Testo completoThe diagnosis of Lyme disease, caused by tick-borne Borrelia bacteria, is difficult due to their low concentration in clinical samples, as well as the existence of several pathogenic species. The recommended diagnostic method is an indirect serological approach, which is associated with the risk of both false-positive and false-negative results. The aim of this PhD work was to develop a methodological proof-of-concept for the ultra-sensitive and highly specific detection of Borrelia burgdorferi using functionalized nanoparticles and inductively coupled plasma mass spectrometry (ICP-MS) analysis. Nanoparticle synthesis and functionalization were carried out and optimized using surface analysis techniques such as XPS and Auger spectroscopy. Several analytical routes were followed, based on the use of different particle types combined with a panel of ICP-MS experimental conditions enabling significant specificity and sensitivity for Borrelia burgdorferi from buffer solutions or artificial blood samples used as model blood matrix
