Abstract (sommario):

Fifteen marine strombidiid species were measured and identified after protargol staining of bottle-cast samples collected during an annual study of the ciliate microzooplankton in the Caribbean Sea. Nine of these are described herein as new species. One new species from the genus Tontonia, T. simplicidens sp. nov., is defined, based on the pattern of the girdle and ventral kinety. Six new species of Strombidium, S. bilobum sp. nov., S. eurystomum sp. nov., S. ioanum sp. nov., S. maedai sp. nov., S. pollostomum sp. nov. and S. sphaericum sp. nov. are defined, based on cell size and shape, the arrangement of the oral ciliature, and the nature of the macronucleus. One new strombidiid genus Cyrtostrombidium gen. nov., is defined by the presence of a unique oral ‘basket’ and the absence of the ventral polykinetidal zone. Two species of Cyrtostrombidium, C. longisomum sp. nov. and C. wailesi sp. nov., are distinguished, based on cell size and macronuclear structure. Assemblages of six previously-described strombidiid species, S. constrictum, S. dalum, S. epidemum, S. inclinatum, S. wulffi, and Laboea strobila are also briefly described.

Abstract (sommario):

This study was carried out in selected poultry farms to determine the prevalence, distribution and antimicrobial resistance patterns in Salmonella serovars in Ogun State, South-western Nigeria. A total of 200 faecal samples were aseptically collected from the four geographical zones of Ogun State, Nigeria. Seventy-eight Salmonella isolates spread across 39 serovars and representing a prevalence of 39% was recovered. Salmonella Urbana (n=7), Salmonella Kingston (n=6) and Salmonella Agama (n=5) serovars were more commonly isolated. Resistance was most common to ciprofloxacin (29.5%; n=23/78). Multi-drug resistance (MDR) was observed in 15.4% (n=12/78) of the isolates spread across 7 serovars: S. Kentucky, S. Telelkebir, S. Virchow, S. Blockley, S. Chomedey, S. Haifa, and S. Isangi. The study showed the diversity of Salmonella serovars and the increasing trend of antimicrobial resistance in poultry farms in Ogun State, Southwestern Nigeria.

This series of poems operates somewhere between the Bronx, Half Way Tree (Kingston), and memory. Indian indentureship in Jamaica is epistemologically eclipsed; queer death is unmemorialized; an opening of sugar packets evokes the violence of empire. These poems reckon with loss—whether through grammar, digitization, or death. Yet there remains an abiding desire to explode the beauty of (extra)ordinary moments and scenes. Diasporic and hyperlocal, these poems entangle language(s), archives, and memory to map constellations of identities formed and complicated by colonization.

Abstract (sommario):

Trickster novels, especially those by Gerald Vizenor and Maxine Hong Kingston, can be used to destabilize and undermine ethnic stereotypes. As many studies show, the trickster him/herself cannot be stable and thus resists the limitations of definition as the embodiment of ambiguity. Both insider and outsider, s/he plays with the whole concept of “sides” so as to erase the distinction between them. The trickster plays the game, including the game of language, in order to break and exploit its rules and thus destabilizes linguistic markers. Kingston and Vizenor use their novels to subvert the rules of the linguistic game and free perception from stereotypic rigidity. Perceptions of race and ethnicity are frequently codified in the form of stereotypes with which we are all familiar. Once established, they, of course, prove remarkably difficult to dismantle however false or misleading they might be with regard to the race or ethnicity in question; and thus they continue to exacerbate the social tensions with which we are equally familiar. Ethnic American literature has frequently addressed this issue; in this essay I intend to look at one narrative strategy which is specifically designed to question, challenge, exploit, and even manipulate perception.

The purpose of this note is to record the availability for research of highly fossiliferous sediment from a late Pleistocene terrestrial deposit in the Caribbean. The Red Hills Road cave in the parish of St. Andrew, northwest of Kingston, Jamaica (for more precise locality information, see Donovan et al., 1995, p. 12-15) is a late Pleistocene, essentially unstratified, cave deposit. The terrestrial fauna from this site includes diverse vertebrates (Savage, 1990; D.A. McFarlane and R.D.E. MacPhee, research in progress) and gastropods, associated with rare arthropods including millipedes (Donovan and Veltkamp, 1994), isopods, ostracodes and claws of non-marine crustaceans (J. S. H. Collins, research in progress).

AbstractElectron impact excitation rates recently calculated by Keenan, McCann and Kingston for transitions in the He-like ions Al XII, Si XIII and S XV are used to derive the electron temperature sensitive emission line ratio G and the density sensitive ratio R in its low density limit (Ro). These ratios are compared with those calculated by other authors and with the values obtained for solar flares using instruments on board the P78-1 and Solar Maximum Mission satellites. In general it is found that our results resolve discrepancies which currently exist between observation and theory, which provides experimental support for the methods and atonic data adopted in the present analysis.

Objective To determine the association of preeclampsia (PE) with the presence of cardiovascular risk (CVR) factors at approximately 11 years postpartum and to assess the longer term effect of PE on childhood development. Study Design A mail-out survey was sent to all women who were previously recruited into the Kingston arm of the Pre-Eclampsia New Emerging Team's prospective cohort (n = 112 PE and n = 150 control). Physical and biochemical CVR markers were compared between the two groups. Physical, social, and cognitive development of the children involved in the pregnancies was evaluated using the Vineland-3 Domain-Level Parent/Caregiver Form. Results Thirteen PE women and 28 control women returned the study questionnaire. Based on the 2017 American Heart Association High Blood Pressure Clinical Practice Guidelines, 9/13 (69.2%) of the PE women, compared with 6/28 (21.4%) of the control women, have hypertension (p < 0.01). The median percentile rank for overall adaptive functioning was 58 (interquartile range [IQR: 21–73]) in the PE children and 81 (IQR: 61–94) in the control children (p < 0.05). Conclusion The development of PE leads to longer term changes in CVR markers and in childhood development at approximately 11 years postpartum. Pregnancy and the postpartum provide an early window of opportunity for early maternal and child screening and intervention for health preservation and disease prevention.

Abstract (sommario):

The Calbuco volcano in southern Chile (41.3° S, 72.6° W) underwent three separate eruptions on 22–23 April 2015. Following the eruptions, distinct layers of enhanced lidar backscatter at 532 nm were observed in the lower stratosphere above Buckland Park, South Australia (34.6° S, 138.5° E), and Kingston, Tasmania (43.0° S, 147.3° E), during a small set of observations in April–May 2015. Using atmospheric trajectory modelling and measurements from the Cloud-Aerosol Lidar with Orthogonal Polarization (CALIOP) space-borne lidar and the Ozone Mapping Profiler Suite (OMPS) instrument on the Suomi National Polar-orbiting Partnership (NPP) satellite, we show that these layers were associated with the Calbuco eruptions. Buckland Park measurements on 30 April and 3 May detected discrete aerosol layers at and slightly above the tropopause, where the relative humidity was well below saturation. Stratospheric aerosol layers likely associated with the eruptions were observed at Kingston on 17 and 22 May in narrow discrete layers accompanied by weaker and more vertically extended backscatter. The measurements on 22 May provided a mean value of the particle linear depolarisation ratio within the main observed volcanic aerosol layer of 18.0 ± 3.0%, which was consistent with contemporaneous CALIOP measurements. The depolarisation measurements indicated that this layer consisted of a filament dominated by ash backscatter residing above a main region having likely more sulfate backscatter. Layer-average optical depths were estimated from the measurements. The mean lidar ratio for the volcanic aerosols on 22 May of 86 ± 37 sr is consistent with but generally higher than the mean for ground-based measurements for other volcanic events. The inferred optical depth for the main volcanic layer on 17 May was consistent with a value obtained from OMPS measurements, but a large difference on 22 May likely reflected the spatial inhomogeneity of the volcanic plume. Short-lived enhancements of backscatter near the tropopause of 17 May likely represented the formation cirrus that was aided by the presence of associated volcanic aerosols. We also provide evidence that gravity waves potentially influenced the layers, particularly in regard to the vertical motion observed in the strong layer on 22 May. Overall, these observations provide additional information on the dispersal and characteristics of the Calbuco aerosol plumes at higher southern latitudes than previously reported for ground-based lidar measurements.

Background While anti-β2 glycoprotein 1 (anti-β2GP1) antibody positivity is included in the diagnostic criteria for antiphospholipid syndrome (APS), the association between anti-β2GP1 and the obstetrical complications of APS has been inconsistently reported and remains unclear. Objective We completed a case–control study nested within the Canadian Ottawa and Kingston (OaK) Birth Cohort to evaluate the association between anti-β2GP1 antibody positivity and placenta-mediated pregnancy complications. Study Design Five hundred cases were randomly selected among pregnant women who experienced any of the following independently adjudicated placenta-mediated pregnancy complications: preeclampsia, placental abruption, late pregnancy loss (≥ 12 weeks' gestation), and birth of a small-for-gestational age (SGA) infant < 10th percentile. Five hundred pregnant women without any placenta-mediated pregnancy complications were selected as controls. Stored blood samples were analyzed for the presence of anti-β2GP1 antibodies by enzyme-linked immunosorbent assay. Results Anti-β2GP1 immunoglobulin G (IgG) and/or immunoglobulin M (IgM) antibodies in titers ≥ 20 G/M units (> 99th percentile) were present in 24 of 497 (4.8%) of controls and 33 of 503 (6.6%) of cases. There was no significant difference between cases and controls for the composite outcome of any placenta-mediated pregnancy complications (odds ratio, 1.38, 95% confidence interval [CI], 0.8–2.37, p = 0.25). Conclusion Our results call into question the association between anti-β2GP1 antibodies and placenta-mediated pregnancy complications, with further research needed.

AbstractThis report of an aircraft crash at a major airport in Kingston, Jamaica examines the response of the local Emergency Medical Services (EMS). Factors that impacted the response are discussed, and the need for more disaster simulation exercises is highlighted. The objective of this case report was to document the response of EMS personnel to the crash of American Airlines Flight 331, and to utilize the information to examine and improve the present protocol.While multiple errors can occur during a mass-casualty event, these can be reduced by frequent simulation exercises during which various personnel practice and learn designated roles. Efficient triage, proper communication, and knowledge of the roles are important in ensuring the best possible outcome. While the triage system and response of the EMS personnel were effective for this magnitude of catastrophe, more work is needed in order to meet predetermined standards. Ways in which this can be overcome include: (1) hosting more disaster simulation exercises; (2) encouraging more involvement with first responders; and (3) strengthening the links in the local EMS system. Hull or Kingston-upon-Hull is a port upon the River Hull. With a population of over 300,000, it is the fourth biggest city in Yorkshire (after Leeds, Sheffield, and Bradford) and the fifteenth biggest in Britain. Yet its name, like those of other English cities (London, Manchester, Leeds, York, Doncaster), has lacked rational explanation until lately. In 2018 the writer proposed that Hull is not (as long asserted) called after the River Hull, supposedly with an obscure pre-English name. The river is instead called after the town, because Hull derives not from some opaque Celtic hydronym but from Old English hula ‘sheds, huts’. Hull is thus a greater namesake of (Much) Hoole ‘shed(s)’ south-west of Preston, Lancashire.1 As for the River Hull, its old name may have been Leven ‘smooth one’, still that of a village near its source. Information on Holocene lacustrine diatom records from Greenland is rare. Here, a complete Holocene diatom succession is described from a sediment core recovered from Basaltsø, an arctic lake on Geographical Society Ø, eastern Greenland. Based on radiocarbon measurements on plant material, the upper 2.42 m of the 9.85 m long sediment sequence represent the Holocene sedimentological record in this area from about 10 000 calibrated (cal.) years BP. The diatom assemblages in Basaltsø are dominated by only a few species. Four stratigraphic diatom zones could be distinguished. During the early Holocene, mainly Staurosira construens (Ehr.) Williams et Round, Nitzschia amphibia Grunow, and Pinnularia mesolepta (Ehr.) W. Smith were the dominant taxa. After this initial stage, between 9600 and 5000 cal. years BP, Staurosirella pinnata (Ehr.) Williams et Round was the most abundant diatom species. From 4500 to 500 cal. years BP, the diatom assemblage was characterized by the co-occurrence of several taxa, including S. construens. S. pinnata, Kolbesia suchlandtii (Hust.) Kingston, and Cyclotella sp. A (rossiitripartitacomensis complex) as the main contributors. Finally, from 500 cal. years BP, the diatom assemblage was clearly dominated by Cyclotella sp. A, Cyclotella pseudostelligera Hustedt, and Sellaphora spp. The Holocene relative frequencies of S. pinnata and S. construens clearly correlate with the total diatom valve concentration, the Betula and Salix pollen percentage in Basaltsø, and the oxygen isotope record from the Renland ice core. This suggests that S. pinnata and S. construens are temperature-sensitive species that responded on the general climatic development in eastern Greenland during the Holocene. The diatom record together with the pollen and geochemical data of Basaltsø indicate to the Holocene climatic optimum on Geographical Society Ø from ca. 9000 to 6500 cal. years BP.Key words: Arctic, Greenland, Holocene, diatoms, paleolimnology, environmental change.

Various occupational groups are often viewed as having a set of distinctive personality characteristics. The purpose of this study was to examine the personality traits of five subject groups: occupational therapy students in first, second, third and fourth years (N = 60) at the School of Rehabilitation Therapy at Queen 's University, and practicing occupational therapy clinicians from the Kingston area (N = 22). The Personality Research Form—E (PRF-E) was used to collect the data. As a combined group, students and clinicians were not distinguishable from the general population in terms of personality. For students notable correlation scores (p .01) between the year of study and the obtained PRF-E scores were observed on two scales: endurance (r = −0.31) and nurturance (r = −0.41). Analysis of variance indicated statistically significant differences (p .01 and p .05) between fourth year students and clinicians on certain personality variables. Fourth year students scored higher on the abasement, affiliation, change, nurturance, play and social recognition scales. Clinicians obtained higher scores for endurance and harm avoidance.

The area of East Anglia in which its traditional dialects are spoken has shrunk significantly over the past few decades and seen a marked decline in the use of traditional features. These include lack of -s marking on third-person singular forms (Kingston, 2000; Potter, 2018), as well as the long-standing distinction between those words descended from Middle English /ɔ:/ and /ɔu/, as in ‘moan’ vs ‘mown’, which failed to become homophonous as part of the Long Mid Mergers (Wells, 1982a). Like other relics, this distinction now only remains in East Anglia's more northern locales (Trudgill & Foxcroft, 1978; Trudgill, 2004; Butcher, 2019). This apparent ‘dialect death’ situation (Trudgill, 1986: 68) is the outcome of continued supralocalisation, a situation in which locally specific linguistic forms lose out to linguistic variants with greater socio-spatial currency, usually as a result of mobility and dialect contact (Britain, 2010). In East Anglia's case, Trudgill (2001) argues that dialect levelling has largely been driven by the impact of London and Home Counties varieties of English. This process is predicted to continue (Trudgill, 1986; Kingston, 2000), though is unlikely to be straightforward. Britain (2011) reports that substantial intra-regional differences are found in relation to route and rate of sound change in East Anglia. This calls for more comprehensive analyses of East Anglian English to better understand the workings of this ‘heterogenous homogenisation’ (Britain, 2011: 57), and how individual case studies relate to the overall shrinkage of the area in which traditional East Anglian dialects are spoken. This paper presents data in this vein from two studies undertaken in Suffolk, southern East Anglia. The first presents data on unstressed vowel tensing, which refers to the alternation between [ə] and [ɪ] in those unstressed syllables where /ɪ/ occurs in other varieties of Southern British English (SBE) e.g. ‘carpet’ [kɑ:pɪt] / [ka:pəʔ]. The second reports data on yod dropping, i.e. the absence of /j/ from sequences of C+/ju/ e.g. ‘huge’ [hju:dʒ] / [hu:dʒ]. Both variables are well known from descriptions of East Anglian dialects but, as we will see later, they appear to behave differently in this study of contemporary East Anglian English.

Abstract The winter operation of an on-stream Stormwater management pond, located in Kingston, Ontario, Canada, was studied with respect to ice cover formation and breakup, density stratification, velocity field under the ice cover, and the risk of sediment washout The pond freezes over in late November, with the ice thickness reaching up to 0.5 m. During the first half of the winter period, the ice thickness was well described by Stefan’s formula with empirical coefficients appropriate for shallow water bodies. The pond was chemically stratified, with total dissolved solids concentrations up to 1200 mg L-1 observed close to the bottom. The meromictic stability of this stratification was calculated as 882 g . cm.cm-2. Measurements of the velocity field under the ice cover indicated a flow pattern comprising a fast flow region, a small dead zone and a large recirculation zone to the north and south of the fast flow trajectory, respectively. Such a pattern agreed well with that simulated by a CFD model (PHOENICS_). During a snowmelt event, the near-bottom velocities reached up to 0.05 m . S-1, but were not sufficient to scour the bottom sediment.

AbstractTheoretical electron temperature sensitive emission line ratios in Si IV involving the 3d2D − 3p2P and 4s2S - 3p2P multiplets at ~1125A and 816A respectively are derived using R-matrix electron excitation rate calculations. A comparison of these with observational data for a solar active region obtained with the Harvard S-055 spectrometer on board Skylab reveals that there is good agreement between theory and observation for the ratio that includes the2D3/2, 5/2 −2p3/2 transition at 1128.3A. This is in contrast to the findings of Keenan, Dufton and Kingston, and provides support for the atomic data adopted in the calculations. However the2D3/2 −2pl/2 line at 1122.5A appears to be severely blended, as it leads to electron temperature estimates that differ significantly from that expected in ionisation equilibrium. The fact that the I(1122.5A)/I(1128.3A) intensity ratios determined from several flare spectra are closer to theory than that for the active region indicates that the blending is probably due to species with relatively low ionisation potentials, such as an Fe III transition at 1122.53A, as suggested previously by Burton and Ridgeley, and Feldman and Doschek. Electron temperatures deduced for a sunspot are much lower than that predicted from ionisation balance calculations, in agreement with earlier results, and imply that a cooling flow may be present.

AbstractKingstonite, ideally Rh3S4, is a new mineral from the Bir Bir river, Yubdo District, Wallaga Province, Ethiopia. It occurs as subhedral, tabular elongate to anhedral inclusions in a Pt-Fe nugget with the associated minerals isoferroplatinum, tetraferroplatinum, a Cu-bearing Pt-Fe alloy, osmium, enriched oxide remnants of osmium, laurite, bowieite, ferrorhodsite and cuprorhodsite. It is opaque with a metallic lustre, has a black streak, is brittle and has a subconchoidal fracture and a good cleavage parallel to [001]. VHN25 is 871–920 kg/mm2. In plane-polarized reflected light, kingstonite is a pale slightly brownish grey colour. It is weakly pleochroic and displays a weak bireflectance. It does not possess internal reflections. The anisotropy is weak to moderate in dull greys and browns. Reflectance data and colour values are tabulated. Average results of twenty electron microprobe analyses on four grains give Rh 46.5, Ir 16.4, Pt 11.2, S 25.6, total 99.7 wt.%. The empirical formula is (Rh2.27Ir0.43Pt0.29)Σ2.99S4.01, based on 7 atoms per formula unit (a.p.f.u.). Kingstonite is monoclinic (C2/m) with a = 10.4616(5), b = 10.7527(5), c = 6.2648(3) Å, β = 109.000(5)°, V = 666.34(1) Å3 (Z = 6). The calculated density is 7.52 g/cm3 (on the basis of the empirical formula and unit-cell parameters refined from powder data). The seven strongest X-ray powder-diffraction lines [d in Å(I) (hkl)] are: 3.156 (100) (310), 3.081 (100) (1̄31), 2.957 (90) (002), 2.234 (60) (202), 1.941 (50) (2̄23), 1.871 (80) (4̄41) and 1.791 (90) (060, 1̄33). The structure of kingstonite was solved and refined to Rp = 3.8%. There are four distinct metal sites with Rh occupancies of 0.64–0.89. Two metal sites are regular RhS6 octahedra that share edges to form a ribbon running parallel to c. The other two metal sites are coordinated by 4 S + 2 Rh and 5 S + 2 Rh and define a puckered Rh6 ring. The ribbons of regular RhS6 octahedra alternate with the columns of Rh6 rings linked by S atoms. S–S bridges also connect the ribbons and columns. As such, the kingstonite structure is essentially that of synthetic Rh3S4. Minor differences in the unit-cell parameters, atom coordinates and displacement parameters of kingstonite and synthetic Rh3S4 arise from the considerable substitution of Ir for Rh. The mineral name honours Gordon Kingston (formerly of Cardiff University) in recognition of his contributions to platinum group element mineralogy and the geology of their mineral deposits.

This paper describes the development of a unique rolling load simulator (ROLLS) for testing bridge superstructure with a footprint up to 4 m ×17 m, and its first application to test a full-scale 1220 mm ×900 mm ×16000 mm B900 prestressed concrete box girder. This facility at Queen’s University in Kingston, Ontario, is the first of its kind in Canada. ROLLS can apply cyclic loading in a controlled laboratory environment, under realistic highway scale ‘rolling wheel loads’, in lieu of the conventional ‘pulsating stationary loads’. It has two half-axles of a large tandem, each comprising a dual 1140 mm diameter air-inflated tires spaced at either 1.2 or 2.4 m. Each half-axle can apply up to 125 kN, representing the heaviest half-axle load of the CL-625 design truck of the Canadian Highway Bridge Design Code (CHBDC). The maximum travel range and speed are 14.9 m and 6 m/s, respectively. A case study involving analysis of a bridge with eight adjacent B900 box girders of 27.6 m span was carried out prior to experimentally testing one of the girders using ROLLS. Load distribution analyses were conducted using both (i) a finite element model of the full bridge under various CL-625 truck loading configurations and (ii) the CHBDC load distribution method, and both agreed well. Scaling analysis of the girder load share was then conducted to account for shortening it to 16 m to fit in the laboratory, resulting in two-115 kN ROLLS design loads, 1.2 m apart. Multiple passes were conducted at various loads of 40%–100% of the design load, at speeds of 1–5 m/s to examine the machine and girder behaviours. It was found that the applied load fluctuates by less than 10% of full capacity and a 0.13 s/cycle time lag occurs. The measured girder deflection and elastic strains were 11%–20% lower than predicted theoretically. With the two half-axles assembly spaced at 1.2 m, the apparatus has the ability to complete three million cycles in approximately 4.5 months if ran continuously at 5 m/s.

Introduction Knowledge about individual and interpersonal correlates of violence in Canadian seniors is limited. This study identifies correlates of current and past violence by intimate partner and family member(s) in community-dwelling Canadian seniors, while accounting for childhood adverse circumstances. Methods We performed logistic regression analysis of baseline data from a longitudinal study of community-dwelling individuals aged 65 to 74 years and living in Kingston (Ontario) and Saint-Hyacinthe (Quebec). Domestic violence was assessed using the Hurt- Insult-Threaten-Scream (HITS) screening tool. Odds ratios (ORs) are reported with 95% confidence intervals (CIs). Results Current violence of a psychological nature was reported by 18% of the sample. Women were at greater risk of current and lifetime violence perpetrated by a family member (current violence: adjusted OR ¼ 1.83; 95% CI: 1.02–3.30) as well as experiencing violence from their intimate partner in their lifetime than were men (adjusted OR ¼ 2.48; 95% CI: 1.40–4.37). Risk factors have accumulated over the life course that were found to be consistently associated with both current and lifetime violence included having witnessed violence at home in childhood (lifetime violence by family member: adjusted OR ¼ 9.46; 95% CI: 5.11–17.52), as well as poor quality of relationships with intimate partners, family and friends. Conclusion Our research documents the ongoing impact of early adversity on subsequent partner and family violence in Canada. Findings identify some preventable factors associated with current psychological violence and past violence among communitydwelling Canadian seniors.

Abstract Introduction Our group and others have reported the murine SHIP1-deficient mouse model of MDS/CMML is characterized by expansion of immunosuppressive, arginase 1 (Arg1)-positive M2-macrophages and myeloid-derived suppressor cells (MDSC) (Rauh et al., Immunity, 2005). Moreover, translational studies confirmed increased Arg1 in bone marrow aspirate cells of subsets of MDS and CMML patients (Rauh et al., Blood Abstract 2010: 1855), although involving impractical Arg1 enzymatic assays and Western blots. Herein, our goals were to 1) confirm this Arg1 immune signature in an independent cohort of patients, using more clinically applicable immunohistochemistry (IHC), 2) determine the associated risk profile with recent prognostic scoring systems, and 3) begin to connect this Arg1 immune signature with recurring MDS/CMML-acquired mutations. Methods With ethics approval, 29 BM biopsies (decalcified, FFPE) and clinical parameters were retrieved from the archives of Kingston General Hospital: 6 controls (4 negative lymphoma staging BM and 2 mild anemia NYD; mean age +/- std = 57 +/- 13 y), 13 MDS (1 RA, 1 RARS, 1 del(5q), 5 RCMD, 3 RAEB-1, 2 RAEB-2; 76 +/- 14 y), and 10 CMML patients (7 CMML-1, 3 CMML-2; 72 +/- 10 y). H&E and anti-human Arg1 IHC (1/2500 dilution, clone HPA003595, Sigma) were conducted under optimized, automated conditions (Ventana). IHC scoring was recorded independently by 2 blinded Hematopathologists. IPSS, IPSS-R (Greenberg), WPSS (Malcovati), CMML PSS (Such) and Mayo CMML (Patnaik) scores were calculated. Floxed TET2 and Vav-Cre mice were obtained from JAX and used according to approved Queen's University Animal Care protocols. TET2 sequences were obtained from genomic DNA using custom AmpliSeq primer pools and the Ion Torrent PGM platform (LifeTech). Linear regression and student's t-tests were conducted with Prism software (GraphPad). Results 1) We demonstrated increased BM biopsy Arg1 IHC expression in CMML (22 +/- 17% Arg1-positive cells; n = 10; p = 0.0093) and low-grade MDS, particularly RCMD (14 +/- 11%; n = 5; p = 0.012) relative to control subjects (0.4 +/- 1%; n = 6) (Figure 1). Significantly increased mean Arg1 expression was not seen in other MDS subtypes (n = 8). These findings were consistent with our previously reported 40-subject (Toronto) BM aspirate Arg1 assay/Western blot cohort (Rauh et al. Blood Abstract 2010:1855), suggesting the reproducibility of these findings and the clinical utility of IHC-based assessment. Parallel Arg1 IHC is underway on the Toronto cohort, to determine correlations with enzymatic assays/Westerns. 2) We previously reported Arg1 over-expression in the Toronto cohort was significantly associated with the lowest IPSS and WPSS MDS clinical risk categories and now extend this to IPSS-R. In our Kingston cohort, only trends to lower MDS risk were observed. In contrast, increased Arg1 expression was not associated with clinical risk (CPSS and Mayo scores) in either CMML cohort. Thus, Arg1 expression associated with neutral risk in CMML and neutral to lower risk in MDS patients. Both the clinical risk profiles and proportions of Arg1 over-expressing MDS/CMML patients were reminiscent of reported TET2 mutation risk profiles/percentages. 3) TET2-deficient mice demonstrated increased monocytes, macrophages and CD11b+Gr1+ splenocytes (immuno-phenotypically consistent with MDSC), phenocopying SHIP1-deficient mice. We are currently determining whether TET2-/- mouse macrophages are similarly M2-skewed. In parallel, were are obtaining TET2 genomic sequences (along with other recurring mutated genes) for our Toronto and Kingston cohorts, in order to determine if the Arg1 immune signature associates with a particular mutation(s). These findings will be discussed. Conclusions 1) Using two independent patient cohorts, we demonstrated significant Arg1 over-expression in CMML and low-grade (RCMD) MDS. Arg1 IHC warrants further investigation as an ancillary diagnostic test. 2) Increased Arg1 expression had neutral prognostic significance in CMML and neutral to low-risk MDS associations. 3) Increased Arg1 expression in MDS/CMML may be driven by mutant TET2, impacting the epigenetics of MDSC and M2-macrophage expansion, controlled by SHIP1 and related signaling networks. Abstract Background: The Oasis program is a model of aging-in-place that targets social connectedness, physical activity and nutritional wellness through member-driven programming. Oasis, first established in an apartment building in Kingston Ontario, has recently been expanded to 6 new communities across Ontario in a participatory action research study. The purpose of this poster is to describe the physical activity patterns of five unique Oasis communities (Original and 4 new) and explore the impact of personal, environmental, and Oasis program characteristics on these patterns. Methods: Participants were recruited from Oasis communities in 3 market-priced apartiments, 1 subsidized apartment, and 1 mobile-home park. Participants wore the ActivPAL3 activity monitor for 7 days. Mobility was measured using the Timed Up and Go. Programming was described by type (e.g. social, exercise, nutrition), frequency, and timing of programming. Results: Participants included 70 older adults aged 79.8(min 62, max 97), community mean age ranged from 66.2 – 83.5 years. TUG score 11.6(SD 4.9) (community range 10.5 to 13.7 s). Average daily step count was 5800(SD 2835) steps, with communities ranging from 4685 to 6472 steps/day. An average of 604(community range 236 – 1056) of steps were taken at a healthy pace(100 steps/min). Only 27% of participants took the recommended 7000 steps/day (with community rates ranging from 9.5% to 37.5%). Conclusions: Older adults within these aging in place communities demonstrated low to moderate levels of physical activity, with activity patterns differing across communities. Impact of community make up and characteristics on activity patterns will be presented.
