Since 2015, the project Sprachalltag II has been running at the Institute of Historical and Cultural Anthropology in Tübingen. In addition to the final processing of the Sprachatlas von Nord Baden-Württemberg (SNBW), begun in 2009, and the creation of a popular, online Sprechender Sprachatlas von Baden-Württemberg, the goal is to research and digitize the extensive material of the Arno-Ruoff archive for further linguistic and ethnologic studies. This project in particular includes the transcription and alignment of the dialect recordings collected by Ruoff and Bausinger since 1955 for the so called Zwirner corpus as well as the publication of the edited texts in a database in cooperation with the IDS Mannheim. After researching mainly on the morphological and syntactic level of the corpus, the project now also enables phonological examinations of the spoken language throughout the federal state of Baden-Württemberg and the area of Bavarian Swabia via the database. This report will introduce the nature and extent of the hitherto edited recordings in more detail and show how, in the sense of a cross-disciplinary collaboration, Empirical Cultural Studies can benefit from the content development of the Tübingen corpus. In the course of editing, the transcripted dialect recordings are assigned to thematic categories by using keywords such as leisure or modernization. This includes the means of enquiry of the corpus to a content-related level that can serve the research fields of Historical and Cultural Anthropology.

At high pressures, optical spectroscopic techniques are the principal means for probing dense solid hydrogen, including both its vibrational and electronic excitations. Ruoff et al. [Phys. Rev.B47, 11039 (1993)] proposed that refractive index gradients of H 2 samples loaded with 5% Al 2 O 3 powder in diamond anvil cells could cause a 4% rise in infrared reflectivity which interfered with the measurements on hydrogen reported by Mao et al. [Phys. Rev. Lett.65, 484 (1990)]. We have analyzed this alternative proposal, including computer simulations of the appropriate sample configuration. Our calculations show that effects of a graded index on the rise of reflectivity from 1.0 to 0.5 eV are less than 0.1% and are therefore insignificant.

A comparative analysis of the principles of the cadastre in the most developed countries and in Russia. It is shown that some of the principles of cadastre, it is advisable to introduce into the Russian legislation. Such principles include the principle of Renzenberger, as well as the principles of Ruoff and Kuranda. The Russian inventory has more than twenty years, however, it cannot be considered complete, as registered in cadastre only 60 % of real estate. Full filling of the cadastre information on real estate is possible, if we abandon the application of the principle of reference. Unlike foreign domestic inventory the inventory contains errors that complicate the procedure of registration of immovable property. In addition, the domestic inventory is not the only source of information about the property that causes the ambiguity of the information about the same object. Important is also that the damage caused inaccurate inventory information bona fide buyer or seller of real property under current law, does not exceed one million rubles, regardless of the value of the lost object. Foreign inventory recognizes the property owner the main participant of the changes, however, the Russian legislation allows for the adjustment of the information object without the application of the property owner. See principles of the foreign inventory is useful for the maintenance of the national cadastre. This will simplify the process of state cadastral accounting of real estate, reduce the time of its formation and to increase the reliability of materials of the Unified state register of real estate.

Abstract Prostate cancers adapt to androgen receptor (AR) pathway inhibitors and progress to castration resistance despite the continued expression and function of the AR. We developed a new approach to antagonize the proliferative activity of AR and inhibit castration-resistant prostate cancer (CRPC) growth using multivalent peptoid conjugates (MPCs). Our strategy is to display multiple copies of the AR antagonist, ethisterone, on a peptoid scaffold. Such a multivalent display increases the ligand’s effective local concentration, thus targeting AR with high affinity and affecting AR interactions with coregulators. Here, we report a new potent MPC called MPC309, which displays three ethisterone groups and binds to AR with nanomolar affinity. MPC309 reduces the proliferation of enzalutamide-resistant prostate cancer cells, including those harboring AR splice variants, AR ligand binding mutations, and non-canonical AR gene expression programs. We provide evidence that MPC309 enters cells via macropinocytosis, which facilitates the fluid-phase uptake of extracellular macromolecules. Macropinocytotic uptake of MPC309 enhances cancer cell-specific delivery, thus limiting systemic toxicities. MPC309 displays favorable pharmacological properties and produced significantly greater tumor suppression in xenograft studies than enzalutamide, the standard of care anti-androgen in clinical use. Mechanistically, MPC309 inhibits prostate cancer growth by eliciting a unique gene expression program through alterations in AR chromatin occupancy compared to dihydrotestosterone (DHT) or enzalutamide. Thus, MPC309 represents a novel AR antagonist that activates an AR anti-proliferative program to repress the growth of CRPC and supports further investigation of the MPC compound class for treating CRPC. Objective:To evaluate the performance of matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) by using 190 Catalase-negative Gram-Positive Cocci (GPC) clinical isolates.Methods:All isolates were identified by conventional phenotypic tests following the proposed scheme by Ruoff and Christensen and MALDI-TOF MS (Bruker Daltonics, BD, Bremen, Germany). Two different extraction methods (direct transfer formic acid method on spot and ethanol formic acid extraction method) and different cut-offs for genus/specie level identification were used. The score cut-offs recommended by the manufacturer (≥ 2.000 for species-level, 1.700 to 1.999 for genus level and <1.700 no reliable identification) and lower cut-off scores (≥1.500 for genus level, ≥ 1.700 for species-level and score <1.500 no reliable identification) were considered for identification. A minimum difference of 10% between the top and next closest score was required for a different genus or species.MALDI-TOF MS identification was considered correct when the result obtained from MS database agreed with the phenotypic identification result.When both methods gave discordant results, the 16S rDNA orsodAgenes sequencing was considered as the gold standard identification method. The results obtained by MS concordant with genes sequencing, although discordant with conventional phenotyping, were considered correct. MS results discordant with 16S orsodA identification were considered incorrect.Results:Using the score cut-offs recommended by the manufacturer, 97.37% and 81.05% were correctly identified to genus and species level, respectively. On the other hand, using lower cut-off scores for identification, 97.89% and 94.21% isolates were correctly identified to genus and species level respectively by MALDI-TOF MS and no significant differences between the results obtained with two extraction methods were obtained.Conclusion:The results obtained suggest that MALDI-TOF MS has the potential of being an accurate tool for Catalase-negative GPC identification even for those species with difficult diagnosis asHelcococcus,Abiotrophia,Granulicatella, among others. Nevertheless, expansion of the library, especially including more strains with different spectra on the same species might overcome potential “intraspecies” variability problems. Moreover, a decrease of the identification scores for species and genus-level identification must be considered since it may improve the MALDI-TOF MS accuracy.

Changes of cortisol, triiodothyronine, and thyroxine content in the blood plasma of perch, ruff, rudd and bleak under the impact of agricultural runoff from the adjacent territories to the water bodies were studied. Under the long-term exposure to the contaminated water, the weight and size of fish their growth rates specimens significantly decreased. The hormonal responses to the polluted waters impact are species-specific and partly depend on the ecological features of the species. Under the chronic impact of the polluted water, the cortisol content in blood plasma decreases and triiodothyronine content increases as compared to the control. The fishes occurring in the surface water layers create the thyroxine reserve in blood, and fishes which prefer the near-bottom layers use it to increase the triiodothyronine content. After treatment of the fields close to the reservoir with pesticides, the ruff and rudd develop the active stress reaction, the cortisol and thyroxine content in blood significantly increases. According to the considered indicators, the bleak is the most resistant to agricultural runoffs or possesses the ability to avoid toxic water pollution.

La importancia de los modelos de simulación para el monitoreo de los recursos naturales cada vez es mayor. El objetivo del presente estudio fue aplicar el modelo SWAT (Soil Water Assessment Tool) para evaluar el impacto del cambio de uso de suelo sobre el escurrimiento y sedimentos en la cuenca del Río Chapingo de 1999 a 2015. Se generaron dos mapas de uso de suelo mediante la fotointerpretación de imágenes Landsat y RapidEye con resoluciones de 15 y 5 m, respectivamente y recorridos de campo para verificación. Ante la falta de datos de escurrimiento confiable y reciente se utilizaron los registros mensuales de los períodos 1964-1970 y 1971-1975 para la calibración y validación del modelo. En este proceso los coeficientes de Nash-Sutcliffe fueron de 0.58 y 0.52, respectivamente. El uso de suelo de la cuenca en el período de estudio cambió: agricultura (-6.9 %) zona urbana (+2.89%), área de minería (+5.04 %) y pastizal (+7.75 %). La aplicación del SWAT calibrado y validado sólo para escurrimiento para el período 1999-2015 no detectó cambios en el escurrimiento (p = 0.2351) y sedimentos (p = 0.4430) en función del cambio de uso de suelo determinado. La correlación entre el escurrimiento y sedimentos anuales simulados con SWAT fue significativa (p = 0.0116) con un R2 bajo (0.36), atribuible a la falta de datos de campo de sedimentos para calibrar y validar el SWAT, además de la presencia de acciones de conservación de suelo y agua en la cuenca estudiada.

Background:Gout is the most common inflammatory arthritis resulting from a chronic deposition of MSU crystals in the joints and other soft tissues. After the process of repeated tissue damage and repair due to gout, tophi could be formed around the affected joints. Intra-articular tophi may sometimes result in bone destruction, joint deformities, and dysfunction which can adversely affect the patient’s quality of life. Furthermore, early-onset tophaceous gout patients are more likely to develop renal dysfunction, however, few studies have investigated if the presence of tophi is related with the progression of renal dysfunction in gout patients.Objectives:We aimed to compare clinical characteristics of patients with and without tophi at the time of the diagnosis of gout and investigate the effect of tophi on the renal function in gout patients.Methods:Data of 257 patients who were first diagnosed with gout at the Kangwon National University Hospital from January 2012 to December 2018 were retrospectively studied. Patients were divided into 2 groups according to the presence of tophi at the diagnosis. We compared clinical characteristics and the progression of renal dysfunction between the two groups.Results:Of all patients, 66 (25.5%) initially presented with tophi. Patients with tophi were older, had a longer duration of symptoms, and had a higher prevalence of multiple joint involvement than those without tophi. The decline in the eGFR was more prominent in patients with tophi than in those without (-4.8±14.5 ml/min/1.73m2vs. -0.7±11.9 ml/min/1.73m2, respectively; P=0.039). In multivariate logistic regression analysis, a prolonged symptom duration (odds ratio [OR], 1.010; 95% confidence interval [CI], 1.004–1.017; P=0.001) and multiple joint involvement (OR, 3.027; 95% CI, 1.831–5.004; P<0.001) were significantly associated with increased risk of formation of tophi. The presence of tophi was significantly associated with a rapid decline in the eGFR (β=-0.141; P=0.035).Conclusion:A prolonged symptom duration and multiple joint involvement were independent risk factors for tophi as the presenting symptom in gout patients. The presence of tophi was associated with a declining renal function. Therefore, an early diagnosis and active treatment are important in tophaceous gout.References:[1]Bardin T, Richette P.
