Abstract (sommario):
Kentucky reported a solid 13.1% growth in certified organic land from 1997 to 2002. The relative lack of research on disease management practices in Kentucky consistent with organic regulations is an issue that needs to be addressed to provide more reliable information to local farmers. Thus, the first objective of this research was to investigate the potential disease control obtained with natural, organically certifiable spray materials against Colletotrichum orbiculare in vitro and in vivo. The second objective was to test certifiable spray materials in combinations to identify synergistic interactions. The third objective was to evaluate Organic Material Review Institute (OMRI)-certified materials for managing Septoria leaf spot and early blight in tomato under field conditions. The fourth objective was to evaluate chitosan-based products against C. orbiculare in vitro and in vivo. Essential oils, Trilogy®, and Actinovate®, failed to suppress C. orbiculare in vitro as well as cucurbit anthracnose. Bicarbonate salts, Regalia®SC, Sonata®, copper based-products, lime sulfur and water-soluble chitosan showed high antifungal activity in vitro. Bicarbonate salts, Sonata®, Serenade Max®, Soil Gard 12G®, copper based-products and lime sulfur reduced anthracnose disease severity in vivo. In the synergism experiments only a limited number of mixtures showed synergistic interactions, but even in those cases, the effect was not consistent between experiments. The main response obtained was antagonism. In field experiments the most effective fungicides for managing Septoria leaf spot and early blight of tomato were copper-based fungicides. None of the biological-based products (Sonata® and Serenade Max®)), plant-based extracts (Trilogy® and Regalia® SC), chitosan, ammonium bicarbonate nor horticultural lime sulfur provided a significant reduction in disease severity. For the fourth objective, water-soluble chitosan with a molecular weight between 3 to 10 kDa (80 and 85% deacetylated) showed the highest antifungal activity among all chitosan-based products evaluated in vitro. Also, combining the in vitro and in vivo results suggest that the antifungal activity of chitosan-based products is molecular weight- and concentration-dependent. These results provide a significant advance in the evaluation of the efficacy of OMRI-certified materials and natural materials to help organic farmers in Kentucky and the USA to manage diseases.

Abstract (sommario):
This work presents the synthesis of new radical probes to investigate enzymatic activity. Those probes, named “shifting nitroxides”, are synthesized to develop new methods for the detection of enzymatic activity using EPR, but also for an application in diagnosis by OMRI. The shifing nitroxides could then be an efficient alternative to the gadolinium based contrast agents. The first part focused on the optimization of the synthesis of the free shifting nitroxyde, a key synthetic intermediate. The yield has been improved from 2 to 13 % over ten synthetic steps. The second part is devoted to the synthesis of several nitroxides based probes from the free shifting nitroxide. Thus, this manuscript describes the synthesis of nitroxydes suitable for the detection of lipases by EPR spectroscopy. Other probes have been synthesized for the early detection of pulmonary inflammations: one is currently used for the development of earth’s magnetic field MRI, and the other has been used to develop the first example of in vivo detection of a disease by OMRI. Finally, the synthesis of the first family of shifting nitroxydes recognized by UPA is also described. These probes are then promising for the early diagnosis of cancers using OMRI

Abstract (sommario):
The CsgD protein is the master regulator of a pathway leading to the formation of curli, in essence regulating the switch between a motile and a sessile lifestyle for bacteria. The 5’-UTR region of the csgD mRNA is a hotspot for multiple regulatory small RNAs (sRNA) involved in a complex regulatory network. Even though it is previously known how the interaction takes place it is unknown how sRNA binding affects the translational activity. In order to suggest a mode of regulation a mutational assay was performed by making changes in the csgD 5’-UTR and investigate what the translational effects were. Mutations in different regions are shown to affect the translation levels in various ways.

Abstract (sommario):
This project is about building a three wheeled robot car with Omni wheels. Omni wheels can make a robot drive sideways without rotating first. They can make a robot rotate at the same time as it goes straight ahead. An Omni wheel robot can for example be good choice for a tracking robot, though it can drive more effective than a robot car with regular wheels. The thing that speaks against Omni wheels is that it has more friction and it takes more power to rotate the wheels. This robot car is an obstacles avoiding robot that should with help of Ultrasonic sensors and IR-sensors be able to drive around in a room without crashing in to objects or walls. With the help of Omni wheels the robot should drive without rotating much which makes it more effective than a robot car with regular wheels.

Abstract (sommario):
This work relates the continuity and advances in the implementation of the Overhauser-enhanced Magnetic Resonance Imaging technique on a 0.2 T scanner. Briefly, OMRI technique is based on polarization transfer of saturated electronic spins from free nitroxide radicals to proton spins of surrounding water molecules in the aim to drastically enhance proton NMR signal. To this technique, our research team has merged specific strategies for proteolytic activity detection. The first strategy relies on a 3D visualization of proteolytic activity happening in intact living cells or in vivo in healthy mice. With an Overhauser switch based upon changes in molecular tumbling time, high Overhauser enhancements of 10-fold were observed in the intestinal tract of mice after that elastolytic activity of our probe: the nitroxide-labeled elastin macromolecule took place. In addition, MRI developments - TrueFISP sequence implementation, undersampling Keyhole method and data reconstruction were carried out for imaging these rapid biological processes. A second exquisite strategy is also described using nitroxides with shifting resonant peaks. Here, a Beta-phosphorylated nitroxide molecule was specifically detected at two distinct frequencies: one for its substrate and the other for its product once hydrolysis took place. This hydrolysis was imaged in 3D in the stomach of living mice with Overhauser enhancements of more than 400% and with a good spatiotemporal resolution. The perspectives of this work lie on a future detection of a pathological proteolytic activity in vivo and eventually and development of very low magnetic field OMRI

Abstract (sommario):
The purpose of the Open Mobile Service Interface (OMSI) is to simplify the device management process for mobile devices from different manufacturers with a single PC application, instead of using one unique application for every manufacturer. The different OMSI use cases include device management for application vendors, points-of-sale, repair centers or for self-service. This will lead to higher service level for end users, faster repair times, better control over service transactions, an open market for compatible applications and an easy plug-and-play installation. Manufacturers are currently in the process of developing their own specific modules and there is an increasing need for test and verification software to certify OMSI conformance. In order for phone manufacturers to be able to efficiently verify that their OMSI modules and catalogs support and comply with the OMSI standard, there is a need for automated module tests and manual catalog tests. Development of such tests is the main purpose of this Master thesis work. The implementation of the different verification processes have been divided into different sub-projects to create a more structured view of the OMSI Test Suite project and easier management. The first part of the thesis work deals with the module verification process, second part with the client verification process while the third and final part deals with the catalog verification process. The thesis work has been performed in a project form, where the development of the project plan was a part of the thesis work. The final version of the Module Interface Verifier was implemented in C#, in a Visual Studio .NET 2003 environment. The software functioned as expected, both towards a sample module and Sony Ericsson’s and Nokia’s respective modules. The Client Interface Verifier was developed in a C++ environment and functioned according to the given specifications. The Catalog Interface Verifier was developed in C# environment, built on an already existing part of the OMSI Implementation Framework.

The Stanford University Satellite Systems Development Laboratory will flight test a telemetry reengineering experiment on its student-built SAPPHIRE spacecraft. This experiment utilizes solar panel current information and knowledge of panel geometry in order to create a virtual sun sensor that can roughly determine the satellite's sun angle. The Omni-Directional Differential Sun Sensor (ODDSS) algorithm normalizes solar panel currents and differences them to create a quasi-linear signal over a particular sensing region. The specific configuration of the SAPPHIRE spacecraft permits the construction of 24 such regions. The algorithm will account for variations in panel outputs due to battery charging, seasonal fluctuations, solar cell degradation, and albedo affects. Operationally, ODDSS telemetry data will be verified through ground processing and comparison with data derived from SAPPHIRE's infrared sensors and digital camera. The expected sensing accuracy is seven degrees. This paper reviews current progress in the design and integration of the ODDSS algorithm through a discussion of the algorithm's strategy and a presentation of results from hardware testing and software simulation.

Abstract (sommario):
This study attempts to identify possibilities for creating a unified offline and online customer experience across communication channels in a retail company. With the current technological development and the trend of customers shifting more to the online channel, the customers also expect more from this communication channel. Within the online channel a development into an omni- channel (unified experience across channels) approach is leading to new possibilities, and keeping up with the development will support companies to keep their competitive advantage. The study reviews a case (Mio) with an offline channel represented by several large scale retail stores in Sweden, and also providing an online channel as a website meant to complement the stores. The study reviews previous literature in the field of customer experience both online and offline, omni channel retailing, and marketing. This is the foundation, together with a benchmark case (comparison case), and a survey to gain further insight and widen the knowledge. The results indicates that there are specific features which could contribute to a more positive and seamless experience across channels for the customer. These are information availability, quality and consistency across channels. As well as, functionality for purchases, order handling and tracking, and interactive customer service online. This could preferentially be fused in one place as a customers personal profile, allowing for a portal with everything the customer needs during all phases of the shopping experience. These findings provide insight into how the online channel could be used to better complement the offline channel, working synergistically. Future research is needed to validate these initial findings.

Abstract (sommario):
A robot with the purpose of utilizing so-called omnidirectional movement, movement in every direction, to conquer tough obstacles and rough terrain in order to conduct exploration and rescue missions has been developed at the Kaneko-Higashimori Laboratory at Osaka University, Japan. On account of the Japanese space agency JAXA it had now been requested to have an improved version of this so called Omni crawler developed, in order to further improve its terrain capabilites and even allow it to be thrown into an area and still resume its mission without getting stuck. In this project an improved variation of this model, called the Omni Tri-crawler, has been developed as a CAD model and a basic physical model, showing how a robot combining a unique type of tracks and a triangular shape could fulfill these requirements and look and behave.

Abstract: The OMNI system provides facilities for the creation, communication and management of processes in an heterogeneous network of computers running UNIX. It extends UNIX concepts by providing similar services on a distributed system. The system's three main modules, called the Name Server, the Communication Ports Module and the Process Manager are briefty described. Every system entity, such as ports, processes or port groups, has a unique, homogeneous, system-wide identification called OMNIid, which identifies it in both time and space. The Name Server is responsible for associating user-defil1ed names with these identifications, so that they can be later retrieved by other processes anywhere on the network, thus providing transparency with regard to actual entity location. The Communication Ports Module provides a mechanism used by processes to communicate with one another by sending messages to ports. Ports can be connection or connectionless. A message can only be sent to a connection port by its connected peer, while any process can send a message to a connectionless port. The system also supports special connectors and port groups, which allow a message to be multicast to many ports or to be delivered to one of several possible ports only. The Process Manager is responsible for creating distributed processes, sending signals to them, detecting and reporting their termination. Most UNIX services are extended, allowing parent processes to be interrupted by a signal at child's death or block until child termination. UNIX process groups and sessions are also extended.

Abstract (sommario):
Omni-channel retailing, the combination of online and traditional store channels, has led to the use of traditional stores as fulfillment centers for online orders. A key aspect of omni-channel fulfillment problems is the tradeoff between cancellations of accepted online orders and profits: a riskier fulfillment policy may result in more online sales but also more cancelled orders. In this dissertation, I will describe two approaches to the fulfillment problem cast generally as a stochastic optimization problem of setting inventory thresholds above which the online channel stays open. In the more traditional approach, we build a stochastic model of the process leading to order cancellations for a single item so that retailers may find inventory and fulfillment policies that effectively use this information along with shipping costs between various locations. We describe iterative algorithms based on Infinitesimal Perturbation Analysis (IPA) that converge to optimal and locally optimal policies within certain flexible policy classes for the multiple-location version of this model, and show their empirical performance on simulated data based on real data from a high-end North American retailer. In a more modern approach, we apply techniques from machine learning and discrete optimization to find fulfillment policies that perform well empirically at maximizing revenues subject to a constraint on cancellations across a large portfolio of items. Using the real data mentioned earlier, we build estimators that predict the cancellation probability and other features of incoming online orders. We formulate and solve an optimization problem based on these estimates to get a fulfillment policy in a separate second step. Then we investigate a joint estimation and optimization model based on a neural network to find both the generative parameters for our estimates as well as the policy that maximizes revenue while limiting cancellations. We show how both the separate and joint estimation and optimization models can be used to account for data truncation. The joint methods typically identify policies that more closely track target cancel rates than the separate estimation and optimization methods. For the joint method, we also custom built a neural network layer to efficiently solve the knapsack optimization problem central to our model and demonstrate substantial empirical improvement in running time and scalability over existing methods.

Abstract (sommario):
This thesis presents the design and development of a unique omni-directional platform known as the Omnibot which was built in the Mechatronic and Robotic Systems Laboratory at UOIT. The Omnibot's layout is novel because its drive axes do not intersect with the geometric center of the body, which is typical for omni-directional platforms using segmented omni-directional wheels. This design enables the center of mass to be lower in the design and increases the stability. A suspension system was designed for each of the four wheels to limit vibrations and to ensure contact between the wheels and operating surface. The Omnibot was built to modularly support many systems, including a robot arm, without altering the mechanical design of the frame. Two control modes were developed: local and global. Commands to drive the Omnibot can be received from either a joystick that can be directly interfaced with the controller or with commands that are sent from other systems that are either on or o of the Omnibot. Both control modes require encoder feedback to ensure commanded velocities are being executed as specified. Global control requires feedback from an indoor localization system to determine the Omnibot's pose. Early implementation of the localization system is discussed. An open source robotics software, known as Robot Operating System (ROS) was selected for implementation of the Omnibot systems. ROS serves as a middleware which allows components, such as the localization system and remote desktop, to communicate with each other through a decoupled messaging system. ROS is modular and exible, allowing for easy adaptation of future components. Test results of the Omnibot in operation are presented.

Abstract (sommario):
The rescue of blocked replication forks is vital to the faithful duplication of a cell's entire genome once per cell cycle. Eukaryotic cells are equipped with the intra-S phase checkpoint that co-ordinates the multitude of cellular responses to replicative stress, and evidence implicate the checkpoint in reflating fork restart by recombination pathways. In this study, I investigate the role of various Schizosaccharomyces pombe checkpoint gene products in the regulation of recombination at forks arrested by a programmed protein barrier, Replication Termination Sequence 1 (RTS1).

Abstract (sommario):

In this work we seek to establish a first contact with Geometric Analysis, aiming the understanding of the Good Shadow Principle of Fontenele and Xavier ([FX11]), which is a generalization of the Omori-Yau Principle. We will expose the basic results that are needed for their comprehension, and extend the study to other topics of Geometric Analysis, as the heat kernel, the existence of exhaustion functions and estimates to the gradient of harmonic functions and subsolutions of the heat equation. Once understood the Good Shadow Principle, we intend to extend it by proving that the class of the second order uniformly bumpable manifolds, introduced by Azagra and Fry in [AF10], also satisfies this principle.

Abstract (sommario):
The Omai goldfield consists of mesothermallode gold mineralisation and associated saprolitealluvial placer deposits, hosted within Palaeoproterozoic granite-greenstone terrain of the Guiana Shield. Total mining reserves are estimated at 44.8 million tonnes, grading at 1.43 glt Au. The goldfield lies along an ESE-trending, regional-scale structure referred to as the Issano-Appaparu shear zone. At Omai, the gold deposits are located in two discrete ore zones - the Omai Stock Zone and the Wenot Lake Zone. The bulk of primary mineralisation is centred on a high-AI, quartz diorite-tonalite boss (the Omai Stock), where wall-rock alteration is dominated by a hydrothermal sericite-carbonate assemblage. The primary ore package of Au-W-Te-S mineralisation is contained in a series of narrow (1-5cm), quartzcarbonate (ankerite) veins. Visible gold is commonly associated with galena and microscopic tellurides. Provisional fluid inclusion studies have indicated that the parent hydrothermal fluid was H20-C02 -bearing (- 5.0 mol% CO2), of low salinity (0-1.8 wt. % NaCI equiv.) and moderate density (0.96 g/cm3). The depositional temperature of the fluid was probably in the region of 200-400oC. Preliminary 6'80 values are consistent with a magmatic andlor metamorphic source. The Majuba Suite greenstones adjacent to the Omai Stock are also partially mineralised. These rocks are dominated by primitive, low-K, high-Fe tholeiitic (HFT) basalts and minor basic intrusives which are characterised by flattish REE patterns of around 10 times chondrite. A subordinate, calc-alkaline series (CAS), of mostly andesitic composition, is intercalated within the volcanic pile. In terms of trace element geochemistry, the Majuba Suite is chemically comparable to volcanic rocks from modern island arc settings. The REE geochemistry, which proved to be extremely diagnostic throughout, indicates that the Omai Stock is genetically related to the regional Tigri pluton. This and other TransAmazonian soda granitoid plutons in the area have chemical affinities akin to calc-alkaline volcanic arc granitoids of Phanerozoic orogenic belts. Several lamprophyric (appinite) pipes are spatially and temporally associated with the Omai Stock. Although the appinites are only weakly mineralised, they may have a genetic significance regarding the source of the gold. Both the Omai Stock and Majuba Suite are cut by a series of partially mineralised, ultrabasic to intermediate dykes (the Gilt Creek Suite) which intruded along shallow-dipping fracture zones. These rocks are typically Mg-rich, LREE enriched (LanlYbn = 7.84-32.6) and appear to be the product of alkaline arc magmatism. A few post-orogenic mafic dykes (POMDs), of probable Mesozoic age, are also identified in the area. These rocks are strictly nonmineralised and are chemically correlatable with tholeiitic basalts from continental rift-related settings. In the Wenot Lake Zone, lode mineralisation is hosted within thin bands of high-silica rhyolites (HSRs), also of the Majuba Suite, close to a sheared contact with phyllitic tuffs. The HSRs have a distinct chemistry characterised by elevated Si02 (> 75.0%), Na, Nb, Zr and REE contents with anomalously low concentrations of Mg, Ti, Ca, K, Rb and Sr. These rocks are further characterised by wing-shaped REE patterns containing deep negative Eu anomalies.

Abstract (sommario):
Cells sense the properties of the surrounding environment and convert this information into changes in gene expression. Bacteria are, in contrast to many multi-cellular eukaryotes, remarkable in their ability to cope with rapid environmental changes and to endure harsh and extreme milieus. Previously, control of gene expression was thought to be carried out exclusively by proteins. However, it is now clear that small regulatory RNAs (sRNA) also carry out gene regulatory functions. Bacteria such as E. coli harbor a large class of sRNAs that bind to mRNAs to alter translation and/or mRNA stability. By identifying mRNAs that are targeted by sRNAs, my studies have broadened the understanding of the mechanisms that underlie sRNA-dependent gene regulation, and have shed light on the impact that this type of regulation has on bacterial physiology. Control of gene expression often relies on the interplay of many regulators. This interplay is exemplified by our discovery of mutual regulation between the sRNA MicF and the globally acting transcription factor Lrp. Through double negative feedback, these two regulators respond to nutrient availability in the environment which results in reprogramming of downstream gene expression. We have also shown that both the transcription factor CsgD, and the anti-sigma factor FlgM, are repressed by the two sRNAs OmrA and OmrB, suggesting that these sRNAs are important players in the complex regulation that allow bacteria to switch between motility and sessility. Bacterial populations of genetically identical individuals show phenotypic variations when switching to the sessile state due to bistability in gene expression. While bistability has previously been demonstrated to arise from stochastic fluctuations in transcription, our results suggest that bistability possibly may arise from sRNA-dependent regulatory events also on the post-transcriptional level.

Abstract (sommario):
This thesis describes a method to create local maps from an omni-directional vision system (ODVS) mounted on a mobile robot. Range finding is performed by a structure-from-motion method, which recovers the three-dimensional position of objects in the environment from omni-directional images. This leads to map-making, which is accomplished using certainty grids to fuse information from multiple readings into a two-dimensional world model. The system is demonstrated both on noise-free data from a custom-built simulator and on real data from an omni-directional vision system on-board a mobile robot. Finally, to account for the particular error characteristics of a real omni-directional vision sensor, a new sensor model for the certainty grid framework is also created and compared to the traditional sonar sensor model.

Abstract (sommario):
Background Omni-channel retailing has developed as an extension of multi-channel retailing. The difference between the concepts is the level of integration, where an omni-channel requires seamless and complete channel integration compared to a multi-channel where the level of integration varies from non-existing to high. Today’s customers expect an integrated shopping experience where they can combine the different channels according to their preferences and omni-channel is a way for retailers to meet this need. Purpose The purpose of this thesis is to explore the concept of omni-channel retailing and its implementation in retail firms operating in the Swedish market. Method This study is based on an interpretive approach and takes the form of an exploratory multiple case study through the investigation of four retailers. Qualitative data was collected through semi-structured interviews and the findings were analyzed abductively. Conclusion The conclusion of this thesis is that the investigated retailers still have, more or less, a long way to go in their integrative efforts. The retailer that has come the furthest in their effort has only implemented nine out of fifteen omni-channel criteria. Three main incentives for implementing an omni-channel strategy were identified. These are to respond to customer demands, to optimize the individual sales channels and to gain a competitive advantage. Additionally, several challenges arise as the retailer moves towards an omni-channel. These include technological investments, change management, channel conflict, return management and consistency. Furthermore, five potential sources of these challenges were identified as technological requirements, organizational changes, sales accreditation policies, in-store returns and finally the franchise business model.

This thesis provides a deep mathematical analysis of the diverse alternatives for routing models considering an Omni-channel supply chain. The natural evolution of supply chains from traditional brick-and-mortar stores to an omni-channel supply chain, encompassing and merging e-commerce together with a multi-channel concept, allows businesses to reach new levels of operational efficiency by leveraging inventory closer to the customer and making decisions on the fly on how to better and more cheaply provide a service/product to the final consumer. The flexibility and benefits, unfortunately, do not come without a certain dose of complexity and further development of the supply chain tactical implementation and systems. New alternatives to fulfill customer orders are available, which require greater screening among the different alternatives. An effective routing model becomes essential to make sure these alternatives are properly considered in order to satisfy both the consumer and retailer objectives, such as on-time delivery of orders, retail stores' service levels, and fulfillment costs.

Abstract (sommario):
This dissertation provides a survey of the available archaeological sources that have been discovered that help illuminate the history of the Northern Kingdom of Israel from King Omri (876 BCE) into the Assyrian Exile (721 BCE) of the Iron II a and b periods. This history is given in the Hebrew Scriptures as found in 1 Kings 16:16 to 2 Kings 17:6. To accomplish this research, several resources have been organized in a systematic approach. The artifacts and physical remains are examined in categories including: pottery, town planning, architecture, literacy and inscriptions, temples, gods, cult objects, metallurgy, weapons and warfare, weights and measures, farming, food preparation, music, jewelry, art, and burial. Photographs of several artifacts are included. With the background developed on the material culture, the next topic of importance to be considered is the archaeological sites and their relationship to this historic period. The relevant archaeological excavations and in-situ artifacts surveyed were found in areas of the former cultures including: Israel, Judah, Sinai, Edom, Moab, Amman, Lebanon, Syria, Aram, Assyria, Babylon and Persia. A vast number of these sited reveal information regarding the time period from King Omri, his son King Ahab and Ahab's wife, Queen Jezebel. There was interaction between Israel and Judah, and the kingdoms in Moab, Ammon, Lebanon, Aram, and Assyria. Eventually in 721 BCE many people were taken from the Israel into Exile and captivity by the Assyrians. The conclusion of this dissertation ties together the findings of the material sources with the historic events. The material sources have given considerable light on the history of the Northern kingdom as was given in the Hebrew Scriptures as is presented in this final section of the dissertation.

In this thesis, a self-localization algorithm and a software platform are developed for an omni-directional robot with omni-directional vision. The research is divided into two parts: robot self-localization and robotics software platform. In the development of robot self-localization algorithm, a recursive particle filter with probabilistic distribution is utilized to find the robot pose in the environments. The particle filter is composed of a motion model and a sensor model. The motion model is constructed based on robot kinematics with motor encoder feedback, while the sensor model is established by using an omni-directional vision system. In the development of robotics software platform, a software module is programmed for the robot mechatronics system to solve the interface problem for robots with heterogeneous hardware, including humanoid robots, two-wheeled robots, and omni-directional robots. The software platform is developed on Window PC-based controllers to resolve the interface and communication problems between these controllers.

Abstract (sommario):
Our perception, of the Omrid kings of the Kingdom oflsrael in the ninth century BCE, is based on the Books of 1 and 2 Kings in the Hebrew Bible. The Biblical author's concentration, on Omrid apostasy rather than on their abilities and accomplishments, has robbed these competant monarchs of the prominence allotted to kings like David and Solomon. Recent archaeological excavations, in conjunction with extra-Biblical sources, have however projected a different image. Excavations at the royal Omrid cities of Samaria, and especially Jezreel, have indicated that Omri, and his son Ahab, had erected immense and grandiose structures. These edifices bear testimony to periods of peace, stability and great economic prosperity. The Omrids deserve new assessments as to their accomplishments, and therefore, by means of visible and tangible structural remains, I wish to promote the persuasion of archaeology as vindication of Omrid grandeur and achievement at Samaria and Jezreel.

A vision-based system for vehicle detection and tracking in video streams is an important research in computer vision. It also plays an important role in ITS (Intelligent Transportation Systems). Mounting high omni-directional camera allows the omni-directional camera to cover a wider area. Motion detection aims to capture the changed region from the omni-directional video image sequences and build a background image. In this paper, a multiple-vehicle counting and tracking framework for intersection traffic surveillance is proposed. In order to accurately locate vehicles, we use Kalman filter to correct vehicle trajectories. According to our experimental results, our approaches can analyze the vehicles at intersection efficiently.

Novel inventions and the hierarchic controls for under-actuated systems are proposed and studied in the thesis. The first invention by cooperating two different sizes of idle wheel can result in a proper-circle Omni wheel. The non-shaft-support type design can enhance the strength of Omni wheel. Locking mechanism only for the contact-floor idle wheel of an Omni wheel can control its lateral sliding effect is the second invention. The third invention is to use Killough wheel with power as the locking mechanism for an Omni wheel. The fourth invention is a spherical robot that uses Killough wheels with power to control rotation of a ball. In the thesis, the proposed control algorithms are derived based on a spherical robot. The conventional state-dependent-switching coefficients are adopted to assemble sliding surfaces or subsystems in the hierarchical structures, so the control inputs with varied ability can overcome the under-actuated problem. However, the switching coefficients are modified to be positive ones in the thesis. Therefore, the proposed control algorithm can only ensure a stable subsystem state, but the stability of the original state cannot be guaranteed. The vector approach and coordinate transformations are adopted into the hierarchical control to result in two vector-coordinate transformation hierarchical controls (VCHCs) and the geometric coordinate hierarchical control (GHC).

The purpose of this research is to construct a comprehensive security system to enhance community safe. The system shall offer accompanying care for the elderly and infirm in the community; friendly guides and personal surveillance for visitors; fire hazard and foreign intrusion detection and intelligent patrol. The system has been specifically designed to overcome the shortcomings of existing surveillance and security systems deployed in community environments such as apartment buildings that are not capable of offering adequate security and protection to residents and visitors. Accompany care is a form of personal surveillance intended for children or the elderly through the application of RFID sensing and positioning technology. It is complemented with online real-time image feeds and mobile devices so that users are able to keep an eye on the movements of children or elderly under surveillance and prevent them from entering potentially dangerous areas. This will effectively reduce the occurrence of accidents for the disadvantaged residents in the community. Accompanying care and guide for visitors make use of RFID positioning technology to monitor visitors’ activities after they have entered the community. If a visitor loses his direction in the community or idles in the community after visiting a specific resident, the system will be able to issue real-time messages to help guide the visitor to the residence he wishes to visit. In addition, an online video feed allows community security to monitor the visitor’s activities in the community, including any possibly suspicious activity. In light of the frequency of incidents such as fire hazards and theft in communities, the system featured in this study is equipped with a surveillance system to capture real time images for fire and skin analysis so that the system may respond immediately by notifying community security in the event of such incidents and effectively prevent hazards and incidents that threaten community safety. Intelligent patrol is a fully-automated patrol system provided to augment existing community security services where the system generates different patrol formats (on-route, reverse route, fixed time and irregular patrols) to eliminate blind spots that security personnel might have missed on their patrol. The combination of different patrol routes will make it difficult for perpetrators to predict the community’s security measures, thereby ensuring highly effective community security. The system proposed in this paper can be easily deployed at communities as an affordable and economic solution. Considering the fact that conventional security measures in communities usually involve a surveillance system, motion detection and manual patrols, such a combination are more susceptible to security loopholes. In this research, we have proposed a better solution that offers comprehensive security protection that further strengthens community safety.

Abstract (sommario):
Transient receptor potential vanilloid-1 (TRPV1) is a member of the TRP family of channels that are responsible for nociceptive, thermal and mechanical sensations. It is primarily associated with neuronal cells, but has been detected in different non-neuronal cells, including leukocytes. Capsaicin (CAP), the active ingredient of hot chilli peppers, is one of a number of related endogenous and plant-derived compounds (broadly termed ‘vanilloid-like agents’) that have been shown to induce apoptosis and inhibit cell proliferation in some cancer cells, through both TRPV1-dependent and -independent mechanisms. The expression and function of TRPV1 in haematological malignancies however, has not been extensively investigated. Specific targeting by vanilloid-like agents toward TRPV1 on cancerous cells in patients with haematological malignancies may represent a novel therapeutic approach to treating these diseases. This thesis investigated the expression and function of TRPV1 in haematological malignancies, using both blood cancer cell lines and blood samples obtained from patients with different blood cancers. The specific aims were to; 1) study the effect of TRPV1 agonists and antagonists on the viability of THP-1, U266B1 and U937 haematological malignant cell lines, 2) validate and optimise Western blotting and flow cytometry protocols to detect TRPV1 expression in leukocytes, 3) investigate TRPV1 expression in THP-1, U266B1 and U937 cells, and 4) compare TRPV1 expression in leukocytes obtained from patients with blood cancers to normal subjects. The thesis begins with a comprehensive review and discussion on TRPV1 structure and function, as well as its expression and role in health and disease. In particular, there is a focus on the role of TRPV1 in cancer, including haematological malignancies (Chapter 1). In Chapter 2, the effect of CAP on the metabolic activity of three malignant haematological cell lines, THP-1, U266B1 and U937, was investigated. Metabolic activity assays were performed using the alamarBlue® method. CAP induced cytotoxicity in all three cell lines in a concentration-dependent manner. A biphasic effect on metabolic activity was observed on THP-1 cells [EC50, IC50 (95% CI) = 32.9 (19.9-54.3), 219 (144-246) μM]. U266B1 cells were more resistant to CAP-induced death than THP-1 and U937 cells. TRPV1 and CB1 antagonists (SB452533 and AM251, respectively) suppressed the CAP-induced increase in THP-1 cell metabolic activity (P<0.001). These experiments suggest that CAP inhibits the metabolic activity of malignant haematological cells through a non-TRPV1-dependent mechanism. Chapters 3 and 4 represent the experimental work and trouble-shooting conducted to develop, validate and optimise methods for the detection of TRPV1 expression in human cells. Western blotting (Chapter 3) and flow cytometric (Chapter 4) methods have been previously published, however few have documented the use of appropriate controls for the detection of TRPV1, suggesting that data in the literature may not necessarily be valid. A problem identified in the current study was the correct application of negative controls, particularly to assess the specificity and therefore suitability of the primary antibody used in these methods. These optimised protocols were then used to investigate the expression of TRPV1 in human malignant haematological cell lines (Chapter 5) and leukocytes obtained from patients with blood cancers (Chapter 6). Increased expression of TRPV1 protein was observed in THP-1, U266B1 and U937 cells compared to normal leukocytes. Furthermore, a TRPV1 dimer was detected in U266B1 cells. Interestingly, TRPV1 was detected in non-haematological cell lines that have previously been used as TRPV1-negative cells for Western blotting, including untransfected- and TRPV1-transfected (without tetracycline to switch TRPV1 transcription off) HEK293 and RAW264.7 cells. This latter finding highlights the need for appropriate negative (and positive) controls in both flow cytometric and Western blotting studies of TRPV1. Expression of TRPV1 in leukocytes obtained from patients with a range of haematological malignancies, including multiple myeloma (MM) and B-cell non-Hodgkin’s Lymphoma (B-NHL), was then investigated (Chapter 6). TRPV1 expression was detected in all patients and controls using flow cytometry, but not Western blotting. Using flow cytometry, a sub-group of patients (4/49=8.2%, MM=2, B-NHL=2) demonstrated increased expression of TRPV1 relative to the remainder of the cohort. TRPV1 was found to be similar to the control group for 91.8% of all patients. There were no significant differences in TRPV1 expression (assessed using flow cytometry) between patients with MM and B-NHL, or between de novo patients and those undergoing treatment. Using Western blotting, TRPV1 (~95kDa) was detected in one MM and four B-NHL patients, although interestingly, a 240kDa band was also detected in both a B-NHL and a MM patient. In addition, although C-reactive protein was elevated (≥5 mg/L) in 25% of all patients, it was not associated with higher TRPV1 expression. These results indicate that TRPV1 expression in leukocytes is relatively increased in a small subset of patients with blood cancers, and is not associated with inflammation. Furthermore, some patients may have a unique isoform of TRPV1 that warrants further investigation. In summary, this study has generated new data and knowledge on the role of TRPV1 in haematological cells, including those from patients with blood cancers. A number of novel findings have been reported. Firstly, the inhibition of cell metabolic activity by the TRPV1 agonist, CAP, was found to be independent of TRPV1 activation in malignant haematological cell lines. Secondly, optimised Western blotting and flow cytometric methods for the detection of TRPV1 expression were developed and successfully validated. Thirdly, increased TRPV1 expression was demonstrated in the THP-1, U266B1 and U937 malignant haematological cell lines. Finally, increased TRPV1 expression was observed in some patients with MM and B-NHL, but was not associated with inflammation. The results presented in this thesis can be used as a basis for future studies of TRPV1 function in other human cells and cancers.

Designs of frequency and polarization reconfigurable microstrip patch antennas with conical radiation are respectively presented in this thesis. For the reconfigurable frequency antenna design, a monopolar patch antenna structure is used to generate conical radiation patterns. The frequency switching is realized by controlling four diodes to reconfigure the size of the shorted rectangular patch of the monopolar patch antenna. Each diode connects one side of the shorted rectangular patch and an open stub. For the reconfigurable polarization antenna design, the used antenna structure is single feed and involves two resonant modes, in which a 4-element-array loop antenna mode, producing horizontal polarization, and a monopolar patch antenna mode, producing vertical polarization. By properly exciting and coupling the two resonant modes, circular polarization radiation can be obtained. Moreover, the polarization switching is achieved by controlling four diodes to reconfigure the number of the shorting walls. Two prototypes for the abovementioned reconfigurable frequency and polarization antennas are implemented, respectively, and their dimensions are obtained by numerical analyzing with HFSS. From the measured results, the operating frequency of the reconfigurable frequency antenna has a tunable range from 1820 ~ 2480 MHz, and good omni-directional radiation patterns are observed across the available operating frequencies. As for the reconfigurable polarization antenna, the measured results exhibit the antenna prototype has the ability of switching polarization, and the operating frequencies of the right-hand and left-hand circular polarization occur at 2900 and 2420 MHz, respectively. Also, both the operating frequencies have a return loss of less than 10 dB and an axial ratio of less than 3 dB.

The main goal of this study is to investigate the time-optimal control problem of an omni-directional mobile robot between two configurations. In the proposed method, the time-optimal control problem is formulated and solved as a constrained nonlinear programming (NLP) one. During the optimization process, the count of control steps is fixed initially and the sampling period is treated as a variable to be determined. The goal is to minimize the sampling period such that it is below a specific minimum value, which is set in advance considering the accuracy of discretization. To generate initial feasible solutions of the NLP problem, a systematic approach is also proposed. Since different initial feasible solutions can be generated, the optimization process of the NLP problem can be started from many different points to find the optimal solution. To show the feasibility of the proposed method, simulation results are included for illustration.

Organo-Modified Reservoir Sludge (OMRS) is a good example for re-use of waste resources. The modification technology of reservoir sludge which as an environmental problem turns the reservoir sludge into waterproof material and develops waterproof products which increase reservoir sludge added-value. Porous Asphalt Concrete(PAC) excludes water quickly with high air void content , but it’s easy to cause stripping and raveling in pavement by moisture damage. To improve the resistance of pavement for moisture damage, OMRS , lime and cement were used in this research as fillers in PAC mixtures. The results showed that there was no effect in the permeability coefficient for adding different fillers in the mixtures. In mechanical properties , the strength of specimens with OMRS was weaker than others. Then, three kinds of fillers had similar results on the first day in residual strength test. And it could be found that the longer the specimens soaked in water, the smaller TSR value had .The PAC mixtures with OMRS had better performance in anti-moisture damage.
