Abstract (sommario):
This is a feasibility study for the application of a novel concept of single-needle device for localized chemotherapy. Systemic chemotherapy has numerous and severe side effects. To conduct localized (electro)chemotherapy, we designed a novel device that does not currently exist on the market. Electrochemotherapy is based on the cell membranes temporary or permanent permeabilization using an electric current of defined characteristics. Electroporation can be reversible, when after a period of opened pores and membrane permeability increasing, membranes and cells return to their original state without damage. Electroporation can be an irreversible process when the pores on the membrane remain permanently open, electrolyte imbalance occurs resulting in cell death. Electrochemotherapy involves a combination of cytostatics and reversible electroporation, when pores on the cell membrane are temporarily opened and, during that short period, a large amount of cytostatic is entered into the cell, which is a macromolecule that would not normally penetrate the cell. After closing the pores, the cytostatic remains trapped in the cell in large quantities, multiplying its effect. In this paper, we present a feasibility study of electroporation application in irreversible mode without the use of cytostatics. Fresh porcine liver tissue was used to show that the constructed equipment was effective, thus opening the way for further investigations using reversible electroporation with the application of cytostatics, which would represent localized electrochemotherapy. We penetrated the virtual tumor area (liver metastases) with a specially designed needle with electrodes that generate an electric field and apply electroporation in the target tissue. We have shown that the constructed novel design single needle equipment for electroporation is effective on the experimental model of isolated porcine liver. Further steps in our study will be the testing of electrochemotherapy in an experimental animal model in vivo.

Abstract (sommario):
Single plate shear connections are designed to have sufficient strength to resist the shear force and moment transferred from the beam. The connections must also provide sufficient ductility to allow the beam end to rotate freely. In the United States the current recommended design method is found in the AISC 13th Edition Steel Construction Manual (2005b). The limited experimental work which led to the current method necessitated additional single plate shear connection investigations. This paper summarizes the results and analysis of eight full scale single plate shear connections tested at Virginia Polytechnic Institute and State University. The test setup consisted of a test beam attached to a test column with a single plate shear connection at one end and supported by a roller at the other end. The single plate was welded to the column flange and bolted to the beam web. Load was applied to the test beam at third points until failure of the connection or test beam. The current design method used in the United States was examined with respect to the connection tests performed. In particular, the ultimate shear strength and the rotational capacity were investigated. Suggestions are made regarding changes to the method and further research.

Purpose: The aim of this study was to examine whether a verb-focused language intervention was effective in increasing children’s verb-vocabulary. In particular, this study investigated whether the treatment resulted in changes to children’s production of target words compared to control words for children who are late talkers. Method: The study utilised a single-subject, multiple baseline across behaviours design. Four children, aged 26-to-39 months who exhibited delayed expressive language development participated in the study. At the beginning of the study, all children had poor expressive language performance indicated by a mean length of utterance two standard deviations below the mean expected for their age and limited vocabulary measured by the New Zealand Communicative Development Inventory: Words and Sentences. New verb-vocabulary items were randomly assigned to intervention and untreated control conditions and probed at regular intervals over a period of eight weeks. Results: All the participants showed increased use of the target verbs compared to the control verbs during the intervention and post-intervention phase. Conclusion: The findings suggest that a verb-focused language intervention was effective in increasing the verb-vocabulary of late talkers. Further research is warranted to determine whether similar results can be found with a larger cohort and whether these gains are sustained over time.

This study presents a set of data analysis approaches for single subject designs (SSDs). The primary purpose is to establish a series of statistical models to supplement visual analysis in single subject research using Bayesian estimation. Linear modeling approach has been used to study level and trend changes. I propose an alternate approach that treats the phase change-point between the baseline and intervention conditions as an unknown parameter. Similar to some existing approaches, the models take into account changes in slopes and intercepts in the presence of serial dependency. The Bayesian procedure used to estimate the parameters and analyze the data is described. Researchers use a variety of statistical analysis methods to analyze different single subject research designs. This dissertation presents a series of statistical models to model data from various conditions: the baseline phase, A-B design, A-B-A-B design, multiple baseline design, alternating treatments design, and changing criterion design. The change-point evaluation method can provide additional confirmation of causal effect of the treatment on target behavior. Software codes are provided as supplemental materials in the appendices. The applicability for the analyses is demonstrated using five examples from the SSD literature.

Acceptance and Commitment Therapy (ACT) has been shown to be an effective alternative therapeutic approach for adults of various presenting problems as well as those with whom traditional forms of cognitive-behavioral therapy have been futile (Asmundson & Hadjstavropolous, 2006; Bach & Hayes, 2002; Clarke, Kingston, James, Bolderston & Remington, 2014; Dimidjian, Hollon, Dobson, Schmaling, Kohlenberg et al., 2006; Martell, Addis, & Dimidjian, 2004). Recently, researchers and theorists have posited about the application of ACT with youth, however, little research has been conducted utilizing quantitative measures of change in psychological flexibility. This article presents a single-subject multiple probe experimental design of ACT adapted for application to adolescents. This study compared outcomes of two therapeutic approaches, ACT modified for adolescents and Treatment as Usual, by assessing changes in values (PVQ II; Blackledge, Ciarrochi, & Bailey, 2005), acceptance (CAMM; Greco & Baer, 2006), defusion (AFQ-Y; Greco, Murrell, & Coyne, 2005), committed action (diary card and direct behavioral observation), and quality of life (YQOL-R; Edwards, Hueber, Connell, & Patrick, 2002). Behavioral changes as observed by parent and teacher report (BASC-2; Reynolds & Kamphaus, 2004) were assessed at both pre- and post-intervention to provide an objective measure. By the end of treatment, the ACT participant showed a significant decrease in internalizing problems compared to the TAU participant, indicating improvements in psychological flexibility. Change in isolated ACT processes was non-significant between participants. Findings provide evidence supporting previous research suggesting that ACT can be successfully adapted to and may improve the psychological flexibility of adolescents.

The experimental study reported upon in this thesis focused on the development of physical data characterizing the behavior of single-shear, laterally-loaded connections when loaded up to and beyond capacity (i.e., maximum resistance). Specimens included a wide array of connection configurations common in wood construction. All connections were tested monotonically in tension under displacement-controlled loading, parallel to the grain. Results of these tests are presented and discussed. Test variables of nailed connections included nail diameter, side member material type, and side member thickness. Test variables of bolted connections included bolt diameter, commercial species grouping of the main and side members, and main member thickness. Conclusions drawn from this research include mechanics-based explanations of numerous connection response trends observed with respect to test variables. Additionally, observed factors of safety and over-strengths of current design values are quantified on a capacity-basis.

Silicon is commonly used in infrared (IR) imaging systems. The surface quality is an important issue in optics manufacturing since surface roughness affects optical performance of imaging systems. Surface quality of an optical component is determined by number of factor, including cutting parameters
cutting speed, depth of cut and feed in radial direction. In this thesis, an experimental study has been performed to investigate the relation between cutting parameters and average roughness of the surface of silicon. In the experiments, silicon specimens, which have a diameter of 50 mm, were face turned by using a 2-axis CNC single point diamond turning machine. The specimens were machined by using either constant spindle speed or constant cutting speed. Two different tools with rake angles of -15 degrees and -25 degrees were used. The attained surfaces were measured by using a white light interferometer, which has a resolution of 0.1nm. The experiments were designed according to the factorial design method, considering cutting parameters. The effects of cutting parameters and tool rake angles on surface quality of silicon were observed. The best average surface roughness obtained was about 1 nm which is quite better than the acceptable average surface roughness level of 25 nm.

Threshold tacking studies involve studying the excitability of a motor unit (MU), the fundamental physiological unit of our motor system. The excitability property of a MU can be parameterised by its threshold and a range of stimulus intensities over which a MU shows a stochastic behaviour. Statistical methods are seldom used for this purpose and judgments are subjective. Two main interests in threshold tracking studies are i) A novel approach that makes it possible to obtain reliable estimation of the MU's excitability parameters, and ii) The efficient use of the available resources. The aim is to cover both goals. The efficiency of both classical and Bayesian approaches for estimation MU's parameters is discussed. A MU exhibits binary responses to given stimulus voltages. This makes the generalised linear models (GLMs) a standard approach to model its behaviour. The standard issue with experimental design for GLMs is the dependency of the design to the unknown model parameters. The need for optimal initial design points is another challenge faces the experimenters. We develop a sequential Monte Carlo (SMC) approach for Bayesian analysis of experimental design for binary response data that selects design point based on an online manne via minimising an expected loss. We will apply this loss function to the estimates of target quantiles from the stimulus-response curve. Through simulation we show our approach is more efficient than the existing sequential design approaches; Wu's improved Robins-Monro, generalised polya urn (GPU) based models, and 2-point design that uses D-optimality criterion. By reducing up to more than halve the length of the threshold tracking experiments, our approach uses priors on unknown parameters to tackle the dependency to the design to unknown parameters. Regardless of how to initialise the our sequential approach, its efficiency is also independent of initial design points.

Background: Prospective memory difficulties are commonly reported in people with dementia. The evidence supporting the use of prospective memory devices among the dementia population remains limited. MindMate is a recently developed smart device application that aims to support individuals with a diagnosis of dementia, improving self-management skills and quality of life. Aims: This study investigated the effectiveness and usability of the reminder tool on the MindMate application as a memory aid. Method: Three participants with a diagnosis of mild Alzheimer’s disease were recruited to this multiple baseline single case experimental design study. Partners of the participants recorded their performance on everyday tasks on weekly monitoring forms during a baseline phase (for between five and seven weeks) and during the intervention phase (five weeks) whilst using MindMate. Results: Two participants successfully used the app throughout the intervention weeks and gave positive usability ratings. Tau-U analysis showed a significant increase in memory performance between baseline and intervention phase (Tau-U = 1, 0.94, p < 0.01). A third participant withdrew from the intervention phase following difficulties turning off the reminders and frustrations with the reminder alert sound. Conclusions: The use of the MindMate app was feasible for people with dementia in the community. It was effective compared to practice as usual, with participants reporting intentions to use in the future. Limitations and implications for future research are discussed.

The monograph considers models of the interaction of radiation fields of single-mode and multimode Gaussian laser beams with the optical system of the eye. Formulas have been obtained that allow calculating the coefficient of the degree of danger of laser radiation both by the criterion of damage to the skin and cornea of the eye, and by the criterion of damage to the retina at any distance from the radiation source. Methods for selecting control points are proposed. Examples of specific methods of dosimetric control of laser radiation at the workplaces of personnel servicing laser installations are given. The theoretical foundations of circuit design solutions for laser dosimeters are considered. Modern requirements for the metrological characteristics of dosimeters, as well as methods and means of their verification are presented. In the interest of understanding contact mechanics, friction and wear processes where plastic deformation occurs between rough surfaces, significant effort has and continues to be applied to understand single asperity elastic-plastic contacts. The main tools used in obtaining experimental data with which to inform and validate simulation methods in this area of study are nano and micro indenters. This article presents some of the less commonly considered phenomena which may affect the interpretation of experimental data from such apparatus. The interpretation of AFM pull off data is briefly discussed and invasive effects of electron imaging are highlighted.

A number of turbulence models offered by the commercial code STAR-CD have been tested on the measurements published in the literature with the objective to compare their capabilities for the simulation of a flow and heat transfer in multiple impinging jets. Numerical predictions of the single jet and jet array air impinging heat transfer have been compared with experimental data. The comparison shows that only turbulence models with additional limiters for turbulence production in the stagnation zone are able to correctly predict the jet impingement heat transfer. Suga’s k-ε turbulence model with Yap-correction, k-ε RNG, V2F and SST turbulence models with different near wall modifications are in acceptable agreement with experiments. The deviations from the experimental data, which provide all the turbulence models, are analyzed.

Abstract When a multi-body system collides with a single body or with another multi-body system, impact dynamics with friction should be considered. This paper presents a general computer oriented analysis of impact dynamics incorporating friction. The presence of friction between sliding contacts during the impact makes the problem difficult since the events such as reverse sliding or sticking, which may occur at different times throughout the impact, must be determined. The boundary representations of the bodies are used to solve for the velocities at the points of contact. Using this information and a classification of the modes of impact, the frictional impact with sliding contact problem is solved. Using a high speed video camera, the resulting computer strategy is experimentally verified. Simulation and experimental results agree.

Abstract In the present paper a single-degree-of-freedom, periodically forced impact oscillator is investigated by numerics and experiments. At first the different types of motion and bifurcation diagrams are determined from experiments and compared to numerical results on the basis of the identified impact model. Also the nonsmooth third order system shows a rich bifurcational behaviour. On the basis of the identified nonsmooth model further analytical and numerical analysis can be applied. The Lyapunov exponents and the winding number are evaluated for the non-smooth system.

There has been extensive research and investment in strategies that aim to improve the quality of education around the world. But despite rigorous evidence, the question of what to do in a specific context usually remains only partially answered at best. As a result, numerous impact evaluations are being conducted with the goal of learning what works. The goal of the guidelines presented here is to summarize the evidence and provide references in a single document in order to save time for those implementing education impact evaluations through randomized control trials. The guidelines focus on supporting evaluations that help policymakers determine where to assign limited resources to improve the quality of education. The document has five sections. The first reviews empirical findings to provide a general idea of why education is important, which inputs matter, and howto conduct an impact evaluation. The second section provides guidance on how to define the impact evaluation hypothesis. The third section presents a methodology for selecting the sample and setting up a randomized impact evaluation. The fourth section provides information on what data to collect for the evaluation, and the final section looks at how to analyze data and strategies to adjust the analysis for changes in the original design. The guidelines are not exhaustive: their main contribution is to present a methodology to design an impact evaluation, outline the necessary inputs to start the design of such an evaluation, and provide a structured source of rigorous references.

This is the last report of a�c reports detailing the small-scale mechanical testing performed on the trial welds in this consolidated program. This report summarizes and compares the results of all of the mechanical tests applied primarily to welds of rounds 1 and 2, including tensile results and their correlation with microstructure, Charpy test results, conventional (through-thickness-notched) toughness tests, and low-constraint toughness tests. The reports contains a summary of the mechanical properties of the experimental single and dual torch GMAW-P X100 pipe welds prepared for this consolidated program. It summarizes the detailed results reported in Final Reports 277-T-05, 277-T-06 and 277-T-07. The intent of this summary is to provide insight and understanding of the significance of the results and the implications for mechanical testing of weldments to extract properties essential for strain-based design (SBD).

Over the past decade, several long-duration subduction earthquakes took place in different locations around the world, e.g., Chile in 2010, Japan in 2011, China in 2008, and Indonesia in 2004. Recent research has revealed that long-duration, large-magnitude earthquakes may occur along the Cascadia subduction zone of the Pacific Northwest Coast of the U.S. The duration of an earthquake often affects the response of structures. Current seismic design specifications mostly use response spectra to identify the hazard and do not consider duration effects. Thus, a comprehensive understanding of the effect of the duration of the ground motion on structural performance and its design implications is an important issue. The goal of this study was to investigate how the duration of an earthquake affects the structural response of special concentric braced frames (SCBFs). A comprehensive experimental program and detailed analytical investigations were conducted to understand and quantify the effect of duration on collapse capacity of SCBFs, with the goal of improving seismic design provisions by incorporating these effects. The experimental program included large-scale shake table tests, and the analytical program consisted of pre-test and post-test phases. The pre-test analysis phase performed a sensitivity analysis that used OpenSees models preliminarily calibrated against previous experimental results for different configuration of SCBFs. A tornado-diagram framework was used to rank the influence of the different modeling parameters, e.g., low-cycle fatigue, on the seismic response of SCBFs under short- and long-duration ground motions. Based on the results obtained from the experimental program, these models were revisited for further calibration and validation in the post-test analysis. The experimental program included three large-scale shake-table tests of identical single-story single-bay SCBF with a chevron-brace configuration tested under different ground motions. Two specimens were tested under a set of spectrally-matched short and long-duration ground motions. The third specimen was tested under another long-duration ground motion. All tests started with a 100% scale of the selected ground motions; testing continued with an ever-increasing ground-motion scale until failure occurred, e.g., until both braces ruptured. The shake table tests showed that the duration of the earthquake may lead to premature seismic failure or lower capacities, supporting the initiative to consider duration effects as part of the seismic design provisions. Identical frames failed at different displacements demands because of the damage accumulation associated with the earthquake duration, with about 40% reduction in the displacement capacity of the two specimens tested under long-duration earthquakes versus the short-duration one. Post-test analysis focused first on calibrating an OpenSees model to capture the experimental behavior of the test specimens. The calibration started by matching the initial stiffness and overall global response. Next, the low-cycle fatigue parameters were fine-tuned to properly capture the experimental local behavior, i.e., brace buckling and rupture. The post-test analysis showed that the input for the low-cycle fatigue models currently available in the literature does not reflect the observed experimental results. New values for the fatigue parameters are suggested herein based on the results of the three shake-table tests. The calibrated model was then used to conduct incremental dynamic analysis (IDA) using 44 pairs of spectrally-matched short- and long-duration ground motions. To compare the effect of the duration of ground motion, this analysis aimed at incorporating ground-motion variability for more generalized observations and developing collapse fragility curves using different intensity measures (IMs). The difference in the median fragility was found to be 45% in the drift capacity at failure and about 10% in the spectral acceleration (Sa). Using regression analysis, the obtained drift capacity from analysis was found to be reduced by about 8% on average for every additional 10 sec in the duration of the ground motion. The last stage of this study extended the calibrated model to SCBF archetype buildings to study the effect of the duration of ground motion on full-sized structures. Two buildings were studied: a three-story and nine-story build that resembled the original SAC buildings but were modified with SCBFs as lateral support system instead of moment resisting frames. Two planer frames were adopted from the two buildings and used for the analysis. The same 44 spectrally-matched pairs previously used in post-test analysis were used to conduct nonlinear time history analysis and study the effect of duration. All the ground motions were scaled to two hazard levels for the deterministic time history analysis: 10% exceedance in 50 years and 2% exceedance in 50 years. All analysis results were interpreted in a comparative way to isolate the effect of duration, which was the main variable in the ground-motion pairs. In general, the results showed that the analyzed SCBFs experienced higher drift values under the long-duration suite of ground motions, and, in turn, a larger percentage of fractured braces under long-duration cases. The archetype SCBFs analysis provided similar conclusions on duration effects as the experimental and numerical results on the single-story single-bay frame.

From one point on the circumference of the pipe, it is therefore proposed to inspect the entire circumferential area of the pipe. This approach leads to inspection efficiency.� Experimental techniques cannot be developed on a trial and error basis. The purpose of this work is therefore to develop a guided wave propagation model and subsequent numerical solutions to assist in the �theoretically driven� sensor design and data acquisition systems for improved nondestructive testing including defect detection, location, and potential classification and sizing analysis in a pipe.

Prior full-scale research with the fire service was primarily designed to isolate specific tactics, most often either ventilation or suppression, which allowed researchers to develop science-based recommendations related to the specific components of fireground operations studied in relatively controlled conditions. The current project went beyond earlier research by conducting twenty experiments in eight acquired, single-family residential structures and that combined fireground tactics to quantify the impact of coordination between ventilation and suppression actions. This experimental series included second-story bedroom fires (14 experiments) and first-floor kitchen fires (6 experiments). The main control variables studied included the position of initial application of water, the ventilation method, and the timing of ventilation relative to water application. The ventilation tactics examined in these experiments included horizontal, vertical, positive pressure, and hydraulic ventilation, while the suppression tactics included both interior water application and initial exterior water application followed by interior water application. While some elements of the experiments (e.g. structure floor plan and weather) resulted in increased variability, the lessons learned highlighted the importance of having a systematic approach to the implementation of tactics. Most importantly, there was no meaningful increase in temperature outside of fire rooms when ventilation tactics were executed in coordination with (shortly after or shortly before) the onset of suppression. The effectiveness of suppression actions in extinguishing the fire were dependent on the ability of those actions to 1) cool surfaces in the fire room and 2) wet unburned fuel. Exterior suppression actions on second-floor bedroom fires resulted in a decrease in temperatures throughout the second floor, followed by regrowth prior to final suppression through interior streams. When exterior suppression was performed on first-floor kitchen fires, where more complete fuel wetting was possible, regrowth was not observed prior to interior suppression. When surface cooling or fuel wetting are not possible due to the elevation of the fire room, missing ceiling, or obstacles, firefighters should consider alternative means of water distribution to improve the effectiveness of suppression actions from outside the fire room. Suppression actions, whether interior or exterior, generally resulted in a decrease in temperatures and gas concentrations at locations where occupants may potentially be located. Conditions improved most quickly at locations closest in proximity to the inlet of the flow path established between the front door and the fire room. For this reason, opening an exterior door to gain access should be thought of as an important ventilation action, both in terms of its potential to cause fire growth and its potential to improve conditions for potentially trapped occupants. After effective suppression, structure ventilation operations should similarly be cognizant of gas flows, with the aim of establishing flow throughout all areas where occupants may be located.
