Dos, Santos Brito Silva Nayane. "Development and implementation of new reference panels for HLA imputation from different populations to accelerate immunogenetic association studies". Electronic Thesis or Diss., Nantes Université, 2024. http://www.theses.fr/2024NANU1034.
Testo completoThe Major Histocompatibility Complex (MHC) is crucial in immune response. The human leukocyte antigen (HLA) genes, within the MHC, encode key molecules that promote immune system activation by directly acting on self/non-self antigen presentation. HLA polymorphisms are linked to many diseases and influence transplant outcomes. To optimize the analysis of these genes and improve SNP and haplotype detection, the hla- mapper methodology was developed in Brazil. This study used this tool to correct genetic alignments and perform a more precise sequencing analysis. As a result, it was possible to identify HLA alleles and SNPs in different worldwide populations, including over 1,000 samples from a Brazilian population, which is absent from databases. The generated data were used to create reference panels within the SNP-HLA Reference Consortium (SHLARC) framework. Although imputation methods are not as accurate as direct sequencing, they offer an efficient and practical way to infer HLA alleles from SNPs in genome-wide association studies. In admixed populations, such as the Brazilian population, models including local samples performed better in predicting HLA alleles, highlighting the importance of multiethnic references for improving imputation

Testo completoThe meeting of three continents : Africa, America and Europe have led to three centuries of human trafficking between Africa and the New World. The Gulf of Benin on the west coast Africa chosen as part of this study was one of supplying men to work in plantations and mines of Brazil, Cuba and the United States. This trade has ravaged the continent while promoting the development of Europe and America. However, the return of former slaves freed in Brazil, Cuba is one of the real contributions of the trade. Aguda called Amaro, Saro Krio or in the case of Sierra Leone or Liberia, Afro-Brazilians were a social group that has spread to coastal cities, giving a new impetus to the development of concerned countries: Nigeria, the former Dahomey Benin and Togo. They have become the driving force behind the economy, embarking on a rotating basis in the slave trade and oil palm. They have initiated, developed and enriched culture Afro-Brazilian still existing today through religion, architecture and material culture. Educated, speaking European languages, they took part in the protest movement against the colonial system, denouncing newspaper through its abuse and the misey of the African people. Their political struggles have helped to awaken the political consciousness of the masses. Despite the almost total disappearance of this community, its legacy remains visible and indisputable éveille sights. Work of preserving this heritage has begun in recent years by African and American exchange through meeting and seminars
