Toll-like receptors (TLRs) are key players in the innate immune system. Despite the great efforts in TLR structural biology, today we know the spatial structures of only four human TLR intracellular TIR domains. All of them belong to one of five subfamilies of receptors. One of the main bottlenecks is the high-level production of correctly folded proteins in soluble form. Here we used a rational approach to find the optimal parameters to produce TIR domains of all ten human TLR family members in soluble form in E. coli cells. We showed that dozens of milligrams of soluble His-tagged TLR2/3/6/7TIR and MBP-tagged TLR3/5/7/8TIR can be produced. We also developed the purification protocols and demonstrated by CD and NMR spectroscopy that purified TLR2/3/7TIR demonstrate a structural organization inherent to TIR domains. This illustrates the correct folding of produced proteins and their suitability for further structural and functional investigations.

Human and murine platelets (PLTs) variably express toll-like receptors (TLRs), which link the innate and adaptive immune responses during infectious inflammation and atherosclerotic vascular disease. In this paper, we show that the TLR9 transcript is specifically up-regulated during pro-PLT production and is distributed to a novel electron-dense tubular system-related compartment we have named the T granule. TLR9 colocalizes with protein disulfide isomerase and is associated with either VAMP 7 or VAMP 8, which regulates its distribution in PLTs on contact activation (spreading). Preincubation of PLTs with type IV collagen specifically increased TLR9 and CD62P surface expression and augmented oligodeoxynucleotide (ODN) sequestration and PLT clumping upon addition of bacterial/viral ODNs. Collectively, this paper (a) tracks TLR9 to a new intracellular compartment in PLTs and (b) describes a novel mechanism of TLR9 organization and signaling in human PLTs.

Abstract In addition to their essential role in antibody production, B cells regulate immune responses by secreting effector cytokines. Interleukin 10 (IL-10) produced by B cells is important for controlling inflammation and autoimmunity, underscoring the importance of identifying mechanisms regulating B cell IL-10 production. We have previously demonstrated that the membrane-cytoskeleton linker protein ezrin regulates B cell antigen receptor organization and signaling. Further, we have shown that ezrin undergoes rapid dephosphorylation in naïve B cells treated with LPS, suggesting that it may regulate TLR4-mediated IL-10 production. In the present study, we demonstrate that conditional deletion of ezrin in B cells increases IL-10 production induced by TLR4, but not TLR2 or TLR9, ligation. The MyD88-independent TRIF-TBK1-IRF3 pathway is required for ezrin-deficient B cells to produce higher IL-10 upon LPS stimulation. Treatment of B cells with a novel small molecule inhibitor of ezrin induces its dephosphorylation, and increases LPS-induced NF-κB and IRF3 activation and IL-10 secretion, indicating a role for Thr567 phosphorylation of ezrin in limiting IL-10. Loss of ezrin in B cells results in dampened pro-inflammatory response to a sub-lethal dose of LPS in vivo, which is dependent on increased IL-10 production. Taken together, our data yield new insights into molecular and membrane-cytoskeletal regulation of B cell IL-10 production, and reveal ezrin as a potential therapeutic target in inflammatory diseases.

COVID-19 is characterized by a heterogeneous clinical presentation and prognosis. Risk factors contributing to the development of severe disease include old age and the presence of comorbidities. However, the genetic background of the host has also been recognized as an important determinant of disease prognosis. Considering the pivotal role of innate immunity in the control of SARS-CoV-2 infection, we analyzed the possible contribution of several innate immune gene polymorphisms (including TLR2-rs5743708, TLR4-rs4986790, TLR4-rs4986791, CD14-rs2569190, CARD8-rs1834481, IL18-rs2043211, and CD40-rs1883832) in disease severity and prognosis. A total of 249 individuals were enrolled and further divided into five (5) groups, according to the clinical progression scale provided by the World Health Organization (WHO) (asymptomatic, mild, moderate, severe, and critical). We identified that elderly patients with obesity and/or diabetes mellitus were more susceptible to developing pneumonia and respiratory distress syndrome after SARS-CoV-2 infection, while the IL18-rs1834481 polymorphism was an independent risk factor for developing pneumonia. Moreover, individuals carrying either the TLR2-rs5743708 or the TLR4-rs4986791 polymorphisms exhibited a 3.6- and 2.5-fold increased probability for developing pneumonia and a more severe disease, respectively. Our data support the notion that the host’s genetic background can significantly affect COVID-19 clinical phenotype, also suggesting that the IL18-rs1834481, TLR2-rs5743708, and TLR4-rs4986791 polymorphisms may be used as molecular predictors of COVID-19 clinical phenotype.

Subcellular compartmentalization of receptor signaling is an emerging principle in innate immunity. However, the functional integration of receptor signaling pathways into membrane trafficking routes and its physiological relevance for immune responses is still largely unclear. In this study, using Lyst-mutant beige mice, we show that lysosomal trafficking regulator Lyst links endolysosomal organization to the selective control of toll-like receptor 3 (TLR3)– and TLR4-mediated proinflammatory responses. Consequently, Lyst-mutant mice showed increased susceptibility to bacterial infection and were largely resistant to endotoxin-induced septic shock. Mechanistic analysis revealed that Lyst specifically controls TLR3- and TLR4-induced endosomal TRIF (TIR domain–containing adapter-inducing interferon β) signaling pathways. Loss of functional Lyst leads to dysregulated phagosomal maturation, resulting in a failure to form an activation-induced Rab7+ endosomal/phagosomal compartment. This specific Rab7+ compartment was further demonstrated to serve as a major site for active TRIF signaling events, thus linking phagosomal maturation to specific TLR signaling pathways. The immunoregulatory role of Lyst on TLR signaling pathways was confirmed in human cells by CRISPR/Cas9-mediated gene inactivation. As mutations in LYST cause human Chédiak-Higashi syndrome, a severe immunodeficiency, our findings also contribute to a better understanding of human disease mechanisms.

Crimean–Congo haemorrhagic fever (CCHF), caused by Crimean–Congo haemorrhagic fever virus (CCHFV), is a disease of worldwide importance (endemic yet not limited to Asia, Middle East, and Africa) and has triggered several outbreaks amounting to a case fatality rate of 10–40% as per the World Health Organization. Genetic diversity and phylogenetic data revealed that the Asia-1 genotype of CCHFV remained dominant in Pakistan, where 688 confirmed cases were reported between the 2012–2022 period. Currently, no approved vaccine is available to tackle the viral infection. Epitope-based vaccine design has gained significant attention in recent years due to its safety, timeliness, and cost efficiency compared to conventional vaccines. In the present study, we employed a robust immunoinformatics-based approach targeting the structural glycoproteins G1 and G2 of CCHFV (Asia-1 genotype) to design a multi-epitope vaccine construct. Five B-cells and six cytotoxic T-lymphocytes (CTL) epitopes were mapped and finalized from G1 and G2 and were fused with suitable linkers (EAAAK, GGGS, AAY, and GPGPG), a PADRE sequence (13 aa), and an adjuvant (50S ribosomal protein L7/L12) to formulate a chimeric vaccine construct. The selected CTL epitopes showed high affinity and stable binding with the binding groove of common human HLA class I molecules (HLA-A*02:01 and HLA-B*44:02) and mouse major histocompatibility complex class I molecules. The chimeric vaccine was predicted to be an antigenic, non-allergenic, and soluble molecule with a suitable physicochemical profile. Molecular docking and molecular dynamics simulation indicated a stable and energetically favourable interaction between the constructed antigen and Toll-like receptors (TLR2, TLR3, and TLR4). Our results demonstrated that innate, adaptive, and humoral immune responses could be elicited upon administration of such a potent muti-epitope vaccine construct. These results could be helpful for an experimental vaccinologist to develop an effective vaccine against the Asia-1 genotype of CCHFV.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is the etiologic agent of the ongoing pandemic of coronavirus disease 2019 (COVID-19), a public health emergency of international concerns declared by the World Health Organization (WHO). An immuno-informatics approach along with comparative genomics was applied to design a multi-epitope-based peptide vaccine against SARS-CoV-2 combining the antigenic epitopes of the S, M, and E proteins. The tertiary structure was predicted, refined and validated using advanced bioinformatics tools. The candidate vaccine showed an average of ≥90.0% world population coverage for different ethnic groups. Molecular docking and dynamics simulation of the chimeric vaccine with the immune receptors (TLR3 and TLR4) predicted efficient binding. Immune simulation predicted significant primary immune response with increased IgM and secondary immune response with high levels of both IgG1 and IgG2. It also increased the proliferation of T-helper cells and cytotoxic T-cells along with the increased IFN-γ and IL-2 cytokines. The codon optimization and mRNA secondary structure prediction revealed that the chimera is suitable for high-level expression and cloning. Overall, the constructed recombinant chimeric vaccine candidate demonstrated significant potential and can be considered for clinical validation to fight against this global threat, COVID-19.

Abstract Tubercle granulomas are highly organized immune structures that protect the host against Mycobacterium tuberculosis (Mtb) infection. The organization of immune cells around infected macrophages serves to sequester Mtb and enhance macrophage microbicidal responses. In addition, the granuloma localizes inflammation, thereby preventing damage to surrounding lung tissue. Although there is controversy regarding the involvement of Toll-like receptor 2 (TLR2) in host defense against Mtb, our findings indicate that TLR2 may be playing a key role in regulating these two opposing functions of the Mtb granuloma. Comparative studies between wild type and TLR2 gene-deficient mice demonstrated that the absence of TLR2 did not affect the magnitude of the Th1 effector response generated following aerosol infection with Mtb or the induction of recall Th1 memory immunity in response to Mtb challenge. However, the consequence of TLR2 absence was manifested at the level of the granuloma. Analysis of lungs from chronically-infected TLR−/− mice revealed increased bacterial burden and loss of granuloma integrity in comparison with infected WT mice. The infected lungs of TLR2−/− mice also exhibited enhanced inflammation and reduced Foxp3+ regulatory T cells. Together, these findings provide insights into how TLR2 might regulate the tubercle granuloma.

Chinese officials have reported the novel coronavirus to the world health organization, which is called the SARS-CoV-2. Toll-like receptor 3 (TLR3) induces antiviral immune responses via the production of type I interferons and inflammatory cytokines. In this study, we aimed to examine TLR3 c.1377C/T and -7C/A polymorphisms in COVID-19 and the association between some clinical parameters. We investigated the frequencies of TLR3 (c.1377C/T and -7C/A) polymorphisms in 150 patients with COVID-19 and 171 healthy individuals as controls. We performed polymerase chain reaction (PCR) based on restriction fragment length polymorphism (RFLP). We also investigated whether TLR3 c.1377C/T and-7C/A were associated with the severity of COVID-19. In addition, CHAID tree-based classification algorithm was created to investigate the severity of the patients in our study. TLR3 c.1377C/T TT genotype frequencies were statistically significant between cases and controls (p= 0.02). For TLR3 -7C/A polymorphism, the findings showed a statistically significant difference in A allele frequencies (p= 0.03). There was a statistically significant difference in the distribution of TLR3 -7C/A CA genotype frequency (p= 0.04). Our findings suggest that TLR3 c.1377C/T and -7C/A polymorphisms may be important on susceptibility or clinical course of COVID-19.

Mikroglia sind residente Makrophagen-artige Zellen des Zentralnervensystems (ZNS), die das Gewebe kontinuierlich auf Anzeichen homöostatischer Störungen überwachen. Als die wesentlichen immunkompetenten Effektorzellen im Hirnparenchym exprimieren sie eine Vielzahl von Rezeptoren für pathogen-assoziierte molekulare Strukturmuster (pathogen-associated molecular patterns, PAMPs). Zu diesen Rezeptoren zählt der Toll-like receptor (TLR) 4, der nicht nur Reaktionen der Mikroglia auf bakterielle Infektionen, sondern auch auf Gewebe Schädigungen ermöglicht. Stimulation des TLR4 mit bakteriellem Lipopolysaccharid (LPS) und endogenen schädigung-sassoziierten molekularen Strukturen (damage-associated molecular patterns, DAMPs), die durch Gewebebeeinträchtigung freigesetzt werden, löst sowohl TRIF- als auch MyD88-abhängige Signalkaskaden aus. Die damit induzierte Freisetzung von Zytokinen und Chemokinen rekrutiert und instruiert periphere Immunzellen für eine Protektion und unterstützende Geweberegeneration des ZNS. Wir zeigen hier, dass der TLR4-Korezeptor CD14 ein essenzieller gate keeper für die Generierung von Immunantworten im ZNS ist, die durch LPS oder E. coli-Verabreichung, aber auch durch mechanisches Trauma und ischämischen Schlaganfall ausgelöst werden. In gewissem Gegensatz zu extraneuralen Makrophagen nutzen Mikroglia CD14 zur Erlangung einer extremen Sensitivität gegenüber sehr geringen LPS-Mengen. Gleichzeitig schützt CD14 Mikroglia vor überschießenden Reaktionen auf hohe LPS-Dosen und verhindert dabei insbesondere die exzessive Produktion von CXCL1, eines chemoattraktiven Signals für neutrophile Granulozyten. Entsprechend unterstützt CD14 die ZNS-Rekrutierung von Monozyten und Neutrophilen durch niedrige LPS-Dosen, während es die verstärkte Einwanderung von Neutrophilen durch hohe Dosen von LPS oder E. coli verhindert. Als eine besonders wichtige Funktion beschreiben wir dabei die absolute CD14-Abhängigkeit DAMP-ausgelöster und TLR4-vermittelter Immunreaktionen. CD14-Defizienz (unter cd14-/--Bedingungen) oder CD14 Blockade (durch Antikörper) löschen mikrogliale Reaktionen, die durch Plasma-Fibronektin (als repräsentatives DAMP-Molekül) ausgelöst werden können, komplett aus und beeinträchtigen die Leukozyten-Infiltration nach ZNS-Trauma. Bei einer ischämischen ZNS-Schädigung weisen cd14-/--Mäuse im Gehirn nicht nur weniger Monozyten auf, sondern gleichzeitig ein vergrößertes Infarktvolumen. Wir konnten für Interferon (IFN) b eine Schlüsselfunktion in der CD14-vermittelten Eindämmung der CXCL1-Synthese darstellen, die auf eine negative CD14/TLR4-TRIF-IFNβ-INAR1-Jak- Rückkopplung für MyD88-getriebene Chemokine schließen lässt. Obwohl CD14 somit TLR4-vermittelte Reaktionen auf infektiöse und nicht-infektiöse Agenzien orchestriert, wird seine Expression durch verschiedene TLR-Liganden und Zytokine reguliert. Letztlich unterliegen damit CD14-kontrollierte Funktionen selbst einer komplexen Kontrolle durch ZNS-residente und eingewanderte periphere Zellen. Diese Regulationen können über die Einbeziehung oder den Ausschluss der Kapazitäten des TLR4-Komplexes für eine Schadenserkennung während der ZNS-Reaktionen in unterschiedlichsten pathologischen Szenarien entscheiden.

The aging problem in Taiwan has made us one of the most accelerating country in the world. The government has implemented policies for the aging problem during the last 20 years, specially in year 2016, the "Long-Term Care System 2.0" was implemented. Besides its own limited resources, the government encourages enterprises and non-profit organizations to join the Long term care system, in order to make the system more complete. With exploring and consolidating various research papers, this research has used the "case study method" and the "in-depth interview method" as the research methods. The result of this research reveals that: 1. when the organization utilizes its original resources to implement local long term care services, it can present the organization's values and its importance to the community. When an organization inputs its resources to the long term care courses planning, it will enhance its own characteristics in this area. And this in term will enhance the volunteers to have internal cohesion to the organization, which is a non-duplicable resource as to other organizations. This will also help the organization to increase the efficacy of its resources, and supplement the deficiency of the government. 2. The organization utilizes its tangible assets and invisible assets to satisfy the needs of the community care system, so that its resources can create values to the community. Its leaders and ability are the scarce internal resources, and the service given is not copyable by the other organizations. The funds needed come mostly from internal donations or charity sales, in order to supplement the deficiency from the government subsidy. 3. Under the care of organization volunteers and coworkers, the elderly people can improve their interaction skills, learning skills, and self image. The volunteers in the community care system can accumulate professional skills and coordination skills. The sense of honor gained from serving the elderlies cannot be obtained from anywhere else. These volunteers are often unpaid or with very low salaries, the support of one's s family is essential to the modern society. 4. The organization utilizes its young people and teenagers to care the elderlies, this rare model shows the convenience of local care system, to provide immediate service. The unique courses designed by the organization, is not copy able by other organizations. The testimonies of the elderly people cannot be replaced. This research can be an example for the long term care system.

This chapter reports on efforts to create a sustainable model to increase engagement, success, and retention in a MOOC for learning computer programming, for a U.S.-based national vocational program. In 2014, the training organization was one a few national and regional organizations who were awarded scholarships by a telecommunication's company to participate in a MOOC whose curriculum was informed and designed by multinational corporations to try and address the dearth of young computer programming talent. The vocational training program aimed to convert MOOC registrations into active and supported participation, with a view to increase completion. Theoretical frameworks were employed to ramp up knowledge of an unknown subject area and skill. Social connectedness methods were used to create teaching and learning communities (TLC) of support. Key results allowed the organization's trainees to outperform all other participating organizations. Resulting in the organization being awarded 500 more scholarships for computer programming that could be used over a three-year period.

Existential crises question the organization's readiness to withstand uncertainties. Geopolitics and natural disruptions have changed human behavior into a “new normal.” In improving business continuity, HEIs must strategically deal with operating models, resilience, and agility and be better prepared for disruptions. Typical HEI reactions of online home TLR lack addressing the human capacity/capability needs, technologies/infrastructure, academic/student services support, and TLR evaluation/assessments that cover the HEI broader approach of education values, systems, mechanisms and pedagogies, and student outcomes assurance. It includes HEI human, infor, and organizational capital capacities and capabilities of these systems. Being resilient and agile calls for faster decision-making focused on HEI priorities. This chapter addresses the HEI strategic readiness through a strategic organizational readiness model (SORM) managing the HEI organization readiness. The SORM evolves around the HEI six thematic systems, with resilience and agility as vital analytical and assessment parameters.

The International Society for Performance Improvement (ISPI) Human Performance Technology model was used as a guideline for this case study and applied to assess and evaluate the resource allocation at the Technology and Learning Center (TLC). This model has proven to be a useful guideline as a process to be followed during the project. A team of instructional systems technology graduate students served in a consulting role on this project to help the TLC allocate resources and redesign processes on how support tickets were handled. The project team conducted performance analysis through extensive stakeholder interviews and extant data review to perform organizational, environmental, gap and cause analysis. Through these analyses, performance issues were isolated, the causes behind them were identified and concluded with the recommendation of interventions to the client.

Throughout history, terrorist organizations have demonstrated capacity to adapt and evolve in order to further their ideological and political goals. Cyber Terrorism is increasingly becoming a top five national security priority for Nation States. The purpose of this chapter is to evaluate the threat Cyber Terrorism poses to the stability of the international community. It will explore the concept of Cyber Terrorism, its interpretations and terminology. This chapter seeks to identify potential attacks made possible through IT and ICT technologies (like SCADA and Industrial Automation, while it includes those ICT standards used in the field of Transport, Automotive, and Social Networks as well) and to classify all the possible actors, needed skills and relevant goals, thanks to the currently available public documentation and relevant case studies.

The need to introduce complex medical technologies in low- and middle-income countries (LMIC) is increasing, to improve quality of life and life expectancy. Despite improvements in medical infrastructure, many countries do not have the skills required to implement such technologies and need support from overseas organizations. Transplant Links (TLC) is a UK charity supporting the development of sustainable kidney transplant programs in LMIC countries. TLC's experience demonstrates health literacy challenges encountered in introducing a complex medical technology, of which transplantation is a good example. Such challenges were found to be wide-ranging, in a specialty already associated with low levels of health literacy. Nevertheless, such challenges need to be addressed and measures undertaken to improve the level of understanding of renal failure and transplantation by patients, families, healthcare workers, and other stakeholders including politicians, healthcare managers, and the media. These measures may be transferable to the development of other complex medical technologies.

Introduction: Long RNAs are non-coding RNAs with more than 200 nucleotides in length, with essential regulatory roles in several biological processes, including in breast cancer (BC). The human genome contains 481 ultraconserved regions, which are genomic stretches of over 200 base pairs conserved among humans, rats, and mice. Most of these regions are transcriptionally active (T-UCRs), and several are differentially expressed in tumors. Some T-UCRs have been functionally characterized, but few have been associated with BC. Objectives: In this study, we aimed to expand the knowledge of T-UCRs in BC and characterize the lnc-uc.147, a long RNA transcribed from an ultraconserved region. Methods: We evaluated the expression level of 481 T-UCRs and their association with clinical parameters from TCGA data. For confirmation, 102 Brazilian BC samples were analyzed by RT-qPCR. Cytosolic and nuclear cell fractions and RT-qPCRs were done to determine the cell compartment of the transcript. Northern blotting and RACE were performed to determine the sequence and precise size of lnc-uc.147. Using two luminal cell lines (CAMA and BT474), a siRNA-based approach was applied to investigate the effects of lnc-uc.147 knockdown in cell viability, colony formation, and apoptosis level. To understand the interactions of lnc-uc.147 and proteins, we performed a pull-down assay. Results: Using TCGA (The Cancer Genome Atlas) data, we found 302 T-UCRs related to clinical features in BC: 43% were associated with molecular subtypes, 36% with estrogen-receptor positivity, 17% with HER2 expression, 12% with stage, and 10% with overall survival. We found that uc.147 is highly expressed in luminal A and B patients, which was also confirmed in Brazilian samples. For luminal A, a subtype usually associated with better prognosis, high uc.147 expression was associated with a poor prognosis and suggested as an independent prognostic factor. The lncRNA from uc.147 (lnc-uc.147) is in the nucleus. Northern blotting results show that uc.147 is a 2,8 kb monoexonic transcript. The silencing of uc.147 increases apoptosis, arrests the cell cycle and reduces cell viability and colony formation in luminal BC cell lines. Additionally, we identified 19 proteins that interact with uc.147 through mass spectrometry. These proteins are mainly involved in cytoskeletal and centrosome organization as well as in epithelial-mesenchymal transition. Conclusions: We show herein evidence that neoplastic BC cells exhibit a unique expression profile of T-UCRs. This study characterized the lnc-uc.147, a transcript that has never been described before. Indeed, lnc-uc.147 has an oncogenic effect in the luminal BC cell line and can interact with proteins. Furthermore, uc.147 has the potential as a BC prognostic marker in luminal patients.

Abstract In south-east field, the normalized number of running swellable packers is 7 to 8 swellable packer, and yet with this number holds up occur and could raise other operational issues. In the Onshore operated field, a designated well was nominated to trial an Autonomous inflow control valve (AICV) which is a technology that improves oil production by reducing unwanted gas and water breakthrough. To have the proper implementation of the trial, the development department required 15 swellable packers to be run in the horizontal hole part of the lower completion to assure segmentation isolation and a proper positioning of the AICV. The requirement was challenging, as it was not experienced in the specified site to run this amount of swellable packers, as each swellable packer could create a force which could cause hold up and even a stuck. Part of the well planning phase, torque and drag simulation, hydraulics simulations and risk assessments were prepared and analyzed to study and evaluate the possibilities of running the 17 swellable packers along a horizontal hole section. During the execution phase, a proper drift run was in place to provide an indication of a smooth run of the lower completion. In addition to that, to assure well bore trajectory accessibility and suitability for the lower completion, a Mechanical Caliper Log run on Tubing Log conveyed (TLC) was in place. As a contingency during running the lower completion, a remote actuated sleeve was placed at the bottom of the lower completion to avoid having a close system in the hole with no circulation, which could case difficulties in releasing the string in case of any hold up or stuck. The completion deployment was successful, which was a result of a proper planning and communication from all the parties involved. The combined skills of the Operations and Engineering team was key to the safe and efficient execution of the project as a proper and continues follow up was in place by a high-competent members during the execution phase which assured the completion of the well as planned. This job was a start to encourage other fields to implement new technologies after evaluating each case separately to get the full benefit of it. In case any high risk/ unexperienced circumstances operations, with the proper planning and study, alternative procedures, processes, and technologies can be implemented to add a bigger value to the organization.
