Abstract (sommario):
Dual-modal imaging by combining magnetic resonance (MR) and near-infrared (NIR) fluorescence can integrate the advantages of high-resolution anatomical imaging with high sensitivity in vivo fluorescent imaging, which is expected to play a significant role in biomedical researches. Here we report a dual-modality imaging probe (NIR/MR-MSNs) fabricated by conjugating NIR fluorescent heptamethine dyes (IR-808) and MR contrast agents (Gd-DTPA) within highly aminated mesoporous silica nanoparticles (MSNs-NH2). The dual-modality imaging probes NIR/MR-MSNs possess a size of ca. 120 nm. The NIR/MR-MSNs show not only near-infrared fluorescence imaging property with an emission peak at 794 nm, but also highly MRT1relaxivity of 14.54 mM−1 s−1, which is three times more than Gd-DTPA. In vitro experiment reveals high uptake and retention abilities of the nanoprobes, while cell viability assay demonstrates excellent cytocompatibility of the dual-modality imaging probe. After intratumor injection with the NIR/MR-MSNs, MR imaging shows clear anatomical border of the enhanced tumor region while NIR fluorescence exhibits high sensitive tumor detection ability. These intriguing features suggest that this newly developed dual-modality imaging probes have great potential in biomedical imaging.

Gadolinium chelate and Quantum Dots (QDs) decorated single walled carbon nanotubes (SWNTs) nanohybrids (SWNT-Gd-QDs) were designed for dual-modal cellular imaging via magnetic resonance imaging (MRI) and optical imaging. The nanohybrids were prepared via a simple and novel layer-by-layer (LBL) assembly in combination with covalent connection strategy. The SWNT-Gd-QDs nanohybrids showed a strong near-infrared photoluminescence (PL) emission at room temperature and enhanced T1-weighted and T2-weighted MRI signals in water. Thus, the nanohybrids combine magnetic and fluorescent properties and can serve as dual-modal MRI/optical imaging contrast agent.

The design of near-infrared fluorescent (NIRF) probes that are activated by specific proteases has, for the first time, allowed enzyme activity to be imaged in vivo. In the current study, we report on a method of imaging enzyme activity using two fluorescent probes that, together, provide improved quantitation of enzymatic activity. The method employs two chemically similar probes that differ in their degradability by cathepsin B. One probe consists of the NIRF dye Cy5.5 attached to a particulate carrier, a crosslinked iron oxide nanoparticle (CLIO), through cathepsin B cleavable l-arginyl peptides. A second probe consists of Cy3.5 attached to a CLIO through proteolytically resistant d-arginyl peptides. Using mixtures of the two probes, we have shown that the ratio of Cy5.5 to Cy3.5 fluorescence can be used to determine levels of cathepsin B in the environment of nanoparticles with macrophages in suspension. After intravenous injection, tissue fluorescence from the nondegradable Cy3.5–d-arginyl probe reflected nanoparticle accumulation, while fluorescence of the Cy5.5–l-arginyl probe was dependent on both accumulation and activation by cathepsin B. Dual wavelength ratio imaging can be used for the quantitative imaging of a variety of enzymes in clinically important settings, while the magnetic properties of the probes allow their detection by MR imaging.

Dual or multimodal imaging probes have emerged as powerful tools that improve detection sensitivity and accuracy in disease diagnosis by imaging techniques. Two imaging techniques that are complementary and do not use ionizing radiation are magnetic resonance imaging (MRI) and optical fluorescence imaging (OFI). Herein, we prepared metal-free organic species based on dendrimers with magnetic and fluorescent properties as proof-of-concept of bimodal probes for potential MRI and OFI applications. We used oligo(styryl)benzene (OSB) dendrimers core that are fluorescent on their own, and TEMPO organic radicals anchored on their surfaces, as the magnetic component. In this way, we synthesized six radical dendrimers and characterized them by FT-IR, 1H NMR, UV-Vis, MALDI-TOF, SEC, EPR, fluorimetry, and in vitro MRI. Importantly, it was demonstrated that the new dendrimers present two properties: on one hand, they are paramagnetic and show the ability to generate contrast by MRI in vitro, and, on the other hand, they also show fluoresce emission. This is a remarkable result since it is one of the very few cases of macromolecules with bimodal magnetic and fluorescent properties using organic radicals as the magnetic probe.

We report a reliable and facile procedure to prepare near infrared fluorescence (NIRF)-magnetic resonance dual-modal imaging probes (DMIPs) using Cy5-labeled, Gd–Al co-doped mesoporous silica nanoparticles. High intensity NIRF and a relaxation rate (17.7 mM−1s−1) can be observed. Furthermore, the DMIPs in saline were injected into mice through the tail vein for imaging.

L’imagerie par fluorescence (IF) et l’imagerie de résonance magnétique (IRM) comptent parmi les outils de diagnostic les plus efficaces. Dans ce contexte, des QDs possédant à la fois des propriétés fluorescentes et magnétiques sont d’un grand intérêt en tant que sondes bimodales. Dans ce travail, des QDs Ag-In-Zn-S (AIZS) et Ag-In-Ga-Zn-S (AIGZS) ont été préparés et dopés afin de développer de nouvelles sondes de sondes bimodales pour l’IF et l’IRM. Des QDs AIZS très fluorescents ont été préparés en milieu organique à l’aide de DDT et d’OAm comme ligands. Les QDs Mn:AIZS possèdent des propriétés paramagnétiques et superparamagnétiques. Les QDs AIZS et Mn:AIZS ont également été transférés en phase aqueuse à l’aide du polymère amphiphile PMAO. Par la suite, des QDs AIZS dopés Mn, Gd ou Fe ont été préparés en milieu. Des études toxicologiques et d’imagerie ont montré une bonne biocompatibilité avec les cellules KB ainsi que le fort potentiel de ces nanocristaux pour l’IF. Dans la dernière partie de ce travail, des QDs AIGZS and Mn:AIGZS QDs ont été préparés via un procédé de décomposition thermique n’utilisant qu’un seul précurseur. Ces QDs possèdent de très bonnes propriétés optiques et magnétiques. Les QDs dopés Mn ont été transférés en phase aqueuse et ont montré un fort potentiel comme agent de contraste pour l’imagerie T1 et T2
Since fluorescence imaging (FI) and magnetic resonance imaging (MRI) are among the most effective diagnostic tools, QDs with fluorescent and magnetic properties are of great interest as dual-modal probes. In this work, undoped and doped Ag-In-Zn-S (AIZS) and Ag-In-Ga-Zn-S (AIGZS) QDs were synthesized and investigated as bimodal probes for FI and MRI. Highly fluorescent AIZS QDs were prepared in organic media using DDT and OAm as capping ligands. Mn:AIZS QDs showed paramagnetic and superparamagnetic properties. AIZS and Mn:AIZS QDs were also transferred into aqueous phase using the amphiphilic PMAO polymer. Further, Mn, Gd or Fe-doped AIZS QDs were prepared in aqueous media, showed low cytotoxicity toward KB cells, and demonstrated potential as fluorescent probes for FI. Finally, AIGZS and Mn:AIGZS QDs, synthesized via a novel single precursor thermal decomposition method, showed high fluorescence and paramagnetic/superparamagnetic properties. Mn-doped aqueous transferred AIGZS QDs increased contrast in both T1-weighted and T2-weighted images with increasing in Mn loading
