Copper is an essential macronutrient for living organisms. Nevertheless, at high concentrations, it is toxic to most forms of life, including microorganisms. In this research, we examined the biofilm formation ability and identified the molecular characteristics of copper-resistant bacteria isolated from PT Freeport Indonesia’s tailings. Four bacteria isolates from PT Freeport Indonesia’s tailings were used in this study. Qualitative analysis of biofilm formation by copper-resistant bacteria was performed using the Scanning Electron Microscopy (SEM) method and Microtiter Plate Biofilm Assay. The results showed that the C53 isolate could be categorized as a strong biofilm former, and three other isolates (C38, C40, and C43) as medium biofilm formers. The identity of the selected isolates was based on 16S rRNA gene sequence analysis: C38 isolate had a 99% similarity to Bacillus cereus strain HM85, C43 isolate had a 99% similarity to Bacillus subtilis strain EN16, C40 isolate had a 99% similarity to Lycinibacillus fusiformis strain MB52, and C53 isolate had a 98% similarity to Pseudomonas aeuruginosa strain GGRJ21. The capability of the C53 isolate to form strong biofilm can be exploited in bioremediation processes aiming to remove copper from tailings.

Abstract The search for antileishmanial plants used in traditional medicine led to the choice of CH2Cl2–MeOH (1:1) crude extract of the fruits and stem bark of Pentadesma butyracea Sabine (Clusiaceae) which displayed good activity in vitro against Leishmania donovani 1S (MHOM/SD/62/1S) promastigotes during preliminary screening with IC50 values 5.96 and 26.43 μg mL−1, respectively. The fractionation of both extracts using flash chromatography yielded active fractions with IC50 values ranging from 2.71 to 18.88 μg mL−1. Fourteen compounds (1–14) were isolated from the obtained fractions using successive column chromatographies and their structures were elucidated based on the analysis of their NMR and MS data. Daphnifolin (1), epicathechin (3), α-mangostin (9) and 9-hydroxycalabaxanthone (14) exhibited potent antileismanial activity against L. donovani 1S (MHOM/SD/62/1S) promastigotes with IC50 values of 2.01, 9.09, 3.37, and 6.87 μg mL−1, respectively and good selectivity towards Raw 264.7 macrophage cells (SI > 2.4). Extracts, fractions and some isolates were also assessed in vitro for their antibacterial activity against six bacterial strains [Salmonella typhi (CPC), Enterobacter cloacae (CPC), Pseudomonas aeruginosa HM801, Staphylococcus aureus ATCC 43300, Streptococcus pneumoniae ATCC 491619, Escherichia coli ATCC 25322] using serial microdilution method. Among the tested samples, the stem bark extract of P. butyracea as well as compounds 2 and 8 showed good to moderate activity against the aforementioned bacterial strains with MIC ≤ 250 μg mL−1.

Mobile augmented reality has become an influential tool for digital content representation and visualization of media content in terms of enhancing users' experience and improving the adaptability and usability of typical augmented reality applications, such as in e-commerce shopping, virtual museum, or digital heritage scenarios. This research proposes a new Service Oriented Mobile AR Architecture called SOMARA, which includes a novel mobile AR client application. SOMARA takes advantage the ability to integrate third party content through service orientation. The SOMARA architecture enhances traditional standalone mobile AR applications with embedded media content by uniquely integrating a web service framework into an augmented reality client application to create more efficient and flexible mobile augmented reality applications that efficiently supports novel media content acquisition and visualization through appropriate access parameters. The proposed architecture requires access to media content through specific media content service providers, e.g. a museum commissioning an augmented reality based museum interactive — predetermined media content, or any third party with their own service APIs, e.g. the Victoria and Albert Museum API — related external media content. This approach allows relevant third party media content to be ‘mashed' via their public API with museums' augmented reality interactive's ‘embedded' media content in the SOMARA mobile AR client. In this way novel mobile AR interactive applications, such as a museum augmented reality interactive, can be created based on particular museum environment scenarios that integrate a museum visitor's experience with the interactive's cultural objects. Such experiences based on a SOMARA type museum augmented reality interactive can also be saved allowing visitors to take home their museum experience. SOMARA thus allows museum interactive experiences based on visualization of museums and third party media content physically located in the museum to be migrated to the visitor's home environment for further study, enjoyment and understanding. This unique feature, ability to effectively replay the experience at home, of the proposed system utilizes service-orientation to integrate third party media content, which is currently deficient from commercial augmented reality solutions.

This research examines the social use of the Internet at the level of specified online users in conservative Arab societies, who are called in this thesis online lead users. These specified lead users are the ones who are influential in shaping the innovation processes due to their leading role of practice in finding solutions to their needs and problems they have experienced before the emergence of online social networks such as the lack of public sphere and independent civic organizations etc., which led them to utilize the Internet as their new public space to pursue their goals through their online initiatives and collective actions to apply the social change they seek for their societies under the existence of society social norms and censorship. The research has been carried out as mixed-method study, the gathered data done through means of quantitative and qualitative methods, and the chosen country to conduct the research were Saudi Arabia. A survey questionnaire link were distributed among the targeted online lead users in the country under study, and the author chose three cases form the country under study for the qualitative part. The research sought to examine to what extent society social norms and censorship influence online lead user's expression and behaviours, and their effect on the formulation of their online activities and collective actions, and analyzing the characteristics of online lead user's. The significance of this study lies in the fact that it contribute to the gap in the literature on the use of Internet in conservative Arab societies by providing insight into the roles that social norms play in influencing online behaviour, particularly in communities that are seeking to discuss social issues or mobilize collective action, and analysing online lead users characteristic to distinguish between conservative and cosmopolitan ones using the Right-Wing authoritarian measurement.

This thesis presents my original contribution to knowledge, a combination of critical media and performance theories to analyse the production and augmentation of postdigital experiential spaces in Punchdrunk Theatre Company. Distributed agency is key to Punchdrunk's work, with makers within the company and audiences both being active participants in meaning-making, across complex and detailed interfaces. In order to investigate the making cultures on ‘both sides' of the interface, I undertook a two-year participant study as a researching designer within the company during the build of the productions The House Where Winter Lives and The Drowned Man in 2011-2014, gathering field data in the form of extensive interviews with members of the company and audience participants, supported by diary notations and photographs. I studied the processes and methods that extend, distribute and regulate agency to both audiences and makers within the company, and identified devices and features of the interaction design of the company that produce the immanent subject-event relationships that support immersion in their work. A core aspect of this research concerns the relationship between immersion and the sublime, and how subject-event relationships (immanent vs. transcendent) contribute to engendering sublime interactive experiences. I have analysed the consequences of this for the modelling of participation in interaction design, and how it influences conditions of possibility within interactive systems across physical, digital and blended media. The conclusion of this research includes the definition of a postdigital sublime, and proposes a delinquent system aesthetic that integrates proxies for gravity through articulation of the ‘shadow side' of interaction design.

This thesis is a study of inquiry. Drawing upon the work of American pragmatist John Dewey, this work seeks to contribute to our understanding of the construction of knowledge within the social system of communities of inquiry. The process of inquiry that is traced in this work is one effected in the course of architectural designing. An ethnographically informed study of an architectural masterplan project is used to illustrate Dewey's ideas and how they are played out in design practice. This thesis is understood to correspond with the growing interest of the students of organisational learning and knowledge management in knowledge creation and the underlying social processes. It is further seen as a response to the claim that the key processes of knowledge creation remain largely an enigma. Agreement has been established with Dewey that knowledge is not just an end in itself but a form of action, a medium of change and social transformation. The formation of knowledge, however, within the operation of inquiry is not a matter that “naturally” runs its course. The process of inquiry as studied both in theory and practice has shown just how much its results, which by definition constitute knowledge, are shaped by the institution and control of a problem. A problem is a social construct and the product of the purposeful selection and arrangement of pieces of information. Inquiry is therefore considered a process of controlled knowledge formation. That which counts as knowledge in the realm of social phenomena has been shown to be a matter not so much of agreement between actions and their consequences but agreement in terms of intellectual acceptance. What “satisfies” as a solution (such as the final masterplan) has therefore been shown to be not necessarily a question of its logical status.

Through the evolution of digital media technology, social networks and more recently Web 3.0 (e.g. Cloud-based) technologies, culture and memory is being transformed, both in relation to how memories are represented, and how they may be engaged with or re-accessed. As digital technology alters ways in which knowledge is produced, stored, connected and shared, new terrains, tools and artefacts are formed; new cultural practices alter the ways in which we remember and the ways in which memory is processed, destabilising traditional “historically encoded social habits: religion, authority, morality, traditional values, or political ideology” (Diamantaki 2013). This doctoral project consists of two parts exploring questions of memory in contemporary time. The practice work submitted develops various imaginaries and investigates how to enable mnemonic practices so that works function as memory palaces where bodies and ‘collective' and ‘networked memories' (Hoskins, 2010) can be realised. The work, briefly summarised, includes communal activities in public spaces (a series of workshops and heritage day events, Rendezvous, centrally social activities organised between Fabrica and various charitable organisations in Brighton). It includes a series of installation works, as a transitional process of memory between body, object, an investigation of ubiquitous technology, are investigated – iremembr (2009-15); Rendezvous (2010-15); Untitled#21 (2012). And it leads to the development of an installation piece, 200.104.200.2 (2013-15), that seeks to offer or extend the possibilities of the act of remembering, of memory, as a post-Internet experience; a complex temporal, social, spatial and material, overlapping and merging human and silicon memory. In this, the written component of the combined and larger project, questions concerning memory and digital technology, and how to explore them, are taken up in theoretical terms, and the works I have produced returned to and explored in these contexts. A central project here has been to locate new forms of qualities of ‘digital' memory in a memory map or topology that builds on adapts, and develops other models. Aspects of zones of memory are explored centrally in each of the later thesis chapters each of which also takes up a particular aspect of my practice. The intention – and the contribution to the development of critical thinking around the digital – particularly critical thinking that comes through digital media art practice, is to question how digital technology intervenes in the process of memory; how the concept of digital memory is being thought about; leading me to investigate what does this new digital terrain do as it overlaps and re-writes to some extent the older ones? How does it change ‘how memory happens'.

In the participatory era of new media, a multidisciplinary approach to new media user agency is expected since the increasing diffusion of new media use in various social fields has generated multifarious user roles and activities. This new approach is also expected to overcome the insufficiencies of methodological individualism and wholism which have long dominated the research discipline of new media user agency and activity. This thesis applies the insights of practice theories to the study of YouTube user agency through an 18 months' participant observation study within the Australian context (23 semi-structured interviews and a self-reflexive participant observation through uploading YouTube videos in person). Unlike individualistic and wholistic approaches to human agency, practice theories insist that the social and collective organization of practices is the everyday pivot between structure and agency. Based on this rationale, I develop a new practice approach in my thesis which understands YouTube users as social actors who carry out YouTubing practice in social life. I suggest that YouTube user agency is decided neither by individualistic parameters nor by structural forces but by the organization of YouTubing practice. I examine three issues in my thesis which are concerned with YouTube user agency from a practice perspective. Firstly, I investigate people's motivations for uploading and sharing videos on YouTube through examining the process through which my respondents participate in YouTubing practice. The result reveal how the ways in which YouTubing practice relate to other social practices in my respondents' world influence their YouTubing participation. Secondly, I find that YouTubing practice is organized by two key constitutive elements: a sense of audience and individuals' beliefs concerned with a sense of audience. I then demonstrate how these two key constitutive elements are developed by the joint forces of my respondents' intelligibilities and the elements involved in YouTubing practice. These findings help us understand how YouTube user agency is rooted in the organization of YouTubing practice. This thesis lastly identifies four types of YouTubing performances which represent my respondents' differentiated participation levels and engagement.

This narrative-based qualitative research investigated the distinct journeys of eight cross-cultural artists (four artists from South Korea and four artists from China). Utilizing a variety of theoretical frameworks surrounding cross-cultural research, this dissertation examined current discussions on cross-cultural challenges and their implications in the field of art education. Methods of data collection focused primarily on interviews and were examined through the lens of Bandura’s (1997) self-efficacy theory. Evaluating the lived experiences of artists illuminated nuances in cross-cultural environments, specifically, how socio-cultural transitions influenced their artwork and professional lives. The findings of this research correlate with previous literature surrounding current challenges in the lives of cross-cultural students. These challenges were discussed in the context of how art educators can best confront issues that emerge in the classroom. The analysis and discussion presented in this thesis seeks to provide insights into the experiences of cross-cultural artists, while highlighting the educational implications for both artists and educators.

Both the user of the system and the dialog controller communicate using language. The user’s language consists of strings of English words ordered according to the rules of English syntax. The dialog controller’s language is made from strings of ground logic terms, variables, and punctuation symbols all connected according to the syntax of Prolog expressions. This chapter describes the design of a parser whose task is to translate strings from the user’s language into strings with approximately the same meaning in the dialog controller’s language. Some difficulties encountered by the parser follow. 1. Because of the less than flawless performance of speech recognizers, the parser will not know exactly what the user has said. Instead, the parser will receive as input one or more estimates of what was spoken, none of which may be absolutely correct. 2. Even what was spoken might not be what the user was thinking. The user may have mispronounced part of the utterance, or may make simple grammatical errors in the utterance. 3. The user may deliberately omit small structure words, such as “the” and “of”, from what is said. This is a natural and subconscious response of native speakers when speaking to a machine which has less than perfect language skills. 4. The parser’s grammar of the English language is probably not identical to the user’s. Thus, even without recognition errors, mispronunciations, or omitted words;, the input to the parser may not be syntactically well-formed. 5. The mapping from the user’s language to the dialog controller’s language is not one-to-one. Many inputs will have multiple meanings, and will thus need to be translated into two or more outputs. Conversely, a particular output may result from several syntactically unsimilar inputs. Together with [EM81], [HHCT86], [HM81], and [YHW+90], we argue that due to the above problems, the traditional natural language parsing techniques that accept written or typed input are not adequate for systems which accept speech input. New parsing architectures are required to deal with the high level of uncertainty that is inherent to speech input.
