Abstract (sommario):
Throughout history, people have made or acquired vessels from which to sip their favourite beverage. In the contemporary domestic setting, households frequently accumulate multiples of the same type of object in numbers that are considerably greater than is necessary and practical for use alone. Of these many objects there are often individual pieces that have special significance for the owner or user. Some are so valued that they may even be removed and set aside because of their perceived importance. The research was initiated by a previous study of tea drinking vessels coupled with a desire, as an object maker and collector, to find out why people have special items that they designate as personally important. The aim was to identify how significance could be recognised in specific objects and whether the notion that a group of features used to gauge such objects could be conveyed into studio based work. The research outcomes are evidenced in a text-based document (which articulates the theoretical and empirical elements of the enquiry) and a body of creative studio work developed in response to aspects of the investigation. The document encompasses two components of the study. The first references material from the fields of museum and cultural studies, pivotal in focusing the enquiry. This contributed to the compilation of a general and speculative inventory of qualities that might pertain to objects deemed ???significant???. During these early investigations it became evident that a more in depth and contemporary analysis of significant drinking vessels, their owners and/or users was required. A Survey Questionnaire regarding personal use and special drinking vessels preceded a series of Interviews with a selected group of Australia curators, artists, academics and collectors who discussed and analysed their association with a personally significant drinking vessel. Subsequently, the content of these interviews became central to the focus of the research and outcomes. The research isolates a number of attributes that are commonly identified in objects that, whatever their condition, are deemed ???significant???. These describe the maker, usage, ownership, association and historical context. The perceived value or worth of the object for its owner, is recognised as a consequence of significance and declares the object as distinctive. This outcome is clearly validated by the interviews. The studio work develops from the fusion of personal narrative that has been enhanced by findings of the research. In particular, it references the cherished object, most especially those pieces that have been retained despite the ravages of time and use. The resulting work was exhibited as Trace Elements ??? Marking Time: Significance, the Vessel and the Domestic at Kudos Gallery, Paddington in April 2004.

This thesis proposes a reconstruction and reconceptualisation of the cultural significance provisions of the Protection of Movable Cultural Heritage Act 1986 (Cth). It takes the form of a critical study of the role, function and legal effectiveness of these provisions, and proposes the development of new understanding of what is involved in the construction of cultural significance by law. This thesis is being carried out in both the conceptual and practical domain. However, it is neither a theoretical study nor an exercise in doctrinal exegesis on its own, but relies on both to undertake a practical and functional study into the operation of this legislation and its practices. By drawing on the insights of cultural legal studies and museums studies in which to critically examine the law and its practices, it may be characterised as an intersectional and interdisciplinary study that provides an exchange between law and the policy and practices of the field with which it is engaging. The cultural significance provisions of the Protection of Movable Cultural Heritage Act 1986 (Cth) are used to decide whether culturally significant objects should be kept in Australia for the benefit of the national interest. If an object is 'culturally significant enough', it will be refused an export permit. Because the legislation intrudes on the rights of property owners, decisions to refuse export permits may be reviewed by the Administrative Appeals Tribunal ('the AAT'). However, this thesis argues that the interpretation of the cultural significance provisions by the AAT has reconstructed their meaning into existing legal forms and categories, by imposing law's analytical conception of objects. This method analytically separates the physical manifestation of the object from the discursive factors on which its cultural significance is based. As a result, the cultural significance provisions cannot effectively explain what makes an object culturally significant, in particular when a physically insignificant, but highly culturally significant object has to be tested against these provisions. As a result the conceptual basis on which the legislation rests is in an unsatisfactory state. At one level, the problems with the interpretation of the cultural significance provisions are, in part, derived from the tension between the market and the national interest that characterises this area of law. The clash between the market and national interest was at the forefront of the policy debates surrounding the introduction of the legislation into Australia. A case study shows how political and commercial pressures may influence decision-making when no criteria exist to decide whether objects are culturally significant or not. This case study considers how paintings and other fine art objects are dealt with under the cultural significance provisions, focussing on a dispute about the protection of contemporary indigenous art that entered the public arena in 1999, which leaves the impression that cultural significance decision-making operates in a vacuum that will allow for any outcome that may be commercially or politically palatable. At a more prosaic level, this study reveals that fundamental problems exist in the practical functioning of the legislation and its administration. The cultural significance provisions themselves are cumbersome and difficult to follow. The legislation relies on the advice and input of specialists in a range of areas of expertise relating the Australia's cultural heritage to assist with the process of decision-making. However, the practices of cultural significance decision-making and the legislative structure are disconnected, meaning that the experts in the field are making recommendations based in one discourse, while law's analysis follows a different method. This thesis argues that the 'cultural significance provisions' can only be effectively construed and interpreted if the narrative and discursive framework for 'cultural significance assessment' used by the museums sector are overtly embraced within law's conception of what makes an object 'culturally significant'. It sets out and explains the cultural significance assessment techniques used by Australian museums to evaluate the cultural significance of objects. The 'discursive techniques' of the museological method are tested against existing cultural significance decision-making. It will be shown that this method provides reasoned explanations for an object's cultural significance, and the adoption of this methodology would lead to an improved decision-making process. However, this thesis concludes by observing that fundamental policy issues will impact on any proposals for change.

The desire for preserving a portion of this country's cultural resources has forced archaeologists and resource managers to make determinations about the relative significance of archaeological sites. Decisions are made regarding which sites are to be preserved and which must be dismissed in the name of progress. The first half of this thesis traces both the history and evolution of the significance concept, and briefly reviews some of the means and strategies archaeologists use in making significance evaluations in the face of continued economic development and a rapidly changing theoretic and methodological discipline. The second half of this thesis demonstrates a proposed significance preservation model for the known archaeological resources using data from the Upper Yalobusha River Basin in north central Mississippi. Unlike preservation strategies that evaluate resource significance on a site-by-site basis, this model establishes preservation areas based on a twenty percent representative sample of the known cultural resources in the region.

Introduction: Testicular microlithiasis is an uncommon condition characterized by calcifications within the seminiferous tubules. The true prevalence in a normal population has not been defined. Methods: A review of the literature with emphasis on the connection between testicular microlithiasis and testicular malignancy was carried out. Results: Testicular microlithiasis is associated with different testicular pathologies, including testicular cancer. However, a direct causative connection between testicular microlithiasis and testicular pathologies is not supported by the literature. Conclusions: Patients with testicular microlithiasis should be followed up regularly. Further investigations concerning the etiology of testicular microlithiasis remain to be done

This dissertation is about fairness and the role it plays in political and personal morality. Specifically, I investigate when it is appropriate to rely on considerations of fairness to draw substantive conclusions about what we should do. In Chapter 1 ("Numbers, Fairness and Beneficence") I discuss the "numbers problem," the problem of explaining why you should save more people rather than fewer when forced to choose. Existing non-consequentialist approaches to the problem appeal to fairness to explain why. I argue that this is a mistake and that we can give a more satisfying answer by appealing to requirements of beneficence or generosity. In Chapter 2 ("Fairness, Distributive Justice and Global Justice") I discuss justice in the distribution of resources, both within states and across different states. On one influential view, it is always unjust for one person to have less than another through no fault of her own. State borders, on this account, have no importance in determining which distributions are just. I show that an alternative approach is needed. I argue that distributions of wealth are only unjust in so far as they issue from unfair treatment. It follows that not all inequalities in the distribution of goods are unjust. I use these results to explain how state borders do play a role in determining which inequalities are unjust, since some of these inequalities issue from unfair treatment of citizens by the state. In Chapter 3 ("Contractualism, Politics and Morality") I discuss Rawls' contractualist theory of social justice and Scanlon's extension of it to provide a theory of "rightness", or morality more generally.
(cont.) I argue that while there is some justification for adopting a contractualist theory of social justice, this justification does not support a contractualist theory of rightness. This is because social justice is centrally a matter of cooperative fairness whereas rightness is not.

In this thesis, I investigate the nature of metaphysics and the role it plays in our broader theoretical pursuits. In doing so, I defend it against various criticisms and offer a novel conception of why metaphysical disputes are important. In Chapter One I address the general question of when disputes are defective and when they are worth taking seriously. I first criticize one popular way of answering this question that appeals to the difference between verbal and factual disputes. Verbal disputes involve divergence in what the participants mean by their terms and some think that metaphysical disputes are defective in this way. I argue that this approach fails because the verbal/factual distinction is incapable of doing the work this view requires of it. I then offer an alternative view where the status of a dispute depends on its role in our theorizing. Worthwhile (or, as I call them, significant) disputes are those with appropriate connections to the rest of our theorizing while defective (or insignificant) disputes are insular, with no implications for anything beyond themselves. In Chapter Two I apply the framework developed in the previous chapter to a pair of ontological disagreements: those over the existence of concrete possible worlds and coincident material objects, like a statue and its clay. The question is whether ontological disputes like these have the requisite theoretical connections to render them significant. I argue that they do. I then address some general reasons for doubting their significance, arguing that they are not compelling. In Chapter Three I contrast my approach with some other recent defenses of metaphysics, with particular focus on the views developed by Theodore Sider. Metaphysics is, on these views, an inquiry into the world's fundamental structure. I argue that this approach is unsatisfactory because it cannot guarantee that metaphysical disputes are significant in the way I describe. It thus threatens to render metaphysics irrelevant to our other theoretical activities, undermining many of its legitimate successes, like the role theorizing about metaphysical modality played in the development of modal logics applicable in many different fields.

The study investigates the relevance of public, historic buildings in an urban context with specific focus on the old ZAR Staatsmuseum in Pretoria. The project aims to find a way, to re-establish the Staatsmuseum as a significant b uilding, a b uilding with dignity, through making it relevant within the urban context, once again. The heritage response that will be investigated is an adaptive re-use approach, which refers back to the statement of significance of the building, as formulated as part of this study. The programmatic response is associated with the initial program of the building, as a Life Sciences museum will be introduced in the building. The concepts of plinth, entrance, exhibition (including light and movement) and courtyard are specific design drivers, that will be investigated and responded to as part of the design process.

Noncovalent interactions are at the center of many biological and chemical processes. There has been a conscious effort in recent times to exploit these interactions in order to achieve specific targets. However, to fully unravel and exploit the potential of these weak interactions, it is necessary to understand their effects and efficacy in greater detail. Noncovalent interactions that have garnered maximum attention in terms of the ubiquity, bond-strength, and practical applications are those that are dominated by electrostatic contributions. Noncovalent interactions are defined to be the combination of some physically well-defined contributors: electrostatics, dispersion, donor-acceptor charge transfer, polarization and Puali repulsion. Recent literature reveals that the polarization and donor-acceptor charge transfer are electrostatic in nature. According to the Feynman interpretation, even the dispersion interaction is electrostatic in origin. Therefore, proper treatment of electrostatic interactions is adequate to describe all kinds of noncovalent interactions fully. This thesis is dedicated to the studies of those noncovalent interactions that are dominated by electrostatic contributions.

Work in this thesis has focused on identifying a biological role for the incidence of osteocyte apoptosis, the identity of osteocyte-derived resorption targeting signals and addresses the design of methods of blockade of the excess apoptosis observed during the use of glucocorticoids. An early event in the bone resorption process is the clearance of osteoblasts; hence I have studied the effects of osteocyte apoptotic bodies on osteoblastic cells. Apoptotic bodies derived from different cell types were also used to determine behavioural responses in osteoblast and other potential target cells. Osteoblasts were shown to undergo apoptosis upon contact with osteocyte apoptotic bodies, while apoptotic products derived from different cell types, did not induce osteoblast death. Furthermore, target cells other than osteoblasts, did not undergo apoptosis in the presence of osteocyte or other cell type apoptotic products. These data indicated a specific role for osteocyte apoptosis in the bone microenvironment that might be directing the behaviour of osteoblasts in the bone remodelling process.  In addition, the demonstration of phenotypic specificity in the apoptotic body-derived signals points to a further level of physiological “meaning” to apoptosis. The physical interaction of apoptotic osteocytes with osteoblasts and the resultant osteoblast death were dependent t on membrane changes in the apoptotic body and appeared to involve the phagocytic receptors CD14 and scavenger receptor A as evidenced by using gene knockout animal models. Having identified an important potential role for apoptotic osteocytes in targeting osteoblast removal at sites of remodelling, mechanisms that could interfere with the induction of osteocyte apoptosis in response to Dexamethasone were investigated. Dexamethasone-induced apoptosis was associated with activation of the Fas death receptor and ERK1/2 pathways. Bisphosphonates suppressed the pro-apoptotic stimuli, independently of their detailed structure and their in vivo anti-resorptive potency. These data suggested that bisphosphonates could provide therapeutic approaches against excess osteocytic death observed in glucocorticoids-induced osteoporosis in order to maintain a balance between osteocyte viability and death, which might ultimately lead to stabilised bone turnover activity and better bone quality.

Debate concerning the relationship between ethics and aesthetics has re-emerged in contemporary aesthetic literature. All of the major contemporary positions, I argue, treat this relationship as existing between the "moral value" of art and its aesthetic value. Throughout this thesis I analyse the various "value- based" positions (ethicism, moderate moralism, and contextualism) and examine whether their accounts of this relationship hold. My aim is to explore whether an alternative account - in which the aesthetic value of art can be enhanced or negated through its "moral significance", rather than its "moral value" - is plausible. I argue, that given the failure of these value- based positions we should favour a "significance- based contexutalist" approach that is better equipped to account for the complexity of both our engagement with art, and the moral reflection that it invites.

Diese Dissertation unterstreicht die Rolle von Informationen bei der Erforschung ökonomischer Entscheidungen. Das erste Kapitel beschäftigt sich mit dem Zugang zu akkuraten Informationen über die Wirksamkeit medizinischer Interventionen. Unserem Model zufolge besteht in vielen entwickelten Ländern Nachfrage für Zugang zu unabhängigen medizinischen Informationen wie Cochrane Reviews. Wir schätzen, dass für viele Länder diese Nachfrage zu moderaten oder geringen Kosten erfüllt werden kann. Das zweite Kapitel beschäftigt sich mit der Kommunikation solcher Informationen und untersucht den unterstützenden Effekt natürlicher Häufigkeiten bei der Berechnung von A-posteriori Wahrscheinlichkeiten (Gigerenzer & Hoffrage, 1995). Durch eine Meta-Analyse erklären wir Konzepte und entflechten die Effekte von 15 Studienmerkmalen. Im einfachsten Studiendesign führen natürliche Häufigkeiten zu 24 Prozent korrekten Antworten verglichen mit 4 Prozent bei konditionellen Wahrscheinlichkeiten. Die finalen beiden Kapitel analysieren Satisficing-Strategien für unsichere Entscheidungsumgebungen in denen Agenten eine vollumfassende, probabilistische Beschreibung des Entscheidungsproblems fehlt. Simon (1955) zufolge nutzen Satisficing-Strategien ein Anspruchsniveau um die Suche nach weiteren Entscheidungsalternativen zu beenden. Das dritte Kapitel beschreibt wie solche Strategien in der ökonomischen Literatur als Präferenz modelliert werden um Entscheidungen zu erklären die der Nutzenmaximierung unterlegen sind während die Kognitionswissenschaften diese Strategien als Lösungen für Inferenzprobleme betrachten. Wir erklären die Divergenz mit unterschiedlichen Annahmen über die vorliegenden Informationen der Agenten. Das letzte Kapitel untersucht Satisficing-Strategien unter Taxifahrern. Wir stellen fest, dass die Stundenlöhne von Taxifahrern kaum vorhersagbar sind und ihre Entscheidungen Schichten zu beenden am besten durch einfache Satisficing-Strategien vorhergesagt werden können.
This doctoral thesis emphasizes the significance of informational conditions in studying economic decisions. The first chapter concerns access to accurate probabilistic information in the domain of medical interventions. We estimate that, in many developed countries, there appears to be demand for governments to grant citizens free access to impartial reviews of medical evidence, as provided in Cochrane Reviews. For these countries, we estimate that this demand can be met at low costs. The second chapter concerns the communication of such information and examines the facilitating effect of natural frequencies on the derivation of posterior probabilities, as delineated by Gigerenzer and Hoffrage (1995). In a meta-analysis, we clarify concepts and disentangle the effects of 15 study characteristics. We find that in the simplest study design, 4 percent correct solutions when presented with conditional probabilities and 24 percent when presented with natural frequencies. The final two chapters examine the satisficing class of strategies for uncertain decision environments in which agents lack a full probabilistic description of the decision problem. According to Simon (1955), satisficing strategies use aspiration levels to terminate search for suitable alternatives. The third chapter describes how in economics, satisficing is modeled as a preference structure or as a decision rule that yields choices inferior to utility maximization, whereas in cognitive science, satisficing strategies use aspiration levels to solve inference problems. We explain the divergence, noting that they refer to risky and uncertain environments, respectively. The final chapter examines satisficing in an applied setting, studying taxi drivers' shift termination behavior. We find that drivers' hourly earnings are very uncertain and drivers’ behavior is best predicted by simple satisficing strategies that terminate shifts when reaching an aspired shift income or shift duration.

Civility has many critics. Some challenge its distinctness as a virtue, others its moral significance. In this essay, I attempt to meet both challenges by offering an account of civility that stands distinct from other concepts and holds great value, both intrinsically and instrumentally. I claim that we show civility toward others when we dispute their ideas in ways that respect those persons' intrinsic worth. My account pays particular attention to the connections between civility, clarity and reconciliation--connections that make civility vital for modern pluralistic societies. I then consider a recent alternative to this conception of civility advanced by Calhoun (2000), arguing that it secures distinctness at the cost of moral significance.

As fosfatidiletanolaminas constituem a segunda classe de fosfolípidos mais abundantes nos organismos. Elas estão presentes nas membranas biológicas e nas lipoproteínas. As alterações estruturais dos fosfolípidos, ocorrem devido ao stress oxidativo e podem manisfestar-se em alterações das suas propriedades e funções. Já são conhecidas algumas condições fisiopatológicas, nas quais os fosfolípidos oxidados estão envolvidos, por exemplo sinalização celular, resposta imunitária, apoptose e doenças relacionadas com o envelhecimento. Por esse motivo, o interesse no estudo dos fosfolípidos oxidados e suas funções tem crescido nos últimos anos. Contudo, a maioria dos estudos realizados, focam a oxidação das fosfatidilcolinas, tendo sido dedicada pouca atenção a outras classes de fosfolipídos, como as fosfatidiletanolaminas. As fosfatidiletanolaminas, podem ainda sofrer outras modificações, devido ao grupo amina livre presente na cabeça polar, como por exemplo a glicação. As fosfatidiletanolaminas glicadas já foram detectadas em condições de hiperglicemia, em pacientes diabéticos, e tem correlação com a hemoglobina glicada. Sabe-se que a glicação de biomoléculas, pode aumentar as modificações oxidativas, que por sua vez, podem ser responsáveis pelo estado inflamatório, existente na diabetes mellitus. Tanto o stress oxidativo, como a inflamação estão relacionados com a diabetes e as suas complicações. A espectrometria de massa tem sido utilizada como uma importante tecnologia na detecção e caracterização de modificações oxidativas de fosfolípidos. Assim, neste trabalho pretendeu-se estudar as modificações oxidativas induzidas em fosfatidiletanolaminas glicadas, e os seus efeitos biológicos nos monócitos e células dendríticas do sangue periférico. Pretendeu-se ainda, estudar as alterações que ocorreram nas espécies de fosfatidiletanolaminas do fígado de ratos diabéticos. Os resultados obtidos permitiram identificar vários produtos de oxidação de fosfatidiletanolaminas glicadas, nomeadamente novos produtos formados pela oxidação da cabeça polar glicada. A oxidação na cabeça polar glicada foi, ainda, confirmada pela realização de experiências com spin traps combinadas com espetrometria de massa. Posteriormente, as fosfatidiletanolaminas oxidadas, glicadas e glicoxidadas demonstraram ter efeitos pró-inflamatórios, confirmados pelo aumento da estimulação monócitos e de células dendríticas, expresso no aumento do número de células produtoras de citocinas em comparação com o estado basal. As diferentes modificações de fosfatidiletanolaminas induziram estímulos distintos nos dois tipos de células. Sendo as fosfatidiletanolaminas glicadas e as glicoxidadas, os compostos que induziram um maior estímulo. Estes resultados sugeriram que as fosfatidiletanolaminas glicadas e as glicoxidadas podem estar associadas com o estado inflamatório que decorre da hiperglicemia crónica. Ainda, a avaliação do perfil lipídico de extratos de fígado de ratos diabéticos demonstrou que a hiperglicemia induz inúmeras alterações das espécies de fosfatidiletanolaminas e das espécies de outras classes de fosfolípidos, em simultâneo com sinais de lesão hepática. Em conclusão, este trabalho demonstra a relação existente entre, a hiperglicémia, o stress oxidativo, a glicação e oxidação de fosfatidiletanolaminas e ainda a inflamação e compliações diabéticas. Portanto a contribuição da lipidómica é importante para compreender os efeitos prejudiciais da hiperglicemia não controlada, e por isso, merece ser mais explorado.
Phosphatidylethanolamines (PE) are the second most abundant phospholipids found in biological membranes and in lipoproteins. In cases of oxidative stress, phospholipids may undergo structural changes that lead to alterations of phospholipids properties and functions. Oxidized phospholipids are known to be involved in various physiopathological conditions, such as signaling of inflammation, immune response, apoptotic events and in age related diseases. Because of such implications, the interest in the study of this subject is increasing. However, most of the studies performed only focused the role phosphatidylcholines and little attention has been paid to other phospholipid families, like phosphatidylethanolamines. Also, phosphatidylethanolamines can undergo other modifications, due to the free amino group present at polar head, such as glycation. Glycation of phosphatidylethanolamines was already reported to occur under hyperglycemia, in diabetic patients and to have correlation with glycated hemoglobin. The glycation of biomolecules increase oxidative modifications, which can be responsible for the low inflammatory state, typical of diabetes mellitus. Both oxidative stress and inflammation are related to diabetes and can be associated with diabetic complications. Mass spectrometry has been used as an important tool in the detection and characterization of these phospholipids’ oxidative modifications. So work aimed to study the oxidative modifications induced in glycated phosphatidylethanolamines, and their biological effects in blood monocytes and dendritic cells. It also aimed to evaluate the alterations of liver PE species under hyperglycemia in diabetic rats. The results allowed identifying various oxidation products of glycated phosphatidylethanolamines, including some new products with oxidation in polar head and in glucose. Oxidation in glycated polar head was confirmed by spin trapping experiments combined with mass spectrometry. Furthermore the oxidized, glycated and glycoxidized phosphatidylethanolamines showed pro-inflammatory effects, confirmed by the increase monocytes and dendritic cell stimulation with increased frequency of cytokine-producing cells, relative to the basal level. The different PE modifications induced different stimulus in the different cells, being the glycated and glycoxidized compounds the ones that induced higher stimulus. The results suggested that glycated and glycoxidized phosphatidylethanolamines could be associated with inflammatory status present in chronic hyperglycemia. In addition, the evaluation of lipid profile of liver extracts from diabetic rats showed that under hyperglycemia occurred changes in PE and other phospholipid species accompanied by signs of hepatic damage. In conclusion, this work reenforces the network between hyperglycemia, oxidative stress, PE glycation and oxidation, inflammation and diabetic complications. Thus the contribution of lipidomic to understand deleterious effects of uncontrolled hyperglycemia is a promising field of research that deserves to be further explored.

Some governments chose to ban cryptocurrency completely, whereas others opted for regulation.

We propose a prototype model of market dynamics in which all functional relationships are linear. We take into account three borders, defined by linear functions, which are intrinsic to the economic reasoning: non-negativity of prices; downward rigidity of capacity (depreciation) and a capacity constraint for the production decision. Given the linear specification, the borders are the only source for the emerging of cyclical and more complex dynamics. In particular, we discuss centre bifurcations, border collision bifurcations and degenerate flip bifurcations - dynamic phenomena the occurrence of which are intimately related to the existence of borders.

Approximate computing has been an emerging programming and system design paradigm that has been proposed as a way to overcome the power-wall problem that hinders the scaling of the next generation of both high-end and mobile computing systems. Towards this end, a lot of researchers have been studying the effects of approximation to applications and those hardware modifications that allow increased power benefits for reduced reliability. In this work, we focus on runtime system modifications and task-based programming models that enable software-controlled, user-driven approximate computing. We employ a systematic methodology that allows us to evaluate the potential energy and performance benefits of approximate computing using as building blocks unreliable hardware components. We present a set of extensions to OpenMP 4.0 that enable the programmer to define computations suitable for approximation. We introduce task-significance, a novel concept that describes the contribution of a task to the quality of the result. We use significance as a channel of communication from domain specific knowledge about applications towards the runtime-system, where we can optimise approximate execution depending on user constraints. Finally, we show extensions to the Linux kernel that enable it to operate seamlessly on top of unreliable memory and provide a user-space interface for memory allocation from the unreliable portion of the physical memory. Having this framework in place allowed us to identify what we call the refresh-by-access property of applications that use dynamic random-access memory (DRAM). We use this property to implement techniques for task-based applications that minimise the probability of errors when using unreliable memory enabling increased quality and power efficiency when using unreliable DRAM.

Type II diabetes is one of the leading cause of morbidity in the U.S. and other parts of the world. Insulin resistance which precedes Type II diabetes is a complex state of the body where the body fails to respond to insulin. Its complexity lies in its multifactorial origin that is to say various environmental and polygenic components come into play. Here we try to dissect one of these components - `Alox8' in transgenic mice and try to see if it affects blood glucose homeostasis. Comparison of glucose tolerance and insulin sensitivity among sixteen mice comprising of six wild type, five heterozygous and five knockout mice with respect to Alox8 gene showed that wild type mice had relatively more glucose tolerance than knockout mice and this corresponded with relatively more insulin sensitiveness of wild type mice with respect to the knock out. However, these findings were not significant statistically at p=0.05. In search of any relevant biological significance, periodic acid schiff staining of the liver sections from these mice in three independent repeated experiments revealed that the knockout phenotype led to accumulation of glycogen deposits as compared to the wild type mice, an indication of insulin resistance. Taken together, our data suggests that these findings when extrapolated to human which carries ALOX15B instead of mice orthologue Alox8, could lead to a benefit of administration of lower doses of insulin in the wild type phenotype as compared to its polymorphic alleles carrying individuals.

By examining stories from LGBTQ+ individuals of places that matter most to them in Philadelphia, this thesis argues that historic preservationists can expand their definition of significance to include personal testimony and broaden their practices to better engage the communities whose histories they seek to preserve.

För att undersöka vad medveten närvaro kan betyda i dialektisk beteendeterapi (DBT) meningskoncentrerades nio intervjuer. Medveten närvaro hade en positiv betydelse för samtliga deltagare. Det centrala för deltagarna var färdigheterna observera och en sak i taget. Färdigheterna hjälpte patienterna att stanna upp i ett negativt händelseförlopp. Behandlarna menade att medveten närvaro hjälpte vid stressrelaterade situationer. Samtliga deltagare underströk betydelsen av färdighetsträningen i grupp och praktiska övningar. Det fanns ett tydligt behov av att förenkla teorin kring medveten närvaro i DBT samt att medveten närvaro behövde få ett större utrymme. Om medveten närvaro kan hjälpa patienter i DBT att stanna upp i ett destruktivt händelseförlopp så är det en verkan av stor betydelse för patienterna såväl som för anhöriga och sjukvården.

During photosynthesis sunlight is absorbed by photosynthetic pigments and converted into organic compounds, such as carbohydrates. Photosynthesis needs to be highly regulated, since both too much and too little light are harmful to plant. If too little light is absorbed, a plant cannot store enough energy, which will have effects on growth and fitness of the plant. With too much light absorbed, a dangerous side reaction of photosynthesis, the production of reactive oxygen species can happen. These reactive oxygen species can damage the proteins in the chloroplast and the lipids of the chloroplast.

To avoid the production of reactive oxygen species, plants have evolved many mechanisms, which act on different time-scales and different levels of organization. As a first measure, when the absorbed light is exceeding the capacity for its utilization, is to switch the light-harvesting antenna from efficient light harvesting to energy dissipation. This process is called feedback de-excitation (FDE). The protein PsbS is essential for this process as well as a functioning xanthophylls cycle with the enzyme violaxanthin de-epoxidase (VDE).

I have investigated the effects of plants with changes in their ability to dissipate excess excitation energy in the model plants species Arabidopsis thaliana. Three genotypes with either increased or decreased capacity for FDE were used during my experiments. The first genotype over-expresses the PsbS gene, having approximately two-fold increased amounts of PsbS and FDE. The second is a PsbS deletion mutant with no PsbS protein and no FDE. The third genotype cannot perform the conversion of violaxanthin to zeaxanthin, because the enzyme VDE is missing. This mutant has some FDE left.

Arabidopsis thaliana is an annual plant, which flowers only once in its lifetime. Therefore, when counting the seeds produced an estimation of fitness can be made from the amount of seeds produced. This was done during my experiments and shown that FDE is a trait and that plants with increased FDE have a higher fitness and vice versa.

This was also the case for a collection of plants lacking a single protein from the light harvesting antenna. All of these genotypes had a fitness reduction, proving that their function is not redundant.

In an attempt to explain why the fitness is reduced in plants with altered FDE, photosynthetic measurements, as well as a determination of the transcriptome and the metabolome was performed. Plants lacking FDE had higher levels of photoinhibition, leading both to lower rates of photosynthesis and to higher repair cost. This could in part explain the reduction in fitness. These plants also had major changes in their transcriptome and their metabolome. Primary metabolism was most effected, for example carbohydrate and amino acid metabolism. But there were also changes in secondary metabolism such as an up regulation of the biosynthesis of anthocyanins.

This is a dissertation on how certain cognitive limitations inform a theory of knowledge. Explanations in terms of the pure indexical "I" indicate a class of cognitive limitations. "I" cannot be completely eliminated from any successful explanation of how the world is intelligible to me and only I can refer to myself with the indexical "I." This raises the possibility that there are thoughts that I can think that cannot be thought by anyone else. Given what an epistemological theory must say about the definition, structure, and instances of knowledge and epistemic merit in general, such limits to cognitive access must arise both in its explanations of ordinary cases and its specialized theoretical concepts. The main contention of this dissertation is that it must be possible for an epistemological theory to plausibly account for these limitations.

The purpose of this thesis is to give an account of the significance of time in Kant&rsquo
s Critique of Pure Reason by discussing its role in the unification of sensibility and understanding. I primarily investigate the role that time plays in the constitution of objective knowledge. I discuss that since time is the necessary condition for objects to be given to our sensibility, without it any representation would be without a temporal order and perhaps would not make any sense at all. Kant claims that it is imagination that enables the connection between sensibility and understanding possible. After investigating the relation between imagination and time, I argue for the following thesis: what lies at the very heart of the possibility of the connection between sensibility and understanding must be time itself. Finally, I focus on the kinship between time and the self. I argue that when it comes to knowledge of the phenomenal world, time together with transcendental self stands as the condition of all human knowledge.

Morphological changes including fibrosis, vascular degeneration and loss of mesothelium occur to the peritoneal membrane during peritoneal dialysis. The loss of mesothelium is thought to effect peritoneal homeostasis and host defence, as these cells play a pivotal role in these organs response to inflammation. Cancer antigen 125 (CA125), a mucilaginous high molecular weight glycoprotein, has been used as a marker of peritoneal mesothelial cells mass/turnover and membrane functional integrity in peritoneal dialysis patients. The use of new more biocompatible dialysis solutions is associated with an increase in CA125 effluent concentration, however it's precise molecular nature, function and regulation is poorly defined. This study investigates the transcriptional mechanisms regulating CA125 expression in human peritoneal mesothelial cells as well as CA125's regulation in response to inflammation. Additionally, CA125's function in the process of mesothelial cell repair is examined using a novel mesothelial wound healing system. These investigations have: Successfully reconstructed the genomic structure of CA125 and characterised its proximal promoter region. Demonstrated regulation of CA125 cell surface expression and shedding in response to IL-1 p and IL-6 trans-signalling. Shown the retardation of human peritoneal mesothelial cell CA125 shedding in response to high levels of glucose degradation products present within peritoneal dialysis fluids. Shown the improved wound healing of mesothelial cells in response to elevated CA125 levels. These investigations have clarified existing discrepancies regarding CA125 regulation during inflammation by demonstrating its altered expression at the cell surface, in response to inflammatory mediators. Additionally, this work has linked clinical Observations regarding CA125 effluent levels during peritoneal dialysis to a potential role of CA125 in wound healing of the mesothelium.

This thesis aimed to provide improved understanding of the spatial and temporal variability of stream temperature, the influence of riparian woodlands on thermal regimes and examples of the ecological consequences of different thermal regimes.  First, the spatial and temporal variability of temperature within a large montane river basin at nested spatial scales was established.  It was from this analysis that (i) variation that can occur at all spatial scales was observed, (ii) different physical factors are controlling these variations at all scales, and (iii) greatest differences occur between different tributaries.  The extent of riparian forestry proved to be in important factor in determining river temperatures.  The results suggest that broadleaved tree species within the riparian zone are more able to reduced incoming solar radiation and consequently control stream temperature.  This highlights the potential of riparian plantings in mitigation efforts against high temperatures under future climate scenarios.  To complement these empirical assessments, the ecological significance of observed thermal variability was assessed in a laboratory study of the growth of juvenile Atlantic salmon.  The effects of the temperature regime on growth were in the range of about 3% reduction in final length in the variable regime compared to the constant one.  Relations between invertebrate growth, adult emergence patterns and thermal regime differences related to riparian cover were assessed in relation to the mayfly Baetis rhodani.  Statistically significant differences in mean size and size distribution of Baetis were found in the Girnock, between all sites separated by no more than 5 km.

Abstract (sommario):
This thesis is, in part, a defence of a broad-based approach to epistemology. We should be wary of taking too narrow a focus and thus neglecting important aspects of knowledge. If we are too focused on one methodology then we are likely to miss insights that can come about from a different perspective. With this in mind, I investigate two particular methodologies in detail: Kornblith’s naturalism and Craig’s ‘genealogical’ approach. Kornblith emphasises the importance of looking at knowledge in the context of the natural world, thus stressing the continuity between animal and human knowledge. Craig, on the other hand, focuses on a distinctly human aspect of knowledge: the importance of enquiry and the sharing of information. As such, the two theories of knowledge that are developed have different emphases. I argue that by bringing them together we can better understand what knowledge is. This leads us to the other main contribution of this thesis, which is a defence of the role of reflection in epistemology. This has often been neglected in contemporary epistemology, primarily because of the effectiveness of externalist theories of knowledge. The focus on externalism has lead to reflection being sidelined. I do not argue that reflection is necessary for knowledge, but rather want to bring back attention to the important role that it plays in human life. Reflectively accessible justification is necessary for our knowledge claims and therefore plays a vital role in enquiry. If we add reflectively accessible justification to knowledge then it is both more stable and more valuable. Even if it is not necessary for knowledge, reflection should not be neglected.

Plumage polymorphism is displayed in a number of diverse species and therefore, is presumed to have evolved independently on multiple occasions. To date, no study has established whether any of the hypotheses proposed to explain its evolution in single species are relevant across all avian taxa. Using comparative analyses which included all species displaying a plumage polymorphism involving melanin pigmentation, I found no support for either sexual or apostatic selection being responsible for the evolution of plumage polymorphism. Weak support for a role for disruptive selection was indicated in certain species but not when all polymorphic species were considered together. Contrary to previous studies, this indicates that plumage polymorphism may not confer any selective advantage but may in fact be selectively neutral or perhaps simply a product of historical biogeographic processes. Many polymorphic species have clinal morph-ratio distributions. In the polymorphic New Zealand fantail, Rhipidura fuliginosa, I established that the black morph was more common in central parts of the South Island of New Zealand than in the South. This was not the pattern expected based on previous studies and no environmental factor was found to correlate with the distribution. Polymorphism may not confer a selective advantage to a species as a whole, but the fact that two or more often very differently coloured morphs can persist within a population suggests that balancing selection may be in operation. I demonstrated that black morph fantails suffered reduced feather damage over the course of the year and that black and pied fantails differed slightly in foraging behaviour. The benefits of black and white plumage whilst foraging were further elucidated through plumage manipulation experiments. Thus, a trade-off between feather wear and foraging between the two morphs of the fantail, produced due to the different selective advantages provided by their plumage colouration, seems likely to be part of the balancing selection mechanism responsible for maintaining the plumage polymorphism in this species. Plumage polymorphism may be better understood if the mechanisms controlling plumage evolution in general were unravelled. Therefore, I reconstructed plumage characters of the genus Rhipidura onto a molecular phylogeny that I created based on the cytochrome b gene. Species that were divergent in plumage were also found to be more genetically divergent. This suggested that, unlike in other genera, the plumage characters of the Rhipidura spp. may be a good estimator of phylogenentic relationships.

Background/Aim: Little is known about the clinical significance of phantosmia. The literature on phantosmia indicates that this symptom has a wide range of differential diagnoses. However, most cases of phantosmia remain of unknown origin. Our goal was to follow up patients with idiopathic phantosmia, with special regard to improvement rates and possible severe health conditions preceded by phantosmia of unknown origin. Methods: Forty-four patients with idiopathic phantosmia which had consulted our Ear-Nose-Throat Smell and Taste Clinic over the last 10 years were contacted by telephone and underwent a structured medical interview. Results: None of the patients had eveloped any severe health condition or Parkinson’s disease. More than 5 years after the occurrence of hantosmia, more than 50% of the patients experienced disappearance (31.8%) or improvement (25%). In the remaining cases, phantosmia did not change (38.7%) or became worse ( ! 5%). Conclusion: The main findings of the present study were that idiopathic phantosmia improves or disappears in almost two thirds of the patients after more than 5 years, and that idiopathic phantosmia seems to be more likely a harmless symptom rather than a reliable predictor of early Parkinson’s disease or other severe diseases

An investigation of architecturally significant buildings constructed within the last fifty years, in the Midwest and the application of preservation theories to these significant buildings, to aid in evaluation strategies and preservation implementation.Recognition of recent past construction as candidates for architecturally significant cultural resources and the application of preservation theories to these buildings make for good design and preservation practice. Part of our responsibility, as preservation professionals is to promote the preservation of significant structures, regardless of the date of construction. Since there is no established process to aid in preserving recently constructed buildings of architectural significance, it is necessary to develop strategies to assist in the evaluation process.
