041), hippocampus (mean=1009.5 + 58.6, p=0.005) and all NAWM (mean= 860.3 + 57.0, p<0.0001) in the MMD affected compared to the contra-lesional hemisphere. Conclusions: In pediatric patients with MMD, we found elevated ADC values in deep gray and white matter structures in the diseased hemisphere indicating alterations in the underlying microstructure. Our future analysis will integrate hemodynamic profiles and cognitive outcomes.

Abstract Disclosure: N. Pascuali: None. F. Tobias: None. K. Valyi-Nagy: None. S. Salih: None. A. Veiga-Lopez: None. The incidence of infertility has been consistently increasing over the past few decades, with a high proportion of these cases caused by ovarian-related dysfunction. Metabolic disorders such as hyper- or dyslipidemia have been associated with subfertility (e.g., follicular fluid dyslipidemia is associated with lower IVF success). Lipids play a critical role in the structure and function of ovarian tissue. However, while several studies have investigated transcript- and protein-level alterations in ovarian pathologies, less is known about alterations in the lipid landscape. The first step in determining if ovarian disorders are associated with ovarian dyslipidemia - and thus use lipids as potential biomarkers of ovarian pathology - is to understand the physiological distribution of ovarian lipids. Matrix-Assisted Laser Desorption Ionization Time-Of-Flight (MALDI-TOF) mass spectrometry imaging (MSI) enables label-free in situ analysis of molecules such as lipids and metabolites directly within tissue sections. We aimed to begin mapping the distribution of lipids in human and mouse ovaries using high spatial resolution MALDI MSI. To this end, and after informed consent, two fresh ovarian biopsies from a 26-year-old patient were frozen, cryosectioned into 10-μm sections, and mounted onto ITO-coated slides. A set of 21-day-old mouse ovaries (n=10) were gelatin-embedded and cryosectioned. Slides were sprayed with 1,5-diaminonaphthalene as the matrix. MALDI MSI was performed using a Bruker RapifleX MALDI Tissuetyper (20-μm resolution) in the negative ion mode. Data were processed using the SCiLS Lab software. We identified for the first time in human ovaries over 100 lipid species, with most of them displaying distinct spatial distributions. Specifically, we detected 3 lipids specific to theca cells (m/z: 701.6; 1050.8; 1263.9), 6 lipids specific to granulosa cells (m/z: 279.3; 697.5; 861.7; 883.7; 911.7; 1448.0), 3 lipids specific to the stromal region (m/z 392.1; 753.5; 835.7), and one specific to luteal cells (m/z: 615.3). As for mouse ovaries, we detected 5 lipids specific to follicular cells (theca or granulosa) (m/z: 327.1: 343.2; 552.6; 632.6; 703.5), 5 lipids specific to stromal cells (m/z: 715.6; 729.6; 743.6; 771.7; 799.6), 5 lipids specific to oocytes (m/z: 778.5; 863.7; 1063.8; 1225.7; 1336.0) and 7 lipids specific to the ovarian cortex (m/z: 721.5; 748.5; 776.6; 792.6; 810.6; 869.6; 885.7). To our knowledge, this is the first evaluation of the human ovarian lipidome and characterization of region- or cell-layer-specific lipids. This approach has allowed us to demonstrate that compartments as small as oocytes (∼70 μm in diameter) have a unique lipid signature. Harnessing the unique specificity of the lipid signature per ovarian region and/or cell layer in this study could help pave the way to develop novel biomarkers of ovarian dysfunction.
