Abstract (sommario):
In this article, we construct the exact dynamical wave solutions to the Date–Jimbo–Kashiwara–Miwa equation with conformable derivative by using an efficient and well-established approach, namely: the two-variable G’/G, 1/G-expansion method. The solutions of the Date–Jimbo–Kashiwara–Miwa equation with conformable derivative play a vital role in many scientific occurrences. The regular dynamical wave solutions of the abovementioned equation imply three different fundamental functions, which are the trigonometric function, the hyperbolic function, and the rational function. These solutions are classified graphically into three categories, such as singular periodic solitary, kink soliton, and anti-kink soliton wave solutions. Furthermore, the effect of the fractional parameter on these solutions is discussed through 2D plots.

Abstract (sommario):
We discuss some new aspects of the theory of the Jimbo–Miwa–Ueno tau function which have come to light within the recent developments in the global asymptotic analysis of the tau functions related to the Painlevé equations. Specifically, we show that up to the total differentials the logarithmic derivatives of the Painlevé tau functions coincide with the corresponding classical action differential. This fact simplifies considerably the evaluation of the constant factors in the asymptotics of tau functions, which has been a long-standing problem of the asymptotic theory of Painlevé equations. Furthermore, we believe that this observation is yet another manifestation of L. D. Faddeev’s emphasis of the key role which the Hamiltonian aspects play in the theory of integrable system.

Abstract (sommario):
ABSTRACT IMPACT: MiaA has a human homolog known as TRIT1. Mutations in TRIT1 have been associated with rare diseases such as MELAS and MERRF syndromes. These diseases are associated with mitochondrial disfunction.Understanding the mechanisms of bacterial sRNAs, and the miRNAs associated with these diseases could potentially afford the insight into effective cures. OBJECTIVES/GOALS: The aim is to investigate the regulation and function of tRNA isopentyladenine transferase enzyme in Escherichia coli. We aimed to execute screens for the identification of small RNA regulators of MiaA. The study will also investigate if i6A tRNA modification is necessary for the expression of major heat shock and mitochondrial proteins. METHODS/STUDY POPULATION: We constructed a chromosomal miaA-lacZ translational fusion driven by the arabinose responsive PBAD promoter and used it to screen against an Escherichia coli small RNA library. Using CsrB, one of our candidate sRNA regulators from our genetic screen, we measured the steady state levels of MiaA by Northern Blot in a PBAD-miaA2(P2HS)-lacZ translational fusion strain whereby pBR-pLac-csrB, pBR-pLac-csrA and the pBR-pLac vector are over-expressed, and under the control of an IPTG inducible promoter. Additionally, and in the same PBAD-miaA2(P2HS)-lacZ translational fusion strain background, we measured the steady state levels of MiaA in the wild type, csrA:zeo mutant strain, and csrA:zeo pBR-pLac-csrA complementation strain to determine if a combination of the pair would restore the wild-type genotype. RESULTS/ANTICIPATED RESULTS: Upon measuring the effect of small RNAs on miaA expression using quantitative b-galactosidase assays, we saw a 5-fold decrease in the expression of MiaA in the miaA-lacZ translational fusion containing sRNA CsrB, suggesting that this sRNA may play a role in the regulation of post-transcriptional expression of MiaA.From our northern blotting analysis, we observed a 6-fold decrease in MiaA expression in the absence of csrA, suggesting that csrA is essential for MiaA expression. DISCUSSION/SIGNIFICANCE OF FINDINGS: Identifying, mapping and characterizing how MiaA is regulated post-transcriptionally will give us an increased understanding in the maintenance and regulation of the normal function of E.coli to conserve homeostasis and translation fidelity.

Abstract (sommario):
NKG2D and its ligands, MICA and MICB, are known as the key regulators of NK cells. NK cells are the first reconstituted cells after the allogeneic hematopoietic stem cell transplantation (HSCT); therefore, it is crucial to understand their role in HSCT outcome. In the presented study, we investigated the single amino acid changes across the exons 2–4 of MICA and MICB genes, and point mutations within the NKG2D gene, which defines the type of NKG2D haploblock (HNK/LNK) in the donors (n = 124), as well as in patients with acute myeloid leukemia (n = 78). In our cohort, we found that graft from a donor with at least one MICA allele containing glycine at position 14 (MICA-14Gly) is significantly associated with deterioration of a patient’s overall survival (OS) (p < 0.05). We also observed a negative effect of MICB-58 (Lys → Glu) polymorphism on relapse-free survival (RFS), although it was not statistically significant in multivariate analysis (p = 0.069). To our knowledge, this is the first work describing the role of MICA-14 and MICB-58 polymorphisms on HSCT outcome.

Abstract (sommario):
A γ-tubulin mutation in Aspergillus nidulans, mipA-D159, causes failure of inactivation of the anaphase-promoting complex/cyclosome (APC/C) in interphase, resulting in failure of cyclin B (CB) accumulation and removal of nuclei from the cell cycle. We have investigated the role of CdhA, the A. nidulans homologue of the APC/C activator protein Cdh1, in γ-tubulin–dependent inactivation of the APC/C. CdhA was not essential, but it targeted CB for destruction in G1, and APC/CCdhA had to be inactivated for the G1–S transition. mipA-D159 altered the localization pattern of CdhA, and deletion of the gene encoding CdhA allowed CB to accumulate in all nuclei in strains carrying mipA-D159. These data indicate that mipA-D159 causes a failure of inactivation of APC/CCdhA at G1–S, perhaps by altering its localization to the spindle pole body, and, thus, that γ-tubulin plays an important role in inactivating APC/CCdhA at this point in the cell cycle.

Abstract (sommario):
Abstract MICA (major histocompatibility complex class I chain-related genes A) is known to be associated with susceptibility to various autoimmune diseases. Recent studies have also demonstrated that MICA*A5.1, one allele of MICA, contributed to the resistance of microorganism infection. The aim of this study was to investigate the association between MICA*A5.1 allele and ankylosing spondylitis (AS). A total of 144 AS patients selected from south China and 110 ethnically matched unrelated healthy controls were typed for MICA*A5.1 polymorphism in addition to HLA-B27 typing by polymerase chain reaction using sequence-specific primers. Statistical analysis revealed: (1) a significantly higher frequency of HLA-B27 in the patient group (96.5%) than in the control group (8.2%) (P < 0.0001); (2) the distribution of MICA*A5.1 genotype was significantly different between the patients and controls (P = 0.029), the frequency of MICA*A5.1 homozygote was significantly decreased in the patient group (2.1%) as compared with the controls (8.9%) (P = 0.023); (3) the association between MICA*A5.1 genotype and AS is not independent of HLA-B27 status (P = 0.94). These results suggest that MICA*A5.1 allele may play a potential role in the protection against AS.

Abstract (sommario):
Abstract The polymorphic products of major histocompatibility complex class I–related chain A (MICA) genes are important in solid organ transplantation rejection. MICA expression is limited to gut epithelium and may play a role in triggering acute graft-versus-host disease (aGVHD). A total of 236 recipients of unrelated donor transplantation were studied. Donor-recipient human leukocyte antigen (HLA) match was 10/10 human leukocyte antigen (HLA-A, -B, -C, -DRB1, -DQB1) in 73% and MICA mismatch in 8.4%. Because of physical vicinity of the loci, MICA mismatch was significantly associated with mismatch at HLA-B and HLA-C. A higher rate of grade II-IV aGVHD was seen in MICA-mismatched patients (80% vs 40%, P = .003) irrespective of degree of HLA matching (HLA 10/10 match: 75% vs 39%, P = .02) and HLA any mismatch (83% vs 46%, P = .003). The rate of grade II-IV gastrointestinal aGVHD was also higher in MICA-mismatched patients (35% vs 17%, P = .05). We conclude that MICA may represent novel a transplantation antigen recognized by human allogeneic T cells. This study was registered at ClinicalTrials.gov (Identifier NCT00506922).

Abstract (sommario):
Inflammatory rheumatic diseases are characterized by inflammation resulting from the immune dysregulation that usually attacks joints, skin and internal organs. Many of them are considered as complex disease that may be predisposed by multiple genes and/or genetic loci, and triggered by environmental factors such as microbiome and cellular stress. The major histocompatibility complex class I chain-related gene A (MICA) is a highly polymorphic gene that encodes protein variants expressed under cellular stress conditions, and these MICA variants play important roles in immune activation and surveillance. Recently, accumulating evidences from both genetic and functional studies have suggested that MICA polymorphisms may be associated with various rheumatic diseases, and the expression of MICA variants may attribute to the altered immune responses in the diseases. The objective of this review is to discuss potential genetic associations and pathological relevance of MICA in inflammatory rheumatic diseases that may help us to understand pathogenesis contributing to the development of these diseases.

Abstract (sommario):
Membrane blebs are specialized cellular protrusions that play diverse roles in processes such as cell division and cell migration. Blebbing can be divided into three distinct phases: bleb nucleation, bleb growth, and bleb retraction. Following nucleation and bleb growth, the actin cortex, comprising actin, cross-linking proteins, and nonmuscle myosin II (MII), begins to reassemble on the membrane. MII then drives the final phase, bleb retraction, which results in reintegration of the bleb into the cellular cortex. There are three MII paralogues with distinct biophysical properties expressed in mammalian cells: MIIA, MIIB, and MIIC. Here we show that MIIA specifically drives bleb retraction during cytokinesis. The motor domain and regulation of the nonhelical tailpiece of MIIA both contribute to its ability to drive bleb retraction. These experiments have also revealed a relationship between faster turnover of MIIA at the cortex and its ability to drive bleb retraction.

Abstract (sommario):
Previous studies on the Ke’eryin pegmatite-type lithium ore field in the Songpan–Ganzi Orogenic Belt have explored the characteristics of the parent rock but have not precisely determined its magma source area. This uncertainty limits our understanding of the regularity of lithium ore formation in this region. In this study, to address the issue of the precise source area of the parent rock of lithium mineralization, a detailed analysis of the Li isotope composition of the ore-forming parent rock (Ke’eryin two-mica monzogranite) and its potential source rocks (Triassic Xikang Group metamorphic rocks) was conducted. The δ7Li values of the Ke’eryin two-mica monzogranite, Xikang Group metasandstone, and Xikang Group mica schist are −3.3–−0.7‰ (average: −1.43‰), +0.1–+6.9‰ (average: +3.83‰), and −9.1–0‰ (average: −5.00‰), respectively. The Li isotopic composition of the Ke’eryin two-mica monzogranite is notably different from the metasandstone and aligns more closely with the mica schist, suggesting that the mica schist is its primary source rock. The heavy Li isotopic composition of the two-mica monzogranite compared to the mica schist may have resulted from the separation of the peritectic garnet into the residual phase during the biotite dehydration melting process. Moreover, the low-temperature weathering of the source rocks may have been the main factor leading to the lighter lithium isotope composition of the Xikang Group mica schist compared to the metasandstone. Further analysis suggests that continental crust weathering and crustal folding and thickening play crucial roles in the enrichment of lithium during multi-cycle orogenies.

Abstract (sommario):
Abstract In a hunt for Th2-bias regulatory loci in mice, we have used interval-specific congenic mapping to positionally clone a gene called Mina, encoding an immunoregulatory member of the JmjC protein family. Th2-bias is a genetic trait defined as the propensity of naïve T helper cells to develop in vitro under neutral conditions into T helper 2 (Th2) effector cells. Th2 cells play a critical role in host protection from multicellular parasites such as intestinal helminths and when disregulated contribute to allergic diseases such as asthma. The Th2-bias trait is known to vary across different inbred mouse strains and correlates inversely with the relative abundance of intracellular Mina protein. Adding further support for a causal link between Mina protein level and Th2-bias, Mina was shown to act as a dose-dependent transcriptional corepressor of the gene encoding interleukin-4 (IL4), a key regulator of Th2 development. We have now identified a SNP that explains differential Mina expression level in high and low Th2-biased strains such as BALB/c and C57BL/6. Most interestingly, this SNP acts by modulating SMAD3 binding to a TGFβ-regulated intronic Mina enhancer. We are currently tying this into its physiological role in pulmonary inflammation and intestinal helminth infection using Mina conditional KO mice, which exhibit dramatic phenotypes in both models.

Abstract (sommario):

Abstract (sommario):
Background: Patients undergoing carotid endarterectomy (CEA) may experiment intraoperative neurologic deficits (IND) during carotid cross-clamping. This work aimed to assess the impact of the Gupta Perioperative Myocardial Infarct or Cardiac Arrest (MICA) risk calculator in the IND. Methods: From January 2012 to April 2021, patients undergoing CEA with regional anaesthesia for carotid stenosis with IND and consecutively control operated patients without IND were selected. A regressive predictive model was created, and a receiver operating characteristic (ROC) curve was applied for comparison. A multivariable dependence analysis was conducted using a classification and regression tree (CRT) algorithm. Results: A total of 97 out of 194 included patients developed IND. Obesity showed aOR = 4.01 (95% CI: 1.66–9.67) and MICA score aOR = 1.21 (1.03–1.43). Higher contralateral stenosis showed aOR = 1.29 (1.08–1.53). The AUROC curve was 0.656. The CRT algorithm differentiated obese patients with a MICA score ≥ 8. Regarding non-obese patients, the model identified the presence of contralateral stenosis ≥ 55% with a MICA ≥ 10. Conclusion: MICA score might play an additional role in stratifying patients for IND in CEA. Obesity was determined as the best discrimination factor, followed by a score ≥ 8. A higher ipsilateral stenosis degree is suggested to have a part in avoiding procedure-related IND. Larger studies might validate the benefit of MICA score regarding the risk of IND.

Abstract (sommario):
Abstract Natural killer (NK) cells play important roles in anti-tumor immunity. Here, we demonstrate that the unfolded protein response (UPR) attenuated the sensitivity of hepatoma cells to NK cell cytotoxic activity by decreasing the expression levels of MHC class I-related chain A/B (MICA/B), CD112 and CD155 in human hepatocellular carcinoma (HCC) cells. The decreased expression levels of MICA/B, CD112 and CD155 were due to the involvement of the activating transcription factor 6 (ATF6) and inositol-requiring enzyme 1α (IRE1α) pathways. In addition, the IRE1α pathway contributed to the increased expression levels of the ER-associated degradation (ERAD)-related molecule HRD1 and facilitated the degradation of MICA/B, CD112 and CD155. Moreover, we found that low levels of MICA/B, CD112 and CD155 expression were significantly associated with poor prognosis in patients with HCC. Thus, our results provide molecular, cellular and clinical evidence demonstrating a novel NK cell-associated immune evasion mechanism, and indicate that targeting this immune evasion pathway may be meaningful in treating patients with HCC.

Abstract (sommario):
Cytokinins (CK) play an important role in the formation of nitrogen-fixing root nodules. It has been known for years that rhizobia secrete CK in the extracellular medium but whether they play a role in nodule formation is not known. We have examined this question using the photosynthetic Bradyrhizobium sp. strain ORS285 which is able to nodulate Aeschynomene afraspera and A. indica using a Nod-dependent or Nod-independent symbiotic process, respectively. CK profiling showed that the most abundant CK secreted by Bradyrhizobium sp. strain ORS285 are the 2MeS (2-methylthiol) derivatives of trans-zeatin and isopentenyladenine. In their pure form, these CK can activate legume CK receptors in vitro, and their exogenous addition induced nodule-like structures on host plants. Deletion of the miaA gene showed that transfer RNA degradation is the source of CK production in Bradyrhizobium sp. strain ORS285. In nodulation studies performed with A. indica and A. afraspera, the miaA mutant had a 1-day delay in nodulation and nitrogen fixation. Moreover, A. indica plants formed considerably smaller but more abundant nodules when inoculated with the miaA mutant. These data show that CK produced by Bradyrhizobium sp. strain ORS285 are not the key signal triggering nodule formation during the Nod-independent symbiosis but they contribute positively to nodule development in Aeschynomene plants.

Abstract (sommario):
This article addresses the issue of the types of poetic figures (shabdalankar) in the poetry of the famous poetess of the medieval socio-religious movement bhakti Mira Bai, namely, an important category of Hindi poetry – alankars, which help to express aesthetic emotion – racu. Therefore, their study is of great importance for characterizing the work of Mira Bai in general. Alankars are usually called rhetorical figures, and their close translation – "decorations" – corresponds to the most important European rhetorical category fully, which covers the concept of figures and paths ornatus. They adorn the sound and meaning of the word and help to show aesthetic emotion – racu. It was suggested to study the types of shabdalankar (form decorations) in the poetry of Mira Bai. It was reviewed such shabdalankars as anupras (alliteration), yamaka (lexical repetition – "consonance"), vacrocti and shlesha (some types of paronomasia – "combination"). It was found that in the verses of Mira Bai there are mainly two of them – anupras and vipsa. A feature of the formal side of Mira Bai's works is miserly use of poetry adornments, since the poetry's verses are a cry of the soul, they reflect the unbearable pain of separation from her beloved Krishna. In such a state of mind, Mira Bai was not up to artificial decoration and play on words. If individual alancars appear in her poetry, it happens completely at ease. Only one of several shabdalankar dominates in the verses of Mira Bai, namely: anuprasa, which helps the poetess through the melody of the verse to create the mood she needs and clearly show the aesthetic emotion of love – the shringar's racu.

Abstract (sommario):
The mass media, and the media in general, play an extremely important role in every society, as well as in people's daily lives. This role was crucial even before the number and type of media, and thus the way they work, were multiplied by digitalization and migration to online digital spaces. Today, we have a situation where a certain number of people prefer to be informed through social rather than traditional media, even about issues that affect life and health. With all this in mind, it is logical to think that the media can act for the purpose of achieving any proclaimed goal, and that this action will be successful, and without significant limits in the possibilities of their action. However, when we talk about creating and promoting a culture of peace, by analyzing the most important elements, such as the ontological and phenomenological nature of peace, and war as the opposite, by analyzing and defining the ethics of peace and war, we conclude that human nature, and beyond, the nature of life on Earth, as well as history and inherited experiences from the past, in juxtaposition with the phenomenological and ontological features of the media, shows significant limitations of the media of mass communication in order to achieve this humane and desirable goal. These limits are deeply rooted in the phenomenon of peace, in the phenomenon of war, in the nature of people and society, as well as in the nature of the mass media. However, what the media can do and are not limited to is the promotion of elements of a culture of peace and non-violence, such as tolerance, dialogue, public discourse that promotes and respects differences, as well as other elements that work to calm tensions and promote a nonviolent approach to conflict prevention and resolution.

Abstract (sommario):
Abstract Natural killer (NK) cells play a central role in the immune surveillance of cancer. NK cells express multiple inhibitory and activating surface receptors, including the activating receptor, NKG2D. Engagement of NKG2D by the stress-induced ligands MICA/MICB stimulates NK cell activation and subsequent target cell lysis. While MICA/MICB expression is restricted in normal tissues, expression is strongly induced upon viral infection, genotoxic stress or malignant transformation. In cancer patients, the tumor microenvironment (TME) facilitates evasion of immune surveillance by NK cells. One mechanism by which tumor cells can escape NKG2D-mediated lysis is via proteolytic release of MICA/MICB from the cell surface due to proteases present in the TME. High concentrations of shed MICA have been observed in serum from patients across multiple tumor types and correlate with poor survival. In mouse models, expression of MICA on tumor cells delayed tumor growth, while the absence of NKG2D accelerated tumor growth. Given the established role of the MICA/MICB-NKG2D axis in the immune surveillance of tumor cells, a therapeutic agent that effectively restores this axis is a compelling approach to harness the immune system for treating cancer. CLN-619 is a humanized anti-MICA/MICB monoclonal IgG1 antibody being developed for the treatment of multiple cancer indications. The antibody was selected based on its high-affinity binding to MICA/MICB, potent in vitro activity and robust anti-tumor activity in xenograft models. MICA/MICB is highly polymorphic, with >150 MICA and 47 MICB alleles in humans, and expression level, binding affinity to NKG2D, and degree of MICA/MICB shedding is thought to be allele-dependent. CLN-619 demonstrated high affinity binding to all common allelic variants of MICA, including the most prevalent MICA allele, MICA*008, and the canonical allelic variant of MICB. CLN-619 was able to prevent proteolytic cleavage of MICA/MICB as evidenced by the reduction of shed MICA/MICB in cell culture supernatants and the corresponding increase in surface MICA/MICB on cancer cells. In addition to the ability to restore the MICA/MICB-NKG2D axis, CLN-619 enhanced the binding of MICA to NKG2D on NK cells. This was enabled by both FcγR engagement on NK cells as well as an intrinsic enhancement of MICA binding to NKG2D. Finally, CLN-619 was shown to elicit potent ADCC of MICA/MICB expressing tumor cells. Consistent with the multimodal mechanisms of action described above, CLN-619 treatment resulted in robust anti-tumor activity at low doses in mice bearing MICA/MICB-expressing human tumor xenografts. Given the pan-cancer expression of MICA/MICB in both solid and hematological malignancies, CLN-619 is expected to have broad anti-tumor activity. CLN-619 is currently being investigated in a Phase 1 clinical trial for the treatment of patients with advanced solid tumors. Citation Format: Kerry A. Whalen, Naveen K. Abstract (sommario):
MICA and MICB are non-classical MHC molecules that indicate cellular stress. They act as ligands for NKG2D receptors found on NK cells, thereby triggering a cytotoxic response against damaged, infected, or transformed cells. The production of soluble forms of MICA/MICB occurs via the cleavage of their extracellular domains (ECDs). The expression of MICA/MICB molecules in tumor sections or the levels of their soluble forms in blood have potential as diagnostic tools for cancer. They can predict important clinical outcomes for cancer patients, such as overall and recurrence-free survival. However, their extensive molecular polymorphism complicates the development of monoclonal antibodies (mAbs) for diagnostic use. Therefore, the diagnostic value of mAb-based assays may vary depending on the frequencies of allelic variants in local human populations. We examined the ECD amino acid sequences of more than 280 MICA and 50 MICB allelic variants. Additionally, we identified 172 and 58 single nucleotide polymorphisms (SNPs) located in the coding regions of the respective genes and resulting in amino acid replacements. The most frequent amino acid replacements (> 10%) in the ECD occur at 11 and 4 sites of MICA and MICB, respectively. We found that the frequencies of SNPs in the identified hot spots strongly correlate with each other in different human populations, despite the diversity of allelic variant frequencies. The functional role of only one site is known. The replacement of valine with methionine at position 152 enhances the affinity of MICA to NKG2D receptor. As the hot spots are dispersed throughout the entire ECD sequences, they may play a role other than modulating affinity with the NKG2D receptor interaction. We recommend that Ag sets used to validate anti-MICA/MICB mAbs meet two criteria. First, they should include both MICA and MICB alleles, as these genes have highly similar sequences. Second, the alleles should cover the variability observed in the identified hot spots.

Abstract (sommario):
Abstract NKG2D ligands (NKG2DLs) mark malignant cells for recognition by natural killer (NK) cells and cytotoxic T lymphocytes via the activating immunoreceptor NKG2D. This led to the hypothesis that NKG2DLs play a critical role in tumor immune surveillance. The human NKG2DLs MICA and MICB are expressed on tumors of epithelial origin in vivo. For the other recently described set of human NKG2DLs, the UL16-binding proteins (ULBPs), expression in vivo is as yet undefined. In this study we investigated expression and function of NKG2DLs in leukemia using a panel of newly generated NKG2DL-specific monoclonal antibodies. We report that leukemia cells from patients variously express MIC and ULBP molecules on the cell surface with MICA most frequently detected. Patient leukemia cells expressing MICA were lysed by NK cells in an NKG2D-dependent fashion. Sera of patients, but not of healthy donors, contained elevated levels of soluble MICA (sMICA). We also detected increased sMICB levels in patient sera using a newly established MICB-specific enzyme-linked immunosorbent assay. Reduction of leukemia MIC surface expression by shedding may impair NKG2D-mediated immune surveillance of leukemias. In addition, determination of sMICA and sMICB levels may be implemented as a prognostic parameter in patients with hematopoietic malignancies.

Abstract (sommario):
Natural killer (NK) cells play a pivotal role in cancer immune surveillance, and activating the receptor/ligand interaction may contribute to control the development and evolution of hepatocellular carcinoma (HCC). We investigated the role of the natural killer group 2 member D (NKG2D) activating receptor and its ligand, the major histocompatibility complex class I chain-related protein A and B (MICA/B) in patients with cirrhosis and HCC subjected to surgical resection, patients with cirrhosis and no HCC, and healthy donors (HD). The NKG2D-mediated function was determined in peripheral blood (PB), in tumor-infiltrating lymphocytes (NK-TIL), and in matched surrounding liver tissue (NK-LIL). A group of patients treated with sorafenib because of clinically advanced HCC was also studied. A humanized anti-MICA/B monoclonal antibody (mAb) was used in in vitro experiments to examine NK cell-mediated antibody-dependent cellular cytotoxicity. Serum concentrations of soluble MICA/B were evaluated by ELISA. IL-15 stimulation increased NKG2D-dependent activity which, however, remained dysfunctional in PB NK cells from HCC patients, in line with the reduced NKG2D expression on NK cells. NK-TIL showed a lower degranulation ability than NK-LIL, which was restored by IL-15 stimulation. Moreover, in vitro IL-15 stimulation enhanced degranulation and interferon-γ production by PB NK from patients at month one of treatment with sorafenib. Anti-MICA/B mAb associated with IL-15 was able to induce PB NK cytotoxicity for primary HCC cells in HD and patients with HCC, who also showed NK-TIL degranulation for autologous primary HCC cells. Our findings highlight the key role of the NKG2D-MICA/B axis in the regulation of NK cell responses in HCC and provide evidence in support of a potentially important role of anti-MICA/B mAb and IL-15 stimulation in HCC immunotherapy.

Abstract (sommario):
AbstractThe kaolinized granites of St. Austell, England, are worked to produce a range of china clay products, for some of which the kaolin has to meet stringent particle shape and size specifications. Systematic petrographic study indicates that kaolin occurs in the form of two textural types: (i) finely crystalline kaolin (typically <5 µm in average diameter), which infills dissolution porosity of granitic feldspars, and (ii) coarsely crystalline vermiform aggregates (up to 100 µm or more in length), which are closely associated with expanded micas. The vermiform aggregates are characterized by an intergrowth of mica and kaolin crystals, which can be observed at scales of resolution offered by TEM. Textural and chemical evidence suggest that the expanded mica texture is probably the result of preferential precipitation of kaolin along mica cleavage planes and is not simply a process of chemical replacement.Petrographic examination of kaolin slurries sampled at different points in a typical refinery circuit indicates that platy products with high aspect ratio are derived exclusively from raw materials rich in vermiform aggregates. The fine scale intergrowth of kaolin and mica within the aggregates results in circumstances where mica persists through to fine grained products. Furthermore, the absence of Fe or other chemical components in the kaolin structure suggests that any iron reported for the final products may be a consequence of the presence of Fe-bearing mica within a very fine grained intergrowth.

Abstract (sommario):
Prokaryotic communities play key roles in biogeochemical transformation and cycling of nutrients in the productive mangrove ecosystem. In this study, the vertical distribution of rhizosphere bacteria was evaluated by profiling the bacterial diversity and community structure in the rhizospheres of four mangrove species (Sonneratia alba, Rhizophora mucronata, Ceriops tagal and Avicennia marina) from Mida Creek and Gazi Bay, Kenya, using DNA-metabarcoding. Alpha diversity was not significantly different between sites, but, significantly higher in the rhizospheres of S. alba and R. mucronata in Gazi Bay than in Mida Creek. Chemical parameters of the mangrove sediments significantly correlated inversely with alpha diversity metrics. The bacterial community structure was significantly differentiated by geographical location, mangrove species and sampling depth, however, differences in mangrove species and sediment chemical parameters explained more the variation in bacterial community structure. Proteobacteria (mainly Deltaproteobacteria and Gammaproteobacteria) was the dominant phylum while the families Desulfobacteraceae, Pirellulaceae and Syntrophobacteraceae were dominant in both study sites and across all mangrove species. Constrained redundancy analysis indicated that calcium, potassium, magnesium, electrical conductivity, pH, nitrogen, sodium, carbon and salinity contributed significantly to the species–environment relationship. Predicted functional profiling using PICRUSt2 revealed that pathways for sulfur and carbon metabolism were significantly enriched in Gazi Bay than Mida Creek. Overall, the results indicate that bacterial community composition and their potential function are influenced by mangrove species and a fluctuating influx of nutrients in the mangrove ecosystems of Gazi Bay and Mida Creek.

Abstract (sommario):
Located in the Dnieper valley and dated to ca. 32,000–31,000 cal BP, the site of Mira is characterized by the high level of conservation of human activity evidence. An important specificity of the site is its position in area without lithic raw material outcrops. The lithic industry of Mira, Layer I with confidence can be regarded as homogenous and containing no admixtures. At the same time, it is multi-component and includes specific Middle and Upper Palaeolithic tool types. Bifacial products constitute the essential archaic feature and are the focus of the presented paper. As a pivotal issue several related tasks are seen: to evaluate whether intensive use and recycling can distort the set of bifacial artefacts; to understand what role (core vs. tool) bifacial artefacts did play in technological chaîne opératoire under the terms of shortage of raw materials; to examine which analogies for bifacial products of Mira, Layer I can be seen, based on their technology and morphology. The analysis of available data allows concluding that the initial set of bifacial products was definitely distorted: some bifaces were used as cores, whilst the others were significantly reshaped and reduced. At the same time, no indication exists that the bifaces served as mobile cores from the onset. Instead, they actually played a role of occasional situational cores. Techno-morphological features of Mira, Layer I bifacial products provide reasons for searching the nearest analogies in local Micoquian-related Middle Palaeolithic records.

Abstract (sommario):
In this paper we interpret the elements of the fantasy and ballad principle in Satirova svirala [La flûte de Pan] (1945) for solo flute written by Petar Konjović (1883-1970) from the point of view of the author's poetics and musical and non-musical analogies with Claude Debussy's (1862-1918) composition Syrinx (1913). Aspects of the fantasy principle are equally manifested when considering Satirova svirala as a unique musical entity that musicalizes the myth of Pan's flute, as well as in the context of its dramaturgical position, as being the central movement (Andante più sostenuto e molto rubato) in the five-movement Concert Suite for Wind Quintet. The variation principle, ballad-like 'narrativity' of the flute part, improvisational character, and rubato spaces of the Svirala's musical flow emanate the Other logic and "free play" inseparable from fantasy, resonating, at the same time, with the mythical narrative.

Abstract (sommario):
Linguistic diversity is present in many audiovisual productions and has given rise to fruitful research on translation of multilingualism and language variation. Monsoon Wedding (Mira Nair, 2001) is a prototypical film for translation analysis, since multilingualism is a recurrent feature, as the film dialogue combines English (L1) with Hindi and Punjabi (L3), which creates an effect of code-switching. This article analyses how the multilingualism and the cultural elements present in the source text (ST) have been transferred to the Spanish translated text (TT) La boda del monzón. The results show that in the Spanish dubbed and subtitled versions, few Indian cultural elements are left, and little language variation is preserved. Thus, L3 does not play a central role as it does in the source text. In the translation, only a few loan words from Hindi or Punjabi are kept, mainly from the domains of food and cooking, as well as terms of address and greetings, or words related to the wedding ceremony. The results also show that when L3 is not fully rendered in translation, otherness is still conveyed through image and music, thus (re)creating a different atmosphere for Spanish audiences.

Abstract (sommario):
Abstract The human MHC class I -related chain gene A (MICA), a highly polymorphic MHC-encoded cell-surface glycoprotein, is a stress-induced and transformation related molecule absent from most normal tissues. Expression of MICA has been reported on a variety of mostly epithelial tumors. MICA can activate Natural Killer (NK) cells via interacting with the immunoreceptor NKG2D. Soluble MICA (sMICA), when shedded from tumor by the action of matrix-metalloproteinases (MMPs), can be detected at high levels in the sera of cancer patients. High levels of sMICA can down-regulate NKG2D expression and lead to functional impairment of NK cells thus providing for a mechanism of tumor escape. Gene array expression profiling of bone marrow biopsies revealed high levels of MMP2 and MMP9 in the bone marrow microenvironment, but not in the purified myeloma cells of the same patients. Since we are developing allogeneic NK cell therapy as a novel treatment for high-risk myeloma we decided to study whether the high activity of MMP2 and MMP9 resulted in 1) elevated sMICA levels in myeloma, and 2) affected NKG2D expression by NK cells. We found by ELISA assays that 28% (13/46) of patients with MM at diagnosis contained elevated serum levels of sMICA (median: 265.2 pg/ml; range: 125.9 – 806.5 pg/ml). sMICA levels were low in 16 control healthy donors tested (median: 0.8 pg/ml; range: 0 – 91 pg/ml). Next, we correlated sMICA levels with indicators of tumor burden and prognosticators of outcome in MM. We observed that sMICA levels in the serum of MM patients was not associated with Ras associated oncogene (RAN) expression, presence of abnormal cytogenetics, elevated CRP, elevated b2-microglobulin or elevated serum M-protein levels. There was also no correlation between MICA gene expression in purified MM cells and increased sMICA levels in the serum. We therefore examined several myeloma cell lines and found that high MICA gene expression does not always correlate with MICA expression at the cell surface as detected by flow cytometry. We hypothesize that the lack of correlation between MICA RNA expression and sMICA may be due to variation in translation of the MICA mRNA or failure to transport the MICA protein to the cell surface. Further, we tested the expression of NKG2D on CD3−/CD56+ cells of 5 MM patients with elevated sMICA serum levels and detected no down-regulation of NKG2D on NK cells compared to controls comprising MM patients with normal sMICA levels (n=3) or normal donors (n=5). Recent studies have suggested that tumor-membrane bound MICA may also play a role in the down-regulation of NKG2D on NK cells. We next incubated normal donor NK cells with a) the high membrane bound MICA expressing MM cell line U266 or b) serum from MM patients containing high MICA levels neither of which resulted in down-regulation of NKG2D. We conclude that despite the high expression of MMP2 and MMP9 both soluble or membrane bound MICA do not down-regulate NKG2D on NK cells and hence do not adversely affect NK cell function in MM. One explanation may be that the sMICA levels we found in myeloma patients were in the order of pg/ml, whilst the levels reported in epithelial tumors are 2–3 logs higher.

Abstract (sommario):
Abstract Large granular lymphocyte leukemia (LGL) is a disease of semiautonomous proliferation of cytotoxic T-cells (CTL) often accompanied by immune cytopenias, particularly neutropenia. LGL related cytopenias have been attributed to LGL cellular cytotoxicity or proapoptotic cytokines rather than intrinsic properties of the neutrophils. The association of LGL with autoimmunity suggests that genetic predisposition may contribute to disease pathogenesis. We studied 69 patients with LGL leukemia using a case-control approach; control populations included ethnically matched healthy individuals (N=82) and disease controls of aplastic anemia (N=48) and kidney transplant recipients (N=48). Initially, we applied the Illumina 12K non-synonymous SNP array to a subcohort of 36 LGL patients and 54 healthy controls (training set). Results were subjected to independent hypothesis-generating biostatistical algorithms. First, Exemplar automated analysis determined disease prediction based on independent χ2 analysis for each SNP. As expected, no SNP in this underpowered study reached Bonferroni corrected statistical significance, but our analysis allowed for ranking based on p-value. Second, Random Forests, a nonparametric tree method was applied, whereby all SNP information was calculated multivariately to predict disease. In a non-Mendelian inherited disease, this method more closely reflects the biology of complex polygenic traits; remarkably, those SNP identified by Random Forest were among the highest ranking SNP by Exemplar. Our initial hypothesis-generating set identified 1 SNP in unknown gene C8orf31 and 4 SNP within the MHC class I related-chain A (MICA) gene. We focused on MICA, a non-peptide presenting, tightly regulated stress response HLA molecule that could play a role in pathogenesis of neutropenia in LGL. To further substantiate our finding, the initial training set results were subjected to technical validation; fidelity was rechecked by PCR genotyping with 93% concordance. Biological validation was determined by confirmation in an independent test set consisting of 33 LGL patients and additional 28 controls. As only limited numbers of SNP were tested, there was no need for α-error adjustment. MICA SNP rs1063635 was found to have the most predictive value in both the training set (PPV=56%, NPV=89%) and test set (PPV=64%, NPV=86%). Overall, the control frequency of this SNP in homozygous form was 12% vs 60% in LGL (p<.01, OR=9.1). MICA alleles have been implicated in autoimmune diseases and malignancies. Although this SNP may not define a particular MICA genotype, it is possible that it is in linkage disequilibrium with genotype-defining polymorphisms. To study the functional consequences of our findings, flow cytometric analysis using anti-MICA antibodies was performed, which identified higher expression of MICA in neutrophils from patients as compared to controls (p=.04). MICA overexpression decreased after immunosuppressive therapy (p<.01). While the mechanism of MICA induction is unknown, we stipulate that the presence of MICA alleles leads to a persistent stimulatory signal in LGL predisposing to clonal outgrowth. In sum, our findings suggest that MICA polymorphisms may represent a predisposition factor in LGL and/or LGL-associated neutropenia.

Abstract (sommario):
The study considers the strength and structural characterization of lateritic soil road base in order to increase the strength of low-volume sealed road construction. Sugar Cane Bagasse Ash (SCBA) and Saw Dust Ash (SDA), mixed with soil and in combination with different percentages of Ordinary Portland Cement (OPC), were utilized in the current study. Structural and mechanical characterization of the investigated samples was performed by X-Ray Fluorescence (XRF), X-Ray Diffraction (XRD), Standard Proctor Test (SPT), Unconfined Compression Strength (UCS) Test, and California Bearing Ratio (CBR) Test. The observed increase in strength may be due to the reduction of mica, quartz, and calcite in the investigated samples. CaO and SiO2 contribute to the development of strength in cement, while SCBA, and SDA-stabilized lateritic soils. The microstructural study revealed that the mica, quartz, and calcite phases play a very important role in maintaining the strength and stability of the investigated samples.

Abstract (sommario):
Vasectomized mice play a key role in the production of transgenic mice. However, vasectomy can cause great physical and psychological suffering to mice. Therefore, there is an urgent need to find a suitable replacement for vasectomized mice in the production of transgenic mice. In this study, we generated C57BL/6J mice (Piwil1 D633A-INS99, Piwil1mt/mt) with a 99-base insertion in the Miwi (Piwil1) gene using CRISPR/Cas9 technology and showed that Piwil1mt/+ heterozygous mice were normally fertile and that homozygous Piwil1mt/mt males were sterile and females were fertile. Transplantation of normal fertilized eggs into wild pseudopregnant females following mating with Piwil1mt/mt males produced no Piwil1mt/mt genotype offspring, and the number of offspring did not differ significantly from that of pseudopregnant mice following mating and breeding with ligated males. The CRISPR‒Cas9 system is available for generating Miwi-modified mice, and provides a powerful resource to replace ligated males in assisted reproduction research.

Abstract (sommario):
Abstract Background: Graft-versus-host disease (GVHD) is a major cause of mortality after unrelated hematopoietic stem cell transplantations (HSCT). Despite the development of modern immunosuppressive strategies, a nearly perfectly controlled compatibility of the classical HLA genes (HLA-A, -B, -C, -DRB1, -DQB1, and -DPB1) and availability of numerous so-called minor histocompatibility antigens (e.g. HY or HA-1), its incidence remains largely unexplained to date. MIC genes (MHC class I chain-related) - a distinct lineage of MHC class I genes – are promising candidates to explain, at least partially, the incidence of GVHD in HLA-matched transplantations. MICA and MICB are highly polymorphic (100 alleles for MICA and 40 for MICB) and encode functional cell-surface glycoproteins up-regulated by cell stress. They interact with NKG2D, an activating receptor expressed on the surface of cytotoxic αβ CD8+ and γδ T lymphocytes and natural killer cells. MIC genes are already known to have a HLA-independent effect on solid graft outcomes and may play a similar role in HSCT by triggering GVHD. Objective: The objective of the present study was to determine the impact of donor/patient matching at the MICA and MICB loci on the incidence of GVHD in patients undergoing unrelated HSCT. Methods: We retrospectively analyzed a multicenter cohort of 1072 unrelated transplantations performed between 1996 and 2013. All donor-recipient pairs were fully typed at high resolution for HLA-A, -B, -C, -DRB1, -DQB1, and -DPB1 and were matched for ten of ten HLA alleles (HLA 10/10 matched). High resolution genotyping of MICA and MICB was performed by sequenced-based typing in order to define matching grades between donors and patients. The endpoints of the study were acute and chronic GVHD. Apart from HLA-DPB1 matching, statistical models were adjusted for major clinical variables which have been shown to be associated with outcome (patient’s age, patient’s and donor’s sex, patient’s and donor’s serological status for cytomegalovirus, year of transplantation, time to transplantation, transplantation center, source of stem cells, conditioning regimen, GVHD prophylaxis, treatment with anti-thymocyte globulin, disease category and severity at transplantation). Results: Of the 1072 transplantations, 134 (12.5 %) and 380 (35.4 %) were mismatched at the MICA and MICB locus, respectively. Both MICA and MICB mismatches were significantly associated with an increased incidence of severe acute GVHD (grades III-IV) in univariate and multivariate models (multivariate model: HR = 2.32, 95 % CI = 1.84-2.92; p=0.0003 for MICA and HR = 1.49, 95 % CI = 1.24-1.79; p=0.03 for MICB). At day 100 post-HSCT severe acute GVHD incidences in mismatched vs. matched transplantations were 19.62 % vs. 15.08 % and 20.00 % vs. 14.84 % for MICA and MICB, respectively (Figure 1). Chronic GVHD was associated with MICA and MICB mismatches in univariate analysis (HR = 1.55, 95 % CI = 1.27-1.89; p=0.029 for MICA and HR=1.38, 95 % CI = 1.19-1.62; p=0.03 for MICB), but showed only a trend for association in multivariate models. Figure 1 Estimated cumulative incidence curves of grades III–IV acute GVHD according to MICA (panel A) and MICB (panel B) matching status. The solid and dashed lines represent MIC matched and mismatched grafts, respectively. The Fine and Gray model was used with relapse and death considered as competing risks. Figure 1. Estimated cumulative incidence curves of grades III–IV acute GVHD according to MICA (panel A) and MICB (panel B) matching status. The solid and dashed lines represent MIC matched and mismatched grafts, respectively. The Fine and Gray model was used with relapse and death considered as competing risks. Conclusion: To date this is the largest reported MICA and MICB sequence analysis whether in HSCT or solid organ transplantation. Inclusion of MICA and MICB typing in the donor selection process may be a practical clinical strategy for lowering the risks of severe acute GVHD after unrelated HSCT. Disclosures No relevant conflicts of interest to declare.
