The twin-arginine translocation (Tat) pathway transports folded proteins across energetic membranes. Numerous Tat substrates contain co-factors that are inserted before transport with the assistance of redox enzyme maturation proteins (REMPs), which bind to the signal peptide of precursor proteins. How signal peptides are transferred from a REMP to a binding site on the Tat receptor complex remains unknown. Since the signal peptide mediates both interactions, possibilities include: i) a coordinated hand-off mechanism; or ii) a diffusional search after REMP dissociation. We investigated the binding interaction between substrates containing the TorA signal peptide (spTorA) and its cognate REMP, TorD, and the effect of TorD on the in vitro transport of such substrates. We found that Escherichia coli TorD is predominantly a monomer at low micromolar concentrations (dimerization KD > 50 μM), and this monomer binds reversibly to spTorA (KD ≈ 1 μM). While TorD binds to membranes (KD ≈ 100 nM), it has no apparent affinity for Tat translocons and it inhibits binding of a precursor substrate to the membrane. TorD has a minimal effect on substrate transport by the Tat system, being mildly inhibitory at high concentrations. These data are consistent with a model in which the REMP-bound signal peptide is shielded from recognition by the Tat translocon, and spontaneous dissociation of the REMP allows the substrate to engage the Tat machinery. Thus, the REMP does not assist with targeting to the Tat translocon, but rather temporarily shields the signal peptide.

Introduction. The relevance of the study is determined by the active implementation of digital technologies in all life activities of people, including in the education system. Reforming the modern school in a digital environment has both a positive and a negative impact to the subjects of the educational process. The purpose of the study was to identify the relationship between the attitude of students and teachers of Russian universities to digital transformations of education and the characteristics-motivational sphere of the subjects of the educational process, namely the degree of significance and satisfaction of deficit and everyday needs. Materials and methods. The empirical study involved 150 cadets and students of the Voronezh Institute of the Ministry of Internal Affairs of Russia and the Siberian Fire and Rescue Academy of the State Fire Service of the EMERCOM of Russia; 51 lecturers from a number of Russian universities. To study the need-motivational sphere of the subjects of the educational process, the «A. Maslow's Motivational Questionnaire» was used. Revealing the attitudes of students and teachers to the digital processing process was carried out using the scale modified for research purposes, the Technological Readiness Index (TRI). Results. It was revealed that motivators and inhibitors of digital readiness of subjects of the educational process are associated with the significance and satisfaction of different needs. Students' optimism and innovativeness correlate with the significance of existential needs (respectively, remp = 0.201, remp > r0,01; remp = 0.237, remp > r0.01) and their realization (respectively, remp = 0.197, remp > r0,05; remp = 0.223, remp > r0.01). At the same time, inverse relationships were found between discomfort and satisfaction of scarce needs (remp = -0,160, remp > r0.05), as well as distrust of digital technologies and the realizability of existential needs (remp = -0,163, remp > r0.05). Among teachers, motivators of digital readiness correlate with the realization of deficit needs (optimism: remp = 0.285, remp > r0.05; innovativeness: remp = 0.298, remp > r0.05). The innovativeness of teachers in relation to the digitalization of education correlates with the significance and satisfaction of existential needs (respectively, remp = 0.276, remp > r0.05; remp = 0.353, remp > r0.05). Conclusion. The study confirmed the hypothesis about the correlation between the personality traits of the need-motivational sphere and the attitude of the educational process to the digitalization of education. In view of the fact that a significant part of students and teachers experience discomfort and insecurity associated with the possible consequences of digital processing in education, the integration of traditional and online education systems, the preservation and development of humanistic ideas of pedagogy in the digital era are important in the educational process.

The aim of the study was to evaluate REM sleep parameters, especially the temporal characteristics of rapid eye movement activity, in depressed patients, and to compare three different methods for scoring of REM density. The sleep of 15 nonmedicated depressed patients and 13 healthy controls was recorded during two consecutive nights. Sleep recordings were scored by raters blinded to the diagnosis. In comparison to healthy controls depressed patients showed an increased REM density and increased REM activity. Both groups differed also regarding the pattern of REM density changes between REM sleep periods (REMPs). Whereas in healthy controls REM density in the first REMP was significantly lower than in the successive REMPs, no such difference was found in depressed patients. On visual inspection we failed to find any significant differences in the time course of REM activity within the first REMP in depressed patients. All applied methods for scoring of REM density distinguished depressed patients from healthy controls with comparable accuracy.

An accurate description of the electron correlation energy in closed- and open-shell molecules is shown to be obtained by a second-order perturbation theory (PT) termed REMP. REMP is a hybrid of the Retaining the Excitation degree (RE) and the Møller–Plesset (MP) PTs. It performs particularly encouragingly in an orbital-optimized variant (OO-REMP) where the reference wavefunction is given by an unrestricted Slater determinant whose spin orbitals are varied such that the total energy becomes a minimum. While the approach generally behaves less satisfactorily with unrestricted Hartree–Fock references, reasonable performance is observed for restricted Hartree–Fock and restricted open-shell Hartree–Fock references. Inclusion of single excitations to OO-REMP is investigated and found—as in similar investigations—to be dissatisfying as it deteriorates performance. For the non-multireference subset of the accurate W4-11 benchmark set of Karton et al. [Chem. Phys. Lett. 510, 165–178 (2011)], OO-REMP predicts most atomization and reaction energies with chemical accuracy (1 kcal mol−1) if complete-basis-set extrapolation with augmented and core-polarized basis sets is used. For the W4-11 related test-sets, the error estimates obtained with the OO-REMP method approach those of coupled-cluster with singles, doubles and perturbative triples [CCSD(T)] within 20%–35%. The best performance of OO-REMP is found for a mixing ratio of 20%:80% MP:RE, which is essentially independent of whether radical stabilization energies, barrier heights, or reaction energies are investigated. Orbital optimization is shown to improve the REMP approach for both closed and open shell cases and outperforms coupled-cluster theory with singles and doubles (CCSD), spin-component scaled Møller-Plesset theory at second order (SCS-MP2), and density functionals, including double hybrids in all the cases considered.

Objectives: Based on findings from our multi-center, double-blinded randomized controlled trial comparing arthroscopic Bankart repair with/without remplissage for the treatment of traumatic recurrent shoulder instability there is a greater risk of postoperative recurrent instability in patients without a remplissage at two years follow-up. The objective of the current study is to determine whether our short-term findings change with medium-term follow-up. Methods: This was a double-blinded, randomized clinical trial with two 1:1 parallel groups conducted at two sites. Patients were randomized intra-operatively to an arthroscopic Bankart with remplissage (REMP) or isolated Bankart repair (NO REMP) between 2011 and 2017. Inclusion criteria were patients 14 years or older diagnosed with traumatic anterior shoulder instability with a glenoid defect >15% and the presence of a Hill-Sachs defect (of any size). Patients were contacted by telephone (Spring 2020) and asked standardized questions to determine whether any additional information regarding subluxations, dislocation or additional surgery occurred since their two-year follow-up. “Recurrent instability” was defined as patient-reported dislocation, or two or more episodes of subluxation. Kaplan-Meier survival analyses were performed to assess survival distribution between groups. For the time factor in the survival analysis, the number of months from time of surgery to outcome (either failure or no failure) was based on the time of the long-term follow-up phone call, or from the time of last reported outcome based on clinical or study follow-ups, whichever was the greatest. These were conducted for: 1) recurrent instability and 2) dislocations. Odds ratios with 95% confidence intervals were also calculated. Results: Study groups were similar at baseline with respect to age, gender and BMI. Fifty-four patients were randomized to each study group at the time of the original study. , with 52 REMP and 50 NO REMP included in the analyses up to 24-months post-operative. Of those, 36 from each group were available for mid-term follow-up. Mean months (mean) from surgery to last follow-up was 53.8 for REMP and 49.3 for NO REMP. The rate of postoperative recurrent instability at medium term follow-up was 10% (5/52) in the REMP group at an average of 24 months and 30% (15/50) in NO REMP at an average of 19.5 months (p=0.010). The odds ratio of recurrent instability in the NO REMP group relative to the REMP group was 4.029 (1.337-12.135; p=0.010) and the survival curve was significantly different favouring REMP (χ2 = 6.958, P=0.008; Figure 1). With respect to dislocations only, the odds ratio in the NO REMP group relative to REMP was 3.385 (0.999-11.463; p=0.041) and the survival curve was also significantly different favouring the REMP (χ2 = 4.412, p=0.036; Figure 2). Conclusions: At medium-term follow-up, patients undergoing a Bankart repair with remplissage have a better rate of survival than those with an isolated Bankart repair. Objective To document that regional anesthesia of the infraorbital and inferior alveolar nerves would abolish reflex-evoked muscle action potentials (REMP) in the digastricus muscle during noninvasive stimulation of tooth pulp in halothane-anesthetized dogs. Design Prospective study. Animals 9 healthy female dogs between 2 and 6 years old. Procedure Dogs were anesthetized using halothane. An alligator clip anodal electrode was attached to the tooth to be stimulated, and a platinum needle cathodal electrode was inserted in adjacent gingival mucosa. The cathodal and anodal electrodes were moved to the left upper and lower canine, fourth premolar, and first molar teeth for sequential stimulation. Baseline recording of REMP was made for each tooth. Catheters were inserted percutaneously in the infraorbital and mandibular canals. Saline (0.9% NaCl) solution was injected at each catheterized site in 3 control dogs, and chloroprocaine hydrochloride was injected at each catheterized site in 6 test dogs. Each tooth was stimulated every 10 minutes for 90 minutes (test dogs) or every 10 minutes for 30 minutes and at 90 minutes (control dogs), and REMP was recorded. Results REMP was abolished within 10 minutes in all test dogs, except during stimulation of the lower first molar in 1 dog. In 4 dogs, duration of blockade was less than 90 minutes. The REMP was not restored within 90 minutes for the upper teeth in 1 dog and within 2 hours for all teeth in another dog. At 24 hours, REMP was restored for all teeth except the lower left canine in 1 dog. The REMP was restored for the lower left canine in that dog at 96 hours. The REMP was not abolished at any time in control dogs. Clinical Implications Regional anesthesia of the infraorbital and inferior alveolar nerves may effectively provide analgesia for dental procedures in dogs. (J Am Vet Med Assoc 1997;211:1403–1405)

Background: Surgical management of anterior shoulder instability continues to evolve. The clinical effectiveness of arthroscopic Bankart repair with remplissage (REMP) in patients with increasing glenoid bone loss (GBL) has not been thoroughly compared with Latarjet. Purpose/Hypothesis: The purpose of this study was to compare 2-year outcomes of REMP versus open Latarjet in patients with >15% GBL. We hypothesized that there would be no difference. Study Design: Cohort study; Level of evidence, 3. Methods: A multicenter, retrospective study was performed on a consecutive series of patients who underwent primary REMP or Latarjet by 4 shoulder specialists between August 2013 and December 2019 for >15% GBL. Procedure performance was based on shared decision making between the surgeon and patient. Baseline and 2-year postoperative range of motion (ROM) and the following patient-reported outcomes (PROs) were collected: Western Ontario Shoulder Instability Index, Single Assessment Numeric Evaluation, and visual analog scale for pain. Return to sport, satisfaction, complications, recurrence, and revisions were reviewed. Results: A total of 47 patients were available for study, including 22 who underwent REMP and 25 who underwent Latarjet. Baseline demographic characteristics, ROM, and PROs were similar between the groups. Mean preoperative GBL (REMP 25.8% ± 7.8% vs Latarjet 25.1% ± 9.0%; P = .800) and off-track lesions (REMP 59.1% vs Latarjet 44.0%; P = .302) were similar. Postoperative change in external rotation favored Latarjet (10° ± 22° vs –4° ± 13°; P = .017). Postoperative PROs, return to the sports (86.4% vs 87.5%; P = .99), and satisfaction (95.5% vs 95.8%; P = .99) were similar between groups. There were no recurrent dislocations in either group. One infected hematoma was encountered in the Latarjet cohort. Conclusion: At short-term follow-up, the hypothesis was upheld. REMP and Latarjet provided similar outcomes for patients with >15% GBL, although REMP patients experienced slightly decreased external rotation. Although larger series and longer-term follow-ups are required, remplissage may be considered in patients with >15% glenoid bone loss.

Abstract Polymers exhibiting cyclic topology have attracted great interest over the past 30 years. Macrocycles or cyclopolymers with more than 20 repeating monomer units are considered exceptional candidates for thermoplastic engineering application due to unique properties in comparison to their linear analogues including large hydrodynamic radii and functional group density, heat resistance, good insulating ability and low intrinsic viscosity. Cyclic polymers are thus expected to exhibit improved physical and mechanical properties for certain applications due to the absence of end groups. Although synthetic challenges have historically limited research on cyclic polymers, recent developments in ruthenium catalyzed ring-expansion metathesis polymerization (REMP) have enabled the synthesis and the preliminary investigation of structure-property relationships of high molecular weight macrocycles. In REMP reactions the polymer formation is proposed to proceed through a transient macrocyclic complex in which both ends of the growing polymer chain remain attached to the Ru center. This article summarizes the recent discoveries on the field of REMP assisted cyclopolymer based material development.

Higher education institutions (HEIs) in South Africa are facing challenges arising as a result of the shift from traditional teaching activities to a combination of research and teaching. Increasing emphasis on research, an integral part of this transformation of higher education, has required HEIs to develop and implement capacity development strategies to enable those new to research to engage in research output; and mentorship is a strategy that is enjoying increasing popularity. This article explores the challenges faced in the implementation of a formal research mentorship programme (REMP), using the University of Johannesburg as a case study. A quantitative approach was adopted to obtain the perceptions of academic staff of REMP as a strategy for building research capacity. The findings reveal that whilst the REMP is strongly favoured by academics as a means of guiding novice researchers, key factors need to be addressed to ensure its success. This article examines the challenges for both mentors and mentees and suggests actions to help mentorship programmes make sustainable contributions to the development of research capacity.

Objectives: The purpose of this prospective randomized, double blinded controlled trial was to compare patient-reported outcomes and clinical results between arthroscopic Bankart repair with and without arthroscopic infraspinatus remplissage in patients with anterior shoulder instability with a Hill-Sachs lesion. Failure to recognize and address large Hill Sach’s defects during arthroscopic stabilization surgery for glenohumeral instability is known to lead to high rates of recurrence. Arthroscopic remplissage has evolved in recent years as a reproducible technique with a proposed benefit of decreased dislocations. However, there are no high level clinical studies to conclusively support its efficacy in reducing redislocations. Methods: One hundred and four patients, aged 14 years and older, with a confirmed Hill Sach’s lesion on ultrasound, CT or MRI, were randomized intraoperatively after confirming an engaging Hill Sach’s lesion to either undergo arthroscopic infraspinatus remplissage (REMP) or no remplissage during arthroscopic Bankart repair (NO REMP). Exclusion criteria included a glenoid defect >15% of the AP glenoid diameter, significant shoulder arthropathy, infection, or medical comorbidities. The primary outcome measure was the Western Ontario Shoulder Instability score (WOSI). Secondary outcomes included the Simple Shoulder Test (SST), the American Shoulder and Elbow Society standardized assessment of shoulder function (ASES), active range of motion, stability tests, and incidence of revision surgery. Study time points were pre-, 3-, 6-, 12-, and 24-months post-operative. Significance level was 0.05. Results: A summary of demographics and outcomes are presented in Table 1. There were 53 patients (45 men, 8 women) randomized to REMP and 52 (46 men, 6 women) patients to NO REMP. The groups were comparable with regard to age, body mass index (BMI), and gender distribution). Both groups demonstrated a similar improvement in all subjective scores over time to 12-months post-operative with no difference between the groups (collection of data to 24-months post-operative is ongoing until 2019). The WOSI significantly improved from pre- to 12-months post-operative in both study groups. There were no differences between groups at any time point. ASES scores and SST scores followed a similar pattern. Additionally, there were no differences in range of motion between groups at any time point. There were 2/53 re-dislocations in REMP postoperatively compared to 6/52 in NO REMP; this difference was not significant (p=0.161). There were no differences between groups in reports of limitations in participation in sport attributed to the operated shoulder up to 12-months post-operative. Conclusion: Based on this study, there is no difference in subjective outcome scores and redislocations rates between remplissage and no remplissage for an engaging Hill Sach’s lesion while performing arthroscopic Bankart stabilization. As data continues to be gathered to 24-months post-operative including MRI, longer term benefits or drawbacks may become evident. [Table: see text]

AbstractProtein folding and assembly into macromolecule complexes within the living cell are complex processes requiring intimate coordination. The biogenesis of complex iron sulfur molybdoenzymes (CISM) requires use of a system specific chaperone – a redox enzyme maturation protein (REMP) – to help mediate final folding and assembly. The CISM dimethyl sulfoxide (DMSO) reductase is a bacterial oxidoreductase that utilizes DMSO as a final electron acceptor for anaerobic respiration. The REMP DmsD strongly interacts with DMSO reductase to facilitate folding, cofactor-insertion, subunit assembly and targeting of the multi-subunit enzyme prior to membrane translocation and final assembly and maturation into a bioenergetic catalytic unit. In this article, we discuss the biogenesis of DMSO reductase as an example of the participant network for bacterial CISM maturation pathways.

Complex cofactor-containing Tat substrates acquire their cofactors prior to export, and substrate proteins actually require to be folded before transport can proceed. Thus, it is very likely that mechanisms exist to prevent wasteful export of immature Tat substrates or to curb competition between immature and mature substrates for the transporter. Here we assess the primary sequence relationships between the accessory proteins implicated in this process during assembly of key respiratory enzymes in the model prokaryote Escherichia coli. For each respiratory enzyme studied, a redox enzyme maturation protein (REMP) was assigned. The main finding from this review was the hitherto unexpected link between the Tat-linked REMP DmsD and the nitrate reductase biosynthetic protein NarJ. AbstractMulti-homed devices such as smartphones, tablets and laptops are enabled with multiple heterogeneous interfaces available for transmission. Those interfaces can be utilized for simultaneous transmission of a single TCP flow using Multipath TCP (MPTCP). MPTCP is a protocol that is designed to increase end-to-end throughput and reliability of communications by splitting data through multiple parallel paths. Although delay in MPTCP enhanced significantly in the recent years, high number of data transmissions remains an issue. In this paper, we reduce MPTCP delay by reducing the number of transmissions using Opportunistic Routing (OR) technique. OR is a routing model used to increase the delivery rate and reliability of data transmission in wireless networks by using the broadcasting method. This enables each subflow data to be delivered by multiple relays. We adapted OR on a number of MPTCP protocols namely, traditional MPTCP, Multipath TCP Traffic Splitting Control (MPTCP-TSC) and Redundant MPTCP (ReMP TCP) in an Internet of Things (IoT) environment. The results show that OR-based MPTCP schemes outperform existing schemes. We further compared the OR-based MPTCP protocols in terms of startup delay and energy efficiency. We found that ReMP TCP is better than other schemes in all scenarios.

Abstract In children, sleep disordered breathing (SDB) is associated with changes in cardiac and vascular remodeling and hence may alter cardiac rhythm. Heart rate variability (HRV) measured during different sleep stages and at discreet times across the night, where vascular tone is known to change, provides an opportunity to better understand the effect of SDB on the cardiac function. 50 children diagnosed with SDB and 51 healthy children underwent overnight polysomnography to determine sleep staging. HRV (mean NN, SDNN, RMSSD, LF, HF, and LF:HF) was determined for the following segments pre-sleep; 3 slow wave sleep and 3 segments during rapid eye movement sleep (SWS1, SWS2, REM3, REM2, REM1). Children with SDB demonstrated higher heart rate (decreased mean NN) in all sleep segments. All HRV variables were similar between groups pre-sleep and REM3 and SWS3. LF and LF:HF were significantly lower in SWS1&2 and REM1 while as were SDNN and rMSSD were lower in the SDB group in REM1&2. LF remained low in the SDB group but rose to pre sleep levels in the control group. Children with SDB have increased heart rate across the night even when HRV is similar between the groups. This suggests intrinsic changes to the cardiac components that determine heart rate. The HRV difference between groups was greatest post acrophase (body temperature dropping) and post nadir (body temperature rising) of the circadian cycle. We propose that impaired peripheral vascular control and sustained cardiac remodelling may underlie the heart rate and HRV changes observed in children with SDB.

This paper investigates the effects of the lagged real earnings management on the firms’ future profitability using a panel dataset (for the years 2012–2017) from the Jordanian industrial companies listed in the Amman Stock Exchange (ASE). We follow Roychowdhury (2006) to measure real earnings management using two proxies: abnormal sales (REMS) and abnormal production (REMP) in the regression analysis. Our findings reveal that real earnings management through abnormal sales has an insignificant effect on the firms’ future profitability. However, we document evidence that firms’ future profitability is adversely and significantly affected by real earnings management through abnormal production. We contribute to the ongoing debate in the literature of real earnings management and its ramifications on firms’ profitability, specifically in the context of developing countries. This research provides implications for policymakers, investors and managers regarding the potential consequences of channel stuffing practices at the different stages of the supply chain on the firm’s future profitability. Future research is suggested to focus on how real earnings management can possibly disrupt supply chains.

In Bacillus subtilis, the biofilm matrix is composed of an extracellular polysaccharide and the secreted protein TasA. Expression of both of the matrix components is repressed by the DNA-binding master regulator, SinR. Here we identify two small protein regulators of the extracellular matrix: RemA (formerly YlzA) and RemB (formerly YaaB). Mutation of RemA or RemB impairs pellicle formation, complex colony architecture, and motility inhibition in a sinR mutant background. Both proteins are required for the activation of the matrix biosynthesis operons and appear to act in parallel to SinR and two other known biofilm regulators, AbrB and DegU.

Measurement of monocarboxylate transport kinetics in a range of cell types has provided strong circumstantial evidence for a family of monocarboxylate transporters (MCTs). Two mammalian MCT isoforms (MCT1 and MCT2) and a chicken isoform (REMP or MCT3) have already been cloned, sequenced and expressed, and another MCT-like sequence (XPCT) has been identified. Here we report the identification of new human MCT homologues in the database of expression sequence tags and the cloning and sequencing of four new full-length MCT-like sequences from human cDNA libraries, which we have denoted MCT3, MCT4, MCT5 and MCT6. Northern blotting revealed a unique tissue distribution for the expression of mRNA for each of the seven putative MCT isoforms (MCT1-MCT6 and XPCT). All sequences were predicted to have 12 transmembrane (TM) helical domains with a large intracellular loop between TM6 and TM7. Multiple sequence alignments showed identities ranging from 20% to 55%, with the greatest conservation in the predicted TM regions and more variation in the C-terminal than the N-terminal region. Searching of additional sequence databases identified candidate MCT homologues from the yeast Saccharomyces cerevisiae, the nematode worm Caenorhabditis elegans and the archaebacterium Sulfolobus solfataricus. Together these sequences constitute a new family of transporters with some strongly conserved sequence motifs, the possible functions of which are discussed.

Using our new 3D relativistic electromagnetic particle (REMP) code parallelized with MPI, we investigated long-term particle acceleration associated with a relativistic electron–positron jet propagating in an unmagnetized ambient electron–positron plasma. We have also performed simulations with electron-ion jets. The simulations were performed using a much longer simulation system than our previous simulations in order to investigate the full nonlinear stage of the Weibel instability for electron–positron jets and its particle acceleration mechanism. Cold jet electrons are thermalized and ambient electrons are accelerated in the resulting shocks for pair plasma case. Acceleration of ambient electrons leads to a maximum ambient electron density three times larger than the original value for pair plasmas. Behind the bow shock in the jet shock strong electromagnetic fields are generated. These fields may lead to time-dependent afterglow emission. We calculated radiation from electrons propagating in a uniform parallel magnetic field to verify the technique. We also used the new technique to calculate emission from electrons based on simulations with a small system with two different cases for Lorentz factors (15 and 100). We obtained spectra which are consistent with those generated from electrons propagating in turbulent magnetic fields with red noise. This turbulent magnetic field is similar to the magnetic field generated at an early nonlinear stage of the Weibel instability.

PURPOSE This study compares outcomes of autologous bone marrow transplantation (ABMT) in patients with acute myeloid leukemia (AML) in untreated first relapse (REL1) or in second complete remission (REM2). PATIENTS AND METHODS Forty-seven patients with AML in REL1 (n = 21) or in REM2 (n = 26) were treated with busulfan (BU) and cyclophosphamide (CY) with or without total-body irradiation (TBI) followed by ABMT. All REL1 patients and four REM2 patients had marrow stored during first remission (REM1). Twenty-seven had marrow stored with and 20 without treatment in vitro with 4-hydroperoxycyclophosphamide (4-HC). Eighteen patients received BU and CY and 29 received BU, CY, and TBI. REL1 patients relapsed within a median of 9 months (range, 2 to 26) after marrow harvest and were transplanted a median of 30 days (range, 9 to 87) from detection of relapse. RESULTS With a median follow-up of 2.1 years (range, 0.4 to 5.3), 19 patients survive in remission (10 of 21 in REL1; nine of 26 in REM2). The actuarial probabilities of relapse-free survival at 2 years for patients transplanted in REL1 and REM2 were 45% +/- 22% and 32% +/- 18%, respectively (P = .33). The corresponding probabilities of relapse were 30% +/- 26% and 44% +/- 23%, respectively (P = .45). No conclusions could be drawn about the benefits of adding TBI to BU plus CY. There were no significant differences in neutrophil or platelet recovery or in posttransplant probabilities of relapse and nonrelapse mortality between patients who received marrow treated or not treated with 4-HC. CONCLUSION These results suggest that ABMT may produce long-term leukemia-free survival in approximately one third of patients with AML in REL1 or in REM2. There is no apparent clinical advantage in attempting to obtain second remissions in relapsed patients before ABMT if marrow has been cryopreserved during REM1. Although a strategy of transplantation in REL1 has advantages for the patient, such an approach involves the storage of marrow, which may not be used, and is impractical without the coordinated support of the treating physician, the patient, and the marrow transplant center.

ABSTRACT The ability to attach to host ligands is a well-established pathogenic factor in invasive Staphylococcus aureusdisease. In addition to the family of adhesive proteins bound to the cell wall via the sortase A (srtA) mechanism, secreted proteins such as the fibrinogen-binding protein Efb, the extracellular adhesion protein Eap, or coagulase have been found to interact with various extracellular host molecules. Here we describe a novel protein, the extracellular matrix protein-binding protein (Emp) initially identified in Western ligand blots as a 40-kDa protein due to its broad-spectrum recognition of fibronectin, fibrinogen, collagen, and vitronectin. Emp is expressed in the stationary growth phase and is closely associated with the cell surface and yet is extractable by sodium dodecyl sulfate. The conferring gene emp (1,023 nucleotides) encodes a signal peptide of 26 amino acids and a mature protein of a calculated molecular mass of 35.5 kDa. Using PCR,emp was demonstrated in all 240 S. aureusisolates of a defined clinical strain collection as well as in 6S. aureus laboratory strains, whereas it is lacking in all 10 S. epidermidis strains tested. Construction of an allelic replacement mutant (mEmp50) revealed the absence of Emp in mEmp50, a significantly decreased adhesion of mEmp50 to immobilized fibronectin and fibrinogen, and restoration of these characteristics upon complementation of mEmp50. Emp expression was also demonstrable upon heterologous complementation of S. carnosus. rEmp expressed in Escherichia coli interacted with fibronectin, fibrinogen, and vitronectin in surface plasmon resonance experiments at a K d of 21 nM, 91 nM, and 122 pM, respectively. In conclusion, the biologic characterization of Emp suggests that it is a member of the group of secreted S. aureus molecules that interact with an extended spectrum of host ligands and thereby contribute to S. aureuspathogenicity.

Besides, mistakes and irregularities in between classification of same data can be recurrent. Therefore, there is a great use of automatic scoring system to support reliable classification. The proposed work provides an insight to use the automatic scheme which is based on real time EMG signals. EMG is an electro neurological diagnostic tool which evaluates and records the electrical activity generated by muscle cells. The sleep scoring analysis can be applied by recording Electroencephalogram (EEG), Electromyogram (EMG), and Electrooculogram (EOG) based on epoch which is defined as a period of 30 second length segments, and this method of sleep scoring system is also called polysomnography test or PSG test. The standard database of EMG signals was collected from different hospitals in sleep laboratory which gives the different stages of sleep. These are Waking, Non-REM1 (stage-1), Non-REM2 (stage-2), Non-REM3 (stage-3), REM. The main motive of the proposed work is the synchronization of EEG, EMG, EOG in order to understand different stages of sleep when they are simultaneously recorded. The procedure can be useful in clinics, particularly for scientists in studying the wakefulness and sleep stage correlation and thus helps in diagnosing some sleep disorders.

Though Africa is rich in a wide range of energy resources including solar, bio, natural gas, oil, coal and Uranium, the continent is far from energy self-sufficiency. Addressing climate change will require deep and quick reductions in fossil fuel use so that the systems developed around producing, transporting, and consuming energy are decarbonized by the middle of the century. In the ongoing age, sustainable power source has taken another swing to limelight on the planet, particularly in developed and emerging nations, as it assumes a noteworthy part both in economy and the general job of the world. Significantly, Nigeria an oil-rich country, comes as no surprise that almost all of her energy consumption comes from non-renewable energy sources as coal, natural gas and oil, and as such it is highly vulnerable to shocks due to overdependence on the fossil sources; often time is controlled by the international market. On the whole, the fossil fuel is expected to span only but a millennium (1700-2700) of human civilization while the imperative of an energy shortage situation is felt in every sector of the country considering the poor electricity consumptions in the country, which has reflected on the country’s economy and productivity rate. In revamping the economic sectors in Nigeria, the need for an alternative energy sources that is augmentable in supply keeping in view sustainable development as the hallmark for all sector development. Thus far, Nigeria ought to likewise be opened to universal investments as this would help support the improvement of its assets. This paper, therefore, supports no other sources but renewable energy in promoting the countries productivity at all segments. It further stressed on the implementation of the country’s Renewable Energy Master Plan (REMP) to meet global competitors by the year 2030. Similarly, senior political figures, policymakers and CEOs should engage in a policy dialogue by identifying unique opportunities and best practices for developing and investing in Nigeria and in Africa’s energy markets for “…without this energy supply, the sophisticated skills of the industrial world are merely a burden in the struggle for survival.”

DNA methylation is an epigenetic modification involved in lineage specification and cellular differentiation that has distinct patterns by cell type. Once defined, cell-specific DNA methylation patterns can be leveraged as biomarkers of cell types in deconvolution approaches where cell proportions are inferred from measured composite DNAm signals in common biospecimens like blood and tumor. While epigenetic modifications, specifically methylation of DNA, are known to contribute to carcinogenesis and disease progression, to date there has been limited application of approaches accounting for cellular heterogeneity in the ovarian tumor microenvironment. Adjusting for cellular heterogeneity addresses potential confounding by cell type in bulk DNA methylation analyses. Using publicly available HGSOC tumor data (GSE133556) and GTEx normal ovarian data, we applied Hierarchical Tumor Immune Microenvironment Epigenetic Devonvolution (HiTIMED) to profile cell types in the tumor microenvironment. We also performed repetitive element methylation prediction using the REMP package and copy number variation analysis using the conumee package in R. DNA methylation data from all tumor (n = 99) and nontumor normal samples (n=111) was deconvoluted into six major cell types: tumor, lymphocyte, myeloid, endothelial, epithelial, and stromal. Data from HGSOC tumors were divided into tumor high (<25% tumor burden) and tumor low cell proportions (>25% tumor burden). Assessing non-tumor cell proportions, we identified significant differences in lymphocyte, endothelial, epithelial, and stromal cell type proportions in HGSOC samples with a high tumor burden compared to normal, and we identified significant differences in lymphocyte, endothelial, epithelial, stromal, and myeloid cell type in HGSOC samples with low tumor burden compared to normal. Assessing repetitive element methylation, we found that Alu repetitive elements are significantly hypomethylated in HGSOC samples in comparison to normal ovarian tissue. Performing copy number variation analysis, we identified regions of copy number gain and loss associated with HGSOC. Our study identifies tumor-specific DNA methylation alterations in high-grade serous ovarian carcinoma samples compared to normal ovarian tissue and demonstrates differences in specific cell proportions in the tumor ovarian microenvironment, which may play a greater role in ovarian cancer etiology. Abstract Background: Fetal cell origin DNA methylation signature (FCO) estimates the proportion of cells in a mixture of cell types that are of fetal origin. Machine learning-based methods can predict DNA methylation of locus-specific repetitive elements (RE) by learning surrounding genetic and epigenetic information. In this study, we applied the FCO signature and RE methylation prediction to clear cell renal cell carcinoma (ccRCC) samples to study FCO and RE alterations in ccRCC carcinogenesis and survival. Methods: The FCO algorithm was applied to 47 ccRCC tumor samples and four adjacent normal samples from the Dartmouth Renal Tumor Biobank. 319 ccRCC tumor samples and 160 adjacent normal samples from The Cancer Genome Atlas (TCGA) were used for external validation. Linear regression models were used to compare the FCO signal between ccRCC tumor and normal samples adjusting for sex and age. Cox proportional hazard models were employed to study FCO with overall survival outcome in ccRCC samples, adjusting for sex, age, and tumor stage. The survival groups were dichotomized to FCO = 0% vs. FCO > 0%. The REMP package from Bioconductor was used to project LINE1 element methylation in 47 ccRCC samples. LINE1 median level methylation was compared by tumor grade. Cox proportional hazard models were employed to study LINE1 methylation level as a continuous variable with overall survival in ccRCC samples, adjusting for sex, age, and tumor stage. Results: The FCO signal is significantly lower in Dartmouth Renal Tumor Biobank archived ccRCC tumor samples compared to adjacent normal samples (Δ = 42.9%, p = 5.7e-11). By comparing the overall survival between FCO = 0% and FCO > 0% groups, we observed significantly worse survival outcomes in ccRCC patients with zero levels of FCO (HR: 2.46, 95% CI: 1.16, 5.18). Similar results were observed in TCGA samples. A significant decrease of FCO was found in ccRCC tumor samples compared to adjacent normal samples (Δ = 32.9%, p = 2.2e-308). A 1% increase in FCO is significantly associated with a better survival outcome (HR: 0.97, 95%CI 0.95, 0.99). For RE methylation prediction, we observed that the median LINE1 methylation level decreases with ccRCC tumor grade. A higher level of LINE1 median methylation is significantly associated with a better survival outcome (p = 0.01). Conclusion: Normal kidney samples possess a fetal niche reflected by DNA methylation, while ccRCC tumorigenesis depletes the fetal cells in normal kidneys. The depletion of FCO is associated with worse survival outcomes in ccRCC. The repetitive element LINE1 methylation level decreases with tumor grade and is related to ccRCC survival outcomes.
