AbstractThe diverse activity of flavoenzymes in organic transformations has fascinated researchers for a long time. However, when applied outside an enzyme environment, the isolated flavin cofactor only shows largely reduced activity. This highlights the importance of embedding the reactive isoalloxazine core of flavins in defined surroundings. The latter include crucial non-covalent interactions with amino acid side chains or backbone as well as controlled access to reactants such as molecular oxygen. Nevertheless, molecular flavins are increasingly applied in the organic laboratory as valuable organocatalysts. Chemical modification of the parent isoalloxazine structure is of particular interest in this context in order to achieve reactivity and selectivity in transformations, which are so far only known with flavoenzymes or even unprecedented. This review aims to give a systematic overview of the reported designed flavin catalysts and highlights the impact of each structural alteration. It is intended to serve as a source of information when comparing the performance of known catalysts, but also when designing new flavins. Over the last few decades, molecular flavin catalysis has emerged from proof-of-concept reactions to increasingly sophisticated transformations. There are various barriers to home modifications to prevent falls among the older population. Several strategies may be necessary to overcome these barriers and implement effective home modifications. The need for home modification should be assessed, which requires a home evaluation by a specialist. In Japan, welfare housing environment coordinators have been trained to provide advice on home modifications suitable for people with disabilities. In addition, in Japan, home assessment and advice on home modification before discharge from acute care hospitals for older people is allowed as a medical reimbursement, and a system for effective home modification is well established. Human resource training and medical policy arrangements on home modifications could improve the cost-effectiveness. In Japan, a system has been established to support the costs of home modification and environmental maintenance. Financial support has reduced the barrier to home modification. Fixed grab bars or shower chairs can be rented, which may be more cost-effective than purchasing them and may shorten the time required for installation. There may be psychological barriers to home modification for older population. Since many older people do not recognize the importance of home modification, promotion to convey the value of home modification may be necessary. Training of staff to engage in home modification, public financial support for modification, and ideas for reducing psychological hesitation may help to reduce the barriers for home modification and to enable effective home modification.

SUMMARY One of the first hurdles to be negotiated in the postgenomic era involves the description of the entire protein content of the cell, the proteome. Such efforts are presently complicated by the various posttranslational modifications that proteins can experience, including glycosylation, lipid attachment, phosphorylation, methylation, disulfide bond formation, and proteolytic cleavage. Whereas these and other posttranslational protein modifications have been well characterized in Eucarya and Bacteria, posttranslational modification in Archaea has received far less attention. Although archaeal proteins can undergo posttranslational modifications reminiscent of what their eucaryal and bacterial counterparts experience, examination of archaeal posttranslational modification often reveals aspects not previously observed in the other two domains of life. In some cases, posttranslational modification allows a protein to survive the extreme conditions often encountered by Archaea. The various posttranslational modifications experienced by archaeal proteins, the molecular steps leading to these modifications, and the role played by posttranslational modification in Archaea form the focus of this review.

How much can government-driven mortgage modification programs reduce the mortgage default rate? I compare an economy without a modification option to one with easy modifications, and evaluate the impact of these loan modifications on the foreclosure rate. Through loan modification, mortgage servicers can mitigate their losses and households can improve their financial positions without having to walk away from their homes. When modifying loan contracts is prohibitively costly, the default rate increases 1.5 percentage points in response to a 2007-style unexpected drop in housing prices of 30%. I calibrate the cost of modification after the financial crisis to match the Home Affordable Modification Program (HAMP) modification rate of 0.68%. My quantitative exercises show that current government efforts to promote mortgage modifications reduce the mortgage default rate by 0.63 percentage points.

Abstract Dynamic and reversible RNA modifications such as N6-methyladenosine (m6A) can play important roles in regulating messenger RNA (mRNA) splicing, export, stability and translation. Defective mRNA modification through altered expression of the methyltransferase and/or demethylases results in developmental defects and cancer progression. Identifying modified mRNAs, annotating the distribution of modification sites across the mRNA, as well as characterizing and comparing other modification features are essential for studying the function and elucidating the mechanism of mRNA modifications. Several methods including methylated RNA immunoprecipitation and sequencing (MeRIP-seq) are available for the detection of mRNA modifications. However, a convenient and comprehensive tool to annotate diverse kinds of mRNA modifications in different species is lacking. Here, we developed RNAmod (https://bioinformatics.sc.cn/RNAmod), an interactive, one-stop, web-based platform for the automated analysis, annotation, and visualization of mRNA modifications in 21 species. RNAmod provides intuitive interfaces to show outputs including the distribution of RNA modifications, modification coverage for different gene features, functional annotation of modified mRNAs, and comparisons between different groups or specific gene sets. Furthermore, sites of known RNA modification, as well as binding site data for hundreds of RNA-binding proteins (RBPs) are integrated in RNAmod to help users compare their modification data with known modifications and to explore the relationship with the binding sites of known RBPs. RNAmod is freely available and meets the emerging need for a convenient and comprehensive analysis tool for the fast-developing RNA modification field.

Background Pathogens can impact host RNA modification machinery to establish a favorable cellular environment for their replication. In the present study, we investigated the effect of Toxoplasma gondii infection on host RNA modification profiles and explored how these modifications may influence the host-parasite interaction. Methodology/principal findings We analyzed the modification levels of ∼ 80 nt tRNA and 17–50 nt sncRNAs in mouse liver, spleen, and serum using liquid chromatography and tandem mass spectrometry analysis. The results revealed alterations in RNA modification profiles, particularly during acute infection. The liver exhibited more differentially abundant RNA modifications than the spleen. RNA modification levels in serum were mostly downregulated during acute infection compared to control mice. Correlations were detected between different RNA modifications in the liver and spleen during infection and between several RNA modifications and many cytokines. Alterations in RNA modifications affected tRNA stability and protein translation. Conclusions/significance These findings provide new insight into the role of RNA modifications in mediating the murine host response to T. gondii infection.

Post-translational modifications of histone proteins in conjunction with DNA methylation represent important events in the regulation of local and global genome functions. Advances in the study of these chromatin modifications established temporal and spatial co-localization of several distinct ‘marks’ on the same histone and/or the same nucleosome. Such complex modification patterns suggest the possibility of combinatorial effects. This idea was originally proposed to establish a code of histone modifications that regulates the interpretation of the genetic code of DNA. Indeed, interdependency of different modifications is now well documented in the literature. Our current understanding is that the function of a given histone modification is influenced by neighbouring or additional modifications. Such context sensitivity of the readout of a modification provides more flexible translation than would be possible if distinct modifications function as isolated units. The mechanistic principles for modification cross-talk can originate in the modulation of the activity of histone-modifying enzymes or may be due to selective recognition of these marks via modification of specific binding proteins. In the present chapter, we discuss fundamental biochemical principles of modification cross-talk and reflect on the interplay of chromatin marks in cellular signalling, cell-cycle progression and cell-fate determination.

DNA phosphorothioate (PT) modification is a novel modification that occurs on the DNA backbone, which refers to a non-bridging phosphate oxygen replaced by sulfur. This exclusive DNA modification widely distributes in bacteria but has not been found in eukaryotes to date. PT modification renders DNA nuclease tolerance and serves as a constitute element of bacterial restriction–modification (R–M) defensive system and more biological functions are awaiting exploration. Identification and quantification of the bacterial PT modifications are thus critical to better understanding their biological functions. This work describes three detailed methods derived from iodine-induced specific cleavage-an iodine-induced cleavage assay (ICA), a deep sequencing of iodine-induced cleavage at PT site (ICDS) and an iodine-induced cleavage PT sequencing (PT-IC-Seq)-for the investigation of PT modifications. Using these approaches, we have identified the presence of PT modifications and quantized the frequency of PT modifications in bacteria. These characterizations contributed to the high-resolution genomic mapping of PT modifications, in which the distribution of PT modification sites on the genome was marked accurately and the frequency of the specific modified sites was reliably obtained. Here, we provide time-saving and less labor-consuming methods for both of qualitative and quantitative analysis of genomic PT modifications. The application of these methodologies will offer great potential for better understanding the biology of the PT modifications and open the door to future further systematical study.

The modification of wood cell walls is based on the characteristics of the chemical composition and structure of the cell wall. Various physical and chemical modifications to these characteristics enhance the original properties of the cell wall and give additional functionality. Through complex modification, wood has also obtained the opportunity to become a multifunctional material. Scholars have paid more attention to the microscopic properties of the cell wall with continuous enrichment of modification methods and improvement of modification mechanisms. This article summarizes the methods of cell wall modification in recent years and proposes prospects for future development: (1) innovation of modifiers and combination with modification mechanism, as well as improvement of cell wall permeability; (2) the application directions of cell wall structures; and (3) the application of nano-technologies in cell wall modification. This review provides further ideas and technologies for wood modifications.

Die Produkte von Maschinen- und Anlagenbauunternehmen stellen in der Regel langlebige, kundenspezifische Investitionsgüter dar. Je umfangreicher die Konstruktions- und Montagezeiten des Investitionsgutes, umso frühzeitiger muss der Kunde sich festlegen, welche Leistungsmerkmale gefordert werden und unter welchen Bedingungen die Maschine oder Anlage arbeiten muss. Hierbei kommt es regelmäßig zu Divergenzen zwischen dem Bedarf des Kunden zum Lieferzeitpunkt, der Nachfrage zum Zeitpunkt der Kaufentscheidung und dem Angebot des Anbieters. Um diese Divergenzen ab dem Zeitpunkt des Kaufs zu beheben, sind mehr oder weniger kostenintensive technische Leistungsveränderungen – so genannte Modifications – erforderlich. Die Arbeit zeigt zum einen Erkenntnisse aus der Untersuchung von Modifications. Zum anderen bietet sie ein theoretisch und insbesondere ein empirisch orientiertes, umsetzbares System (Modification- Management), mit dem dieser Problematik nachhaltig begegnet bzw. der Umgang damit optimal gestaltet werden kann.

La SUMOylation est une modification post-traductionnelle essentielle pour toutes les cellules eucaryotes. C’est un processus enzymatique qui permet la liaison covalente du polypeptide SUMO sur des résidus lysine de protéines cibles. La SUMOylation est un processus réversible sous l’action de désumoylases appelées SENP. Il est critique de maintenir un équilibre entre forme modifiée et non modifiée d’un substrat donné. En effet, la dérégulation de la balance SUMOylation/déSUMOylation a été mise en évidence dans plusieurs pathologies cérébrales. La synapse est le point de contact entre les neurones où s’effectue la communication synaptique. Ce sont des structures très denses où le processus de SUMOylation régule l’interaction et la fonction de multiples protéines. Durant ma thèse, j'ai combiné l’utilisation de l'imagerie en temps réel sur cellules vivantes avec des approches biochimiques et pharmacologiques pour identifier les mécanismes de régulation du transport de SENP1. J'ai ainsi démontré que l'activation neuronale augmente les niveaux synaptiques de SENP1. Cette augmentation synaptique résulte de la modification de la vitesse de diffusion de l’enzyme SENP1 entre les dendrites et les synapses d’une part, et d’autre part, de l’augmentation importante du temps de rétention synaptique de l’enzyme. Je rapporte également que ce mécanisme de régulation dynamique de SENP1 implique l'activation des récepteurs métabotropiques du glutamate. De plus, je suggère la participation du processus de phosphorylation dans cette régulation synapto-dendritique de SENP1 mettant ainsi en lumière un nouveau mécanisme de régulation de la balance neuronale entre SUMOylation et déSUMOylation
Sumoylation is a vital eukaryotic posttranslational modification. Sumoylation occurs as an enzymatic cycle that conjugates SUMO proteins to target proteins. SUMO proteases (SENP) deconjugate SUMO from modified proteins and thus maintain balanced levels of SUMOylated and un-SUMOylated proteins required for physiological homeostasis. Neuronal synapses are protein-rich structures that underlie synaptic transmission and plasticity. Strong evidence exists that sumoylation occurs in synapses and regulates the function of synaptic proteins. Indeed, distortion of the SUMO balance has been linked to several pathologies of the synapse. Gaining a deeper understanding into the molecular mechanisms regulating the SUMO balance is a prerequisite to envisaging the development of novel therapies. In my PhD work, I used a combination of live-cell confocal imaging, protein biochemistry and pharmacological approaches to identify SENP1 regulatory mechanisms at synapses. I provided evidence that synaptic activation increases SENP1 protein levels at synapses. I showed that the increase in synaptic SENP1 upon synaptic activation is a result of two processes: Although (a) fewer SENP1 proteins enter into spines at low diffusion speed (b) a significant proportion of SENP1 becomes immobile and is retained in spines. I demonstrate that the regulatory mechanisms of SENP1 dynamics involve a direct activation of mGlu1/5 receptors. Moreover, I suggest that phosphorylation may play an important regulatory role in SENP1 synapto-dendritic diffusion. Altogether, I propose a novel mechanism driving for the SUMO balance at synapses

In this thesis investigations into chemically modified graphene structures are presented. Chemical functionalization of graphene is the chemical attachment of molecules or atoms to the graphene surface via covalent or Van der Waals bonds, this process offers a unique way to tailor the properties of graphene to make it useful for a wide range of device applications. One type of chemical functionalization presented in this thesis is fluorination of graphene which is the covalent attachment of fluorine to the carbon atoms of graphene and the resultant material is fluorographene which is a wide band-gap semiconductor. For low fluorine coverage the low temperature electron transport is through localized states due to the presence of disorder induced sub-gap states. For high fluorine coverage the electron transport can be explained by a lightly doped semiconductor model where transport is through thermal activation across an energy gap between an impurity and conduction bands. On the other hand, at low temperatures the disorder induced sub-gap density of states dominates the electrical properties, and the conduction takes place via hopping through these localized states. In this thesis it is also shown that electron beam irradiation can be used to tune the coverage of fluorine adatoms and therefore control energy gap between the impurity and conduction bands. Futhermore, electron beam irradiation also offers a valuable way to pattern conductive structures in fluorinated graphene \textit{via} the irradiation-induced dissociation of fluorine from the fluorinated graphene. This technique can be extended to the patterning of semiconducting nano-ribbons in fluorinated graphene where the spatial localization of electrons is just a few nm. The second type of chemical functionalization presented in this thesis is the intercalation of few layer graphene with ferric chloride which greatly enhances the electrical conductivity of few layer graphene materials making them the best known transparent conductors.

Poly(vinyl alcohol) (PVA) is a polymer used in numerous applications, principally those in which its high water solubility is a desirable asset. However there are also areas where PVA is limited by its inherent solubility (for example some specific environments in the biomedical field). This work has sought to overcome such limits by manipulating the surface of PVA in order to propose various means by which the surface solvent resistance might be increased while maintaining the bulk properties of the polymer. Both chemical and physical modifications have been tried and in each case progress has been made towards insolubilizing a single surface of the polymer when in film form. Grafting various species onto the surface of PVA was successfully performed. It is believed that such species bonded to the PVA via attachment to the hydroxyl groups (though this has not been proven conclusively). The data contained herein has led to the conclusion that the primary factor in reducing solubility this way is the removal of the hydroxyl groups, and not the attachment of specifically highly hydrophobic molecules. Introducing permanent cross-links into the surface region has been attempted via various routes. The data recorded shows promise however the system is far from optimised. The biggest challenge remaining is to optimise the depth of material cross-linked. Some steps have been made towards understanding and controlling this parameter though there is much scope for further investigation. The methods used have built on those used for bulk cross-linking and as such are new for the case of surface specific treatment. An interesting phenomenon in some semi-crystalline polymers reported in recent years is that of surface specific crystallization. This effect has been successfully induced and observed in PVA to produce what is believed to be a highly crystalline surface layer, and crystalline regions of PVA are generally accepted to be more water resistant than amorphous ones. In summary, in this work several surface-specific treatments for PVA have been trialled, providing options for post-film forming modification to reduce the surface water sensitivity whilst retaining the bulk properties of the polymer.

The avoidance and response prevention (RP) literatures are reviewed. This review highlighted a number of persistent issues, revealed a need for the development of sensitive fear assessment measures and showed a number of parameters that enhance response prevention's efficacy in reducing conditioned fear. This thesis examined (i) a number of RP parameters across escape-from-fear and passive avoidance baselines, using in the latter situation, multivariate fear assessment measures; and (ii) the psychopharmacological actions of the benzodiazepine, diazepam, and beta-adrenergic blockers, propranolol and atenolol, across passive avoidance, conditioned suppression of licking and signal detection behavioural baselines. In experiment one, escape-from-fear behaviour was found to be insensitive to 100 - and 200 - non-reinforced 5 second CS presentations (RP). Massed RP was reported to be superior to distributed RP and protracted RP (2 hr) more efficacious than 1 hr RP in reducing conditioned fear and hastening avoidance extinction in experiment two. Social facilitation of RP (experiment three) enhanced RP effects when assessed by the time fear assessment measure but had less effect on RP when assessed by the approaches and first entry latency measures. This differential sensitivity of the fear assessment measures was also reported for diazepam, propranolol and atenolol - assisted RP (experiments three and four). Experiment five examined the separate and combined administration of diazepam and propranolol on disinhibiting licking behaviour. Diazepam was more effective than the combined treatment, which in turn was more effective than propranolol alone, with atenolol having little effect in disinhibiting licking behaviour. Experiment six established a signal detection behavioural baseline which was used to independently assess possible diazepam - and propranolol-induced changes in stimulus discrimination from possible drug-induced changes in response-bias (experiment seven). Both diazepam and, propranolol failed to affect response bias, whereas stimulus discrimination was attenuated by propranolol but unaffected by diazepam administration. The response prevention results were discussed in terms of a modified two process theory presented in chapter two, which emphasised the relative sensitivity of and the relationship between the fear assessment measures in discriminating RP effects on conditioned fear and avoidance behaviour. The drug results were discussed in terms of the respective modes of anxiolytic action of the benzodiazepines and beta-adrenergic blockers. This signal detection results were discussed in terms of matching model and signal detection analyses of choice behaviour. Implications of these results to avoidance theory and research as well as to the assessment and treatment of fear motivated human neurotic behaviours were discussed with suggestions for future research being presented.

Stress corrosion cracking of Alloy 600 has lead to the modification and replacement of many nuclear power plant components. Among these components are the Bottom Mounted Nozzles (BMN) of the Reactor Pressure Vessel (RPV). Modifications of these components have been performed on an emergent basis. Since that time, Framatome ANP has developed state-of-the-art modification methods for the repair of BMNs using the Electrical Power Research Institute (EPRI) managed Materials Reliability Program (MRP) attributes for an ideal repair as a basis for evaluation of modification concepts. These attributes were used to evaluate the optimal modification concepts and develop processes and tooling to support future modification activity. This paper details the BMN configurations, modification evaluation criteria, several modification concepts, and the development of the tooling to support the optimal modification scenarios.

A modification of an involute gearing is presented by a modification of an involute and a longitudinal modification along the face width. In the contribution both of the modifications are presented and theoretical and practical methods for determination of their parameters are shown. Experimental methods of setting up of input data for design of modifications are brought in then. These data are prepared on the basis of measuring of deformations of gearwheels bodies, shafts and shafts supports. An attention is given to the influence on a size and position of a zone of contact of meshing teeth. A separate part deals with the appreciation of an influence of modification on a size of a noise and vibrations of involute gears. As far as a loading capacity of modified involute gearings, the attention is first given to the loading capacity in a contact stress between teeth faces. The comparison is made between loading capacity of an involute gearing with no modification and a gearing modified by various types of modifications. This comparison is made both by using a FEM and by experiment. Experiment is based on lifetime test of these gearing.

Modified wood is wood that is improved with special modifiers in order to increase its strength, durability and water-, bio-, fire- and corrosion resistance, which, unlike natural wood, makes it more resistant to various environmental factors. This wood is good to use in construction, and it will be effective for further development, as the buildings will be used longer, and less likely to be subject to repair and reconstruction, which will reduce deforestation. The most popular methods of modification are thermomechanical modification with preliminary steaming of wood, its heating, or with preliminary impregnation with mineral oils; thermochemical modification; chemical modification; radiation-chemical modification. Each type of the above modifications differs in its properties, and also have their own impact on a particular type of wood. The most common and most effective method of modification is achieved by thermomechanical processing. Since it is when the wood is heated, after the introduction of the necessary modifiers into it, the polymers begin to heat up, due to which the vessels of the wood harden, which affects its density. To avoid the destruction of wood, it is very important to monitor the temperature and prevent it from reaching high heating levels.

An analysis of three modifications of the Scarpi method is given in order to assess their applicability to calculating Hadamard matrices of high orders with structural symmetries. Descriptions of modifications are presented, the results of Hadamard matrix calculation are demonstrated, confirming the conclusion about the significance of the Balonin-Seberry modification. The computational experiment shows that there are no results refuting the existence of matrices symmetric structures calculated by the Balonin-Seberry modification.

This Evidence Check review explored factors contributing to effective referral to population based lifestyle modification programs for people with chronic disease. It considered factors which influence practitioner referrals to such programs and factors influencing the likelihood of the patient engaging with and completing a program. The reviewers identified barriers and facilitators to referrals by health professionals and to uptake, engagement and completion of programs by patients. The research suggests that for health professionals, involvement in program design, educational activities and practitioner advocates and receiving feedback on program effectiveness may increase referrals. For patients, factors related to the method of referral and personal, environmental and social factors appear to influence likelihood of engagement with and completion of programs.
