We investigate the case of disagreement between predicted and observed in-situ parameters of the recurrent high-speed solar wind streams (HSSs) existing for Carrington rotation (CR) 2118 (December 2011) in comparison with CRs 2117 and 2119. The HSSs originated at the Sun from a recurrent polar coronal hole (CH) expanding to mid-latitudes, and its area in the central part of the solar disk increased with the rotation number. This part of the CH was responsible for the equatorial flank of the HSS directed to the Earth. The time and speed of arrival for this part of the HSS to the Earth were predicted by the hierarchical empirical model based on EUV-imaging and the Wang-Sheeley-Arge ENLIL semi-empirical replace model and compared with the parameters measured in-situ by model. The predicted parameters were compared with those measured in-situ. It was found, that for CR 2117 and CR 2119, the predicted HSS speed values agreed with the measured ones within the typical accuracy of ±100 km s−1. During CR 2118, the measured speed was on 217 km s−1 less than the value predicted in accordance with the increased area of the CH. We suppose that at CR 2118, the HSS overtook and interacted with complex ejecta formed from three merged coronal mass ejections (CMEs) with a mean speed about 400 km s−1. According to simulations of the Drag-based model, this complex ejecta might be created by several CMEs starting from the Sun in the period between 25 and 27 December 2011 and arriving to the Earth simultaneously with the HSS. Due to its higher density and magnetic field strength, the complex ejecta became an obstacle for the equatorial flank of the HSS and slowed it down. During CR 2117 and CR 2119, the CMEs appeared before the arrival of the HSSs, so the CMEs did not influence on the HSSs kinematics.

Abstract Factor V (fV) binds to phospholipid (PL) membranes via a motif localized to the C2 domain. We and others have shown that PL binding is mediated by two pairs of hydrophobic residues, each displayed at the tips of β-hairpin turns. The homologous hydrophobic residues in the C2 domain of factor VIII also contribute to PL binding. We hypothesized that the solvent-exposed hydrophobic residues of the fV C2 domain make specific contacts that influence membrane affinity and activity of fV. To test this hypothesis we have prepared fVIII/fV hybrid mutants in which amino acid(s) of the fV C2 domain were changed to the homologous residues of fVIII (Mutants #1 W/W 2063/2064 M/F, #2 L/S 2116/2117 L/L, and #3 W/W/S 2063/2064/2117 M/F/L). Mutants were expressed in COS-1 cells and purified by FPLC. The specific activity of the fV/FVIII hybrids #2 and #3 exceeded those of wild type factor V by approx. 10-fold and approx. 4-fold, respectively, in a prothrombin time assay with factor V deficient plasma. Apparent PL affinites were evaluated in a prothrombinase complex assay with limiting phospholipid. The apparent affinities are 9.8 and 21-fold higher than wild type factor V for mutants #2 and #3 which contain the S→L change in the second hydrophobic spike. An unexpected result was that mutants 1–3 supported prothrombinase activity in the absence of added phospholipid, in contrast to wild type fV. We hypothesized that this activity resulted from phospholipid that was not dissociated from fV during purification. This hypothesis was supported because activity was eliminated by incubation of the factor V mutants with phospholipase A2 or by incubation with lactadherin. The tenacity of the phospholipid binding was further investigated by washing immobilized wild type fV and fV mutants with CHAPS prior to elution from an FPLC column. fV mutants #2 and #3 retained activity after the CHAPS wash, free of added PL and activity remained inhibitable by lactadherin, further illustrating the tenacious phospholipid affinity. We utilized Mutants #1 & 2 in the absence of added PL to evaluate the mechanism through which soluble phosphatidylserine with 6-carbon acyl chains (C6PS) enhances activity of the prothrombinase complex. In the absence of phospholipid vesicles C6PS enhanced activity of Mutant 1 equivalent to wild type fV (> 20 fold). However, enhancement of mutant 2 activity, which presumably retained phospholipid via residues 2116/2117, was < 3-fold. This suggests that C6PS functions to activate wild type fV, in part, by engaging the free LS 2116/2117 hydrophobic spike. Together, these data indicate that the hydrophobic spikes of factor V influence the specific activity of factor V, that the high affinity reversible binding of fV to phospholipid membranes is readily perturbed by mutations, and that activation of the prothrombinase complex by C6PS and phospholipid membranes likely involves engagement of amino acids 2116/2117.

Com o objetivo de avaliar a tolerância dos híbridos de milho BRS 3060, 3101, 2114 e 2110 a doses crescentes do herbicida nicosulfuron, foi instalado um ensaio de campo na área experimental do Centro de Pesquisa da Embrapa Milho e Sorgo, município de Sete Lagoas-MG, durante o ano agrícola 1998/1999. A densidade de plantio foi de 50.000 plantas por hectare, e as doses do nicosulfuron foram de 50, 60 e 70 g i.a. ha-1, mais um tratamento testemunha que não recebeu aplicação do produto, mas que permaneceu limpo durante todo o ciclo da cultura. O delineamento experimental adotado foi em blocos ao acaso com três repetições, em esquema fatorial 4 x 4. A aplicação do produto foi realizada quando as plantas de milho se encontravam no estádio de quatro a seis folhas, ou seja, 20 dias após emergência das plantas. As avaliações de fitotoxicidade foram feitas aos 7, 14 e 21 dias após aplicação dos tratamentos herbicida. Somente o BRS 3060 permaneceu com efeito fitotóxico muito leve nas avaliações realizadas aos 14 e 21 dias após aplicação, sem comprometer o rendimento de grãos. Os resultados indicam que o herbicida nicosulfuron pode ser aplicado para o controle de plantas daninhas em híbridos de milho BRS 3060, 3101, 2114 e 2110 nas doses de 50 a 70 g i.a. ha-1, sem causar danos significativos ao desenvolvimento e à produção da cultura.

Thirty-three advanced breeding lines of peanut were evaluated during kharif, 2019 using Mahalanobis D2 statistics for 19 characters including physiological, yield and quality traits and were grouped into nine clusters, where cluster IV was the largest containing 11 genotypes followed by cluster III and II with 8 and 7 genotypes respectively. Highest inter-cluster distance was observed between cluster VII and IX (118.10) followed by cluster V and VI (113.29) and cluster V and IX (106.42), respectively. The maximum intra-cluster distance was observed in cluster IV (31.37) followed by cluster III (20.11) and II (13.17). Hundred kernel weight contributed maximum towards genetic divergence followed by sound mature kernel (%), specific leaf area at 45 DAS, oil content and plant height. The crosses viz., TCGS-2141 × TCGS-2117, Narayani × TCGS-1887, Narayani × TCGS-2117, Narayani × TCGS-2111 and TCGS-1887 × TCGS-2141 could be recommended to get desirable transgressive segregants for physiological, yield and quality traits.

The article provides an integral assessment of the competitiveness of Ukrainian-made cars (compared with the direct competitors on the automobile market). An integral assessment of the competitiveness of Ukrainian-made cars compared with the direct competitors on the automobile market (for ZAZ Lanos – Hyundai Accent and Kia Rio; for ZAZ Lanos Cargo – Bohdan-2310 pickup and IZH 27175; for ZAZ Sens – Geely CK and Bohdan-2110; for ZAZ Forza − Chevrolet Aveo and Hyundai Accent; for ZAZ Vida – Fiat Albea and Bohdan-2111; for Bohdan-2110 – Geely CK and ZAZ Sens; for Bohdan-2111 – Fiat Albea and ZAZ Vida; for Bohdan-2310 pickup – ZAZ Lanos Cargo and IZH 27175; for Bohdan 2312 – ZAZ Lanos Cargo and IZH 27175) is based on an expert method. The ranks (I; II; III) provided by 15 experts are subjected to the normalization procedure, and a single synthetic rank is formed from the normalized ranks according to a certain comparative criterion. Comprehensive indicator of the competitiveness of cars in terms of non-price parameters is formed from the obtained synthetic ranks according to the comparative criteria “quality”, “assortment (modifications)” and “service maintenance”. The integral indicator of the competitiveness of cars is obtained by synthesizing the values of complex indicators according to the price parameter and non-price parameters. The medium level of competitiveness of Ukrainian-made cars (an exception is ZAZ Lanos, which demonstrates a high level of competitiveness) on the automobile market is determined by: mostly constructive influence of the “Price” and “Service maintenance” parameters; mostly destructive influence of the “Quality” and “Assortment (modifications)” parameters. The prospects for increasing the competitiveness of Ukrainian-made cars are: maintaining of the competitive positions by the “Price” and “Service maintenance” parameters; improvement of competitive positions by the “Quality” and “Assortment (modifications)” parameters.

A resistência de híbridos de milho foi avaliada com relação ao ataque de Sitophilus zeamais em condições de armazenamento. Amostras de grãos de onze híbridos de milho, BRS 3003, BRS 1030, BRS 3123, BRS 2020, BRS 2223, BRS 2110, BRS 3060, BRS 2114, BRS 3151, BRS 1010 e BRS 1001, foram infestadas com adultos de S. zeamais e mantidas em condições de laboratório a 27°C e 12 horas de fotofase. Após trinta dias da infestação foram avaliados os seguintes parâmetros de resistência: índice de suscetibilidade, perda de massa seca dos grãos, número de insetos emergidos e ciclo biológico. Os híbridos mais suscetíveis ao ataque de S. zeamais foram: BRS 3003 e BRS 1030, enquanto que os mais resistentes foram: BRS 1010 e BRS 1001.

The crystal structure of strontium diarsenate has been reinvestigated from single-crystal X-ray diffraction data. In contrast to the previous determinations of this structure [Weilet al.(2009).Solid State Sci.11, 2111–2117; Edhokkaret al.(2012).Mater. Sci. Eng.,28, 012017] and to all isotypicA2B2O7compounds that crystallize in the space groupP41, the current redetermination revealed theP43enantiomorph of Sr2As2O7with a purity of 96.3 (8)%. The crystal structure is made up from two eclipsed As2O7diarsenate groups (symmetry 1) with characteristically longer As—O bridging bonds [1.756 (4)–1.781 (4) Å] than the terminal As—O bonds [1.636 (4)–1.679 (4) Å] and four Sr2+sites with coordination numbers ranging from seven to nine. The building units are arranged in sheets parallel to (001).

BackgroundEnterococcus faecalisandEnterococcus faeciumare ubiquitous opportunistic pathogens found in the guts of humans and farmed animals. This study aimed to determine the occurrence, antimicrobial resistance, virulence, biofilm-forming ability and genotypes ofE. faecalisandE. faeciumfrom swine farms. Correlations between the genotypes, virulotypes, antibiotic resistance, and the environmental factors such as locality of farms and farm hygiene practice were explored.MethodsE. faecalisandE. faeciumstrains were isolated from the oral, rectal and fecal samples of 140 pigs; nasal, urine and fecal samples of 34 farmers working in the farms and 42 environmental samples collected from seven swine farms located in Peninsular Malaysia. Antibiotic susceptibility test was performed using the disk diffusion method, and the antibiotic resistance and virulence genes were detected by Polymerase Chain Reaction. Repetitive Extragenic Palindromic-Polymerase Chain Reaction and Pulsed-Field Gel Electrophoresis were performed to determine the clonality of the strains. Crosstab/Chi-square test and DistLM statistical analyses methods were used to determine the correlations between the genotypes, virulence factors, antibiotic resistance, and the environmental factors.ResultsA total of 211E. faecalisand 42E. faeciumwere recovered from 140 pigs, 34 farmers and 42 environmental samples collected from seven swine farms in Peninsular Malaysia. Ninety-eight percent of the strains were multidrug-resistant (resistant to chloramphenicol, tetracycline, ciprofloxacin and erythromycin). Fifty-two percent of the strains formed biofilms. Virulence genesefa, asaI, gelE,esp,cylandacegenes were detected. Virulence genesefaandasaI were most prevalent inE. faecalis(90%) andE. faecium(43%), respectively. Cluster analyses based on REP-PCR and PFGE showed the strains were genetically diverse. Overall, the strains isolated from pigs and farmers were distinct, except for three highly similar strains found in pigs and farmers. The strains were regional- and host-specific.DiscussionThis study revealed alarming high frequencies of multidrug-resistant enterococci in pigs and swine farmers. The presence of resistance and virulence genes and the ability to form biofilm further enhance the persistence and pathogenicity of the strains. Although the overall clonality of the strains were regionals and host-specific, strains with high similarity were found in different hosts. This study reiterates a need of a more stringent regulation to ensure the proper use of antibiotics in swine husbandry to reduce the wide spread of multidrug-resistant strains.

However, when assessing the general grounds of refusal, the specific function of certification must always be kept in mind. In three different cases, the Fifth Board of EUIPO decided that the certification marks Manuka honey and Bio-Mineralwasser (as word and figurative mark) are non-distinctive and descriptive.

Проблема и цель. Для достижения поставленной цели проведены стендовые испытания двигателя на определение его мощности и крутящего момента на колесе при различных значениях искрового зазора свечи зажигания. В результате определена оптимальная величина зазора свечи, при которой испытуемый двигатель имеет максимальные динамические характеристики. Методология. Для определения влияния изменения искрового зазора свечи на динамические параметры двигателя нами были проведены стендовые испытания двигателя ВАЗ 2112 при зазорах свечи от 0,3 до 1,8 мм с интервалом через 0,1 мм на стенде Cartec 2020 LPS 4WD по условиям, определенным ГОСТ 14846-81 «Двигатели автомобильные Методы стендовых испытаний». При проведении испытаний были использованы новые свечи NGK BCPR6E-11. В ходе испытаний в режиме Pmax автомобиль разгонялся на прямой передаче до максимально возможной скорости. После достижения предварительно заданного порогового значения скорости, стенд начинает оказывать сопротивление вращению колёс, имитируя реальные дорожные условия движения автомобиля. Измеренные в режиме Pmax значения представлены в виде графиков изменения мощности и крутящего момента. Результаты. С повышением искрового зазора свечи зажигания от 0,3 мм и больше мощность двигателя и крутящий момент на колесе плавно увеличиваются, достигая своих максимумов 97,2 л.с. и 290,3 Н·м соответственно при величине зазора 1,5 мм. При дальнейшем увеличении зазора происходит ухудшение динамических параметров двигателя. Это свидетельствует о том, что наилучшие динамические параметры двигатель имеет при величине межэлектродного зазора свечи, равной 1,5 мм, которая является оптимальной для двигателя ВАЗ-2112. Очевидно, при таком зазоре свечи между электродами образуется устойчивая длинная искра, способствующая наиболее полному сгоранию рабочей смеси, следовательно, и повышению динамических параметров двигателя. Заключение. Определен оптимальный размер межэлектродного зазора свечи зажигания NGK BCPR6E-11 для двигателя ВАЗ-2112, установленного на автомобиле ВАЗ-2111, равный 1,5 мм. С таким искровым зазором двигатель имеет максимальные динамические параметры при работе со свечами маркировки BCPR6E-11. Problem and purpose. To achieve this goal, bench tests of the engine were carried out to determine its power and torque on the wheel at various values of the spark gap of the spark plug. As a result, the optimal value of the spark plug gap was determined, at which the tested engine has maximum dynamic characteristics. Methods. To determine the effect of changing the spark plug gap on the dynamic parameters of the engine, we carried out bench tests of the VAZ 2112 engine with spark plug gaps from 03 to 1.8 mm with an interval of 0.1 mm on the Cartec 2020 LPS 4WD stand under the conditions defined by GOST 14846-81 "Automobile engines Bench test methods". During the tests, new NGK BCPR6E-11 spark plugs were used. During tests in Pmax mode, the car accelerated in direct gear to the maximum possible speed. After reaching the pre-set threshold speed, the stand begins to resist the rotation of the wheels, simulating the real road conditions of the car. The values measured in the Pmax mode are presented in the form of power and torque curves. Results. With an increase in the spark gap of the spark plug from 0.3 mm or more, engine power and torque on the wheel increase smoothly, reaching their maximums of 97.2 hp. and 290.3 Nm, respectively, with a gap of 1.5 mm. With a further increase in the gap, the dynamic parameters of the engine deteriorate. This indicates that the engine has the best dynamic parameters when the electrode gap of the spark plug is 1.5 mm, which is optimal for the VAZ-2112 engine. Obviously, with such a spark plug gap between the electrodes, a stable long spark is formed, which contributes to the most complete combustion of the working mixture, and, consequently, to an increase in the dynamic parameters of the engine. Conclusion. The optimal size of the interelectrode gap of the NGK BCPR6E-11 spark plug for the VAZ-2112 engine installed on the VAZ-2111 car was determined, equal to 1.5 mm. With such a spark gap, the engine has maximum dynamic parameters when working with candles marked BCPR6E-11.

The Thai specimen shows morphological characters similar to C. elok as follows: 12-14 supralabials, 10-11 infralabials, seven tubercles across midbody, 49 ventral scales, enlarged femoral scales absent, tubercles on forelimbs absent, ventrolateral fold poorly defined, and large tubercles of dorsolateral caudal rows. The species was previously known from the discovery made in Malaysia. This addition brings the number of Thai prehensile-tailed species to three. Morphological and distributional maps are provided.

Chinese hamster cells (V79) resistant to high concentrations of Cd2+ in the medium were obtained by using the procedure of Beach & Palmiter [(1981) Proc. Natl. Acad. Sci. U.S.A. 78, 2110-2114], which in mouse led to amplification of metallothionein (MT) genes and to an enrichment in cellular MT. The Cd-resistant V79 clones isolated were significantly more resistant than parental cells to oxidative stress by extracellular H2O2 or a mixture of H2O2 and superoxide anion (O2-) generated by xanthine oxidase plus acetaldehyde. On a per-cell basis, there was no difference between the two cells in their total H2O2-decomposing or O2-(-)dismutating activity. The most likely explanation is that an enrichment in MT content in the Cd-resistant cells was responsible for this effect, because of the antioxidant properties already described for this protein.

Nowadays the hybrid imaging technologies which combine the modern equipments of radiology and nuclear medicine play an important role in both the translational research process and clinical diagnostics. Among the routine diagnostic imaging procedures positron emission tomography and single photon emission computed tomography combined with computed tomography or magnetic resonance imaging currently belong to the most advanced techniques allowing that functional and morphological images can be superimposed on each other in the same position. The hybrid imaging equipments provide useful information about the pathological processes in the body due to their high sensibility and resolution. Furthermore, with the help of these imaging modalities we can get acquainted with the biochemical and pathobiochemical processes that are essential for understanding and treating diseases, or getting acquainted with the behaviour of a new drug candidate. With the help of the clinical and preclinical non-invasive in vivo molecular imaging systems the drug developing process can be shortened and its costs can be reduced. Orv. RESUMO A pinta preta do tomateiro é uma das principais doenças da cultura. Assim, o objetivo deste trabalho foi caracterizar 49 subamostras de tomateiro do Banco de Germoplasma de Hortaliças da Universidade Federal de Viçosa (BGH-UFV) quanto à pinta preta. O experimento ocorreu em Viçosa, MG, entre dezembro de 2007 e abril de 2008. Utilizou-se delineamento em blocos casualizados com três repetições e as testemunhas Débora, Fanny, Santa Clara e IPA-6. Realizaram-se cinco avaliações com base na severidade da doença. Com os valores de severidade calculou-se a área abaixo da curva de progresso da doença (AACPD), possibilitando uma melhor visualização dos resultados. Foi realizada a análise de variância, seguido do agrupamento das médias de AACPD pelo teste de Scott-Knott, a 5% de significância. As subamostras BGH-2109, BGH-2110, BGH-2115, BGH-2122, BGH-2134, BGH-2135 e BGH-2142 apresentaram menor média de AACPD em relação às testemunhas, podendo ser utilizadas em futuros programas de melhoramento.

Kota Solo saat ini perkembangannya sangat pesat dimana banyak bermunculan perhotelan, pertokoan serta perkantoran. Penghasilan masyarakat kota Solo setiap tahun meningkat sehingga pemakaian energi listrik juga semakin bertambah banyak. Letak pusat pembangkit tenaga listrik kepada konsumen jaraknya sangat jauh. Untuk menyalurkan energi listrik dari pusat pembangkit tenaga listrik ke konsumen terlebih dahulu menaikan tegangan dengan trafo step up kemudian ditransnisikan ke pusat-pusat beban listrik. Digardu induk tegangan diturunkan menggunkan trafo step down. Setelah tegangan energi listrik diturunkan pada gardu induk kemudian didistribusikan melalui trafo distribusi agar sampai kepada konsumen listrik. Selama penyaluran listrik dari pusat pembangkit tenaga listrik ke pelanggan listrik akan terjadi pengurangan muatan listrik. Faktor terjadi berkurangnya kapasitas tenaga listrik disebabkan ada sejumlah unsur seperti jarak penyaluran dari pusat pembangkit ke konsumen sangat jauh, ketaksimetrisan muatan listrik menyebabkan penghantar menghasilkan temperatur tinggi. Tujuan dari penelitian ini memanisfestasikan turun daya listrik di kota Solo dibawah 10 % supaya energi listrik dapat dinikmati oleh warga kota Solo. Model yang digunakan dalam penelitian ini berupa model perhitungan rugi daya dan hasil pada jaringan distribusi dengan memakai struktur kurva daya dan dikaitkan dengan kurva beban pada masing-masing stasiun distribusi. Rugi daya saluran teganggan medium diperhitungkan sesuai dengan jumlah arus beban yang mengalir pada kawat penghantar. Pada saat yang sama, rugi daya trafo dihitung berdasarkan kondisi beban. Berdasarkan hasil perhitungan untuk beban tegangan medium yang terkecil pada gardu SKA 2114 sebesar 21,701 KVA dan terbesar pada gardu SKA 2104 sebesar 74,36 KVA. Sedangkan hasil perhitungan untuk arus nominal trafo yang terkecil yaitu pada gardu SKA2101, SKA2102, SKA2103, SKA2110, SKA 2111, SKA2112, SKA2113, SKA2114, SKA2115, SKA2116, SKA2117 yaitu sebsar 5,28 Ampere dan untuk yang terbsar pada gardu SKA2107 sebesar 11,56 Amper. Hasil perhitungan prosentase rugi trafo pada saat pembebanan yang terkecil yaitu pada gardu SKA 2104 sebesar 1,17% dan terbesar pada gardu SKA 2114 sebesar 4,56%.

Introduction: In the medical diagnostics of bacteria, the rapid detection of pathogenic microorganisms from body fluids is one of the most important tasks. The majority of the modern measuring techniques are based on specific labels bound to the bacteria. However, this strategy usually assumes a rather time-consuming procedure involving several steps (e.g., the widely used enzyme-linked immunosorbent assay normally consists of 5 consecutive steps). Hence, there is an urgent need for the elaboration of rapid, “label-free” techniques, that are often based on Lab-on-a-chip devices. Aim: In this paper, the authors report on the development of a biosensor based on a miniature, integrated optical Mach–Zehnder interferometer. Method: Functionalization of the measuring arm of the sensor by antibodies, made the rapid and specific label-free detection of pathogens feasible. Results: Using the combination of the interferometer with a microfluidic system, the device was able to detect Escherichia coli bacteria at concentrations as low as 106 colony forming unit/ml within minutes. Conclusions: This makes the newly developed biosensor a promising device for a wide range of applications, not only in medical microbiology, but microbial forensics, criminal investigations, bio-terrorism threats and in environmental studies as well. Tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) is a potent cancer cell apoptosis-inducing factor that can induce apoptosis in a variety of cancer cells. However, resistance to TRAIL in cancer cells is a huge obstacle in creating effective TRAIL-targeted clinical therapies. Thus, agents that can either enhance the effect of TRAIL or overcome its resistance are needed. In this study, we combined TRAIL with SNX-2112, an Hsp90 inhibitor we previously developed, to explore the effect and mechanism that SNX-2112 enhanced TRAIL-induced apoptosis in cervical cancer cells. Our results showed that SNX-2112 markedly enhanced TRAIL-induced cytotoxicity in HeLa cells, and this combination was found to be synergistic. Additionally, we found that SNX-2112 sensitized TRAIL-mediated apoptosis caspase-dependently in TRAIL-resistant HeLa cells. Mechanismly, SNX-2112 downregulated antiapoptosis proteins, including Bcl-2, Bcl-XL, and FLIP, promoted the accumulation of reactive oxygen species (ROS), and increased the expression levels of p-JNK and p53. ROS scavenger NAC rescued SNX-2112/TRAIL-induced apoptosis and suppressed SNX-2112-induced p-JNK and p53. Moreover, SNX-2112 induced the upregulation of death-receptor DR5 in HeLa cells. The silencing of DR5 by siRNA significantly decreased cell apoptosis by the combined effect of SNX-2112 and TRAIL. In addition, SNX-2112 inhibited the Akt/mTOR signaling pathway and induced autophagy in HeLa cells. The blockage of autophagy by bafilomycin A1 or Atg7 siRNA abolished SNX-2112-induced upregulation of DR5. Meanwhile, ROS scavenger NAC, JNK inhibitor SP600125, and p53 inhibitor PFTα were used to verify that autophagy-mediated upregulation of DR5 was regulated by the SNX-2112-stimulated activation of the ROS-JNK-p53 signaling pathway. Thus, the combination of SNX-2112 and TRAIL may provide a novel strategy for the treatment of human cervical cancer by overcoming cellular mechanisms of apoptosis resistance.

Scrapie is considered an endemic disease in both sheep and goats in Greece. However, contrary to sheep, in goats more than one prion protein (PrP) polymorphism has been recognized as a candidate for resistance breeding against the disease. For an impression, candidates which are circulating, (i) brain samples (n = 525) from scrapie-affected (n = 282) and non-affected (n = 243) animals within the national surveillance program, and (ii) individual blood samples (n = 1708) from affected (n = 241) and non-affected (n = 1467) herds, in a large part of mainland Greece and its islands, were collected and assayed. A dedicated Taqman method was used to test for amino acid polymorphisms 110T/P, 146N/S/D, 211R/Q, and 222Q/K. Highly prevalent genotypes were 110TT, 146NN, 211RR, and 222QQ. The frequencies of polymorphisms in blood and negative brain samples for codons 110P, 211Q, and 222K were 4.0%, 3.0%, and 1.9%, respectively, while 146D (0.7%) was present only on Karpathos island. Codon 110P was exclusively found in scrapie-negative brains, and homozygous 110P/P in two scrapie-negative goats. It is concluded that breeding programs in Karpathos could focus on codon 146D, while in other regions carriers of the 110P and 222K allele should be sought. Case-control and challenge studies are now necessary to elucidate the most efficient breeding strategies.

Neurophysiol. 79: 2111–2118, 1998. Monosynaptic perforant path responses evoked by subicular stimulation were recorded from CA3 pyramidal cells of rat hippocampal slices. These monosynaptic responses were isolated by using low intensities of stimulation and by placing a cut through the mossy fibers. Perforant path–evoked responses consisted of both excitatory and inhibitory components. Excitatory postsynaptic currents (EPSCs) were mediated by both α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acidreceptors (AMPAR) and N-methyl-d-aspartate receptors (NMDAR).Inhibitory postsynaptic currents consisted of γ-aminobutyric acid-A (GABAA-) and -B (GABAB)-receptor–mediated components. At membrane potentials more positive than -60 mV and at physiological [Ca2+]/[Mg2+] ratios, >30% of perforant path evoked EPSC was mediated by NMDARs. This value varied as a function of the membrane voltage and external [Mg2+]. Two types of responses were observed after low-intensity stimulation of the perforant path. The first type of response showed paired-pulse facilitation and was reduced by 2-amino-4-phosphonobutyric acid (AP4). The second type of response showed paired-pulse depression and was reduced by baclofen. Electrophysiological and pharmacological characteristics of these two types of responses are similar to the properties of lateral and medial perforant path–evoked EPSPs in the dentate gyrus.

Background and objective: polycystic ovary syndrome(PCOS) is a common cause of infertility and is associated with chronic anovulation and hyperandrogenism.The aim of current study is to compare the effect of combined letrozole metformine to that of metformin-laparoscopic ovarian drilling (LOD) for ovulation induction in patients with PCOS.Materials and Methods: on the whole 138 ovarian cycles were studied in 60 patients with clomiphene citrate resistant PCOS in AL-Diwaniyah Maternity and Pediatrics Teaching Hospital during the period of June 2112-June 2114. All patients (n=60) received metformin 500mg /3times per day ,group A(n=30)received letrozole 5mg daily for 5 days repeated for 6 cycles, group B(n=30) underwent LOD and followed for six cycles. Outcome measures were ovulation rates, endometrialthickness, pregnancyrates, live birth rates and miscarriage rate.Results: ovulation rate was higher in group A than group B(63.33% versus 30.0%),a significant increase in endometrial thickness in group A than group B(9.9mm versus 9.03 mm).Live birth rate was higher in group A 83.33% compared to 33.33% in group B. There were no significant differences regarding miscarriage rates between both groups. No multiple pregnancy or ovarian hyper stimulation syndrome or congenital abnormality among live births were reported.Conclusions: Combinedletrozolemetformin seems to be a suitable second line ovulation inducing alternative to LOD in women with PCOS who do not respondwith clomiphene citrate.

Plants possess a battery of specific pathogen resistance ( R- )genes. Precise R- gene regulation is important in the presence and absence of a pathogen. Recently, a microRNA family, miR482/2118, was shown to regulate the expression of a major class of R- genes , nucleotide-binding site leucine-rich repeats ( NBS-LRRs ). Furthermore, RNA silencing suppressor proteins, secreted by pathogens, prevent the accumulation of miR482/2118, leading to an upregulation of R- genes. Despite this transcriptional release of R -genes, RNA silencing suppressors positively contribute to the virulence of some pathogens . To investigate this paradox, we analysed how the regulation of NBS-LRRs by miR482/2118 has been shaped by the coevolution between Phytophthora infestans and cultivated and wild tomatoes. We used degradome analyses and qRT-PCR to evaluate and quantify the co-expression of miR482/2118 and their NBS-LRR targets. Our data show that miR482/2118-mediated targeting contributes to the regulation of NBS-LRRs in Solanum lycopersicum. Based on miR482/2118 expression profiling in two additional tomato species—with different coevolutionary histories with P. infestans —we hypothesize that pathogen-mediated RNA silencing suppression is most effective in the interaction between S. lycopersicum and P. infestans . Furthermore, an upregulation of miR482/2118 early in the infection may increase susceptibility to P. infestans .

Abstract Background The interleukin (IL)-23 pathway is a pathogenic driver in psoriasis, psoriatic arthritis, and inflammatory bowel disease (IBD). There are currently no approved oral therapies selectively targeting the IL-23 pathway. JNJ-77242113 (JNJ-2113) is a targeted oral peptide that binds the IL-23 receptor with high affinity, and potently and selectively inhibits IL-23 proximal signaling and downstream cytokine production. Oral JNJ-2113 showed dose-dependent inhibition of IL-23–induced IL-17A production in rat blood and attenuated signs of disease in a rat colitis model. A Phase 1 study showed inhibition of IL-23–induced IFN-γ production in blood from participants dosed with JNJ-2113 (Fourie et al. ISID. 2023). Methods We evaluated IL-23–induced IL-22 expression in colon tissue after exposure to JNJ-2113 or JNJ-2100 (a tool molecule with similar affinity and potency). IL-22 mRNA was measured after IL-23 stimulation in rat colon biopsies treated with JNJ-2100 in vitro or in colon tissue from rats 6 h after oral JNJ-2100 dosing. Human colon explants from donors were treated in vitro with JNJ-2100 or JNJ-2113, stimulated with IL-23, and IL-22 mRNA was measured. Colon and rectum biopsies from healthy participants dosed for 10 days with oral JNJ-2113 or placebo in a Phase 1 study were obtained, stimulated with IL-23 ex vivo, and IL-22 mRNA was evaluated. Results Overall, inhibition of IL-23–induced IL-22 expression was observed in colon tissue exposed to JNJ-2100 or JNJ-2113. Dose-dependent inhibition of IL-22 expression was observed in rat colon biopsies exposed to JNJ-2100 in vitro. Colon explants from rats dosed with 0.3, 3, and 30 mg/kg oral JNJ-2100 showed dose-dependent inhibition of IL-22 expression (52%, 88% and 96%, respectively) compared with explants from rats treated with vehicle. Human colon explants exposed to JNJ-2100 or JNJ-2113 in vitro also showed dose-dependent inhibition of IL-23–induced IL-22 expression, with &gt;90% inhibition at the highest doses. Importantly, in colon biopsies from Phase 1 participants dosed with oral JNJ-2113 versus placebo, significant inhibition of IL-23–induced IL-22 expression was seen (P&lt;0.05), with an average of 65% and 66% inhibition observed in sigmoid colon and rectum biopsies, respectively. Conclusion These results demonstrate pharmacodynamic activity of JNJ-2100 in vitro and ex vivo in rat colon tissue, of JNJ-2100 and JNJ-2113 in vitro in human colon explants, and of JNJ-2113 in colon biopsies from healthy human volunteers. These data support development of JNJ-2113 for the treatment of IBD. Of note, a Phase 2b dose-ranging study has demonstrated clinical activity of JNJ-2113 in moderate-to-severe plaque psoriasis (NCT05223868).

Abstract Heat-shock protein 90 (hsp90) is a important molecular chaperone required for protein folding, assembly, and maintenance of conformational integrity of several client proteins regulating cell survival, proliferation, and apoptosis. In this study, we investigate whether targeting hsp90 induces cytotoxicity of multiple myeloma (MM) cells in the bone marrow (BM) microenvironment using the small molecule hsp90 inhibitor SNX-2112. SNX-2112 induces growth inhibition in both MM cell lines and patient MM cells resistant to conventional therapeutic agents, with IC50 of 0.019-7.339 uM. Interestingly, SNX-2112 is more potent against all MM cell lines than 17-allylamino-17-demethoxy-geldanamycin (17-AAG) and orally bioavailable. These data suggest its potential utility to overcome conventional drug resistance in MM. MM cell apoptosis triggered by SNX-2112 is mediated via caspase-8,-9,-3 and PARP cleavage. In addition, SNX-2112 significantly inhibits Akt and extracellular signal-related kinase (ERK) signaling pathways induced by interleukin-6 (IL-6) and insulin-like growth factor 1 (IGF-1), which mediate cell proliferation and survival in MM cells, both known client proteins of hsp90. Moreover, SNX-2112 overcomes the survival and growth advantages conferred by IL-6, IGF-1 and bone marrow stromal cells (BMSC). Importantly, SNX-2112 inhibits in vitro capillary-like tube formation by human umbilical vein endothelial cells (HUVEC) via abrogation of endothelial nitric oxide (eNOS)/Akt pathway which is essential cascade in angiogenesis. It also markedly inhibits osteoclast formation associated with downregulation of ERK/c-fos, p38MAPK, and PU.1 pathways, which mediate osteoclastogenesis. Furthermore SNX-5422, a prodrug of SNX-2112, significantly inhibits MM tumor growth and prolongs survival in vivo in a xenograft murine model. We also confirm that SNX-5422 triggers MM cell apoptosis in vivo by TUNEL assay, associated with an inhibitory effect on microvessel density (MVD), evidenced by immunohistochemical analysis for CD34 expression. Therefore, targeting hsp90 by novel small molecule inhibitor SNX-2112 not only inhibits MM cell growth, but also acts in the BM microenvironment to block angiogenesis and osteoclast formation associated with downregulation of Akt and ERK signaling. Taken together, our data provide the preclinical rationale for clinical evaluation of SNX-2112 to improve patient outcome in MM.

Designing novel symmetric-key primitives for advanced protocols like secure multiparty computation (MPC), fully homomorphic encryption (FHE) and zero-knowledge proof systems (ZK), has been an important research topic in recent years. Many such existing primitives adopt quite different design strategies from conventional block ciphers. Notable features include that many of these ciphers are defined over a large finite field, and that a power map is commonly used to construct the nonlinear component due to its efficiency in these applications as well as its strong resistance against the differential and linear cryptanalysis. In this paper, we target the MPC-friendly ciphers AIM and RAIN used for the post-quantum signature schemes AIMer (CCS 2023 and NIST PQC Round 1 Additional Signatures) and Rainier (CCS 2022), respectively. Specifically, we can find equivalent representations of 2-round RAIN and full-round AIM, respectively, which make them vulnerable to either the polynomial method, or the crossbred algorithm, or the fast exhaustive search attack. Consequently, we can break 2-round RAIN with the 128/192/256-bit key in only 2111/2170/2225 bit operations. AbstractHeat-shock protein 90 (Hsp90) acts as a molecular chaperone required for maintaining the conformational stability of client proteins regulating cell proliferation, survival, and apoptosis. Here we investigate the biologic significance of Hsp90 inhibition in multiple myeloma (MM) and other hematologic tumors using an orally available novel small molecule inhibitor SNX-2112, which exhibits unique activities relative to 17-allyamino-17-demethoxy-geldanamycin (17-AAG). SNX-2112 triggers growth inhibition and is more potent than 17-AAG against MM and other malignancies. It induces apoptosis via caspase-8, -9, -3, and poly (ADP-ribose) polymerase cleavage. SNX-2112 inhibits cytokine-induced Akt and extracellular signal-related kinase (ERK) activation and also overcomes the growth advantages conferred by interleukin-6, insulin-like growth factor-1, and bone marrow stromal cells. Importantly, SNX-2112 inhibits tube formation by human umbilical vein endothelial cells via abrogation of eNOS/Akt pathway and markedly inhibits osteoclast formation via down-regulation of ERK/c-fos and PU.1. Finally, SNX-2112, delivered by its prodrug SNX-5422, inhibits MM cell growth and prolongs survival in a xenograft murine model. Our results indicate that blockade of Hsp90 by SNX-2112 not only inhibits MM cell growth but also acts in the bone marrow microenvironment to block angiogenesis and osteoclastogenesis. Taken together, our data provide the framework for clinical studies of SNX-2112 to improve patient outcome in MM and other hematologic malignancies.

Background. Diabetic retinopathy (DR), including retinal angiogenesis and endothelial cell proliferation and migration, is a serious complication in diabetic patients. It has been reported that ginsenoside Rg1 can prevent retinal damage. However, the mechanism by which Rg1 prevents retinal damage is unknown. Therefore, the aim of the present study was to investigate the mechanism by which Rg1 inhibits high glucose-induced complications through the regulation of the lncRNA SNHG7/miR-2116-5p/SIRT3 axis. Methods. Under high glucose (HG) conditions, human retinal endothelial cells (HRECs) were cultured to simulate a DR environment, and Rg1 was added after 48 h. Negative control (NC), miR-2116-5p mimic, si-SNHG7, pc-DNA SIRT3, and miR-2116-5p inhibitor were transfected into HRECs, and CCK-8 assay was used to detect the cell viability. Angiogenesis and transwell assays were used to evaluate angiogenesis and cell migration, respectively. qRT–PCR and Western blot were used to detect the expression of related genes and proteins. Luciferase reporter assays and bioinformatics were used to analyze the target binding sites of miR-2116-5p to lncRNA SNHG7 and SIRT3. Results. The proliferation, migration and angiogenesis of HRECs were induced by HG. As expected, HG upregulated miR-2116-5p and VEGF expression but downregulated lncRNA SNHG7 and SIRT3 expression. Importantly, Rg1 inhibited HG-induced HREC proliferation, migration, and angiogenesis by upregulating the lncRNA SNHG7, and miR-2116-5p had a target regulatory relationship with both lncRNA SNHG7 and SIRT3. Conclusion. Rg1 inhibits HG-induced proliferation, migration, angiogenesis, and VEGF expression in retinal endothelial cells through the lncRNA SNG7/miR-2116-5p/SIRT3 axis. This finding provides theoretical evidence for the clinical application of Rg1 in DR.

A Gram-variable, motile, endospore-forming, slightly halophilic bacterial strain, designated SW-211T, was isolated from a tidal flat of the Yellow Sea in Korea, and was characterized taxonomically by using a polyphasic approach. The organism grew optimally at 37 °C and in the presence of 2–3 % NaCl. Comparative 16S rRNA gene sequence analysis showed that strain SW-211T forms a distinct phylogenetic lineage within the radiation of the cluster comprising Bacillus species. Strain SW-211T had cell-wall peptidoglycan based on meso-diaminopimelic acid. The predominant menaquinone was MK-7 and the major fatty acids were anteiso-C15 : 0 (34·8 %), iso-C15 : 0 (15·6 %), iso-C16 : 0 (12·5 %) and iso-C14 : 0 (10·0 %). The DNA G+C content was 35·2 mol%. Strain SW-211T exhibited levels of 16S rRNA gene sequence similarity of <96·2 % with respect to the type strains of recognized Bacillus species. On the basis of its phenotypic properties and phylogenetic distinctiveness, strain SW-211T (=KCTC 3898T=DSM 16303T) was classified in the genus Bacillus as the type strain of a novel species, for which the name Bacillus litoralis sp. nov. is proposed.

This paper presents an experimental study on the encapsulation of Mahkota Dewa extracts by maltodextrin using spray drying. The bioactive compound from dried Mahkota Dewa was obtained using a subcritical water extraction process prior to a spray drying process. The effect of feed flow rate (485 to 2115 ml/h) was investigated using one-factor-at-a-time (OFAT). It was observed that the mean particle size increase varied from 3.55 to 8.38 µm when the feed flow rate increased from 485 to 2115 ml/h. Moisture content increased 4.88 to 6.83% as the feed flow rate increased from 485 to 2115 ml/h, whereas the antioxidant activity increased slightly from 90.48 to 91.65%. The findings from this study showed that decrease in feed flow rate reduces antioxidant activity, moisture content, and particle size. ABSTRAK: Kertas kerja ini adalah berkenaan kajian eksperimen melalui proses pengkapsulan ekstrak buah Mahkota Dewa dengan menggunakan maltodekstrin melalui teknik pengeringan semburan. Sebatian bioaktif dari buah Mahkota Dewa kering diperoleh melalui teknik pengekstrakan air subkritikal sebelum proses pengeringan semburan. Kesan kadar aliran masuk (485 hingga 2115 ml/jam) dikaji menggunakan konsep satu-faktor-pada-satu-masa (OFAT). Dapatan kajian mendapati bahawa purata saiz zarah meningkat dari 3.55 kepada 8.38 µm ketika kadar aliran masuk meningkat dari 485 sehingga 2115 ml/jam. Kandungan kelembapan meningkat dari 4.88 kepada 6.83% dengan kenaikan kadar aliran masuk dari 485 hingga 2115 ml/jam, sedangkan aktiviti antioksida meningkat sedikit dari 90.48 hingga 91.65%. Dapatan kajian menunjukkan bahawa penurunan kadar aliran masuk berkurang melalui aktiviti antioksida, kandungan kelembapan dan saiz zarah.

The total synthesis of two decahydroquinoline poison frog alkaloids ent-cis-195A and cis-211A were achieved in 16 steps (38% overall yield) and 19 steps (31% overall yield), respectively, starting from known compound 1. Both alkaloids were synthesized from the common key intermediate 11 in a divergent fashion, and the absolute stereochemistry of natural cis-211A was determined to be 2R, 4aR, 5R, 6S, and 8aS. Interestingly, the absolute configuration of the parent decahydroquinoline nuclei of cis-211A was the mirror image of that of cis-195A, although both alkaloids were isolated from the same poison frog species, Oophaga (Dendrobates) pumilio, from Panama.

Chinese hamster ovary (CHO) cells manifesting striking cytopathogenic changes in culture were investigated to determine the causative agent. Electron microscopic analyses revealed viral particles of about 40 nm in diameter, displaying typical calicivirus morphology. To date, this virus, designated isolate 2117, exclusively replicates in CHO cells, achieving only moderate titres. After cloning, the coding region of 7928 nucleotides, the 3′ non-coding region and the poly(A) tail were sequenced. The genome consists of three open reading frames (ORFs), with the first and second ORF having the same reading frame. The overall genomic organization as well as the nucleotide sequence of isolate 2117 is most similar to that of a recently described canine calicivirus, but also shows significant similarity to the sequences of mink calicivirus and other caliciviruses within the genus Vesivirus. In Western blots, using antibodies against the viral protease, a stable, unprocessed 3CD protein of 68 kDa was identified in homogenates of 2117-infected CHO cells. Furthermore, antibodies raised against ORF 3 reacted with the respective protein in 2117-virions, demonstrating that this predicted 9 kDa protein is a minor structural component of the virion. In addition, an RT-PCR assay was established to detect 2117 viral RNA in biological products such as foetal bovine serum, which will aid the discovery of the origin and host of the virus.

Ca2+-regulated heat-stable protein of 24 kDa (CRHSP-24) is a serine phosphoprotein originally identified as a physiological substrate for the Ca2+-calmodulin regulated protein phosphatase calcineurin (PP2B). CRHSP-24 is a paralog of the brain-specific mRNA-binding protein PIPPin and was recently shown to interact with the STYX/dead phosphatase protein in developing spermatids (Wishart MJ and Dixon JE. Proc Natl Acad Sci USA 99: 2112–2117, 2002). Investigation of the effects of phorbol ester (12- o-tetradecanoylphorbol-13-acetate; TPA) and cAMP analogs in 32P-labeled pancreatic acini revealed that these agents acutely dephosphorylated CRHSP-24 by a Ca2+-independent mechanism. Indeed, cAMP- and TPA-mediated dephosphorylation of CRHSP-24 was fully inhibited by the PP1/PP2A inhibitor calyculin A, indicating that the protein is regulated by an additional phosphatase other than PP2B. Supporting this, CRHSP-24 dephosphorylation in response to the Ca2+-mobilizing hormone cholecystokinin was differentially inhibited by calyculin A and the PP2B-selective inhibitor cyclosporin A. Stimulation of acini with secretin, a secretagogue that signals through the cAMP pathway in acini, induced CRHSP-24 dephosphorylation in a concentration-dependent manner. Isoelectric focusing and immunoblotting indicated that elevated cellular Ca2+ dephosphorylated CRHSP-24 on at least three serine sites, whereas cAMP and TPA partially dephosphorylated the protein on at least two sites. The cAMP-mediated dephosphorylation of CRHSP-24 was inhibited by low concentrations of okadaic acid (10 nM) and fostriecin (1 μM), suggesting that CRHSP-24 is regulated by PP2A or PP4. Collectively, these data indicate that CRHSP-24 is regulated by diverse and physiologically relevant signaling pathways in acinar cells, including Ca2+, cAMP, and diacylglycerol.

The existence of global warming is common knowledge. However, it can be predicted that there may be cooling zones worldwide based on the mechanism of terrestrial biophysical processes. Here, the Theil–Sen median trend, the Mann–Kendall trend test method, continuous wavelet transformation, and the Hurst exponent were used to study the cooling trends, abrupt change times, periodicity, and future sustainability of temperature changes in different cooling zones since the 1900s based on the CRU dataset. We found an amazing result; 8,305,500 km2 of land surface had been cooling since the 1900s, covering five continents and 32 countries, accounting for 86% of land area in China, and distributed over 16 zones. The average cooling rate of the cooling zones was −0.24 °C/century. The maximum cooling rate was −1.40 °C/century, and it was 1.43 times the average rate of global land warming (0.98 °C/century). The cooling zones near the sea were greatly influenced by ocean currents and were mainly affected by a small time scale periodicity of less than 30 years, whereas the cold zones located relatively far from the sea and less affected by ocean currents were mainly affected by medium time scales of more than 30 years. Moreover, 32.33% of the cooling zones, involving 2,684,900 km2, will be continuously cooling in the future, and the rest will probably warm up in 2114, 2041, 2096, 2099, 2119, 2073, 2048, and 2101, respectively. The study will help us to further understand the essential characteristics of global climate change, and to find more theoretical bases for mitigating global warming and exploring surface cooling mechanisms.

The primary creep behavior at ambient temperature of typical h.c.p., b.c.c. and f.c.c. metals and alloys of annealed state was surveyed and the deformation mechanism of CP-Ti was discussed through transmission electron microscopy. Only h.c.p. metals and alloys demonstrated significant creep at ambient temperature. Arrays of straight screw dislocations of b=1/2<2110> and <2110> direction were observed in the crept CP-Ti.
