Kettin is a large actin-binding protein with immunoglobulin-like (Ig) repeats, which is associated with the thin filaments in arthropod muscles. Here, we report identification and functional characterization of kettin in the nematode Caenorhabditis elegans. We found that one of the monoclonal antibodies that were raised against C. elegans muscle proteins specifically reacts with kettin (Ce-kettin). We determined the entire cDNA sequence of Ce-kettin that encodes a protein of 472 kDa with 31 Ig repeats. Arthropod kettins are splice variants of much larger connectin/titin-related proteins. However, the gene for Ce-kettin is independent of other connectin/titin-related genes. Ce-kettin localizes to the thin filaments near the dense bodies in both striated and nonstriated muscles. The C-terminal four Ig repeats and the adjacent non-Ig region synergistically bind to actin filaments in vitro. RNA interference of Ce-kettin caused weak disorganization of the actin filaments in body wall muscle. This phenotype was suppressed by inhibiting muscle contraction by a myosin mutation, but it was enhanced by tetramisole-induced hypercontraction. Furthermore, Ce-kettin was involved in organizing the cytoplasmic portion of the dense bodies in cooperation with α-actinin. These results suggest that kettin is an important regulator of myofibrillar organization and provides mechanical stability to the myofibrils during contraction.

Kettin is a giant muscle protein originally identified in insect flight muscle Z-discs. Here, we determined the entire nucleotide sequence of Drosophila melanogaster kettin, deduced the amino acid sequence of its protein product (540 kD) along with that of the Caenorhabditis elegans counterpart, and found that the overall primary structure of Kettin has been highly conserved in evolution. The main body of Drosophila Kettin consists of 35 immunoglobulin C2 domains separated by spacers. The central two thirds of spacers are constant in length and share in common two conserved motifs, putative actin binding sites. Neither fibronectin type III nor kinase domains were found. Kettin is present at the Z-disc in several muscle types. Genetic analysis showed that kettin is essential for the formation and maintenance of normal sarcomere structure of muscles and muscle tendons. Accordingly, embryos lacking kettin activity cannot hatch nor can adult flies heterozygous for the kettin mutation fly.

Kettin is a high molecular mass protein of insect muscle that in the sarcomeres binds to actin and α-actinin. To investigate kettin's functional role, we combined immunolabeling experiments with mechanical and biochemical studies on indirect flight muscle (IFM) myofibrils of Drosophila melanogaster. Micrographs of stretched IFM sarcomeres labeled with kettin antibodies revealed staining of the Z-disc periphery. After extraction of the kettin-associated actin, the A-band edges were also stained. In contrast, the staining pattern of projectin, another IFM–I-band protein, was not altered by actin removal. Force measurements were performed on single IFM myofibrils to establish the passive length-tension relationship and record passive stiffness. Stiffness decreased within seconds during gelsolin incubation and to a similar degree upon kettin digestion with μ-calpain. Immunoblotting demonstrated the presence of kettin isoforms in normal Drosophila IFM myofibrils and in myofibrils from an actin-null mutant. Dotblot analysis revealed binding of COOH-terminal kettin domains to myosin. We conclude that kettin is attached not only to actin but also to the end of the thick filament. Kettin along with projectin may constitute the elastic filament system of insect IFM and determine the muscle's high stiffness necessary for stretch activation. Possibly, the two proteins modulate myofibrillar stiffness by expressing different size isoforms.

with the economic development, the three-kettle condensation process is more and more widely applied. This paper introduces three-kettle condensation and its features and advantages. Comparing the changes of steam consumption and solvent residue in rubber before and after the modification of the cis-1,4-polybutadiene rubber condensation system, the obtained result shows that compared with the two-kettle condensation, the three-kettle condensation system can steam by 25%, and also can reduce 50% of the solvent residual liquid in rubber. The results after the process optimization on three-kettle condensation system show that the bottom temperature of No.1 kettle is 85 °C, and the top pressure is 48 kPa; the bottom temperature of No. 2 kettle is 102 °C, and the top pressure is 90 kPa; and the top pressure of No. 3 kettle is 10 kPa. In addition, here points out the differences in optimization, and gives suggestions to the optimization.

Kettil Bruun is best known for the total consumption model introduced in 1975 in the collective work “Alcohol control policy in public health perspective”. There were three tracks in particular that led Bruun's research towards the total consumption model: research on the distribution of power, research on international drug policies, and his contributions to criminology and social policy. Kettil Bruun’s lifework shows that the innovation, intersectionality, and synergy which are now rehearsed in pompous research strategies were a non-question 40 years ago. These aspects were a natural part of the scientific pursuit of answers. Maybe in a much more modest and homemade form, but all the more import in terms of applicability at the general level of principle.

Kettle holes are small water bodies of glacial origin which mostly occur in agricultural landscapes. They provide numerous ecosystem services (ES), but their supply may be negatively affected by agricultural management. We conducted a literature review to identify which ES are supplied by kettle holes and to analyze feedbacks with agricultural management. Taking Germany as a test case, we also analyzed how kettle holes are addressed in policy documents and for which ES they are regulated. This was done to identify the societal value officially associated with kettle holes. The literature review found eight ES attributed to kettle holes, of which hydrological cycle and flood control, chemical condition of freshwaters, nursery populations and habitats and biotic remediation of wastes were addressed most often. In contrast, only the provision of habitat service was addressed in German policy documents related to kettle holes. We identified types of agricultural management that negatively affected the supply of ES by kettle holes, in particular artificial drainage, high levels of pesticide and fertilizer application, and management where tillage and erosion result in elevated sediment inputs. Additionally, climate change may lead to an increased drying up of kettle holes. Based on our finding, we conclude that the intensity of agricultural management around kettle holes threatens the supply of all ES while only the service of providing habitats for biodiversity is addressed in German policy regulations. Further regulation is required to induce agricultural management change towards a conservation of all ES supplied by kettle holes.

Abstract Kettle holes are found in young moraine landscapes and serve as an important habitat for amphibians. The loss of amphibians has been dramatic in recent decades, mainly because of the increase in land use intensity and deterioration of habitats e.g., kettle holes in agricultural landscapes. We monitored amphibian species on three organically managed farms in north-eastern Germany to get an overview of their occurrence and proof of reproduction to develop effective protection strategies. From 2016 to 2020, we investigated 50 kettle holes in cultivated fields. In 2018, we implemented the nature conservation measure ‘cutting back dense wooded belts’ in six of these kettle holes. Here, we focused on seven species considering four highly endangered species. We found six to seven species in up to 17 kettle holes in the 44 kettle holes without the measure ‘cutting back dense wooded belts’. Bombina bombina occurred at the most kettle holes (57%). The number of kettle holes where amphibians reproduced differed strongly. On average, at least one species reproduced at 58% of the kettle holes. Many kettle holes become overgrown with negative effects for amphibians due to the reduction in solar irradiation and higher water consumption. The nature conservation measure increased the number of species on average from two to four and the number of species with reproduction from one to three. It is one of more than 100 measures in the ‘Farming for Biodiversity’ project that farmers can choose to receive a nature conservation certificate, which can be used for marketing purposes.

Abstract Kettle holes are small depressional wetlands and because of the high variability of site factors they are potential hotspots of biodiversity in the monotone arable land. We investigated eight kettle holes and two agrarian reference biotopes for carabid beetles and spiders. The animals were captured with pitfall traps from May to August 2005, along with surveys of the soil and vegetation. We asked whether each kettle hole has specific ecological properties which match with characteristic carabid beetle and spider coenoses and whether they represent isolated biotopes. Differences in the composition of ecological and functional groups of carabid beetles and spiders between the plots were tested with an ANOVA. The impact of the soil variables and vegetation structure on the distribution of species was analyzed with a Redundancy Analysis. The assemblage similarities between the kettle hole plots were calculated by the Wainstein-Index. Ecological groups and habitat preferences of carabid beetles had maximal expressions in seven different kettle holes whereas most of the ecological characteristics of the spiders had maximal expression in only two kettle holes. High assemblage similarity values of carabid beetle coenoses were observed only in a few cases whereas very similar spider coenoses were found between nearly all of the kettle holes. For carabid beetles, kettle holes represent much more isolated habitats than that for spiders. We concluded that kettle holes have specific ecological qualities which match with different ecological properties of carabid beetles and spiders and that isolation effects affect carabid beetles more than spiders.

Hepatic carnitine palmitoyltransferase (CPT) turnover was studied in control and in non-ketotic hyperglycaemic streptozotocin-diabetic rats. The degradation constant (kd) and half-life (t1/2) did not appear to be altered by mild diabetes. The hepatic CPT (micrograms/g of liver) was not increased by the mild, non-ketotic, diabetes. However, the total hepatic CPT (micrograms/liver) was 37% greater in the diabetic animals, owing to the increased liver weight. This resulted from a 40% increase in the synthesis constant (ks). Hepatic CPT activity (total detergent-solubilized) and translation rates were measured in fed, starved (48 h), non-ketotic diabetic, ketotic diabetic and diethylhexyl phthalate (DEHP)-treated rats. CPT activity (m units/mg of mitochondrial protein) was not significantly increased with non-ketotic diabetes (44% increase, but non-significant), but was increased approx. 2-fold with starvation and ketotic diabetes, and 3.5-fold with DEHP treatment. CPT expressed as units/liver was increased non-significantly (23%) in non-ketotic and starved rats, similar to the turnover study, but was significantly increased with ketotic diabetes and with DEHP treatment. mRNA-translation activity for CPT was elevated in all states to a somewhat greater extent than was activity. It was concluded that protein synthesis as a product of increased CPT-mRNA translation activity is a major means of long-term regulation.

An ethylmethane sulfonate (EMS) mutagenesis of Drosophila melanogaster aimed at discovering novel genes essential for neuromuscular development identified six embryonic lethal alleles of one genetic locus on the third chromosome at 62C. Two additional lethal P element insertion lines, l(3)S02001 and l(3)j1D7, failed to complement each other and each of the six EMS alleles. Analysis of genomic sequence bracketing the two insertion sites predicted a protein of 16,215 amino acid residues, encoded by a 70 kb genomic region. This sequence includes the recently characterized kettin, and includes all known partial D-Titin sequences. We call the genetic locus, which encodes both D-Titin and kettin, D-Titin. D-Titin has 53 repeats of the immunoglobulin C2 domain, 6 repeats of the fibronectin type III domain and two large PEVK domains. Kettin appears to be the NH2-terminal one third of D-Titin, presumably expressed via alternative splicing. Phenotype assays on the allelic series of D-Titin mutants demonstrated that D-Titin plays an essential role in muscle development. First, D-Titin has an unsuspected function in myoblast fusion during myogenesis and, second, D-Titin later serves to organize myofilaments into the highly ordered arrays underlying skeletal muscle striation. We propose that D-Titin is instrumental in the development of the two defining features of striated muscle: the formation of multi-nucleate syncitia and the organization of actin-myosin filaments into striated arrays.

Purpose – The purpose of this paper is to establish a technology that is suitable for the production of the 80,000 MTPY series of epoxy resins on an industrial scale. Design/methodology/approach – This paper introduces the synthesis and processing of epoxy resins, including liquid epoxy resins (DGEBA) (E-51 and E-44), brominated bisphenol A-based epoxy resins (EX-23-80A), semisolid epoxy resins based on DGEBA (E-39D) and solid epoxy resins based on DGEBA (E-21). The study analyses the theoretical raw materials of epoxy resins per tonne, and lists an example of a distributed control system (DCS) to explain that the production process of 80,000 MTPY epoxy resins can be automated. Findings – A two-step method was used to produce either E-51 or E-44 sequentially in the pre-reactor, the reactor, the recycling kettle, refining kettle (1), the refining kettle (2) and the desensitizing kettle or the falling-film evaporator. An advancement process was adopted to manufacture EX-23-80A, E-39D and E-21 using E-51 as a raw material in the predominating kettle and the mixing kettle, the adding kettle (1) and the adding kettle (2), separately. All the processes were controlled automatically by DCS to yield the products. Originality/value – The results support the assertion that the technology developed by the authors ' company to produce 80,000 MTPY epoxy resins results in fewer side reactions and higher yield production.

Understanding the ecosystem functions and services of central European kettle holes (small wetlands) requires knowledge about their spatiotemporal dynamics. A lot of existing research has been conducted on the wet–dry cycles of North American potholes, but much less is known about kettle holes. Based on the extreme weather conditions between 2015 and 2020, we aimed to quantify differences among dominant plant communities of kettle holes using unmanned aerial systems. Different dominant plant communities were differently affected by dry and wet intervals with a major increase in terrestrial plants. Multivariate analysis showed strong variability in plant community composition for permanent and semi-permanent kettle holes, where hydrophytes decreased and nitrophilous perennials increased. Although we cannot provide quantitative results in succession over a five-year observation period, we found indications of progressive succession towards irreversible alternative stable states with woody plants for some kettle holes, especially of the “storage type”. Therefore, we assume stronger changes in proportion of wetland types in kettle holes compared to potholes and we expect the proportion of wood-dominated kettle holes to increase in the central European landscape in the future, leading to enhanced homogenization of the landscape accompanied by a loss of ecosystem functions and services.

Namun belum banyak masyarakat yang mengetahui kegiatan komunikasi ini. Sehingga pengabdi memiliki solusi menggunakan kesenian sebagai media penyampaian dan pengantar gagasan yang cukup menarik. Pengabdi melihat dan menyadari kesempatan bahwa Ketjil Bergerak sedang membutuhkan bantuan publikasi melalui video klip lagu baru mereka yang berjudul “Ada Cinta di Desa” untuk program Sekolah Desa. Pembuatan video klip ini juga sekaligus media promosi kepada masyarakat yang belum mengenal Sekolah Desa yang dijalankan oleh komunitas Ketjil Bergerak. Hasilnya, pembuatan video klip dilakukan di Desa Ngrawan, Getasan, Semarang, Jawa Tengah. Pengabdian ini dilakukan pada tanggal 4 – 6 Juni 2021 dengan melibatkan masyarakat setempat sebagai talent. Pemilihan lokasi ini karena Desa Ngrawan adalah salah satu desa wisata di Jawa Tengah dan merupakan desa binaan dari komunitas Ketjil Bergerak. Proses pembuatan video klip ini adalah pra produksi seperti diskusi dengan komunitas Ketjil bergerak, pembagian tugas, penentuan ide konsep video klip, membuat storyboard, dan casting talent. Produksi, yaitu proses pengambilan gambar. Pasca produksi, melakukan proses editing dan revisi, kemudian serah terima video klip kepada Ketjil Bergerak melalui zoom.

The Disability Equipment Assessment Centre, Southampton, funded by the Medical Devices Directorate, Department of Health, which undertakes a rolling programme of equipment evaluation, has recently completed an evaluation which included kettle tippers. All commercially available kettle tippers (n=6) were evaluated by subjects (n=25) with impaired grip or mild tremor, following a standardised test procedure. The mechanical functioning of the tippers was also examined. A higher proportion of subjects experienced difficulty in pouring as the force required to do so was increased. A high proportion of subjects rated all tippers as stable, with the exception of the Sunflower tipper with a kettle filled to minimum capacity. The majority of subjects (88%) found using a kettle in conjunction with the tippers easier than using a kettle on its own, but important individual differences between the tippers are described.

This study aims to determine what factors influence consumer satisfaction in using eucalyptus oil in a Walbarua kettle in Ubung village, Buru Regency. This research is quantitative descriptive. The research is located in the kettle Walbarua in Ubung village. The research object is consumers having bought eucalyptus oil repeatedly in a Walbarua kettle in Ubung Village. The research period is a month from April to May 2020. The data collection techniques in this study are observation and questionnaires. Data analysis in this study using multiple linear regression analysis. The results showed that there was an effect of product quality on consumer satisfaction who had used eucalyptus oil in a Walbarua kettle in Ubung village of 41.9%, and there was an effect of price on consumer satisfaction who had used eucalyptus oil in a Walbarua kettle in Ubung village of 38.1 %. Therefore, the affordable eucalyptus oil price is one of the main factors to attract the attention of consumers for buying eucalyptus oil in Ubung Village.

The formation of kettle peatlands is a typical vertical peatland terrestrialization. There are few studies on the plant composition and dynamics of kettle peatlands, especially the long-term observation in freshwater lake wetlands. In this study, we observed the plant community composition of typical kettle peatlands in Xingkai Lake, Heilongjiang Province and followed strict rules to carry out field investigation, recording and laboratory analysis. This dataset contains the plant community composition, dominant plants, above-ground and belowground biomass, and plant density in the kettle peatlands from 2012 to 2018. Users can search the data by the survey year and plant community composition. On the one hand, the preparing and sharing of the long-term dataset about plant community in kettle peatlands could enrich the peatland plant community dataset. On the other, the data can be used as a support for students and researchers working in physical geography and ecology, and as a reference for ecological restoration and sustainable management of wetlands.

Senyawa antimikroba didefinisikan sebagai senyawa biologis atau kimia yang dapat menghambat pertumbuhan dan aktivitas mikroba. Tujuan dari penelitian ini adalah untuk mengetahui bioaktivitas anti bakteri ekstrak etanol (C2H6O) dari tiga jenis tumbuhan Pulutan (Urena lobata L), Kadamba (M. speciosa) dan Buah Ketitir (Brucea javanica L. Merr). terhadap bakteri Streptococcuss mutans dan Escherichia coli. Metode pengujian antibakteri dilakukan dengan metode difusi agar sumuran dengan modifikasi dengan menggunakan ekstrak etanol. Ekstrak etanol tumbuhan obat pulutan (Urena lobata L), kadamba (Mitragyna spesiosa), dan buah ketitir (Brucea javanica L. Merr) memberikan penghambatan lemah sampai dengan kuat terhadap pertumbuhan bakteri Escherichia coli dan Streptococcuss mutans. Konsentrasi ekstrak etanol 200 (μg/well) memberikan penghambatan dengan klasifikasi kuat.

AbstractThe KetelD dominant female-sterile mutations and their ketelr revertant alleles identify the Ketel gene, which encodes the importin-β (karyopherin-β) homologue of Drosophila melanogaster. Embryogenesis does not commence in the KetelD eggs deposited by the KetelD/+ females due to failure of cleavage nuclei formation. When injected into wild-type cleavage embryos, cytoplasm of the KetelD eggs does not inhibit nuclear protein import but prevents cleavage nuclei formation following mitosis. The Ketel+ transgenes slightly reduce effects of the KetelD mutations. The paternally derived KetelD alleles act as recessive zygotic lethal mutations: the KetelD/- hemizygotes, like the ketelr/ketelr and the ketelr/- zygotes, perish during second larval instar. The Ketel maternal dowry supports their short life. The KetelD-related defects originate most likely following association of the KetelD-encoded mutant molecules with a maternally provided partner. As in the KetelD eggs, embryogenesis does not commence in eggs of germline chimeras with ketelr/- germline cells and normal soma, underlining the dominant-negative nature of the KetelD mutations. The ketelr homozygous clones are fully viable in the follicle epithelium in wings and tergites. The Ketel gene is not expressed in most larval tissues, as revealed by the expression pattern of a Ketel promoter-lacZ reporter gene.

Abstract Ketosis is a major metabolic disorder of high-yielding dairy cows during the transition period. Although metabolic adaptations of the adipose tissue are critical for a successful transition, beyond lipolysis, alterations within adipose tissue during ketosis are not well known. The objective of this study was to investigate the adipose tissue proteome of healthy or ketotic postpartum cows to gain insights into biological adaptations that may contribute to disease outcomes. Adipose tissue biopsy was collected on 5 healthy and 5 ketotic cows at 17 (±4) d postpartum and ketosis was defined according to the clinical symptoms and serum β-hydroxybutyrate concentration. Morphology micrographs stained by hematoxylin–eosin showed that adipocytes were smaller in ketotic cows than in healthy cows. The isobaric tag for relative and absolute quantification was applied to quantitatively identify differentially expressed proteins (DEP) in the adipose tissue. We identified a total of 924 proteins, 81 of which were differentially expressed between ketotic and healthy cows (P < 0.05 and fold changes >1.5 or <0.67). These DEP included enzymes and proteins associated with various carbohydrate, lipid, and amino acid metabolism processes. The top pathways differing between ketosis and control cows were glycolysis/gluconeogenesis, glucagon signaling pathway, cysteine and methionine metabolism, biosynthesis of amino acids, and the cGMP–PKG signaling pathway. The identified DEP were further validated by western blot and co-immunoprecipitation assay. Key enzymes associated with carbohydrate metabolism such as pyruvate kinase 2, pyruvate dehydrogenase E1 component subunit α), lactate dehydrogenase A , phosphoglucomutase 1, and 6-phosphofructokinase 1 were upregulated in ketotic cows. The expression and phosphorylation state of critical regulators of lipolysis such as perilipin-1 and hormone-sensitive lipase were also upregulated in ketotic cows. Furthermore, key proteins involved in maintaining innate immune response such as lipopolysaccharide binding protein and regakine-1 were downregulated in ketotic cows. Overall, data indicate that ketotic cows during the transition period have altered carbohydrate, lipid metabolism, and impaired immune function in the adipose tissue. This proteomics analysis in adipose tissue of ketotic cows identified several pathways and proteins that are components of the adaptation to ketosis.

Under present-day conditions of market economy, the requirements to the quality and quantity of livestock products increase significantly, and this leads to significant intensification of production and, as a consequence, to the development of metabolic disorders including ketosis in highly productive cows. The research goal was to study protein and mineral metabolism in ketotic cows before and after calving. The studies were conducted on the Training Farm «Prigorodnoye» in the City of Barnaul in autumn and winter in comparable Black-Pied cows. Two groups of cows were formed: the trial group consisted of ketotic cows; the control group consisted of apparently healthy cows. These cows underwent clinical and biochemical examination. The biochemical study of blood determined the following: total protein, albumins, alpha-globulins, beta-globulins, gamma-globulins, total calcium, inorganic phosphorus and ketone bodies. The biochemical blood tests were run 4 times: 2 months and 1 month before calving, and in 10 days and in 1 month after calving. It was found that the change of protein metabolism indices in ketotic cows showed a profound disorder of the protein synthesis function of liver both before and after calving. The level of total calcium in the blood of ketotic cows was characterized by lower values as compared to that in healthy comparable cows. At the same time, the dynamics of inorganic phosphorus concentration change did not have significant differences between ketotic and healthy cows. Total calcium content in ketotic cows increased one month before calving while in apparently healthy cows this index increased after calving only. The concentration of ketone bodies in the blood of ketotic cows was significantly higher than that of apparently healthy cows throughout the study.

The intensification of agriculture over the last few decades has caused habitat loss, which poses a significant threat to the survival of populations and species. Where habitats are connected, populations may escape the destruction of their habitat by migrating to another one. Consequently, the functional connectivity of landscapes has become an important focus for species conservation. Kettle holes are hotspots of biodiversity that provide suitable conditions for wildlife species (i.e., amphibians, insects, aquatic plants) and contribute to landscape heterogeneity. They are also considered to function as stepping stone habitats that contribute to habitat connectivity. This study assesses the contribution of kettle holes for (i) habitat provision and (ii) the functional connectivity of three amphibian species with different movement ranges, and (iii) the study identifies areas where the creation of stepping stone biotopes could improve functional connectivity. The contribution of kettle holes was assessed using GIS-based clustering within three research areas in Germany. It was found that the importance of kettle holes for providing amphibian habitats in the three studied areas was equal to or higher than that of other wetland habitats. The state of functional connectivity and the contribution of kettle holes differed strongly depending on the species’ range. For the short-range species, landscapes were highly fragmented, and the contribution of kettle holes was much smaller than that of corridor habitats. For the long-range species, all habitats suited for amphibian reproduction were connected, and the contribution of kettle holes was similar to that of corridor habitats. However, the contribution of both was mostly redundant. Overall, the results showed that kettle holes play a crucial role in habitat provision and function as important stepping stone biotopes in agricultural landscapes. The clustering method applied in this study provides a simple tool for landscape planning and environmental protection agencies, which can be easily adapted to analyze functional connectivity and habitat interactions for different species or landscapes.
