Recent advances in tissue engineering and regenerative medicine offer a long-term solution through biological repair, replacement of damaged teeth or maintenance and improvement of tissue and organ function through the use of stem cells. Stem cells or also called universal cells, progenitor cells or precursor cells; they are primitive, undifferentiated, clonogenic cells that are characterized by their self-renewal capabilities and that can be differentiated into more specialized cells with specific functions. Currently many sources are known from where you can obtain stem cells, one of which are those obtained from oral or dental tissues, called dental stem cells (DSC), from where it has been possible to identify, isolate and characterize around 8 unique populations: dental pulp stem cells (DPSC), human exfoliated deciduous tooth stem cells (SHED), periodontal ligament stem cells (PLDSC), dental follicle stem cells (DFSC), stem cells derived from bone alveolar (CMHA), the stem cells of the apical papilla (SCAP), the stem cells of the dental germ (DGSC) and the gingival stem cells (GSC). These DSC have attracted attention in recent years due to their accessibility, plasticity and high proliferation capacity. Currently, DSC have shown that they can be used in endodontic and periodontal regenerative therapy, in the regeneration of dentin and bone and in dental bioengineering. Tissue engineering methodologies combined with a greater understanding of the biology of DSCs will provide powerful tools for a broader spectrum of their application in various future therapeutic strategies.

In this paper COM+ based on Microsoft .NET is adopted to realize a Distributed Collaborative System (DCS). This system adopts a complex architecture combined C/S with B/S to reduce the remote communication and improve the performance. By means of IE or other browsers, a client can download client tool, register or login, search or publish information via the collaborative website. After installing the client tool, a client can automatically receive the collaborative requests or invitations from the collaborative server and collaborate with other participators. The system utilizes COM+ services to realize the Distributed Synchronous Communication (DSC) and Distributed Asynchronous Communication (DAC). Loosely Coupled Event (LCE) service can make DSC true easily and effectively. Transaction service, Queued Component (QC) service and Windows NT Service can be applied to realize DAC efficiently and reliably.

This study aims to discuss the effectiveness of “Demand-Supply Coordinated control” in reducing the power consumption required for conveyance by the heat transport medium in District Cooling and Heating (DHC) systems. The problem with DHC systems is that increased conveyance power is required to provide heating to consumers. As one of the measures to solve this problem, Demand-Supply Coordinated (DSC) control is introduced; however, its effectiveness and limitations have not been clarified so far. In this paper, first, the fundamental characteristics of a DHC system under DSC control are numerically examined. The results showed that the conveyance power consumption of DHC systems under DSC control can be classified into three regions, depending on the relative rate of demand change against the load-following capability of the heat source. Next, the authors compared the conveyance power of DSC control with that of Constant Supply Pressure (CSP) control adopted in conventional DHC, and showed regularity for each of the three regions mentioned above. Finally, the authors show the appearance frequency of these three regions of the practical DHC system under a real heat load in Japan. The results showed that the conveyance power required for DSC control is markedly lower than that of CSP control.

Abstract Direct Bonded Copper (DBC) is the most popular solution for conventional high-power modules because of superior thermal/electrical/mechanical performance and mature manufacturing. To meet the rising demand of power density and power rating, a Double-Sided Cooled (DSC) sandwich structure using dual insulated metal-clad substrates was proposed and DBC still dominated the substrate selection of DSC power modules. However, there are several long-existing reliability challenges of conventional DBC-based power modules and the cost of DBC is relatively high compared with organic and metal (e.g. lead frame) substrates. This study proposes a DSC 1.2 kV half-bridge power module using dual epoxy-resin Insulated Metal Substrate (eIMS) for solving DBC-based power module issues and providing a cost-effective solution. The thermal performance outperforms traditional Alumina (Al2O3) DBC-based DSC power module due to moderate thermal conductivity (10 W/mK) and thin (120 μm) epoxy-resin composite dielectric layer compared with Alumina. The breakdown voltage of this high thermally conductive organic dielectric is 5 kVAC (@ 120 μm) and the Glass Transition Temperature (Tg) is 300°C which is indispensable for Wide-Band-Gap (WBG) devices and high-power applications. In terms of thermal-mechanical reliability, the organic-based DSC power module can pass the thermal cycling test over 2000 cycles by optimizing the mechanical properties of the encapsulant material. In conclusion, this paper not only proposes a competitive organic-based power module but also a methodology of evaluation for thermal and mechanical performance.

Differential scanning calorimetry (DSC) has been used to estimate the effect of disc disease on the collagen helix-coil transition and morphology for tissue extracted from patients during surgical operation. Forty discs were obtained from patients with degenerative disc disease undergoing surgery for low back pain. The patients were in the age between 20 and 70 years old. The specimens were kept wet during DSC experiment. The data allow the comparison between thermal stability of collagen tissue from healthy patients and from patients suffering from disc disease. In the paper the comparison between thermal helix-coil transition for collagen fibers from patients suffering from disc disease and collagen fibers from healthy organisms has been discussed. The heating rate has an influence on the position on denaturation temperatures of collagen in disc tissues. Higher helix-coil transition temperature of collagen in degenerated disc suggests that additional intermolecular cross linking of collagen fibers occurs. Denaturation temperatures of collagen in degenerated male disc possess smaller values than in female ones. Disc disease induces changes in collagen structure and leads to formation of additional crosslinks between collagen fibers.

The disc stack centrifuge (DSC) in the virgin olive oil (VOO) industry is widely used for oil purification at a final stage of extraction process. The obtained oil should be commercialized directly as turbid or sent to settling and/or filtration. Despite technology is strongly consolidated, few literature is available on the effect of DSC on efficiency and quality of VOO, specially compared with alternative methods. Alternative technologies to DSC like settling or direct filtration are suitable only for very small production scale. Application of advanced technology already available for DSC like hermetic design is applicable also to VOO but more research is necessary to establish effect on quality and cost/benefit ratio. Improvement of installation module and a more appropriate use of DSC on VOO industry are recommended to reduce water consumption and consequent loss of VOO quality.

Lidar is a key sensor of autonomous driving systems, but the spatial distribution of its point cloud is uneven because of its scanning mechanism, which greatly degrades the clustering performance of the traditional density-based spatial clustering of application with noise (DSC). Considering the outline feature of detected objects for intelligent vehicles, a DSC-based adaptive clustering method (DAC) is proposed with the adoption of an elliptic neighborhood, which is designed according to the distribution properties of the point cloud. The parameters of the ellipse are adaptively adjusted with the location of the sample point to deal with the uniformity of points in different ranges. Furthermore, the dependence among different parameters of DAC is analyzed, and the parameters are numerically optimized with the KITTI dataset by considering comprehensive performance. To verify the effectiveness, a comparative experiment was conducted with a vehicle equipped with three IBEO LUX8 lidars on campus, and the results show that compared with DSC using a circular neighborhood, DAC has a better clustering performance and can notably reduce the rate of over-segmentation and under-segmentation.

The mechanical properties after age hardening heat treatment and the kinetics of related phase transformations of high strength AlZnMgCu alloy AA 7068 were investigated. The experimental work includes differential scanning calorimetry (DSC), differential fast scanning calorimetry (DFSC), sophisticated differential dilatometry (DIL), scanning electron microscopy (SEM), as well as hardness and tensile tests. For the kinetic analysis of quench induced precipitation by dilatometry new metrological methods and evaluation procedures were established. Using DSC, dissolution behaviour during heating to solution annealing temperature was investigated. These experiments allowed for identification of the appropriate temperature and duration for the solution heat treatment. Continuous cooling experiments in DSC, DFSC, and DIL determined the kinetics of quench induced precipitation. DSC and DIL revealed several overlapping precipitation reactions. The critical cooling rate for a complete supersaturation of the solid solution has been identified to be 600 to 800 K/s. At slightly subcritical cooling rates quench induced precipitation results in a direct hardening effect resulting in a technological critical cooling rate of about 100 K/s, i.e., the hardness after ageing reaches a saturation level for cooling rates faster than 100 K/s. Maximum yield strength of above 600 MPa and tensile strength of up to 650 MPa were attained.

We have used Segment Anything model (SAM) by Meta AI and based fine-tuned model MedSAM for segmentation of optic disc in retinal fundus images. We have used Indian Diabetic Retinopathy dataset (IDRiD) segmentation part. It consists of 81 original colour fundus images in jpg files split into train and test set. Ground-truth images for the lesions (Microaneurysms, Haemorrhages, Hard Exudates and Soft Exudates divided into train and test set - TIF Files) and Optic Disc (divided into train and test set - TIF Files). This dataset is feed into the deep learning neural network model; Segment Anything Model (SAM) and MedSAM for the image segmentation task. The Dice Similarity Coefficient (DSC) is used in the experiment for observing the model performance. We have used 80% for training and 20% for testing of Indian Diabetic Retinopathy Dataset (IDRiD) in both models. After 100 epochs with 32 batch size it is observed that average Dice Similarity Coefficient (DSC) of SAM and MedSAM models in the optic disc segmentation task are, 85.97 % and 90.15 %. Also, in finding it is observed that SAM and MedSAM both have very low DSC where the fundus images are having very high brightness.

Arenga pinnata starch (APS) production from APS industrial centers in Klaten, Indonesia, produces Arenga pinnata starch mill effluent (APSME). This waste will undoubtedly harm the environment, significantly reducing river water quality. On the other hand, in Gunungpati, Semarang, Indonesia, durian fruit is abundant, it leaves durian skin waste, which may cause environmental pollution. However, durian skin contains cellulose, which has the potential to be valorized for various applications. Therefore, this study evaluated the preparation of natural coagulant from durian skin cellulose crosslinked with glutaraldehyde for treating APSME from APS industrial centers in Klaten, Indonesia. Durian skin flour (DSF), durian skin cellulose (DSC), and glutaraldehyde-crosslinked durian skin cellulose (DSC-G) coagulants were characterized by proximate composition, UV-Vis spectroscopy, Fourier-transform infrared (FTIR) spectroscopy and scanning electron microscopy (SEM). The effects of coagulant dose, pH, and mixing speed on removal efficiency and sludge volume in DSF, DSC, and DSC-G were compared with polyaluminum chloride (PAC). The study assessed the process efficiency in terms of percentage removals for chemical oxygen demand (COD), biological oxygen demand (BOD5), total dissolved solids (TDS), and total suspended solids (TSS) as 71.38%, 78.23%, 94.79%, and 96.12%, respectively, with a percentage sludge volume of 24%. The results indicated that the optimum DSC-G dosage was 2500 mg/L with an optimum working area of pH at 5.5 and a mixing speed of 90 rpm. DCS-G has a floc stability of -12.33 mV. This study indicated that DSC-G has the potential to be used as a coagulant for the treatment of APSME.

A series of Co(RR′dsc)3 and [Co2(RR′dsc)5]+ complexes (R, R′ = two alkyl groups or one heterocyclic group; dsc = NCSe2) have been synthesized and their redox behaviour, chemical reactivity and spectroscopic properties compared with the corresponding dithiocarbamate (RR′dtc) complexes. Electrochemical oxidation of Co(RR′dsc)3 in dichloromethane at platinum electrodes occurs at potentials about 0.34 V less positive than for Co(RR′dsc)3. The formally cobalt(IV) complexes [Co(RR′dsc)3]+ can be identified as a product which is then converted into [Co2(RR′dsc)5]+ via dimerization and an internal redox reaction. Despite the enhanced thermodynamic stability implied by the redox potentials, [Co(RR′dsc)3]+ has similar kinetic stability to the analogous dithiocarbamate complexes. Co(RR′dsc)3 is reduced at fairly negative potentials on both platinum and mercury electrodes with extremely rapid loss of [RR′dsc]-. [Co(RR′dsc)3]- is therefore thermodynamically and kinetically more unstable than [Co(RR′dtc)3]- . The [Co2(RR′dsc)5]+ complexes are also more readily oxidized and harder to reduce than the sulfur analogues. Oxidation of [Co2(RR′dsc)5]+ produces [Co2(RR′dsc)5]2+ at low temperatures and fast scan rates, but no stable reduced form of the dimer is accessible on the voltammetric time scale examined. The reduction process for the dimer is consistent with the reaction [Co2(RR′dsc)5]+ +e- → Co(RR′dsc)3+ Co(RR dsc)2. Electrochemical oxidation data obtained at mercury electrodes for the diselenocarbamate complexes are complicated by adsorption but are similar to that found at platinum electrodes. This contrasts with the dithiocarbamates where a mercury electrode specific pathway is observed. Cobalt-59 n.m.r. spectroscopy in dichloromethane shows the non- equivalence of the two cobalt atoms in [Co2(RR′dsc)5]+. The chemical shifts for Co(RR′dsc)3 complexes exhibit similar substituent effects to the dithiocarbamates in cobalt-59 n.m.r. measurements as was the case in oxidative electrochemistry. Cobalt-59 n.m.r. spectroscopy and mass spectrometry demonstrate that exchange, substitution and redox reactions can lead to the formation of mixed ligand diselenocarbamate complexes and mixed dithiocarbamate/diselenocarbamate complexes for both the cobalt(III) monomers and dimers.

AbstractNovel types of tandem solar cells (TSC) based on dye-sensitized (DSC) and perovskite (PSC) solar cells including DSC/DSC and DSC/PSC configurations with a common counter electrode were fabricated and investigated. The measurements of PV parameters for tandem solar cells under AM1.5 light intensity conditions have shown that the highest power conversion efficiency (PCE) of 14.5% was obtained for the DSC/DSC tandem configuration. At the same time, investigations of DSC/PSC tandem solar cells demonstrated the prospective benefits of this tandem system for obtaining high PCE values.

Disturbed state concept theory (DSC) utilizes constitutive models to depict material responses. The response is gained by weight-averaging reference responses. DSC also can superpose disturbance factor on reference responses to reach actual response values. Suitable constitutive models give reference responses and DSC revises them to be acceptable outputs. Disturbance factor is independent to constitutive model. On DSC application on seepage problem, combination of DSC and Biot’s consolidation is forwarded by introducing disturbance factor into equilibrium function and extended cam-bridge model provides reference response. On DSC application on anisotropy, anisotropic disturbance factor is applied on to obtain stress anisotropy. Hierarchical single surface model is utilized to provide reference response for further anisotropic revision. Detailed formula derivation for DSC-seepage or for DSC-anisotropy is accomplished. On both applications, the key combination of DSC and other theory depends on the introduction of disturbance factor into equilibrium equation. Outcomes of DSC show readjustment of parameters of disturbance factor guaranties quantity of response descriptions. DSC takes into account of outside effects without changing inside. Existing models and existing methods can be used to describe newly responses.

Abstract Silicon-Carbide (SiC) power devices have become a promising option for traditional Silicon (Si) due to the superior material properties. To fully take advantage of the SiC devices, a high-performance power device packaging solution is necessary. This study proposes a cost-effective double-sided cooled (DSC) 1.2 kV SiC half-bridge power module using organic epoxy-resin composite dielectric (ERCD) substrates. The high mechanical and thermal performance of the power module is achieved by the low-modulus, moderate thermal conductivity, and relatively thin (120 μm) layer of ERCD material compared with traditional metal-clad ceramic approaches. This novel organic dielectric can withstand high voltage (5 kV @ 120 μm) and operate up to 250°C continuously, which is indispensable for high power applications. The thermal modeling results show that the equivalent thermal resistance junction-to-case (Rjc_eq) of the DSC power module using dual direct bonded copper (DBC) is 17% higher than the dual ERCD configuration. Furthermore, a non-insulated DSC power module concept is proposed for maximizing thermal performance by considering thermal vias in the ERCD substrate and direct-soldered heat sink. A thought process for optimization of thermal via design is demonstrated and it shows up to 24% of improvement on thermal performance compared with the insulated DSC power module.

Abstract This systematic review evaluated if different cryopreservation protocols could affect biological properties (Cell survival rate (CSR), proliferation, differentiation, maintenance of stem cell markers) of stem cells obtained from dental tissues (DSC) post-thaw. An electronic search was carried out within PubMed and ISI Web Science by using specific keyword. Two independent reviewers read the titles and abstracts of all reports respecting predetermined inclusion/exclusion criteria. Data were extracted considering the biological properties of previously cryopreserved DSCs and previously cryopreserved dental tissues. DSCs cryopreserved as soon as possible after their isolation presents a CSR quite similar to the non-cryopreserved DSC. Dimethyl sulfoxide (DMSO) [10%] showed good results related to cell recovery post-thaw to cryopreserve cells and tissues for periods of up to 2 years. The cryopreservation of DSC in a mechanical freezer (-80°C) allows the recovery of stem cells post-thaw. The facilities producing magnetic field (MF), demand a lower concentration of cryoprotectant, but their use is not dispensable. It is possible to isolate and cryopreserve dental pulp stem cell (DPSC) from healthy and diseased vital teeth. Cryopreservation of dental tissues for late DSC isolation, combined with MF dispensability, could be valuable to reduce costs and improve the logistics to develop teeth banks.

Decentralized stochastic control (DSC) is a stochastic optimal control problem consisting of multiple controllers. DSC assumes that each controller is unable to accurately observe the target system and the other controllers. This setup results in two difficulties in DSC; one is that each controller has to memorize the infinite-dimensional observation history, which is not practical, because the memory of the actual controllers is limited. The other is that the reduction of infinite-dimensional sequential Bayesian estimation to finite-dimensional Kalman filter is impossible in general DSC, even for linear-quadratic-Gaussian (LQG) problems. In order to address these issues, we propose an alternative theoretical framework to DSC—memory-limited DSC (ML-DSC). ML-DSC explicitly formulates the finite-dimensional memories of the controllers. Each controller is jointly optimized to compress the infinite-dimensional observation history into the prescribed finite-dimensional memory and to determine the control based on it. Therefore, ML-DSC can be a practical formulation for actual memory-limited controllers. We demonstrate how ML-DSC works in the LQG problem. The conventional DSC cannot be solved except in the special LQG problems where the information the controllers have is independent or partially nested. We show that ML-DSC can be solved in more general LQG problems where the interaction among the controllers is not restricted.

China is a nation that is affected by a multitude of natural disasters, including droughts and floods. In this paper, the variations of extreme drought and pluvial patterns and their relations to the large-scale atmospheric circulation have been analyzed based on monthly precipitation data from 483 stations during the period 1958–2010 in China. The results show the following:(1)the extreme drought and pluvial events in China increase significantly during that period. During 1959–1966 timeframe, more droughts occur in South China and more pluvial events are found in North China (DSC-PNC pattern); as for the period 1997–2003 (PSC-DNC pattern), the situation is the opposite.(2)There are good relationships among the extreme drought and pluvial events and the Western Pacific Subtropical High, meridional atmospheric moisture flux, atmospheric moisture content, and summer precipitation.(3)A cyclone atmospheric circulation anomaly occurs in North China, followed by an obvious negative height anomaly and a southern wind anomaly at 850 hPa and 500 hPa for the DSC-PNC pattern during the summer, and a massive ascending airflow from South China extends to North China at ~50∘N. As for the PSC-DNC pattern, the situation contrasts sharply with the DSC-PNC pattern.

Emerging digital transformation in industry is noticeable among others in Supply Chain Management (SCM). For instance, applying new-generation digitalized technologies in the Dairy Supply Chain (DSC) enables an increase of manufacturing productivity, improves planning and forecasting, and also enhances competitive capabilities according to Industry 4.0 assumptions. It is worth mentioning, that in modern DSC, high visibility of raw materials, components, products, and processes by all contributors on all stages of DSC is crucial. This article focuses on the transparency aspect of the DSC supported by IoT-based technologies enabling interoperability among all DSC participants. The paper addresses the problem of effective integration of heterogeneous data sources, i.e., deployed new technological IoT solutions with traditional SCM systems and a third-party software component. The main objective of this report is to propose the IoT-based DSC model comprising four chain stages: milk production, milk transportation, milk processing, and dairy products distribution. Moreover, the comprehensive DSC domain ontology as a knowledge model is formulated and described. The ontology aims on improvement of the DSC management efficiency by facilitating interoperability within DSC. The applicability of the proposed ontological model is verified using a sustainable-oriented case study, which estimates the environmental footprint at the milk transportation stage of the DSC.

Introducción: la asociación entre infección e inflamación se considera la causa principal de complicaciones como la pre-eclampsia. Las células deciduales estromales (DSC) constituyen el principal componente celular de la decidua. Las DSC son las principales protectoras frente a bacterias Gram negativas y señales de peligro que emanan de inflamación severa con o sin infección. Por medio de la expresión de TLR4 las DSC regulan la repuesta inmune durante la infección. Objetivo: Determinar la expresión de receptores TLR4, así como citoquinas producidas por las DSC de pacientes con pre-eclampsia, que podrían actuar como marcadores diagnósticos de activación inmunológica. Materiales y métodos: Estudio descriptivo. Las DSC fueron aisladas y cultivadas a partir de tejido placentario, de pacientes con parto vaginal sin complicaciones (PVSC, n=6), parto por cesárea electiva (PCE, n=6) y parto por cesárea con pre-eclampsia (PCPE, n=6). La caracterización fenotípica de las DSC se efectuó mediante análisis por citometría de flujo. La determinación de citoquinas se realizó mediante ELISAs. La expresión génica de TLR4 se valoró por qRT-PCR. Resultados: la pureza de las DSC se determinó por la expresión de antígenos característicos como: CD10, CD44, CD73 y CD90. La ausencia de CD45 y CD62p, descartó la contaminación con células leucocitarias y células epiteliales. La secreción de IL-6 por las DSC fue mayor en PVSC, que en PCE y PCPE. La expresión génica del TLR4 fue similar en las DSC. Conclusiones: Se confirmó la pureza de las DSC. La expresión génica de TLR4 en las DSC de PVSC, PCE y PCPE, no presentó diferencias significativas. Las DSC secretan IL-6 e IL-10.

In this study, inclusion complexes of DDC-Cu with hydroxypropyl beta-cyclodextrin (HP) or sulfobutyl ether beta-cyclodextrin (SBE) were prepared and investigated as an approach to enhance the apparent solubility of DDC-Cu. Formulations were prepared by simple mixing of DDC-Cu with both cyclodextrin (CDs) at room temperature. Phase solubility assessments of the resulting solutions were performed. DDC-Cu CD solutions were freeze-dried for further characterisations by DSC, thermogravimetric analysis (TGA) and FT-IR. Stability and cytotoxicity studies were also performed to investigate the maintenance of DDC-Cu anticancer activity. The phase solubility profile deviated positively from the linearity (Ap type) showing significant solubility enhancement of the DDC-Cu in both CD solutions (approximately 4 mg/mL at 20% w/w CD solutions). The DSC and TGA analysis confirmed the solid solution status of DDC-Cu in CD. The resulting solutions of DDC-Cu were stable for 28 days and conveyed the anticancer activity of DDC-Cu on chemoresistant triple negative breast cancer cell lines, with IC50 values less than 200 nM. Overall, cyclodextrin DDC-Cu complexes offer a great potential for anticancer applications, as evidenced by their very positive effects against chemoresistant triple negative breast cancer cells.

Background: Delayed sternal closure (DSC) has been used for patients who develop bleeding, chest wall edema, and malignant arrhythmia following cardiopulmonary bypass. Multiple factors can influence the timing of when to perform DSC. We aimed to describe our DSC experience in neonates and infants by comparing outcomes between patients undergoing early (<48 hours) versus late DSC (> 48 hours). We explored the associations between specific clinical and laboratory variables and the timing of DSC. Methods: Retrospective chart review of neonates and infants (<one-year-old) with DSC after heart surgery from December 2012 to December 2018. Patients requiring extracorporeal membrane oxygenation were excluded. Results: A total of 121 patients were included in the analysis, 34% (n = 41) met late DSC criteria. The overall cohort had a 75% survival rate and a median time for open sternum of 42.5 hours (Q1:23-Q3:65). The median time for open sternum in the early and late DSC groups was 24 hours (Q1:21-Q3:43) and 93 hours (Q1:65-Q3:141), respectively ( P < .01). There was no statistical difference in mortality rate between groups. Patients with late DSC endured longer intensive care unit stays (median 24.3 days [Q1:13-Q3:35.3] vs 36.8 [Q1:23.9, 73.6]; P< .01) and a two-fold longer hospital stay compared to the early DSC group (multivariable analysis: relative risk = 2, 95% CI: 1.5-2.7; P < .01). Univariate analysis revealed patients with late DSC had higher median lactates both intraoperatively (7.6 [Q1:5.9-Q3:10.7] vs 9.3 [Q1:7.5-Q3:12.1]; P < .01) and 24 hours postoperatively (6.5 [Q1:4.3-Q3:10.3] vs 8.7 [Q1:5.7-Q3:14.70]; P = .03). A higher vasoactive inotrope score at 36 hours was associated with late DSC (odds ratio = 1.1, 95% CI: 1.01-1.2; P = .02). Conclusions: Future research that explores additional clinical and laboratory variables that can help guide DSC decision-making and timing is needed.

In wild-type HIV-1-infected H9 cells, the DRC fractionated with the soluble cytoplasmic fraction, whereas the DSC was associated with the membrane fraction. The DRC was also detected in the cytoplasmic fraction in H9 cells expressing HIV-1 Myr− mutant Gag. However, little of the unmyristylated Gag and Gag-Pol proteins was found in the membrane fraction. Furthermore, HIV-1 Gag proteins synthesized in vitro in a rabbit reticulocyte lysate system in the absence of exogenous lipid membrane were able to assemble into a viral Gag complex similar to that of the DRC identified in infected H9 cells. The density of the viral Gag complex was not altered by treatment with the nonionic detergent Triton X-100, suggesting a lack of association of this complex with endogenous lipid. Formation of the DRC was not significantly affected by mutations in assembly domains M and L of the Gag protein but was drastically inhibited by a mutation in the assembly I domain. Purified DRC could be disrupted by high-salt treatment, suggesting electrostatic interactions are important for stabilizing the DRC. The Gag precursor proteins in the DRC were more sensitive to trypsin digestion than those in the DSC. These findings suggest that HIV-1 Gag and Gag-Pol precursors assemble into DRC in the cytoplasm, a process which requires the protein-protein interaction domain (I) in NCp7; subsequently, the DRC is transported to the plasma membrane through a process mediated by the M domain of the matrix protein. It appears that during this process, a conformational change might occur in the DRC either before or after its association with the plasma membrane, and this change is followed by the detection of virus budding structure at the plasma membrane.
