In this paper, a novel approach is proposed for learning visual object classifiers with only positive images. Both positive and negative examples are collected from the positive images, and the false negative examples are filtered out by the Ramp Loss function, which has a strong ability of suppressing the influence of outliers. Meanwhile, the Latent Structural Support Vector Machines are adopted to estimate the best bounding boxes due to the unavoidable annotation errors. In learning the sub-problems, an extremely fast learning algorithm, CCCP-SGD, is proposed to optimize the Ramp Loss-based SVMs based on the Stochastic Gradient Descent algorithm. Experiments demonstrate that the CCCP-SGD algorithm can reduce the computing time both in the training and predicting phases, especially with a significant speedup in training, whilst it can also yield robust generalization performance when dataset is noisy. Besides, the visual object classifiers learnt by our approach with only positive images almost have the same performance as the classifiers learned with both positive and negative examples in object detection tasks.

Abstract (sommario):
Ensemble techniques have recently been used to enhance the performance of machine learning methods. However, current ensemble techniques for classification require both positive and negative data to produce a result that is both meaningful and useful. Negative data is, however, sometimes difficult, expensive or impossible to access. In this thesis a learning framework is described that has a very close relationship to boosting. Within this framework a method is described which bears remarkable similarities to boosting stumps and that does not rely on negative examples. This is surprising since learning from positive-only data has traditionally been difficult. An empirical methodology is described and deployed for testing positive-only learning systems using commonly available multiclass datasets to compare these learning systems with each other and with multiclass learning systems. Empirical results show that our positive-only boosting-like method learns, using stumps as a base learner and from positive data only, successfully, and in the process does not pay too heavy a price in accuracy compared to learners that have access to both positive and negative data. We also describe methods of using positive-only learners on multiclass learning tasks and vice versa and empirically demonstrate the superiority of our method of learning in a boosting-like fashion from positive-only data over a traditional multiclass learner converted to learn from positive-only data. Finally we examine some alternative frameworks, such as when additional unlabelled training examples are given. Some theoretical justifications of the results and methods are also provided.

La Leucémie Lymphoïde Chronique (LLC) est caractérisée par l’accumulation dans le sang de lymphocytes B/CD5+ qui sont déficients pour leur programme de mort apoptotique. Malgré les progrès thérapeutiques récents, la LLC est une maladie toujours incurable. La stratégie de rétablissement des processus apoptotiques semble pertinente pour améliorer les traitements des patients, et cette stratégie nécessite l’identification de cibles spécifiques. Dans ce but, nous avons développé une recherche du mécanisme d’action de l’hyperforine, un phloroglucinol purifié à partir de la plante Millepertuis, qui est un inducteur d’apoptose des cellules de LLC ex vivo par la voie mitochondriale dépendante des caspases. Les résultats montrent que le traitement des cellules primaires de LLC régule positivement Noxa, une protéine de la famille Bcl-2 du type « BH3-only » qui est un ligand antagoniste spécifique de Mcl-1, la protéine de survie majeure pour le défaut d’apoptose dans les LLC. Nous montrons que Noxa interagit effectivement avec Mcl-1, ce qui entraine la dissociation du complexe que Mcl-1 forme avec Bak, et l’activation de cette protéine responsable de la libération par la mitochondrie des facteurs d’activation des caspases. La régulation positive de Noxa ne résulte apparemment pas d’une activation transcriptionnelle mais plutôt d’une régulation au niveau protéique comme le suggère la capacité de l’hyperforine à inhiber l’activité protéolytique du protéasome dans les cellules de LLC. Par ailleurs, l’apoptose induite par l’hyperforine est réduite en partie par l’extinction de Noxa avec des ARN interférents. Nos résultats indiquent donc que la régulation positive de Noxa est un des mécanismes qui permet à l’hyperforine de déclencher l’apoptose des cellules de LLC. Ce mécanisme semble être commun à d’autres composés d’origine végétale, comme nous l’avons observé avec l’extrait M2Yn dans la seconde partie de notre travail qui a permis de mettre en évidence que cet extrait de plantes d’Asie centrale est capable lui aussi d’induire l’apoptose des cellules de LLC par la voie mitochondriale dépendante des caspases. Enfin, nous avons contribué à la mise en évidence des propriétés pro-apoptotiques de l’hyperforine dans un autre modèle de leucémie, la leucémie myéloïde aiguë, grâce en partie à la régulation positive de Noxa ainsi qu’à l’inhibition de l’activité kinasique de Akt-1.En conclusion, nos travaux de thèse décrivent des nouvelles étapes dans le mécanisme d’action de l’hyperforine et permettent d’établir que la protéine « BH3-only » Noxa est une cible majeure pour la stratégie thérapeutique du rétablissement d’apoptose dans les LLC, ce qui devrait susciter le développement de nouveaux agents capables d’imiter spécifiquement l’action de Noxa
Chronic lymphocytic leukemia (CLL) is characterized by blood accumulation of CD5+/B lymphocytes which are deficient in their apoptotic program. Despite recent therapeutic advances, CLL remains an incurable disease. Treatment strategy based on the re-establishment of apoptotic processes has retained much attention, and such a strategy requires the identification of specific targets. To this aim, we have investigated the mechanism of action of hyperforin, a phloroglucinol purified from St John’s wort, capable of inducing apoptosis of CLL cells ex vivo via the caspase-dependent mitochondrial pathway. Our results show that treatment of CLL patients’ cells upregulates the BH3-only protein Noxa that is a specific antagonist ligand of Mcl-1, the crucial prosurvival protein for apoptosis deficiency in CLL. We find that hyperforin provokes both the interaction of Noxa with Mcl-1 and the dissociation of Mcl-1/Bak complex, leading to Bak activation responsible for the release of apoptogenic factors from mitochondria. Noxa upregulation does not seem to result from transcription activation but rather from a regulation at the protein level, as suggested by the ability of hyperforin to inhibit proteasome activity in CLL cells. Using siRNA, Noxa silencing partially reduces hyperforin-induced apoptosis. Our data indicate that Noxa upregulation is one of the mechanisms through which hyperforin triggers CLL cell apoptosis. This mechanism appears to be shared with other plant-derived compounds, as observed with M2Yn, an extract from central Asia plants, that we show here to be an inducer of apoptosis in CLL cells by activating the caspase-dependent mitochondrial pathway. Finally, we have participated in highlighting the proapoptotic properties of hyperforin in another leukemia model, acute myeloid leukemia, via in part Noxa upregulation and Akt1 kinase activity inhibition.In conclusion, our data describe new steps in the mechanism of action of hyperforin and favor that the BH3-only protein Noxa is a major target for the therapeutic strategy based on apoptosis induction in CLL. Thus, this thesis should prompt the development of new BH3 mimetics specific for Noxa

O Vírus da Imunodeficiência Humana (HIV) é o agente etiológico da Síndrome da Imunodeficiência Adquirida (AIDS). A AIDS é uma doença de distribuição mundial, e estima-se que existam atualmente pelo menos 36,9 milhões de pessoas infectadas com o vírus. Durante o seu ciclo replicativo, o HIV promove diversas alterações na fisiologia da célula hospedeira a fim de promover sua sobrevivência e potencializar a replicação. A rápida progressão da infecção pelo HIV-1 em humanos e em modelos animais está intimamente ligada à função da proteína acessória Nef. Dentre as diversas ações de Nef está a regulação negativa de proteínas importantes na resposta imunológica, como o receptor CD4. Sabe-se que esta ação resulta da indução da degradação de CD4 em lisossomos, mas os mecanismos moleculares envolvidos ainda são totalmente elucidados. Nef forma um complexo tripartite com a cauda citosólica de CD4 e a proteína adaptadora 2 (AP-2), em vesículas revestidas por clatrina nascentes, induzindo a internalização e degradação lisossomal de CD4. Pesquisas anteriores demonstraram que o direcionamento de CD4 aos lisossomos por Nef envolve a entrada do receptor na via dos corpos multivesiculares (MVBs), por um mecanismo atípico, pois, embora não necessite da ubiquitinação de carga, depende da ação de proteínas que compõem os ESCRTs (Endosomal Sorting Complexes Required for Transport) e da ação de Alix, uma proteína acessória da maquinaria ESCRT. Já foi reportado que Nef interage com subunidades dos complexos AP-1, AP-2, AP-3 e Nef não parece interagir com subunidades de AP-4 e AP-5. Entretanto, o papel da interação de Nef com AP-1 e AP-3 na regulação negativa de CD4 ainda não está totalmente elucidado. Ademais, AP-1, AP-2 e AP-3 são potencialmente heterogêneos devido à existência de isoformas múltiplas das subunidades codificadas por diferentes genes. Todavia, existem poucos estudos para demonstrar se as diferentes combinações de isoformas dos APs são formadas e se possuem propriedades funcionais distintas. O presente trabalho procurou identificar e caracterizar fatores celulares envolvidos na regulação do tráfego intracelular de proteínas no processo de regulação negativa de CD4 induzido por Nef. Mais especificamente, este estudo buscou caracterizar a participação do complexo AP-1 na modulação negativa de CD4 por Nef de HIV-1, através do estudo funcional das duas isoformas de ?-adaptina, subunidades de AP-1. Utilizando a técnica de Pull-down demonstramos que Nef é capaz de interagir com ?2. Além disso, nossos dados de Imunoblot indicaram que a proteína ?2-adaptina, e não ?1-adaptina, é necessária no processo de degradação lisossomal de CD4 por Nef e que esta participação é conservada para degradação de CD4 por Nef de diferentes cepas virais. Ademais, por citometria de fluxo, o silenciamento de ?2, e não de ?1, compromete a diminuição dos níveis de CD4 por Nef da membrana plasmática. A análise por imunofluorêsncia indireta também revelou que a diminuição dos níveis de ?2 impede a redistribuição de CD4 por Nef para regiões perinucleares, acarretando no acúmulo de CD4, retirados por Nef da membrana plasmática, em endossomos primários. A depleção de ?1A, outra subunidade de AP-1, acarretou na diminuição dos níveis celulares de ?2 e ?1, bem como, no comprometimento da eficiente degradação de CD4 por Nef. Além disso, foi possível observar que, ao perturbar a maquinaria ESCRT via super-expressão de HRS (uma subunidade do complexo ESCRT-0), ocorreu um acumulo de ?2 em endossomos dilatados contendo HRS-GFP, nos quais também detectou-se CD4 que foi internalizado por Nef. Em conjunto, os resultados indicam que ?2-adaptina é uma importante molécula para o direcionamento de CD4 por Nef para a via ESCRT/MVB, mostrando ser uma proteína relevante no sistema endo-lisossomal. Ademais, os resultados indicaram que as isoformas ?-adaptinas não só possuem funções distintas, mas também parecem compor complexos AP-1 com diferentes funções celulares, já que apenas a variante AP-1 contendo ?2, mas não ?1, participa da regulação negativa de CD4 por Nef. Estes estudos contribuem para o melhor entendimento dos mecanismos moleculares envolvidos na atividade de Nef, que poderão também ajudar na melhor compreensão da patogênese do HIV e da síndrome relacionada. Em adição, este trabalho contribui para o entendimento de processos fundamentais da regulação do tráfego de proteínas transmembrana no sistema endo-lisossomal.
The Human Immunodeficiency Virus (HIV) is the etiologic agent of Acquired Immunodeficiency Syndrome (AIDS). AIDS is a disease which has a global distribution, and it is estimated that there are currently at least 36.9 million people infected with the virus. During the replication cycle, HIV promotes several changes in the physiology of the host cell to promote their survival and enhance replication. The fast progression of HIV-1 in humans and animal models is closely linked to the function of an accessory protein Nef. Among several actions of Nef, one is the most important is the down-regulation of proteins from the immune response, such as the CD4 receptor. It is known that this action causes CD4 degradation in lysosome, but the molecular mechanisms are still incompletely understood. Nef forms a tripartite complex with the cytosolic tail of the CD4 and adapter protein 2 (AP-2) in clathrin-coated vesicles, inducing CD4 internalization and lysosome degradation. Previous research has demonstrated that CD4 target to lysosomes by Nef involves targeting of this receptor to multivesicular bodies (MVBs) pathway by an atypical mechanism because, although not need charging ubiquitination, depends on the proteins from ESCRTs (Endosomal Sorting Complexes Required for Transport) machinery and the action of Alix, an accessory protein ESCRT machinery. It has been reported that Nef interacts with subunits of AP- 1, AP-2, AP-3 complexes and Nef does not appear to interact with AP-4 and AP-5 subunits. However, the role of Nef interaction with AP-1 or AP-3 in CD4 down-regulation is poorly understood. Furthermore, AP-1, AP-2 and AP-3 are potentially heterogeneous due to the existence of multiple subunits isoforms encoded by different genes. However, there are few studies to demonstrate if the different combinations of APs isoforms are form and if they have distinct functional properties. This study aim to identify and characterize cellular factors involved on CD4 down-modulation induced by Nef from HIV-1. More specifically, this study aimed to characterize the involvement of AP-1 complex in the down-regulation of CD4 by Nef HIV-1 through the functional study of the two isoforms of ?-adaptins, AP-1 subunits. By pull-down technique, we showed that Nef is able to interact with ?2. In addition, our data from immunoblots indicated that ?2- adaptin, not ?1-adaptin, is required in Nef-mediated targeting of CD4 to lysosomes and the ?2 participation in this process is conserved by Nef from different viral strains. Furthermore, by flow cytometry assay, ?2 depletion, but not ?1 depletion, compromises the reduction of surface CD4 levels induced by Nef. Immunofluorescence microscopy analysis also revealed that ?2 depletion impairs the redistribution of CD4 by Nef to juxtanuclear region, resulting in CD4 accumulation in primary endosomes. Knockdown of ?1A, another subunit of AP-1, resulted in decreased cellular levels of ?1 and ?2 and, compromising the efficient CD4 degradation by Nef. Moreover, upon artificially stabilizing ESCRT-I in early endosomes, via overexpression of HRS, internalized CD4 accumulates in enlarged HRS-GFP positive endosomes, where co-localize with ?2. Together, the results indicate that ?2-adaptin is a molecule that is essential for CD4 targeting by Nef to ESCRT/MVB pathway, being an important protein in the endo-lysosomal system. Furthermore, the results indicate that ?-adaptins isoforms not only have different functions, but also seem to compose AP-1 complex with distinct cell functions, and only the AP-1 variant comprising ?2, but not ?1, acts in the CD4 down-regulation induced by Nef. These studies contribute to a better understanding on the molecular mechanisms involved in Nef activities, which may also help to improve the understanding of the HIV pathogenesis and the related syndrome. In addition, this work contributes with the understanding of primordial process regulation on intracellular trafficking of transmembrane proteins.

A comercialização de produtos florestais não madeireiros (PFNMs) popularizou-se como atividade promotora do desenvolvimento socioeconômico de comunidades florestais com baixo impacto ambiental. Há evidências, contudo, de que a exploração possa produzir efeitos ecológicos e socioeconômicos negativos, sugerindo que é necessário monitorar tais iniciativas. A comercialização frequentemente ocorre em áreas remotas e em contextos de pobreza, como é o caso de muitas daquelas da Amazônia brasileira. Desta forma, é necessário que o monitoramento restrinja-se a avaliar os efeitos evidenciados como mais comuns em estudos anteriores. Este estudo revisa e sintetiza as evidências científicas dos efeitos da exploração de PFNMs sobre parâmetros ecológicos e socioeconômicos e, a partir daí, indica aqueles mais relevantes ao monitoramento. O estudo também levanta até que ponto o monitoramento é implementado no contexto da Amazônia brasileira e avalia quais os parâmetros importantes e viáveis de monitoramento neste caso específico. Para isso, foram realizadas revisões sistemáticas da literatura e a consulta a profissionais da área através do método Delphi. Os resultados indicam que efeitos ecológicos negativos são frequentes, principalmente quando se coletam folhas ou cascas. Alterações em órgãos ou processos fisiológicos e a taxa de sobrevivência dos espécimes explorados são parâmetros que devem ser monitorados, em particular quando se coletam frutos e partes vegetativas. Para todos os tipos de PFNMs, o tamanho e a estrutura populacional são parâmetros prioritários ao monitoramento. A riqueza de espécies da comunidade explorada merece atenção, principalmente quando se coletam frutos. Para os aspectos socioeconômicos, efeitos positivos foram mais frequentes que negativos. A contribuição da renda monetária obtida com o comércio na renda total, a regularidade de ingresso desta renda e o papel dos PFNMs como recursos de salvaguarda são parâmetros do capital financeiro prioritários ao monitoramento. Para o capital social, o empoderamento feminino, a coesão de grupo e o acesso aos benefícios gerados pela comercialização devem ser monitorados. Na Amazônia brasileira são poucas as iniciativas de implementação do monitoramento da comercialização de PFNMs. Embora este seja considerado importante, existem dificuldades que derivam principalmente da falta de apoio institucional, políticas de incentivo e de recursos financeiros. Os profissionais participantes do Delphi consideram que os parâmetros ecológicos mais importantes a monitorar neste contexto são o tamanho e a estrutura populacional do recurso explorado, o aumento da taxa de mortalidade, a quantidade total de recurso extraída e a técnica de coleta utilizada. Para os parâmetros econômicos, aspectos do mercado, como o preço pago ao coletor, a demanda e a qualidade do produto, bem como a renda monetária obtida pelos indivíduos são os parâmetros considerados mais importantes. Por fim, para os aspectos sociais, os efeitos na cultura, na qualidade de vida e na organização interna da comunidade foram priorizados. Os profissionais indicam que é viável estabelecer o monitoramento dos parâmetros levantados.

The Adaboost learning algorithm can train an effective and reliable classifier for object detection. Traditional approaches used both positive and negative samples of target object on training. Such approaches are in fact to divide world objects in to two categories, i.e., target one and non-target one. Hence, a face-detection learning algorithm, for example, needs to gather discriminant features both for face and non-face objects respectively from given positive/negative samples for ‘classification’. Such an approach is, in fact, to do objection detection in classification sense. Detection theory or signal detection theory is a means to measure the ability to differentiate between information-bearing patterns (e.g., facial images) and random patterns that distract from the information (e.g., almost any random images or sub-images are non-facial images). This implies that samples for information-bearing patterns (positive samples) are scarce resources in comparison with the opposite ones (negative samples). One may say that almost ‘nothing is not noise’ in discourse of detection theory. Hence, to learn detection, negative samples seemingly plays an irrelevant role for feature extraction on the target object. For example, teaching children to know the appearance of giraffes, we only show them giraffe pictures, and not include any non-giraffe ones. Such a learning scheme will not cause any difficulties for children to learn the concept of giraffes. This motivates us to develop a learning scheme for object detection based mainly on positive samples of the target object. This thesis proposes the so called PSO-Adaboost learning algorithm. It is a abbreviation of Positive-Sample-Only Adaboost learning algorithm. The concept will be described by training a face detector. During training, the weak classifiers of the face detector are determined iteratively based on the feature entropy. It is computed by the probability distribution of the so-called NBBP (Neighboring-Block Binary Pattern) features among the given positive samples. Some experimental results and analytical information will be provided in the thesis. It is shown that PSO-Adaboost is more learning effective than traditional approaches, and the resulting ROC curve is also comparable with them. Furthermore, PSO-Adaboost is allowed to associate with a refining scheme for improvement. It also facilitates to develop an incremental Adaboost learning algoirithm as as to build an object detector incrementally.

Most previous research have indicated that life domain satisfaction can promote global well-being. However, the main purpose of this study is to examine the effect of balanced life satisfaction on global well-being and to prove the mechanism of balanced life. In study 1, results indicated that the standard deviation of life satisfactions across life domains had significant influence on the Global Well-being. Multiple regression analysis showed that this remained significant when the average of satisfaction was controlled for. In study 2, the author measured “need fulfillment” in multiple life domains for capturing the satisfaction in multiple life domains. Then we created three imbalanced indicators. Simple correlation showed higher imbalanced scores were associated with less Global Well-being. Multiple regression demonstrated that the ceiling effect and the scarcity hypothesis are the mechanism of balanced life. Finally, further discussion and recommendation were proposed.

Chile's FONDEF and FONTEC R&D grant programs both boost the productivity of direct beneficiaries, increasing total factor productivity (TFP) by around 4.2 percent. However, spillover effects are contingent on program design. Only FONDEF funded projects (requiring collaboration between firms and research centers) generate positive spillovers. FONTEC projects, which fund R&D within the firm, do not. Spillover effects are nonlinear according to the share of firms within a sectorregion receiving subsidies. Positive knowledge spillovers dominate when the share of treated firms is small. However, if the program supports a large share of a firms rivals, spillovers decline as a result of a business-stealing effect.

While many people get HIV-related counseling and testing, only those who receive pre- and post-test counseling, and test voluntarily, are participating in voluntary counseling and testing (VCT). The high rates of HIV infection among youth make it crucial to find programs to prevent infection. Because there is evidence that many adults benefit from VCT, there is increasing interest in extending these services to young people. VCT counseling helps adolescents evaluate their own behavior and its consequences. A negative test result offers the opportunity to recognize vulnerabilities and develop risk-reduction plans to adopt safe behaviors. Young people who test HIV-positive can receive referrals for care and have opportunities to discuss and understand what their HIV status means and what responsibilities they have to themselves and others as a result. Young women who are pregnant and test HIV-positive should be offered special care to safeguard their own health and minimize the risk of passing the virus to the baby. This report assesses the available evidence about the current status of VCT and youth in developing countries.

Expanding small-scale interventions without lowering quality and attenuating impact is a critical policy challenge. Community monitoring overs a low-cost quality assurance mechanism by making service providers account-able to local citizens, rather than distant administrators. This paper provides experimental evidence from a home visit parenting program implemented at scale by the Nicaraguan government, with two types of monitoring: (a) institutional monitoring; and (b) community monitoring. We find d a positive intent-to-treat effect on child development, but only among groups randomly assigned to community monitoring. Our findings show promise for the use of community monitoring to ensure quality in large-scale government-run social programs.

The empirical accounts of the costs and benefits of quasi-parity codetermined supervisory boards, a very special German institution, have long been inconclusive. A valid economic analysis of a particular legal regulation must take the legal specificities seriously, otherwise it will be easily lost in economic fictions of functional equivalence. At its core the corporate actor “supervisory board” has no a priori objective function to be maximised – the corner stone of the theory of the firm – but its objective function will only be brought about a posteriori – should negotiations result in an agreement (E. Fraenkel). With this understanding,the paper presents six recent quasi-experimental studies on the economic (dis) advantageousness of the German codetermination laws that try to follow the rules of causal inference despite the lack of random variation. By and large they refute the hold-up model of codetermination by showing positive or nonnegative effects even on shareholder wealth – and a far-reaching improvement of the well-being of the core workforce. In conclusion, indications are offered that the shareholder primacy movement has only weakened, but not dissolved the “Deutschland AG”.

Evasion of labor market regulations in middle income countries is systemic. This is generally known as informality. In Latin America, where less than 50% of workers are registered with social security, this is a permanent phenomenon and encompasses a variety of economic realities ranging from subsistence self-employment to evasion of certain regulations including social security contributions. In this study we analyze the role of enforcement in curbing informality in large formal firms in Peru, where informality levels are around 70%. Through the Peruvian National Labor Control Superintendence (SUNAFIL) we randomly sent 697 letters to formal Peruvian firms of more than 50 workers, indicating their obligation to enroll workers in social insurance systems (health and pensions). Two types of letters were sent, one with a deterrence message and one emphasizing the benefits of formalization. One year after the letters were sent, we found a positive and statistically significant effect on the number of workers enrolled in social security (9.8% on average). Only strict deterrence messages had a significant impact, and only in very large firms. This evidence suggests that there is room for improvement in compliance with labor regulations through more proactive monitoring and behavioral tools such as reminders, but effects could be concentrated in the largest firms.

Violent extremism has a trauma problem. Psychological trauma plays a role in the genesis, perpetration, and resolution of violent extremism. Despite evidence attesting to the positive effects of trauma-informed rehabilitation programs and trauma recovery support for armed combatants and criminal offenders, there has been limited donor interest or coordinated policy effort to meaningfully integrate trauma recovery into the design of rehabilitation and reintegration of violent extremists specifically. Research in global mental health confirms trauma is not only relevant to the emergence of violent extremism but is also a consequence of participation in violent extremism. While there is a general dearth of data on the psychobiological markers of trauma among extremist populations specifically, the limited data we have from child recruits, as well as from other conflict-affected populations calls for a more prominent role of psychological rehabilitation and trauma recovery in the reintegration of violent extremists.
