Pełny tekst źródłaIn the context of the National Program of Control and Eradication of Brucellosis and Tuberculosis (PNCEBT), the necessity of an efficient bacteriologic characterization of the infected herds as a cornerstone of the monitoring system and the difficulties faced by the laboratories regarding the methods for Mycobacterium bovis isolation led to a growing interest for scientific studies, especially molecular, of M. bovis isolates. To use these molecular techniques it is necessary to have an abundant bacillary mass, obtained through the maintenance of isolates in laboratory and replication in culture media. However the fastidious growth of M. bovis in culture media brings out great difficulties for these activities. Thus, the present study has the following objectives: First, to compare three decontamination methods for organ homogenates, phase that precedes the sowing in culture media, 60 samples of tissues with granulomatosis injuries, proceeding from bovine slaughterhouses in the State of São Paulo, were obtained, immersed in sodium borato saturated solution and transported to the Laboratório de Zoonoses Bacterianas of the VPS-FMVZ-USP, where they were processed up to 60 days after the sampling. These samples were submitted to three methods of decontamination: Basic NaOH 4%, Acid (H2SO4 12%) and 1- Hexadecylpyridinium chloride (HPC) 1.5% and a simple dilution with saline solution (control method). The results were analysed by means of the &chi test to compare proportions, and it was verified that HPC method presented the smallest proportion of contamination (3%) and the greatest proportion of success for M. bovis isolation (40%). Second, to compare three different cryopreservation media for M. bovis, 16 isolates identified by the technique of spoligotyping were used. Each one of these isolates was solubilized in three media (original saline solution, 7H9 and 7H9 modified), and stored in three different temperatures (-20ºC, -80ºC and -196ordm;C), and defrosted in three different time periods (45, 90 and 120 days of freezing). Before the freezing and after the unfreezing, quantitative cultivations in Stonebrink Leslie media were carried out. The proportions of Colony-Forming Units (CFU) loss in the different conditions were calculated and compared with one another through parametric and non-parametric methods. The results obtained were: in the analysis of the variable time, at 90 days of freezing a bigger proportion of CFU loss was observed when compared to 120 days (p=0,0002); in the analysis of the variable temperature, a statistically significant difference was observed between the average proportions of CFU loss for the temperatures of -20ºC and -80ºC (p<0,05); in the analysis of the variable media, a significant difference was observed (p=0,044) between the media A and C, for 45 days of freezing and -20ºC of cryopreservation temperature. Althougth the medium ones of the proportion of losses of CFU to always have been inferior 4,2%, had allowed to suggest that the best protocol for cryopreservation of M. bovis isolates is to solubilize them in 7H9 modified medium and to keep them at a temperature of -20ºC

As abordagens de Biologia Sintética possuem potencial de aumentar a produção de ferramentas diagnósticas adequadas para utilização na prática, como os biossensores de DNA. O controle dos sistemas biológicos permite o aprimoramento de tecnologias em uso e surgimento de novas tecnologias, como o diagnóstico molecular através da identificação de sequências específicas de DNA. A maioria dos métodos moleculares de diagnóstico de DNA se baseiam em hibridização. Contudo, atualmente, as proteínas ligadoras de DNA tem tido destaque, devido às suas numerosas vantagens. As principais vantagens são a identificação de sequências em DNA dupla-fita, eliminando o passo de desnaturação, e a alta especificidade pelo alvo. Neste contexto, o objetivo deste trabalho foi desenvolver uma proteína sintética quimérica para aplicação em sistema biossensor de DNA que permitirá rápida identificação de uma sequência específica de DNA de Corynebacterium striatum, um importante patógeno emergente. Foi construída, neste estudo, uma proteína quimérica biossensora composta por: (I) um domínio de ligação ao DNA derivado de proteínas TALEs (proteínas efetoras do tipo ativadores transcricionais) o qual reconhece especificamente uma sequência de 18 pb de um gene único de C.striatum codificador de uma permease para transporte de ferro; combinado a (II) uma cromoproteína tsPurple (Tinsel Purple) para permitir detecção visual da sequência alvo. Para construção do gene codificador da proteína TALE específica, o gene específico de C. striatum foi analisado com a ferramenta Tal Effector Nucleotide Targeter (Cornell University) e uma sequência-alvo de 18pb foi definida. A construção de domínios TAL foi realizada utilizando a ferramenta GeneArtTM Perfect Match TAL (Thermo Scientific); o gene sintético encontra-se clonado no plasmídeo pTALE. Para construir as sequências gênicas quiméricas, foram desenhados primers de PCR para isolamento da sequência da tsPurple com a adição de sítios artificiais de restrição para EcoRI e BamHI; os produtos de PCR digeridos foram clonados no plasmídeo pTALE, downstream à sequência da TALE específica. Essas sequências fusionadas foram então sub-clonadas em um vetor de expressão pDESTTM17, através de recombinação utilizando tecnologia Gateway para expressão das proteínas quiméricas. A expressão da proteína quimérica foi induzida pela adição de 1mM de IPTG. A expressão da proteína quimérica foi confirmada através de análises de SDS PAGE 10% , que demonstraram a superexpressão da proteína esperada de 151kDa; além disso, foi observada a forte coloração roxa no precipitado bacteriano, em razão da funcionalidade da cromoproteína TsPurple.

Nowaday, the onchip radio front-end contains passive components such like, spiral inductors and capacitors. Thinking about integrating (in terms of microelectronic integration) the antenna including its switching capabilities, seems to be interesting in the field of Smart Deviees operating at high frequency ranges. The aim of this study, is to simulate, design and measure dipole antennas directly coupled to VCO's, using standard (industrial type) silicon BiCMOS technology. The considered operating frequencies are in the range of 10 to 20 GHz. By achieving this study, the technological issues for antenna integration will be highlighted and the capabilities of the considered standard BiCMOS microelectronic technologies will be investigated to allow on-chip antenna implementation with acceptable performances

Pełny tekst źródłaThe elastic and inelastic scattering reactions of an 8He beam on a proton target were measured at GANIL. The 8He beam produced by the SPIRAL facility at an energy of 15. 6 A. MeV impinged on a proton target and the recoil proton was detected in an array of eight telescopes, the wall of strip-detectors, MUST, devoted to the detection of light charged. The experimental setup was also by a plastic scintillator wall for the detection of heavy projectile, and by two beam tracking detectors CATS for the measure event by event of the incident position and angle of the beam on the target. This setup allowed also to measure the one and two neutron transfer reactions 8He(p,d)7He and 8He(p,t)6He. The excitation energy spectrum for 6,7,8He and the angular distributions associated to each reaction were measured. The observed excited states are in agreement with the data of the literature. We indicate the presence of a first excited state for 7He at 0. 9 MeV and we give for the first time the position of a second excited state in 8He; the presence of such state was just suggested in a previous experiment. From CCBA calculations a spectroscopic factor of 4. 4±1. 4 for the pick-up of one neutron from 8He to 7He was extracted from the data. This value is in agreement with a closed p3/2 sub-shell for 8He. To analyse successfully the angular distributions for elastic and inelastic scattering, it was necessary to take into account in the formalism, used to describe the reactions, the couplings to the one neutron transfer reaction. Pełny tekst źródłaLos sistemas fotosintéticos naturales y artificiales en los cuales la energía de la luz es colectada y almacenada como energía química, es un proceso de conversión que es iniciado con la transferencia fotoinducida de un electrón, que es seguida por otras etapas químicas que permiten la separación y estabilización cinética de los llamados “sistemas de carga separada”. Este es el caso de las 7H-dibenzo[de,h]quinolin-7-onas, conocidas como oxoisoaporfinas, que al ser fotorreducidas en presencia de aminas generan fotoproductos iónicos metaestables, capaces de almacenar una fracción apreciable de la energía de fotoexcitación. Los fotoproductos que se forman vía una transferencia secuencial de electrón-protón-electrón, revierten térmicamente a las oxoisoaporfinas originales, generando productos de oxidación de la amina. La reconversión del fotoproducto metaestable permite la reutilización del sistema en tanto exista amina remanente que reaccione con la oxoisoaporfina regenerada. Se estudiaron los transientes de la fotorreducción por aminas de la oxoisoaporfina y 5-metoxi-oxoisoaporfina, el triplete excitado, el anión radical y el radical neutro. Los intermediarios transientes fueron generados por láser flash-fotólisis, en presencia y ausencia de aminas terciarias, con y sin hidrógenos alfa. Si la amina no es capaz de donar protones, la reacción procederá sólo hasta la formación de A-·. Se identificaron y se caracterizaron los tripletes y radicales de las dos oxoisoaporfinas en estudio y se determinaron los parámetros cinéticos
Natural and artificial photosynthetic systems in which the energy of the light is collected and stored as chemical energy, is a process beggining with a photoinduced single electron transfer, PET, followed by other stages wich allow the separation and kinetic stabilization of “separate-charge systems”. This is the case of 7H-dibenzo[de,h]quinolin-7-onas, well-known as oxoisoaporphines, wich are photoreduced by amines, generating ionic metastable photoproducts capable to store an appreciable fraction of the photoexcitation energy. The photoproducts, formed via a sequential transfer of electron-proton-electron, revert thermally to the original oxoisoaporphines generating amine photooxidation products. The back reaction of the metastable photoproducts allows the reuse of the system as long as amine remains in the system, to react with the regenerated oxoisoaporphine. In this work we studied the transient species involved in the photoreduction by amines of oxoisoaporphine and 5-methoxy-oxoisoaporphine; the excited triplet state of oxoisoaporphines, the radical anion formed after electron transfer and the neutral hydrogen radical, generated by proton transfer. Photoreduction intermediary transients were generated by laser flash-photolysis, in the presence and absence of tertiary amines, with and without transferable α- hydrogen. If the amine is not able to donate protons, the reaction will only proceed until the formation of A-·, allowing its characterization. With amines capables to donate an α- H, photoreduction continues to the formation of neutral hydrogen radical AH·, and eventually to metastable photoproducts. In this work triplet excited states were characterized, radical anions and neutral hydrogen radicals for both oxoisoaporphines and the kinetic parameters through evolve were also determined
