Abstract (sommario):
Many psychoanalytic writers have viewed acting out and somatization as alternate routes for the discharge of uncontained aggressive impulses in borderline patients. In order to investigate this empirically, we tracked the occurrence of two related behaviors—acting up and physical illness—over the course of one patient's long-term hospitalization. Daily scores on these variables were analyzed using a time series procedure. Results indicated that as treatment progressed, acting up and physical illness became more synchronized, and this occurred just prior to symptom remission. Material from psychotherapy suggested that these behavioral changes were associated with the patient's increased ability to verbalize aggressive impulses in the context of fantasy and memory.

Abstract (sommario):
Purpose: The purpose of this study was to test a conceptual model that specified relationships among perceived organizational support (POS), emotional labor, job satisfaction, and affective commitment. Methods: The participants included 297 physical educators who completed an online survey. The data were analyzed using confirmatory factor analysis and structural equation modeling. Results: After verifying the factor structure, the results of structural equation modeling supported the conceptual model, χ2(157) = 225.09, p < .001; χ2/df = 1.43; root mean square error of approximation = .055 (90% confidence interval [0.045, 0.064], p < .001); standardized root mean square residual = .042; nonnormed fit index = .984; comparative fit index = .987. POS related negatively to surface acting and positively to deep acting and genuine expression. Affective commitment related positively to POS, deep acting, and genuine expression, and negatively to surface acting. Job satisfaction related positively to POS and negatively to surface acting. Discussion/Conclusion: The findings are discussed within the framework of affective events theory, and recommendations are made for helping in-service and preservice physical educators to develop emotional labor strategies.

Abstract (sommario):
Little evidence links emotional labor to either psychological or physical health. This study determined whether the two types of emotional labor (i.e., surface vs. deep acting) were significantly associated with depressive symptoms and work-related musculoskeletal disorders in call center workers. A cross-sectional study was conducted with 274 workers recruited from a call center in Seoul, South Korea. In adjusted regression models, levels of surface, but not deep, acting were significantly and positively associated with depressive symptoms. Higher surface acting levels were significantly and positively associated with low back pain; higher deep acting levels were significantly and inversely associated with low back pain. Study findings could inform occupational health nurses as they delineate differentiated strategies, according to the nature of surface and deep acting, to promote psychological and physical health in call center workers.

Abstract (sommario):
The paper presents a physical model of a single-acting hydroimpact device. Its design can be used to create a hydraulic punch for drilling wells in soft soils. A stand with a clamping device has been developed to simulate the friction force acting on the punch housing when it moves in the soil. Methods of recording dynamic characteristics of working cycles of the hydraulic device are worked out. Tests were carried out with changes in the force acting on the housing from the clamping device. Oscillograms of the characteristics of working cycles (displacements of the housing and striker, pressures in the working chambers and the accumulator) are given.

Abstract (sommario):
The most important clarification we bring in our reply to commentators concerns the problem of the “explanatory gap”: that is, the gulf that separates physical processes in the brain from the experienced quality of sensations. By adding two concepts (bodiliness and grabbiness) that were not stressed in the target article, we strengthen our claim and clarify why we think we have solved the explanatory gap problem, – not by dismissing qualia, but, on the contrary, by explaining why sensations have a “feel” and why “feels” feel the way they do. We additionally clarify our views on: internal representations (we claim internal representations cannot explain why sensation has a feel), on behaviorism (we are not behaviorists), on perception and action (we believe there can be perception without action), and on the brain (we believe the brain does do something important in perception).

This review summarizes the occurrence, properties and role in mucin O-glycosylation pathways of the various members of glycoprotein sulphotransferase families. Although a number of sulphotransferases have been cloned that act on mucin-type substrates in vitro, it is still difficult to determine exactly which enzymes are responsible for mucin sulphation in vivo. Sulphotransferases play a critical role in determining the chemical, physical and biological properties of mucins. Several of these enzymes have been shown to differ in expression and activity in cancer and inflammation.

To the Editor.— Recent discussions in Pediatrics (1985;75:785-786 and 1986;77:783-784) have pointed out the problems associated with intravascular injection of long-acting penicillins. There is an obvious need for continued education of health care providers concerning the physical properties of these agents and their delivery systems. Wyeth, the manufacturer of the Tubex cartridge system, has recently redesigned their product so that the flow of any aspirated blood is directed to one side of the glass barrel, immediately past the hub, where it can be more easily seen.

Testo completo
Abstract (sommario):
The Russian School of Acting was a term encountered amongst theatre-practitioners in 1990s' Moscow. Its roots lay in Stanislavsky's system with influences from Michael Chekhov and Jerzy Grotowski. In defining the phrase, this investigation follows Stanislavsky's psycho-physical technique through three practices: (1) pre-determining a production through a detailed mise-en-scene; (2) analysing the play through round the table discussions and (3) exploring the Method of Physical Actions. Chekhov's `creative individuality' and Grotowski's via negativa are also considered. The contemporary Russian School of Acting is examined through experience of actor-training at the State Institute of Cinematography in Moscow, 1993 - 1994, in terms of: (a) work on oneself, as explored through Vladimir Ananyev's Scenic Movement; (b) work in the ensemble, as explored through Katya Kamotskaya's Actor Training programme and (c) work on the role, as explored by Albert Filozov's rehearsal of playtexts. An attempt is made to apply the elements of this psycho-physical actor-training to two British repertory productions at the Swan Theatre, Worcester - The Seagull (February 1995) and Steaming (March 1996). In conclusion, an assessment is made of the relevance of the process-orientated Russian technique to the essentially result-orientated British repertory environment

Richardson, Andrew. "Acting the Absurd: Physical Theatre for Text/Text for Devising". VCU Scholars Compass, 2015. http://scholarscompass.vcu.edu/etd/3744.

Testo completo
Abstract (sommario):
This paper considers two purposes for actor training—textual interpretation and devising original works—through the teaching of a class based on contemporary theatrical clown and physical theatre exercises which are then applied to Samuel Beckett’s Waiting for Godot. Devised work can be used to interpret a script, and a script can be used as a jumping-off point to devise new works. Beginning with an explanation of the teaching methods for the class, the paper then gives a background of clowns who performed in Beckett’s plays, and analyzes various productions' use of games to enliven text. Exercises from the class are used as examples of exploring the uncovering of clown personas and the application of games to both Beckett scene-work and invented theatre pieces. The students’ final performances are examined to demonstrate the effectiveness of the classwork, confirming that textual interpretation and devising are complementary instead of opposing practices.

Abstract (sommario):
The purpose of this thesis is to showcase the importance of God-inspired Theatre and to manifest the transformative effects of living in accordance to the Word of God. In order to share my vision for theatre such as this, I will examine the biblical elements in Zora Neale Hurston's short story Sweat (1926). I will write a stage adaptation of the story, while placing emphasis on the biblical lessons that can be used for God-inspired Theatre. When viewing the stage adaptation based on Sweat, the audience members will understand how God-inspired Theatre aims to help members of society utilize their gifts and abilities to assist others in achieving spiritual stability. The members of the audience will also be informed of my vision to use this piece to inspire others to embrace cultural awareness and sensitivity. This is my vision--helping others to walk in their God-ordained destiny. With this in mind, I am using Sweat as a proposed play because it is closely related to the creation account as recorded in the Old Testament Book of Genesis. In this play, Adam and Eve are replaced with the characters Sykes and Delia Jones. The creation account is a very influential testimony because it is known throughout humanity. Its popularity is due to the fact that the Old Testament is the commencement of the Christian Bible. For those of the Jewish faith, the collection encompasses the Torah, the first five books of the bible--the law for everyday living-- as well as the history of God's promise to them. For Christians, the Old Testament is just as sacred, but they view its religious meaning as incomplete without the life, teachings, death, and resurrection of Jesus Christ detailed in the New Testament. Also, Muslims trace their religious roots to some of the figures in the Old Testament although they deny the religious significance of the work as a whole. In essence, the Old Testament is crucial to Western Civilization. This is why Sweat is so powerful. It takes an extremely familiar testimony and shares pertinent messages that help people to become productive members of society. In order to show how effective Sweat is in helping others to live spirit-filled lives, I will use creative staging that will place the characters in the personal space of the audience members. I will achieve this by: having the actors enter and exit from the audience; allowing certain scenes to take place within the audience; and having the actors deliver some lines to various audience members. I feel that by making the audience a part of the production, it will cause them to see that they are not any different from the characters in the play. At some point in their lives, theatergoers have encountered--or been intimate with--an Adam, an Eve, a Sykes or a Delia. This will cause them to not see Sweat as just a play, but as a valuable life lesson, triggering self-examination and initiating renovated thinking that helps people to become culturally aware and spiritually sound. It is imperative that the biblical messages in Sweat are conspicuous. Whereas the narrator normally describes Delia's facial expression or feelings, I plan to write in scenes where her thoughts are audible. Some of her thoughts will include moments when she is praising and worshipping God. This is apparent because she starts to emerge as a woman of strength as the story progresses. Her relationship with God is cultivated on a daily basis. This is why she is able to tolerate her husband's foolishness. Her husband, Sykes, does not commune with God. To demonstrate his lack of communion with God, I will stage him being resistant to her times of worship--as he normally is according to the narrator. Clearly, the marriage is unbalanced. One partner is trying to please God, and the other is trying to please self. This is not how God intended marriage to be. In the New Testament Book First Peter, it states in the third chapter and seventh verse "husbands are to dwell with them in understanding, giving honor to the wife … being heirs together of the grace of life." Showing the burdensome consequences of destructing God's original design will pull on the hearts of audience members because they have encountered or known someone who is presently dealing with the consequences of this disobedient act. I will further reiterate the need for living a spirit-filled life by using costumes, scenic devices, and lighting to convey the godly and ungodly character traits that are embodied within the story. Through the use of colors and patterns, I will project the internal state of the character as in relation to God's instructions. I will work with a lighting designer in order to help convey the moods of the various scenes. The lighting techniques we choose will help to establish the thoughts and personalities of the characters. These feelings will transcend the minds of the audience and cause them to take the biblical messages into very deep consideration. The actors are the final ingredients in making Sweat an awe-inspiring, informative piece. Words are what they are, what one perceives them to be, while on paper. It is the job of the actor to give life to these words, cause them to live in the atmosphere, and to make the character come alive. Until the actor embodies the very heart of a character, the message in God-inspired Theatre will not be able to come forth and propel audience members to have a spiritual awakening. This is why people cannot just read Sweat. They must see the trials and journeys in order to receive life-changing revelations from the testimonies within the play.

Ferreira, de Mendonça Guilherme Abel. "Acting theory as poetic of drama : a study of the emergence of the concept of 'motivated action' in playwriting theory". Thesis, Brunel University, 2012. http://bura.brunel.ac.uk/handle/2438/7331.

Testo completo
Abstract (sommario):
Playwriting theory has, from its beginning, been concerned with the search for the essential nature of dramatic writing. Early playwriting treatises (poetics) defined the essential aspects of drama as being the plot (creation of sequences of fictional events), the moral character of its heroes, the idea of enactment, or the rhetorical and lyrical qualities of the text. These categories were kept through later treatises with different emphasis being put on each category. An understanding of drama as a sequence of fictional events (plot) has been central in acting theory. Modern theories and techniques centred on Stanislavsky’s ideas rely heavily on rehearsal methods that carefully establish the sequence of actions of the characters in a play as a result of psychological motivations. This method was described by Stanislavsky in An Actor’s Work on a Role, published in 1938, and is known as the Method of Physical Actions. This thesis reassesses the definition of playwriting as consisting essentially in the creation of a plot populated by suitable characters. Rather than discussing playwriting theory in isolation it attempts a bridge between acting theory and playwriting theory by using the Method of Physical Actions as an equivalent to plot. Acting theory is thus considered as a theoretical justification for the centrality of plot. The method used is hermeneutic — a systematic interpretation of poetics, unveiling in almost an archaeological manner the relevance of the essential definitions of drama, such as character, source, genre, and language to the concept of plot. The chronological path of development of dramatic theories is shown to be gradual: from the strict obedience to the narrative line imposed by the mythic sources, in classical treatises; through to an interest in the lyrical expression of the predicament of specific characters, in neoclassical theory; to an awareness of specific social types in the eighteenth century; and, finally, to the conception of the plot as a product of the mental life of individual characters in modern theory.

Madureira, Ana Mafalda. "(Re)acting the city. Physical planning practices and challenges in urban development projects of the Entrepreneurial City". Doctoral thesis, Blekinge Tekniska Högskola, Institutionen för fysisk planering, 2014. http://urn.kb.se/resolve?urn=urn:nbn:se:bth-00583.

Testo completo
Abstract (sommario):
The aim of this dissertation is to traceand discuss the practices and challenges of physical planning within an Entrepreneurial City approach to urban policy. The research aim is addressed by focusing on three questions: 1) how have the practices of physicalplanning been influenced by the context of an Entrepreneurial City approach to urban policy 2) how has physical planning responded to this urban policy context, and 3) which potential dilemmas for physical planning practice derive from this new context? By an Entrepreneurial City approach tourban policy I understand an approach whereby there is an attention placed over strategies to promote local economic growth and attract investments, companies and specific types of people in to the city. Arguably urban policies focus less on welfare-related and redistribution strategies. There is an adoption of private sector discourses and tools to promote the city as a place to live, work and invest in. These discourses and tools pass through place-making strategies, marketing, engagement in speculative, risk-taking market-led projects, and seeking partners with whom to establish alliances that will serve to promote the city. The strong emphasis of Entrepreneurial City approaches on interventions over the built environment of a city or neighborhood implies a greater attention to what is happening to the practices of physical planning in municipalities that have adopted this approach. Existing studies tend to emphasize that it signifies a decrease in the scope of influence for public sector, and by extention for physical planning, in the governance and steering of these projects. The dissertation focuses on large-scale urban development projects – Brunnshög, in Lund, and Bo01, Norra Sorgenfri and Hyllie, in Malmö. The projects were chosen due to their likelihood toillustrate physical planning practices marked by an Entrepreneurial City approach. The main findings of this thesis refute the idea of a turn in urban policy towards entrepreneurial city approaches, and illustrate instead a process by which new practices and values coincide with previously established settings and practices. Physical planning is adopting the discourses of an urban policy approach where intercity competition for new industries (preferably in knowledge-intensive sectors) and residents(preferably the “creative classes”) guide urban development projects. The governance setting is marked by the need to establish working networks and partnerships that will create the capacity to act. Experimentation, piece-meal approaches and inter-project learning mark the adaptation strategies to an urban policy context that is still changing. Potential dilemmas lie in the fragmented character of the partnerships required to execute the projects, and in the assumption that these projects will result in the rehabilitation of the socio-economic trends of the city and promote local economic growth. Additionally the resulting built environments are prone to processes of gentrification and displacement, and spatial and socioeconomic polarization.

Testo completo
Abstract (sommario):
Bodydialogue is a coherent and simple system of exercises, rehearsal techniques, principles and aesthetic values which in application enhance the actor's ability to physicalise dramatic action and behaviour. It can be applied directly within a rehearsal process to heighten the physical life of a play or performance event, or it can be taught separately as a system for providing student actors with concrete skills in movement, stagecraft and physical characterisation. Unlike many other movement systems taught in drama schools, such as Mime, Dance, Acrobatics or Alexander, which are grounded in their own discipline base, Bodydialogue is grounded in Stanislavsky's Acting through the Method of Physical Action, and as such is centered in the discipline of text-based Acting. It is thus first and foremost an approach to Acting via Physical Action and Physical Behaviour, rather than a study of Movement, or a movement genre. This thesis describes the development and application of Bodydialogue physicalisation techniques to a workshop production of miss julie downunder - an adaptation of Strindberg's Miss Julie - and situates the place of these techniques within contemporary Acting discourse.

Abstract (sommario):
This thesis is an exploration of movement pedagogy as a continuation of basic acting lessons from Stanislavski. Using the example of an introductory semester of movement instruction, physical acting and movement concepts are explained in terms of their connection to and derivation from universally accepted acting terminology and ideas. This is put forth as a way to facilitate the synthesis of movement instruction with other acting curriculum, as well as providing a new way to view some familiar acting concepts. Several specific examples are explored in more depth as case studies in physical equivalents to the intellectual, visual, or emotional techniques familiar to all with a basic knowledge of Stanislavski based acting principles.

Abstract (sommario):
This study investigated the reported benefits of adolescent students with an intellectual disability as tutors or teacher assistants in physical education. The personal experience and meaning of five young leaders with an intellectual disability was reported by means of an interview, video activity, picture activity, and field observations. An interpretative phenomenological analysis (IPA) was used. The analysis revealed different combinations of benefits for each participant. All participants expressed positive affects from helping. Four participants identified pride and accomplishment and connectedness. Three participants demonstrated career ambitions and self-identity. Two participants addressed responsibility towards others, being a role model, and modification of behavior. One student addressed altruism as a benefit. The young leaders' experiences were discussed in terms of their roles (Allen & Feldman, 1976), role theory (Thomas & Biddle, 1966), previous tutoring literature, and Hellison's responsibility model (2003).

Abstract (sommario):
Fluidelastic vibration of tube arrays caused by cross-flow has recently been highlighted by a practical event. There have been many studies on fluidelastic instability, but almost all works have been devoted to the tube-vibration in the transverse direction to the flow. For this reason, there are few data on the fluidelastic forces for the in-flow movement of the tubes, although the measured data on the stability boundary has gradually increased. The most popular method to estimate the fluidelastic force is to measure the force acting on tubes due to the flow, combined with the movement of the tubes. However, this method does not give the physical explanation of the root-cause of fluidelastic instability. In the work reported here, the in-flow instability is assumed to be a nonlinear phenomenon with a retarded or delayed action between adjacent tubes. The fluid force acting on tubes are estimated, based on the measured data in another paper for the fixed cylinders with distributed pressure sensors on the surface of the cylinders. The fluid force acting on the downstream-cylinder is assumed in this paper to have a delayed time basically based on the distance between the separation point of the upstream-cylinder to the re-attachment point, where the fluid flows with a certain flow velocity. Two models are considered: a two-cylinder and three–cylinder models, based on the same dimensions as our experimental data to check the critical flow velocity. Both models show the same order of the critical flow velocity and a similar trend for the effect of the pitch-to-diameter ratio of the tube arrays, which indicates this analysis has a potential to explain the in-flow instability if an adequate fluid force is used.

Abstract (sommario):
Hydraulic energy is constructed from real and imaginary energies. Their acting directions are normal to each other. Their physical properties are quite different. All the physical parameters, such as force, velocity, and acceleration therefore consist of two different type real and imaginary functions. Physically, there are three different types of fluid particles rotational motion: straightly forward non-rotational motion, which is based upon kinetic real physical parameters, circularly forward rotational motion, which is based upon un-kinetic imaginary physical parameters, and their combined rotational motion. Their interrelation is shown in diagram.

Abstract (sommario):
This paper deals with a vibratory problem of fluid-structure interaction. It considers the two-dimensional case of a rigid, smooth and circular cylinder undergoing transverse sinusoidal oscillations and immersed in a viscous fluid otherwise at rest. Our work is focused on the in-line force acting on the cylinder in unsteady laminar flow. The aim is to understand the variations of the force with time according to the configuration of the physical system. For that the analysis will also use an energetic approach based on the power balance. The physical system can be characterized by two non-dimensional numbers: the Reynolds number (Re) compares the importance of the fluid viscosity to its inertia, and the Keulegan-Carpenter number (Kc) measures the amplitude of the cylinder displacement compared to its diameter. First the incompressible Navier-Stokes equations are solved numerically by means of a finite elements method. The flow structure is analyzed by determining the evolution with time and throughout the computational domain of flow quantities, such as pressure field, vorticity field or stream lines. We also calculate the values versus time of the different terms occurring in the mean force balance and power balance. We compare these results for several pairs (Kc, Re) of “extreme” values. Thus it appears three characteristic configurations: the inertial Euler case (Kc≪1 and inviscid fluid), the Stokes case (Kc≪1 and Re≫1) and the drag case (Kc≫1). For these three reference configurations the physical mechanisms operating in the system are identified. But in intermediate cases, particularly when Kc>1, every mechanisms interact. Consequently the evolution of the force acting on the cylinder versus time is more complex and its interpretation becomes less straightforward. That is why a quantitative energetic analysis is carried out. We define a measure of the dissipative energy present in the flow. Then we compare the values of that coefficient for different cases throughout the map (Kc, Re).

Abstract (sommario):
All the physical parameters, such as energy, force, velocity, and acceleration are constructed from two different kinds; one is real and the other is imaginary. Their acting directions are normal to each other. The former acts horizontal direction and causes visible kinetic movement on fluid particle. All the supplied energy is utilized and consumed. The latter acts vertical direction but does not cause any visible kinetic movement on fluid particle. All the energy transfer from mechanical to hydraulic and from hydraulic to mechanical is caused by the imaginary parameters in vertical direction.

Abstract (sommario):
This rapid review updates a previous report (Fraser and Martineau-Searle, 2018) with evidence from 2018 onwards. It finds an evidence base on online gender-based violence (OGBV) covering a wider range of countries than the previous report. Some key findings on the nature and prevalence of OGBV include: The most recent surveys show a prevalence of OGBV ranging from 16% to 58%; Men and boys also experience online abuse in high numbers, but it is less likely to be gender-based; Several studies from different countries identify Facebook as the top location for incidents of OGBV; Higher levels of online harassment and abuse are faced by people with intersecting inequality factors; According to victim-survivors, perpetrators are more likely to be unknown and acting alone, but large numbers are known to the victims. Perpetrators themselves report divergent, multifaceted and often over-lapping motivations for their actions; Analysis of underlying drivers of OGBV highlights an overarching theme of power and control, and heteronormative expectations around gender roles and sexual practice. Many authors recommend that OGBV be understood as part of a continuum of abuse where normalised behaviours, such as sexual harassment in public spaces, shade into behaviours widely recognized as criminal, such as physical assault. The societal impact of OGBV includes: Media freedom is compromised; Democracy being undermined; Economic losses resulting from lost productivity; A ‘climate of unsafety’ prevails. Evidence base: The number of surveys about self-reported experiences with online harassment has increased rapidly. The majority of the research found during the course of this rapid review came from international and domestic non-governmental organisations and think-tanks. Academic research studies were also found, including several literature reviews.

Abstract (sommario):
This work deals with the new, relativistic direction in quantum econophysics, within the bounds of which a change of the classical paradigms in mathematical modelling of socio-economic system is offered. Classical physics proceeds from the hypothesis that immediate values of all the physical quantities, characterizing system’s state, exist and can be accurately measured in principle. Non-relativistic quantum mechanics does not reject the existence of the immediate values of the classical physical quantities, nevertheless not each of them can be simultaneously measured (the uncertainty principle). Relativistic quantum mechanics rejects the existence of the immediate values of any physical quantity in principle, and consequently the notion of the system state, including the notion of the wave function, which becomes rigorously nondefinable. The task of this work consists in econophysical analysis of the conceptual fundamentals and mathematical apparatus of the classical physics, relativity theory, non-relativistic and relativistic quantum mechanics, subject to the historical, psychological and philosophical aspects and modern state of the socio-economic modeling problem. We have shown that actually and, virtually, a long time ago, new paradigms of modeling were accepted in the quantum theory, within the bounds of which the notion of the physical quantity operator becomes the primary fundamental conception(operator is a mathematical image of the procedure, the action), description of the system dynamics becomes discrete and approximate in its essence, prediction of the future, even in the rough, is actually impossible when setting aside the aftereffect i.e. the memory. In consideration of the analysis conducted in the work we suggest new paradigms of the economical-mathematical modeling.

Testo completo
Abstract (sommario):
The Golog action programming language is a powerful means to express high-level behaviours in terms of programs over actions defined in a Situation Calculus theory. In particular for physical systems, verifying that the program satisfies certain desired temporal properties is often crucial, but undecidable in general, the latter being due to the language’s high expressiveness in terms of first-order quantification and program constructs. So far, approaches to achieve decidability involved restrictions where action effects either had to be contextfree (i.e. not depend on the current state), local (i.e. only affect objects mentioned in the action’s parameters), or at least bounded (i.e. only affect a finite number of objects). In this paper, we present a new, more general class of action theories (called acyclic) that allows for context-sensitive, non-local, unbounded effects, i.e. actions that may affect an unbounded number of possibly unnamed objects in a state-dependent fashion. We contribute to the further exploration of the boundary between decidability and undecidability for Golog, showing that for acyclic theories in the two-variable fragment of first-order logic, verification of CTL properties of programs over ground actions is decidable

Abstract (sommario):
Original objectives Elucidation of PABP's role in crTMV148 IRES function in-vitro using wheat germ extract and krebs-2 cells extract. Fully achieved. Elucidation of PABP's role in crTMV148 IRES function in-vivo in Arabidopsis. Characterization of the physical interactions of PABP and other potential ITAFs with crTMV148 IRES. Partly achieved. To conduct search for additional ITAFs using different approaches and evaluate the candidates. Partly achieved. Background of the topic The power of internal translation via the activity of internal ribosomal entry site (IRES) elements allow coordinated synthesis of multiple gene products from a single transcription unit, and thereby enables to bypass the need for sequential transformation with multiple independent transgenes. The key goal of this project was to identify and analyze the IRES-trans-acting factors (ITAFs) that mediate the activity of a crucifer-infecting tobamovirus (crTMV148) IRES. The remarkable conservation of the IRES activity across the phylogenetic spectrum (yeast, plants and animals) strongly suggests that key ITAFs that mediate its activity are themselves highly conserved. Thus, crTMV148 IRES offers opportunity for elucidation of the fundamental mechanisms underlying internal translation in higher plants in order to enable its rational manipulation for the purpose of agricultural biotechnology. Major conclusions and achievements. - CrTMV IRES requires PABP for maximal activity. This conclusion was achieved by PABP depletion and reconstitution of wheat germ- and Krebs2-derived in-vitro translation assays using Arabidopsis-derived PABP2, 3, 5, 8 and yeast Pab1p. - Mutations in the internal polypurine tract of the IRES decrease the high-affinity binding of all phylogenetically divergent PABPs derived from Arabidopsis and yeast in electro mobility gel shift assays. - Mutations in the internal polypurine tract decrease IRES activity in-vivo. - The 3'-poly(A) tail enhances crTMV148 IRES activity more efficiently in the absence of 5'-methylated cap. - In-vivo assembled RNPs containing proteins specifically associated with the IRES were purified from HEK293 cells using the RNA Affinity in Tandem (RAT) approach followed by their identification by mass spectroscopy. - This study yielded a list of potential protein candidates that may serve as ITAFs of crTMV148 IRES activity, among them are a/b tubulin, a/g actin, GAPDH, enolase 1, ribonuclease/angiogenin inhibitor 1, 26S proteasome subunit p45, rpSA, eEF1Bδ, and proteasome b5 subunit. Implications, both scientific and agriculture. The fact that the 3'-poly(A) tail enhances crTMV148 IRES activity more efficiently in the absence of 5'-methylated cap suggests a potential joint interaction between PABP, the IRES sequence and the 3'-poly(A). This has an important scientific implication related to IRES function in general.

Abstract (sommario):
A knowledge-based program defines the behavior of an agent by combining primitive actions, programming constructs and test conditions that make explicit reference to the agent’s knowledge. In this paper we consider a setting where an agent is equipped with a Description Logic (DL) knowledge base providing general domain knowledge and an incomplete description of the initial situation. We introduce a corresponding new DL-based action language that allows for representing both physical and sensing actions, and that we then use to build knowledge-based programs with test conditions expressed in the epistemic DL. After proving undecidability for the general case, we then discuss a restricted fragment where verification becomes decidable. The provided proof is constructive and comes with an upper bound on the procedure’s complexity.

Abstract (sommario):
This report summarizes the primary output of this project, a book of COVID-era street reconfiguration case studies called Rethinking Streets During COVID-19: An Evidence-Based Guide to 25 Quick Redesigns for Physical Distancing, Public Use, and Spatial Equity. COVID-era needs have accelerated the process that many communities use to make street transformations due to: a need to remain physically distanced from others outside our immediate household; a need for more outdoor space close to home in every part of every community to access and enjoy; a need for more space to provide efficient mobility for essential workers in particular; and a need for more space for local businesses as they try to remain open safely. This project is the third in a series of NITC-supported case study books on best practices in street reconfigurations for more active, sustainable, and in this case, COVID-supportive uses. The full, 154-page book is available for free download from the National Institute for Transportation and Communities (NITC).

Abstract (sommario):
Disruptive socio-natural transformations and climatic change, where system invariants and symmetries break down, defy the traditional complexity paradigms such as machine learning and artificial intelligence. In order to overcome this, we introduced non-ergodic Information Physics, bringing physical meaning to inferential metrics, and a coevolving flexibility to the metrics of information transfer, resulting in new methods for causal discovery and attribution. With this in hand, we develop novel dynamic models and analysis algorithms natively built for quantum information technological platforms, expediting complex system computations and rigour. Moreover, we introduce novel quantum sensing technologies in our Meteoceanics satellite constellation, providing unprecedented spatiotemporal coverage, resolution and lead, whilst using exclusively sustainable materials and processes across the value chain. Our technologies bring out novel information physical fingerprints of extreme events, with recently proven records in capturing early warning signs for extreme hydro-meteorologic events and seismic events, and do so with unprecedented quantum-grade resolution, robustness, security, speed and fidelity in sensing, processing and communication. Our advances, from Earth to Space, further provide crucial predictive edge and added value to early warning systems of natural hazards and long-term predictions supporting climatic security and action.
