Testo completoP-glycoprotein Pgp is a transmembrane transporter protein that protects cells from potentially toxic exogenous compounds. This protein acts as a molecular pump and is overexpressed in cancer cells. So via that mechanism, tumor cells exhibit intrinsic or acquired cross-resistance to diverse chemotherapeutic agents, resulting in the failure of chemotherapy for some cancers. The modulation of the efflux activity of that protein can be one of the mean that could increase anticancer treatments efficiency. Pgp has different substrates sites, two of which are spatially close : the steroid site and the ATP site, the latter is in charge of ATP hydrolysis which give the required energy for the Pgp efflux activity. Our goal was to synthesize bivalent modulators steroids/linker/nucleosides. First we have obtained purine bases analogues and particularly didéazapurines such as 5- and 6-azaindoles and the corresponding trioxopyrrolopyridines, then we have carried out the glycosylation of those compounds. Then, steroids modulators have been synthesized : some of them have shown a good modulation activity toward Pgp. Finally, polyethylene glycols chains with different length have been grafted on adenine so that they could be attached to the steroids via an amide function

Testo completoThe overall objective was to assess the genetic diversity of a collection of native potato varieties originating from the island of Chiloe, to characterize the resistance of these varieties to late blight (Phytophthora infestans) and to inquire about the status of in-situ conservation. The following specific topics was developed: a) conservation in-situ of native Solanum within the great isle of Chiloe and its impact on diversity, b) evaluation of the morphological diversity of the native potatoes that we have collected c) assessment of genetic diversity of the collection by the mean of microsatellite and AFLP markers, d) characterization of field and in-vitro resistances to late blight. Based on the results of the surveys, we can conclude that in-situ maintenance of native potato diversity i is not well preserved, due to strong social and economic changes on the island of Chiloe. There is a clear process of replacing native varieties for commercial cultivars in response to market conditions. Native varieties are present in 80.5% of the farms of Chiloe, but the diversity of varieties has been significantly reduced. The results morphological diversity showed the formation of groups under the popular name and local attributes assigned by the farmers themselves. Molecular evaluation of the collection (SSRs and AFLP) reveals a high degree of genetic diversity. Both markers were consistent in classifying Solanum fernandezianum as the more distant genotype compared to all the others. The SSRs allowed the estimation of the polymorphic information content for seven loci which values ranged between 0.63 to 0.89. Both types of markers, did not provide the same groupings among the accessions. The SSR-based study showed a low differentiation between native potatoes and improved cultivated varieties. The analysis of diversity based on the AFLP was not inconsistent with this result because despite containing only one cultivar (Desiree), the cultivar was grouped with the native varieties. In relation to resistance to late blight, most accessions of native potato fall into the ranks moderately resistant to moderately susceptible

The numerical simulations were performed to identify the range and location of the maximal stress and strain within the pump stator. The experimental tests allowed to characterize the mechanical behaviour of rubber under cyclic solicitation at several temperatures. Testo completoInfant infections are a major concern in terms of global morbidity and mortality, significantly contributing to infant deaths, particularly through respiratory infections. Risk factors for severe infections in infants are multifactorial, primarily related to in utero development, prematurity, low birth weight, as well as immunological, genetic, and environmental factors. Recent data in the literature suggest that modifications in the microbiome can have immunological implications and potentially increase the risk of infection. Therefore, changes in the maternal-fetal microbiome could have consequences for the infant. Recent literature suggests associations between in utero exposure to antibiotics and an increased risk of serious infections in early childhood. These studies have been made possible, in particular, through the development of pharmacoepidemiology using medical-administrative databases, such as the National Health Data System (SNDS), allowing the creation of substantial cohorts of pregnant women. The overall objective of this thesis was to evaluate the role of medication exposures during pregnancy and the risk of serious infections in the first year of life. The research work focused on two pharmacological classes due to their frequent use during pregnancy and their impact on the maternal-fetal microbiome: antibiotics and proton pump inhibitors. Firstly, a national cohort study, including 2.8 million full-term infants, evaluated the association between in utero exposure to systemic antibiotics and the occurrence of serious infections in full-term infants during their first year of life. The results revealed a moderate increase in the incidence of serious infections in infants exposed in utero to systemic antibiotics. Associations were similar regardless of the trimester of exposure, antibiotic class, and infection sites. However, infants exposed to broad-spectrum antibiotics or three or more antibiotic courses appeared to have a slightly increased risk, supporting a potential causal relationship. Secondly, another study, stratified by the use of proton pump inhibitors during the first three months of life and including 2.1 million full-term infants, explored the impact of prenatal exposure to proton pump inhibitors on the occurrence of serious infections during their first year of life. The results ruled out a significant association between the use of proton pump inhibitors during pregnancy and the occurrence of serious infections in infants. Nevertheless, even after adjusting for several confounding factors, this study did not exclude a limited residual risk, restricted only to infants with proton pump inhibitors use in early life. In conclusion, this thesis confirmed existing data on an association between exposure to antibiotics during pregnancy, which strongly affects the microbiome, and the occurrence of serious infections in infants. The results are more reassuring concerning proton pump inhibitors; although a low risk cannot be completely ruled out. These findings need confirmation through further studies. They have contributed to expanding the knowledge regarding the safety of medication use during pregnancy

Testo completoThe urbanisation process in China brings a high pressure on the environment. The highest potential to reduce these impacts corresponds to decisions made during the building’s design phase, which can be supported by numerical simulation. This thesis is dedicated to the study of three boundary conditions related to the energy and environmental performance evaluation of buildings in China: - The ground: a ground coupled heat pump model is proposed integrating a fast calculation ground heat exchanger model for a large-scale boreholes field. This model can be used to improve the energy performance of the system in the design and operation phases. - The microclimate: a site-specific weather file generation method which can provide local hourly air temperature is proposed, accounting for the urban heat island effect. The effects of the microclimate on the building’s energy performance are quantitatively investigated. - The background system for life cycle assessment: the effects of the spatial and temporal variation of the electricity production mix in China on the environmental impacts are investigated. The environmental database is adapted to the Chinese national and local context. The results show that the environmental impacts of buildings could be more reasonably evaluated by considering these three boundary conditions
