The stela contains information about the family of the Bearer of Soil Iy at Abydos in the late Middle Kingdom, adding to Simpson’s ANOC 19. The archaeological context of the object is explored, and a full translation of E.31 is provided, as well as discussion of a heavily fragmented stela (321d) found in the same context by Garstang.

Introduction: Psychogenic coma, generally is one of the most anxious and irritant problems in clinical medicine for all medical practitioners. Using recurrent painful or mischievous stimuli is contraindicated in a psychogenic coma. Method of study and patients: My study was technical, prospective, observational, and interventional for 321 cases. The study was conducted in a physician outpatient clinic, Fraskour Central Hospital, and Ras-Al-bar Central Hospital. The author reported the 321 cases of psychogenic coma over nearly 5 years and 7-months, started from August 07, 2015, and, ended on March 07, 2021. Three selective groups were included in the study. Three groups were selected and evaluated for safety or complications and efficacy or responses. Suggesting hypothesis: Yasser’s maneuver can regaining the consciousness in a psychogenic coma. The research objectives to evaluate this hypothesis might include: What is psychogenic coma? What is Yasser’s maneuver? How can Yasser’s maneuver do improvement of psychogenic coma? Is the study supported by past publicized literature studies? Results: The range of age in the study was 16-55 years with an insignificant P-value (0.231). There is a female sex predominance for all groups (67%). The response for the group I was: (97.35%)) vs. (85.85%)) in group II, and (81.37%) in group III. The most common associated risk factor was psychogenic hyperventilation syndrome (HVS); in the group, I was 79.6%, in group II was 78.4%, and in group III was 96.2±1.7 with no statistical significance (P-value 0.74). Conclusions: The author concluded thatYasser’s maneuver is easy, available, quick, non-costive, time-saving, and extremely safe in the psychogenic coma. Very few and mild few mild complications for this maneuver encourage the generalizing use in the psychogenic coma.

Field crickets were collected from agricultural fields and the surrounding vegetation of district Jacobabad through traditional insect nets and by hand picking. The collected specimens were 321, which were sorted out into single sub-family Gryllinae falling into 05 genera and 07 species, i.e. Gryllodes sigillatus (Walker, 1869), Gryllodes supplicans (Walker, 1859), Gryllus bimaculatus (De-Geer, 1773), Acheta burdigalensis (Latreille, 1804), Acheta domesticus (Linnaeus, 1758), Gryllopsis histrio (Saussure, 1877) and Teleogryllus commodus (Walker, 1869). The maximum population of Acheta domesticus (Linnaeus, 1758) was 36%, and Gryllodes supplicans (Walker, 1859) was 24%. While the minimum population of Gryllopsis histrio (Saussure, 1877) was 3%, and Acheta burdigalensis (Latreille, 1804) was 5%, respectively. Additionally, a detailed description of species and distribution is provided. Finding field crickets from this area will be instrumental for the farmers and researchers concerned with the biodiversity of this group.

A descriptive study was conducted on HIV patients who had started antiretroviral therapy (ART). This study included a sample of 321 patients selected from the cohort of HIV-infected patients followed at the Medical Center for the Follow-up of Blood Donors (CMSDS) of Abidjan (RCI), in the period from 2005 to 2012. These patients were regularly treated, according to their status, with seven (07) ARV therapeutic combinations which are AZT-3TC-EFV AZT-3TC-NVP AZT-3TC-NFV AZT-3TC-LOP/RIT DT4-3TC-EFV DT4-3TC-NVP FTC-TDF-EFV. Regular follow-ups were performed during 36 months of treatment and clinical parameters (Karnofsky Index, Body Mass Index, Brachial Perimeter) were measured every six (6) months. The evolution of these three parameters (KI, BMI and BP) is a good indicator of the effectiveness of the treatments, as they allow patients to maintain a good clinical condition.

Некоторые спортсмены неправильно трактуют влияние молочной кислоты (в т. ч. лактата и связанных с ним ионов водорода) на организм, что ведет к неверным целям мышечного развития. В статье проводится анализ научной литературы, в которой рассматривается влияние молочной кислоты на развитие мышечной выносливости при разных типах тренировок – аэробных и анаэробных. В конце статьи сделан вывод, о том, что молочная кислота не имеет никакого влияния на развитие общей выносливости, поскольку не используется в процессе аэробных тренировок. В то же время молочная кислота является одним из главных химических компонентов анаэробных тренировок, которые развивают специальную выносливость.

Research in recent decades on specialised languages – the status and methods of terminology, terminological definitions, the characteristics of terms, the relationship between term and word, etc. – shows how radical changes have occurred in the field. We would like to note, however, that the stylistic and poetic metalanguage of the English language still remains outside the interests of researchers. So far we do not have studies, which would elucidate the systemic and functional complexity of English stylistic-poetic terminology. These specific languages are necessary, important, especially in the areas of interpretation, decoding, exegesis and hermeneutics of text. They have always been part of the linguistic terminology developed by various schools: in Paris, London, Heidelberg, Prague. The field of metalanguage, which is of interest to us, its lexicographical aspect has not been the subject of research in English. Terms such as anesis, antanagoga, antistasis, catabasis, colon, cycloid and many others have remained omitted, some ignored, others replaced. Our investigation aims to identify these terms, define them lexicographically and present them as integral elements of stylistic and poetic metalanguage that cannot, but do not deserve, to be marginalized.

O processo de expansão da Palma de Óleo na Amazônia foi possibilitado pelas condições edafoclimáticas e de ocupação do espaço rural amazônida, o artigo presente traz como objetivo expor a relação da mudança de uso e ocupação da terra e a expansão da Palma de Óleo na Amazônia Oriental, dessa forma metodologicamente se utilizou, principalmente, de imagens de satélite Landsat 07 e 08, para os anos de 2002 e 2020, sendo adotadas classes para a melhor compreensão das diferenças de ocupação do solo entre estes dois períodos, adotando-se 10 classes. Os resultados demonstram que houve um crescimento de 321% relacionado à ocupação da Palma, e que aproximadamente 30% da conversão se deu de territórios que já foram ocupados por atividades agropecuárias e solo exposto por outras atividades, também se traz um quadro que expõe políticas e ferramentas que podem auxiliar na compreensão da expansão dessa monocultura. The dynamics of oil palm expansion - elaeis guineensis jacq in an eastern Amazonian territory A B S T R A C TThe process of expansion of oil palm in the Amazon was made possible by the edaphoclimatic conditions and situation of the occupation of the Amazonian rural space. This article aims to expose the relation between the changes in land use and occupation and the expansion of the oil palm in the Eastern Amazon. Thus, methodologically, mainly Landsat 07 and 08 satellite images were used, for the years 2002 and 2020, with the adoption of classes to better understand the differences in land occupation between these two periods; 10 classes were adopted. The results show that there was a growth of 321% related to the occupation of oil palm, and that approximately 30% of the conversion took place in territories that were already occupied by agricultural activities and soil exposed by other activities. The research also indicates policies and tools that can assist in understanding the expansion of this monoculture. It was concluded that the history of occupation of the towns may differ; however, the occupation by the oil palm can be analyzed in a unified way, as done in this study, and, in addition, a simultaneous joint work of the different fields of interest on the expansion of palm can lead to less harmful results to the inserted environment and the impacted social group.Key Words: land use change, geotechnologies, northeast of Pará.

The effect of different sources of Arbuscular mycorrhiza (AM) fungi on cabbage seedlings (Atlas-70) were conducted. Eight sources of AM fungi viz. Jessore (AM-01), Rahmatpur (AM-02), Joydebpur (AM-03), Ullapara (AM-04), Jamalpur (AM-05), Hathazari (AM-06), Ishurdi (AM-07), Rajshahi (AM-08) from different AEZs of Bangladesh were studied along with a control and mixed sources on cabbage seedlings. Soil based AM inoculum at the rate of 2.0 kg m-2 was used. Biomass yield of cabbage (Atlas-70) increased from 28.0% to 130.7% in 2007-08 and 44.8% to 96.9% in 2008-09 over control by inoculation with different sources of AM. The highest biomass yield (503 mg seedling-1 in 2007-08 and 321 mg seedling-1 in 2008-09) of cabbage was observed with Jamalpur source which was identical to all AM source except Ishurdi source in 2007-08 and with Joydebpur source which was identical to all Jamalpur source in 2008-09. Uptake of all the nutrients by cabbage seedlings was also improved by inoculation with AM fungi. High trait positive affect (PA) protects against depressive symptoms through cognitive responses such as rumination. However, how rumination in response to positive emotions (positive rumination) protects against depressive symptoms while rumination in response to negative emotions (brooding) predicts depressive symptoms is poorly understood. We hypothesized that (a) positive rumination and brooding represent a shared cognitive process of affect amplification on distinct affective content and (b) less brooding and greater positive rumination would distinctly mediate greater trait PA in predicting fewer depressive symptoms. Our prospective design among 321 adults first compared three confirmatory factor analysis models of the relationship between brooding and positive rumination. We then utilized structural equation modeling to examine whether brooding and positive rumination mediated the relationship between trait PA and depressive symptoms, controlling for baseline depressive symptoms, trait negative affect (NA), and the distinct effects of each mediator. Results supported a conceptualization of brooding and positive rumination as distinct but related constructs, represented as a common process of affect amplification to explain how rumination may amplify resilience or risk in predicting depressive symptoms (χ = 195.07, Δχ = 8.78, p < .001, CFI = .91, RMSEA = .07). Furthermore, positive rumination and brooding were distinctly predicted by trait PA, suggesting that trait PA exerts distinct effects on protective and risk forms of rumination. Less brooding mediated the relationship between greater trait PA and fewer depressive symptoms (β = -.04, p = .012), but positive rumination did not (β = .02, p = .517). Rumination may represent a protective and a risk factor, which may better enable individuals who brood to redirect their rumination on positive content and thereby reduce their risk of depressive symptoms.

Purpose We investigated whether thymidylate synthase (TS) expression is a predictive marker for the clinical outcome of pemetrexed/cisplatin in patients with nonsquamous non–small-cell lung cancer. Patients and Methods Eligible patients were tested for TS expression by immunohistochemistry and stratified into either a TS-negative or a TS-positive group. After stratification, patients in each group were randomly assigned (1:1 ratio) to receive either pemetrexed/cisplatin or gemcitabine/cisplatin for a maximum of six cycles until disease progression. The primary end point was evaluation of the interaction between TS groups and treatment allocation for objective response rate. Results Of 321 enrolled patients with nonsquamous non–small-cell lung cancer, 315 received at least one dose of study chemotherapy and were analyzed. By investigator assessment, response rates were 47% for the pemetrexed/cisplatin arm and 21% for the gemcitabine/cisplatin arm in the TS-negative group and 40% and 39%, respectively, for the TS-positive group (interaction P = .0084). By independent reviewers, response rates of pemetrexed/cisplatin and gemcitabine/cisplatin were 39% and 21%, respectively, in the TS-negative group and 40% and 48% in the TS-positive group (interaction P = .0077). The median progression-free survival times for the pemetrexed/cisplatin and the gemcitabine/cisplatin arms were 6.4 and 5.5 months, respectively, in the TS-negative group and 5.9 and 5.3 months in the TS-positive group (interaction P = .07). Conclusion With regard to response rate and progression-free survival, pemetrexed/cisplatin was superior to gemcitabine/cisplatin in the TS-negative group but not in the TS-positive group, indicative of TS expression as a potential predictive marker. Additional prospective studies involving larger cohorts are warranted to confirm the predictive role of TS expression.

OBJECTIVES/GOALS: We aim to develop an understanding of how polysubstance use (PSU) relates to the general psychopathology factor (p-factor), as well as to individual components of the Hierarchical Taxonomy of Psychopathology (HiTOP) model (e.g., fear, distress). This insight can help identify treatment targets related to substance use and psychopathology. METHODS/STUDY POPULATION: Psychopathology and substance use data, collected at a Baltimore treatment center over several years, will be analyzed. The center aids about 6000 underserved clients per year, and the population is primarily African American clients of all genders. Structural equation modeling (using Mplus software) will be used to develop the latent models and identify relationships between psychopathology and PSU (i.e., direct and indirect pathways). The current latent HiTOP model was developed from symptom checklists completed upon entry at the treatment center. The PSU latent factor will be developed from a biopsychosocial assessment where clients list their drug of choice. Due to the varying organizations of the datasets, smaller-scale preliminary models will be developed to ensure an accurate large-scale final model. RESULTS/ANTICIPATED RESULTS: Current models being tested are derived from January to September 2023 data (i.e., completed months' data), with an N of 1,564. From symptom checklist data collected at the treatment center, a preliminary HiTOP model was derived with reasonable fit (χ2 = 4532.35 (df = 321, p<.001), CFI = .77, SRMR = .07, RMSEA = .09 (.089, .094)). Data analysis is being conducted to derive the PSU factor before relating PSU to the HiTOP model. Given previous work at a local treatment center (Pavuluri etal., 2022) and with the National Comorbidity Survey-Replication data, we expect all positive direct relationships, negative indirect relationships between internalizing factors (fear and distress) and PSU when accounting for p-factor, and a positive indirect relationship between antagonism and PSU when accounting for p-factor. DISCUSSION/SIGNIFICANCE: Given our previous work to develop such models, we want to establish proof of concept in alarger treatment center population. This confirmation will help provide a path towards conducting therapeutic trials to target psychopathology when treating substance use given the shared relations, some of which are less understood (e.g., fear and PSU).

The competitiveness of three phenotypically different sugarcane cultivars with bermudagrass was determined in field trials. In trial one, bermudagrass biomass was 22% less in CP 70-321 than in HoCP 85-845 in the plant-cane crop, but biomass was 130 to 170% greater in CP 70-321 than in the other two cultivars during the second-ratoon crop. CP 70-321 emerges quickly following planting, which might have reduced bermudagrass growth in the plant-cane crop, but the lower stalk population of CP 70-321 might have promoted bermudagrass survival and growth during the second-ratoon crop. In trial two, there were no differences in bermudagrass biomass when comparing its establishment in the different cultivars. Sugarcane, averaged across cultivar, produced fewer stalks and was shorter when competing with bermudagrass. In the plant-cane crop, stalk populations were reduced 13 to 23%. In the first-ratoon crop, stalk population was reduced 8 to 15%. In the second-ratoon crop, stalk population was reduced 8 to 10%. Bermudagrass interference reduced sugar yields by 8 to 32% in the plant-cane crop, with reductions of no more than 9% in the first- and second-ratoon crops. The greater yield loss in the plant-cane crop in the first production year shows the importance of controlling bermudagrass in the summer fallow period prior to planting and during establishment of the plant-cane crop.

ABSTRACT Treatment of immature (22-day-old) female Sprague–Dawley rats with pregnant mare serum gonadotrophin (PMSG) resulted in a significant increase in hepatic microsomal cytochrome P-450 concentrations when compared with animals treated with 0·9% (w/v) NaCl solution. Cytochrome P-450 concentrations in microsomes from control and PMSG-treated animals were 0·49 ± 0·07 and 0·78 ± 0·18 nmol/mg microsomal protein respectively (mean ± s.d.). When hypophysectomized rats were treated with PMSG no significant difference was observed between saline-treated and PMSG-treated animals (0·84 ± 0·06 vs 0·88± 0·08 nmol/mg microsomal protein). Cytochrome b5 concentration and NADPH–cytochrome c reductase and ferricyanide reductase activities were not significantly affected by PMSG treatment. Similar results were obtained when immature and hypophysectomized rats were treated with oestradiol benzoate. Hepatic glutathione (GSH) S-transferase activity was significantly stimulated over control values when hypophysectomized or immature animals were treated with PMSG, oestradiol benzoate or 3-methyl-cholanthrene. Treatment of animals simultaneously with PMSG or oestradiol benzoate and 3-methyl-cholanthrene resulted in higher transferase activity than did either treatment alone. Oestradiol benzoate or PMSG treatment of immature female rats resulted in an approximate twofold increase in hepatic aryl hydrocarbon hydroxylase (AHH) activity. However, AHH activity in hypophysectomized animals was not significantly increased by PMSG or oestradiol benzoate treatment. It is concluded that PMSG or oestradiol benzoate treatment of immature female rats resulted in increased total cytochrome P-450 concentration and AHH activity in hepatic microsomes. Glutathione S-transferase activity was also stimulated. Neither PMSG nor oestradiol benzoate stimulated cytochrome P-450 levels or AHH activity in hypophysectomized rats. However, GSH S-transferase activity was stimulated similar to that in immature animals. Conceivably, the PMSG effect observed in immature rats may be due to PMSG-stimulated increases in plasma oestradiol concentrations which interact at the hypothalamo-pituitary axis. The ability of oestradiol benzoate to mimic the effects of PMSG and the known action of PMSG to increase plasma oestradiol levels in immature rats would tend to support this hypothesis. The lack of increase in cytochrome P-450 concentrations and AHH activity in hypophysectomized female rats would further support the involvement of the pituitary gland in the response to PMSG rather than a direct effect of PMSG or oestradiol benzoate on the liver. Abstract INTRODUCTION: The search for more personalized treatments for breast cancer patients has been increasingly frequent. In this sense, the search for the de-escalation of the therapies used, especially the cytotoxic ones, should be prioritized, with the application of genetic signatures, in patients with luminal tumors, allowing more adjusted therapies and avoiding serious and even fatal outcomes in patients who will not benefit from chemotherapy. The MINDACT study evaluated the use of MammaPrint™, but in a population of nine exclusively European countries. The study of the application of this genetic signature in other populations, especially in low- and middle-income countries, may determine greater safety in the universalization of the test.OBJECTIVE: To analyze the results of the application of MammaPrint™ in the Brazilian population, and to compare with data from the MINDACT study, when applied to breast cancer patients with high clinical risk.METHODOLOGY: Cross-sectional study comparing the prevalence of low and high genomic risk in a population with breast carcinoma, of high clinical risk, in the populations of the MINDACT study and of a Brazilian cohort, evaluated by the genetic signature MammaPrint™, between 2016 and 2020. The simple and relative frequencies of the variables were calculated in relation to the classification in low and high risk of the populations of the AGEMA-BRA and MINDACT studies. Then, the chi-square test was used to verify the differences between the proportions. To measure the intensity of differences/associations, relative risks (RR) and their 95% confidence intervals (95% CI) were calculated. The tests were considered significant when p&lt;0.05.RESULTS: The presence of the signature of 70 genes with low genomic risk in the total AGEMA-BRA sample was found in 57.1% (542 patients), compared with 46.2% in the MINDACT study (RR 1.23 (1.16-1.32 p &lt;0,001). In the analysis of subgroup by age group; &lt;35 years in 46.4% (13 patients), 35-49 years in 53.4% (151 patients), 50-70 years in 59.2% (321 patients), &gt;70 years in 60.7% (54 patients), compared with the MINDACT study where they were found respectively 24 % (RR 1.97 (1.13-3.43 p= 0.038), 44.1% (RR 1.20 (1.06-1.37 p= 0.006), 48.1% (RR 1.23 p &lt;0,001), 61.5%. (RR 0.98 (0.69-1.39) p= 0.936). When assessed according to menopausal status, the low genomic risk represented 52.7% (164 patients, 43% in MINDACT, RR 1.23 (1.09-1.39 p =0.002)) in premenopausal and 59.3% (375 patients, 48% in MINDACT, RR 1.23(1.14-1.33) p &lt;0,001) postmenopausal.CONCLUSION: The evaluation of the results of the AGEMA-BRA study, when compared to the MINDACT study, demonstrated a higher prevalence of patients with low genomic risk in all age groups (except &gt;70 years), indicating that the possibility of treatment de-escalation is even more significant in the Brazilian population than in the European population. Evaluation of outcomes regarding relapse-free survival and overall survival, an ongoing study, is necessary to confirm the data obtained. ABSTRACT The partnering of a β-lactam with a β-lactamase inhibitor is a highly effective strategy that can be used to combat bacterial resistance to β-lactam antibiotics mediated by serine β-lactamases (EC 3.2.5.6). To this end, we tested two novel penem inhibitors against OXA-1, a class D β-lactamase that is resistant to inactivation by tazobactam. The Ki of each penem inhibitor for OXA-1 was in the nM range (Ki of penem 1, 45 ± 8 nM; Ki of penem 2, 12 ± 2 nM). The first-order rate constant for enzyme and inhibitor complex inactivation of penems 1 and 2 for OXA-1 β-lactamase were 0.13 ± 0.01 s−1 and 0.11 ± 0.01 s−1, respectively. By using an inhibitor-to-enzyme ratio of 1:1, 100% inactivation was achieved in ≤900 s and the recovery of OXA-1 β-lactamase activity was not detected at 24 h. Covalent adducts of penems 1 and 2 (changes in molecular masses, +306 ± 3 and +321 ± 3 Da, respectively) were identified by electrospray ionization mass spectrometry (ESI-MS). After tryptic digestion of OXA-1 inactivated by penems 1 and 2, ESI-MS and matrix-assisted laser desorption ionization-time-of-flight MS identified the adducts of 306 ± 3 and 321 ± 3 Da attached to the peptide containing the active-site Ser67. The base hydrolysis of penem 2, monitored by serial 1H nuclear magnetic resonance analysis, suggested that penem 2 formed a linear imine species that underwent 7-endo-trig cyclization to ultimately form a cyclic enamine, the 1,4-thiazepine derivative. Susceptibility testing demonstrated that the penem inhibitors at 4 mg/liter effectively restored susceptibility to piperacillin. Penem β-lactamase inhibitors which demonstrate high affinities and which form long-lived acyl intermediates may prove to be extremely useful against the broad range of inhibitor-resistant serine β-lactamases present in gram-negative bacteria.

ABSTRACT We describe here an ∼40-kDa Plasmodium vivax tryptophan-rich antigen (PvTRAg40) which contains 321 amino acids and 11.4% tryptophan residues. This protein shows 65% homology (35% identity) with the previously described PvTRAg, besides sharing 23 of 27 positionally conserved tryptophan residues and similar genomic organization. The nucleotide sequence of the entire tryptophan-rich domain of PvTRAg40 was identical among 35 P. vivax clinical isolates. The protein is expressed by ring, trophozoite, and schizont stages of the parasite. The cDNA covering exon 2 of PvTRAg40 was cloned and expressed in the pPROEXHTa vector, and recombinant protein was purified. A high humoral immune response (90.7% seropositivity; n = 43) against this recombinant protein was detected in humans during the course of natural P. vivax infection. Eighty percent of the total of 20 P. vivax-exposed individuals exhibited lymphoproliferative responses against this antigen. The T cells of these individuals produced larger amounts of interleukin-12 (IL-12), IL-4, and IL-10 than gamma interferon and tumor necrosis factor alpha cytokines in response to the recombinant protein. Production of Th2-biased cytokines, conserved T- and B-cell epitopes, and an enhanced humoral immune response indicate that PvTRAg40 could possibly induce antibody-mediated immune protection against infection.

ABSTRACT A cohort of 321 children was followed from birth up to 2 years of age to determine the incidence of enterotoxigenic Escherichia coli (ETEC) in Bangladesh. The average number of diarrheal days and incidence rates were 6.6 and 2.3/child/year, respectively. ETEC was the most common pathogen and was isolated in 19.5% cases, with an incidence of 0.5 episode/child/year. The prevalence of rotavirus diarrhea was lower (10%). ETEC expressing the heat-stable enterotoxin (ST) was predominant. Strains isolated from diarrheal cases were positive for colonization factors (CFs) in higher frequency (66%) than from healthy children (33%) (P < 0.001). The heat-labile toxin (LT)-positive strains from healthy children were more often CF negative (92%) than those isolated from children with diarrhea (73%) (P < 0.001). In children with symptomatic or asymptomatic infections by CFA/I, CS1 plus CS3, CS2 plus CS3, or CS5 plus CS6 strains, a repeat episode of diarrhea or infection by the homologous CF type was uncommon. Repeat symptomatic infections were noted mostly for LT- and ST-expressing ETEC. ETEC diarrhea was more prevalent in children in the A and AB groups than in those in the O blood group (P = 0.032 to 0.023). Children with ETEC diarrhea were underweight and growth stunted at the 2-year follow-up period, showing the importance of strategies to prevent and decrease ETEC diarrheal morbidity in children.

ABSTRACT UL9, a superfamily II helicase, is a multifunctional protein required for herpes simplex virus type 1 replication in vivo. Although the C-terminal 317-amino-acid DNA binding domain of UL9 exists as a monomer, the full-length protein behaves as a dimer in solution. Thus, it has been assumed that the N-terminal 534 residues contain a region necessary for efficient dimerization and that UL9 dimers are in a head-to-head configuration. We recently showed, however, that residues in the N terminus could modulate the inhibitory properties of UL9 by decreasing the DNA binding ability of the C terminus (S. Chattopadhyay and S. K. Weller, J. Virol. 80:4491-4500, 2006). We suggested that a direct interaction between the N- and C-terminal portions of UL9 might exist and serve to modulate the DNA binding activities of the C terminus. In this study, we used a coimmunoprecipitation assay to show that the N-terminal portion of UL9 can indeed directly interact with the C terminus. A series of truncation mutant proteins were used to show that a region in the N terminus between residues 293 and 321 is necessary for efficient interaction. Similarly, a region in the C terminus between residues 600 and 800 is required for this interaction. The simplest model to explain these data is that UL9 dimers are oriented in a head-to-tail arrangement in which the N terminus is in contact with the C terminus.

AbstractBackgroundChildhood GH deficiency, suspected in the presence of decreased height velocity and short stature, is usually characterized by low IGF-I and IGF-binding protein-3 (IGFBP-3) serum concentrations and is conventionally confirmed by diminished GH peak responses to pharmacological stimuli.ObjectiveWe evaluated the agreement between different IGF-I (IGFBP-3) assays in predicting GH deficiency and tested whether variability between growth factor screening and pharmacological testing could be diminished by reassessment of growth factor and GH peak concentrations in a single laboratory.DesignUsing the Tuebingen IGF-I (IGFBP-3) RIA, 317 (321) sera from children evaluated for growth disorders in 19 centers were reanalyzed. In 103 children with insulin hypoglycemia and arginine tests, we evaluated how the association between the outcome of growth factor screening and functional testing would change if different assays were employed.ResultsLocally measured IGF-I correlated better than IGFBP-3 with the results of the central laboratory (Tuebingen) assay (slope of the regression curve 1.05; 95% confidence interval (95% CI) 1.01–1.1 versus 1.18; 95% CI 1.09–1.3). Agreement between local and central laboratory assays in predicting GH deficiency was better for IGF-I than for IGFBP-3 assays (κ=0.59 versus κ=0.47). The poor agreement between growth factor screening and GH pharmacological testing was not improved when hormone concentrations were remeasured in the central laboratory (κ local=−0.0031, central=0.12).ConclusionsIn children with impaired growth, growth factor screening reflects different aspects of GH insufficiency than does functional testing. Agreement between these approaches is poor and could not be improved by reduction of assay-related variability.

PurposeThe purpose of this study is to investigate the determinants of consumers' intention to adopt or continue to use Internet-only banks based on the benefit–risk framework and network externality theory. It also examines the difference in the determinants between pre- and postadoption stages of innovation.Design/methodology/approachThe proposed research model was tested by using online survey data collected from a South Korean sample, which was divided into two subgroups of 321 nonadopters and 351 existing users.FindingsIn both pre- and postadoption stages, the number of services provided and trust had a significant positive impact on consumers' behavioral intentions, while security risks had a negative impact. Critical mass in the preadoption stage and convenience and economic efficiency in the postadoption stage had positive effects on consumer's adoption intention and continuance intention, respectively.Practical implicationsInternet-only banks must reduce the security risks for consumers and increase their trust. In addition, to facilitate the adoption of nonadopters, focus should be on securing a critical mass; on the other hand, to promote the continued use of existing users, the focus should be on enhancing benefits such as convenience and economic efficiency.Originality/valueThe results of this study confirm the influence of network externalities on consumers' adoption and use of financial technology services and show differences in consumer decision-making according to the innovation diffusion process.

AbstractThe human T-cell leukemia virus type 2 (HTLV-2), an oncogenic retrovirus closely related to HTLV-1, produces a lifelong infection whose possible association to certain human diseases is still debated. Although some viral products can influence the expression and action of cellular genes, very little is known about the molecular mechanisms involved. Here we show that the AIR-1-encoded human major histocompatibility complex (MHC) class II transactivator (CIITA) strongly inhibits viral replication, but not virus entry, in human B- and T-cell susceptible targets. This effect results from CIITA inhibiting the Tax-mediated transactivation of the HTLV-2 long-term repeat. Further molecular analysis shows that the N-terminal region of CIITA encompassing the first 321 amino acids is responsible for the inhibitory effect on viral replication. This region is crucial for the transactivation of human MHC class II genes and includes the activation domain as well as domains interacting with coactivators that also are used by the viral transactivator Tax to modulate cellular functions. These results represent the first evidence that a cellular transcriptional activator, controlling the coordinate expression of the entire family of MHC class II antigen-presenting molecules, inhibits HTLV-2 viral replication by a distinct mechanism. In this new role CIITA may represent a new tool for therapeutic strategies aimed at counteracting HTLV-2 replication and spreading. (Blood. 2004;103:995-1001)

Purpose This study aims to check which scale of love brand developed by Carroll and Ahuvia (2006) or Mohammadian and Karimpour (2014) have better adhesion to the perception of Aspirin love brand, that is, a functional brand. Design/methodology/approach The conceptual models and associated hypotheses are tested with a sample of 321 consumers. Data were analyzed through a structural equation model. Findings The results demonstrate strong relationships between the three antecedents (brand engagement, confidence and overall attitude) and brand love and between brand love and its consequences (brand loyalty, positive word of mouth and brand purchase intention) in both scales. Research limitations/implications The main limitation of the study relates to the sample, which is only of Portuguese consumers and does not match the main socio-demographic characteristics of the population. Therefore, the study should be seen as exploratory on the brand love in the case of functional brands. Practical implications The knowledge that a consumer can establish an emotional relationship with a functional brand, in a highly competitive sector as the pharmaceutical sector, in particular in drugs that are sold without a prescription, can help managers in defining their communication strategy appealing to the emotions and long-term involvement with the consumed. Originality/value Few studies about the background of the brand love and those that exist are linked to hedonic product categories and self-expressive brands. It is the first time that the brand love for a medicine is evaluated.

Purpose The purpose of this paper is to explain the multi-stage dynamic process of financial distress. An attempt is made to explore multiple adverse heterogeneous events of financial distress leading a firm closer to bankruptcy progressively. Design/methodology/approach Sample comprises 321 ongoing, 54 suspended and 91 delisted non-financial firms from Pakistan Stock Exchange. Financial distress is segregated into three stages, i.e. profit reduction, mild liquidity (ML) and severe liquidity (SL). Flow diagrams are used to explain the transition of healthy firms through proposed stages of financial distress. Findings Results showed that firms liquidated/winding-up by court documented SL problems and closed their operations well before the delisting year. It is found that healthy firms are more likely to face SL when faced ML problem at first stage. Distressed firms can recover to a healthy position at any stage, however after approaching to SL, recovery is less expected. Practical implications The proposed process will provide a foundation for future studies to develop more relevant, robust and accurate early warning system of corporate failure that will help stakeholders to respond potential crisis accordingly and timely. Originality/value Previously, most of the studies used the ex post definition of bankruptcy that is criticized due to the contextual application, sample bias and non-segregation by the degree of liquidity problems. The originality of the proposed ex ante model is its segregation into a three-stage process that can be generalized regardless of specific bankruptcy law.

Purpose The purpose of this paper is to illustrate the maintenance prioritising for facilities services in high-rise residential buildings in Peninsular Malaysia. Maintenance prioritisation is becoming more prominent in the building maintenance industry due to budget constraints, poor maintenance management and to yield better maintenance performance. Design/methodology/approach Two main categories with eleven facilities services that require maintenance were identified through extensive literature review. A total of 321 returned questionnaires were analysed to distinguish the relationship between the maintenance priority and cost variance. Semi-structured interviews were then conducted to validate the findings. Findings The findings revealed that five essential facilities services were significantly correlated to cost variance and a regression model which examines the probability of over-budget was developed. Meanwhile, the interviews recognised that maintenance prioritisation has impact towards maintenance cost. Research limitations/implications This research focusses on the maintenance priorities of facilities services and their effects to maintenance cost. However, it is undeniable that the maintenance cost can be affected by other factors, contributing to a lower percentage of the total variance in the regression model. Thus, it creates research opportunity to study the factors (i.e. manpower, materials, wear and tear, etc.) affecting the variance of maintenance cost. Practical implications This study is useful to property managers in efforts to enhance the cost performance via appropriate maintenance prioritisation. The essential facilities services should be highly prioritised compared to the value-added facilities services. Originality/value The paper signifies the importance of maintenance prioritisation. It serves as a guide to plan and execute maintenance planning in a more logical way within budget and time constraints.

ABSTRACT Very little is known about the role that evolutionary dynamics plays in diseases caused by mammalian DNA viruses. To address this issue in a natural host model, we compared the pathogenesis and genetics of the attenuated fibrotropic and the virulent lymphohematotropic strains of the parvovirus minute virus of mice (MVM), and of two invasive fibrotropic MVM (MVMp) variants carrying the I362S or K368R change in the VP2 major capsid protein, in the infection of severe combined immunodeficient (SCID) mice. By 14 to 18 weeks after oronasal inoculation, the I362S and K368R viruses caused lethal leukopenia characterized by tissue damage and inclusion bodies in hemopoietic organs, a pattern of disease found by 7 weeks postinfection with the lymphohematotropic MVM (MVMi) strain. The MVMp populations emerging in leukopenic mice showed consensus sequence changes in the MVMi genotype at residues G321E and A551V of VP2 in the I362S virus infections or A551V and V575A changes in the K368R virus infections, as well as a high level of genetic heterogeneity within a capsid domain at the twofold depression where these residues lay. Amino acids forming this capsid domain are important MVM tropism determinants, as exemplified by the switch in MVMi host range toward mouse fibroblasts conferred by coordinated changes of some of these residues and by the essential character of glutamate at residue 321 for maintaining MVMi tropism toward primary hemopoietic precursors. The few viruses within the spectrum of mutants from mice that maintained the respective parental 321G and 575V residues were infectious in a plaque assay, whereas the viruses with the main consensus sequences exhibited low levels of fitness in culture. Consistent with this finding, a recombinant MVMp virus carrying the consensus sequence mutations arising in the K368R virus background in mice failed to initiate infection in cell lines of different tissue origins, even though it caused rapid-course lethal leukopenia in SCID mice. The parental consensus genotype prevailed during leukopenia development, but plaque-forming viruses with the reversion of the 575A residue to valine emerged in affected organs. The disease caused by the DNA virus in mice, therefore, involves the generation of heterogeneous viral populations that may cooperatively interact for the hemopoietic syndrome. The evolutionary changes delineate a sector of the surface of the capsid that determines tropism and that surrounds the sialic acid receptor binding domain.

Purpose The purpose of this study is to examine the influence of workplace happiness and work engagement on employee service innovative behavior from the perspective of positive psychology. The study also examines if work engagement mediates the relationship between workplace happiness and employee service innovative behavior. Finally, it investigates how co-worker socializing and the service climate of the organization moderate the relationship between work engagement and employee service innovative behavior. Design/methodology/approach The study used survey data from 321 frontline employees working in the service sector in the United Arab Emirates (UAE). Six major hypotheses were established and examined using the SPSS Statistics V22.0 Process. The measurement model was analyzed using Amos 22. Findings Workplace happiness and work engagement are found to be important factors affecting employee service innovative behavior. Workplace happiness influences employee service innovative behavior directly and indirectly through work engagement. Both service climate and co-worker socializing play a significant moderating role in the relationship between work engagement and employee service innovative behavior among frontline employees. Interestingly, service climate erodes this relationship while co-worker socializing enhances it. Practical implications This study provides guidelines for managers and practitioners in the service industry to promote frontline employee service innovative behavior. Specifically, the findings provide guidance for decision-makers on how to use workplace happiness to trigger the innovative service behaviors of frontline employees, taking into consideration the conditional role of service climate and co-worker socializing. Originality/value The literature on factors affecting the service innovative behavior of frontline employees in the service sector from the perspective of positive psychology is limited, especially in the context of the UAE. The data, framework and outcomes of this research address this gap and contribute to the current body of knowledge. Specifically, the study contributes to the broaden-and-build theory of positive emotions in the field of positive psychology by validating the applicability of the theory in a wider organizational context.

Abstract Objective To evaluate the role of time since injury on the factor structure of the Post-Concussion Symptom Scale (PCSS) for athletes after sustaining a sport-related concussion (SRC). Methods Adolescent athletes (n = 782) were dichotomized based on time since injury: 0–7 (EARLY; n = 321, age: 15.4 ± 1.9 years, 51.7% female), 8–14 (MIDDLE; n = 281, age: 15.8 ± 2.2 years, 54.8% female) and 15–30 days (LATE; n = 180, age: 15.6 ± 1.8 years, 52.8% female). An EFA was performed on the 22 variables of the PCSS for each of the three cohorts (primary factor loading of 0.6 to retain each item). Results EARLY had a five-component model (64% of total variance): 1) GLOBAL (18.0%; headache/dizziness/photosensitivity/phonosensitivity/difficulty concentrating), 2) FATIGUE/HYPERSOMNIA (16.4%; fatigue/drowsiness/mental fogginess/slowed down/hypersomnia), 3) AFFECTIVE (12.6%; more emotional/sadness), 4) INSOMNIA (9.0%; insomnia/trouble falling asleep), 5) SOMATIC (8.0%; vomiting/numbness). MIDDLE had a four-component model (63.1% of variance): 1) GLOBAL (19.4%; photosensitivity/headache/phonosensitivity/nausea/dizziness), 2) INSOMNIA/AFFECTIVE (14.4%; insomnia/more emotional/nervousness/trouble falling asleep), 3) SOMATIC/MEMORY (12.2%; difficulty remembering/numbness), and 4) HYPERSOMNIA (12.0%; drowsiness/hypersomnia). LATE had a four-component model (65.7% of variance): 1) VESTIBULAR/OCULAR (18.2%; vision/difficulty remembering/balance/fogginess/dizziness), 2) MIGRAINE (16.6%; photosensitivity/phonosensitivity/fatigue/headache), 3) AFFECTIVE (16.1%; sadness/nervousness/more emotional), and 4) INSOMNIA (7.6%; insomnia/trouble falling asleep). Conclusions The results of this study suggest greater time since injury modifies symptom factor structure in adolescent athletes with SRC. Specifically, symptom factors become more distinct with longer time since injury before first clinical visit. A GLOBAL symptom factor was observed for both EARLY and MIDDLE, but not in LATE. SOMATIC (i.e., numbness) was observed in EARLY/MIDDLE, but not LATE. Symptom factors in LATE seem to mirror concussion clinical sub-types from previous research (e.g., vestibular/ocular, migraine, affective, sleep).

Abstract INTRODUCTION The CeTeG/NOA-09 trial demonstrated superior median overall survival (mOS, 48.1 months) in MGMT-methylated glioblastoma treated with lomustine/temozolomide compared to temozolomide. We retrospectively analyzed an off-study cohort of patients treated with lomustine/temozolomide to gather real-world data on this new regimen. METHODS Adult patients from 20 centers in Germany, Austria and Switzerland were included. Inclusion criteria were MGMT-methylated IDHwt glioblastoma newly diagnosed prior to end of 2020, and lomustine/temozolomide treatment as part of first-line therapy. RESULTS 321 patients with a median age of 57 years (range, 21-78) and a median follow-up of 19.9 months were included. In the whole cohort, mOS was 41.0 months (95%CI, 33.0 – not reached). In patients starting lomustine/temozolomide immediately upon initiation of radiotherapy strictly following the CeTeG protocol (88%), mOS was 52.8 months (35.8 – not reached) as compared to 24.6 months (17.6 – not reached) in patients starting lomustine/temozolomide after completion of radiotherapy/concomitant temozolomide (12%, logrank test: p = 0.06). Patients with a KPS &lt; 80 had a shorter mOS of 19.7 months (95%CI, 16.6 – not reached) compared to 41.0 months (33.0 – not reached, p = 0.009) in KPS 80-100. Gross total resection (GTR, 53.9%) was associated with longer mOS (52.8 months, 95%CI 24.1 – not reached) compared to partial resection/biopsy (30.5 months, 95%CI 36.8 – not reached, p=0.004). Multivariable Cox regression analysis confirmed GTR (HR 0.66, p = 0.033) and younger age ( ≤ 50 years: HR 0.42, p = 0.001), but not KPS (80-100 vs. lower: HR 0.66, p = 0.12) as independent prognostic factors. DISCUSSION In this real-world multicenter cohort, survival was similar to the promising results of CeTeG/NOA-09. Further analyses should investigate a potentially reduced benefit from lomustine/temozolomide in patients with low KPS/no GTR and a possible detrimental effect from deferred lomustine/temozolomide initiation. The median follow-up is admittedly short, updated data will be presented.

Introdução: a problemática central deste artigo é o cuidado. O cuidado não como expressão única do tecnicismo, mas em suas múltiplas dimensões. Objetivo: discutir o cuidado numa perspectiva ampliada, envolvendo suas expressões subjetivas, espirituais, corporais e ambientais. Metodologia: trata-se de uma revisão bibliográfica sobre as dimensões do cuidado, tendo como base autores da filosofia, sociologia, psicologia e enfermagem. Resultados: as múltiplas dimensões do cuidado foram estruturadas em cinco eixos epistemológicos, quais sejam, cuidar de si, cuidar do outro, cuidar da terra, cuidar da psique e cuidar do espírito. Conclusão: A partir das discussões desta investigação, é perceptível que os indivíduos, em sua magnitude, precisam adquirir uma nova postura diante do cenário atual que o mundo está passando, pois a problemática apontada requer uma visão multidimensional do cuidado.Palavras-chave: cuidado, enfermagem, tecnicismo e multidimensionalidade. Abstract: Introduction: the central proprosition of this article is the care. Care is not a unique expression of technicality, but in its multiple dimensions. Objective: to discuss care in an extended perspective, involving its subjective, spiritual, corporeal and environmental expressions. Methodology: this is a bibliographical review on the dimensions of care, based on authors of philosophy, sociology, psychology and nursing. Results: the multiple dimensions of care were structured in five epistemological axes, namely, caring for oneself, caring for others, caring for the earth, caring for the psyche and caring for the spirit. Conclusion: From the discussions of this research, it is noticeable that individuals, in their magnitude, need to acquire a new posture in view of the current scenario that the world is going through, since the problematic pointed out requires a multidimensional view of care. Chinese radish (Raphanus sativus) is an important vegetable grown widely in China. In 2010 to 2013, surveys for Leptosphaeria spp. on cruciferous vegetables were conducted in 17 counties in Hubei Province, China. Black leg symptoms on tuber roots and basal stems of radish were observed in Hanchuan, Jingmen, and Macheng counties. Disease incidence ranged from 2 to 25% in 10 surveyed radish fields. Five fungal isolates were obtained from diseased radish plants by surface-sterilizing radish tissue (5% NaOCl for 90 s, and then rinsed in sterilized water three times) and plating onto potato dextrose agar (PDA) plates incubated at 20°C. The isolations produced fluffy white colonies with a yellow pigment, and black-brown, globose pycnidia with pink conidial ooze formed after 10 days. Pycnidia were 150 to 200 × 80 to 100 μm. Conidia were hyaline, cylindrical, and 4 to 5 × 2 μm. The cultural and morphological characteristics of the isolates matched the description for Phoma lingam, anamorph of Leptosphaeria maculans and L. biglobosa (3). All five isolates were identified by PCR assay using the species-specific primers LbigF, LmacF, and LmacR (1), and isolate HCLB-1 was identified further by PCR cloning and analysis of the sequences coding for actin, β-tubulin, and the internal transcribed spacer (ITS) region of ribosomal DNA (3,4). Genomic DNA was extracted from mycelium of each isolate harvested from 7-day-old cultures in V8 broth using the CTAB method (5). A 444-bp DNA fragment was detected by PCR assay, suggesting that all five isolates belonged to L. biglobosa rather than L. maculans as the latter generates a 331-bp DNA fragment (1). The HCLB-1 sequences for ITS (587 bp, GenBank Accession. No. KC880981), actin (899 bp, KF307762), and β-tubulin (432 bp, KF220296) genes were 99 to 100% identical to those of L. biglobosa isolates in GenBank. All five isolates were tested for pathogenicity on R. sativus cultivars Duan Ye 13 and Qi Ye Hong. Cotyledons of 10-day-old radish seedlings and post-harvest mature roots were wounded using a sterilized needle, and 10 μl of a conidial suspension (1 × 107 conidia/ml) of each isolate was pipetted onto the wounded area on each cotyledon or root, respectively, with 12 cotyledons (= 24 wounded sites) and 1 root (= 6 wounded sites) inoculated/isolate. One wounded root and 12 wounded cotyledons inoculated with water were used as control treatments. Treated roots and seedlings were incubated at 20°C and 100% RH in the dark for 7 days, and under a 12 h light/12 h dark cycle for 12 days, respectively. While the control roots and cotyledons remained asymptomatic, the roots and cotyledons inoculated with all five test isolates formed black cankers and necrotic lesions, respectively, in the inoculated wounds. A fungus re-isolated from symptomatic roots and cotyledons resembled the original isolates in colony morphology and the 444-bp DNA fragment detected by PCR assay. No fungus was isolated from control seedlings or roots. Thus, L. biglobosa appears to be the causal agent of black leg observed on radish in Hubei, China. ABSTRACT: This study aims to examine the effect of health, education, and labor force moderated by urban population on the Human Development Index. This study uses a quantitative approach and a Moderated Regression Analysis technique with a sample of 25 OIC countries from 2010-2019 taken by purposive sampling method. The results showed that health, education, and the labor force affected the Human Development Index. While the urban population moderates the effect of health and the workforce on the Human Development Index, the urban population does not moderate the effect of education on the Human Development Index. This study has a novelty in the results of the study, namely the basic education variable which has a significant but negative effect on the Human Development Index. This research is limited to a sample of OIC member countries because more than 30% of poor countries with low income according to World Bank data are in OIC member countries, so it is hoped that this research will be able to provide references related to increasing the human development index to save them from poverty. Penelitian ini menggunakan pendekatan kuantitatif dan teknik Moderated Regression Analysis dengan sampel 25 negara OKI dari tahun 2010-2019 yang diambil dengan metode purposive sampling. Hasil penelitian menunjukkan kesehatan dan angkatan kerja berpengaruh positif signifikan terhadap Human Development Index, namun pendidikan berpengaruh negatif signifikan terhadap Human Development Index. Urban population memoderasi pengaruh antara kesehatan dan angkatan kerja terhadap Human Development Index, namun urban population tidak memoderasi pengaruh pendidikan terhadap Human Development Index. Penelitian ini memiliki kebaruan dalam hasil penelitian, yaitu variabel pendidikan dasar yang justru berpengaruh namun negatif signifikan terhadap Human Development Index. Penelitian ini terbatas dilakukan pada sampel negara anggota OKI sebab lebih dari 30% negara miskin dengan pendapatan rendah menurut Data Bank Dunia adalah ada pada negara anggota OKI, sehingga diharapkan penelitian ini mampu memberikan referensi terkait peningkatan indeks pembangunan manusia untuk menyelamatkannya dari kemiskinan. El vermifiltro con la especie Eisenia Foetida para tratar aguas residuales de uso doméstico tuvo un caudal de 217 L/d, una Carga Hidráulica (CH) de 150 ml/m2.min y Tiempo de Retención Hidráulico (TRH) de 5.7 días. Se analizaron muestras de agua residual sin tratamiento y con tratamiento, en tres fechas, cada 7 días y se obtuvieron los siguientes resultados: Sólidos Suspendidos Totales (SST) de 1708 mg/L a 32 mg/L en la primera fecha, 321 mg/L a 34 mg/L en la segunda y 154 mg/L a 2.5 mg/L en la tercera fecha. Improving bituminous material using recycling waste (Crumb rubber) as modifier was emphasized in this paper. Crumb rubber (CR) waste was added by volume to the bitumen in 0, 5, and 7 %. The volumetric properties tests were conducted using the Marshall Test (ASTM D1559), others includes Characterization of Binder and Aggregate materials in HMA, (FTIR Analysis), Fundamental mechanical properties tests, also Rheological properties according with ASTM D7369-11, Dynamic Modulus Test, according with AASHTO R83-17, Rutting Test according with AASHTO T324-04, Fatigue Cracking Evaluation according with AASHTO TP124 and Flexural Beam Fatigue Beam Test according with AASHTO T 321-07. The result indicates that crumb rubber is very suitable as a modifier, with bitumen of 60/70 PEN grade. From the results, at the designed production temperature 135oC for unmodified 60/70 Bitumen, the modified admixed samples (0% and 5% CR) at the same binder content show a high void percentage for mixes (5% CR). This is an indication that the modified asphalt mixtures exhibited volumetric properties with higher voids and lower density due to change in viscosity caused by modification and requires compaction at higher temperature to be at par with regular unmodified bitumen grade. However, the air voids of mixtures fall within the specification range of 3% to 5%, and as such acceptable for use as a modifier in 60/70 PEN grade bitumen.

Abstract Background Blonanserin (BNS) had been undergoing post-market surveillance (PMS) since September 2018. Using the surveillance data, we did this analysis to assess the safety and effectiveness of different doses of BNS to explore a sufficient dose range of BNS in Chinese patients with schizophrenia (SZ). Methods A 12-week, prospective, observational, single-arm, multicenter, open-label PMS was conducted. In this analysis, we divided the patients from PMS into low, medium to high, and high dose groups based on the dose of BNS they received, with medium to high dose group being the focus. The Brief Psychiatric Rating Scale (BPRS) scores at week 2 or 4, 6 or 8, and 12 were calculated to evaluate the effectiveness of BNS in improving psychiatric symptoms. The safety of BNS was reported as the incidence of adverse drug reactions. Results 364 patients were included in the medium to high dose group, of which 321 completed the surveillance, with a dropout rate of 11.8%. The mean daily dose was 15.1 ± 1.92 mg. The BPRS total score was 50.1 ± 11.95 at baseline and decreased to 26.6 ± 7.43 at 12 weeks (P < 0.001). When compared with other groups, the median to high dose group achieved significantly more reduction in BPRS score at week 12 (P = 0.004 versus low dose and P = 0.033 versus higher dose). Extrapyramidal symptoms [EPS (46.4%)] were the most common adverse reactions in the medium to high group. The average weight gain during the surveillance was 0.5 ± 2.56 kg and prolactin elevation occurred in 2.2% patients. Most adverse reactions were mild. Conclusions BNS at medium to high doses (mean 15.1 mg/d) significantly improved symptoms of SZ and was well-tolerated. Most ADRs were mild, and the likelihood of causing metabolic side effects and prolactin elevations was low. Medium to high dose of BNS is a more potent treatment choice for SZ. AbstractBackgroundDysfunctional dopamine signaling and disruptions in neurotrophic support are potential exacerbators of Alzheimer’s disease (AD) pathophysiology. A single nucleotide polymorphism (SNP) in the dopamine transporter gene (rs6347 in DAT1/SLC6A3) is associated with greater ventricular expansion and lower scores on the Mini Mental State Exam (MMSE; Roussotte et al., 2015). A polymorphism in the brain derived neurotrophic factor gene (Val66Met in BDNF) is related to greater hippocampal atrophy and more severe episodic memory decline for amyloid positive Met carriers (Lim et al., 2013). However, it is unclear whether and how these polymorphisms directly relate to AD pathology.MethodWe tested a moderated mediation model (Fig. 1) involving both BDNF and DAT1 polymorphisms in a sample of cognitively normal older adults (n = 321) from the Alzheimer’s Disease Neuroimaging Initiative (ADNI). This mediation involved DAT1 (independent variable), b‐amyloid (Ab; mediator), and tau pathology (dependent variable) with BDNF as a moderator. Ab and tau were measured both cross‐sectionally and longitudinally with positron emission tomography (PET) tracers [18F]Florbetapir/[18F]Florbetaben and [18F]Flortaucipir, respectively.ResultA significant moderated mediation demonstrated that DAT1 and BDNF interacted to predict cross‐sectional Ab‐PET (p = .02) and tau‐PET (p = .04), such that carriers of both DAT1 C and BDNF Met alleles demonstrated greater pathology (Fig. 2), consistent with previous research associating these variants with AD symptoms. Longitudinal analyses replicated these cross‐sectional results, with DAT1 C and BDNF Met carriers exhibiting greater accumulation of tau pathology (p = .0002) and marginally increased Ab accumulation (p = .07).ConclusionOur analyses revealed novel interaction effects between genetic polymorphisms in the DAT1 and BDNF genes. These results suggest that carrying DAT1 CC and BDNF Met results in higher Ab, which then contributes to higher tau pathology. While our analyses do not demonstrate causality, these findings are consistent with current hypotheses of AD in which Ab drives increases in tau pathology (Karran et al., 2011).
