Mode shapes estimated from the vehicle responses are normally used to detect bridge damage efficiently for their high spatial resolution. However, an updated baseline finite element model (FEM) is normally required to quantify damages for such an approach. A two-stage damage detection procedure is presented for bridges by utilizing the mode shape estimated from a moving vehicle. Damage locations are first determined through a damage localization index (DLI) defined by regional mode shape curvature (RMSC). Then the relationship between the damage extents and the RMSC changes is investigated by FEM simulation. Finally, an equation set to quantify the single and multiple damages is deduced by combining the RMSCs and the relationship between the damage extents and the RMSC changes established by an un-updated FEM. Numerical and experimental examples are carried out to verify the validity and efficiency of the two-stage method. The results revealed that it can localize and quantify damages with satisfactory precision by using the response measured from one sensor only.

Mesenchymal stem cells (MSCs) have been increasingly tested in cell-based therapy to treat numerous diseases. Genetic modification to improve MSC behavior may enhance posttransplantation outcome. This study aims to test the potential therapeutic benefits of rat bone marrow MSCs overexpressing hypoxia-inducible factor 2α (rMSCsHIF-2α) in a rat hindlimb ischemia model. PBS, rMSCs, or rMSCsHIF-2α were injected into rat ischemic hindlimb. Compared with the injection of PBS or rMSCs, transplantation of rMSCsHIF-2α significantly improved blood perfusion, increased the number of vessel branches in the muscle of the ischemic hindlimb, and improved the foot mobility of the ischemic hindlimb (all P<0.05). rMSCHIF-2α transplantation also markedly increased the expression of proangiogenic factors VEGF, bFGF, and SDF1 and Notch signaling proteins including DII4, NICD, Hey1, and Hes1, whereas it reduced the expression of proapoptotic factor Bax in the muscle of the ischemic hindlimb. Overexpression of HIF-2α did not affect rMSC stemness and proliferation under normoxia but significantly increased rMSC migration and tube formation in matrigel under hypoxia (all P<0.05). RMSCsHIF-2α stimulated endothelial cell invasion under hypoxia significantly (P<0.05). Genetic modification of rMSCs via overexpression of HIF-2α improves posttransplantation outcomes in a rat hindlimb ischemia model possibly by stimulating proangiogenic growth factors and cytokines.

Mesenchymal stem cells (MSCs) can be used in adult stem cell-based gene therapy for vascular diseases. To test the hypothesis that MSCs alone or endothelial nitric oxide synthase (eNOS)-modified MSCs can be used for treatment of erectile dysfunction (ED), syngeneic rat MSCs (rMSCs) were isolated, ex vivo expanded, transduced with adenovirus containing eNOS, and injected into the penis of aged rats. Histological analysis demonstrated that rMSCs survived for at least 21 days in corporal tissue after intracavernous injection, and an inflammatory response was not induced. Intracavernous administration of eNOS-modified rMSCs improved the erectile response in aged rats at 7 and 21 days after injection. The increase in erectile function was associated with increased eNOS protein, NOS activity, and cGMP levels. rMSCs alone increased erectile function of aged rats at day 21, but not at day 7, with the transplanted cells exhibiting positive immunostaining for several endothelial and smooth muscle cell markers. This change in rMSC phenotype was accompanied by upregulation of penile eNOS protein expression/activity and elevated cGMP levels. These findings demonstrate that an adenovirus can be used to transduce ex vivo expanded rMSCs to express eNOS and that eNOS-modified rMSCs improve erectile function in the aged rat. Intracavernous injection of unmodified wildtype rMSCs improved erectile function 21 days after injection through mechanisms involving improved endothelium-derived NO/cGMP signaling and rMSC differentiation into penile cells expressing endothelial and smooth muscle markers. These data highlight the potential clinical use of adult stem cell-based therapy for the treatment of ED.

Aim: Biotechnological culture of hypoxia-conditioned (CH) rat mesenchymal stem cells (rMSC-CH) for testicular failure therapy with low libido improves the functional outcome of the testicle for producing spermatogenic cells and repairs Leydig cells in rats (Rattus norvegicus). Materials and Methods: In the first group (T1), rats with testicular failure and low libido were injected with normoxia-conditioned (CN) rMSCs (21% oxygen); in the second group (T2), rats with testicular failure and low libido were injected with rMSC-CH (1% oxygen); in the negative control group (T–), rats with normal testis were injected with 0.1 mL phosphate-buffered saline (PBS); and in the sham group (TS), rats with testicular failure and low libido were injected with 0.1 mL of PBS. Results: Vascular endothelial growth factor expression, as the homing signal, in the groups T2, T–, T1, and TS was 2.00±0.5%, 2.95±0.4%, 0.33±0.48%, and 0±0%, respectively. The number of cluster of differentiation (CD)34+ and CD45+ cells in the groups T– and TS was <20%, whereas that in T1 and T2 groups was >30% and >80%, respectively, showing the mobilization of hematopoietic stem cells (HSCs). The number of spermatogenic cells (spermatogonia, primary spermatocytes, secondary spermatocytes, and spermatid) decreased significantly (p<0.05) in TS compared with that in T–, T1, and T2, whereas that in T2 did not show a significant (p>0.05) decrease compared to that in T–. The improvement in libido, based on the number of Leydig cells producing the hormone testosterone for libido expression, did not increase in T1, whereas T2 was able to maintain the number of Leydig cells significantly compared to that between TS and T1. Conclusion: rMSC-CH culture for testicular failure with low libido showed improvement in the functional outcome of the testicle and in repairing Leydig cells.

In our previous study, we discussed the possibility of differentiation activity measurement for rat mesenchymal stem cells (RMSC) by Dielectrophoretic (DEP) levitation. Consequently, it was found that the differentiation activity of the RMSC could be evaluated by DEP levitation without the differentiation induction. Thus, we discuss the possibility of differentiation activity evaluation by DEP levitation with cells other than the RMSC. Human mesenchymal stem cells (HMSC) and human adipose tissue-derived stem cells (ASC) were used as the sample cells. The dielectric characteristics (Re[K(ω)]) measurement, the Re[K(ω)] of both the HMSC and the ASC decreased with the increasing passage number. Moreover, to evaluate the differentiation activity of the HMSC and the ASC that had performed the osteoblast differentiation induction, the amount of Alkaline Phosphatase (ALP) was measured. Consequently, the ALP activity of both the HMSC and ASC decreased with increasing the passage number. Therefore, it was found that the differentiation activity of the HMSC and the ASC could be evaluated by measuring the Re[K(ω)] due to the relationship between the Re[K(ω)] and ALP activity.

Abstract The aim of this study was to compare the efficacy of attachment of arginine-glycine-aspartic acid (RGD) peptide to titanium surfaces oxidized by different methods. Titanium surfaces were treated as follows: (1) treatment A: passivation in nitric acid, (2) treatment B: heated in air at 400°C for 1 hour, (3) treatment C: immersed in 8.8 M H2O2/0.1 M HCl at 80°C for 30 minutes, and (4) treatment D: treated as in treatment C and then heated at 400°C for 1 hour. RGD was attached to titanium samples treated as in treatments A through D. The quantity of attached RGD was determined by an enzyme-linked immunoabsorbent assay. Mineralization of a rat bone marrow stromal cell (RMSC) culture on the titanium surfaces after 21 days was determined y atomic absorption spectroscopy. The treatments were ranked according to quantity of RGD attached as C, A, B, and D. Twenty-one days after RMSC culture, the degree of mineralization was significantly higher for treatment C than for treatments A, B, and D and for controls. The efficacy of RGD attachment varies with the oxidation treatment given to titanium. Oxidation in H2O2/0.1 M HCl at 80°C provided the best overall surface for RGD attachment as well as calcified matrix formation of RMSCs.

Birds play a vital and indispensable role in biodiversity and environmental conservation. Protecting bird diversity is crucial for maintaining the balance of nature, promoting ecosystem health, and ensuring sustainable development. The Broad Learning System (BLS) exhibits an excellent ability to extract highly discriminative features from raw inputs and construct complex feature representations by combining feature nodes and enhancement nodes, thereby enabling effective recognition and classification of various birdsongs. However, within the BLS, the selection of feature nodes and enhancement nodes assumes critical significance, yet the model lacks the capability to identify high quality network nodes. To address this issue, this paper proposes a novel method that introduces residual blocks and Mutual Similarity Criterion (MSC) layers into BLS to form an improved BLS (RMSC-BLS), which makes it easier for BLS to automatically select optimal features related to output. Experimental results demonstrate the accuracy of the RMSC-BLS model for the three construction features of MFCC, dMFCC, and dsquence is 78.85%, 79.29%, and 92.37%, respectively, which is 4.08%, 4.50%, and 2.38% higher than that of original BLS model. In addition, compared with other models, our RMSC-BLS model shows superior recognition performance, has higher stability and better generalization ability, and provides an effective solution for birdsong recognition.

Reconstitution of secretory IgA (S-IgA) by the association in vitro of secretory component (SC) and polymeric IgA (pIgA) obtained from hybridomas is a valuable tool in the study of the structure-function relationship in this particular class of antibody. Although dimeric IgA (dIgA) can be obtained and purified from hybridoma clones, SC remains tedious to isolate in sufficient amounts from colostral milk. Several murine models for the study of mucosal immunity are available, which could potentially benefit from the use of cognate IgA antibodies in various molecular forms, including dIgA and S-IgA. We report here on the establishment of two expression systems allowing the production of milligram amounts of pure recombinant murine SC (rmSC) with preserved murine pIgA-binding capability. The first system relies on the use of recombinant vaccinia virus to prompt infected HeLa cells to express the murine SC protein, whereas the second system is based on a stably transfected cell clone exhibiting murine glycosylation. The second source of rmSC will permit the study of the role of its sugar moieties in pathogen-host interactions, and the evaluation of its function in passive protection without risking adverse immune responses. The extensive biochemical characterization conducted in this study demonstrates that rmSC is a dependable and convenient alternative to the natural product, and indicates that the J chain is dispensable in the recognition of pIgA and SCin vitro, whereas it is required for proper pIgA-polymeric Ig receptor interaction in vivo.

Summary Objectives The aim was to evaluate the effects of mesenchymal stem cell (MSC) transfer to periodontal ligament (PDL) on the inhibition and/or repair of orthodontically induced root resorption (OIRR) during and after arch expansion and on the orthodontic tooth movement (OTM) rate of the maxillary first molar teeth of rats. Material and methods Sixty Wistar rats were divided into three groups as the untreated group, MSC and control injections during the expansion period group (EMSC-EC), and MSC and control injections at the retention period group (RMSC-RC). Fifty grams of orthodontic force was applied to the maxillary first molar teeth of the rats for 14 days in the vestibular direction, and then, 20 days of retention was carried out. MSCs and control injections were performed every 3 days in the EC, RC, EMSC, and RMSC groups. At the end of the experiment, samples were prepared for OTM evaluation, mRNA expression analysis, micro-computed tomography measurements, cementum thickness calculations, and structural examinations. Results The amount of OTM in EMSC group was significantly higher than in EC group (P &lt; 0.001). MSC transfer during the expansion and retention periods reduced the number of resorption lacunae, volumetric and linear resorptive measurements, and cyclooxygenase-2 and receptor activator of nuclear factor kappa B ligand (RANKL) mRNA expression levels, and increased the osteoprotegerin (OPG) expression levels, OPG/RANKL ratio, and cementum thickness in the EMSC and RMSC groups. Conclusions MSC transfer to PDL during expansion increased the amount of OTM. Injection of MSC during the retention period was found to be slightly more effective in prevention and/or repair of OIRR than MSC transfer during the expansion period.

The spontaneous expression of neural markers by mesenchymal stem cells (MSCs) has been considered to be a demonstration of MSCs’ predisposition to differentiate towards neural lineages. In view of their application in cell therapy for neurodegenerative diseases, it is very important to deepen the knowledge about this distinctive biological property of MSCs. In this study, we evaluated the expression of neuronal and glial markers in undifferentiated rat MSCs (rMSCs) at different culture passages (from early to late). rMSCs spontaneously expressed neural markers depending on culture passage, and they were coexpressed or not with the neural progenitor marker nestin. In contrast, the number of rMSCs expressing mesengenic differentiation markers was very low or even completely absent. Moreover, rMSCs at late culture passages were not senescent cells and maintained the MSC immunophenotype. However, their differentiation capabilities were altered. In conclusion, our results support the concept of MSCs as multidifferentiated cells and suggest the existence of immature and mature neurally fated rMSC subpopulations. A possible correlation between specific MSC subpopulations and specific neural lineages could optimize the use of MSCs in cell transplantation therapy for the treatment of neurological diseases.

Recently, dense retrieval (DR) models, which represent queries and documents with fixed-width vectors and retrieve relevant ones via nearest neighbor search, have drawn increasing attention from the IR community. However, previous studies have shown that the effectiveness of DR critically relies on sufficient training signals, which leads to severe performance degradation when applied in out-of-domain scenarios, where large-scale training data are usually unavailable. To solve this problem, existing studies adopt a data-augmentation-plus-joint-training paradigm to construct weak/pseudo supervisions on the target domain and combine them with the large-scale human annotated data on the source domain to train the DR models. However, they don't explicitly distinguish the data and the supervision signals in the training process and simply assume that the DR models are mighty enough to capture and memorize different domain knowledge and relevance matching patterns without guidance, which, as shown in this paper, is not true. Based on this observation, we propose a Robust Multi-Supervision Combining strategy (RMSC) that decouples the domain and supervision signals by explicitly telling the DR models how the domain data and supervision signals are combined in the training data with specially designed soft tokens. With the extra soft tokens to store the domain-specific and supervision-specific knowledge, RMSC allows the DR models to conduct retrieval based on human-like relevance matching patterns and target-specific language distribution on the target domain without human annotations. Extensive experiments on zero-shot DR benchmarks show that RMSC significantly improves the ranking performance on the target domain compared to strong DR baselines and domain adaptation methods, while being stable during training and can be combined with query generation or second-stage pre-training.

A pós-graduação em Saúde Coletiva investe na força de trabalho do SUS, visando contribuir para a promoção da saúde da população. Objetivou-se descrever os significados atribuídos por pós-graduandos sobre Competências em Promoção da Saúde. Estudo de natureza descritiva com abordagem quantitativa, com 34 estudantes dos cursos de Residência Multiprofissional em Saúde Coletiva (RMSC) e Mestrado Profissional em Saúde da Família (MPSF) no Ceará. Utilizou-se a Técnica de Associação Livre de Palavras com o termo indutor “Competências para Promoção da Saúde”. Análises prototípica (AP) e de similitude (AS) das evocações foram discutidas à luz da matriz de Competências Essenciais para Promoção da Saúde no contexto brasileiro (CompEPS). Os termos ‘conhecimento’ e ‘habilidade’ foram evidenciados com elevada frequência na AP, compondo o núcleo central de ambos os cursos e, na AS, os termos “cuidado”, “promoção” e “prevenção” foram destaque no MPSF, enquanto “humanização” e “ética” destacaram-se na RMSC. Assim, os significados atribuídos pelos pós-graduandos ora ampliaram ora se distanciaram da CompEPS, implicando compreensões dissonantes e requerendo melhorias na formação pós-graduada.

Mounting evidences have indicated that terminal differentiation-induced lncRNA (TINCR) contributes to various cellular processes, such as proliferation, apoptosis, autophagy, migration, invasion, and metastasis. However, the function of TINCR in regulating migration of MSCs is largely unknown. In this study, the effects of TINCR on the migration of rat MSCs from the bone marrow were studied by Transwell assays and wound healing assays. Our results suggested that TINCR positively regulated migration of rMSCs. miR-761 mimics suppressed rMSC migration, whereas miR-761 inhibitor promoted migration. Target prediction analysis tools and dual-luciferase reporter gene assay identified Wnt2 as a direct target of miR-761. miR-761 could inhibit the expression of Wnt2. Further, the investigation about the function of TINCR in miR-761-induced migration of rMSCs was completed. These results demonstrated that TINCR took part in the regulation of miR-761-induced migration in rMSCs through the regulation of Wnt2 and its Wnt2 signaling pathway. Taken together, our results demonstrate that lncRNA-TINCR functions as a competitive endogenous RNA (ceRNA) to regulate the migration of rMSCs by sponging miR-761 which modulates the role of Wnt2. These findings provide evidence that lncRNA-TINCR has a chance to serve as a potential target for enhancing MSC homing through the miR-761/Wnt2 signaling pathway.

Generally, multimodality data contain different potential information available and are capable of providing an enhanced analytical result compared to monosource data. The way to combine the data plays a crucial role in multimodality data analysis which is worth investigating. Multimodality clustering, which seeks a partition of the data in multiple views, has attracted considerable attention, for example, robust multiview spectral clustering (RMSC) explicitly handles the possible noise in the transition probability matrices associated with different views. Spectral clustering algorithm embeds the input data into a low-dimensional representation by dividing the clustering problem into k subproblems, and the corresponding eigenvalue reflects the loss of each subproblem. So, the eigenvalues of the Laplacian matrix should be treated differently, while RMSC regularizes each singular value equally when recovering the low-rank matrix. In this paper, we propose a multimodality clustering algorithm which recovers the low-rank matrix by weighted nuclear norm minimization. We also propose a method to evaluate the weight vector by learning a shared low-rank matrix. In our experiments, we use several real-world datasets to test our method, and experimental results show that the proposed method has a better performance than other baselines.

This study explores further the impacts of the relationship between autumntime mean sea level pressure (MSLP) over the Indian Ocean and autumntime precipitation over Tasmania. We employ a combination of cross-correlation analysis, Singular Value Decomposition (SVD), and covariance measures to unravel this intricate connection. Our results reveal that MSLP over the Indian Ocean significantly influences Tasmania’s autumn precipitation, particularly in the region extending from 125°E eastward and from 35°S southward. This MSLP-precipitation link is statistically significant, characterized by a Pearson’s coefficient of correlation of 0.63 (p < 0.05). In addition, we calculate the Root Mean Square Covariance (RMSC) to gauge the strength of this association. The RMSC value, calculated at 0.33, surpasses the threshold of 0.1, emphasizing a robust and significant connection between autumntime MSLP and precipitation in Tasmania. Our results emphasize that the first leading mode of SVD accounts for 84% of the variability in both MSLP over the Indian Ocean and precipitation in Tasmania. These results contribute to a more profound understanding of the factors governing autumn precipitation in Tasmania and offer valuable insights for meteorological and climate science.

Abstract The aim of this study was to evaluate injectable, in situ cross-linkable elastin-like recombinamers (ELRs) for osteochondral repair. Both the ELR-based hydrogel alone and the ELR-based hydrogel embedded with rabbit mesenchymal stromal cells (rMSCs) were tested for the regeneration of critical subchondral defects in 10 New Zealand rabbits. Thus, cylindrical osteochondral defects were filled with an aqueous solution of ELRs and the animals sacrificed at 4 months for histological and gross evaluation of features of biomaterial performance, including integration, cellular infiltration, surrounding matrix quality and the new matrix in the defects. Although both approaches helped cartilage regeneration, the results suggest that the specific composition of the rMSC-containing hydrogel permitted adequate bone regeneration, whereas the ELR-based hydrogel alone led to an excellent regeneration of hyaline cartilage. In conclusion, the ELR cross-linker solution can be easily delivered and forms a stable well-integrated hydrogel that supports infiltration and de novo matrix synthesis.

Abstract Abstract 4476 Introduction The therapeutic effects of using mesenchymal stem cells (MSCs) for transplantation in the treatment of spinal cord injury have been previously studied. Astragalus propinquus, also known as Huangqi, is a traditional Chinese herb commonly used to strengthen the immune system and protect neuron. However, the possibility of combining the beneficial effects of both MSC transplantation and Huangqi Injecta in treating spinal cord injury has not been addressed. Methods A total of 120 male, four-week-old, Wistar rats, were randomly divided into six groups: sham group, model group, phosphate buffered saline (PBS) solution control group (PBS group), Huangqi injection control group (Huangqi group), rMSCs transplantation group (rMSCs group), rMSCs and Huangqi injecta treated group (rMSCs + Huangqi group). There were 10 rats in the sham group and 22 rats in the other groups. In the PBS group, rats were injected 5 μl PBS into the cephalic site of spinal cord injured section three days after the operation; in the Huangqi group, rats were intraperitoneally injected with Huangqi Injecta at a dose of 1.2 ml per 100 g of body weight on day three after the operation; in the rMSCs group, 5 μl of rMSC suspension (2.5–5×105 cells) was injected into the cephalic site of the spinal cord injured part in rats on day three after the operation; in the rMSCs + Huangqi group, rats were injected Huangqi Injecta and 5 μl rMSC suspension (2.5–5×105 cells) following the methods mentioned in the Huangqi group and the rMSCs group. Six animals were randomly picked from each groups on day 7, 14, 21 and 28 to receive Basso-Beattie-Bresnahan (BBB) scale grading for motor function checking of the hind legs. HE staining is adopt for the histopathologic examination, and Dual-marked immunohistochemical analysis is applied for cell locating and differentiation determination. Results The average BBB score in the Huangqi group is higher than model group and PBS group, except on day 21. The score in rMSCs group and the rMSCs + Huangqi group is higher than that in the model group, PBS group and the Huangqi group. The rMSCs + Huangqi group has a higher score than the Huangqi group at each time point. Also, except on day 14, the score for the rMSCs + Huangqi group was higher than the rMSCs group, and the difference was significant. HE staining of the spinal cord paraffin sections showed that the degree of degeneration and necrosis in nerve cells was alleviated in the rMSCs and rMSCs + Huangqi groups, compared with the PBS and Huangqi groups. Edema and infiltration of inflammatory cells were obviously reduced, and the proliferation of glial cells and integrated nerve cells was detected in the rMSCs and rMSCs + Huangqi groups. Moreover, the proliferation of glial cells was more active in the rMSCs + Huangqi group than in the rMSCs group. Using immunohistochemical analysis, both in the rMSCs group and the rMSCs + Huangqi group, BrdU marked rMSCs survived in the spinal cord and accumulated in the gray matter more than in the white matter. GFAP + BrdU and BrdU+NF-M positive cells were detected from day 7 after transplantation. The quantity of BrdU + GFAP positive cells in the rMSCs group and rMSCs + Huangqi group is 6.35±1.14 and 8.75±1.16 per visual field relatively, while BrdU + NF-M positive cells in the rMSCs group and rMSCs + Huangqi group is 1.60±0.75 and 3.30±0.98 per visual field relatively. It indicated that the quantity of BrdU + GFAP and BrdU + NF-M positive cells per visual field observed in the spinal cord paraffin section of the rMSCs + Huangqi group, was significantly higher than that in the rMSCs group (P<0.05). Conclusions These findings indicated that MSCs transplantation has the capability of nerve restoration and recovery after spinal cord injury, and it could be enhanced by Huangqi Injecta. The strategy of combining Huangqi Injecta and MSCs, aimed to promote transplanted cell differentiation and their tissue repair capability, provided a simple yet effective way of optimizing cell transplantation therapy. Abstract Here we report the in vitro and in vivo properties of a spontaneously transformed nude rat mesenchymal stem cells (rTMSCs) following culture passages 23. When this cell line was compared with early passages of mesenchymal cells (rMSCs) from the same animal, no apparent differences could be seen with regard to the phenotype and Gap junctions’ connexin 43. However, the telomerase activity in rTMSCs is lower than in the rMSCs. Karyotyping of rTMSC gave a trisome 6 phenotype. Microarry analysis of rTMSC shows over expression of the transforming growth factor family including TGF-beta and BMp6. Both rMSC and rTMSC had equal capability to differentiate into adipocytic and “neural like” linage. In contrast rTMSC was unable to differentiate into bone and only this cell line form spheroids in cultures. Subcutaneously injection of rTMSC in doses down to 2x 104 into nude/nude mice formed tumor in all animals tested. Examination of the tumor assembles morphology like an immature sarcoma. When 100–200 SP + cells were administered to the animals, tumor could also be formed. After intravenous injections and direct injections into left heart ventricle all animals developed metastasis in the lung; abdominal cavity; bone and skin. The rTMSC has been stabled marked with reporter gene GFP and Leucipherase. Currently the proliferation and migration capacity of the linage-differentiated rTMSC is studied in vivo by imaging and these results will be presented. We conclude that our normal and transformed rMSCs could be potential useful for further biological studies on mesenchymal cells both in vitro and in vivo.

Sampling subnet is an important topic of complex network research. Sampling methods influence the structure and characteristics of subnet. Random multiple snowball with Cohen (RMSC) process sampling which combines the advantages of random sampling and snowball sampling is proposed in this paper. It has the ability to explore global information and discover the local structure at the same time. The experiments indicate that this novel sampling method could keep the similarity between sampling subnet and original network on degree distribution, connectivity rate and average shortest path. This method is applicable to the situation where the prior knowledge about degree distribution of original network is not sufficient.

The efficacy of cell therapy is limited by low retention and survival of transplanted cells in the target tissues. In this work, we hypothesize that pharmacological preconditioning with celastrol, a natural potent antioxidant, could improve the viability and functions of mesenchymal stromal cells (MSC) encapsulated within an injectable scaffold. Bone marrow MSCs from rat (rMSC) and human (hMSC) origin were preconditioned for 1 hour with celastrol 1 μM or vehicle (DMSO 0.1% v / v), then encapsulated within a chitosan-based thermosensitive hydrogel. Cell viability was compared by alamarBlue and live/dead assay. Paracrine function was studied first by quantifying the proangiogenic growth factors released, followed by assessing scratched HUVEC culture wound closure velocity and proliferation of HUVEC when cocultured with encapsulated hMSC. In vivo, the proangiogenic activity was studied by evaluating the neovessel density around the subcutaneously injected hydrogel after one week in rats. Preconditioning strongly enhanced the viability of rMSC and hMSC compared to vehicle-treated cells, with 90% and 75% survival versus 36% and 58% survival, respectively, after 7 days in complete media and 80% versus 64% survival for hMSC after 4 days in low serum media ( p < 0.05 ). Celastrol-treated cells increased quantities of proangiogenic cytokines compared to vehicle-pretreated cells, with a significant 3.0-fold and 1.8-fold increase of VEGFa and SDF-1α, respectively ( p < 0.05 ). The enhanced paracrine function of preconditioned MSC was demonstrated by accelerated growth and wound closure velocity of injured HUVEC monolayer ( p < 0.05 ) in vitro. Moreover, celastrol-treated cells, but not vehicle-treated cells, led to a significant increase of neovessel density in the peri-implant region after one week in vivo compared to the control (blank hydrogel). These results suggest that combining cell pretreatment with celastrol and encapsulation in hydrogel could potentiate MSC therapy for many diseases, benefiting particularly ischemic diseases.

In order to effectively evaluate shale gas reservoirs with low porosity, extra-low permeability, and no natural productivity, dipole array acoustic logging, which can provide various types of information including P-wave slowness (DTC) and S-wave slowness (DTS), is widely used. As the dipole array acoustic logging tool has a larger investigation depth and is suitable for complex borehole environments, such as those with a high wellbore temperature, high drilling fluid column pressure, or irregular borehole wall, it has been mainly applied to the evaluation of lithology, gas potential, fractures, and stimulation potential in shale gas reservoirs. The findings from a case study of the Sichuan Basin in China reveal that the acoustic slowness, S-P wave slowness ratio (RMSC), and S-wave anisotropy of the dipole array acoustic logging can be used to qualitatively identify reservoir lithology, gas potential, and fractures. Using the relationship between DTC and the total porosity of shale gas reservoirs, and combined with the compensated neutron (CNL) and shale content (Vsh) of the reservoir, a mathematical model for accurately calculating the total porosity of the shale gas reservoir can be established. By using the relationship between the RMSC and gas saturation in shale gas reservoirs and tied with density log (DEN), a mathematical model of gas saturation can be established, and the determination of gas saturation by the non-resistivity method can be achieved, delivering a solution to the issue that the electric model is not applicable under low resistivity conditions. The DTS, DTC, and DEN of shale can be used to calculate rock mechanic parameters such as the Poisson’s ratio (POIS) and Young’s modulus (YMOD), which can be used to evaluate the shale stimulation potential.

The main features of shallow cracks fracture are considered, and a brief analysis of methods allowing to predict the temperature dependence of the fracture toughness KJC (T) for specimens with shallow cracks is given. These methods include DA-method, (JQ)-method, (J-T)-method, “local methods” with its multiparameter probabilistic approach, GP method uses power approach, and also two engineering methods – RMSC (Russian Method for Shallow Crack) and EMSC (European Method for Shallow Crack). On the basis of 13 sets of experimental data for national and foreign steels, a detailed verification and comparative analysis of these two engineering methods were carried out on the materials of the VVER and PWR nuclear reactor vessels considering the effect of shallow cracks.

Purpose – The purpose of this paper is to examine the relationship between the relational self-identity and prosocial silence in the context of the Egyptian public sector. It also examines the mediating effect of relational maintenance strategies among co-workers (RMSC) on the aforementioned relationship. Design/methodology/approach – The study uses a questionnaire that consists of measures adopted from existing and tested scales. A parallel mediation analysis is conducted using Sobel test for specific indirect effects in regression analysis. Findings – The findings reveal that high relational employees do not engage in prosocial silence unless they are involved in networking and shared tasks strategies. Originality/value – Prosocial silence has received limited attention despite its drastic effects in employee well-being. This study provides initial insights concerning the dynamics of social behaviours which motivate employees to avoid speaking up in order to not embarrass a workplace friend.

Then, most adequate and appropriate model is selected by analyzing diagnostics checks. Later on, by comparing values of Akaike Information Criterion (AIC), Schwarz Information Criterion (SIC), and Standard Error (S.E.) of Regression, Root Mean Square Error (RMSC) and Theil Inequality Coefficient (TIC) for each model, forecast model has been finalized. In the end, forecasts have been made using models and compared these forecast values with the actual values for 2010 in order to check the accuracy of the model.</p>

O distanciamento da Psicologia das demandas das populações do campo é um fato histórico. Mesmo com a sua inserção no contexto das políticas públicas e reorientação da formação a partir das novas diretrizes curriculares, o olhar dessa ciência e profissão manteve-se restrito às demandas do contexto urbano. Em 2011, com a aprovação da Política Nacional de Saúde Integral das Populações do Campo, da Floresta e das Águas (PNSIPCFA), as diferentes profissões da Saúde ganham um marco legal de estímulo à formação de profissionais sensíveis às especificidades dessas populações. A primeira Residência Multiprofissional em Saúde da Família com ênfase na Saúde da População do Campo (RMSC) surge, então, com a missão de formar profissionais de saúde a partir das necessidades e especificidades das populações do campo. O objetivo desse trabalho é refletir sobre a experiência da formação e atuação das (os) residentes de Psicologia inseridas (os) nesse programa, entendendo que a inserção da Psicologia nessa experiência pioneira lança novos olhares para a consolidação tanto da Psicologia Rural, quanto da PNSIPCFA no país.

Road maintenance systems (RMS) are crucial for maintaining safe and efficient road networks. The impact of climate change on road maintenance systems is a concern as it makes them more susceptible to weather events and subsequent damages. To tackle this issue, we propose an RMSDC (Road Maintenance Systems Using Deep Learning and Climate Adaptation) technique to improve road maintenance systems based on Deep learning and Climate Adaptation. RMSDC aims to use the multivariate classification technique and divides the dataset into training and test datasets. The RMSDC combines Convolutional Long Short-Term Memory (ConvLSTM) techniques with road weather information and sensor data. However, in emerging nations, the effects of climate change are already apparent, which makes road networks particularly susceptible to extreme weather, floods, and landslides. Therefore, climate adaptation of road networks is essential, especially in developing nations with limited financial resources. To address this issue, we propose an intelligent and effective RMSDC that utilizes deep learning algorithms based on climate change predictions. The ConvLSTM block effectively captures the relationship between input features over time to calculate the root-mean deviation (RMSD). We evaluate RMSDC performance against frameworks for downscaling climate variables using two metrics: root-mean-square error (RMSE) and mean absolute difference. Through real evaluations, RMSDC consistently outperforms approaches with a reduced RMSE of 0.26. These quantitative results highlight how effective RMSDC is in addressing maintenance systems on road networks leading to proactive road maintenance strategies that enhance traffic safety, reduce costs, and improve environmental sustainability.

The objective of the present study is to evaluate the proteinaceous digestibility of crude flours and derived protein rich product of three varieties of Mucuna pruriens of the male albino rats in growth. 48 rats have been taken randomly split up into 8 groups where 6 each according to their formulated feedings; those foods are: foods without protein (RSP), foods with casein (RC), food with the black macuna derived protein rich product (RMNc), food with the scratch macuna derived protein rich product (RMRc), food with the white macuna derived protein rich product (RMBc), food with the black macuna crude flours (RMNf), food with the scratch macuna crude flours (RMRf) et food with the white macuna crude flours (RMBf). All those foods content the same quantity of azote excepted foods without protein. Rats have been isolated in the semi metabolic cages which allow gathering the remaining of the foods supplied and the faeces to determine the parameter values of consumption. The experiment has taken 28 days. The apparent and reel protein digestibilities obtained in each food are respectively : RC (76,63±0,36% ; 95,86±0,06%), RMNc (71,20±0,77% ; 91,60±0,99%), RMRc (73,68±0,23% ; 93,49±0,85%), RMBc (71,16±0,59% ; 92,03±0,70%), RMNf (33,25±1,91% ; 53,881±1,70%), RMRf (31,3±2,40% ; 52,12±2,22%) and RMBf (31,12±4,00% ; 51,92±3,8%). The results show that derived protein rich product have ameliorated the protein digestibility and could be the alternative for the use of seeds of Mucuna pruriens to feed human beings and animals.Int. The rms2 and rms4 pea (Pisum sativum L.) branching mutants have higher and lower xylem-cytokinin concentration, respectively, relative to wild type (WT) plants. These genotypes were grown at two levels of nitrogen (N) supply for 18–20 d to determine whether or not xylem-cytokinin concentration (X-CK) or delivery altered the transpiration and leaf growth responses to N deprivation. Xylem sap was collected by pressurising de-topped root systems. As sap-flow rate increased, X-CK declined in WT and rms2, but did not change in rms4. When grown at 5.0 mm N, X-CKs of rms2 and rms4 were 36% higher and 6-fold lower, respectively, than WT at sap-flow rates equivalent to whole-plant transpiration. Photoperiod cytokinin (CK) delivery rates (the product of transpiration and X-CK) decreased more than 6-fold in rms4. Growth of plants at 0.5 mm N had negligible (< 10%) effects on transpiration rates expressed on a leaf area basis in WT and rms4, but decreased transpiration rates of rms2. The low-N treatment decreased leaf expansion by 20–25% and expanding leaflet N concentration by 15%. These changes were similar in all genotypes. At sap-flow rates equivalent to whole-plant transpiration, the low N treatment decreased X-CK in rms2 but had no discernible effect in WT and rms4. Since the low N treatment decreased transpiration of all genotypes, photoperiod CK delivery rates also decreased in all genotypes. The similar leaf growth response of all genotypes to N deprivation despite differences in both absolute and relative X-CKs and deliveries suggests that shoot N status is more important in regulating leaf expansion than xylem-supplied cytokinins. The decreased X-CK and transpiration rate of rms2 following N deprivation suggests that changes in xylem-supplied CKs may modify water use.

Diabetes mellitus (DM) is a metabolic disorder characterized by elevated blood glucose levels. There are 2 types of DM where molecular-level diagnosis becomes very important because both have different treatments to avoid treatment errors. An electrochemical aptasensor as a type 2 DM detector with insulin target has been developed. This study aims to determine the interaction and specificity based on the values of RMSD, RMSF, and binding energy between aptamer and insulin when it reaches stability in silico compared to HbA1c and glucose. Docking simulations were performed on the HDOCK webserver and dynamics simulations for 1000 ns on the aptamer and protein molecular models used. The simulation results were analyzed to see the stability and visualized using VMD to see the conformation of the aptamer-ligand complex. The docking result showed higher binding energy between aptamer-insulin compared to other molecules, namely −221.87 kcal/mol. The results of RMSF and RMSD analysis of molecular dynamics simulations show that the system is stable, has the best binding energy value of −9.9510 kcal/mol. The aptamer complex with insulin showed better specificity compared to glucose and HbA1c based on RMSD, RMSF, and binding energy.

Introdução: O rabdomiossarcoma (RMS) é o tumor de tecidos moles mais comum da infância. Pode ser classificado em dois subtipos principais: o rabdomiossarcoma alveolar (RMSa) e o embrionário (RMSe). No RMSa, o prognóstico é desfavorável quando comparado ao RMSe, necessitando de tratamento intensificado; dessa forma, a distinção entre ambos os subtipos é fundamental. Citogeneticamente, o RMSa apresenta translocações cromossômicas envolvendo o gene FOXO1 em 80% dos casos. A metodologia de hibridização in situ por fluorescência (FISH) tem sido muito utilizada para caracterizar o RMSa. Relato do caso: Paciente do sexo feminino, com 7 anos de idade, apresentou ao diagnóstico RMSa parameníngeo, sem metástase ao diagnóstico. A análise por meio de FISH mostrou a translocação envolvendo o gene FOXO1 e uma cópia extra desse gene. A paciente foi incluída no protocolo de tratamento do EpSSG, classificada como grupo de alto risco e recebeu quimioterapia e radioterapia. No final do tratamento, foi observada resposta parcial e iniciada quimioterapia de segunda linha. Não houve resposta clinicorradiológica e a paciente evoluiu com progressão de doença local refratária ao tratamento e óbito após um ano do diagnóstico. Conclusão: De acordo com o nosso conhecimento, é a primeira descrição de um caso de RMSa apresentando a translocação do gene FOXO1 e uma cópia extra desse gene em clones separados. São necessários ainda novos estudos, a fim de compreender melhor o significado prognóstico da presença dessas alterações.

One knows that smart transport is also an integrated part of healthcare technologies. To minimize the pollution for benefiting the healthcare, the traffic throughput on roundabout intersection has to be increased which will reduce wasted time and will also enhance passenger comfort. This paper presents a new approach by use of cooperative vehicular control utilizing VANET without compromising the safety of vehicles. This intersection side unit (ISU)-based system use lane change mechanism. The modular use of lane with lane change in newly designed protocol CARA (collision avoidance at roundabout algorithm) will greatly enhance the capacity utilization of roundabout. A new simulator ‘RoundSim' was also developed exclusively for simulation in roundabout. A new MAC protocol RMAC (roundabout MAC) is also designed which will suit the roundabout management utilizing lane change to minimize sudden jerk to passengers, thus enhancing healthcare of people. This RMAC utilizes message set with different prioritization scheme which results in better utilization of allotted frequency spectrum.

Introdução: Os Programas de Residência Multiprofissional (PRM) em Saúde são mecanismos de cooperação intersetorial que visam a inserção de jovens profissionais da saúde qualificados para exercer suas profissões no mercado de trabalho, particularmente preparados para trabalhar e contribuir para o desenvolvimento e construção do SUS. O Programa de Residência em Saúde Coletiva com foco em gestão e planejamento em saúde permite projetar a habitual rotina reproduzida no dia a dia dos serviços de saúde, como antigas práticas da saúde pública tradicional para uma expectativa de inovação que modifique os modelos de cuidado e gestão em saúde. Objetivo: Avaliar a atuação, atividades, a interação e estrutura organizacional de um PRM. Metodologia: O estudo transversal foi realizado no Alto Sertão da Paraíba com amostra (n=29) participante do Programa de Residência Multiprofissional em Saúde Coletiva (tutores, residentes, preceptores e coordenadores). Esses profissionais responderam a um questionário autoaplicado que envolvia questões avaliativas no âmbito estrutural, organizacional e de atuação do PRM. A análise dos dados foi obtida a partir da frequência absoluta e relativa dos dados. Resultado: Os resultados mostraram dados quantitativos divididos em três diferentes dimensões do programa, a dimensão da gestão, da estrutura organizacional e do processo pedagógico. Quanto ao grau de envolvimento entre tutores, preceptores e IES na implantação do PRMSC, 50% consideraram que houve um envolvimento parcial na implantação. No quesito vínculo entre a IES com a RMSC, a totalidade dos participantes respondeu que esse vínculo se faz presente. Na vertente da participação dos residentes, tutores e preceptores nas avaliações sistemáticas do PRMSC, 42,9% consideraram que existe pouca participação. Sobre o processo de avaliação do PRMSC, 28,6% dos profissionais envolvidos consideraram que as avaliações são previstas e realizadas ao longo do programa, entretanto uma parcela semelhante de profissionais considerou que essas avaliações são previstas e não realizadas. A partir desses dados foi possível a identificação e discussão de fragilidades que devem ser superadas para o fortalecimento do PRMSC. Conclusão: Conclui-se que os instrumentos avaliativos são importantes para a identificação de pontos que precisam ser melhorados dentro do programa, utilizando-se da percepção da amostra inserida no programa para direcionamentos que visem promover melhorias nos pontos de fragilidades encontrados, possibilitando assim o fortalecimento e desenvolvimento do programa. Kanker payudara adalah tumor ganas yang terbentuk dari sel-sel payudara yang berkembang tanpa terkendali. Doxorubicin merupakan antibiotik golongan antrasiklin yang banyak digunakan untuk kemoterapi pada kanker payudara namun menyebabkan kardiotoksisitas. Tujuan penelitian ini adalah menemukan senyawa sebagai kandidat obat antikanker payudara yang berasal dari metabolit sekunder putik bunga saffron (Crocus sativus L) secara in silico. Analisis metabolit sekunder dilakukan dengan prediksi druglikeness, ADME-Tox, dan uji aktivitas metabolit sekunder dengan metode molecular docking menggunakan program AutoDock Vina dengan reseptor SIRT1 (PDB: 4I5I). Tahap selanjutnya dilakukan uji stabilitas dengan molecular dynamic menggunakan program CHARMM-GUI dan GROMACS yang terdiri dari RMSD, RMSF, jari-jari girasi, SASA dan ikatan hydrogen. Hasil molecular docking menunjukkan metabolit sekunder terbaik putik bunga saffron (Crocus sativus L) yaitu Scutellarein 7-methyl ether 6-glucoside (ΔG = -11,53 kkal/mol) terhadap senyawa pembanding doxorubicin (ΔG = -9,46 kkal/mol). Hasil interaksi ikatan senyawa Scutellarein 7-methyl ether 6-glucoside dengan reseptor 4I5I menunjukkan adanya ikatan hydrogen pada asam amino ASP A:348; ILE A:347; ikatan hidrofobik pada asam amino PHE A:273; PHE A:297. Senyawa Scutellarein 7-methyl ether 6-glucoside selama simulasi menunjukkan hasil RMSD yang relatif stabil dengan selisih nilai RMSD pada kisaran 0,15-0,3 Å; nilai RMSF; nilai jari-jari girasi; nilai SASA, ikatan hidrogen yang relatif stabil tanpa adanya fluktuasi selama waktu simulasi berjalan hingga 20ns. Diharapkan pada penelitian lebih lanjut dilakukan uji in vitro terhadap senyawa Scutellarein 7-methyl ether 6-glucoside yang berpotensi sebagai aktivator SIRT1.

Con el objetivo de caracterizar el rendimiento de cinco variedades de alfalfa en Montecillos, Texcoco, estado de México. Se establecieron en el año 2004 praderas con las variedades Cuf-101, Moapa, Oaxaca y Valenciana, en las que se determinó durante cuatro años el rendimiento de materia seca anual (RMSA), rendimiento estacional (RMSE), tasa de acumulación de forraje (TAF) y relación hoja: tallo. Los datos se analizaron por el procedimiento GLM de SAS y comparación de medias por la prueba de Tukey. Se analizaron los cambios por estación, variedad y años y sus interacciones. Se encontraron diferencias (p< 0.05) en rendimiento RMSA, MSE, TAF y relación hoja: tallo entre variedades, estaciones y años. Durante el primer año se observó mayor RMSA, y disminuyo hasta 57% al final del periodo de evaluación. La variedad Oaxaca mantuvo mayor producción al final de la evaluación y Moapa la de menor. En el RMSE se observó que en verano tuvo el mayor rendimiento y en invierno disminuye cerca de 50%, tendencia que se mantuvo durante el periodo de evaluación, esta diferencia va disminuyendo con el paso de los años, con una acumulación de MS más constante en el año. La relación hoja: tallo es mayor en el segundo año donde las variedades Cuf-101 y Valenciana registraron la mayor relación en la estación de invierno. En conclusión, el genotipo de la variedad de alfalfa influye en el rendimiento de materia seca por estación, año y periodo de producción, donde la estación de mayor producción de MS es verano.

Abstract. From a passive satellite remote sensing point of view, the richest set of information on aerosol properties can be obtained from instruments that measure both intensity and polarization of backscattered sunlight at multiple wavelengths and multiple viewing angles for one ground pixel. However, it is challenging to exploit this information at a global scale because complex algorithms are needed with many fit parameters (aerosol and land/ocean reflection), based on online radiative transfer models. So far, two such algorithms have demonstrated this capability at a global scale: the Generalized Retrieval of Atmosphere and Surface Properties (GRASP) algorithm and the Remote sensing of Trace gas and Aerosol Products (RemoTAP) algorithm. In this paper, we present a detailed comparison of the most recent versions of RemoTAP and GRASP. We evaluate both algorithms for synthetic observations, for real PARASOL (Polarization and Anisotropy of Reflectances for Atmospheric Science coupled with Observations from a Lidar) observations against AERONET (Aerosol Robotic Network) for common pixels, and for global PARASOL retrievals for the year 2008. For the aerosol optical depth (AOD) over land, both algorithms show a root mean square error (RMSE) of 0.10 (at 550 nm). For single scattering albedo (SSA), both algorithms show a good performance in terms of RMSE (0.04), but RemoTAP has a smaller bias (0.002) compared to GRASP (0.021). For the Ångström exponent (AE), GRASP has a smaller RMSE (0.367) than RemoTAP (0.387), mainly caused by a small overestimate of AE at low values (large particles). Over ocean both algorithms perform very well. For AOD, RemoTAP has an RMSE of 0.057 and GRASP an even smaller RMSE of 0.047. For AE, the RMSEs of RemoTAP and GRASP are 0.285 and 0.224, respectively. Based on the AERONET comparison, we conclude that both algorithms show very similar overall performance, where both algorithms have stronger and weaker points. For the global data products, we find a root mean square difference (RMSD) between RemoTAP and GRASP AOD of 0.12 and 0.038 over land and ocean, respectively. The largest differences occur over the biomass burning region in equatorial Africa. The global mean values are virtually unbiased with respect to each other. For AE the RMSD between RemoTAP and GRASP is 0.33 over land and 0.23 over ocean. For SSA, we find much better agreement over land (bias = −0.01, RMSD = 0.043 for retrievals with AOD > 0.2) than over ocean (bias = 0.053, RMSD = 0.074). As expected, the differences increase towards low AOD, over both land and ocean. We also compared the GRASP and RemoTAP AOD and AE products against MODIS. For AOD over land, the agreement of either GRASP or RemoTAP with MODIS is worse than the agreement between the two PARASOL algorithms themselves. Over ocean, the agreement is very similar among the three products for AOD. For AE, the agreement between GRASP and RemoTAP is much better than the agreement of both products with MODIS. The agreement of the latest product versions with each other and with AERONET improved significantly compared to the previous version of the global products of GRASP and RemoTAP. The results demonstrate that the dedicated effort in algorithm development for multi-angle polarimetric (MAP) aerosol retrievals still leads to substantial improvement of the resulting aerosol products, and this is still an ongoing process.

The global navigation satellite system (GNSS) is developing rapidly, and the related market applications and scientific research are increasing. Studies based on large low Earth orbit (LEO) satellite constellations have become research hotspots. The global coverage of the LEO constellation can reduce the dependence of navigation satellites on ground-monitoring stations and improve the precise orbit determination (POD) accuracy of navigation satellites. In this paper, we simulate various LEO satellite constellations (with 12, 30, and 60 satellites), along with ground stations’ observation data, to examine the impact of LEO satellites on the precision of the BeiDou-3 Global Navigation Satellite System (BDS-3) in terms of its POD accuracy. Using the simulated observation data of both LEO satellites and ground-monitoring stations, we analyze the integrated orbit determination for the LEO and BDS-3 satellites. The findings reveal that the 3D orbital accuracy of BDS-3 is 9.51 dm by using only seven ground-monitoring stations, and it is improved to a centimeter level after adding the LEO constellations. As the number of LEO constellation satellites increases, the impact on improving accuracy gradually diminishes. In terms of time synchronization accuracy in the BDS-3, compared to the results of clock offset using only ground stations, the addition of 12 LEO satellites resulted in an improvement of 49% for RMS1(root mean square) and 52% for RMS2 (standard deviation), the addition of 30 LEO satellites resulted in an improvement of 66% for RMS1 and 70% for RMS2, and the addition of 60 LEO satellites resulted in an improvement of 87% for RMS1 and 90% for RMS2. The integrated orbit determination of the LEO and BDS-3 satellites constellation greatly improves the accuracy of time synchronization. In addition, we also use simulated inter-satellite link (ISL) data to perform enhanced BDS-3 satellites POD and time synchronization experiments. The experiments showed that the orbit determination accuracy of the seven sta (seven stations) and ISL scheme is comparable to that of the seven sta and LEO12 scheme, and that the time synchronization accuracy of the seven sta and ISL scheme is slightly worse. The preliminary experiments showed that the LEO satellite could enhance the orbit determination accuracy of BDS-3 and obtain a higher time synchronization accuracy.

Objective: In this study, it was aimed to perform in silico studies on the papain-like protease structure of SARS-CoV-2 (PDB: 7JIT) of 1300 FDA-approved drugs downloaded from the ZINC database. Material and Method: A molecular docking study was performed with PLpro (PDB ID: 7JIT) using four different molecular docking programs for a total of 1300 FDA-approved drugs obtained from the ZINC database. Conivaptan and amphotericin B were obtained in docking analysis with AutoDock Vina and Sybyl-X, respectively. Docking analysis with Glide SP and Glide XP resulted in fludarabine and panobinostat, respectively. Molecular dynamics simulations were performed for a period of 120 ns to check the stability of these four drugs. Result and Discussion: The reliability of the results obtained using four different molecular docking programs on the SARS-CoV-2 papain-like protease of 1300 drug molecules was checked by reinserting the co-crystal ligand. Protein-ligand interactions between fludarabine, conivaptan, amphotericin-B, panobinostat, and PLpro were given. In the molecular dynamics study, RMSD, RMSF, Rg, and SASA analyses were performed for four systems. It was observed that RMSD remained constant for all 120 ns for all four systems except for amphotericin B, which deviated slightly towards the end of 120 ns. No significant fluctuation was noticed in the RMSF graphics for all four systems.

Pusat Pengurusan Penyelidikan dan Inovasi (RIMC), UIS telah menambahbaik Sistem Pengurusan Penyelidikan (RMS) selaras dengan keperluan perancangan strategik UIS. Dalam artikel ini, pengkaji membuat perbandingan antara RMS versi pertama (V1) dengan RMS versi kedua (V2). Kajian ini menggunakan kaedah tinjauan keperluan pembangunan RMS V2 kepada pegawai akademik UIS dan analisis kandungan perbandingan pembangunan modul antara RMS V1 dan RMS V2 ini. Kajian mendapati RMS V1 yang dibangunkan mempunyai modul permohonan penyelidikan dan penjanaan senarai tajuk penyelidikan, penerbitan dan inovasi yang tidak mengikut format pentaksiran MyRA secara terperinci. Pembangunan RMS V2 telah melibatkan tambahan modul yang lebih efektif dan efisen iaitu 1) permohonan penyelidikan yang lebih lengkap 2) penilaian proposal oleh panel kluster yang boleh dijalankan secara atas talian 3) pemantauan kemajuan penyelidikan yang lebih komprehensif melalui sistem atas talian, 4) pemprofilan (profiling) pegawai akademik yang lebih terperinci berdasarkan format pentaksiran MyRA dan 5) penjanaan laporan yang dapat membantu RIMC mengurus data aktiviti penyelidikan, pembangunan, pengkomersilan dan inovasi (RDCI) dengan lebih teratur. RMS V2 diharap dapat membantu penyelidik UIS mengembangkan potensi mereka sebagai sarjana dan ahli akademik serta membantu pengurusan UIS dalam membuat keputusan dan memenuhi keperluan sebagai sebuah universiti.

Kanker leukemia merupakan suatu penyakit keganasan pada sistem hematopoiesis yang ditandai dengan akumulasi leukosit ganas di sumsum tulang dan darah tepi. Reseptor CK2-α (Casein Kinase 2-alfa) merupakan molekul penting dalam mengatur kaskade sinyal hematopoiesis yang terkait dalam mendorong pertumbuhan beberapa tumor darah. Senyawa ligan alami (CX-4945) dari reseptor CK2-α yang merupakan senyawa turunan alkaloid naftiridin berdasarkan beberapa penelitian yang telah dilakukan baik secara in vitro maupun secara in vivo terbukti memiliki aktivitas dalam menghambat sel-sel penyebab kanker, khususnya protein CK2-α pada kanker leukemia. Tujuan: Mengkaji interaksi dan afinitas senyawa turunan naftiridin terhadap reseptor CK2-α serta prediksi toksisitasnya sehingga menjadi alternatif pengobatan leukemia dengan resiko efek samping yang minimum. Metodologi: Tahapan studi yang dilakukan antara lain pengkajian parameter fisikokimia, meliputi penetapan koefisien partisi (CLogP), bobot molekul (BM), jumlah donor dan akseptor ikatan hidrogen, jumlah rotasi ikatan, energi HOMO-LUMO, lalu dilakukan analisis interaksi ligan uji terhadap reseptor CK2-α melalui simulasi molecular docking dan molecular dynamic serta prediksi toksisitasnya. Validasi docking dilakukan dengan me-redocking ligan alami CX-4945 dari reseptor CK2-α (kode PDB: 3PE1), diperoleh nilai RMSD sebesar 0,358 Å. Hasil: Pada simulasi molecular docking senyawa turunan naftiridin terhadap reseptor CK2-α dengan Autodock didapatkan 10 ligan uji terbaik dengan nilai energi bebas ikatan (∆G) dan nilai konstanta inhibisi (Ki) yang rendah. Sebelum dilakukan simulasi molecular dynamic, 10 ligan uji dan reseptor target dipreparasi dengan pembuatan topologi. Hasil simulasi molecular dynamic 10 ligan uji terbaik terhadap reseptor CK2-α dengan Gromacs selama 10 nanodetik menunjukkan nilai RMSD dan RMSF yang rendah ditandai dengan kecenderungan grafik yang cukup konstan, dimana nilai RMSD untuk ligan uji 4-Metilaaptamin sebesar 1,4 Å dan nilai RMSF untuk ligan uji Lopokladin A sebesar 1,5 Å menunjukkan kestabilan yang cukup baik diantara ligan uji lainnya. Sedangkan prediksi toksisitas dengan ADMET Predictor menunjukkan bahwa ligan uji Kloronaftiridin diprediksi memiliki resiko toksisitas paling rendah dibandingkan ligan uji lainnya. Berdasarkan hasil penelitian tersebut, diketahui bahwa 4-Metilaaptamin merupakan kandidat senyawa inhibitor CK2-α yang paling berpotensi dengan afinitas tertinggi dan resiko toksisitas yang rendah.

Garcinia adalah genus yang terdiri dari berbagai macam spesies dengan aktivitas beragam. Dua spesies garcinia yang telah diteliti adalah G. dulcis dan G. mangostana. Ekstrak buah bagian pericarp kedua spesies tersebut memiliki aktivitas sebagai antioksidan, antibakteri, anti hiperlipidemia, dan antidiabetes. Namun, aktivitas kedua spesies ini bervariasi, sehingga diperlukan autentikasi salah satu spesies untuk menghindari terjadinya adulterasi (pencampuran). Penelitian ini bertujuan untuk mengautentifikasi G. dulcis dalam campuran G. dulcis dan G. mangostana. Penelitian ini menggunakan metode spektrofotometri kombinasi dengan kemometrik. Peralatan yang digunakan adalah spektrofotometri UV-vis, dan analisis data menggunakan PLSR. Hasil dari penelitian pada panjang gelombang daerah 246,77-224,46 nm model normal RMSEC adalah 3,75 dan R2 0,99 dan panjang gelombang 300,2- 256,56 nm derivatisasi kedua RMSEC adalah 3,21 dan R2 0,9956. Berdasarkan data yang diperoleh dapat disimpulkan model derivatisasi ke-2 pada panjang gelombang 300,2-256,56 nm merupakan model terbaik untuk autentikasi G.dulcis terhadap G.mangostana karena memiliki RMSE terendah dan R2 tertinggi secara kualitatif namun belum mampu mengkuantifikasi perbedaannya.

Coming into the second year of the pandemic, the acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and its variants continue to be a serious health hazard globally. A surge in the omicron wave, despite the discovery of the vaccines, has shifted the attention of research towards the discovery and use of bioactive compounds, being potential inhibitors of the viral structural proteins. The present study aimed at the green synthesis of zinc oxide (ZnO) nanoparticles with seed extracts of Nigella sativa and Pimpinella anisum—loaded nanostructured oil carriers (NLC)—using a mixture of olive and black seed essential oils. The synthesized ZnO NLC were extensively characterized. In addition, the constituent compounds in ZnO NLC were investigated as a potential inhibitor for the SARS-CoV-2 main protease (3CLpro or Mpro) where 27 bioactive constituents, along with ZnO in the nanostructure, were subjected to molecular docking studies. The resultant high-score compounds were further validated by molecular dynamics simulation. The study optimized the compounds dithymoquinone, δ-hederin, oleuropein, and zinc oxide with high docking energy scores (ranging from −7.9 to −9.9 kcal/mol). The RMSD and RMSF data that ensued also mirrored these results for the stability of proteins and ligands. RMSD and RMSF data showed no conformational change in the protein during the MD simulation. Histograms of every simulation trajectory explained the ligand properties and ligand–protein contacts. Nevertheless, further experimental investigations and validation of the selected candidates are imperative to take forward the applicability of the nanostructure as a potent inhibitor of COVID-19 (Coronavirus Disease 2019) for clinical trials.

AbstractAdvanced glycosylation end products (AGEs) are harmful factors that can damage the inner blood–retinal barrier (iBRB). Rat retinal microvascular endothelial cells (RMECs) were isolated and cultured, and identified by anti-CD31 and von Willebrand factor polyclonal antibodies. Similarly, rat retinal Müller glial cells (RMGCs) were identified by H&E staining and with antibodies of glial fibrillary acidic protein and glutamine synthetase. The transepithelial electrical resistance (TEER) value was measured with a Millicell electrical resistance system to observe the leakage of the barrier. Transwell cell plates for co-culturing RMECs with RMGCs were used to construct an iBRB model, which was then tested with the addition of AGEs at final concentrations of 50 and 100 mg/L for 24, 48, and 72 h. AGEs in the in vitro iBRB model constructed by RMEC and RMGC co-culture led to the imbalance of the vascular endothelial growth factor (VEGF) and pigment epithelial derivative factor (PEDF), and the permeability of the RMEC layer increased because the TEER decreased in a dose- and time-dependent manner. AGEs increased VEGF but lowered PEDF in a dose- and time-dependent manner. The intervention with AGEs led to the change of the transendothelial resistance of the RMEC layer likely caused by the increased ratio of VEGF/PEDF.

Background: Inertial measurement units (IMUs) offer the possibility to capture the lower body motions of players of outdoor team sports. However, various sources of error are present when using IMUs: the definition of the body frames, the soft tissue artefact (STA) and the orientation filter. Methods to minimize these errors are currently being used without knowing their exact influence on the various sources of errors. The goal of this study was to present a method to quantify each of the sources of error of an IMU separately. Methods: An optoelectronic system was used as a gold standard. Rigid marker clusters (RMCs) were designed to construct a rigid connection between the IMU and four markers. This allowed for the separate quantification of each of the sources of error. Ten subjects performed nine different football-specific movements, varying both in the type of movement, and in movement intensity. Results: The error of the definition of the body frames (11.3–18.7 deg RMSD), the STA (3.8–9.1 deg RMSD) and the error of the orientation filter (3.0–12.7 deg RMSD) were all quantified separately for each body segment. Conclusions: The error sources of IMU-based motion analysis were quantified separately. This allows future studies to quantify and optimize the effects of error reduction techniques.
