Integrins constitute a family of membrane-spanning, heterodimeric proteins that mediate adhesive interactions between cells and surrounding extracellular matrices (or other cells) and participate in signal transduction. We are interested in assessing integrin functions in the context of developing Drosophila melanogaster. This report, using mutants of the beta PS subunit encoded by the myospheroid (mys) locus, analyzes the relationships between integrin protein structure and developmental functions in an intact organism. As a first step in this analysis, we demonstrated the ability of a fragment of wild-type mys genomic DNA, introduced into the germ line in a P-element vector P[mys+], to rescue phenotypes attributed to lack of (or defects in) the endogenous beta PS during several discrete morphogenetic events. We then produced in vitro a series of modifications of the wild-type P[mys+] transposon, which encode beta PS derivatives with mutations within the small and highly conserved cytoplasmic domain. In vivo analysis of these mutant transposons led to the following conclusions. (1) The cytoplasmic tail of beta PS is essential for all developmental functions of the protein that were assayed. (2) An intron at a conserved position in the DNA sequence encoding the cytoplasmic tail is thought to participate in important alternative splicing events in vertebrate beta integrin subunit genes, but is not required for the developmental functions of the mys gene assayed here. (3) Phosphorylation on two conserved tyrosines found in the C terminus of the beta PS cytoplasmic tail is not necessary for the tested developmental functions. (4) Four highly conserved amino acid residues found in the N-terminal portion of the cytoplasmic tail are important but not critical for the developmental functions of beta PS; furthermore, the efficiencies with which these mutant proteins function during different morphogenetic processes vary greatly, strongly suggesting that the cytoplasmic interactions involving PS integrins are developmentally modulated.

This rejoinder acknowledges the empirical gaps and theoretical/theological disharmony highlighted in the three selected commentaries on Place Spirituality (PS), but we defend our central argument about the developmental pathways of PS. First, we provide an overview of recent studies on PS, highlighting what has been done so far in the field. Second, we draw from the commentaries to advance the understanding of PS in relation to three world religions: Islam, Christianity and Hinduism. Third, we evaluate the normative aspects of PS as a transactional versus transitional phenomenon. Finally, we defend the two contested developmental pathways to PS, involving the compensation and correspondence working models of attachment, while complementing these models using the motivational systems framework. We maintain that these models are relevant for understanding the relationship between religious attachment and place attachment among religious and non-religious people. Recommendations for further studies are made in relation to the broader implications of PS.

Presenilina-1 (PS1), el component catalític de γ-secretasa que regula el processament de múltiples proteïnes transmembrana, es troba mutada en la majoria de casos d’Alzheimer familiar (FAD). Evidències recents indiquen que mutacions en PS1 lligades a FAD redueixen el processament de múltiples proteïnes transmembrana, suggerint un mecanisme de pèrdua de funció. De fet, la inactivació de PS1 durant l’embriogènesi comporta defectes morfològics, mentre que la inactivació genètica d’ambdues PS en el cervell adult causa defectes en memòria dependents d’edat i neurodegeneració. A més, la participació de PS en la proteòlisi de molècules de senyalització implicades en el desenvolupament del sistema nerviós, incloent ErbB4, suggereix que aquestes vies de senyalització podrien contribuir a la neurodegeneració. En aquesta tesi doctoral hem estudiat el paper del processament dependent de PS1/γ‑secretasa d’EphA3 i de la senyalització Nrg1/ErbB4 en el desenvolupament neuronal i la neurodegeneració. Els nostres resultats mostren que PS1/γ-secretasa és necessària pel creixement axonal en el cervell en desenvolupament. PS1/γ-secretasa regula l’elongació axonal a través de l’escissió d’EphA3 en la tirosina 560 que resulta en la generació d’un fragment ICD. EphA3 ICD regula negativament RhoA, interacciona amb la miosina IIA no-muscular (NMIIA) i incrementa la seva fosforilació (S1943) produint el desmuntatge del filament i el creixement axonal. De manera contrària a la senyalització clàssica ephrin-EphA3, la senyalització d’EphA3 dependent de PS/γ-secretasa és independent de lligand. Aquest resultat suggereix per primer cop papers oposats d’EphA3 inhibint (dependent de lligand) o promovent (processament dependent de PS1/γ-secretasa) el creixement axonal en neurones. En segon lloc, mostrem que PS1/γ-secretasa regula l’expressió de Nrg1 tipus III i el processament de Nrg1 tipus III i ErbB4, i regula negativament la sinaptogènesi a través de Nrg1. En conjunt, els nostres resultats mostren que PS1/γ-secretasa regula el creixement axonal i la sinaptogènesi a través de la regulació de la senyalització d’EphA3 independent de lligand i del processament/senyalització de Nrg1/ErbB4, respectivament. La nostra investigació obre el camí a explorar noves relacions entre el neurodesenvolupament i la neurodegeneració, proporcionant evidències sobre l’existència d’una comunicació entre les vies de senyalització implicades en aquests processos.
Presenilin-1 (PS1), the catalytic component of γ-secretase that regulates the processing of multiple transmembrane proteins is mutated in the majority of cases of familial Alzheimer’s disease (FAD). Recent evidence indicates that FAD-linked PS1 mutations reduce the γ-secretase cleavage of several transmembrane proteins, suggesting a loss-of-function mechanism. Indeed, PS1 inactivation during embryogenesis leads to morphological defects, whereas genetic inactivation of both PS in the adult brain causes age-dependent memory impairments and neurodegeneration. Moreover, the participation of PS in the proteolysis of signaling molecules involved in the development of nervous system, including ErbB4, suggest that these signaling pathways could contribute to neurodegeneration. In this doctoral thesis we have studied the role of PS1/γ-secretase-dependent cleavage of EphA3 and Nrg1/ErbB4 signaling in neuronal development and neurodegeneration. Our results show that PS1/γ-secretase is required for axon growth in the developing brain. PS1/γ-secretase mediates axon elongation through the cleavage of EphA3 at Tyr560 resulting in the generation of an ICD fragment. EphA3 ICD regulates negatively RhoA, and interacts with and increases phosphorylation (S1943) of non-muscle myosin IIA (NMIIA) leading to filament disassembly and axon growth. In contrast to the classical ephrin/EphA3 signaling, PS/γ-secretase-dependent EphA3 signaling is independent of ligand. This result suggests for the first time opposite roles of EphA3 on inhibiting (ligand-dependent) and enhancing (PS/γ-secretase-dependent processing) axon growth in neurons. Second, we show that PS/γ-secretase regulates Nrg1 type III expression, mediates the processing of Nrg1 type III and ErbB4 and regulates negatively synaptogenesis through Nrg1. Taken together, our results show that PS1/γ-secretase regulates axon growth and synaptogenesis by regulating ligand-independent EphA3 signalling and Nrg1/ErbB4 processing/signalling, respectively. Our investigation paves the way for exploring new relationships between neurodevelopment and neurodegeneration, providing insights of the existence of a crosstalk among the signaling pathways involved in these processes.

This master thesis was performed at the Mechanical department at Outotec AB,Skellefte˚a. Outotec AB is a technology company as well as a project company, sell-ing complex mining technology and plant projects. Outotec’s technologies are usedfor applications such as producing base metals, processing iron ore etc. Outotec isrunning a program called ‘Fully automated smelter’ where manual hazardous workmust be avoided. A machine which automatically replaces punching bars on thepunching machine is needed. This thesis project aimed to investigate if the punching bar changer will be aproprietary purpose-build machine or an industrial robot. The five-step conceptgeneration method was used for concept development during this thesis project.For gathering knowledge and information about the project area, experts have beeninterviewed to obtain information for the start of the project. The gathered infor-mation and knowledge from interviews and literature studies have been used as abase when the concepts were generated. Seven concepts were generated and evalu-ated using a screening matrix. The concept that best satisfied the set requirementswere further developed, designed and simulated. The two scenarios have been in-vestigated and a design concept of the proprietary purpose-built machine has beenpresented. The result from concept evaluation and simulation indicated that bothalternatives are applicable in this area. But to make a decision a more in-depthcalculation of the associated cost and investigation of the design is necessary.

A global maximum power point tracking (MPPT) algorithm hybrid based on Particle Swarm Fireworks (PS-FW) algorithm is proposed which is formed with Particle Swarm Optimization and Fireworks Algorithm. The algorithm tracks the global maximum power point (MPP) when conventional MPPT methods fail due to occurrence of partial shading conditions. With the applied strategies and operators, PS-FW algorithm obtains superior performances verified under simulation and experimental setup with multiple cases of shading patterns.

This study examined the extent to which coaching facilitates the successful implementation of the Problem-Solving/Response to Intervention (PS/RtI) model in schools, as well as the extent to which coaching enhances the fidelity of implementation of PS/RtI practices in those schools. Data from 34 schools in seven districts participating in three years of a statewide initiative to implement PS/RtI practices with assistance of a PS/RtI coach were used to evaluate the relationship between coaching activities and levels of implementation and integrity outcomes. Data on various coaching-related factors (i.e., perceived coaching quality, coach continuity, frequency and duration of training and technical assistance), educator beliefs and perceived skills, and PS/RtI implementation and fidelity levels were collected and examined utilizing a series of multilevel modeling (MLM) procedures. Results of the analysis suggest that a number of coaching variables were related to growth in specific measures of PS/RtI implementation and fidelity over time. Specifically, shorter, more frequent training sessions were related to higher levels of staff consensus and fidelity of problem analysis implementation over time after controlling for the quality of the coaching delivered. Growth in PS/RtI implementation over time was predicted positively by the continuity (the degree to which coaching was delivered by the same individual over the three years of the study) of the coaching received. Educators' perceptions of their own PS/RtI skill levels related to manipulation of data and use of technology in schools predicted increases in fidelity of problem identification implementation over time after controlling for quality of coaching. Fidelity of program evaluation/RtI implementation was predicted by the quality of coaching received across time. The relationship between coaching and infrastructure development, as well as the relationship between coaching and fidelity of intervention development and implementation, were unclear. Potential explanations for the findings from this exploratory study and implications for future research are discussed.

The amount of taxes paid by multinational enterprises (MNEs) in host and home countries continues to make headline news. Corporate tax regimes, particularly those in many OECD countries, have never been more complex and the competition to attract and retain foreign direct investment (FDI) has perhaps never been so great. All of these political, legal, economic and competitive realities face countries at a time when they need additional budget revenues. At the June 2012 G-20 Summit in Los Cabos, leaders identified base erosion and profit shifting as key fiscal issue to be addressed. Many are expecting this to translate into a new approach to applying existing international tax standards, an increased pressure to eliminate "corporate tax breaks", enact tougher anti-abuse provisions, and less tolerance of aggressive tax planning. There has been an increased critical focus on transfer pricing, corporate restructuring and double tax treaties. Some have suggested that double tax treaties are eroding the domestic tax bases of developing countries, while others conclude that double tax treaties promote development and FDI and thereby expand the tax base. Dividing up a "revenue pie" has never been easy and the implementation of international tax rules to transparently and predictably allocate revenue to avoid double taxation and double non taxation has never been more adversarial between taxpayers and tax authorities and between tax jurisdictions. It was for these reasons that the Global Tax Policy Center of the Institute for Austrian and International Tax Law (Vienna University of Economics and Business) and the International Tax and Investment Center (ITIC) decided to undertake this study. The objective of our study was to look at the development impact of double taxation treaties and, more broadly, how tax policy can help generate economic growth and prosperity. Legally domestic tax laws are normally subordinate to international double taxation treaties, but in reality a double tax treaty only serves a country as well as its domestic tax regime. We've concluded that the problems affecting developing countries lie not with double tax treaties but rather in weak domestic tax legislation. Our study reviews empirical data from 20 developing countries, including LDCs, middle-to-high income developing countries, resource-rich countries, and BRIICS[1] countries. We hope that the empirical analysis and the conclusions that can be drawn from it can help guide policymakers to refocus their policy objectives to boost capital formation, expanding exports, and protect their domestic tax bases. We believe that a country with strong domestic tax legislation can advance their pursuit of the Millennium Development Goals by affectively utilizing double tax treaties and the related international tax rules to more transparently share and grow their tax base. A solid-phase extraction (SPE) method was developed for the determination of organochlorine pesticides, namely aldrin, alpha-BHC, beta-BHC, delta-BHC, dieldrin, endosulfan I, endosulfan II, endosulfan sulfate, endrin, endrin aldehyde, lindane, heptachlor, heptachlor epoxide, 4,4’-DDD, 4,4’-DDE and 4,4’- DDT in water. The effect of extraction conditions, such as the addition of sodium chloride and methanol to the sample prior to loading was studied. The sample was concentrated by a plain polystyrene-divinylbenzene resin, and the extract was eluted by ethyl acetate. Qualification and quantification of the target pesticides were performedby gas chromatography/ mass spectrometry (GC/MS) in the full-scan and selected ion-monitoring mode, respectively, and for better detection of pesticides in field samples the mass spectrometer was altered from electron ionization (EI) to chemical ionization mode (CI). The repeatability of the method for MilliQ-water fortified with pesticides at a level of 0.1 to 0.6 µg/l ranged from 8 to 18%, and the obtained recoveries ranged from 67 to 135%.

The method was evaluated for the determination of organochlorine pesticides in fourteen surface- and groundwater samples taken from locations along King Talal Dam, King Abdullah Canal and Zarqa River in the Jordan Valley. The limit of detection of the pesticides residues in 500-ml field water samples ranged from 0.0009 to 15.7 ng/l. The obtained results confirmed the presence of trace amounts of some organochlorine pesticides in the analyzed samples, i.e. lindane and endosulfan compounds.

The purpose of this dissertation is building a system for one of the key part of TFT LCD process - color filter, how to use the photo spacer and RGB in-line thickness result predicting the final thickness. This investigation will focus on the difference between in-line and final thickness. Still, we will use the relationship to predict the final thickness in advance for reducing the time period of model change and increasing the utilization rate for more competitiveness of industry. For data and information collection, it came from AU Optronics C5A photo engineering measurement machine. The main structure of the analysis belongs to “Teaching the Taguchi method to industrial engineers“. Based on the theory、procedure、and relative sheet we could find the most obvious and effective key factor for this experiment module. Still, according to the result by back-propagation method we could infer the key parameter and put them for in-line verification. Finally, the real time result shows us a good performance for process thickness monitor and improvement of time period when model change. For well controlling of the thickness, engineer used to follow the experiment for recipe tuning without any verification rule. This analysis also made a contribution that the key process factor of the final thickness was solid ratio in photo-resist. Based on the In-line THK data and the contraction percentage of thickness by PS/RGB photo-resist,we got good result for predicting the in specification final thickness data that the error percentage less than only 1%.

Developments in linguistics provided another external source of inspiration for students of ideology. ‘The struggle over political language’ explores the contribution of language to the development of ideology. The meaning of language — in particular the emphasis on grammar and semantics — played a critical role in enabling similar ideologies to be expressed in their own specific terms. Making sense of ideological texts became an increasingly important role for political theorists. From this emerged the four Ps of ideological composition: proximity, priority, permeability, and proportionality. These have provided an important tool for complex comparative analysis of ideologies.

AbstractThe chapter explores the trajectories of the evolution of political science (PS) in four former Soviet Socialist Republics (Estonia and Lithuania, the Republics of Moldova and Belarus) after the USSR collapse. Departing from the premise that PS is appreciated as the science of democracy, the authors claim that its identity and autonomy are particularly important. Research shows that PS in these countries started from the same impoverished basis (“scientific communism”), but it soon took diverse trajectories and currently faces specific challenges. Democracy, pro-Western geopolitical settings and the shorter period of Sovietization contributed to the faster and more sustainable development of PS in two Baltic States. However, in Estonia, political developments have led to the retrenchment of PS and to downsize of universities’ departments and study programmes. In Lithuania, political scientists are very visible in the public sphere. In Moldova, its uncertain geopolitical orientation and a series of internal political conflicts have led to the weak identity of PS and questionable prospects for its further institutionalization. In authoritarian Belarus, PS as an academic discipline exists within a hostile political environment and under a hierarchical system of governance offering practically no degree of academic freedom.

The current article is concerned with the development of Proto-Samoyed *j in intervocalic position in Nganasan. In previous studies, intervocalic PS *j is said to disappear in Nganasan, leading into the formation of secondary vowel sequences in addition to the primary ones inherited from Proto-Samoyed. A more detailed inspection of certain Proto-Samoyed etymologies reveals that *j indeed disappears in intervocalic position unless it is either preceded or followed by a sequence of two vowels, in which case it is preserved and phonetically often strenghtened to d’. The implications of this change are further discussed in light of one particular etymology, that of PS *kåläjə̑ŋ ’mammoth / whale’, for which Nganasan has, in addition to the regular reflex kol’iiŋ ’whale’, several doublet reflexes that appear to be early borrowings from a neighbouring Samoyed language.

Abstract Recent developments in hybrid low pressure thermal spray technologies such as Plasma Spray-Thin Film (PS-TF), PS-PVD, PS-CVD are being increasingly used to develop functional inorganic coatings and films for emerging high end energy applications. The requirements of such coatings and films are more highly specified than those of conventional plasma spray coatings. Successful film deposition therefore requires not only the development and application of novel operating parameters, but also goes hand-in-hand with tailored feedstock materials development. Targeted development by Sulzer Metco has allowed applications to evolve into fields where conventionally competitive manufacturing technologies would be applied; potentially enabling entirely new fields of plasma spray manufacturing to emerge. Such applications include corrosion protection and electrolytic films in SOFC, gas tight mixed electron and ion conducting membranes for gas separation and thin, transparent functional layers in photo-voltaic applications. This paper provides a brief overview of the status of developments of several high end emerging energy applications which are being developed using such hybrid low pressure plasma spray technologies.

The possibility of low-temperature synthesis of graphene on the surface of porous silicon (PS) is associated with the excess surface energies of nc-PS nanocrystallites ; the boundary interface nanocrystallties nc-PS / c-Si monocrystal matrix; the dangling bonds of silicon atoms of nanocrystallites skeleton nc-PS. This opens up new prospects for the development of methods for the low-temperature synthesis of graphene without metal catalysts for the decomposition of carbon precursors, including using the ALD method.

In Israel and in the U.S., Streptococcus iniae is responsible for considerable losses in various fish species. Poor understanding of its virulence factors and limited know-how-to of vaccine formulation and administration are the main reasons for the limited efficacy of vaccines. Our strategy was that in order to Improve control measures, both aspects should be equally addressed. Our proposal included the following objectives: (i) construction of host-pathogen interaction models; (ii) characterization of virulence factors and immunodominant antigens, with assessment of their relative importance in terms of protection and (iii) genetic identification of virulence factors and genes, with evaluation of the protective effect of recombinant proteins. We have shown that two different serotypes are involved. Their capsular polysaccharides (CPS) were characterized, and proved to play an important role in immune evasion and in other consequences of the infection. This is an innovative finding in fish bacteriology and resembles what, in other fields, has become apparent in the recent years: S. iniae alters surface antigens. By so doing, the pathogen escapes immune destruction. Immunological assays (agar-gel immunodiffusion and antibody titers) confirmed that only limited cross recognition between the two types occurs and that capsular polysaccharides are immunodominant. Vaccination with purified CPS (as an acellular vaccine) results in protection. In vitro and ex-vivo models have allowed us to unravel additional insights of the host-pathogen interactions. S. iniae 173 (type II) produced DNA fragmentation of TMB-8 cells characteristic of cellular necrosis; the same isolate also prevented the development of apoptosis in NCC. This was determined by finding reduced expression of phosphotidylserine (PS) on the outer membrane leaflet of NCC. NCC treated with this isolate had very high levels of cellular necrosis compared to all other isolates. This cellular pathology was confirmed by observing reduced DNA laddering in these same treated cells. Transmission EM also showed characteristic necrotic cellular changes in treated cells. To determine if the (in vitro) PCD/apoptosis protective effects of #173 correlated with any in vivo activity, tilapia were injected IV with #173 and #164 (an Israeli type I strain). Following injection, purified NCC were tested (in vitro) for cytotoxicity against HL-60 target cells. Four significant observations were made : (i) fish injected with #173 had 100-400% increased cytotoxicity compared to #164 (ii) in vivo activation occurred within 5 minutes of injection; (iii) activation occurred only within the peripheral blood compartment; and (iv) the isolate that protected NCC from apoptosis in vitro caused in vivo activation of cytotoxicity. The levels of in vivo cytotoxicity responses are associated with certain pathogens (pathogen associated molecular patterns/PAMP) and with the tissue of origin of NCC. NCC from different tissue (i.e. PBL, anterior kidney, spleen) exist in different states of differentiation. Random amplified polymorphic DNA (RAPD) analysis revealed the "adaptation" of the bacterium to the vaccinated environment, suggesting a "Darwinian-like" evolution of any bacterium. Due to the selective pressure which has occurred in the vaccinated environment, type II strains, able to evade the protective response elicited by the vaccine, have evolved from type I strains. The increased virulence through the appropriation of a novel antigenic composition conforms with pathogenic mechanisms described for other streptococci. Vaccine efficacy was improved: water-in-oil formulations were found effective in inducing protection that lasted for a period of (at least) 6 months. Protection was evaluated by functional tests - the protective effect, and immunological parameters - elicitation of T- and B-cells proliferation. Vaccinated fish were found to be resistant to the disease for (at least) six months; protection was accompanied by activation of the cellular and the humoral branches.

This document (PS-125) presents the guidelines that will be used by the Private Sector Department (PRI) to prepare its Expanded Project Supervision Report (XPSR). The XPSR will help IDB to comply with the Evaluation Cooperation Group (ECG) Good Practice Standards (GPS) and to initiate a process of improving good practices in the Inter-American Development Bank (IDB). The XPSR builds on the basic monitoring reports already in place - Project Supervision Reports (PSRs) and Project Performance Monitoring Report (PPMRs) - by adding information requirements that are relevant for a self-evaluation document, such as issues relating to achievement of development outcomes, environmental performance, contribution to the country living standards and lessons learned. The XPSR fulfills the function of a Project Completion Report (PCR).
