The title compound, C18H22N2O5, was synthesized by nitrification of its enol precursor. The pyrrolidine ring plane adopts a twisted conformation about the C—C bond linking the spiro centre and the C=O group remote from the N atom. It makes dihedral angles of 71.69 (9) and 88.92 (9)°, respectively, with the benzene ring plane and the plane defined by the four C atoms that form the seat of the of the cyclohexane chair. At the spiro centre, the NH group is axial and the C=O group is equatorial with respect to the cyclohexane ring. In the crystal, inversion dimers linked by pairs of N—H...O hydrogen bonds generateR22(8) loops. The dimers are linked by C—H...O interactions, generating a three-dimensional network.

Furocoumarins (FCs) are an important class of photoactive compounds which may potentially bind to DNA forming intermolecular complexes, and once excited by UVA light (~ 365 nm) they re able to form photoadducts, which may result in mutagenicity and lethality. However, when the 8-methoxypsoralen (8-MOP) is added to the post-irradiation plating medium increases the sensitization probably by inhibiting repairs in damaged DNA. The FCs also have a protective effect against UVC (~254 nm) attributed to the inhibition of pyrimidine dimers. FCs associated with UVB light (312 nm), remain few known. The aim of the present study was to investigate the lethal effect of UVB light alone, and combined with solutions of 8-MOP, 4,5',8-trimethylpsoralen (TMP) and 3-carbetoxypsoralen (3-CPs) at different concentrations on Staphylococcus aureus growth. We also evaluated the effect of these FCs in the plating medium. Treatment with 8-MOP-UVB and TMP-UVB were more effective in inducing lethality than the UVB treatment alone. Increasing the solution concentration of 8-MOP resulted in a higher mortality while the increase in the concentration of the TMP led to a reduction in the lethality. For other hand, 3-CPs displayed a photoprotective effect against UVB damage in all concentrations tested. The results of FCs in the plating medium showed that the 8-MOP induzed a higher lethal effect and also increased mortality from bacterial strain treated by FC-UVB. The different behaviors shown by FCs may be related with differences in the sequence specificity of binding and photoreaction, inhibition of pyrimidine dimers formation by intercalated molecules and efficacy of repair systems. These results emphasize the need for further studies to elucidate the participation of FCs as photosensitizing and photoprotective agents in biological systems, when combined with UVB.
As furocumarinas (FCs) são uma importante classe de compostos fotoativos que potencialmente podem se ligar ao DNA formando complexos intermoleculares, e uma vez excitados por luz UVA (~365 nm) são capazes de formar fotoadições, que podem resultar em mutagênese e letalidade. Porém, quando a 8-metoxipsoraleína (8-MOP) está adicionada ao meio de plaqueamento pós-irradiação aumenta a sensibilidade provavelmente por inibir reparo de lesões no DNA. As FCs também possuem efeito protetor contra UVC (~254 nm) atribuído à inibição da formação de dímeros de pirimidina. As FCs associadas à UVB (312 nm) permanecem pouco conhecidas. O objetivo do presente estudo foi investigar o efeito da luz UVB, e combinada com soluções de 8-MOP, 4,5 ,8-trimetilpsoraleína (TMP) e 3-carbetoxipsoraleína (3-CPs) em diferentes concentrações, sobre o crescimento de Staphylococcus aureus. Avaliamos também o efeito destas FCs em meio de plaqueamento. O tratamento com 8-MOP-UVB e TMP-UVB foram mais eficazes em induzir letalidade do que o tratamento apenas com UVB. O aumento da concentração de 8-MOP resultou em mortalidade mais elevada enquanto que o aumento na concentração de TMP levou a redução na mortalidade. Por outro lado, 3-CPs exibiu efeito fotoprotetor contra danos causados por UVB em todas as concentrações testadas. Os resultados com FCs no meio de plaqueamento mostraram que a 8-MOP induziu o maior efeito letal e também aumentou a mortalidade da cepa bacteriana tratada por FC-UVB. Os diferentes efeitos apresentados pelas FCs podem estar relacionados com diferenças na especificidade por seqüência de ligação e fotorreação, a inibição da formação de dímeros de pirimidina por moléculas intercaladas e eficácia de sistemas de reparo. Esses resultados reforçam a necessidade de mais estudos para elucidar a participação das FCs como agentes fotossensibilizantes e fotoprotetores em sistemas biológicos, quando combinadas com UVB.

Purpose: Monoamine oxidase (MAO) plays a key role in the treatment of Parkinson‟s disease (PD), since it is the major enzyme responsible for the catabolism of dopamine in the substantia nigra of the brain. Inhibition of MAO-B may conserve dopamine in the brain and provide symptomatic relief. The MAO-B inhibitors that are currently used for the treatment of PD, are associated with a variety of adverse effects (psychotoxic and cardiovascular effects) along with additional disadvantages such as irreversible inhibition of the enzyme. Irreversible inhibition may be considered a disadvantage, since following treatment with irreversible inhibitors, the rate by which the enzyme activity is recovered may be variable and may require several weeks. In contrast, following the administration of reversible inhibitors, enzyme activity is recovered when the inhibitor is cleared from the tissues. There exists therefore, a need to develop new reversible inhibitors of MAO-B which are considered to be safer than irreversible MAO-B inhibitors. Rationale: Recently discovered reversible MAO-B inhibitors include safinamide and (E)-8-(3-chlorostyryl)caffeine (CSC). Safinamide has a benzyloxy side chain, which is thought to be important for inhibition of MAO-B. CSC, on the other hand, consists of a caffeine moiety with a styryl substituent at C-8, which is also a critical feature for its inhibitory activity. In a previous study, the caffeine ring and the benzyloxy side chain were combined to produce a series of 8-benzyloxycaffeine analogues which proved to be potent new MAO-B inhibitors. In this study, caffeine was substituted with the phenoxymethyl functional group at C-8, instead of the benzyloxy moiety. The aim of this study was therefore to compare the MAO-B inhibition potencies of selected 8-(phenoxymethyl)caffeine analogues with the previously studied 8-benzyloxycaffeine analogues. In the current study, 8-(phenoxymethyl)caffeine (1) and nine 8-(phenoxymethyl)caffeine analogues (2-10) were synthesized and evaluated as inhibitors of recombinant human MAOA and –B. These analogues only differed in substitution on C3 and C4 of the phenoxymethyl phenyl ring. The substituents that were selected were halogens (Cl, F, and Br), the methyl group, the methoxy group and the trifluoromethyl group. These substituents are similar to those selected in a previous study where 8-benzyloxycaffeine analogues were evaluated as MAO inhibitors. This study therefore explores the effect that a variety of substituents on C3 and C4 of the phenoxymethyl phenyl ring will have on the MAO-A and –B inhibition potencies of 8-(phenoxymethyl)caffeine. Based on the results, additional 8-(phenoxymethyl)caffeine analogues with improved MAO-A and –B inhibition potencies will be proposed for investigation in future studies. Methods: The target, 8-(phenoxymethyl)caffeine, analogues were synthesized by reacting 1,3- dimethyl-5,6-diaminouracil with the appropriately substituted phenoxyacetic acid in the presence of a carbodiimide coupling agent. Ring closure was catalyzed in basic conditions and methylation of the resulting theophyline intermediates at C-7 was carried out with iodomethane. The structures and purities of all the target compounds were verified by NMR, MS and HPLC analysis. All of the 8-(phenoxymethyl)caffeine analogues were subsequently evaluated as MAO-A and –B inhibitors using the recombinant human enzymes. The inhibition potencies of the analogues were expressed as the IC50 values (concentration of the inhibitor that produces 50% inhibition). In addition, the time-dependency of inhibition of both MAO-A and –B was evaluated for two inhibitors in order to determine if these inhibitors interact reversibly or irreversibly with the MAO isozymes. A Hansch-type quantitative structure-activity relationship (QSAR) study was carried out in order to quantify the effect that different substituents on the phenyl ring of the 8-(phenoxymethyl)caffeine analogues have on MAO-B inhibition activity. Results: The results showed that among the test compounds, several analogues potently inhibited human MAO-B. The most potent inhibitor was 8-(3-bromophenoxymethyl)caffeine with an IC50 value of 0.148 μM toward human MAO-B. There were also inhibitors which displayed inhibition activities towards human MAO-A with IC50 values ranging from 4.59 μM to 34.0 μM. Compared to the 8-benzyloxycaffeine analogues, that were in general non-selective inhibitors, the 8-(phenoxymethyl)caffeine analogues, evaluated here, were selective for MAO-B. For example, 8-(3-bromophenoxymethyl)caffeine was found to be 141 fold more selective as an inhibitor of MAO-B than of MAO-A. Also, compared to the 8-benzyloxycaffeine analogues, the 8-(phenoxymethyl)caffeine analogues were slightly less potent MAO-B inhibitors. For example, 8-benzyloxycaffeine is reported to have an IC50 value of 1.77 μM for the inhibition of human MAO-B while 8-(phenoxymethyl)caffeine was found to have an IC50 value of 5.78 μM for the inhibition of human MAO-B. This study also shows that two selected analogues bind reversibly to MAO-A and –B, respectively, and that the mode of MAO-B inhibition is competitive for one representative compound. Qualitative inspection of the results revealed interesting structure-activity relationships. For the 8-(phenoxymethyl)caffeine analogues, bearing both the C3 and C4 substituents on the phenyl ring, the MAO-B activity significantly increases with halogen substitution. Furthermore, increased MAO-B inhibition was observed with increased electronegativity of the halogen substituent. To quantify these apparent relationships, a Hansch-type QSAR study was carried out. The results showed that the logarithm of the IC50 values (logIC50) correlated with Hansch lipophilicity (π) and the Swain-Lupton electronic (F) constants of the substituents at C-3 of the phenoxymethyl ring. The correlation exhibited an R2 value of 0.87 and a statistical F value of 13.6. From these results it may be concluded that electron-withdrawing substituents at C3 with a high degree of lipophilicity enhance MAO-B inhibition potency. These results are similar to those previously obtained for the series of 8-benzyloxycaffeine analogues. For this series, the MAO-B inhibition potencies correlated with the Hansch lipophilicity (π) and Hammett electronic (σ) constants of the substituents at C-3 of the benzyloxy ring. Similarly to the 8-(phenoxymethyl)caffeine analogues, electron-withdrawing substituents with a high degree of lipophilicity also enhance the MAO-B inhibition potencies of 8-benzyloxycaffeine analogues.

Runowski, Polnische Orgelmusik nach 1945 Zwar kann die moderne polnische Musik aufgrund des seit 1956 existierenden und seinerzeit so spektakulären Festivals Warszawska Jesien [Warschauer Herbst] auch im Ausland als immerhin in groben Grundzügen bekannt gelten. Da aber – wie Runowski selbst hervorhebt– geistliche Musik auf diesem Festival nie eine größere Rolle spielte, blieb dieser Bereich polnischen Musikschaffens nicht nur im Ausland, sondern auch in Polen selbst einem breiteren Publikum verborgen. Umso interessanter ist die Existenz einer durchgehenden polnischen Orgelmusiktraditionseit Kriegsende bis heute, und doppelt interessant ist diese Existenz angesichts der Spannung zwischen katholischem Glauben und kommunistischem System in der Volksrepublik Polen.

Zika (ZIKV), La Crosse (LACV), and Cache Valley (CVV) viruses are mosquito-vectored diseases that cause significant morbidity and mortality in humans and animals. Transmission of these viruses are dependent on numerous factors including vector competence and the effects of mosquito-virus interactions. We conducted vector competence studies of local Aedes and Culex mosquitoes for ZIKV and CVV, and found that all Aedes mosquitoes were competent for CVV and only Aedes albopictus and Aedes japonicus were competent for ZIKV. Vector competence for CVV was dose-dependent, where mosquitoes orally infected with high titers developed higher transmission rates. We also found that vector competence for ZIKV was limited by midgut and salivary gland barriers. Second, we looked at the effects of LACV and ZIKV infection on repellent response in Aedes mosquitoes and found that infected mosquitoes were refractory to low concentrations of DEET, picaridin, and PMD. Increasing concentrations of the repellents to ≥10% was able to increase percent protection (%p) against infected and uninfected mosquitoes. Lastly, we determined the effects of ZIKV and LACV infection on oxidative stress and glutathione-S-transferase (GST) activity in Aedes albopictus. Virus infection had no effect on oxidative stress, but GST activity was significantly different for mosquitoes 3-days post-exposure. We found that oxidative stress levels and GST activity had an inverse relationship for infected and uninfected mosquitoes, where oxidative stress decreased and GST activity increased over the 10-day test period. This indicates that GSTs may aid in controlling byproducts of oxidative stress. The results from this entire study identified competent vectors for emerging arboviruses and demonstrated the behavioral and physiological effects of virus infection in the mosquito vector.
Doctor of Philosophy
Zika (ZIKV), La Crosse (LACV), and Cache Valley (CVV) viruses are transmitted by mosquitoes and can make humans and animals very sick. There are many biological factors that determine if a mosquito can transmit a virus and these viruses can change the biology of a mosquito. We conducted laboratory studies to see if Aedes and Culex mosquitoes can transmit ZIKV and CVV. We found that all Aedes mosquitoes were able to transmit CVV and only the Asian tiger mosquito and Asian rock pool mosquito were able to transmit ZIKV. Mosquitoes infected with high amounts of CVV developed higher transmission rates. We also found that transmission of ZIKV was limited by barriers in the mosquito midgut and salivary glands. Second, we looked at the effects of LACV and ZIKV infection on how Aedes mosquitoes respond to repellents and found that infected mosquitoes were less sensitive to low concentrations of DEET, picaridin, and PMD. Increasing concentrations of the repellents to 10% or higher was able to provide adequate protection against infected and uninfected mosquitoes. Lastly, we determined the effects of ZIKV and LACV infection on oxidative stress and glutathione-S-transferase (GST) activity in the Asian tiger mosquito. Virus infection did not change oxidative stress, but GST activity was higher in infected mosquitoes tested after 3 days after infection. We found that oxidative stress decreased and GST activity increased over the 10-day test period. This indicates that GSTs may help control damaging products from oxidative stress. The results from this entire study identified what mosquitoes were able to transmit emerging mosquito-borne viruses and demonstrated the biological effects of virus infection in the mosquitoes.

Le passage du (1R,2S,5R)-2-diméthylphénylméthyl-5-méthylcyclohexyl carbaldéhyde à l'imine correspondante du glycinate de méthyle, suivi de la déprotonation (diisopropylamidure de lithium), puis de l'alkylation par des halogénures d'alkyles conduit après hydrolyse à des α-aminoacides de configuration (R) avec des e. E. >98%. Il a été montré que pour obtenir les α-aminoacides de configuration (S), il n'était pas nécessaire d'inverser la totalité des centres stéréogéniques de l'aldéhyde initial, l'inversion d'un seul centre étant suffisante. Ainsi, à partir de l'aldéhyde (1S,2S,5R), des α-aminoacides de configuration (S) ont été obtenus avec des e. E. >98%. La méthode a été généralisée à la synthèse d'un α-aminoacide à centre quaternaire la (R)-methyldopa avec un e. E. >98%. Les aldéhydes (1R,2S,5R) et (1S,2S,5R) sont aisément obtenus à partir de la (R)-(+)-pulégone et transformés facilement l'un en l'autre par épimérisation. L'aldéhyde utilisé est donc un auxiliaire chiral à centre d'aiguillage (R,S).

AbstractUnruptured brain AVMs (bAVMs) remain a controversial subject for practicing neurosurgeons, especially in the light of ARUBA and other observational studies. This retrospective study aims to analyze our experience with unruptured bAVMs to see whether it is beneficial in the long-term and how it corresponds to large literature trials. The study comprised 160 adult patients with unruptured bAVMs surgically treated in Burdenko NMC (Moscow) in 2009–2017. Mean age: 33.4 ± 10лет. Clinical presentations were: seizures in 99 (61.9%), chronical headaches—49 (30.6%), ischemic symptoms—4 (2.5%), asymptomatic in 8 (5%) patients. Spetzler-Martin scale: I—18 pt. (11.3%), II—71 pt. (44.4%), III—60 pt. (37.5%), IV—11 pt. (6.8%). Good outcomes (mRS = 0–2) at discharge were achieved in 149 (93.1%), satisfactory (mRS—3)—9 (5.6%). Follow-up was complete for 97 (60.6%) patients, mean—59.3 (13–108 month). Excellent outcomes (mRS = 0–1) reached in 94.8%. For epilepsy patients, Engel I outcome was found in 50 (84.8%); for chronic headaches, 43 (66.1%) patients reported improvement. Postoperative visual field defects were followed in 22 of 55 (40%), complete recovery was reported in 6 (27%) and partial recovery in 8 (36%) patients. Overall, our results support the conclusion that surgery for low-grade bAVMs (S-M I–II) is a beneficial, low-risk option.

In this chapter, the design and implementation of the self-synchronized adaptive linear neural (S-ADALINE) network approach for harmonic detection and the measurement of electrical power components (active power, reactive power, apparent power, power factor, etc.) at non-sinusoidal state have been discussed. A brief of the importance of electrical power measurement has been specified in Section 2. Section 3 is to discuss on the modern techniques of power measurement. The use of ADALINE style for harmonic estimation has been reflected in Section 4. In Section 5, harmonic tracking using S-ADALINE has been presented. Section 6 has been utilized to compare between ADALINE and S-ADALINE. In Section 7, S-ADALINE based power components measurement has been enlightened. A set of simulation tests in terms of accuracy, speed, and convergence of the S-ADALINE has been performed in MATLAB and deliberated in Section 8. The developed prototype is presented in Section 9 with experimental results.

Diesel or JP-8 fueled SOFC power systems are envisioned for both critical and non-critical military use such as auxiliary power units (APUs), portable power, tent cities, camp kitchens, and permanent stationary power [1]. A system that eliminates or minimizes the addition of make-up water is a great advantage in these applications. This paper presents an approach for water recovery from the spent anode exhaust gas for use in the JP-8 steam reformer [2]. A key objective of this on-going effort is to configure an internal water management scheme for the system to recover water for the JP-8 fuel processor steam-reforming requirements. The approach used was to combust the water-laden SOFC anode exhaust gas (AEG), condense and recycle the water to the reformer. HYSYS simulation performed on the configured system indicated that over 91% of fuel processor water requirements can be obtained in this approach to maintain a steam-to-carbon (S/C) ratio of 4. The S/C ratio was set at 4 to ensure a high JP-8 conversion and prevent carbon formation in the reformer. Operational tests at simulated system conditions confirmed >91% water recovery as predicted. System tests of this scheme indicate that it is feasible to combust and recover product water for reforming, minimizing makeup water requirements. The results from the system tests are promising for configuring stationary power systems for military use where water availability is limited.

A comprehensive study of Mississippi Valley-type base-metal deposits across the Canadian Cordillera was done to compare and contrast their features. Extensive dissolution of host rocks is followed by multiple generations of dolomite cements from early, low-temperature, fine-grained to coarser, higher temperature types that overlap with Zn-Pb sulfide minerals; late-stage calcite occludes residual porosity. Dolomite is generally chemically stoichiometric, but ore-stage types are often rich in Fe (<1.3 weight per cent FeO) with small sphalerite inclusions. Sphalerite-hosted fluid inclusions record ranges for homogenization temperatures (77-214°C) and fluid salinity (1-28 weight per cent equiv. NaCl±CaCl2). These data suggest fluid mixing with no single fluid type related to all sulfide mineralization. In situ secondary ion mass spectrometry (SIMS) generated delta-18OVSMOW values for carbonate minerals (13-33 permille) reflect dolomite and calcite formation involving several fluids (seawater, basinal, meteoric) over a large temperature range at varying fluid-rock ratios. Sphalerite and pyrite SIMS delta-34SVCDT values vary (8-33 permille) but in single settings have small ranges (<2-3 permille) that suggest sulfur was reduced via thermochemical sulfate reduction from homogeneous sulfur reservoirs. Collectively, the data implicate several fluids in the mineralizing process and suggest mixing of a sulfur-poor, metal-bearing fluid with a metal-poor, sulfide-bearing fluid.

Highly asymmetric somatic hybrid plants were obtained by PEG/DMSO fusion of gamma irradiated (100, 250, 7500 and 1000 Gy) protoplasts of a (KmR-) interspecific hybrid Lycopersicon esculentum x L. pennellii (EP) with protoplasts of eggplant (E). Somatic hybrid calli were selected based on kanamycin resistance and verified by PCR of the NptII gene, RAPD's and Southern's using potato rDNA pTHG2 probes. Flow cytometry indicated all hybrid calli that did not regenerate shoots were 5-9n. Three asymmetric plants regenerated only from callus close to 4n and such calli oly occurred when EP received 100 Gy. The asymmetric plants had eggplant morphology and regenerated from one hybrid callus with 6.29 average size tomato chromosomes. Limited amounts of EP DNA were found in the three somatic hybrid plants H18-1 to -3 by dot-blot hybridization with probe pTHG2, to be equivalent to 6.23, 5.41, and 5.95 % EP, respectively. RFLP analysis of Lycopersicon esculentum and L. pennellii specific chromosomes revealed that only fragments of 8 to 10 out of the 24 EP chromosomes are present in the asymmetric plants. Transgenic plants 2-3, 2-4 and 10-3 were found resistant to verticillium; suggesting successful transfer of the Ve complex from S. torvum to eggplant.

In this study we investigated coda-wave attenuation (QC) from the eastern Canadian Arctic in Nunavut and the Hudson Bay complex including portions of northern Manitoba, Ontario, Quebec and Labrador. We used earthquake recordings from 15 broadband and 3 short period seismograph stations of the Canadian National Seismic Network (CNSN) and 29 broadband stations of the POLARIS network across the region. Our dataset is comprised of 637 earthquakes recorded between 1985 and 2021 with magnitudes ranging from 1.3 to 6.1, depths from 0 to 20 km and epicentral distances of 5 to 100 km. This gives a total of 246 high signal-to-noise (S/N) traces (S/N[lesser/equal]5.0) useful for QC calculation (with a maximum ellipse parameter, a2, of 100) across the region. Coda windows were selected to start at tc = 2tS (two times the travel time of the direct S wave), and were filtered at center frequencies of 2, 4, 8, 12 and 16 Hz. Our study reveals a consistent pattern. We find that in the northern section of the study area, the highest Q0 values (e.g., Q0 of 110 and 112) are at station POIN and station RES, respectively, which are located in the older Archean province. The lowest Q0 values that we find (e.g., Q0 of 55 and 61) are at station AKVQ and IVKQ respectively, located in northern Quebec. Smaller Q0 values for stations in the south are explained by the younger age of the rocks and proximity to the main fault systems. An average for all the data results in a Q relationship of QC = 82f1.08 for the frequency band of 2 to 16 Hz for the entire region.

Evaluate the capabilities of current technologies that are offered commercially for methane leak detection monitoring using standard pipeline patrol aircraft 2. Provide a test location for development of emerging technologies that are not yet commercially available for pipeline leak detection 3. Assess the feasibility of using airborne sensors to detect staged liquid oil leaks The field study was conducted on two of Enbridge�s operating pipelines located near Mineral Wells, Texas. This location provided realistic conditions for assessing the capabilities and limitations of automated sensor systems that are currently available for leak detection. Three airborne leak detection vendors (Lasen, Pergam, and New Era Technology), and one ground vehicle vendor (Picarro), completed the field study. The field study was performed as part of the 2012 PRCI RAM program that is primarily focused on leak detection technologies using aerial platforms. The study tested a range of sensors designed for gas leak detection using systems mounted on helicopters and fixed wing aircraft. Enbridge staged a series of controlled gas releases simulating pipeline gas leaks. Qualified operators released a series of prescribed natural gas leaks from 8 known locations along the pipelines in order to simulate transmission leaks of varying sizes. Additionally, Enbridge placed 8 liquid petroleum targets along the test pipelines. These gas and oil targets allowed for a direct comparison of technology performance against a known set of conditions. Standard leak patrol methods (ground survey with foot patrol) were also used to identify any emission sources other than the controlled releases. The results from the field study show that 3 of the 4 vendor sensor technologies tested are now feasible alternatives for gas leak detection. The findings for liquid leak detection were promising, but inconclusive due to plume overlaps between gas and oil targets. The sensors used by the vendors in this field test were optimized for detecting methane gas and not oil vapors. Therefore, until additional studies are performed on liquid hydrocarbon volatilization, it is premature to conclude that the tested technologies are or are not appropriate for oil leak detection.

Growth rate of cultured fish from hatching to commercial size is a major factor in the success of aquaculture. The normal stimulus for muscle growth in growing fish is not well understood and understanding the regulation of muscle growth in fish is of particular importance for aquaculture. Fish meat constitutes mostly of skeletal muscles and provides high value proteins in most people's diet. Unlike mammals, fish continue to grow throughout their lives, although the size fish attain, as adults, is species specific. Evidence indicates that muscle growth is regulated positively and negatively by a variety of growth and transcription factors that control both muscle cell proliferation and differentiation. In particular, growth hormone (GH), fibroblast growth factors (FGFs), insulin-like growth factors (IGFs) and transforming growth factor-13 (TGF-13) play critical roles in myogenesis during animal growth. An important advance in our understanding of muscle growth was provided by the recent discovery of the crucial functions of myostatin (MSTN) in controlling muscle growth. MSTN is a member of the TGF-13 superfamily and functions as a negative regulator of skeletal muscle growth in mammals. Studies in mammals also provided evidence for possible interactions between GH, IGFs, MSTN and the musclespecific transcription factor My oD with regards to muscle development and growth. The goal of our project was to try to clarify the role of MSTNs in Sparus aurata muscle growth and in particular determine the possible interaction between the GH-IGFaxis and MSTN in regulating muscle growth in fish. The steps to achieve this goal included: i) Determining possible relationship between changes in the expression of growth-related genes, MSTN and MyoD in muscle from slow and fast growing sea bream progeny of full-sib families and that of growth rate; ii) Testing the possible effect of over-expressing GH, IGF-I and IGF-Il on the expression of MSTN and MyoD in skeletal muscle both in vivo and in vitro; iii) Studying the regulation of the two S. aurata MSTN promoters and investigating the possible role of MyoD in this regulation. The major findings of our research can be summarized as follows: 1) Two MSTN promoters (saMSTN-1 and saMSTN-2) were isolated and characterized from S. aurata and were found to direct reporter gene activity in A204 cells. Studies were initiated to decipher the regulation of fish MSTN expression in vitro using the cloned promoters; 2) The gene coding for saMSTN-2 was cloned. Both the promoter and the first intron were found to be polymorphic. The first intron zygosity appears to be associated with growth rate; 3) Full length cDNA coding for S. aurata growth differentiation factor-l I (GDF-II), a closely related growth factor to MSTN, was cloned from S. aurata brain, and the mature peptide (C-terminal) was found to be highly conserved throughout evolution. GDF-II transcript was detected by RT -PCR analysis throughout development in S. aurata embryos and larvae, suggesting that this mRNA is the product of the embryonic genome. Transcripts for GDF-Il were detected by RT-PCR in brain, eye and spleen with highest level found in brain; 4) A novel member of the TGF-Bsuperfamily was partially cloned from S. aurata. It is highly homologous to an unidentified protein (TGF-B-like) from Tetraodon nigroviridisand is expressed in various tissues, including muscle; 5) Recombinant S. aurata GH was produced in bacteria, refolded and purified and was used in in vitro and in vivo experiments. Generally, the results of gene expression in response to GH administration in vivo depended on the nutritional state (starvation or feeding) and the time at which the fish were sacrificed after GH administration. In vitro, recombinantsaGH activated signal transduction in two fish cell lines: RTHI49 and SAFI; 6) A fibroblastic-like cell line from S. aurata (SAF-I) was characterized for its gene expression and was found to be a suitable experimental system for studies on GH-IGF and MSTN interactions; 7) The gene of the muscle-specific transcription factor Myogenin was cloned from S. aurata, its expression and promoter activity were characterized; 8) Three genes important to myofibrillogenesis were cloned from zebrafish: SmyDl, Hsp90al and skNAC. Our data suggests the existence of an interaction between the GH-IGFaxis and MSTN. This project yielded a great number of experimental tools, both DNA constructs and in vitro systems that will enable further studies on the regulation of MSTN expression and on the interactions between members of the GHIGFaxis and MSTN in regulating muscle growth in S. aurata.

The National Park Service (NPS) began monitoring landbirds at Channel Islands National Park in 1993 as part of its long-term inventory and monitoring program. The park?s landbird monitoring later became part of the NPS Inventory and Monitoring Division?s Mediterranean Coast Network long-term monitoring programs. Consequently, landbird monitoring has been conducted in the park during every breeding season since 1993. In this report, we summarize data collected during the 2020 breeding season. Landbird monitoring was conducted between 1 April and 30 June 2020. Using distance-based sampling methods in a standardized protocol, birds were counted on 7 of 10 permanent line transects (70%) (2 of 3 on Santa Barbara Island, 1 of 1 on East Anacapa Island, and 4 of 5 on San Miguel Island). Two transects were not sampled in 2020 because of nesting California Brown Pelicans (Pelecanus occidentalis) on Santa Barbara Island (i.e., Canyons Transect) and reduced person hours and unfavorable winds on San Miguel Island (i.e., San Miguel Hill Transect). For point counts, 225 of 338 (67%) permanent stations were counted (i.e., 30 of 33 points on Santa Barbara Island, 8 of 8 on East Anacapa Island, 100 of 112 on Santa Cruz Island, 40 of 40 on San Miguel Island, and 47 of 145 on Santa Rosa Island). The 8 Prisoners? Cove points were not counted in 2020. Three points were not counted on Santa Barbara Island due to nesting pelicans: these points and the transect were also not counted in 2016?2019 to avoid disturbing breeding pelicans. Other points (i.e., on east Santa Cruz Island and Santa Rosa Island) were not counted due in large part to the global COVID-19 pandemic. Traveling to and from the park was only granted to essential NPS staff for the majority of the landbird season. Fifty-one bird species were counted from points and transects across all of the islands in 2020; 39 of these are breeding species on the island. Parkwide, the 10 most commonly detected breeding landbirds in 2020 were, in descending order: Horned Lark, Spotted Towhee, Song Sparrow, White-crowned Sparrow, Western Meadowlark, Orange-crowned Warbler, Bewick?s Wren, Island Scrub-Jay, House Finch, and Common Raven. On East Anacapa Island, 26 landbird species have been counted since 1993; 5 species were counted in 2020. No new transient species were detected in 2020; 10 transient or visiting species (nonbreeding, native species recorded only once or twice during surveys) have been counted on the island overall since 1993. On Santa Barbara Island, 49 landbird species have been counted since 1993; 15 species were counted in 2020. The highest number of Horned Lark since 1993 were counted in 2020 (n = 451). Warbling Vireo was a new transient species counted in 2020 on Santa Barbara; 30 transient or visiting species have been counted on the island since 1993. On Santa Cruz Island, 74 landbird species have been counted since 2013; 34 species were counted in 2020. Bullock?s Oriole was a new transient species counted in 2020 on Santa Cruz; 21 transient or visiting species have been counted on this island since 2013. On San Miguel Island, 69 landbird species have been counted since 1993; 14 were counted in 2020. No transient species were counted in 2020; 34 transient or visiting species have been counted on the island since 1993. On Santa Rosa Island, 75 landbird species have been counted since 1994; 26 were detected in 2020. Rose-breasted Grosbeak was a new transient species counted in 2020 on Santa Rosa Island; 30 transient or visiting species have been counted on the island since 1994. Across all the 5 islands, 3 transient or visiting bird species were newly counted in 2020, for a total of 77 such species counted since NPS monitoring began on the islands. Nonnative and invasive birds were counted on only 1 of the 5 islands in 2020: 4 European Starlings on Santa Rosa Island. However, anecdotal sightings of nonnative species occurred much more frequently (i.e., outside of the point and transect counts), and were made on all islands except Anacapa in 2020. The highest numbers of nonnative species detections occurred on Santa Cruz Island, with 33 detections of Eurasian Collared Dove (primarily at the Main Ranch area in the Central Valley), 15 detections of Brown-headed Cowbird (primarily at Scorpion Harbor), and 15 detections of European Starling (primarily at the Main Ranch and Scorpion Harbor). House Sparrows were observed fewer times, but on all islands except Anacapa; cowbirds occurred on all islands except Anacapa and San Miguel; and Rock Pigeon occurred on Santa Barbara and Santa Cruz Islands. In 2020, despite the COVID-19 pandemic, 67% of all points and 77% of all transects were counted among the 5 islands. Santa Rosa received the lightest sampling of points (32%), due to the difficulty of getting observers onto the island. Even with diminished sampling, species richness (number of species) values fell in predictable patterns: richness was greatest on the larger islands (75 on Santa Rosa, 73 on Santa Cruz) and least on the smallest islands (26 on Anacapa, 48 on Santa Barbara). We continue to recommend that nonnative invasive species, such as European Starlings on Santa Rosa Island, be removed before their numbers become harder to manage. Also, because Distance analyses assist statistically with evaluations of trends, we continue to recommend that a trend analysis using program DISTANCE, or newer hierarchical distance analyses, should be used after the 2020 season to assess 5-year trends in breeding species? numbers following the 2015 trend analysis conducted by Coonan and Dye (2016).

Tomato yellow leaf curl geminivirus (TYLCV) is a major pathogen of cultivated tomato, causing up to 100% crop loss in many parts of the world. In Israel, where TYLCV epidemics have been recorded since the 1960' s, this viral disease is well known and has been of economic significance ever since. In recent years, TYLCV outbreaks also occurred in the "New World" - Cuba, The Dominican Republic, and in the USA, in Florida, Georgia and Louisiana. Thus, TYLCV substantially hinders tomato growth throughout the world. Surprisingly, however, little is known about the molecular mechanisms of TYLCV interaction with the host tomato cells. The present proposal, a continuation of the project supported by BARD from 1994, expanded our understanding of the molecular mechanisms by which TYLCV enters the host cell nucleus for replication and transcription and exits it for the subsequent cell-to-cell spread. Our project sought two objectives: I. To study the roles of the viral capsid protein (CP) and host cell factors in TYLCV nuclear import. II. To study the roles of CP and host cell factors in TYLCV nuclear export. Our research toward these goals have produced the following major achievements: . Developed a one-hybrid assay for protein nuclear export and import (#3 in the List of Publications). . Identified a functional nuclear export signal (NES) in the capsid protein (CP) of TYLCV (#3 in the List of Publications). . Discovered homotypic interactions between intact TYLCV CP molecules and analyzed these interactions using deletion mutagenesis of TYLCV CP (#5 in the List of Publications). . Showed developmental and tissue-specific expression of the host factor required for nuclear import of TYLCV CP, tomato karyopherin alpha 1, in transgenic tomato plants (#14 in the List of Publications). . By analogy to nuclear import of TYLCV ,identified an Arabidopsis VIPI protein that participates in nuclear import of Agrobacterium T -complexes via the karyopherin alpha pathway (#4,6, and 8 in the List of Publications). These research findings provided significant insights into (i) the molecular pathway of TYLCV entry into the host cell nucleus, and (ii) the mechanism by which TYLCV is exported from the nucleus for the cell-to-cell spread of infection. Furthermore, the obtained knowledge will help to develop specific strategies to attenuate TYLCV infection, for example, by blocking viral entry into and/or exit out of the host cell nucleus. Also, as much of our findings is relevant to all geminiviruses, new anti- TYLCV approaches developed based on the results of our research will be useful to combat other members of the Geminivirus family. Finally, in addition to the study of TYLCV nuclear import and export, our research contributed to our understanding of general mechanisms for nucleocytoplasmic shuttling of proteins and nucleic acids in plant cells.

Vulnerability assessments are a common approach to identifying conservation priorities for species and informing resource management decisions by evaluating species? exposure (spatial extent and frequency/intensity) and sensitivity (whether and how a species will respond) to a threat. We assessed the vulnerability of the Northern Spotted Owl (NSO; Strix occidentalis caurina) in Marin County to eight distinct threats: 1) competition with Barred Owls (S. varia); 2) climate change; 3) wildfire; 4) sudden oak death (SOD); 5) other habitat loss/disturbance, specifically from urban development or fuels management; 6) noise disturbance; 7) rodenticide poisoning; and 8) disease. Specifically, we reviewed evidence from published literature for the relative vulnerability of NSOs to each potentially relevant threat in the region; surveyed local experts on the relative vulnerability of NSOs to each potential threat; ranked the vulnerability of NSOs to each threat in Marin County; and made conservation recommendations. We identified three threats to which NSOs are currently most vulnerable in Marin County: climate change, competition with Barred Owls, and wildfire (specifically high-severity fires); exposure to each of these threats is expected to increase over the next 30?50 years. We identified rodenticides and SOD as moderate threats, and habitat loss (from urban development and fuels management), noise disturbance, and disease were identified as the lowest relative threats. However, except for urban development, the local experts? confidence in their assessment of the sensitivity of NSOs to the lower-ranked threats was low, suggesting a need for further research. Conservation recommendations include: continued habitat protections that consider NSOs and dusky-footed woodrat habitat on a landscape scale; management of Barred Owls to maintain the current low level of exposure; continued avoidance of noise disturbance to nesting NSOs; and community education and outreach efforts to help reduce the threat of rodenticides, noise, and other types of human disturbance. We identified research needs and uncertainties including evaluating the response of NSOs and their prey in Marin County to: weather and climate, including extreme events; habitat change including to fuels management and SOD; and noise and other human disturbance. Conservation actions to protect NSOs and the habitat and prey on which they depend, and outreach to local communities and land managers will be an essential part of the future health of this population.

Caribou (Rangifer tarandus) are an integral part of the ecological and cultural fabric of northwest Alaska. Western Arctic Herd (WAH) caribou roam over this entire region, including all 5 Arctic Network (ARCN) Inventory and Monitoring Program?s National Park units. Conservation of healthy caribou populations is specifically mentioned within the enabling legislation (Alaska National Interested Lands Conservation Act or ANILCA) for 3 of these park units and is of importance to subsistence hunters. Caribou are, by far, the most abundant large mammal in northwest Alaska and are famous for their long-distance migrations and large population oscillations. For these reasons, the ARCN parks chose WAH caribou as a Vital Sign for long-term monitoring. This report documents the monitoring results of this Vital Sign during its 14th year (September 2022?August 2023) of implementation. Results from the previous years of monitoring are also included for ease of comparison. Periodic syntheses of these data will be performed and reported on as appropriate. National Park Service (NPS) monitoring of the WAH is done in conjunction and cooperation with the Alaska Department of Fish and Game (ADFG). Thanks to a 2015 data sharing agreement, the report includes data funded by the ADFG dating back to September 2013. That particular year was chosen as it represents when the ADFG started deploying a substantial number of GPS collars on an 8-hour relocation schedule. Monitoring of the herd relies heavily on the use of Global Positioning System (GPS) radio telemetry collars that are capable of transmitting location data to a satellite. Given the extremely remote area that the WAH inhabits, this system provides the most efficient and accurate means to track individual caribou. These data are utilized to monitor the timing and location of migrations, as well as seasonal distributions of WAH caribou. Monitoring movement and the phenology of movement is perhaps the simplest means to track the influences of climate change, natural perturbations, development, and other potential impacts on a species?an analysis of which is outside the scope of this current report. This report also documents the NPS commitment and involvement with the WAH Working Group. The group is composed of important stakeholders including representatives for rural villages, sport hunters, conservationists, hunting guides, hunting transporters, and reindeer herders. In addition, all the agencies charged with managing the WAH, including the ADFG, NPS, US Fish and Wildlife Service (FWS) and Bureau of Land Management (BLM), serve as advisors to the group. Information gathered by the Caribou Vital Sign monitoring program are intended to supplement and complement existing data streams gathered by the other cooperating agencies and will be important in future management decisions.

Harlequin ducks (Histrionicus histrionicus) are sea ducks that migrate inland each spring to nest along fast-flowing mountain streams. They are considered one of the most imperiled duck species in North America and occur in two distinct populations on the Atlantic and Pacific coasts. The Pacific coast population includes Washington State, where harlequin ducks breed in the Olympic, Cascade, and Selkirk Mountains. This species is designated as a Management Priority Species by the National Park Service within North Cascades National Park Service Complex (NOCA). This report summarizes harlequin duck surveys conducted during 15 years across a 27-year period (1990 and 2017) on major streams within NOCA, and incidental observations collected from 1968–2021. The primary objectives of these surveys were to 1) document the distribution and abundance of harlequin duck observations within NOCA boundaries, 2) describe productivity (number of broods and brood size), 3) describe breeding chronology of harlequin ducks, and 4) describe habitat characteristics of breeding streams. Sixty-eight stream surveys over 15 years resulted in observations of 623 individual harlequin ducks comprising various demographics, including single adults, pairs, and broods. In addition, we collected 184 incidental observations of harlequin ducks from visitors and staff between 1968–2021. Harlequin ducks were observed on 22 separate second- to sixth-order streams throughout NOCA across the entire 53-year span of data, both incidentally and during harlequin duck surveys by Park staff. Harlequin ducks were detected on 8 of the 13 streams that were actively surveyed. Excluding recounts, 88.7% (n = 330) of individual harlequin duck observations during surveys occurred in the Stehekin River drainage. Between all surveys and incidental observations, 135 unpaired females without broods were sighted across all NOCA waterways. Thirty-nine broods were recorded between NOCA surveys and incidental observations, with a mean brood size of 3.61 (± 1.44 SD; range = 2–10). Breeding pairs were recorded as early as April 5 and were seen on streams until June 15, a period of less than seven weeks (median: May 2), but most pairs were observed within a 3-week span, between April 26 and May 17. Single females (unpaired with a male, with (an)other female(s), or with a brood) were observed on streams between April 26 and August 25 (median: July 3), though most observations were made within a 5-week period between June 12 and July 19. Habitat data collected at adult harlequin duck observation sites indicate that the birds often used stream reaches with features that are characteristic of high-energy running water. While adults occupied all instream habitat types identified, non-braided rapids and riffles were used most frequently, followed by pools and backwaters. Larger instream substrate sizes (cobbles and boulders) were present at most observation sites. Adult harlequin ducks were more often found at locations that lacked visible drifting or lodged woody debris, but drift debris was a slightly more abundant debris type. The presence of gravel bars and at least one loafing site was common. Adult harlequin ducks were more often observed in association with vegetation that offered some cover over the channel, but not where banks were undercut. The average channel width at adult observation sites was 34.0 m (range: 6-80 m; n = 114) and 27.6 m (± 15.7 m; range: 10-60 m; n = 12) at brood observation sites. Compared to adult harlequin duck sites, broods were observed more frequently in low velocity habitat (pools, backwaters), but rarely in rapids. Cobble and boulder substrates were still the most dominant substrate type. Contrary to adult ducks, broods were observed most often observed in meandering stream channels, a morphology indicative of low gradient, low velocity stream reaches.
