e12519 Background: The 21 gene assay Oncotype Dx Breast Recurrence Score (RS) is currently the standard of care to determine if adjuvant chemotherapy is needed in early stage node negative, hormone receptor positive, HER-2 negative breast cancer. In current American society of clinical oncology (ASCO) guidelines there is little or no benefit of adjuvant chemotherapy in patients older than 50 years whose tumors have Oncotype DX RS <26, and for patients 50 years or less whose tumors have Oncotype DX RS <16. We sought to evaluate the percentage of estrogen receptor (ER) expression as a surrogate measure of determining adjuvant chemotherapy by examining the relationship between ER expression and RS. Methods: We identified 301 patients from years 2015 to 2019 from our cancer registry with early stage hormone receptor positive breast cancer and had oncotype DX testing performed. We divided patients into three groups: Group 1 (ERG1) with ER <10%; Group 2 (ERG2) with ER 10-49%; and Group 3 (ERG3) with ER equal or >50%. We also collected information on tumor size (cm), tumor grade, Nottingham score, and ki-67 percentage. A sub-group analysis was performed for patients < 50 years age (n=30). We compared all continuous variables across ER groups using the Kruskal-Wallis rank test and individual between group comparisons using the Wilcoxon rank sum test. All statistical tests performed utilized a two-tailed p value of <0.05 with the Bonferroni correction for multiple comparisons. Results: Among 301 patients with early stage hormone receptor positive breast cancer, 89.1% were ductal, 7.9% lobular, and 2.9% mixed histology. Median age was 68, 58 and 66 for ERG1, ERG2, and ERG3, respectively (p = 0.41). Median RS was 36 (ERG1), 23 (ERG2), and 16 (ERG3) (p = 0.78 for ERG1 vs. ERG2; p = 0.01 for ERG1 vs. ERG3). As expected, tumor grade, tumor size, and Nottingham score decreased significantly from ERG1 to ERG3. For patients <50 years, median age was 44, 46 and 45 for ERG1, ERG2, and ERG3, respectively (p = 0.75). Median RS was 10 (ERG1), 24 (ERG2) and 18 (ERG3) (p = 0.04 for ERG1 vs. ERG2; p = 0.17 for ERG1 vs. ERG3). Conclusions: We found a significant association between estrogen receptor levels and Oncotype Dx recurrence score (RS) in patients with early stage hormone receptor positive breast cancer patients. Further studies are needed to determine the predictive ability of hormone receptor levels on the outcomes of patients treated for early stage hormone receptor positive breast cancer.

ABSTRACT The application of genome-wide expression profiling to determine how drugs achieve their therapeutic effect has provided the pharmaceutical industry with an exciting new tool for drug mode-of-action studies. We used DNA chip technology to study cellular responses to perturbations of ergosterol biosynthesis caused by the broad-spectrum antifungal agent itraconazole. Simultaneous examination of over 6,600 Candida albicans gene transcript levels, representing the entire genome, upon treatment of cells with 10 μM itraconazole revealed that 296 genes were responsive. For 116 genes transcript levels were decreased at least 2.5-fold, while for 180 transcript levels were similarly increased. A global upregulation ofERG genes in response to azole treatment was observed.ERG11 and ERG5 were found to be upregulated approximately 12-fold. In addition, a significant upregulation was observed for ERG6, ERG1, ERG3, ERG4, ERG10, ERG9, ERG26, ERG25, ERG2, IDII, HMGS, NCP1, and FEN2, all of which are genes known to be involved in ergosterol biosynthesis. The effects of itraconazole on a wide variety of known metabolic processes are discussed. As over 140 proteins with unknown function were responsive to itraconazole, our analysis might provide—in combination with phenotypic data—first hints of their potential function. The present report is the first to describe the application of DNA chip technology to study the response of a major human fungal pathogen to drug treatment.

The interstitial cells of Cajal (ICC), as pacemaker cells of the gut, contribute to rhythmic peristalsis and muscle excitability through initiation of slow-wave activity, which subsequently actively propagates into the musculature. An E-4031-sensitive K+ current makes a critical contribution to membrane potential in ICC. This study provides novel features of this current in ICC in physiological solutions and seeks to identify the channel isoform. In situ hybridization and Kit immunohistochemistry were combined to assess ether-a-go-go-related gene (ERG) mRNA expression in ICC in mouse jejunum, while the translated message was assessed by immunofluorescence colocalization of ERG and Kit proteins. E-4031-sensitive currents in cultured ICC were studied by the whole cell patch-clamp method, with physiological K+ concentration in the bath and the pipette. In situ hybridization combined with Kit immunohistochemistry detected m-erg1 and m-erg3, but not m-erg2, mRNA in ICC. ERG3 protein was colocalized with Kit-immunoreactive ICC in jejunum sections, but ERG1 protein was visualized only in the smooth muscle cells. At physiological K+ concentration, the E-4031-sensitive outward current in ICC was different from its counterpart in cardiac and gut smooth muscle cells. In particular, inactivation upon depolarization and recovery from inactivation by hyperpolarization were modest in ICC. In summary, the E-4031-sensitive currents influence the kinetics of the pacemaker activity in ICC and contribute to maintenance of the resting membrane potential in smooth muscle cells, which together constitute a marked effect on tissue excitability. Whereas this current is mediated by ERG1 in smooth muscle, it is primarily mediated by ERG3 in ICC.

However, the development of resistance to this drug inC. albicansbiofilms is very common, especially in immunocompromised individuals. The upregulation of the sterol biosynthetic pathway geneERGand the efflux pump genesCDRandMDRmay contribute to this azole tolerance inCandidaspecies. We hypothesize that farnesol, an endogenous quorum sensing molecule with possible antimicrobial properties which is also the precursor of ergosterols inC. albicans, may interfere with the development of fluconazole resistance inC. albicansbiofilms. To test this hypothesis, MICs were compared and morphology changes were observed by confocal laser scanning microscopy (CLSM) for farnesol-treated and -untreated and fluconazole-resistant groups. The expression of possible target genes (ERG11,ERG25,ERG6,ERG5,ERG3,ERG1,MDR1,CDR1, andCDR2) in biofilms was analyzed by reverse transcription-PCR (RT-PCR) and quantitative PCR (qPCR) to investigate the molecular mechanisms of the inhibitory effects of farnesol. The results showed a decreased MIC of fluconazole and thinner biofilms for the farnesol-treated group, indicating that farnesol inhibited the development of fluconazole resistance. The sterol biosynthetic pathway may contribute to the inhibitory effects of farnesol, as the transcription levels of theERG11,ERG25,ERG6,ERG3, andERG1genes decreased in the farnesol-treated group.

ABSTRACT Two erg3 genes encoding C-5 sterol desaturase enzymes (Erg3A and Erg3B) in Aspergillus fumigatus were characterized with respect to their nucleotide sequences and null mutant phenotypes. Targeted disruption of the erg3A and erg3B genes and a double gene knockout, erg3A − erg3B −, showed that they are not essential for A. fumigatus viability. Mutant phenotypes clearly showed that Erg3B is a C-5 sterol desaturase, but no apparent role for Erg3A in A. fumigatus ergosterol biosynthesis was found. Susceptibility to amphotericin B, itraconazole, fluconazole, voriconazole, and ketoconazole was not altered in isolates in which erg3A and erg3B were knocked out alone and in combination.

Abstract The oncogenic ETS transcription factor ERG is involved in various cellular pathways including developmental regulation, proliferation, and differentiation. In hematopoiesis ERG plays a specific role during normal T-cell differentiation showing high expression levels in stem cells and down regulation in the progenitor compartment. In this regard, it is intriguing that aberrant expression of ERG was found in a subset of patients with acute T-lymphoblastic leukemia (T-ALL) and was associated with an inferior outcome. Furthermore, high level ERG expression was of adverse prognostic significance in patients with newly diagnosed acute myeloid leukemia (AML), thus highlighting ERG’s potential role in myeloid as well as T-lineage leukemogenesis. ERG3 (NM_182918) and ERG2 (NM_004449) represent the main isoforms and show abundant expression in myeloid and lymphoid hematopoietic progenitor cells. The expression pattern of specific ERG isoforms in acute leukemias has yet to be investigated. To further elucidate the nature of aberrant ERG expression we have determined the existence and transcriptional regulation of ERG isoforms in pretreatment bone marrow samples of adult T-ALL (n=21) and AML (n=20) patients as well as in normal CD34+ hematopoietic cells of healthy volunteers (n=5). 5′RACE revealed the presence of a new ERG isoform (ERG3Δex12) characterized by expression of exon 5 and absence of exon 12. Expression of ERG3Δex12 was verified by RT-PCR in AML, T-ALL, and CD34+ cells. In addition, real-time RT-PCR showed concomitant expression of the two main isoforms ERG2 and ERG3 in AML and normal CD34+ cells. In contrast, T-ALL patients lacked expression of ERG isoforms harboring exon 4 (ERG2). Promoter analyses of ERG2 and ERG3 revealed the presence of two CpG islands in the ERG2 promoter region, whereas no CpG island was predicted in the ERG3 promoter. Bisulfit conversion of genomic DNA and sequencing of cloned PCR products revealed a significantly higher degree of methylation of CpG island 2 in T-ALL samples (median: 86.4%, range: 16.0 – 98.8%) as compared to AML (median: 38.1%, range: 10.9 – 60.7%; P-value=0.0002 - two sided T-test). As for CpG island 1, CD34+ cells had the lowest rate of methylation in CpG island 2 (median: 7.7%, range: 2.4 – 20.7%). Thus, the differential expression of ERG isoforms is mediated by epigenetic silencing of exon 4 containing transcripts in T-ALL. In conclusion, the identification of the new ERG isoform (ERG3Δex12) suggests the association with different partners as the central exons, including exon 12, guide the interaction with different proteins. Furthermore, the distinct expression of specific ERG transcripts controlled by methylation adds to the complexity of ERG directed downstream pathways in different leukemic subtypes.

ABSTRACT Sterol analysis identified four Candida albicans erg3 mutants in which ergosta 7,22-dienol, indicative of perturbations in sterol Δ5,6-desaturase (Erg3p) activity, comprised >5% of the total sterol fraction. The erg3 mutants (CA12, CA488, CA490, and CA1008) were all resistant to fluconazole, voriconazole, itraconazole, ketoconazole, and clotrimazole under standard CLSI assay conditions (MIC values, ≥256, 16, 16, 8, and 1 μg ml−1, respectively). Importantly, CA12 and CA1008 retained an azole-resistant phenotype even when assayed in the presence of FK506, a multidrug efflux inhibitor. Conversely, CA488, CA490, and three comparator isolates (CA6, CA14, and CA177, in which ergosterol comprised >80% of the total sterol fraction and ergosta 7,22-dienol was undetectable) all displayed azole-sensitive phenotypes under efflux-inhibited assay conditions. Owing to their ergosterol content, CA6, CA14, and CA177 were highly sensitive to amphotericin B (MIC values, <0.25 μg ml−1); CA1008, in which ergosterol comprised <2% of the total sterol fraction, was less sensitive (MIC, 1 μg ml−1). CA1008 harbored multiple amino acid substitutions in Erg3p but only a single conserved polymorphism (E266D) in sterol 14α-demethylase (Erg11p). CA12 harbored one substitution (W332R) in Erg3p and no residue changes in Erg11p. CA488 and CA490 were found to harbor multiple residue changes in both Erg3p and Erg11p. The results suggest that missense mutations in ERG3 might arise in C. albicans more frequently than currently supposed and that the clinical significance of erg3 mutants, including those in which additional mechanisms also contribute to resistance, should not be discounted.

ABSTRACT Two clinical Candida albicans isolates that exhibited high-level resistance to azoles and modest decreases in susceptibility to amphotericin B were cultured from unrelated patients. Both isolates harbored homozygous nonsense mutations in ERG3, which encodes an enzyme, sterol Δ5,6-desaturase, involved in ergosterol synthesis. Extraction and analysis of the sterols from both isolates confirmed the absence of sterol Δ5,6-desaturase activity. Although the loss of sterol Δ5,6-desaturase activity is known to confer resistance to azoles, this mechanism of resistance has rarely been seen in clinical isolates, suggesting that such mutants are at a competitive disadvantage. To test this hypothesis, the virulence of the erg3 mutants was assayed by using a mouse systemic infection model. The mutants were significantly less virulent than the wild-type comparator strains. However, the kidney fungal burdens in mice infected with the erg3 mutants were similar to those in mice infected with the wild-type strains. Similar results were obtained by using a laboratory-generated homozygous erg3 deletion mutant (D. Sanglard et al., Antimicrob. Agents Chemother. 47:2404-2412, 2003). Reintroduction of a wild-type ERG3 allele into the homozygous deletion mutant restored virulence, ergosterol synthesis, and susceptibility to azoles, confirming that these phenotypic changes were solely due to the inactivation of Erg3p.

The calcium/calcineurin signalling pathway is required for cell survival under various environmental stresses. Using Saccharomyces cerevisiae, we explored the mechanism underlying calcium-regulated homeostasis of intracellular reactive oxygen species (ROS). We found that deletion of acyltransferase Akr1 and C-5 sterol desaturase Erg3 increased the intracellular ROS levels and cell death, and this could be inhibited by the addition of calcium. The hexose transporter Hxt1 and the amino acid permease Agp1 play crucial roles in maintaining intracellular ROS levels, and calcium induced the expression of the HXT1 and AGP1 genes. The cytosolic calcium concentration was decreased in both the akr1Δ and erg3Δ mutants relative to wild-type cells, potentially lowering basal expression of HXT1 and AGP1. Moreover, the calcium/calcineurin signalling pathway also induced the expression of AKR1 and ERG3, indicating that Akr1 and Erg3 might perform functions that help yeast cells to survive under high calcium concentrations. Our results provided mechanistic insight into how calcium regulated intracellular ROS levels in yeast.

A characteristic feature of species of Trichoderma is the production of concentric rings of conidia in response to alternating light-dark conditions. In response to a single burst of light, a single ring of conidia forms at what was the colony perimeter. On the basis of these observations, competency to photoconidiate has been proposed to be due to the age and metabolic rate of the hyphal cell. In this study, conidiation was investigated in five biocontrol isolates (T. hamatum, T. atroviride, T. asperellum, T. virens and T. harzianum) using both a morphological and molecular approach. All five isolates produced concentric conidial rings under alternating light-dark conditions on potato-dextrose agar (PDA), however, in response to a 15 min burst of blue light, only T. asperellum and T. virens produced a clearly, defined conidial ring which correlated with the colony margin at the time of light exposure. Both T. harzianum and T. hamatum photoconidiated in a disk-like fashion and T. atroviride produced a broken ring with a partially filled in appearance. On the basis of these results, it was postulated that competency to photoconidiate is a factor of the metabolic state of the hyphal cell rather than chronological age or metabolic rate. The influence of the source of nitrogen on photoconidiation was assessed on pH-buffered (pH 5.4) minimal medium (MM) amended with glutamine, urea or KNO₃. In the presence of glutamine or urea, T. asperellum and T. harzianum conidiated in a disk, whereas, when KNO₃ was the sole nitrogen source, a ring of conidia was produced. Further, in the presence of increasing amounts of glutamine, the clearly defined photoconidial ring produced on PDA by T. asperellum became disk-like. These results clearly demonstrated that primary nitrogen promotes photoconidiation in these isolates and strongly suggests that competency of a hyphal cell to conidiate in response to light is dependent on the nitrogen catabolite repression state of the cell. The experiments were repeated for all five isolates on unbuffered MM. Differences were apparent between the buffered and unbuffered experiments for T. atroviride. No photoconidiation was observed in T. atroviride on buffered medium whereas on unbuffered medium, rings of conidia were produced on both primary and secondary nitrogen. These results show that photoconidiation in T. atroviride is influenced by the buffering capacity of the medium. Conidiation in response to light by T. hamatum and T. virens was absent in all nitrogen experiments, regardless of the nitrogen source and buffering capacity, whereas both isolates conidiated in response to light on PDA. These results imply that either both sources of nitrogen are required for photoconidiation, or a factor essential for conidiation in these two isolates was absent in the minimal medium. Mycelial injury was also investigated in five biocontrol isolates of Trichoderma. On PDA, all isolates except T. hamatum conidiated in response to injury. On nitrogen amended MM, conidiation in response to injury was again observed in all isolates except for T. hamatum. In T. atroviride, injury-induced conidiation was observed on all medium combinations except the pH-buffered MM amended with glutamine or urea and T. virens conidiated in response to injury on primary nitrogen only, regardless of the buffering capacity. These results have revealed conidiation in response to injury to be differentially regulated between isolates/species of Trichoderma. On unbuffered MM amended with glutamine or urea, conidiation in response to injury occurred at the colony perimeter only in T. atroviride. It was hypothesised that the restriction of conidiation to the perimeter may be due to changes in the pH of the agar. The experiment was repeated and the pH values of the agar under the growing colony measured at the time of light induction (48 h) or injury (72 h). The areas under the hyphal fronts were acidified to below the starting value of the medium (pH 5.4) and the centres of the plates were alkalinised. The region of acidification at the time of stimuli correlated with the production of conidia, which implicates a role for crossregulation of conidiation by the ambient pH. The influence of the ambient pH on injury-induced conidiation was investigated in T. hamatum and T. atroviride on MM amended with glutamine and PDA, pH-buffered from pH 2.8 to 5.6. Thickening of the hyphae around the injury site was observed at the lowest pH values on MM in both T. atroviride and T. hamatum, however no conidia were produced, whereas both Trichoderma species conidiated on pH-buffered PDA in a strictly low pH-dependent fashion. This is the first observation of injury-induced conidiation in T. hamatum. The influence of the ambient pH on photoconidiation was assessed in T. hamatum, T. atroviride and T. harzianum using both buffered and unbuffered PDA from pH 2.8 to 5.2. On buffered PDA, no conidiation in response to light was observed above pH 3.2 in T. hamatum, above 4.0 in T. atroviride and above 4.4 in T. harzianum, whereas on unbuffered PDA it occurred at all pH values tested. It was postulated that conidiation at pH values above 4.4 on unbuffered PDA was due to acidification of the agar. The pH values of the agar under the growing colony were measured at the time of light exposure and in contrast to the MM with glutamine experiments, alkalisation of the agar had occurred in both T. atroviride and T. hamatum. No change in medium pH was recorded under the growing T. harzianum colony. These results indicate that low pH-dependence of photoconidiation is directly related to the buffering capacity of the medium. Recent studies have linked regulation of conidiation in T. harzianum to Pac1, the PacC orthologue. In fungi, PacC regulates gene expression in response to the ambient pH. In these studies pH-dependent photoconidiation occurred only on buffered PDA and on unbuffered PDA conidiation occurred at significantly higher ambient pH levels. It is proposed that the influence of ambient pH on conidiation in the isolates used in this study is not due to direct Pac1 regulation. The T. harzianum isolate used in this study produced profuse amounts of the yellow anthraquinone pachybasin. Production of this secondary metabolite was strictly pH-dependent, irrespective of the buffering capacity of the medium. Studies in T. harzianum have linked Pac1 regulation to production of an antifungal α-pyrone. pH-dependence on both buffered and unbuffered media strongly suggests that pachybasin production may also be under the control of Pac1. Photoconidiation studies on broth-soaked filter paper, revealed rhythmic conidiation in the pachybasin producing T. harzianum isolate. Diffuse rings of conidia were produced in dark-grown cultures and, in cultures exposed to light for 15 min at 48 h, the rings were clearly defined. These results show that conidiation is under the control of an endogenous rhythm in T. harzianum and represent the first report of circadian conidiation in a wild-type Trichoderma. A Free-Running Rhythm (FRR) assay was used to investigate rhythmic gene expression in T. atroviride IMI206040 and a mutant derivative, in which the wc-2 orthologue, blr-2, was disrupted. Over a 3 d period, expression of gpd, which encodes the glycolytic enzyme glyceraldehyde-3-phosphate dehydrogenase, oscillated with a period of about 48 h. In the Δblr-2 mutant, the gpd rhythm was absent. These results revealed that in T. atroviride, gpd expression is under the control of an endogenous clock and that clock-regulated expression of gpd is associated with a functional BLR complex. Using degenerate primers, a portion of frq, which encodes the N. crassa clock oscillator FREQUENCY, was isolated from T. atroviride and used to probe the FRR assay northern blots. No frq expression was detected at any time point, which suggests that the circadian clock in Trichoderma does not involve FREQUENCY. In a concurrent study, orthologues of rco-1 (rcoT) were isolated and sequenced from T. atroviride and T. hamatum using a combination of degenerate, inverse and specific PCR. RcoT is an orthologue of the yeast global co-repressor Tup1 and in the filamentous fungi, RcoT orthologues have been demonstrated to negatively regulate conidiation. Genomic analysis of all available rcoT orthologues revealed the conservation of erg3, a major ergosterol biosynthesis gene, upstream from rcoT in ascomycetous filamentous fungi, but not in the ascomycetous yeast or in the basidiomycetes. These studies have significantly contributed to our understanding of the regulatory factors controlling conidiation in Trichoderma and have multiple implications for Trichoderma biocontrol; most notable the promotion of conidiation by primary nitrogen and low pH. Incubation conditions can be altered to suit the nitrogen and pH preferences of a biocontrol strain in order to promote cost effective conidial production, however this is not easily achieved in the soil, where the biocontrol strain must perform in a highly buffered environment optimised for plant growth. Successful use of Trichoderma biocontrol strains may involve the screening and targeting of strains to the appropriate pH conditions or the selection of new strains on the basis of capacity to perform under a given range of conditions.

Os projetistas de web sites são continuamente confrontados com um desafio: tornar seus web sites atraentes aos usuários e garantir que estes sejam capazes de realizar suas interações de maneira fácil, eficiente e agradável. Em muitos casos, a rentabilidade financeira das empresas que utilizam a internet como um meio de difusão, ou até mesmo de comércio, de seus produtos está fortemente dependente da qualidade das interfaces humano-computador desenvolvidas. Nesse contexto, uma técnica de avaliação da usabilidade de web sites tem um importante papel, que é o de fornecer aos avaliadores as melhores definições para que estes sejam capazes de influir no ciclo de desenvolvimento da usabilidade dos produtos avaliados. Este trabalho propõe a definição e implementação de um protótipo para um Sistema de Monitoramento (batizado por Ergo-Monitor) para a coleta de informações que possam nortear a elaboração de laudos quantitativos acerca da usabilidade de web sites (com ênfase em sistemas de comércio eletrônico). O ambiente Ergo-Monitor baseia-se na coleta de dados diretamente das interações dos usuários que se encontram armazenados nos server log files. Após essa coleta, as ferramentas do ambiente fazem o tratamento desses dados produzindo taxas e métricas de usabilidade que devem ser, na seqüência, confrontadas pelo avaliador com valores característicos de boas e más interações para web sites que estejam na mesma categoria do web site sendo avaliado. O ambiente Ergo-Monitor apóia uma interação produtiva entre laudos quantitativos e qualitativos de avaliação de usabilidade de web sites e os laudos quantitativos produzidos pelo Ergo-Monitor podem vir a confirmar diagnósticos qualitativos elaborados previamente, ou vir a suscitar novas avaliações qualitativas da usabilidade dos web sites monitorados. Acima de tudo, o Ergo-Monitor está sendo proposto como um ambiente de avaliação "invisível" aos usuários e cujos tratamentos dos dados coletados sejam feitos de forma objetiva, automática e econômica. Assim, seus resultados não são influenciados por ruídos nas observações, como a presença de avaliadores e equipamentos, os usuários não se sentem eles mesmos o foco da avaliação e os custos de uma avaliação in loco e a posteriori podem ser minimizados.

Įvairios komunikacijos technologijos yra tapusios neatsiejama žmogaus gyvenimo dalimi. Jas žmonės naudoja dažnai net nesusimąstydami kaip tai pakeičia natūralią jų būklę ir kokią įtaką tai daro kasdieniam gyvenimui. Šiame darbe nagrinėjami pagrindiniai žmogiškosios komunikacijos būdai kultūroje, apžvelgiama komunikacijos technologijų raida ir jų vaidmuo socialiniame gyvenime, galop nagrinėjama viena naujausių komunikacijos technologijų – internetas, žmonėms pasiūlęs beprecedentes sąveikos galimybes. Viena iš naujausių interneto galimybių yra interneto dienoraštis, dar kitaip vadinamas blogu. Tai istoriškai susiklostęs žmogaus sąveikos būdas, sujungiantis ankstesnes sąveikos tradicijas ir siūlantis naujas galimybes savęs pateikimui ir savęs kūrimui. Šiame darbe nagrinėjama, kokį poveikį interneto dienoraštis turi žmogui šių laikų situacijoje, kokios yra interneto dienoraščio kaip sąveikos būdo savybės, bei kokius poreikius ir kaip žmogus realizuoja naudodamasis interneto dienoraščiu. The main theme of the thesis – Blog (or internet diary) as the unique way of human communication. The main objective is to analyze various means and ways of communication, which are of significance in respect to analyzing the nature of blog. Various communication technologies have become an irreplaceable part of human life. Human usually use them without even having a thought how these means change their natural way of being and what influence they have on one’s daily life. In the thesis the main manners of human communication in the culture are analyzed, the development of communication technologies and their role in the society is described, lastly, one of the newest technologies – the internet having offered unprecedented capabilities for people to interact, is approached. One of the newest scopes of the internet is the blog. It is historically evolved mean of communication, integrating previous traditions of interaction and enabling new means for self-presentation and self-creation. In this work the influence of the blog to human is analyzed, what are the aspects of the blog as the mean of interaction, and what kind of needs and want one is able to realize using the blog. In this work particular interpretational scheme is presented, according to which the blog might find its place among other means and ways of communication. This thesis describes the work process and the results for the development of a newkind of ergonomic cleaning mop, ranging from basic concept to a virtually productionready digital prototype. The project was made in cooperation with, Ömer Altun, whowas also the project supervisor, and whose plan it is to launch this new product onthe market in a foreseeable future. The mop is meant to be used by both professionalcleaning staff and ordinary home users. What makes this product unique is that it hasa movable center piece that simplifies the cleaning under low furniture.The work began with the establishment of a product requirements specification andlater a study of similar products and existing functional models was conducted. Afterthese surveys the work began with refining the existing concepts in order to makethem more complete. In consultation with Ömer, further refining of one of these newconcepts was conducted, but was split into seven smaller steps.After the initial refining process, effort shifted into the investigation and selection ofappropriate materials and manufacturing methods. At this stage, two experimentswere also conducted and subsequently used to support the final decisions concerningthe choice of materials and design.When the materials and methods were adopted, a detailed CAD model of the wholeproduct and related technical drawings was made and also a number of FEM-analyseson all critical details, to ensure that performance requirements were met, wasconducted. These CAD models are closer described in detail in the chapter“Detaljkonstruktion”.In the final sections of discussion and evaluation are suggestions for furtherimprovements. The object of the research – the service marketing of the insurance company „Ergo Lietuva“. The aim of the research – to formulate the strategic aspects of the insurance company‘s „Ergo Lietuva“ services marketing. The AP1000 is two-loop 1100 MWe advanced pressurized water reactor (PWR) that uses passive safety features to enhance plant safety and to provide significant and measurable improvements in plant simplification, reliability, investment protection and plant costs. The AP1000 uses proven technology, which builds on over 30 years of operating PWR experience. The AP1000 final design certification was approved by the NRC in December, 2005. A total of 34 Emergency Operating Procedures (EOPs) for operation of the AP1000 simulator have been prepared based on the AP1000 Emergency Response Guidelines (ERGs), background information documents and detailed plant information. These include 28 EOPs at power and 6 EOPs during shutdown. The AP1000 ERGs were developed by using the generic ERGs for the low pressure reference PWR plant as a basis. The AP1000 design differences from the reference plant were reviewed and reflected in the process of developing operational steps in each ERG. The provisions of the AP1000 PRA were also reviewed and incorporated into the ERGs. Although the AP1000 design does not require operator actions for the first 72 hours after accidents, the operator actions with both safety-related and nonsafety-related equipment have an important role to mitigate the consequence of accidents. In the event of a steam generator tube rupture (SGTR), although the AP1000 is designed so that no operator actions are required to recover from the event, there are actions that can be taken by the operator to limit the release of radioactive effluents from the ruptured SG. These actions include isolation of the ruptured SG and depressurization of the reactor coolant system (RCS) to terminate primary-to-secondary leakage, restoring reactor coolant inventory to ensure adequate core cooling and plant pressure control. It is expected that these operator actions should be incorporated into the ERG to reduce the fission product release. To support the development of the AP1000 ERGs, several transient and accident analyses were performed. These include analyses for LOCA, post-LOCA cooldown and depressurization, passive safety system termination, SGTR and faulted SG isolation. These analyses results were incorporated into the ERG background information documents. In the event of SGTR, several cases were analyzed, including consideration of operator recovery actions. These cases were modeled using the best-estimate state-of-art RELAP5 code. The analyses results show that operator recovery actions are effective for SGTR to be placed under operator control.

A prominent application of knowledge graph (KG) is document enrichment. Existing methods identify mentions of entities in a background KG and enrich documents with entity types and direct relations. We compute an entity relation subgraph (ERG) that can more expressively represent indirect relations among a set of mentioned entities. To find compact, representative, and relevant ERGs for effective enrichment, we propose an efficient best-first search algorithm to solve a new combinatorial optimization problem that achieves a trade-off between representativeness and compactness, and then we exploit ontological knowledge to rank ERGs by entity-based document-KG and intra-KG relevance. Extensive experiments and user studies show the promising performance of our approach.

Abstract (sommario):
The foundation for new Emergency Operating Instructions (EOIs) being developed at several plants in the Ukraine is the Westinghouse Owners Group (WOG) Emergency Response Guidelines (ERGs) developed in the U.S. The ERGs were chosen as a base for the new EOIs for several reasons. First the overall structure and format was adaptable to VVER Pressurized Water Reactor (PWR) designs. Second, the ERGs have served as a base for many plant EOIs in both the U.S. and internationally. Third, key information supporting the ERGs was available. This paper describes the method used at one of the Ukrainian plants to provide an analytical justification for their EOIs. The method being employed by a second plant is very similar, differing only slightly in how it is implemented. The WOG ERG development program, which started shortly after the accident at Three Mile Island Unit 2, used many sources of technical information on plant and system transient response, which were available in support of the plant design and licensing efforts. In addition, operating experience from many operating PWR plants in the U.S. and around the world was used. For example, design basis accident (DBA) analyses, documented in a plant’s Safety Analysis Report (SAR) and other design documents, had been performed by Nuclear Steam Supply System (NSSS) vendors, utilities, or the Architect/Engineer. All relevant sources were considered in the development of the ERGs. Limited Probabilistic Risk Assessment (PRA) analyses were available during that time period. When a technical basis for a recovery strategy and associated operator actions was not available, an analysis was defined and performed. In general, these analyses were performed on a generic basis, and addressed the different categories of design (e.g., number of reactor coolant loops and/or low/high pressure safety injection system design). U.S. Nuclear Power plants that were in the WOG program were responsible for implementing the generic ERGs. This required the utilities to review the generic analyses to ensure that they were applicable and to justify any deviations from the ERG methodology. Modern PRA analyses are similar to the analyses supporting the ERGs since they address multiple failures and assume better estimate or expected assumptions for equipment availability and operator performance. The process being employed by Ukrainian plants is similar to the WOG. That is, available analyses and operating experience are being reviewed and pertinent information extracted to assist in the analytical justification of the EOIs. This includes the use of recently updated PRA and DBA analyses, other “original” design information and operating experience. A systematic review of the EOIs is being conducted to identify items requiring analytical justification. For each analysis identified, the specific purpose of the analysis is being documented. The analysis needs are then compared to the available analyses and operating experience. From this review, new analyses needed to justify the EOIs are developed, and a basis for using existing analyses is established. The work is being conducted in two phases. The first phase performs all of the reviews and assessments necessary to determine the new analyses required to justify the EOIs. In the second phase, these new analyses will be conducted and documented, and the EOI Analytical Justification (AJ) report will be written.

The gas engine-gas turbine combined cycle was developed as the ETCS (Engine Turbo Compound System) that has a potential to be a future high performance combined cycle. The reciprocating engine operates with a maximum pressure and temperature in the cylinder, higher than that of the conventional gas turbines. When the gas engine is integrated with a gas turbine instead of a turbocharger, the concept of the ETCS with ERGT (Engine Reheat Gas Turbine) is available. In order to attain a better ETCS performance, a natural gas firing RGT (Regenerative Gas Turbine) was selected as the core gas turbine. For the system integration, the recuperator of RGT was exchanged for a gas engine. Focusing on the effect of engine exhaust temperature, we found that the ETCS cycle with ERGT has the potential to achieve a higher thermal efficiency than that of a re-generative cycle gas turbine with no change of TIT (Turbine Inlet Temperature). The engine exhaust temperature of 900°C increases the system power generation efficiency from 39% of RGT to 45% in ERGT (GT-Gas Engine) and up to 59% in ETCS (GT-Gas Engine-Steam Turbine) (TIT 1200°C).
