Objectives: To assess the sensitivity of the various discrimination indicesas screening test in beta-thalassemia trait patients in our population. Study design: Crosssectional descriptive study. Period: 1-Sep 2012 to 31-Jan 2013 (5 months). Settings: PathologyDepartment, District Head Quarters (DHQ) Hospital, Rawalpindi. Materials and Methods: Atotal of 150 diagnosed cases of beta thalassemia trait were included in the study. Study wasdone from 1-Sep 2012 to 31-Jan 2013 in DHQ hospital, Rawalpindi. Four discrimination indicesi.e. Mentzer, Shine & Lal, Srivastava, and Red cell Distribution Width Index were calculated forall the patients. The number of correctly identified cases were determined and sensitivity ofeach discrimination index was calculated accordingly. Results: The sensitivity in diagnosingbeta thalassemia trait patients was highest for Shine and Lal index (95%), followed by Mentzerindex (52% sensitivity), and then Srivasava index (46% sensitivity). Red cell distribution widthindex had poor sensitivity of 2% in our study. None of the discrimination indices showed 100%sensitivity. Conclusion: We concluded that Shine & Lal and Mentzer index are the most sensitiveindices. They can be used for cheap and quick screening of beta-thalassemia trait patients inlaboratories where advanced investigation tools are not available. Patients who give positiveresult for beta thalassemia trait with these indices may then be referred for further workup toconfirm the diagnosis.

Abstract (sommario):

Since 2017, a field experiment has been conducted in a dry area with 11 Czech hop varieties. The highest yields were achieved by the Uran variety (2.83 t/ha) and Saaz Shine variety (2.76 t/ha), which also exhibited the lowest yield variability, at 11.17% (Saaz Shine) and 12.53% (Uran). The lowest hop yield was recorded by the Harmonie variety (1.17 t/ha), which also showed high yield variability (30.92%). The Bohemie variety had the highest yield variability of hops at 35.26%. While the Gaia (14.66% w/w) and Uran (13.53% w/w) varieties had the highest alpha acid content. Conversely, the Mimosa variety showed the lowest alpha acid content (1.56% w/w) and also the lowest alpha/beta acid ratio (0.29). The Rubín variety reached the highest alpha/beta acid ratio (3.00). The Uran variety exhibited the lowest variability in alpha (6.16%) and beta (6.28%) acid content. In contrast, the Harmonie variety had the highest variability in alpha (59.02%) and beta (38.18%) acid content. The Uran and Saaz Shine varieties are the most suitable for the dry area. Additionally, the Gaia and Saaz Brilliant varieties showed good performance parameters. The hop varieties Vital, Gaia, Saaz Brilliant, and Saaz Shine had the most intense hop aroma, while the Boomerang and Uran varieties had a high intensity of spicy aroma.

Abstract (sommario):

Four new aroma hop varieties – Saaz Brilliant, Saaz Comfort, Saaz Shine and Mimosa – were registered in the Czech Republic in 2019. All of the new hop varieties have significantly higher yields than the traditional Saaz aroma variety. Saaz Comfort has the significantly highest content of alpha acids (5.59%) whereas Mimosa has the significantly lowest content of alpha acids (1.90%). The Saaz Comfort and Saaz Brilliant varieties show a variability of alpha acid content below 20%. The other hop varieties, Saaz Shine and Mimosa, as well as Saaz, have a variability of alpha acid content above 25%. Mimosa has the significantly highest content of beta acids (6.07%). Mimosa shows the highest average cohumulone content (29.29% rel.) whereas Saaz Comfort has the lowest cohumulone content (18.04% rel.). Saaz Comfort and Saaz Shine have the highest average contents of hop essential oils (0.84% w. and 0.75% w., respectively). The Saaz, Saaz Shine and Saaz Brilliant varieties show the significantly highest farnesene contents (13.47% rel., 12.50% rel. and 12.38% rel., respectively), which are higher than those of Saaz Comfort and Mimosa.

Beta thalassemia trait are more likely to have anemia which leading to intrauterine hypoxia and adverse uterine conditions. Adverse intrauterine environment has been shown to have a significant impact on the risk of low birth weight. Therefore, this study aims to investigate association between Shine and Lal index β-thalassemia trait screening results with anaemia and lowbirth weight. A retrospective study design was conducted based on medical records of mothers and their infants born in 2022 at a regional hospital, in East Nusa Tenggara. The inclusion criteria were being live birth, singleton births and gestational age >28 weeks. Mothers didn’t have complete records were excluded from study. Analysis uses chi-square test to evaluate the relationship between dependent and independent variables, This study identified 738 eligible samples. The incidence of suspected β-thalassemia trait according Shine and Lal Index was about 42.5%. There is a significant relationship between Shine and Lal β-thalassemia trait screening results with maternal anaemia and no relationship between Shine and Lal index β-thalassemia trait screening results and LBW

Background: Iron deficiency anemia (IDA) and beta thalassaemia trait (BTT) are two of the most common causes of microcytic anemia.It is essential to differentiate between the two, so as to avoid unnecessary iron therapy which is contraindicated in beta thalassaemia .We retrospectively evaluated the reliability of various indices for differential diagnosis of microcytosis and 𝛽-TT in the same patient groups .Methods: A total of 200 patients were evaluated. We calculated 6 discrimination indices in all patients with anemia or suspected beta thalassemia. None of the subjects had combined IDA and 𝛽-TT. The patient groups were evaluated according to the Mentzer, Shine and Lal, England and Fraser,Srivastava, Green and King, Ricerca.Results: According to percentage correctly diagnosed criteria, Ricerca and Mentzer index are considered as the two best discrimination indices in differentiation between β thalassemia trait and iron deficiency anaemia .Youden’s index showed the following ranking with respect to the indices’ ability to distinguish between 𝛽-TT and IDA, Ricerca index > Mentzer index > Green and King index > Srivastava index > Shine and Lal index > England and Fraser index.Conclusions: Ricerca and Mentzer index are considered as the two best discrimination indices in differentiation between β thalassemia trait and iron deficiency anaemia.Keywords: Iron deficiency anemia (IDA) and beta thalassaemia trait (𝛽-TT)

Abstract IL-6 is a major regulator of acute phase protein synthesis in the liver. It exerts its action via a plasma membrane receptor consisting of two subunits, a ligand binding 80-kDa glycoprotein and a 130-kDa glycoprotein involved in signal transduction. We genetically generated a soluble form of the 80-kDa subunit of the human IL-6R (shIL-6R) in mouse fibroblasts (NIH/3T3 cells). The shIL-6R added to human hepatoma cells (HepG2) amplified the induction of alpha 1-antichymotrypsin and haptoglobin by IL-6 at the mRNA and protein level. Moreover, a model for a liver permanently exposed to high IL-6 concentrations has been developed; HepG2 cells were stably transfected with human IL-6-cDNA; 10(6) of the transfected cells (HepG2-IL-6) synthesized and secreted 2 micrograms of IL-6 within 24 h. Incubation of these cells with endogenous or exogenous IL-6 did not result in acute-phase protein induction. However, these IL-6-desensitized cells responded to other cytokines such as leukemia inhibitory factor, transforming growth factor beta 1, and IFN-gamma, known to modulate acute phase protein synthesis in the liver. Incubation of HepG2-IL-6 cells with shIL-6R reconstituted their responsiveness to IL-6 in a dose- and time-dependent manner. The possible biologic role that might be played by the shIL-6R in disease is discussed.

Abstract (sommario):

A method of organizing an information cyber- physical system to assist in the mooring process has been considered in the article. It describes ways to organize the mooring of ships to the berth. The problems that occur during this operation have been listed. As the mooring process is labor- intensive and depends on the human factor, a system that will provide up-to-date real-time information to facilitate the mooring operation has been regarded. The structure and stages of development of the proposed solution have been described. The features of the selected elements have been indicated. Obtaining preliminary sensory results from two ports makes it possible to begin the next stage of system development.

The Lanna region, the main part of northern Thailand, is a place of ethnic diversity. In this study, we investigated phak-gard-dong (PGD), or pickled mustard green (Brassica juncea L. Czern.), for its beneficial bacteria content and to analyse the variations in bacterial compositions among the PGD of three different ethnolinguistic groups, the Karen, Lawa, and Shan. DNA was extracted from the PGD pickled brine, and 16S rRNA gene Illumina sequencing was performed. Metagenomic data were analysed and the results demonstrated that the dominant bacterial species were Weissella (54.2%, 65.0%, and 10.0%) and Lactobacillus (17.5%, 5.6%, and 79.1%) in the PGD of the Karen, Lawa, and Shan, respectively. Pediococcus was found only in the PGD of the Karen and Shan. Bacterial communities in PGD of the Lawa were distinctive from the other ethnic groups, both in the alpha and beta diversity, as well as the predicted functions of the bacterial communities. In addition, overall network analysis results were correlated to bacterial proportions in every ethnic PGD. We suggest that all ethnic PGDs have the potential to be a good source of beneficial bacteria, warranting its conservation and further development into health food products.

A correlation between increased beta-1,6 branching of N-linked carbohydrates and the ability of a cell to metastasize or to form a tumor has been observed in several experimental models. Lec9 Chinese hamster ovary (CHO) mutants exhibit a drastic reduction in tumorigenicity in nude mice, and this phenotype directly correlates with their ability to attach an increased proportion of beta-1,6-branched carbohydrates to the G glycoprotein of vesicular stomatitis virus (J. Ripka, S. Shin, and P. Stanley, Mol. Cell. Biol. 6:1268-1275, 1986). In this paper we provide evidence that cellular carbohydrates from Lec9 cells also contain an increased proportion of beta-1,6-branched carbohydrates, although they do not possess significantly increased activity of the beta-1,6 branching enzyme (GlcNAc-transferase V). Biosynthetic labeling experiments show that a substantial degree of underglycosylation occurs in Lec9 cells and that this affects several classes of glycoproteins. Lec9 cells synthesize ca. 40-fold less Glc3Man9GlcNAc2-P-P-lipid and ca. 2-fold less Man5GlcNAc2-P-P-lipid than parental cells do. In addition, Lec9 cells possess ca. fivefold less protein-bound oligosaccharide intermediates, and one major species is resistant to release by endo-beta-N-acetylglucosaminidase H (endo H). Membranes of Lec9 cells exhibit normal mannosylphosphoryldolichol synthase, glucosylphosphoryldolichol synthase, and N-acetylglucosaminylphosphate transferase activities in the presence of exogenous dolichyl phosphate. However, in the absence of exogenous dolichyl phosphate, mannosylphosphoryldolichol synthase and glucosylphosphoryldolichol synthase activities are reduced in membranes of Lec9 cells, indicating that membranes of Lec9 cells are deficient in lipid phosphate. This was confirmed by analysis of lipids labeled by [3H]mevalonate, which showed that Lec9 cells have less lipid phosphate than parental CHO cells. Mechanisms by which a defect in the synthesis of dolichol-oligosaccharides might alter the degree of beta-1,6 branching in N-linked carbohydrates are discussed.

Abstract (sommario):

Carrier screening for early detection of thalassemia in the general population needs a careful approach, especially inÂ areas with limited health services. Various erythrocyte indices have been introduced as options for the first stage; however,Â the low-reliability value of these erythrocyte indices remains the problem. This study aimed to determine the most reliableÂ index for screening beta-thalassemia carriers and distinguish it from iron deficiency anemia. A cross-sectional study wasÂ designed to explore thalassemia carrier status among medical students. Inter-rater reliability value of various indices wasÂ compared to Shine and Lal index. The Cohen's Kappa coefficient was calculated using SPSS v. 25.0. Among 320 respondents,Â 295 subjects were non-anemic and 25 were anemic subjects, whereas 105 subjects had low MCV and/or MCH values.Â Cohen's Kappa value showed moderate reliability results compared to Shine and Lal index for example Mentzer index (0.58),Â Ehsani index (0.57), Srivastava index (0.53), and Bordbar index (0.41), but showed very low-reliability results with Green andÂ King index (0.04). New cut-off indices based on Kumar et al. were also compared, resulting in moderate reliability results.Â Since there was no Hb-electrophoresis test, the sensitivities and specificities of those indices could not be calculated. For thisÂ reason, a complete blood count can only be used for the early stages of screening for beta-thalassemia carriers, whereasÂ Hb-electrophoresis and DNA tests were considered necessary to perform to confirm a diagnosis.

Abstract (sommario):

Ship maneuvering implements sailor skills in maneuvering ships from a departure place to a destination safely, effectively, and efficiently. Ship maneuvers are carried out in waters, but ship maneuvers in icy waters have serious challenges and must be executed carefully. The ice-bound maneuvering procedure must consider the technique used. Besides that, the weather factor can cause obstacles in the movement. Thus, skill in ship maneuvers and accurate analysis are needed to reach a berth position in icy waters. This research aims to analyze whether there is a significant influence between ship maneuvering techniques and weather simultaneously on the ship's berthing position in ice waters. This study uses a quantitative descriptive method. The data sources were obtained through observation, the ship's bell book, and documentation, which were then processed into numbers describing the research variables. These numerical data are used in research to test hypotheses regarding the partial and simultaneous influence of ship maneuvering techniques and weather on ship berthing positions with conditions of icy waters. The study results indicate that the ship's maneuvering techniques and the weather have a significant effect partially or simultaneously on the position of the ship's berth in icy waters.

Abstract (sommario):

In this paper, we introduce a new continuous probability distribution with five parameters called the modified beta Gompertz distribution. It is derived from the modified beta generator proposed by Nadarajah, Teimouri and Shih (2014) and the Gompertz distribution. By investigating its mathematical and practical aspects, we prove that it is quite flexible and can be used effectively in modeling a wide variety of real phenomena. Among others, we provide useful expansions of crucial functions, quantile function, moments, incomplete moments, moment generating function, entropies and order statistics. We explore the estimation of the model parameters by the obtained maximum likelihood method. We also present a simulation study testing the validity of maximum likelihood estimators. Finally, we illustrate the flexibility of the distribution by the consideration of two real datasets.

Abstract Measurements of the spatial distribution of radon isotopes were carried out from April 2021 to August 2021 in the foothills of the Trans-Ili Alatau of the Tien Shan in the Almaty region at various heights above sea level: from 500 to 2500 meters. They were carried out using electronic radiometric equipment: beta-dosimeter “RKS-01B-SOLO”; gamma dosimeter “RKS-01G-SOLO”; radiometer of radon and its daughter decay products “RAMON- 02” in the field. As a result, preliminary assessment schemes were built for route measurements of the 222Rn radon isotope, beta and gamma radiation fields from natural daughter products of decay of radon isotopes and radionuclides located in the surface atmospheric layer.

Background: Iron deficiency anemia (IDA) and beta thalassemia trait (BTT) are the two most common and important causes of microcytic hypochromic anemia in India. It is very difficult to differentiate between the two. Many different types of techniques have been proposed for the same. While some are invasive like bone marrow examination others are not available at all centers, like electrophoresis. Hence different indices come into play. Aims and Objective: This study was undertaken to compare the efficacy of Shine and Lal index and Mentzer index in differentiating between IDA and BTT. Materials and Methods: A total of 407 anemia cases were studied over a period of 18 months and their blood samples were subject to different hematological and biochemical assays to diagnose the type of anemia. Results: Based on these tests 92.1% cases were found to be of IDA whereas 3.7% cases were found to be of BTT. Then both the indices were applied in the above mentioned cases. Conclusion: While Shine and Lal index was found to have better sensitivity, Mentzer index was found to have better specificity.

In this paper, the authors discuss a fully automatic berthing system and actual tests using a small training ship, Shioji Maru. The ship is about 440 gross tonnage and furnished with a controllable pitch propeller, a bow thruster and a stern thruster to which a computer can easily be linked. The system developed is composed of an electro-optical position measuring system and the computer software necessary for manoeuvring the ship.The electro-optical position measuring system can measure a distance between the ship and a target on a berth within 4 cm accuracy and cover an area out to about 100 m range. The manoeuvring is divided into two stages. The first one is from a start line to a transition point off the final berth. At this point, the ship must be stopped in an alongside attitude to the berth. In the second stage, the ship is pushed by the thrusters in an athwartships direction towards the berth and the final speed at the berth must be limited to within 0.5 ms. The computer software generates an appropriate strategy following an approach line which has been determined in advance.According to the actual tests under slightly rough sea conditions, the system could manoeuvre the ship along the approach line by a direct path and maintain her final speed within the predetermined limit.

Objective: To assist in the differential diagnosis of beta thalassemia trait and iron deficiency anaemia, many alternative red blood cell index-based formulae were examined. Methods: The Rawalpindi PEMH performed this study from June 2021 to March 2022. For individuals with beta- thalassemia trait and iron deficient anaemia, age and gender were not considered. More than five millilitres of blood were drawn from each patient in order to determine the haemoglobin content, the number of red blood cells, how they were distributed, and the average cell volume. Five alternative formulas may be used to differentiate these two circumstances. Shine and Lal index, Mentzer index, Srivastava index and Green & King index are among the most well- known ones. Youden's index was included in the calculation of sensitivity, specificity, and positive and negative predictive values (PPVs). Results: The iron deficiency anaemia rate was 70%, and the beta thalassemia phenotype was 30% among the 1500 participants. This indicator has a sensitivity of 100% and specificity of 93.3% when it comes to discriminating between beta-thalassemia trait and iron deficiency anaemia. Conclusion: The red cell distribution width index may be used to differentiate between beta thalassemia trait and iron deficiency anaemia. If a haemoglobin electrophoresis is not available, a diagnosis of beta thalassemia may still be made. Keywords: Srivastava index, beta-thalassemia, Anemia, Cell redness, Hb electrophoresis

The efficiency of loading and unloading operations depends very much on the berth allocation. However, due to certain events such as weather factors and ship failures, ships are often delayed in arriving at the port, which severely impacts the berth allocation plan. To effectively deal with the effects of ship delay on the berth allocation plan, this paper studies the berth allocation problem of mainline and feeder vessels, considering the impact of uncertainty.

We describe here the properties of a mutant of Chinese hamster ovary cells that expresses a conditional-lethal mutation affecting dense lysosomes. This mutant, termed V.24.1, is a member of the End4 complementation group of temperature-sensitive mutants selected for resistance to protein toxins (Colbaugh, P. A., C.-Y. Kao, S.-P. Shia, M. Stookey, and R. K. Draper. 1988. Somatic Cell Mol. Genet. 14:499-507). Vesicles present in postnuclear supernatants prepared from V.24.1 cells harvested at the restrictive temperature had a 50% reduction in acidification activity, assessed by the ATP-stimulated accumulation of the dye acridine orange in acidic vesicles. To investigate whether specific populations of vesicles were impaired in acidification, we measured acidification activity in three subcellular fractions prepared from Percoll gradients: one containing endosomal and Golgi markers, one containing buoyant lysosomes, and the third containing dense lysosomes. Activity in dense lysosomes was reduced by 90%, activity in the buoyant lysosome fraction was unaffected, and activity in the endosome-Golgi fraction was mildly reduced. The activity of three lysosomal enzymes--beta-hexosaminidase, beta-galactosidase, and beta-glucocerebrosidase--was also reduced in dense lysosomes but nearly normal in the buoyant lysosome fraction. However, beta-hexosaminidase and beta-glucocerebrosidase activity was increased two- to threefold in the endosome-Golgi fraction. We conclude that the lesion selectively impairs dense lysosomes but has little effect on properties of buoyant lysosomes.

Erythrocyte indices have been used as the first indicator in screening for beta-thalassemia minor. However, iron deficiency in hemoglobinopathies can reduce the sensitivity of screening, and the spectrum of mutations of different hemoglobinopathies between populations will result in discontinuation of the results of different erythrocyte indices. The research objective in this study was to determine the description of the erythrocyte indices used to screen for beta-thalassemia trait in Ciamis. The research method used in this study was a quantitative descriptive, using a cross-sectional study, data collection had been done during March-May 2021. The subjects of this study were 104 parents of a beta-thalassemia trait who have children with a beta-thalassemia major in Ciamis. The examination parameters used were complete blood count, with processing twenty-six erythrocyte formula index values. Ninety-two respondents were included in the inclusion criteria. The results of the erythrocyte indices in beta-thalassemia trait are England and Fraser, RBC, Mentzer, Srivastava, Shine and Lal, Ricerca, Green and King, Das Gupta, Jayabose, Telmissani-MCHD, Telmissani-MDH, Huber-Herklotz, Kerman I, Kerman II, Sirdah, Ehsani, Keikhaei, Nishad, Wongprachum, Sehgal, Pornprasert, Sirachainan, Bordbar, Matos and Carvalho and CRUISE has cut-off value in that range according to previous studies. Only the Bessman erythrocyte indices has a value of 17, exceeding the cut-off. Twenty-five erythrocyte indices with cut-off value according to previous studies can be used for beta-thalassemia screening in Ciamis, except for the Bessman erythrocyte indices.

Our definition relies on a canonical measure $\mu _{\rm can}$ on the $F$ -analytic manifold associated to $\operatorname {M}_{\beta,\chi }$ and the $p\mathrm {BPS}$ invariants are integrals of natural ${\mathbb {G}}_m$ gerbes with respect to $\mu _{\rm can}$ . A similar construction can be done for meromorphic and usual Higgs bundles on a curve. Our main theorem is a $\chi$ -independence result for these $p\mathrm {BPS}$ invariants. For one-dimensional sheaves on del Pezzo surfaces and meromorphic Higgs bundles, we obtain as a corollary the agreement of $p\mathrm {BPS}$ with usual $\mathrm {BPS}$ invariants through a result of Maulik and Shen [Cohomological $\chi$ -independence for moduli of one-dimensional sheaves and moduli of Higgs bundles, Geom. “Double-Line Ship Mooring” (DLSM) mode has been applied as an initiative operation mode for solving berth allocation problems (BAP) in certain giant container terminals in China. In this study, a continuous berth scheduling problem with the DLSM model is illustrated and solved with exact and heuristic methods with an objective to minimize the total operation cost, including both the additional transportation cost for vessels not located at their minimum-cost berthing position and the penalties for vessels not being able to leave as planned. First of all, this problem is formulated as a mixed-integer programming model and solved by the CPLEX solver for small-size instances. Afterwards, a particle swarm optimization (PSO) algorithm is developed to obtain good quality solutions within reasonable execution time for large-scale problems. Experimental results show that DLSM mode can not only greatly reduce the total operation cost but also significantly improve the efficiency of berth scheduling in comparison with the widely used single-line ship mooring (SLSM) mode. The comparison made between the results obtained by the proposed PSO algorithm and that obtained by the CPLEX solver for both small-size and large-scale instances are also quite encouraging. To sum up, this study can not only validate the effectiveness of DLSM mode for heavy-loaded ports but also provide a powerful decision support tool for the port operators to make good quality berth schedules with the DLSM mode.

Addressing the automatic berthing task for vessels, this study introduces the Flow Matching Double Section Bezier Berth Method (FM-DSB) for handling downstream and upstream berthing instructions. By considering the orientation relationship between the direction of water flow and the berth, combined with berthing modes, the algorithm determines the vessel’s entry angle into the berth and plans the berthing path using double-section Bezier curves. Effective control of vessel speed post-path determination is essential. Therefore, based on the response of vessels to propeller inputs, this study introduces the Berthing Path Velocity Matching Method (BPVM). The BPVM ensures speed matching along the berthing path through analysis of vessel acceleration and deceleration capabilities. Subsequently, simulation experiments are conducted to validate the planning algorithm for both long-distance and short-distance berthing. Furthermore, the feasibility and effectiveness of the berthing path are verified using a dual-loop path tracker based on the planned results. Experimental outcomes illustrate the adaptability of the proposed algorithm in planning berthing paths that align with vessel motion characteristics, effectively guiding vessels into berths through the designed dual-loop control system.

Container berths, which are an essential component of the marine wharf, are exposed to loads from various sources, such as self-weight, operation loads and the environment loads, but the ship load is considered to have the greatest impact on the structure. The container berth in Um Qaser Port was chosen as a model for the study. This paper was based on numerical simulation which was performed by using ABAQUS program to investigate the response of container berth structure under the impact dynamic load.. Studies have dealt with choice eight different diameters of the piles, starting from 800mm to 1500mm. The study included three different cases of pile diameter. The first case is the effect of changing the spacing between the piles. The second case is the effect of the pile depth embedded in the soil. The third case is the change in thickness of pile diameter. The aim of this study is to know the behavior and performance of the pavement structure by changing some parameters, including the diameter of the pile, which is considered the basic and important part in the formation of the structure.

Overexpression of Trop2 is associated with aggressive disease progression and poor prognosis. Recently, Trop2 targeting antibody-drug conjugate such as sacituzumab-govitecan (Trodelvy®) was approved for the treatment of triple-negative breast cancer and urothelial cancer (UC) patients. Interferon-beta (IFN-beta), a pleiotropic cytokine, can induce direct/in-direct tumor killing and activate anti-tumor immune response in tumor microenvironments. However, it is challenging to develop IFN-beta as an anti-cancer agent due to its low biophysical properties, short half-life, and the systemic toxicity. Previously, we developed IFN-beta mutein (ABN102), which showed improved stability and biophysical properties compared to wild-type IFN-beta. We hypothesis to reduce any toxicity of ABN102 by fusing with tumor targeting antibody. In this study, we generated antibody cytokine fusion protein (ABN202), anti Trop2-interferon-beta mutein, and evaluated the efficacy of ABN202 against Trop2-positive UC cell line models. Method & Result: We aimed to determine the direct anti-cancer efficacy of ABN202 using Trop2-positive UC cell lines and the indirect immune cell mediated effect of ABN202 using Trop2-positive UC cell lines co-cultured with human peripheral blood mononuclear cell (PBMC). The in vivo efficacy of ABN202 was further confirmed in Trop2 positive tumor mouse model. Our results revealed that ABN202 directly inhibited proliferation and activates the apoptosis-related signaling mechanisms via Type 1 IFN signaling, showing better efficacy than either anti Trop2 antibody (sacituzumab) or ABN102 alone. ABN202 induces the activation of PBMC subsets and enhanced immune cell-mediated cancer cell killing when UC cell lines were co-cultured with PBMC. The anti-cancer efficacy of ABN202 were confirmed in Trop2-positive tumor mouse model. Conclusion: We evaluated the direct and indirect anti-cancer efficacy of ABN202 (anti Trop2-interferon-beta mutein) in Trop2-positive urothelial cancer. Our results support that ABN202 is a potent drug candidate in Trop2-positive urothelial cancer by directly inhibiting tumor growth and activating anti-cancer immune responses through type 1 IFN signaling. Abstract (sommario):

The flexible operating and changing of the complex one-of-a-kind shipbuilding environment has to be based on adequate concepts and instruments to handle the related controlling, planning and implementation tasks. Product modeling defines the physical and application driven product-related information. It is of basic importance to support this environment, especially the concurrent engineering functions, during the whole product life cycle. Process modeling supports the implementation and operation of complex CIM (Computer Integrated Manufacturing) oriented processes. In this paper some modeling applications within European shipbuilding research and development projects will be highlighted from the viewpoint of an integrated product and process modeling approach. The following projects referenced: NEUTRABAS (Neutral Database for Complex Multifunctional Systems) explores a broad application field of product information for ship steel structure and outfitting systems; ROCOCO (Real Time Monitoring and Control of Construction Site Manufacturing) develops a CIM application and demonstrates this application within the pipe outfitting environment of a ship berth construction site; and MARIN-ABC (Marine Industry Applications of Broadband Communication) demonstrates new applications and services in the maritime transportation business based on future mobile satellite networks.
