The long-term success of osseointegrated trans-tissular medical devices, such as dental implants and intraosseous transcutaneous amputation prosthesis, is usually compromised by a poor fixation between material and surrounding soft tissue, leading to a breach open to bacterial colonization and infection. These complications have been associated with increased morbidity and high medical costs, making this condition a major public health issue. Thin films formed by the Layer-by-Layer (LbL) technique are an attractive surface functionalization approach, since they may act as reservoir of bioactive molecules that guide cellular behavior on the material surface. In this regard, the aim of this thesis has consisted in the development of bifunctional biomimetic LbL coatings, able to enhance soft tissue integration while providing bactericidal activity to trans-tissular implants.LbL films have been built using the extra-cellular matrix glycoprotein fibronectin (Fn), due to its pro-adhesive properties, and a low molecular weight α-poly-L-lysine (30 residues, PLL30), for its intrinsic antimicrobial properties. The Fn-enriched PLL30-Fn films promoted human gingival fibroblasts (HGF) adhesion and proliferation but presented weak antibiofilm effect against Staphylococcus aureus (S. aureus) and Aggregatibacter actinomycetemcomitans (A. actinomycetemcomitans), and a moderate action towards Streptococcus mutans (S. mutans) (2,8 log10 reduction). The co-culture of both procaryotic and eucaryotic cells in a pre-implantation scenario proved that the coatings are able to inhibit bacterial adhesion and protect and boost HGF proliferation. In a post-implantation infection model, the films did not avoid bacterial colonization, but the capacity of S. aureus to invade/infect? HGF significantly decrease . LbL films were also built on two Ti alloys, proving that the material and topographic characteristics do not compromise thin film assembly.The antibacterial properties of LbL films were not improved when PLL30 was combined with poly-L-glutamic acid (PGA), a pure biodegradable polyelectrolyte. PLL was then emulsified with carvacrol, an antimicrobial essential oil, and the emulsion in solution exerted strong efficiency against S. aureus biofilms, particularly when a high molecular weight PLL (70-150 KDa) was utilized (5,7 log10). However, this effect was lost when the particles were incorporated into PGA-based films. Finally, the adsorption of a synthetic antiseptic polyhexamethylene biguanide (PHMB) within (PLL30-PGA)(PHMB-PGA) films, at a concentration of 80 mg/ml, inhibited the growth of S. aureus (-4,2 log10), and had moderate action for A. actinomycetemcomitans (-2,6 log10) and S. mutans (1,3 log10). The cytocompatibility of these coatings towards HGF should be now further enhanced

Degenerative shoulder pathologies are currently treated by total shoulder arthroplasty (joint replacement) or hemiarthroplasty. Despite the success of this treatment, the complication rate reaches 22 % due to the complex nature of joint mechanics and limited bone stock. In this context, for young patients with degenerative glenohumeral joints, a new generation of interposition implant has been developed. The implant, designed with an original spherical shape, is inserted without fixation between the glenoid cartilage and a surgically created humeral bone cavity. Cobalt-chromium (CoCr) is the most used biomaterial in shoulder arthroplasty; however, Pyrocarbon (PyC) is selected for the new implant for its elastic modulus similar to that of cortical bone. Short-term clinical results showed minimal bone and cartilage wear and good bone remodelling in contact with the implant. However, the origin of these results is not yet well understood. In this framework, the previous study on human explants in the laboratory showed that humeral bone remodelling involves the synthesis of a neocartilaginous tissue, which partly explains the favourable clinical results. However, these results are more controversial on the scapula side because of cartilage erosion. Therefore, the effect of PyC and the geometry of the implant on the remodelling of the surrounding tissues need investigation. Thus, this thesis aims to understand the origin of tissue remodelling on the bone and cartilage side. The first step of our strategy is to carry out three parallel studies to dissociate the role of the biology (cellular response), the impact of the material (PyC vs CoCr), and the involvement of mechanical stresses transmission (living tribological triplet). The second step is to validate the results thus obtained by associating the different aspects in an in vitro model based on tissue bioengineering principles. Consequently, our methodology was based on 1) the expertise of retrieved explants; 2) the analysis of murine primary chondrocytes cultures in contact with PyC and CoCr; 3) the testing of in vitro biotribological simulations; and 4) the design and validation of a simulator allowing to combine the biological, physicochemical and tribological results obtained previously. The final results showed better chondrogenic and osteogenic cell activity in the case of PyC compared to CoCr. They were correlated on the one hand with better adsorption of the lubricating molecules, phospholipids, on the surface of PyC and the other hand with an optimal transmission of the mechanical stress due to its properties and geometry. Furthermore, the in vivo control of transmission of mechanical stresses is essential to guarantee good results in the long term. Besides, this thesis opens up fresh perspectives on the control of this transmission through biomaterials and their geometries for cartilage regeneration

In this thesis we study bioactive materials and their properties to create a favourable environment for the osseous regeneration. More precisely, we are interested in the local and early physicochemical reactions, which occur at the interface between the bioactive glass and the biological environment. The characterisation methods used are transmission electron microscopy associated with energy dispersive X-rays spectroscopy (EDXS) and energy loss electron spectroscopy (EELS). We chose the Cliff and Lorimer method to quantify the elements by EDXS. To optimise the absorption corrections for the light elements, we developed an iterative process for absorption corrections using the experimental value of the local relative specimen thickness measured by EELS. Thus we determined precisely the O/Si atomic ratio in the different areas of the sample. To confirm these measurements, we used EELS to analyse the chemical environment of silicon by the study of the ELNES structures recorded on the L2,3 threshold of silicon. These two approaches allow to demonstrate the existence of many peripheral layers at the bioactive glass (A9) / biological fluid (DMEM) interface: a SiO2 rich layer, a fine temporary layer containing Si(OH)4 groups leading to bonds with calcium ions. Finally by the arrival of phosphorus groups, a calcium and phosphorus rich layer is formed. As this layer precipitates in an apatite, the fine layer containing of the Si(OH)4 groups disappears quickly by repolymerisation in SiO2

Ces réseaux tridimensionnels résultent d'interactions hydrophobes interchaînes entre des chaînes thermosensible d'un copolymère aléatoire d'unité oxyde d'éthylène et oxyde de propylène (POEP) greffées sur le squelette polysaccharidiques. Ces interactions sont induites par chauffage.
Afin d'évaluer l'influence des greffons des dérivés polysaccharides possédant différents degrés de substitution, le phénomène de thermoassociation a été étudié en milieu aqueux par des mesures rhéologiques en écoulement et en régime dynamique, de calorimétrie différentielle à balayage et de spectroscopie RMN 13C.
Ces systèmes sont sensibles à la variation de divers paramètres externes tels que la force ionique du milieu aqueux ou la concentration en polymères.
Les hydrogels obtenus au cours de ce travail présentent des applications potentielles dans les domaines biomédical, pharmaceutique et cosmétique.
