An extremely thermophilic bacterium, Thermus thermophilus, possesses two glutamate dehydrogenase (GDH) genes, gdhA and gdhB, putatively forming an operon on the genome. To elucidate the functions of these genes, the gene products were purified and characterized. GdhA showed no GDH activity, while GdhB showed GDH activity for reductive amination 1.3-fold higher than that for oxidative deamination. When GdhA was co-expressed with His-tag-fused GdhB, GdhA was co-purified with His-tagged GdhB. Compared with GdhB alone, co-purified GdhA–GdhB had decreased reductive amination activity and increased oxidative deamination activity, resulting in a 3.1-fold preference for oxidative deamination over reductive amination. Addition of hydrophobic amino acids affected the GDH activity of the co-purified GdhA–GdhB hetero-complex. Among the amino acids, leucine had the largest effect on activity: addition of 1 mM leucine elevated the GDH activity of the co-purified GdhA–GdhB by 974 and 245 % for reductive amination and oxidative deamination, respectively, while GdhB alone did not show such marked activation by leucine. Kinetic analysis revealed that the elevation of GDH activity by leucine is attributable to the enhanced turnover number of GDH. In this hetero-oligomeric GDH system, GdhA and GdhB act as regulatory and catalytic subunits, respectively, and GdhA can modulate the activity of GdhB through hetero-complex formation, depending on the availability of hydrophobic amino acids. This study provides the first finding, to our knowledge, of a hetero-oligomeric GDH that can be regulated allosterically.

This regulation was found to be transcriptional, and an upstream activation site (GDH2 UASc) sufficient for activation of transcription during respiratory growth conditions was identified. This UAS was found to be separable from a neighboring element which is necessary for the nitrogen source regulation of the gene, and strains deficient for the GLN3 gene product, required for expression of NAD-GDH during growth with the activating nitrogen source glutamate, were unaffected for the expression of NAD-GDH during growth with activating carbon sources. Two classes of mutations which prevented the normal activation of NAD-GDH in response to growth with nonfermentable carbon sources, but which did not affect the nitrogen-regulated expression of NAD-GDH, were found and characterized. Carbon regulation of GDH2 was found to be normal in hxk2, hap3, and hap4 strains and to be only slightly altered in a ssn6 strain; thus, in comparison with the regulation of previously identified glucose-repressed genes, a new pathway appears to be involved in the regulation of GDH2.

ABSTRACT Two linked mutations affecting glutamate dehydrogenase (GDH) formation (gdh-1 and rev-2) had been isolated at a locus near the trp cluster in Klebsiella aerogenes. The properties of these two mutations were consistent with those of a locus containing either a regulatory gene or a structural gene. The gdhA gene from K. aerogenes was cloned and sequenced, and an insertion mutation was generated and shown to be linked to trp. A region ofgdhA from a strain bearing gdh-1 was sequenced and shown to have a single-base-pair change, confirming that the locus defined by gdh-1 is the structural gene for GDH. Mutants with the same phenotype as rev-2 were isolated, and their sequences showed that the mutations were located in the promoter region of the gdhA gene. The linkage of gdhA totrp in K. aerogenes was explained by postulating an inversion of the genetic map relative to other enteric bacteria. Strains that bore high-copy-number clones of gdhAdisplayed an auxotrophy that was interpreted as a limitation for α-ketoglutarate and consequently for succinyl-coenzyme A (CoA). Three lines of evidence supported this interpretation: high-copy-number clones of the enzymatically inactive gdhA1 allele showed no auxotrophy, repression of GDH expression by the nitrogen assimilation control protein (NAC) relieved the auxotrophy, and addition of compounds that could increase the α-ketoglutarate supply or reduce the succinyl-CoA requirement relieved the auxotrophy.

Studies were initiated in 1989 to characterize phonological events with corresponding growth and development phenomena of `Eagle' and `Provider' snap beans (Phaseolus vulgaris L.) Ten plantings at approximately 15 day intervals were made at Knoxville, TN from April 17 through July 27. Days to reach growth stages V0 thru R7 were recorded for each cultivar for each planting date. Air temperature, total radiant energy, wind speed and relative humidity were recorded hourly throughout the 171 day test period. Growing degree days (GDD) computed by 8 methods and growing degree hours (GDH) computed by 2 methods were regressed against plant developmental stages. GDD and GDH, along with pod size and pod fiber content, will be discussed as possible indices for predicting harvest maturity. With the methods used to calculate heat summation in this study, GDD and GDH from planting to pod maturity ranged from approximately 550 to 975 and 9,700 to 20,000, respectively.

ABSTRACT The NAD+-dependent glutamate dehydrogenase (NAD-GDH) from Pseudomonas aeruginosa PAO1 was purified, and its amino-terminal amino acid sequence was determined. This sequence information was used in identifying and cloning the encodinggdhB gene and its flanking regions. The molecular mass predicted from the derived sequence for the encoded NAD-GDH was 182.6 kDa, in close agreement with that determined from sodium dodecyl sulfate-polyacrylamide gel electrophoresis of the purified enzyme (180 kDa). Cross-linking studies established that the native NAD-GDH is a tetramer of equal subunits. Comparison of the derived amino acid sequence of NAD-GDH from P. aeruginosa with the GenBank database showed the highest homology with hypothetical polypeptides from Pseudomonas putida, Mycobacterium tuberculosis, Rickettsia prowazakii, Legionella pneumophila, Vibrio cholerae, Shewanella putrefaciens, Sinorhizobium meliloti, andCaulobacter crescentus. A moderate degree of homology, primarily in the central domain, was observed with the smaller tetrameric NAD-GDH (protomeric mass of 110 kDa) fromSaccharomyces cerevisiae or Neurospora crassa. Comparison with the yet smaller hexameric GDH (protomeric mass of 48 to 55 kDa) of other prokaryotes yielded a low degree of homology that was limited to residues important for binding of substrates and for catalytic function. NAD-GDH was induced 27-fold by exogenous arginine and only 3-fold by exogenous glutamate. Primer extension experiments established that transcription of gdhB is initiated from an arginine-inducible promoter and that this induction is dependent on the arginine regulatory protein, ArgR, a member of the AraC/XyIS family of regulatory proteins. NAD-GDH was purified to homogeneity from a recombinant strain of P. aeruginosa and characterized. The glutamate saturation curve was sigmoid, indicating positive cooperativity in the binding of glutamate. NAD-GDH activity was subject to allosteric control by arginine and citrate, which function as positive and negative effectors, respectively. Both effectors act by influencing the affinity of the enzyme for glutamate. NAD-GDH from this organism differs from previously characterized enzymes with respect to structure, protomer mass, and allosteric properties indicate that this enzyme represents a novel class of microbial glutamate dehydrogenases.

During chronic metabolic acidosis, the adaptive increase in rat renal ammoniagenesis is sustained, in part, by increased expression of mitochondrial glutaminase (GA) and glutamate dehydrogenase (GDH) enzymes. The increase in GA activity results from the pH-responsive stabilization of GA mRNA. The 3′-untranslated region (3′-UTR) of GA mRNA contains a direct repeat of an eight-base AU-rich element (ARE) that binds ζ-crystallin/NADPH:quinone reductase (ζ-crystallin) with high affinity and functions as a pH-response element. RNA EMSAs established that ζ-crystallin also binds to the full-length 3′-UTR of GDH mRNA. This region contains four eight-base sequences that are 88% identical to one of the two GA AREs. Direct binding assays and competition studies indicate that the two individual eight-base AREs from GA mRNA and the four individual GDH sequences bind ζ-crystallin with different affinities. Insertion of the 3′-UTR of GDH cDNA into a β-globin expression vector (pβG) produced a chimeric mRNA that was stabilized when LLC-PK1-F+cells were transferred to acidic medium. A pH-responsive stabilization was also observed using a βG construct that contained only the single GDH4 ARE and a destabilizing element from phospho enolpyruvate carboxykinase mRNA. Therefore, during acidosis, the pH-responsive stabilization of GDH mRNA may be accomplished by the same mechanism that affects an increase in GA mRNA.

The relationship between heat units trends and reproductive development in olive (Olea europaea, L.) was studied over a 3-year period (1999-2001) in 15 areas in the southern Italian regions of Campania, Calabria, Puglia, and Sicily. Heat units were calculated using GDH and GDD formulas and the flowering phases in the olive groves were studied using volumetric pollen traps that aspirate fixed quantities of air. With this method, the olive pollen release and flowering trends were determined. The main objective of the study was to correlate the spring heat unit amounts and the phases of maximum pollen emission with the date of flowering. Moreover, the ranges of GDH and GDD in the different study areas were calculated to identify maps of olive pollen release.

The enzyme glutamate dehydrogenase (GDH; Glud1) catalyzes the (reversible) oxidative deamination of glutamate to α-ketoglutarate accompanied by a reduction of NAD+ to NADH. GDH connects amino acid, carbohydrate, neurotransmitter and oxidative energy metabolism. Glutamine is a neurotransmitter precursor used by neurons to sustain the pool of glutamate, but glutamine is also vividly oxidized for support of energy metabolism. This study investigates the role of GDH in neuronal metabolism by employing the Cns- Glud1−/− mouse, lacking GDH in the brain (GDH KO) and metabolic mapping using 13C-labelled glutamine and glucose. We observed a severely reduced oxidative glutamine metabolism during glucose deprivation in synaptosomes and cultured neurons not expressing GDH. In contrast, in the presence of glucose, glutamine metabolism was not affected by the lack of GDH expression. Respiration fuelled by glutamate was significantly lower in brain mitochondria from GDH KO mice and synaptosomes were not able to increase their respiration upon an elevated energy demand. The role of GDH for metabolism of glutamine and the respiratory capacity underscore the importance of GDH for neurons particularly during an elevated energy demand, and it may reflect the large allosteric activation of GDH by ADP.

The NADP-specific glutamate dehydrogenase (NADP-GDH) activity of Aspergillus nidulans was rapidly lost from cultures starved for a carbon source. This loss of NADP-GDH was blocked by protein synthesis inhibitors. Glutamate repressed NADP-GDH but did not cause rapid loss of activity. Since NADP-GDH is involved in the participation of ammonium in the regulation of nitrogen metabolism, the loss of NADP-GDH activity accompanying carbon starvation may be important in the interaction between carbon and nitrogen metabolism. Increased NAD specific glutamate dehydrogenase activity (NAD-GDH) was observed when mycelium was transferred to medium lacking glucose. The increase in NAD-GDH activity was greatest when glutamate was present. Protein synthesis inhibitors did not prevent this increase in activity. Two mutants, amdT102 and amdT19, which are altered in regulation of nitrogen metabolism, are similar to the wild-type strain with regard to regulation of NADP-GDH and NAD-GDH.

A Administração Central do Sistema de Saúde (ACSS), é uma entidade do Sistema Nacional de Saúde que tem como principal missão a gestão dos recursos humanos e financeiros bem como promover a qualidade organizacional das entidades prestadoras de saúde. O Modelo de Planeamento criado pela Siemens SA surge como o sistema de informação que vem colmatar a falta de informação necessária para a ACSS fazer um ajuste adequado dos recursos à procura por parte da população. No entanto, o mercado onde o Modelo se insere caracteriza-se por estar em constante alteração/actualização exigindo ao Modelo a necessidade de se adaptar para continuar a responder adequadamente. No seguimento desta necessidade surge este trabalho com o principal objectivo de adequar o Modelo à recente criação do conceito de Grupo de Diagnóstico Homogéneo (GDH) para Ambulatório. O conceito de GDH surgiu no mercado da saúde como um sistema de classificação de episódios agudos, associados a Internamento, sendo ainda utilizado para definir operacionalmente a produção de um Hospital. A adaptação deste conceito ao Ambulatório surge na consequência da evolução tecnológica permitir, no presente,tratar doentes num período inferior a 24 horas. Pela análise do impacto do conceito no Modelo, realizado no projecto, foi possível confirmar que o problema está no facto de ser a base de dados dos GDHs que alimenta o Internamento no Modelo, levando a que GDHs de Ambulatório fossem introduzidos erradamente nesta linha de produção. Para corrigir esta situação foi crucial estabelecer regras que impedissem a entrada desses episódios em Internamento. Desta forma, a partir da ferramenta Oracle Warehouse Builder, responsável pela manipulação da informação no Modelo, aplicaram-se as regras necessárias para transferir estes dados, considerando o tempo de internamento e o próprio GDH. Para além da actualização do Modelo ao conceito de GDH Ambulatório foi possível trabalhar a outros níveis promovendo um sistema de informação de qualidade, actual e de alta contribuição para quem é responsável pela distribuição e afectação de recursos no sistema de saúde.

Muitos peixes conseguem sobreviver a elevados níveis de amónia ambiental; entre eles está o Misgurnus anguillicaudatus, este peixe tem também a capacidade invulgar de volatilizar a amónia. Uma dos métodos de tolerar níveis elevados de amónia é através da regulação do metabolismo da glutamina. As reacções de catabólise e anabólise deste metabolismo são catalizadas pela glutaminase (Gase), glutamina sintetase (GSase) e glutamato deshidrogenase (GDH) e permitem o consumo ou síntese de amónia. Estas reacções metabólicas poderão estar à volatilização da amónia no intestino do Misgurnus anguillicaudatus. O objectivo deste trabalho consistia em caracterizar a expressão dos três genes e do seu possível envolvimento na tolerância e volatilização da amónia. A expressão genética foi analisada no decurso de duas experiências. Numa primeira vários tecidos (cérebro, guelra, rim, fígado, e três partes do intestino), de animais controlo, seriam analisados de forma a determinar uma linha base da expressão dos genes. Na segunda experiência os níveis de expressão genética, para os três genes, em duas porções do intestino (intestino anterior e posterior), de animais expostos a elevados níveis de amónia ambiental, seriam quantificados ao longo do tempo para poder determinar possíveis alterações na expressão. Adicionalmente conduzir‐se‐ia a análise das sequências genéticas e comparação filogenética das sequências obtidas no decurso do trabalho. Os resultados da sequenciação mostraram elevada identidade entre as sequências do Misgurnus anguillicaudatus e os seus homólogos noutras espécies, mas na maior parte dos casos também havia distanciação evolutiva entre as sequências das espécies. Os resultados da expressão genética nos vários tecidos mostraram níveis significativamente elevados para a expressão da Gase e GSase no cérebro, estando de acordo com a importância destas enzimas na reciclagem do glutamato e na eliminação de amónia neste órgão. A expressão dos genes no intestino em resposta à exposição à amónia ao longo do tempo n mostraram alterações significativas, isto sugere que o metabolismo da glutamina não é importante para o processo de volatilização ou que regulação não se dá ao nível da transcrição. ABSTRACT: Many fishes are able to cope with high levels of environmental ammonia; among them is the oriental weatherloach (Misgurnus anguillicaudatus), which also has the unusual ability to volatilize ammonia. One proposed method of ammonia tolerance in this species is through the regulation of the glutamine metabolism. Glutaminase (Gase), glutamine synthetase (GSase) and glutamate dehydrogenase (GDH) catalyze reaction in which catabolysis and anabolysis lead to the release or consumption of ammonia, and can possibly be associated with ammonia volatilization in the gut of the weatherloach. This work intended to characterize the expression of these three genes and their possible involvement in ammonia tolerance and ammonia volatilization in the gut of the weatherloach. In order to determine the gene expression levels two experiments were analyzed separately. In one, several tissue samples (brain, gill, liver, kidney and three portions of gut) from control animals would be analyzed in order to determine baseline expression levels of the three genes. In the second experiment gene expression levels for the three genes in two sections of the gut (foregut and hindgut) from animals that had been exposed to high environmental ammonia over time would be analyzed in order to determine possible changes in gene expression. Additionally gene sequence analysis and phylogenic comparison would be carried out on the gene sequences obtained during the work. Sequencing results showed high identity between weatherloach and their homologues in other species for each gene, but in most cases with significant evolutionary branching from other species. The results for gene expression in several tissues showed significantly elevated levels of gene expression for Gase and GSase in the brain, which correlates with the importance of these enzymes in glutamate recycling and ammonia detoxification in this organ. The results for gene expression in gut in response to ammonia exposure over time showed no significant changes suggesting that glutamine metabolism is not important in ammonia volatilization or that regulation is not at the transcriptional level.

Since the 1980's, the study of nucleon (proton or neutron) spin structure has been an active field both experimentally and theoretically. One of the primary goals of this work is to test our understanding of Quantum Chromodynamics (QCD), the fundamental theory of the strong interaction. In the high energy region of asymptotically free quarks, QCD has been verified. However, verifiable predictions in the low energy region are harder to obtain due to the complex interactions between the nucleon's constituents: quarks and gluons. In the non-pertubative regime, low-energy effective field theories such as chiral perturbation theory provide predictions for the spin structure functions in the form of sum rules.;Spin-dependent sum rules such as the Gerasimov-Drell-Hearn (GDH) sum rule are important tools available to study nucleon spin structure. Originally derived for real photon absorption, the Gerasimov-Drell-Hearn (GDH) sum rule was first extended for virtual photon absorption in 1989. The extension of the sum rule provides a unique relation, valid at any momentum transfer ( Q2), that can be used to study the nucleon spin structure and make comparisons between theoretical predictions and experimental data.;Experiment E97-110 was performed at the Thomas Jefferson National Accelerator Facility (Jefferson Lab) to examine the spin structure of the neutron and 3He. The Jefferson Lab longitudinally-polarized electron beam with incident energies between 1.1 and 4.4 GeV was scattered from a longitudinally or transversely polarized 3He gas target in the Hall A end station. Asymmetries and polarized cross-section differences were measured in the quasielastic and resonance regions to extract the spin structure functions g1(x, Q2) and g2(x, Q2) at low momentum transfers (0.02 < Q2 < 0.3 GeV2). The goal of the experiment was to perform a precise measurement of the Q2 dependence of the extended GDH integral and of the moments of the neutron and 3He spin structure functions at low Q2. This Q 2 range allows us to test predictions of chiral perturbation theory and check the GDH sum rule by extrapolating the integral to the real photon point. This thesis will discuss preliminary results from the E97-110 data analysis.

Resumo: A glutamato desidrogenase (GDH) é uma enzima mitochondrial que cataliza a reação reversível de glutamato a ?-cetoglutarato. Nas ilhotas pancreáticas, está associada com a secreção de insulina por aumentar a concentração de ATP. Ratos alimentados com dieta hipoprotéica apresentam secreção de insulina diminuída. A suplementação com leucina (LEU) aumenta a secreção de insulina em resposta a agentes insulinotrópicos. O presente estudo investigou a influência da suplementação com LEU na expressão da GDH e seu envolvimento com a secreção de insulina em ratos desnutridos e suplementados com LEU. Ratos machos foram alimentados com dietas normo- (17%, NP) ou hipoprotéicas (6%, LP) por oito semanas. Após, foram divididos e suplementados com LEU (1,5%) na água de beber (NPL e LPL) pelas quatro semanas seguintes. O conteúdo protéico de GDH no cérebro, fígado, rim e músculo esquelético não diferiu entre os grupos. Nas ilhotas LP, a expressão da GDH estava diminuída e a suplementação com LEU aumentou a expressão de RNAm restaurando o conteúdo protéico a valores similares a NP. A secreção de insulina estimulada por agentes insulinotrópicos ou inibidores, combinados ou não, estava diminuída em ilhotas LP comparada com NP. A suplementação com LEU aumentou a secreção de insulina a valores similares a NP, exceto quando as ilhotas LPL foram incubadas com EGCG. Ilhotas LP tiveram diminuição na [Ca2+]i quando expostas a GLN+BCH. A suplementação com LEU restaurou esses parâmetros aos valores de NP. Frente a esses resultados, podemos concluir que a diminuição na expressão da GDH induzida pela dieta LP foi central ao pâncreas endócrino e está associada à redução da secreção de insulina observada nas ilhotas LP. A suplementação com LEU foi capaz de restaurar a expressão da GDH, contribuindo para o aumento da secreção de insulina observado nas ilhotas LPL. Além disso, a GDH pode, ainda, estar associada com a secreção de insulina pelo acoplamento estímulo/secreção via regulação da [Ca2+]i
Abstract: Glutamate dehydrogenase (GDH) is a mitochondrial matrix enzyme that catalyzes the reversible reaction of glutamate to ?-ketoglutarate. In the pancreatic islets, this enzyme is associated with insulin secretion by augmenting ATP levels. Protein malnourished rats displayed reduced insulin secretion. Leucine (LEU) supplementation augments the insulin secretion response to insulinotropic agents. The present study investigated the influence of LEU supplementation on GDH expression and its involvement with insulin secretion in malnourished rats supplemented with LEU. Male rats were fed normal- (17%, NP) or low-protein diet (6%, LP) for eight weeks. Half of rats of each group were supplemented with LEU (1.5%) in drinking water for the following four weeks (NPL and LPL groups). GDH protein content in brain, liver, kidney and skeletal muscles was not different in any group. GDH RNAm and protein content was reduced in LP islets and LEU supplementation augmented RNAm expression restoring protein content similar to NP. Insulin secretion was reduced in LP islets compared with NP when stimulated by insulinotropics agents or inhybitors, combinaned or not. LEU supplementation augmented insulin secretion to similar values as NP, an effect that was blunted when LPL islets were incubated with EGCG. LP islets showed lower [Ca2+]i when exposed to GLN+BCH. LEU supplementation augmented these patterns similar to NP. Taken together, we may conclude that diminution in GDH expression induced by LP diet was central to endocrine pancreas and was associated with reduced insulin secretion observed in LP rats. LEU supplementation was able to restore GDH expression and it was capable to restore insulin secretion via GDH restoration. Yet, GDH may contribute to insulin secretion via Ca2+ regulation stimulus/secretion coupling

A insuficiência cardíaca é uma doença associada a altas taxas de mortalidade e de internamentos, que afecta sobretudo a população com mais de 65 anos. A sua prevalência encontra-se em crescimento em diversos países, devido em grande parte ao incremento na esperança de vida e ao desenvolvimento de medicamentos mais eficazes. Diversos estudos sugerem que os internamentos hospitalares constituem o principal fator para o aumento dos custos de tratamento associados a esta patologia e, como tal, seria benéfico a definição de estratégias para a sua redução. O objectivo deste trabalho é comparar o consumo de medicamentos dentro dos mesmos Grupos de Diagnósticos Homogéneos (GDH) em hospitais diferentes, nomeadamente o Hospital Garcia de Orta e o Hospital Prof. Dr. Fernando Fonseca. Para tal, foram utilizados métodos de regressão linear múltipla para investigar a influência de diversas variáveis que descrevem o episódio de internamento sobre o custo com medicamentos. Os resultados sugerem que o consumo de medicamentos nos dois hospitais é similar para um dado GDH, o que é uma das questões fulcrais deste estudo. Após a análise de dados, apurámos através da análise exploratória e ajuste de modelos que os custos com medicamentos aumenta em média com o número de dias de internamento, sendo isso mais notório para o Hospital Garcia da Orta. No entanto, a idade e o sexo não parecem ter um efeito apreciável no custo. Adicionalmente, em algumas regressões foi encontrada evidência de heterocedasticidade, problema que teve de ser solucionado através da estimação robusta de White.
Heart failure is a disease associated with high mortality rates and hospital admissions. It mostly affects the population aged 65 or more and its prevalence is growing in many countries, mainly due to the increase of life expectancy and the development of new and more effective treatments. Previous research suggests that a large proportion of the treatment's cost of this pathology is comprised of hospital internments. Thus, it would be of interest to set out strategies to its reduction. The purpose of the present study is to compare the associated with cost of medication within the same Diagnosis Related Groups (DRG) in different hospitals, namely in Hospital Garcia da Orta and Hospital Prof. Dr. Fernando Fonseca. To do so, we use multiple linear regression methods to select the variables that explain the medicines' cost. The results suggest that a patient of a given DRG uses approximately the same resources regardless of the hospital where (s)he receives medical assistance. Thus, we conclude the cost associated with medication is the same for a given DRG. In addition, we find that the average associated with medication increases with the length of stay. This seems to be particularly true for Hospital Garcia da Orta. On the other hand, explanatory variables such as age and sex do not seem have a significant effect in the medication cost. To correct the standard errors for the presence of heteroskedasticity we used White's robust covariance matrix.

Abstract (sommario):
Presented in this paper is the transient analysis of a Group Distribution Header (GDH) following a guillotine break at the end of the header. The GDH is the most important component of reactor safety in case of accidents. Emergency Core Cooling System (ECCS) piping is connected to the GDH piping such that, during an accident, coolant passes from the GDH into the ECCS. The GDH that is propelled into motion after a guillotine break can impact neighboring GDH pipes or the nearest wall of the compartment. Therefore, two cases are investigated: • GDH impact on an adjacent GDH and its attached piping; • GDH impact on an adjacent reinforced concrete wall. A whipping RBMK-1500 GDH along with neighboring concrete walls and pipelines is modeled using finite elements. The finite element code NEPTUNE used in this study enables a dynamic pipe whip structural analysis that accommodates large displacements and nonlinear material characteristics. The results of the study indicate that a whipping GDH pipe would not significantly damage adjacent walls or piping and would not result in a propagation of pipe failures.

A growing body of scholarly literature has attempted to measure and value unpaid care work in various countries, but perhaps only the government statistical agencies in the United States and the United Kingdom have seriously undertaken periodic and systematic measures of the time spent on unpaid work at the national level, and partially incorporated those values into their gross domestic product(GDP). One country that has been ahead of its time on aspects of societal welfare measurement is Bhutan, which produces the Gross National Happiness (GNH) Index. However, until the first GNH Survey, in 2008, Bhutan did not have any sense of the size and distribution of unpaid work, despite its strong societal norms about the value of volunteering and community work. This paper is the first to estimate the value of unpaid care work in Bhutan. It shows the pros and cons of various approaches and their equivalent measures of unpaid care work as a share of GDP. As with similar studies on the topic, this paper also finds that women spend more than twice as much time as men performing unpaid care work, regardless of their income, age, residency, or number of people in the household. The paper also provides recommendations for improving the measurement of unpaid care work in Bhutan.
