Abstract The activity of Src-family kinases (SFKs), which phosphorylate immunoreceptor tyrosine-based activation motifs (ITAMs) upon receptor ligation, is a critical factor regulating myeloid cell activation. The SFK Lyn has roles in activating myeloid cells, by phosphorylating ITAMs, and also in inducing suppressive signaling via phosphatase activation. Alternative splicing of lyn mRNA produces two proteins, LynA and LynB, and emerging evidence from our lab and others suggests that LynA and LynB interact with different proteins and preferentially activate different signal cascades. In macrophages LynA is uniquely susceptible to rapid ubiquitin-mediated degradation and functions as a rheostat that regulates ITAM signal strength. Phosphorylation of tyrosine 32 in the unique region of LynA preferentially targets LynA for polyubiquitination by the E3 ligase c-Cbl. In contrast to macrophages, mast cells express little c-Cbl. Upon activation, LynA is not rapidly degraded in mast cells, and SFK-mediated signaling is amplified relative to macrophages. Previous studies of LynA have been hampered by knockout models that lack both LynA and LynB. To understand the role LynA plays in immune development and activation, we used CRISPR-Cas9 gene editing to generate novel mouse strains that express either LynA or LynB. Preliminary studies in these mice have demonstrated novel roles for LynA. Notably, LynA−/− mice have an enrichment in splenic dendritic cells and alveolar macrophages. Thus, LynA is not only an important regulator of myeloid cell activation, tuning cell-specific signaling thresholds, but also helps regulate the function of these cells. Further studies will investigate the impact of LynA deletion in myeloid inflammation sensing.

With the omnipresence of media in the digital age, understanding its impact on individuals' health information perception and behaviors has become paramount. This study presents the meticulous development and rigorous validation of the Media Influence on Health Information Perception and Behavior Scale (MIHIPBS) using the Content Validity Index (CVI) method. The scale development process comprised several critical phases, including item generation, expert panel formation, content validity assessment, and pilot testing. A panel of nine experts, encompassing diverse backgrounds such as psychology, public health, communication studies, and measurement development, rigorously examined each item's relevance to the construct of "Media Influence on Health Information Perception and Behavior." These experts assigned ratings on a 4-point Likert scale, ranging from "Not relevant" (1) to "Highly relevant" (4). To determine content validity, the study adhered to established recommendations for acceptable cut-off scores which were contingent upon the number of experts involved Lynn, Davis, Polit & Beck, and Polit et al.2006. Various validity indices, including the Item-Level Content Validity Index (I-CVI), Scale-Level Content Validity Index based on the Average Method (S-CVI/Ave), and Scale-Level Content Validity Index based on the Universal Agreement Method (S-CVI/UA), were meticulously calculated. The outcomes of the content validity assessment were resoundingly positive. All items within the MIHIPBS attained remarkably high I-CVI scores, significantly exceeding the established acceptable threshold. Moreover, the S-CVI/Ave, calculated as the mean of the I-CVI scores for all items, unequivocally demonstrated robust content validity (Hamie et al. Ozair et al., Lau et al., and Marzuki et al.2018. Of particular significance, the S-CVI/UA, representing the proportion of items achieving the highest relevance rating by all experts, decisively met the prescribed cut-off score. These findings unequivocally affirm the MIHIPBS as an exceptionally valid and comprehensive instrument for capturing the complex dynamics of media's influence on health information perception and behavior. The MIHIPBS emerges as an invaluable and validated tool poised to make substantial contributions to research, practice, and policymaking within the domains of health communication, media literacy, and public health. Its versatility extends to assessing the efficacy of media-based health interventions, evaluating the impact of media campaigns on health decision-making, and informing targeted health communication strategies. Future endeavors should explore opportunities for cross-cultural validation, longitudinal assessments, and adaptations to account for emerging media modalities.

Lydia joined ASIS&T ready to lead after Dick Hill's 27 years of service as executive director. Also announced are the keynote speakers for the 2017 Annual Meeting: Richard Marks, head of PlayStation Magic Lab at Sony Interactive Entertainment, and William Powers, best‐selling author and research scientist at MIT Media Lab. The Annual Meeting will also have an 80th Anniversary Welcome Reception and panel session, and work is underway for an interactive history exhibit at the meeting, coordinated by Kathryn La Barre. Lynn Silipigni Connaway was invited to spend April 26 through May 7 in Hong Kong as a visiting professor working with the master of library and information management program at the University of Hong Kong. Connaway and past president Nadia Caidi also attended the Council for Scientific Society Presidents in Washington, D.C., May 2017, and ASIS&T partnered with the American Association for the Advancement of Science and the Earth Day Network for the March for Science.

Abstract (sommario):

The extremely thermophilic bacterium Thermus thermophilus HB27 synthesizes lysine through α-aminoadipate (AAA). In this study, a T. thermophilus gene encoding the enzyme that catalyses transamination of AAA was cloned as a mammalian kynurenine/AAA aminotransferase (Kat2) gene homologue. A T. thermophilus mutant with disruption of the Kat2 homologue required a longer lag phase for growth and showed slower growth in minimal medium. Furthermore, addition of AAA or lysine shortened the lag phase and improved the growth rate. The Kat2 homologue was therefore termed lysN. LysN recognizes not only 2-oxoadipate, an intermediate of lysine biosynthesis, but also 2-oxoisocaproate, 2-oxoisovalerate and 2-oxo-3-methylvalerate, intermediates of leucine, valine and isoleucine biosyntheses, respectively, along with oxaloacetate, a compound in the TCA cycle, as an amino acceptor. These results suggest multiple roles of LysN in several cellular metabolic pathways including lysine and branched-chain amino acid biosyntheses.

CURE courses were developed for Deaf and Hard-of-Hearing (D/HH) students in the Laboratory Science Technology program (Lynn et al. 2020) at the National Technical Institute for the Deaf of Rochester Institute of Technology. Notably, the developed CUREs enrolled all D/HH students and also fulfilled a requirement for the two-year degree program. Attending to accessibility, the courses were taught in American Sign Language and focused on best practices for working with D/HH students in the lab.

Abstract (sommario):

The Canada Lynx (Lynx canadensis) is the most common and widespread member of the cat family in Canada. Lynx are distributed throughout forested regions of Canada and Alaska and into portions of the northern contiguous United States, closely paralleling the range of its primary prey, the Snowshoe Hare (Lepus americanus). They are most common in the boreal, sub-boreal and western montane forests, preferring older regenerating forests (>20 years) and generally avoiding younger stands, and occupy roughly 95% of their former range in Canada. Lynx population size fluctuates 3–17 fold over an 8–11 year cycle, tracking the abundance of Snowshoe Hares with a 1–2 year lag. During increasing and high hare abundance, lynx have high reproductive output and high kit and adult survival. The decline phase is characterized by reproductive failure, increased natural mortality, and high rates of dispersal. Dispersal distances of over 1000 km have been recorded. During the cyclic low, kit recruitment essentially fails for 2–3 years, and is followed by several years of modest reproductive output. Reproductive parameters in southern lynx populations appear similar to those found during the cyclic low and early increase phase in more northern populations. Trapping is a significant source of mortality in some areas. Field studies have documented from 2–45 lynx/100 km2 at various times in the cycle and in various habitats. Although the amplitude of the cyclic fluctuations in lynx numbers may have decreased somewhat in recent decades, there is no evidence to suggest a significant decline in numbers in Canada. Lynx are managed as a furbearer in Canada, with harvest regulated primarily by seasons, quotas, and closures. The harvest over the past decade has declined concurrent with declining pelt prices, and is currently a fraction of historic levels. Lynx are fully protected in less than 2–3% of their range in Canada. There is no evidence to suggest that overall lynx numbers or distribution across Canada have declined significantly over the past two decades, although loss of habitat through increased urbanization and development and forestry is likely affecting lynx populations along the southern fringe of its range. Its high potential to increase in numbers and propensity to disperse long distances suggest that the species is relatively resilient to localized perturbations and reductions, given time and removal of the factors that cause the initial decrease. Lowered lynx harvests, coupled with a greater awareness of the need for proactive lynx management, suggests that the overall future of lynx in Canada is secure.

We evaluated hypotheses of the dynamics of predators (lynx) relative to prey (snowshoe hares) and predator abundance in the Yukon, Canada. The hypotheses were that predator (lynx) dynamics are influenced by prey density, or by both prey and predator densities. Annual lynx population growth rate (r), estimated from lynx counts, was positively related to previous hare density and negatively related to previous lynx density, as described by the best-fitting additive model (R2 = 0.85). Annual lynx growth rate (r) estimated from lynx tracks was positively related to the ratio of hares per lynx in the best-fitting model (R2 = 0.55). There was most support for the prey- and predator-dependent hypothesis of predator dynamics. Projected lynx tracks showed similar trends to observed abundance but lagged one year, emphasising the need for evaluation of projected predator trends.

Abstract Rheumatoid arthritis (RA) is a chronic, systemic inflammatory disorder characterized by infiltration of innate immune cells into the synovial joints, whose etiology is unclear. While the Fcγ receptor (FcγR) aggregation on innate inflammatory cells by auto-antibodies is a critical stimulus, much of FcγR signaling remains unknown. Previous research from our lab has shown that FcγR-mediated innate immune cell activation is antagonistically controlled by the Src family kinases, Fyn and Lyn. Our current study investigates the Fyn/Lyn dichotomy in controlling the severity of the inflammatory arthritis using K/BxN model or RA. Our preliminary studies show that Lyn deletion exacerbates joint swelling and clinical arthritis scores, while Fyn deletion reduces plasma inflammatory cytokine levels. These data indicate an antagonistic role for Fyn and Lyn kinases in autoimmune arthritis and support further study of the mechanisms by which these Src family kinases regulate inflammation.

Optimised farm crop productivity requires careful management in response to the spatial and temporal variability of yield. Accordingly, combination of crop simulation models and remote sensing data provides a pathway for providing the spatially variable information needed on current crop status and the expected yield. An ensemble Kalman filter (EnKF) data assimilation framework was developed to assimilate plant and soil observations into a prediction model to improve crop development and yield forecasting. Specifically, this study explored the performance of assimilating state observations into the APSIM-Wheat model using a dataset collected during the 2018/19 wheat season at a farm near Cora Lynn in Victoria, Australia. The assimilated state variables include (1) ground-based measurements of Leaf Area Index (LAI), soil moisture throughout the profile, biomass, and soil nitrate-nitrogen; and (2) remotely sensed observations of LAI and surface soil moisture. In a baseline scenario, an unconstrained (open-loop) simulation greatly underestimated the wheat grain with a relative difference (RD) of −38.3%, while the assimilation constrained simulations using ground-based LAI, ground-based biomass, and remotely sensed LAI were all found to improve the RD, reducing it to −32.7%, −9.4%, and −7.6%, respectively. Further improvements in yield estimation were found when: (1) wheat states were assimilated in phenological stages 4 and 5 (end of juvenile to flowering), (2) plot-specific remotely sensed LAI was used instead of the field average, and (3) wheat phenology was constrained by ground observations. Even when using parameters that were not accurately calibrated or measured, the assimilation of LAI and biomass still provided improved yield estimation over that from an open-loop simulation.

By the early 2000s, Eurasian lynx (Lynx lynx) were nearly extirpated in Lithuania. To determine their status, we used snow-track counts in 2006–2012 and volunteer monitoring in 1999–2005 and 2015–2018. Using simple questionnaires, we collected incidental observations from hunters, foresters, and other interested citizens to estimate lynx distribution, abundance and extent of breeding. Citizen scientists provided 206 reports of 278 individual lynx that suggested expanding lynx distributions in central, western, southwestern, and southern parts of Lithuania. A decrease in range fragmentation has also been observed since 2015. Nationwide, the lynx population quadrupled from 2010 to 162 individuals in 2018. The number of breeding individuals reported was just seven in both 2007 and 2008, limited to two forests in the very north and central part of the country. This increased to 46 in 2015–2018. Our results indicate that the lynx population in Lithuania has increased and its conservation status should be reconsidered.

The use of two scales within an environmental impact assessment study of the impacts on the Iberian Lynx (Lynx pardina Linnaeus) is described. The differences in information content and assessment ability are evaluated. One scale is based on the register of observations along the study corridor of the highway (the "object" approach), and the second on a broad regional landscape ecological analysis assessing the entire Lynx population and the viability for survival of the individual populations that are potentially affected by the highway (the "context" approach). Other data quality and type variations and its influence in the evaluation are also analysed.

Abstract (sommario):

The Turkestan lynx (Lynx lynx isabellina Blyth, 1847) is a rare and understudied subspecies of the Eurasian lynx occupying the mountains of Central and South Asia. This elusive felid’s northwestern range includes the Tien Shan and Zhetisu Alatau mountains in the border region of Kazakhstan, China, Kyrgyzstan, and Uzbekistan. As the first step to conserve this vulnerable carnivore, we have conducted the first full-scale research from 2013 until 2022 on its distribution in this region. Using 132 environmental predictors of 359 lynx sightings, we have created species habitat distribution models across the lynx’s northwestern range using machine learning approaches (Maximum Entropy—MaxEnt). Additionally, we created species distribution forecasts based on seven bio-climatic environmental predictors with each three different future global climate model scenarios. To validate these forecasts, we have calculated the changes in the lynx distribution range for the year 2100, making the first species distribution forecast for the Turkestan lynx in the area. Additionally, it provides insight into the possible effects of global climate change on this lynx population. Based on these distribution models, the lynx population in the Northern and Western Tien Shan and Zhetisu Alatau plays a significant role in maintaining the stability of the whole subspecies in its northwestern and global range, while the distribution forecast shows that most lynx distribution ranges will reduce in all future climate scenarios, and we might face the Turkestan lynx’s significant distribution range decline under the ongoing and advancing climate change conditions. For a future (year 2100) warming scenario of 3 deg. C (GCM IPSL), we observe a decrease of 35% in Kazakhstan, 40% in Kyrgyzstan, and 30% in China as the three countries with the highest current predicted distribution range. For a milder temperature increase of 1.5–2 deg. C. (GCM MRI), we observe an increase of 17% Kazakhstan, decrease of 10% in Kyrgyzstan, and 57% in China. For a cooling scenario of approx. 1–1.5 deg. C (GCM MIROC), we observe a decrease of 14% Kazakhstan, increase of 11% in Kyrgyzstan, and a decrease of 13% in China. These modeled declines indicate the necessity to create new and expand the existing protected areas and establish ecological corridors between the countries in Central and South Asia.

Abstract Context The use of baits for reducing the populations of harmful animal species, eradicating invasive species, vaccination, contraception or producing conditioned aversion, is widespread worldwide. However, baiting programs are often not successful enough and affect non-target species, requiring new approaches for baiting methods. Aims The aim of the present study was to evaluate two attractants used in carnivore studies to improve bait intake probability by red foxes and minimise bait intake by non-target species. Methods Non-toxic baits were distributed across 1000 ha, with bait intake monitored by camera traps during 3-week trials. Baits were assigned to two treatments with lures (lynx urine and Fatty Acid Scent – FAS) and one control. Bait intake by red foxes and non-target species was analysed using Generalised Linear Mixed Model (GLMM) and Kaplan–Meier survival analyses. Key results Lynx urine significantly increased the bait intake by red foxes (58.8%) compared with control (5.7%) and FAS (16.7%) treatment. However, FAS did not significantly increase the bait intake by red foxes compared with control. Bait intake by non-target species differed significantly between treatments, with lower intake in lynx urine (23.5%) treatment than control (54.7%), but not regarding FAS (36.7%), and neither between FAS and control. The probability of bait persistence after the 3-week trial period differed significantly among treatments, being lower in lynx urine treatment (0.18) than FAS (0.50) and control (0.43). All baits taken by foxes with lynx urine treatment (58.8%) occurred within the first 10 days, whereas intake by non-target species (23.5%) stopped after Day 7. Conclusions The use of lynx urine lure increased the proportion of baits consumed by red fox and reduced bait intake by non-target species. Implications Lures can serve to optimise bait delivery methods for red foxes in their different applications, such as conditioned aversion studies, vaccination, live trapping or predator control, while minimising risks to non-target species and reducing the costs and application time.
