Streszczenie:

Pulmonary hypertension (PH) is a deadly vascular disease characterized by multiple disparate molecular pathways controlling vasoconstriction and hyperproliferation throughout the pulmonary vasculature. Importantly, the selection and proliferation of hardier pulmonary endothelial cells in PH may describe the origins of the anti-apoptotic and hyperproliferative phenotypes characterizing severe PH pathogenesis. However, the importance of cellular survival in mediating intercellular transfer of regulatory factors during PH progression has yet to be clearly defined. MicroRNAs (miRNAs) may coordinately regulate PH progression, but their integrative functions have been difficult to describe with conventional methods. Recently, using a network-based bioinformatics approach, we identified the miR-130/301 family as a master regulator governing pulmonary vascular proliferation as well as vasoconstriction and vessel stiffening in PH through repression of its direct target gene PPARγ. In this study, we additionally identify miR-130/301 as a regulator of cellular survival and show that the dysregulation of miR-130/301 in PH increases apoptotic activity in pulmonary arterial endothelial cells. Furthermore, we identify NCOA3 and PTEN as direct targets of miR-130/301 through which this miRNA family may control apoptotic signaling in the pulmonary vasculature. Our observations provide critical insight into the systems-level regulation of both cellular survival and proliferation by the miR-130/301 family in PH pathogenesis. Specifically, this model establishes a mechanistic framework describing the importance of miR-130/301 in the initiating apoptotic events of PH development. Moreover, our study suggests broad propositions for miRNA-based therapeutics for treating PH and further endorses the application of in silico network theory to decipher the combinatorial molecular origins of complex diseases such as PH.

Neoliberalism has been on the rise at least since the mid-1980s. The economization of the social and the increasing globalization of capital show all the characteristics of a neoliberal governmentality, as Michel Foucault has analyzed it (cf. Foucault 2008). Gilles Deleuze described this process as a new transformation of the disciplinary society into a control society, which he briefly sketched and described in his far-sighted and ‘prophetic’ essay (cf. Deleuze 1992). The authorities in charge of managing our Andean basins work in unimaginable water-powered landscapes from the Peruvian legislator point of view. They should generate and apply peculiar mechanisms. Why? Because the national legislator is traditionally positioned on the coast, a region very different to the Andean highlands and punas. The use of ethnographic evidence coming from a drainage basin in the Central Andes (Mantaro River, Junin) lets me depict in detail how public officers appeal to mechanisms of location, adaptation and regulation in order to adjust the official regulations, to cover the needs of the farming organizations of watering, and to affirm their positions as representatives of the Peruvian government. The consequence is that they exceed and also violate the regulatory channels that control their administrative functions. In this way, they incorporate a new function to their official position, that is, to develop peculiar administrative patterns to process the tightness between the inflexibility of the State Water Right and the demands of recognition and balance that the andean watering organizations propose to the Government.
Las autoridades encargadas de gestionar nuestras cuencas andinas ejercen sus funciones en paisajes hidráulicos inimaginables para el legislador peruano. Para ello deben generar y aplicar mecanismos peculiares. ¿Por qué? Porque el legislador nacional se encuentra tradicionalmente parapetado en la costa, una región muy diferente a las sierras y punas andinas. El uso de evidencia etnográfica proveniente de una cuenca hidrográfica de los Andes Centrales (Río Mantaro, Junín) me permite detallar cómo los funcionarios estatales apelan a los mecanismos de localización, adaptación y regularización con el fin de modular la normatividad oficial, atender las necesidades de las organizaciones campesinas de riego y afirmar su papel como representantes del Estado peruano. El resultado es que desbordan y hasta violan los cauces normativos que regulan sus funciones administrativas. De este modo, incorporan una nueva función a su papel oficial que consiste en desarrollar peculiares prácticas administrativas para procesar la tensión entre la rigidez del derecho estatal de aguas y las demandas de reconocimiento y articulación que los regantes andinos le plantean al Estado.

A search for neutrinoless double-beta decay (0vBB) is performed using 36.55 g of the isotope Nd-150 with 5.25 y of data from the NEMO experiment. A complete background model for this source foil is constructed using a binned likelihood fit across several observables in multiple control channels. The 2vBB decay half-life is measured to be T(2v) = 9.34 +/- 0.22 (stat.) [+0.60, -0.59] (syst.) x 10^18 y, which represents the most precise measurement for this isotope to date. A multivariate analysis is performed in the search for 0vBB decay. As no evidence for 0vBB decay is observed, lower limits are obtained on the half-lives for several underlying beyond standard model mechanisms. The observed lower limit assuming a light Majorana neutrino exchange is T(0v) > 18.96 x 10^21 y at 90% CL, corresponding to an upper limit on the effective neutrino mass of < (2.64--8.64) eV, representing the most stringent limit obtained with the isotope Nd-150.

Streszczenie:

People naturally produce iconic gestures when they speak. Iconic gestures that depict people's actions may influence the way children process action events. This dissertation investigates experimentally whether seeing such iconic gestures promotes 3-year-old children's action event memory and verb learning. Chapter 1 introduces the topic and presents an outline of the dissertation. Chapter 2 provides a working de nition of gesture and a literature review on iconicity. Chapter 3 describes the development and norming of a large database that contains stimuli videos of actions events and iconic gestures. Action videos showed actors moving in unusual manners and iconic gestures depicted how the actors moved. Chapter 4 examines whether children remember action events differently when they see iconic gestures while encoding these events. Seeing iconic gestures that depicted how actors moved while encoding action events boosted children's memory of actors and their actions. Specifically, children showed better memory for event aspects that were depicted in gesture. Chapter 5 asks whether prior action knowledge promotes verb learning and whether seeing iconic gestures influences this process. Pre-exposure to unlabeled actions facilitated verb learning when those actions were shown with iconic gestures and when children were shown two actors performing the same actions simultaneously, but children performed better in the iconic-gesture condition. Chapter 6 investigates whether children learn that verbs typically refer to actions from seeing iconic gestures that depict individual verb meanings. Children who were taught verbs with iconic gestures demonstrated such word-category knowledge about verbs in an immediate and delayed novel verb learning task in which different novel verbs were taught without iconic gestures. Chapter 7 discusses theoretical and practical implications of the experimental findings. Iconic gestures are meaningful social cues that help children individuate people's actions, encode and remember complex action events, acquire individual verb meanings, and generate word-category knowledge about verbs.

A group G is called a barely transitive group if it acts transitively and faithfully on an infinite set and every orbit of every proper subgroup is finite. A subgroup H of a group G is called a permutable subgroup, if H commutes with every subgroup of G. We showed that if an infinitely generated barely transitive group G has a permutable point stabilizer, then G is locally finite. We proved that if a barely transitive group G has an abelian point stabilizer H, then G is isomorphic to one of the followings: (i) G is a metabelian locally finite p-group, (ii) G is a finitely generated quasi-finite group (in particular H is finite), (iii) G is a finitely generated group with a maximal normal subgroup N where N is a locally finite metabelian group. In particular, G=N is a quasi-finite simple group. In all of the three cases, G is periodic.

This thesis is a contribution to determine the absolute irreducibility of polynomials via their Newton polytopes. For any field F
a polynomial f in F[x1, x2,..., xk] can be associated with a polytope, called its Newton polytope. If the polynomial f has integrally indecomposable Newton polytope, in the sense of Minkowski sum, then it is absolutely irreducible over F
i.e. irreducible over every algebraic extension of F. We present some new results giving integrally indecomposable classes of polytopes. Consequently, we have some new criteria giving infinitely many types of absolutely irreducible polynomials over arbitrary fields.

A group G is called locally finite if every finitely generated subgroup of G is finite. In this thesis we study the centralizers of subgroups in simple locally finite groups. Hartley proved that in a linear simple locally finite group, the fixed point of every semisimple automorphism contains infinitely many elements of distinct prime orders. In the first part of this thesis, centralizers of finite abelian subgroups of linear simple locally finite groups are studied and the following result is proved: If G is a linear simple locally finite group and A is a finite d-abelian subgroup consisting of semisimple elements of G, then C_G(A) has an infinite abelian subgroup isomorphic to the direct product of cyclic groups of order p_i for infinitely many distinct primes pi. Hartley asked the following question: Let G be a non-linear simple locally finite group and F be any subgroup of G. Is CG(F) necessarily infinite? In the second part of this thesis, the following problem is studied: Determine the nonlinear simple locally finite groups G and their finite subgroups F such that C_G(F) contains an infinite abelian subgroup which is isomorphic to the direct product of cyclic groups of order pi for infinitely many distinct primes p_i. A long-standing conjecture states that if A is a finite group acting fixed point freely on a finite solvable group G of order coprime to jAj, then the Fitting length of G is bounded by the length of the longest chain of subgroups of A. If A is nilpotent, it is expected that the conjecture is true without the coprimeness condition. Tetrahedral meshes are becoming more and more important for geo-modeling applications. The presented work introduces new algorithms for efficient visualization and modeling of tetrahedral meshes. Visualization consists of a generic framework that includes the extraction of geological information like stratigraphic columns, fault block boundaries, simultaneous co-rendering of different attributes and boolean operations of Constructive Solid Geometry with constant complexity. Modeling can be classified into geometric and implicit modeling. Geometric modeling addresses local mesh refinement to increase the numerical resolution of a given mesh. Implicit modeling covers the definition and manipulation of implicitly defined models. A new surface reconstruction method was developed to reconstruct complex, multi-valued surfaces from noisy and sparse data sets as they occur in geological applications. The surface can be bounded and may have discontinuities. Further, this work proposes a new and innovative algorithm for rapid editing of implicitly defined shapes like horizons based on the GeoChron parametrization. The editing is performed interactively on the 3d-volumetric model and geological constraints are respected automatically.

Les installations de chimie separative a l'echelle 1 ont ete mises en place au crn de strasbourg. La premiere partie de l'experience comprenant la purification des 300 kg d'uranium vis-a-vis des ultra-traces de #2#3#8pu et la mesure des noyaux de plutonium extraits a ete realisee. Les resultats du comptage des sources de pu ont permis de valider la procedure chimique. Ils montrent egalement une forte contamination de l'experience en #2#3#8pu

Previous research has suggested a role for nutrients in several aspects of psychological functioning. Based on this research the present thesis explored the role of omega-3 fatty acids, vitamins and minerals in cognition, mood and the perception of food. Divided into three sections, the first section of the thesis used qualitative methods to explore factors that affected food choice and the decision to consume a healthy diet (i.e. a diet high in omega-3, vitamins and minerals). In addition, factors that affected supplement use, specifically the factors that affected the use of omega-3 fatty acid, vitamin and mineral supplements were explored. From the data, two models were developed: one depicting factors that influenced food choice and healthy eating, and one depicting factors that influenced supplement use. After the examination of the factors that affected food choice, healthy eating and the consumption of omega-3 fatty acids, vitamins and minerals, the importance of these nutrients in psychological health and cognition was explored in a double-blind, randomized controlled trial. Specifically the second section explored the effect of vitamins, minerals, and/or omega-3 fatty acids in mild psychiatric symptoms, stress, aggression, impulsivity, fatigue, hostility, anxiety, confusion, confidence, reaction time, memory and vigilance. A positive response was found to DHA with regard to aggression and response inhibition; in addition there was a trend for those taking only DHA to report feeling more clearheaded the taking of vitamins/minerals alone resulted in feeling more clearheaded. Supplementation with vitamins and minerals was found to significantly reduce stress. With the General Health Questionnaire the difference between the placebo and vitamins/minerals groups approached statistical significance. There was no effect of omega-3, vitamins and minerals of any cognitive domain (memory, reaction time and vigilance). Although it was concluded that on occasions either vitamins/minerals or DHA when tested alone had a positive effect on mood, stress and mild psychiatric symptoms, there was no synergistic interaction; rather on occasions the interaction between these supplements had negative consequences. The third section explored the role of omega-3 fatty acids, vitamins and minerals in the perception of food, specifically the effects of enrichment, health claim and gender on three variables involved in consumer behaviour: the perceived healthiness, the desirability of supplementation and the likelihood of purchase of foods. The main findings were that i) health claims increased the perceived healthiness of unhealthy foods; ii) the likelihood of purchase increased mostly after the enrichment of healthy foods (which is consistent with some findings but not others), iii) males were more likely to purchase healthy, high protein products than females. Besides these findings there was no consistent effect of enrichment, health claim or gender on the 3 facets of consumer behaviour, suggesting that the effects on the 3 consumer variables should be considered individually. In addition, when examining the role of gender males and females should be considered separately.

O objetivo dessa tese é contribuir para o entendimento da precipitação de compostos intermetálicos e da reversão da martensita por meio de modelos cinéticos, tanto em experimentos isotérmicos no aço maraging 350 (350B) como em estudos não-isotérmicos nos aços maraging 300 (300A), maraging 350 (350C). Além da cinética das transformações de fase, foram estudadas também as mudanças da microestrutura e dos mecanismos de endurecimento decorrentes de tratamentos térmicos de envelhecimento para o aço maraging 350B. Para estas finalidades, foram usadas diversas técnicas complementares de caracterização microestrutural, como microscopia ótica (MO), microscopia eletrônica de varredura (MEV) com espectroscopia por dispersão de energia de raios X (EDS), microdureza Vickers, difração de raios X (DRX) e ferritoscopia. Já a calorimetria exploratória diferencial (DSC) foi usada para estudar a precipitação de compostos intermetálicos e reversão da martensita em experimentos não-isotérmicos. Os resultados dos experimentos não-isotérmicos de DSC com os aços maraging 300 e 350 evidenciaram que a precipitação ocorre em duas etapas. A primeira relacionada à difusão de soluto no volume com energia de ativação próxima da difusão do níquel e molibdênio na ferrita, enquanto a segunda acontece por meio da difusão de soluto ao longo das discordâncias com energia de ativação menor que a difusão do níquel e do molibdênio na ferrita. Observou-se também que a reversão da martensita pode ocorrer em duas etapas. A primeira etapa foi associada à difusão de soluto, enquanto a segunda foi relacionada ao mecanismo de cisalhamento. Já as observações microestruturais, por meio de microscopia óptica e de microscopia eletrônica de varredura, evidenciaram que a austenita revertida formou-se nas regiões de interface, como os contornos de grão, contornos de pacote e contornos de ripas da estrutura martensítica para temperaturas a partir de 520 °C, enquanto a austenita revertida encontrada no interior das ripas da martensita formou-se a partir de 560 °C. O estudo da cinética de precipitação e do comportamento da curva de envelhecimento em um aço maraging 350 (350B), para tratamentos isotérmicos entre 440 e 600 °C, mostrou que as medidas de microdureza podem ser muito úteis para estudos dessa natureza nesses aços. A análise cinética da precipitação, realizada por meio do ajuste dos dados experimentais aos modelos JMAK e Austin-Rickett, mostrou que eles se ajustam bem a esses modelos com coeficiente de correlação próximo de 1. Entretanto, a interpretação dos valores de n, obtidos pela equação Austin-Rickett, mostrou que eles têm maior concordância com as mudanças microestruturais observadas nos aços maraging, em estudos anteriores, se comparados com aqueles estimados por meio da equação JMAK. A interpretação das constantes n, usando a equação Austin-Rickett, permitiu estabelecer diversas etapas para a precipitação. Na primeira ocorre a precipitação nas discordâncias para 440 °C, seguida pelo crescimento de cilindros longos e finitos em comparação com a distância de separação deles para 480 °C e, por fim, o crescimento de precipitados partindo de dimensões pequenas com taxa de nucleação zero para 520 e 560 °C. Já o estudo do comportamento da curva de envelhecimento para diversos tempos entre 440 e 600 °C em aço maraging 350 (350B) mostrou que esse aço apresenta uma etapa de endurecimento e outra de amolecimento. Essa etapa de endurecimento, comumente atribuída à formação de fases intermetálicas coerentes e semicoerentes, pode subdividir-se em dois estágios para as temperaturas de envelhecimento de 440 e 480 ºC ou apresentar um único estágio para 520 e 560 ºC. Já a etapa de amolecimento é associada não somente ao mecanismo clássico de superenvelhecimento em que a queda na resistência mecânica ocorre em virtude da perda de coerência e do engrossamento de precipitados, mas também como consequência da formação de austenita revertida a partir da martensita, especialmente, para temperaturas entre 520 e 600 ºC.
The purpose of this thesis is to contribute to the understanding of precipitation of intermetallic compounds and reversion of martensite through kinetic models, as in isothermal experiments in maraging 350 steel (350B) as in non-isothermal studies in maraging steels 300 (300A) maraging 350 (350C). In addition to kinetics of phase transformation, they were also investigated both the changes of the microstructure and the mechanisms of hardening due to aging heat treatments for the maraging steel 350B. For these purposes, we used several complementary techniques for microstructural characterization, such as optical microscopy (OM), scanning electron microscopy (SEM) with energy dispersive X-ray spectroscopy (EDS), Vickers hardness, X-ray diffraction (XRD) and feritscope, while differential scanning calorimetry (DSC) was used to study the precipitation of intermetallic compounds and reversion of martensite in non-isothermal experiments. The results of the non-isothermal DSC experiments with the maraging steel 300 and 350 showed that precipitation occurs in two steps. First stage related to the diffusion of solute in bulk with activation energy next to nickel diffusion and molybdenum in the ferrite, while second stage occurs through the solute diffusion along dislocations with lower activation energy than the diffusion of nickel and molybdenum in ferrite. It was also observed that the reversion of the martensite can occur in two steps. First stage was associated with the solute diffusion, while the second stage is related to the shear mechanism. Microstructural observations by optical microscopy and scanning electron microscopy showed that austenite reverted was formed in the interface regions, such as grain boundaries, packet boundaries and lath boundaries of martensitic structure for temperatures from the 520 °C, while the reverted austenite found within the martensite laths formed from 560 °C. Study of the kinetics of precipitation and aging hardening behavior in a 350 maraging steel (350B), by isothermal treatments between 440 and 600 °C, showed that the microhardness measurements can be very useful for such studies in these steels. Kinetics of precipitation analysis was carried out by adjusting the experimental data to JMAK and Austin-Rickett models. It showed that they fit well to these models with a correlation coefficient close to 1. However, the interpretation of the n values, obtained by Austin-Rickett equation, they have showed higher agreement with the observed microstructural changes in the maraging steel, in previous studies, when compared with those estimated by JMAK equation. The interpretation of the constants, using the Austin-Rickett equation, revealed several steps to precipitation. In the first precipitates on dislocations for 440 °C followed by growth of long and finite cylinders in comparison with the distance their separation for 480 °C and, finally, the growth of precipitates starting from small dimensions with nucleation rate zero for 520 and 560 °C. Study of aging hardening behavior curve for various times between 440 and 600 °C in maraging steel 350 (350B) showed that the steel has a hardening step and another softening. This step of hardening, commonly attributed to the formation of intermetallic phases coherent and semicoerentes, it can be divided into two stages to the aging temperatures of 440 and 480 °C or present a single stage 520 and 560 °C. Since the softening stage is associated not only to classic overaging mechanism in which a drop in mechanical strength occurs due to loss of coherence and precipitate coarsening, but also as a consequence of austenite formation reverted from the martensite especially to temperatures between 520 and 600 °C.
