This paper uses data from the Human Gene Mutation Database to contrast two hypotheses for the origin of short DNA repeats: substitutions and insertions that duplicate adjacent sequences. Because substitutions are much more common than insertions, they are the dominant source of new 2-repeat loci. Insertions are rarer, but over 70% of the 2–4 base insertion mutations are duplications of adjacent sequences, and over half of these generate new repeat regions. Insertions contribute fewer new repeat loci than substitutions, but their relative importance increases rapidly with repeat number so that all new 4–5-repeat mutations come from insertions, as do all 3-repeat mutations of tetranucleotide repeats. This suggests that the process of repeat duplication that dominates microsatellite evolution at high repeat numbers is also important very early in microsatellite evolution. This result sheds light on the puzzle of the origin of short tandem repeats. It also suggests that most short insertion mutations derive from a slippage-like process during replication.

Cinnamon (Cinnamomum burmannii) is an herbal plant that has a function, one of which is lowering glucose in the blood and warding off free radicals that cause oxidative stress. This study aims to determine the inhibitory activity of the enzyme compound α-glucosidase and the suppression of DPPH free radicals using the first, second, and third replications of cinnamon bark water extract (C. burmannii) as a natural source of antidiabetic drug candidates. The extraction method uses the infundation technique and is carried out using a spectronometer. The highest yield was obtained in the extraction of the first repeat, which is with an average of 0.674%, the yield did not differ markedly (p>0.05) with the second and third repeats. The highest results of total phenolic testing were obtained in the second repeat extraction of 100.374 GAE g-1, the results differed markedly (p<0.05) with the first and third repeats. The highest results of testing antioxidant activity were obtained at the extraction of the first repeat which was 9.012 mg of AAE g-1, the results differed markedly (p<0.05) with the second and third repeats. The highest result of testing of enzyme inhibition activity α-glucosidase was found in the extraction of the first repeat which was 99.650%, the results did not differ markedly (p>0.05) with the second and third repeats.

Abstract Transcription activator-like effectors (TALEs) are bacterial proteins with a programmable DNA-binding domain, which turned them into exceptional tools for biotechnology. TALEs contain a central array of consecutive 34 amino acid long repeats to bind DNA in a simple one-repeat-to-one-nucleotide manner. However, a few naturally occurring aberrant repeat variants break this strict binding mechanism, allowing for the recognition of an additional sequence with a −1 nucleotide frameshift. The limits and implications of this extended TALE binding mode are largely unexplored. Here, we analyse the complete diversity of natural and artificially engineered aberrant repeats for their impact on the DNA binding of TALEs. Surprisingly, TALEs with several aberrant repeats can loop out multiple repeats simultaneously without losing DNA-binding capacity. We also characterized members of the only natural TALE class harbouring two aberrant repeats and confirmed that their target is the major virulence factor OsSWEET13 from rice. In an aberrant TALE repeat, the position and nature of the amino acid sequence strongly influence its function. We explored the tolerance of TALE repeats towards alterations further and demonstrate that inserts as large as GFP can be tolerated without disrupting DNA binding. This illustrates the extraordinary DNA-binding capacity of TALEs and opens new uses in biotechnology.

ABSTRACT Flap endonuclease 1 (FEN1) is a central component of Okazaki fragment maturation in eukaryotes. Genetic analysis of Saccharomyces cerevisiae FEN1 (RAD27) also reveals its important role in preventing trinucleotide repeat (TNR) expansion. In humans such expansion is associated with neurodegenerative diseases. In vitro, FEN1 can inhibit TNR expansion by employing its endonuclease activity to compete with DNA ligase I. Here we employed two yeast FEN1 nuclease mutants, rad27-G67S and rad27-G240D, to further define the mechanism by which FEN1 prevents TNR expansion. Using a yeast artificial chromosome system that can detect both TNR instability and fragility, we demonstrate that the G240D but not the G67S mutation increases both the expansion and fragility of a CTG tract in vivo. In vitro, the G240D nuclease is proficient in cleaving a fixed nonrepeat double flap; however, it exhibits severely impaired cleavage of both nonrepeat and CTG-containing equilibrating flaps. In contrast, wild-type FEN1 and the G67S mutant exhibit more efficient cleavage on an equilibrating flap than on a fixed CTG flap. The degree of TNR expansion and the amount of chromosome fragility observed in the mutant strains correlate with the severity of defective flap cleavage in vitro. We present a model to explain how flap equilibration and the unique tracking mechanism of FEN1 can collaborate to remove TNR flaps and prevent repeat expansion.

Abstract Summary We describe a novel computational method for genotyping repeats using sequence graphs. This method addresses the long-standing need to accurately genotype medically important loci containing repeats adjacent to other variants or imperfect DNA repeats such as polyalanine repeats. Here we introduce a new version of our repeat genotyping software, ExpansionHunter, that uses this method to perform targeted genotyping of a broad class of such loci. Availability and implementation ExpansionHunter is implemented in C++ and is available under the Apache License Version 2.0. ABSTRACT Telomeres in the budding yeast Kluyveromyces lactisconsist of perfectly repeated 25-bp units, unlike the imprecise repeats at Saccharomyces cerevisiae telomeres and the short (6- to 8-bp) telomeric repeats found in many other eukaryotes. Telomeric DNA is synthesized by the ribonucleoprotein telomerase, which uses a portion of its RNA moiety as a template. K. lactistelomerase RNA, encoded by the TER1 gene, is ∼1.3 kb long and contains a 30-nucleotide templating domain, the largest ever examined. To examine the mechanism of polymerization by this enzyme, we identified and analyzed telomerase activity from K. lactiswhole-cell extracts. In this study, we exploited the length of the template and the precision of copying by K. lactistelomerase to examine primer elongation within one round of repeat synthesis. Under all in vitro conditions tested, K. lactistelomerase catalyzed only one round of repeat synthesis and remained bound to reaction products. We demonstrate that K. lactistelomerase polymerizes along the template in a discontinuous manner and stalls at two specific regions in the template. Increasing the amount of primer DNA-template RNA complementarity results in stalling, suggesting that the RNA-DNA hybrid is not unpaired during elongation in vitro and that lengthy duplexes hinder polymerization through particular regions of the template. We suggest that these observations provide an insight into the mechanism of telomerase and its regulation.

It is an effective means for merchants to carry out precision marketing and improve ROI by using historical user behavior data obtained from promotional activities in order to build a model to predict the repeat purchase behavior of users after promotional activities. Most of the existing prediction models are supervised learning, which does not work well with a small amount of labeled data. This paper proposes a BERT-MLP prediction model that uses “large-scale data unsupervised pre-training + small amount of labeled data fine-tuning.” The experimental results on Alibaba real dataset show that the accuracy of the BERT-MLP model is better than the baseline model.

This study aims to analyze sustainable behavior in paying alms through a digital platform. The data was collected using an online questionnaire with a sample of 50 millennial Muslims in Surabaya who had made alms payments through a digital platform. This study uses a quantitative approach and analysis using multiple linear regression. The sampling technique used is purposive sampling. The results of this study indicate that religiosity, perceived ease of use, and e-satisfaction have a positive effect on the sustainable behavior of millennial Muslims in paying alms through digital platforms. The perceived ease of use variable has a small effect on the intention of repeated donations, it is possible that this is due to the lack of internet network in the area and the insufficient balance of funds in the application. Suggestions for further research is to use a qualitative approach in order to obtain more accurate information and use other variables that are more supportive and relevant.

Research background: There are several methods to construct a price index for infrequently traded real estate assets (mainly residential, but also office and land). The main concern to construct a valid and unbiased price index is to address the problem of heterogeneity of real estate or put differently to control for both observable and unobservable quality attributes. The one most frequently used is probably the hedonic regression methodology (classic, but recently also spatial and quantile regression). An alternative approach to control for unobservable differences in assets’ quality is provided by repeat sales methodology, where price changes are tracked based on differences in prices of given asset sold twice (or multiple times) within the study period. The latter approach is applied in renown S&P CoreLogic Case-Shiller house price indices. Purpose of the article: The goal of the paper is to assess the applicability of repeat sales methodology for a major housing market in Poland. Previous studies used the hedonic methodology or mix adjustment techniques, and applied for major metropolitan areas. The most widely known example is the set of quarterly house price indices constructed by NBP — especially for the primary and secondary market. The repeat sales methodology has not been adopted with significant success to date — mainly because of concern regarding relative infrequency of transactions on the housing market in most metropolitan areas (thus a potentially small sample of repeated sales). Methods: The study uses data on repeat sales of residential transactions in Krakow from 2003 to 2015. We apply different specifications of repeat sales index construction and compare respective values to the hedonic price index for Krakow estimated by NBP. Findings & Value added: Findings suggest that repeat sales house sales indices can be used to track price dynamics for major metropolitan areas in Poland. The study suggests problems that need to be addressed in order to get unbiased results — mainly data collection mechanism and estimation procedure.

The present research uses hierarchical modeling to examine the relative contributions of factors about the person, factors about the context, and, most important the interaction of factors about the person and factors about the context in models of both repeat victimization (more than one of the same type of crime) and multiple victimization (two or more different types of crime). Using telephone survey data from a multistage sample of Seattle residents, we estimate separate hierarchical models for repeat property, repeat violent and multiple victimization. Results indicate that repeat victimization of both types varies substantially by neighborhood, whereas multiple victimization seems more determined by individual-level factors. Implications for social disorganization theory, routine activity/lifestyle exposure theory, and future work on repeat victimization are discussed.

This study uses a test tool in the form of IBM SPSS Statistic 26. The population used is Followers Autobase @ohmybeautybank. The data collection used in this research is using a questionnaire. Data analysis in this study used descriptive statistical techniques. The results of the t-test indicate that Customers Experience and Customer Satisfaction have a positive effect on Repeat Purchase Avoskin products at @ohmybeautybank Followers where customers experience has a t-count of 3.250 while Customers Satification has a t-count of 2.123. e18700 Background: Previous studies on mutation calling have documented capture kit batch effects in Whole Exome Sequencing (WES) data from The Cancer Genome Atlas (TCGA) database, hindering direct comparison between samples from different capture kits. For example, in classification, a cancer type exclusively sampled by a specific capture kit in the training set would have very low accuracy if the testing set was sampled by another capture kit. To enable cross-capture-kit between-cancer genotype analyses with the TCGA dataset, a novel read count transformation algorithm is developed to remove capture kit batch effects. This algorithm was tested with our Machine Learning model which uses Tandem Repeat Sequence (TRS) mutation markers as training features. Methods: The proposed algorithm transforms TRS read count data to remove low quality samples, read depth differences, and capture kit batch effects from the dataset. Results: 1) TRS read count of WES samples are investigated. Particularly, we show that TRS site read counts do not correlate across capture kits but correlate within capture kits. This suggests that WES read count is largely independent from an exon’s location in the genome and is more strongly correlated with capture kit probes. 2) TRS detection rate for each sample within each capture kit is found to be normally distributed. Outliers with very low TRS detection rate can be used for quality filtering. 3) The transformation algorithm effectively removes capture kit batch effects from the dataset. At the same time, it retains cancer-specific signals in the samples. Before applying the transformation algorithm, cancer type classification accuracy is low (̃0-25%) if the testing data set uses a different capture kit from the training data set. We show that applying the transformation algorithm allows cancer type classification accuracy to improve by over 65%. Conclusions: We demonstrated that direct comparison of WES TRS read count data across capture kits is possible after application of our transformation algorithm. This opens the path to cross-capture-kit between-cancer genotype analyses with the TCGA dataset, which were previously unfeasible due to capture kit batch effects.

This study aims to determine the effect of trust on the repeat intention to give cash waqf at BWA Surabaya through beliefs which include aspects of attitude, subjective norms, and perceived behavioral control. Retrieval of data in this study using a questionnaire with a total sample obtained by 35 waqif, who had already given money endowment at BWA Surabaya in one past year. The sampling technique are both non probability and simple random sampling. This research uses a quantitative approach with path analysis techniques. The endogenous variable in this study is the repeat intention to give cash waqf, while the exogenous variable used is trust and the intervening endogenous variable is beliefs. The results of this study showed that trust has a positive and significant influence on beliefs. Nevertheless, trusts does not affect the repeat intention to give cash waqf. However, trusts indirectly affects to the repeat intention to give cash waqf through beliefs variables that have a positive and significant effect on the repeat intention to give cash waqf at BWA Surabaya. Suggestions for BWA Surabaya should be maintaining and increasing trusts and beliefs of waqif which can increase the repeat intention of waqif to give cash waqf at BWA Surabaya.Keywords: Trust, Wakaf, Belief

BACKGROUND: Repeat CT scanning is ubiquitous in many clinical situations, e.g. to follow disease progression, to evaluate treatment efficacy, and to monitor interventional CT procedures. However, it incurs in cumulative radiation to the patient which can be significantly reduced by using a region of interest (ROI) and the existing baseline scan. OBJECTIVE: To obtain a high-quality reconstruction of a ROI with a significantly reduced X-ray radiation dosage that accounts for deformations. METHODS: We present a new method for deformable registration and image reconstruction inside an ROI in repeat CT scans with a highly reduced X-ray radiation dose based on sparse scanning. Our method uses the existing baseline scan data, a user-defined ROI, and a new sparse repeat scan to compute a high-quality repeat scan ROI image with a significantly reduced radiation dose. Our method first performs rigid registration between the densely scanned baseline and the sparsely scanned repeat CT scans followed by deformable registration with a low-order parametric model, both in 3D Radon space and without reconstructing the repeat scan image. It then reconstructs the repeat scan ROI without computing the entire repeat scan image. RESULTS: Our experimental results on clinical lung and liver CT scans yield a mean × 14 computation speedup and a × 7.6-12.5 radiation dose reduction, with a minor image quality loss of 0.0157 in the NRMSE metric. CONCLUSION: Our method is considerably faster than existing methods, thereby enabling intraoperative online repeat scanning that it is accurate and accounts for position, deformation, and structure changes at a fraction of the radiation dose required by existing methods.

Abstract Objective Audio is increasingly used to access information on the Internet through virtual assistants and smart speakers. Our objective is to evaluate the distribution of health information through audio. Materials and Methods We conducted 2 studies to compare comprehension after reading or listening to information using a new corpus containing short text snippets from Cochrane (N = 50) and Wikipedia (N = 50). In study 1, the snippets were first presented as audio or text followed by a multiple-choice question. Then, the same information was presented as text and the question was repeated in addition to questions about perceived difficulty, severity and the likelihood of encountering the disease. In study 2, the first multiple-choice question was replaced with a free recall question. Results Study 1 showed that information comprehension is very similar in both presentation modes (53% accuracy for text and 55% for audio). Study 2 showed that information retention is higher with text, but similar comprehension. Both studies show improvement in performance with repeated information presentation. Discussion Audio presentation of information is effective and the format novel. Performance was slightly lower with audio when asked to repeat information, but comparable to text for answering questions. Additional studies are needed with different types of information and presentation combinations. Conclusion The use of audio to provide health information is a promising field and will become increasingly important with the popularity of smart speakers and virtual assistants, particularly for consumers who do not use computers, for example minority groups, or those with limited sight or motor control.

ABSTRACT In most trinucleotide repeat (TNR) diseases, the primary factor determining the likelihood of expansions is the length of the TNR. In some diseases, however, stable alleles contain one to three base pair substitutions that interrupt the TNR tract. The unexpected stability of these alleles compared to the frequent expansions of perfect TNRs suggested that interruptions somehow block expansions and that expansions occur only upon loss of at least one interruption. The work in this study uses a yeast genetic assay to examine the mechanism of stabilization conferred by two interruptions of a 25-repeat tract. Expansion rates are reduced up to 90-fold compared to an uninterrupted allele. Stabilization is greatest when the interruption is replicated early on the lagging strand, relative to the rest of the TNR. Although expansions are infrequent, they are often polar, gaining new DNA within the largest available stretch of perfect repeats. Surprisingly, interruptions are always retained and sometimes even duplicated, suggesting that expansion in yeast cells can proceed without loss of the interruption. These findings support a stabilization model in which interruptions contribute in cis to reduce hairpin formation during TNR replication and thus inhibit expansion rates.

This study aims to determine whether experiential marketing and PayLater features on the Gojek application have on consumer satisfaction and their impact on repurchase interest. The data analysis technique uses path analysis and uses quantitative methods. Quantitative methods include validity and reliability, classic assumption tests, t-test trials, F tests, and the coefficient of determination (R2). The number of samples used in this study were 100 respondents. The results of the study indicate that experiential marketing has a significant positive effect on customer satisfaction, the PayLater feature also has a positive effect on customer satisfaction, and the two independent variables simultaneously have a positive effect on customer satisfaction. The findings also show that experiential marketing has a positive effect on repurchase interest, consumer satisfaction has a positive effect on repurchase interest, and different results are shown by the PayLater feature which has a negative effect on repurchase interest. Simultaneously, experiential marketing variables, PayLater features, and customer satisfaction have a positive effect on repurchase interest.

Development affirmation is considered as a huge endeavor in various applications, particularly in clinical consideration organizations. Among these applications consolidate clinical decisive, seeing of customers' consistently timetable and ID of unusual cases. Here we present an approach for the development affirmation using an accelerometer sensor embedded in a mobile phone. This strategy uses a transparently available accelerometer dataset as the unrefined information signal. The features of the sign are picked subject to the time and repeat space. By then, Principal Component Analysis (PCA) is used to diminish the dimensionality of the features and concentrate the primary ones that can describe human activities. An assessment collaboration is performed between the primary unrefined data and PCA-based features and moreover, time and repeat region features are similarly contemplated using a couple of AI classifiers. The got results show that the PCA-based features get higher affirmation rate while repeat region features have higher exactness, with the speed of 96.11% and 92.10% independently.

Purpose – The purpose of this paper is to suggest a method of selecting efficient customer service programmes and of providing relevant customer information to dealers, based on the analysis of repeat-purchase behaviour data in the automobile industry. Design/methodology/approach – A recurrent event model is proposed and employed to determine which variables affect repurchasing behaviour in consumers' repurchase cycles. Unlike the conventional recurrent event model, the proposed model uses common variables for all strata, as well as stratum-specific variables. Findings – Empirical results show that age, price difference, change in vehicle type, and marketing promotion affect the likelihood of repeat purchase. VIP service centres and repair services are effective marketing tools, and dealers should pay more attention to existing customers having certain characteristics, depending on prior purchase behaviour. Practical implications – Though many customer service programmes are devised and implemented at great cost, Customer Relationship Management (CRM) data reveal that classic car-care services are the most essential. CRM can provide dealers with essential customer information that enables real purchases. Originality/value – Collecting primary data on automobile purchase behaviour and customer service usage is difficult, and therefore, customer behaviour strategy is often formulated using basic principles alone. The paper proposes a method to construct a service strategy and formulate deal guidelines based on CRM data and statistical modelling.

Burglarized locations are at an elevated risk of re-victimization soon after an initial offense. The current study uses a two-mode network approach to examine three possible mechanisms of repeat victimization: (a) The characteristics of a location make the target attractive to all motivated offenders, (b) the same offender returns to the offense location multiple times, or (c) other offender(s), to whom the primary offender is connected, commit subsequent offenses. The results support all three mechanisms, but particularly the same offender returning for subsequent offenses. These “returners” are generally more experienced, more connected burglary offenders. Significantly, however, repeat victimizations not committed by the same offender(s) often involve connected co-offenders, a mechanism for repeat victimization that has not been previously demonstrated empirically.

The rapid development of business and technology has had a positive impact on the business world so that attention to consumer trust and repeat orders is an important thing that must be considered. The purpose of this study is to analyze the role of Islamic business ethics in increasing consumer confidence and the implications for increasing consumer repeat orders. This study uses a qualitative method with a type of phenomenological research. The data sources used are primary and secondary data. Primary data was obtained through observation and direct interviews with 14 informants while secondary data was obtained from relevant literature. The research results show that CV. Sibuh Soppeng Regency focuses its operations on two things, namely service and marketing by implementing Islamic business ethics. Several indicators are used in building consumer trust such as good service, honesty, keeping promises, being responsible for consumers, and working on time which has implications for repeat orders from these consumers. Perkembangan bisnis dan teknologi yang semakin pesat membawa dampak positif dalam dunia bisnis sehingga perhatian terhadap kepercayaan dan repeat order konsumen menjadi hal penting yang harus diperhatikan. Tujuan dari penelitian ini adalah untuk menganalisis peran etika bisnis Islam dalam meningkatkan kepercayaan konsumen serta implikasinya terhadap peningkatan repeat order konsumen. Penelitian ini menggunakan metode kualitatif dengan jenis penelitian fenomenologi. Sumber data yang digunakan yaitu data primer dan sekunder. Data primer diperoleh melalui observasi dan wawancara langsung dengan 14 informan sedangkan data sekunder didapat dari literatur yang relevan. Hasil Penelitian menunjukkan bahwa CV. Sibuh Kabupaten Soppeng memfokuskan operasionalnya pada dua hal yaitu pelayanan dan pemasaran dengan mengimplementasikan etika bisnis Islam. Beberapa indikator digunakan dalam membangun kepercayaan konsumen seperti pelayanan yang baik, kejujuran, menepati janji, bertanggung jawab terhadap konsumen, serta bekerja tepat waktu yang berimplikasi terhadap repeat order dari konsumen tersebut.

Hysterosalpingography (HSG) is a diagnostic modality still commonly used to evaluate tubal occlusion in infertility management. We evaluated the treatment benefit in achieving tubal patency and spontaneous pregnancy in women with bilateral tubal occlusion in our resource poor setting using water soluble contrast medium. Objective: To assess the proportion of women that achieved tubal patency and spontaneous pregnancy following a repeat HSG in the evaluation of infertility and the associated factors. Materials and Method: This is a retrospective study of 37 out of 180 women who achieved tubal patency and pregnancy after repeat HSG in the course of infertility evaluation over a period of 18 months. Result: Out of 180 patients with bilateral tubal occlusion that had the repeat HSG, 37 achieved tubal patency (20.5%) and from which 24 became pregnant. This gave a pregnancy rate of 13.3%. The mean age of the participants was 35.76 ± 4.72 years. The age group of 36-40 years had the highest frequency while most of them were nulliparous and had left tubal occlusion at the distal portion. Conclusion: Tubal patency and spontaneous pregnancy were achieved following repeat HSG in women with bilateral tubal occlusion undergoing infertility evaluation. HSG has both diagnostic and treatment uses in infertility management and it is cost effective.

Abstract: There are over 200 distinct forms of cancer, and all are diagnosed and treated differently. According to the WHO Global Cancer Observatory, 19,292,789 cases new cases of cancer were diagnosed in 2020, with breast (11.7% cases), lung (11.4%), and Colorectum (10% cases) becoming the three most common. Lung cancer is one of the most often diagnosed malignancies and the biggest cause of cancer-related deaths globally, with an estimated 220 million new cases and 179 million deaths every year. It is extremely invasive, quickly spreading, and cause death in both sexes. In light of both core genetic abnormalities and therapy response, lung cancer is a highly diverse ailment. WDR74 protein predominantly controls WNT signaling pathways in lung cancer. Wnt-responsive genes such as c-myc and cyclin D1 have been linked to cell proliferation, whereas caspase 3, caspase 9, and MDR1 have been linked to chemo-resistance and death. WDR74 had regulatory impacts on these genes. In lung cancer cells, WDR74 influenced several biological processes in lung cancer cells by modulating these genes. Abnormal activation of the Wnt/-catenin signalling pathway promotes a number of cellular functions such as proliferation, cell cycle progression, aggressiveness, and chemoresistance, specifically in Lung cancer. Our study uses local alignment to establish WDR74 sequence similarity, and then uses multiple sequence alignment to compare homologous sequences. We then used software to scan the sequence for open reading frames (ORFs) to see if WDR74 had main mutations. The study's findings include a brief summary of the top five protein matches from well-studied reference species in the database, as well as a graphical summary and phylogenetic tree development. The study also suggested the open reading frame with the primary mutation, as well as the start and stop codon positions. The Independent variable of Service Quality has a dominant influence on the Product Sale which has a Beta value of 0,597.

We present a decomposition technique that uses non-deterministic circuits to approximate an arbitrary single-qubit unitary to within distance $\epsilon$ and requires significantly fewer non-Clifford gates than existing techniques. We develop ``Repeat-Until-Success" (RUS) circuits and characterize unitaries that can be exactly represented as an RUS circuit. Our RUS circuits operate by conditioning on a given measurement outcome and using only a small number of non-Clifford gates and ancilla qubits. We construct an algorithm based on RUS circuits that approximates an arbitrary single-qubit $Z$-axis rotation to within distance $\epsilon$, where the number of $T$ gates scales as $1.26\log_2(1/\epsilon) - 3.53$, an improvement of roughly three-fold over state-of-the-art techniques. We then extend our algorithm and show that a scaling of $2.4\log_2(1/\epsilon) - 3.28$ can be achieved for arbitrary unitaries and a small range of $\epsilon$, which is roughly twice as good as optimal deterministic decomposition methods.

At a time when criminal justice systems in Europe and North America increasingly seek to utilize the epistemic authority of a variety of sciences in support of the apprehension and prosecution of suspects and offenders, genetic science and recombinant DNA technology are often singled out for particular approbation. Indeed, the development and application of DNA profiling has been widely described as the “greatest breakthrough in forensic science since fingerprinting.”

The Covid-19 pandemic which has spread in various countries has a severe impact on all sectors in every country, especially the economic and tourism sectors. One of which is Singapore, which has closed access to foreign tourism with a lockdown policy and has progressively implemented a series of steps to reduce the risk of a wider spread of the COVID-19 pandemic in the community. Singapore has managed to maintain sustainable tourism over the years. Several tourist attractions in Singapore have resumed operations after being closed for several months. Indonesia was the market for the first largest source of income from tourism for Singapore for five consecutive years in 2016. This study aims to determine the perception of Indonesian travelers to return to tourism after the Singapore government reopened tourism to foreign countries. The research method used is quantitative, the sampling technique uses non-probability sampling and a sample of 285 respondents is obtained. The analytical tool used Path analysis and the hypothesis uses a significance test using the Partial Least Square Structural Equation Model (PLS SEM). The results of this study indicate that destination attractiveness, tourist motivation, accessibility, and risk perception can directly revisit the intention of Indonesian tourists to travel in Singapore. Then, destination attractiveness and tourist motivation can affect the revisit intention of Indonesian tourists to travel back to Singapore after the Covid-19 outbreak.

Few complete mitochondrial genomes representing limited families in the order Hymenoptera have been sequenced. Here, we sequenced the complete mitochondrial genome of Diadegma semiclausum (Hymenoptera: Ichneumonidae). This genome is 18 728 bp long, the second largest hexapod mitochondrial genome sequenced in its entirety and that with the highest A+T content at 87.4%. Four tRNAs are rearranged compared with the ancestral arrangement. Gene rearrangement mechanisms are different among all three rearranged regions. Six tRNAs have a large variable loop, which is not found in other metazoan mitochondrial genomes. trnS(AGY) uses the abnormal anticodon TCT but trnK uses the normal CTT. The A+T-rich region is very long (2161 bp). An extremely A+T-rich (99.1%) 1515 bp tandem repeat region with three types of repeat elements is located between cox1 and cox2, and the most likely ancestral element originated from the 3′ end of cox1. Independent tandem duplications followed by mutation–insertion–deletion is the best model to explain the formation of this region. These results indicate that independent evolutionary events occurred extensively, such as gene rearrangement events, gene rearrangement mechanisms, derivation of tRNA variable loops, and tandem repeat region evolutionary processes, all of which likely contribute to the diversified features of hymenopteran mitochondrial genomes.

Haemophilus influenzae uses phase variation (PV) to modulate the activity of its defence systems against phage infection. The PV of the restriction–modification (R-M) system HindI, the main defence system against phage infection and incoming chromosomal and phage DNA in H. influenzae Rd, is driven by changes of the pentanucleotide repeat tract within the coding sequence of the hsdM gene and is influenced by lack of Dam methylation. Phase-variable resistance/sensitivity to phage infection correlates with changes in lipooligosaccharide (LOS) structure and occurs by slippage of tetranucleotide repeats within the gene lic2A, coding for a step in the biosynthesis of LOS. The lack of Dam activity destabilizes the tetranuclotide (5′-CAAT) repeat tract and increases the frequency of switching from sensitivity to resistance to phage infection more than in the opposite direction. The PV of the lgtC gene does not influence resistance or sensitivity to phage infection. Insertional inactivation of lic2A, but not lgtC or lgtF, leads to resistance to phage infection and to the same structure of the LOS as observed among phase-variable phage-resistant variants. This indicates that in the H. influenzae Rd LOS only the first two sugars (Glc-Gal) extending from the third heptose are part of bacterial phage receptors.

BACKGROUND: Detecting and interpreting changes in the images of follow-up CT scans by the clinicians is often time-consuming and error-prone due to changes in patient position and non-rigid anatomy deformations. Thus, reconstructed repeat scan images are required, precluding reduced dose sparse-view repeat scanning. OBJECTIVE: A method to automatically detect changes in a region of interest of sparse-view repeat CT scans in the presence of non-rigid deformations of the patient’s anatomy without reconstructing the original images. METHODS: The proposed method uses the sparse sinogram data of two CT scans to distinguish between genuine changes in the repeat scan and differences due to non-rigid anatomic deformations. First, size and contrast level of the changed regions are estimated from the difference between the scans’ sinogram data. The estimated types of changes in the repeat scan help optimize the method’s parameter values. Two scans are then aligned using Radon space non-rigid registration. Rays which crossed changes in the ROI are detected and back-projected onto image space in a two-phase procedure. These rays form a likelihood map from which the binary changed region map is computed. RESULTS: Experimental studies on four pairs of clinical lung and liver CT scans with simulated changed regions yield a mean changed region recall rate > 86%and a mean precision rate > 83%when detecting large changes with low contrast, and high contrast changes, even when small. The new method outperforms image space methods using prior image constrained compressed sensing (PICCS) reconstruction, particularly for small, low contrast changes (recall = 15.8%, precision = 94.7%). CONCLUSION: Our method for automatic change detection in sparse-view repeat CT scans with non-rigid deformations may assist radiologists by highlighting the changed regions and may obviate the need for a high-quality repeat scan image when no changes are detected.

This study aims to see how the level of effectiveness of content marketing communication to consumers at the Bajumantanmoo online store in driving repeat purchase decisions. This research uses quantitative methods with a descriptive approach. Samples were taken using purposive sampling technique and data were collected through questionnaires to the sample. The data then tested using the validity test with the t table value (0.3297) and the reliability test with a Cornbach alpha value of 1.08. Based on the research results, it was found that the persuasion indicator was the content marketing indicator with the least value in encouraging consumers to make repeat purchases.

We distinguish between models using pre-steady state fluorescence kinetics to visualize transiently misaligned DNA intermediates and nucleotide incorporation products formed by DNA polymerases adept at making small frameshift mutationsin vivo. Human polymerase (pol) μ catalyzes Streisinger slippage exclusively in repetitive DNA, requiring as little as a dinucleotide repeat.Escherichia colipol IV uses dNTP-stabilized misalignment in identical repetitive DNA sequences, revealing that pol μ and pol IV use different mechanisms in repetitive DNA to achieve the same mutational end point. In non-repeat sequences, pol μ switches to dNTP-stabilized misalignment. pol β generates –1 frameshifts in “long” repeats and base substitutions in “short” repeats. Thus, two polymerases can use two different frameshift mechanisms on identical sequences, whereas one polymerase can alternate between frameshift mechanisms to process different sequences.

Direct visualization of bile and pancreatic duct pathology is proving to be beneficial in patients where previous techniques have failed. Recent advancements in technology and the development of the SpyGlass system have led to an increased use of cholangioscopy. It is already known that SpyGlass is beneficial in patients with difficult bile duct stones and indeterminate biliary lesions through the use of targeted lithotripsy and visually guided biopsy. Cholangioscopy allows the visualization of hidden stone and guide wire placement across difficult strictures and selective cannulation of the intrahepatic and cystic ducts. It is also demonstrating its utility in investigational applications such as post-liver transplant and primary sclerosing cholangitis stricture treatment, evaluation of hemobilia, and guided radiofrequency ablation of ductal tumors. In addition to having clinical utility, cholangioscopy may also be cost-effective by limiting the number of repeat procedures. Cholangioscopy overall has similar complication rates compared to other standard endoscopic retrograde cholangioscopy (ERCP) techniques, but there may be higher rates of cholangitis. This could be mitigated with prophylactic antibiotic use, and overall, cholangioscopy has similar complication rates compared to other techniques.

Kumala Island is a leading tourist attraction in Kutai Kartanegara Regency. The purpose of this study was to determine the effect of attractions, accessibility, amenities, and quality of service on the interest in repeat visits to Kumala Island, Kutai Kartanegara Regency. This study uses primary data by distributing questionnaires. The population in this study are visitors who have visited Kumala Island, with a sample of 100 respondents using thetechnique Accidental Sampling. The data analysis of this research uses multiple linear analysis using SPSS version 25.The results of this study prove that the Attraction variable partially has a positive and significant effect on Tourist Revisit Interest. Then the accessibility variable partially has a positive and significant effect on Tourist Revisit Interests. The Amenity variable partially has a positive and significant effect on Tourist Revisit Interests. Furthermore, the service quality variable partially has no positive and insignificant effect on Tourist Revisit Interests. Furthermore, simultaneously attractions, accessibility, amenities and quality of service partially have a positive and significant effect on Tourist Revisit Interests. The conclusion of this study explains that partially between the variables of attraction, accessibility, amenity have a positive and significant influence on the interest of repeat tourists. While the quality of service does not have a positive and insignificant effect on the interest of repeat tourists, but simultaneously the variables of attraction, accessibility, amenity and service quality have a positive and significant influence on the interest of repeat tourists.

ABSTRACT Mus dunni endogenous virus (MDEV) can be activated fromM. dunni cells by exposing the cells to hydrocortisone or 5-iodo-2′-deoxyuridine. Interference analysis has revealed that MDEV uses a receptor for cell entry that is different from those used by other murine retroviruses. The entire genome has now been sequenced, revealing a long terminal repeat (LTR)-gag-pol-env-LTR structure typical of simple retroviruses of the murine leukemia virus genus, with no additional open reading frames between env and the 3′ LTR. The LTRs and other noncoding regions of MDEV are most closely related to those of VL30 elements, while the majority of the coding sequences are most closely related to those of gibbon ape leukemia virus. MDEV represents the first example of a naturally occurring, replication-competent virus with sequences closely related to VL30 elements. The U3 region of MDEV contains six nearly perfect 80-bp repeats and the beginning of a seventh, and the region expected to contain the packaging sequence contains approximately four imperfect 33-bp repeats. The receptor specificity domains of the envelope are unique among retroviruses and show no apparent similarity to regions of known proteins.

Long-read-only bacterial genome assemblies usually contain residual errors, most commonly homopolymer-length errors. Short-read polishing tools can use short reads to fix these errors, but most rely on short-read alignment which is unreliable in repeat regions. Errors in such regions are therefore challenging to fix and often remain after short-read polishing. Here we introduce Polypolish, a new short-read polisher which uses all-per-read alignments to repair errors in repeat sequences that other polishers cannot. Polypolish performed well in benchmarking tests using both simulated and real reads, and it almost never introduced errors during polishing. The best results were achieved by using Polypolish in combination with other short-read polishers.

DNA can assemble into non-B form structures that stall replication and cause genomic instability. One such secondary structure results from an inverted DNA repeat that can assemble into hairpin and cruciform structures during DNA replication. Quasipalindromes (QP), imperfect inverted repeats, are sites of mutational hotspots. Quasipalindrome-associated mutations (QPMs) occur through a template-switch mechanism in which the replicative polymerase stalls at a QP site and uses the nascent strand as a template instead of the correct template strand. This mutational event causes the QP to become a perfect or more perfect inverted repeat. Since it is not fully understood how template-switch events are stimulated or repressed, we designed a high-throughput screen to discover drugs that affect these events. QP reporters were engineered in the Escherichia coli lacZ gene to allow us to study template-switch events specifically. We tested 700 compounds from the NIH Clinical Collection through a disk diffusion assay and identified 11 positive hits. One of the hits was azidothymidine (zidovudine, AZT), a thymidine analog and DNA chain terminator. The other ten were found to be fluoroquinolone antibiotics, which induce DNA-protein crosslinks. This work shows that our screen is useful in identifying small molecules that affect quasipalindrome-associated template-switch mutations. We are currently assessing more small molecule libraries and applying this method to study other types of mutations.

The low rate of repeat visits in exclusive dental clinics is an interesting problem to study. From the available data, as well as interviews with several owners of exclusive dental clinics in cities of Jakarta, Surabaya, Samarinda and Banjarmasin in Indonesia as well as from the initial observations at exclusive dental clinics in South Jakarta, we found that the rate of repeat visits for existing patients was much lower than for new patients. From previous studies, it was also found that patient visits to the dental clinic were very low. This was influenced by the low level of patient satisfaction with the dental clinic. Their article mentions that retaining existing patients who make repeat visits at health clinics is about five times more profitable than serving new patients. This study aims to determine the effect of quality, service price and patient satisfaction on patient loyalty which influences patients to make repeat visits to exclusive dental clinics in South Jakarta, Indonesia. The subjects of this study were patients who had made at least two visits to the same exclusive dental clinics in South Jakarta, as well as patients who had previously visited other exclusive dental clinics. In this study, the approach used is a quantitative approach, and the type of data used in this study uses quantitative data in the form of questionnaire tabulations and qualitative data in the form of the research object general observations. We collect primary data through respondents' answers to questionnaires and secondary data sources through information on the population of DKI Jakarta Selatan and the characteristics of the middle-upper economic community.

At a time when criminal justice systems in Europe and North America increasingly seek to utilise the epistemic authority of a variety of sciences in support of the apprehension and prosecution of suspects and offenders, genetic science and recombinant DNA technology are often singled out for particular approbation. Indeed, the development and application of DNA profiling has been widely described as the “greatest breakthrough in forensic science since fingerprinting.”Prior to the implementation of PCR based extraction and amplification methods in the 1990's, the initial uses of DNA fingerprinting (based on Multiple and Single Locus Probes) were largely confined to reactive forensic casework.

Phytopathogenic pseudomonads are widespread in the world and cause a wide range of plant diseases. In this work, we describe the Pseudomonas phage Pf-10, which is a part of the biopesticide “Multiphage” used for bacterial diseases of agricultural crops caused by Pseudomonas syringae. The Pf-10 chromosome is a dsDNA molecule with two direct terminal repeats (DTRs). The phage genomic DNA is 39,424 bp long with a GC-content of 56.5%. The Pf-10 phage uses a packaging mechanism based on T7-like short DTRs, and the length of each terminal repeat is 257 bp. Electron microscopic analysis has shown that phage Pf-10 has the podovirus morphotype. Phage Pf-10 is highly stable at pH values from 5 to 10 and temperatures from 4 to 60 °C and has a lytic activity against Pseudomonas strains. Phage Pf-10 is characterized by fast adsorption rate (80% of virions attach to the host cells in 10 min), but has a relatively small number of progeny (37 ± 8.5 phage particles per infected cell). According to the phylogenetic analysis, phage Pf-10 can be classified as a new phage species belonging to the genus Pifdecavirus, subfamily Studiervirinae, family Autographiviridae, order Caudovirales.

Patterned fabrics are generally constructed from the periodic repetition of a primitive pattern unit. Repeat pattern segmentation of printed fabrics has a very significant impact on the pattern retrieval and pattern defect detection. In this paper, we propose a new approach for repeat pattern segmentation by employing the adaptive template matching method. In contrast to the traditional method for template matching, the proposed algorithm first selects an adaptive size template image in the repeat pattern image based on the size of the original image and its local maximum edge density. Then it uses the sum of absolute differences as the matching features to identify the matched regions in the original image, and the minimum envelope border of the primitive pattern, typically as a parallelogram, can be determined from the results of the four adjacent matched templates. Finally, image traversal base on the obtained parallelogram is implemented over the original image using minimum information loss theory to produce a well-segmented primitive pattern with a complete edge structure. The results from the experiments conducted using an extensive database of real fabric images show that the proposed algorithm has the advantage of rotation invariance and scaling invariance and will not be affected when the background or foreground color is changed.

Speech-recognition tests are a routine component of the clinical hearing evaluation. The most common type of test uses recorded monosyllabic words presented in quiet. The interpretation of test scores relies on an understanding of the variance of repeated tests. Confidence intervals are useful for determining if two scores are significantly different or if the difference is due to the variability of test scores. Because the response to each test item is binary, either correct or incorrect, the binomial distribution has been used to estimate confidence intervals. This method requires that test scores be independent. If the scores are not independent, the binomial distribution will not accurately estimate the variance of repeated scores. A previously published dataset with repeated scores from normal-hearing and hearing-impaired listeners was used to derive confidence intervals from actual test scores in contrast to the predicted confidence intervals in earlier reports. This analysis indicates that confidence intervals predicted by the binomial distribution substantially overestimate the variance of repeated scores resulting in erroneously broad confidence intervals. High correlations were found for repeated scores, indicating that scores are not independent. The interdependence of repeated scores invalidates confidence intervals predicted by the binomial distribution. Confidence intervals and confidence levels for repeated measures were determined empirically from measured test scores to assist in interpreting differences between repeat scores.

ABSTRACT Bacillus anthracis is one of the most genetically homogeneous pathogens described, making strain discrimination particularly difficult. In this paper, we present a novel molecular typing system based on rapidly evolving variable-number tandem repeat (VNTR) loci. Multiple-locus VNTR analysis (MLVA) uses the combined power of multiple alleles at several marker loci. In our system, fluorescently labeled PCR primers are used to produce PCR amplification products from eight VNTR regions in the B. anthracisgenome. These are detected and their sizes are determined using an ABI377 automated DNA sequencer. Five of these eight loci were discovered by sequence characterization of molecular markers (vrrC 1, vrrC 2,vrrB 1, vrrB 2, and CG3), two were discovered by searching complete plasmid nucleotide sequences (pXO1-aat and pXO2-at), and one was known previously (vrrA). MLVA characterization of 426 B. anthracis isolates identified 89 distinct genotypes. VNTR markers frequently identified multiple alleles (from two to nine), with Nei's diversity values between 0.3 and 0.8. Unweighted pair-group method arithmetic average cluster analysis identified six genetically distinct groups that appear to be derived from clones. Some of these clones show worldwide distribution, while others are restricted to particular geographic regions. Human commerce doubtlessly has contributed to the dispersal of particular clones in ancient and modern times.

This research aims to analyze the influence of company performance elements, auditor reputation and repeat audit on audit duration. The length of the audit is measured from the closing date of the financial year to the date the audit report was issued. The data used in this study are secondary data and sample selection using a purposive sampling method which consists of 320 companies listed on the Indonesia Stock Exchange (BEI) and publishes financial statements consistently in the 2014-2018 period. The method of analysis of this study uses multiple linear regression analysis. Based on the estimation results used it can be seen that only the auditor's reputation variable has no significant effect on the length of the audit, while the other variables are proven to have significant influence as follows: (i) Profitability has a significant negative effect on the length of the audit, (ii) Solvency has a positive influence and significant to the length of the audit, (iii) the size of the company has a negative and significant effect on the length of the audit, (iv) repeat audit has a negative and significant effect on the length of the audit.
