In the context of platforms, an open architecture is instrumental in enabling innovation by complementors. But as complementors increasingly deplete the innovation opportunities that the platform architecture affords, the platform architecture must evolve to reinvigorate the platform’s generative capacity. This article underscores the role of complementors in the process of platform architecture evolution by introducing the notion of architectural generativity. Architectural generativity involves actively soliciting and selectively incorporating contributions from complementors to help evolve the platform in unforeseen ways. In the case of the Mozilla Firefox web browser platform, complementors contributed new ways to facilitate access to the platform’s technological components and suggested better ways to control the platform and its architecture.

The paper focuses on the new field of philosophy of architecture in analytic tradition. The research presented in this paper is part of an ongoing doctoral research concerning the connection between ethics and aesthetics in architecture. The connection between architecture and philosophy is not a novelty. Architectural theory has always looked up to philosophy for inspiration but only recently philosophers have started to study architecture in detail. Architectural theory is still a field that is in search of a better conceptual frame after the failure of the theoretical premises of the Modern Movement and the rise of the "theory speak". Architecture's features as a public art ask for a certain amount of objectivity. The philosophy of architecture in analytic tradition can contribute to a more objective conceptual frame. The principal concerns are familiar to those acquainted with continental philosophy: What makes architecture special among the arts? What is the essence of an architectural work? How we can better define architecture's social mission? But the way of answering them is different. The ethical dimension of architecture is one of the most debated subjects among architects in recent years. There are several contributions on the connection between ethics and aesthetics in the philosophy of architecture in analytic tradition. They have the potential to have an impact on architectural theory and practice.

To investigate the decay mechanisms of red oak (Quercus rubra) and hemor (Schima spp.) woods in the old architectural structure of Xichuan Guild Hall, chemical composition changes were determined and analyzed with X-ray photoelectron spectroscopy (XPS). The results showed that decaying resulted in a noticeable decrease of the O/C from 0.59 to 0.42 in the red oak wooden components. The increase of C1 contribution, decrease of C4 contribution, increase of O1 and O3 contributions, and decrease of O2 contribution indicated that the carbohydrates in red oak wooden components can be easily degraded by fungi compared with lignin. Moreover, decaying resulted in a slight decrease of the O/C from 0.49 to 0.47 in the hemor wooden components. The results of increase of C1 contribution, decrease of C3 and C4 contributions, increase of O1, and decrease of O2 and O3 contributions indicated that carbohydrate and lignin were all degraded by fungi.

PurposeThe highly contagious coronavirus and the rapid spread of COVID-19 disease have generated a global public health crisis. Crises are being addressed at various local and global scales through social distancing measures and guidelines, emerging working and living patterns and the utilisation of technology to partially replace physical learning environments. The purpose of this article is to capture the key messages of the contributions published in this special edition of Archnet-IJAR: International Journal of Architectural Research, Volume 15, Issue 1, March 2021. Reviewing more than 70 submissions, 15 articles have been identified that are contributed by 35 scholars, educators and practitioners from 12 countries. The article calls for the need to embed trans-disciplinarity in current and future built environment research.Design/methodology/approachDriven by the fact that architecture, urban design and planning and built environment studies interact and have direct correlation with public health and virus spread. The approach to develop and present the key messages of the contributions is premised on three areas: (a) the pandemic condition as it relates to the built environment, (b) analytical reflections on the emerging themes and (c) the diversity and complexity embedded in these themes.FindingsWhile some contributions speak to the particularities of their contexts, others address regional or global parameters. The enquiry into architectural research, architectural education and architectural design indicates some of the important methods and tools to address the accelerated adoption, adaption and redesign needed to create a new and better normal which embeds flexibility, adaptability and continuous learning. The papers represent brilliant investiture to address the momentous insinuations the COVID-19 condition has on the built environment.Research limitations/implicationsThe diversity of implications reveals potential alternative futures for urbanity and society and the associated education and practice of future built environment professions. While the contributions invite us to critically envisage possibilities for future research and collective action, critical fast-track empirical research is needed to address how health is an integral component in the production of architecture and urban environments.Originality/valueThe diversity, complexity, depth and breadth of the contribution convey important insights on people, health and the spatial environments that accommodate both. Trans-disciplinarity, as it relates to research and action and to the production of urban environments, is viewed as a form of learning involving co-operation among different parts of society, professionals and academia in order to meet complex challenges of society such this pandemic condition. This approach has enabled the identification of three future research areas in architecture urbanism that include implications of virus spread on urban environments, how spatial and social distancing measures and protocols are altering our understanding of spatial design.

This essay reviews three books on contemporary Chinese urbanism. The first book, Designing Reform, understands the exceptionalism of Chinese reform through architectural evidence. The second book, The Shenzhen Experiment, further argues for the exceptionalism and non-replicability of Shenzhen reform. The third book, The City After Chinese New Towns, similarly highlights the unprecedentedness of Chinese new towns. These three recent publications collectively aim to demystify contemporary Chinese urbanism, and have done so with a sensibility towards space, geography, materiality, and infrastructure. These authors and editors are mainly architectural researchers leveraging disciplinary contributions to the discussion of post-socialist China. In turn, they have also contributed to the field of architecture and urban design with rich case studies in China. This review essay aims to understand (i) their geographical and spatial perspectives, (ii) their architectural design perspectives, and (iii) how they address social justice issues. It seeks to formulate contemporary Chinese urbanization as an object of inquiry, to “exceptionalize” Chinese phenomena, and to argue for the pedagogical contribution of Chinese case studies to architecture and urban design.

The activity of Branko Popović is primarily defined in historiography through valorizations of his artistic oeuvre and his role as an art critic, one that he continuously undertook during the first half of the 20th century. Given that Popović's contributions to architectural criticism and the interpretation of contemporary architectural phenomena and problems have often lain outside the main scope of particular research endeavours, instead often being presented in summarial form within monographs and anthologies, there is an acute need for elaboration of this area of his outputs. Popović primarily received his higher education at the Technical Faculty in Belgrade, where he later held the role of professor of history of art and architecture, and during the Second World War he held the position of Dean. The aim of this work is to present, through the review and analysis of articles published in pre-war and inter-war periodicals, another aspect of Popović's interpretations and criticisms, based on perceptions of and participations in contemporary events in the field of architecture, which would complete his contribution to the shaping of artistic thought.

This research is an inquiry into how architecture can inform the practice of composition. As an architect and composer, I try to find strategies for musical composition In architectural practice and thought, by reframing and confronting concepts from both disciplines. My research aims at an expanded practice (and analysis) of musical creation that transverses different conceptions of space-from the score based pitch space to the social and political spaces of music's production, performance and reception. This practice based PhD research consists of a portfolio of nine works that were developed in a dialectical relation to these ideas. The works are presented in a framework composed of five conceptual tools used to articulate music and architecture. These are; Material, Site. Drawing, Programme and Use. With the notion of Material. I explore how the acoustic behaviour of a performance space, or of a performatlve device' affects the musical work. Architectural materials become musical ones as they are implicated in the listening experience. The discussion about Site brings to music the notions of place. the local, and everyday life, embracing soundscapes so many times excluded from musical discourse. Musical sites are also architectural sites, always related to their present environment, and their everyday contlngenclt;ls. Drawing Is a tool for developing ideas, but also the main mediator between architect and builder. or composer and performer. Programme exposes the constraints and conditions of the creation process, while also revealing the sociopolitical relations between musicians and audiences, institutions and composers, composers and performers. Programming as framing can be a platform to expand what the work concerns. Through a consideration of Use, the work becomes dispersed in a pJurality of agents that converge in a useful event. Thus composition. as architecture, moves from being about conditioning design to designing conditions where musical events may happen.

The purpose of this thesis is to document, through observation and research, architectural elements of a culture that have contributed to the development of another. As such it represents a way of thinking about the impact of architecture on culture and of culture on architecture. The len~ for this focus is the contributions of African people brought to the Caribbean during the European Slave Trade (1520's--1860's). As it essentially was a world region where the influence of indigenous people was negligible due to their early demise and general lack of an extensively developed architecture, it may almost be regarded as a 'clean slate' upon which the Europeans and Africans devised an architecture based on climate, materials, and tradition modified by colonization/enslavement. As climate and materials were generally similar to those found in Africa, tradition may be considered the most important aspect of this process and is the basis for this thesis and for the exploration of the following questions: What did Africans do to contribute to the architecture of the Caribbean; how did they say, "We are here?" What may have been present in the physical landscape, built and natural, that gave Africans brought to the New World any sense of familiarity of place in an otherwise alien environment? What are some of their contributions that have survived, either through transferal or transformation, physically tangible or attitudinal, that are present in the Caribbean today? In general, what do people do to affect the physical environment when they are in positions of social and economic powerlessness?

Le thème de recherche abordé dans cette HDR porte sur la conception d'architectures matérielles spécialisées, et sur leur utilisation dans le domaine des systèmes embarqués (par exemple les réseaux de capteurs) et du calcul haute performance (bioinformatique, recherche dans les flux multimédias). Ce thème de recherche est développé sur trois axes interdépendants, brièvement décrits ci-dessous. Tout d'abord la conception de plateformes matérielles de type " systèmes de stockage intelligents " basées sur des technologies de logique programmable. L'objectif est de proposer des systèmes capables de traiter les données à la volée directement en sortie du support de stockage (disque magnétique, mémoire Flash), en utilisant des filtres mis en œuvre sous la forme de coprocesseurs spécialisés. Ce travail à donné lieu à la réalisation de deux prototypes de machines (RDISK et REMIX) ainsi qu'à leur validation sur plusieurs cas d'études. Ensuite, la conception d'accélérateurs matériels spécialisés et massivement parallèles, pour des applications de calcul intensif issues de la bioinformatique et du traitement de données multimédia. Nous nous sommes en particulier intéressés à des algorithmes de recherche de similarités entre séquences, basés sur la notion de profil de Markov (logiciel HMMER), ainsi qu'à des algorithmes de recherche d'images par le contenu. Enfin, les outils d'aide à la conception d'architectures spécialisées à partir de spécifications de haut niveau. Dans ce contexte, nous avons proposé un flot de conception complet pour la conception de nœuds de réseau de capteurs ultra faible consommation. Nous nous sommes également intéressés aux problèmes de génération automatique d'accélérateurs pour nids de boucles, notamment au travers d'un flot de transformation source à source de boucles ciblant en sortie des outils de synthèse de haut niveau " C to hardware ".

NcRNA represent most of primary transcripts RNA in higher eukaryotes and tune gene expression via diverse mechanisms. They adopt 3D structures composed at 70% by WC bp forming A-form helices linked by RNA motifs. We identified the pk-turn, a new RNA motif related to k-turns that allow for the formation of a bend of 60° between stems P16 and P17 from the bacterial RNaseP. Yet it features different sequence and structural requirements than k-turns. The 2nd ribozyme which got my attention is the LCrz inserted in GIR2, a group I intron. This twintron is observed in the pre-rRNA 18S of the small subunit of the eukaryoteD. iris. LCrz catalyzes a reaction equivalent to the first step of splicing by group II introns, but in a structural context related to group I introns. We solved the 2.5 Å crystal structure of the LCrz and confirmed the unexpected shape by means of SAXS experiments. This work emphasizes the relationship between structure and function in the evolution of ribozymes.

Cette thèse s'inscrit dans le cadre d'études visant à améliorer la qualité de service d'un serveur internet, mono-serveur ou en grappe, en cas de surcharge ou en cas de panne. En premier lieu, nous recommandons une architecture orientée session pour le traitement efficace de trafic réseau, offert à une grappe de serveurs internet. En effet, il s'avère que, d'un point de vue économique, le traitement à l'échelle session est plus approprié pour l'évaluation de critères de performance d'un serveur internet. Nous proposons ensuite une architecture flexible garantissant aux services internet une performance adéquate, en cas de panne du serveur légitime. En deuxième lieu, nous nous focalisons sur l'évaluation de mécanismes de contrôle d'admission implicites, opérant à l'échelle session. Nous montrons que le paradigme orienté session améliore le profit de l'opérateur et garantie une QdS aux sessions utilisateurs offertes, indépendamment de leur durée
This thesis focuses on the contributions of edge-network processing devices in improving the perceived end-to -end QoS. Our goal is to recommend an efficient scheme for improving the percieved end-to-end QoS, either under server overload, or when legitimate processing server experiences failures. Firstly, we have described a fully client/server transparent methodology for engineering fault tolerant network services. The proposed architecture adds non additional complexity to a legitimate processing server during failsafeperiods, and enhances the robustness of internet servers across failures. At a second step, we have advocated a methodology, for efficiently improving the salability of high performance cluster-based internet servers, subjected to multiple flow-based sessions. Secondly, we have shown that , responsive session aware admission control contributes in providing improved QoS to subscribers, under overload

Depuis l'apparition des premiers ordinateurs, l'arithmetique flottante a enormement evolue. La norme ieee 754 a permis de fixer les caracteristiques de l'arithmetique des ordinateurs modernes, mais les scientifiques perdent de plus en plus vite le controle de la validite de leurs calculs. Malgre l'enorme travail associe a la definition des operations, la validation des calculs ne peut toujours pas etre assuree de facon certaine par l'arithmetique implantee sur les ordinateurs. Je presente dans la premiere partie de cette etude deux prolongements qui visent a augmenter la marge de validite des operations: un nouveau mode d'arrondi pour les fonctions trigonometriques et un codage efficace des intervalles accessible facilement a l'utilisateur. L'arithmetique en-ligne permet de gerer efficacement les problemes de precision, mais les operateurs elementaires utilises sont peu adaptes aux architectures modernes de 32 ou 64 bits. L'implantation efficace d'un operateur en-ligne ne peut que passer par la description d'un circuit de bas niveau. Les prediffuses actifs, terme francais utilise pour field programmable gate array, sont des composants speciaux programmables au niveau des portes logiques. Ils permettent d'abaisser les couts de production en evitant de fabriquer un prototype. Nous avons implante grace a ces technologies les operateurs simples de calcul en-ligne: addition, normalisation, etc le noyau arithmetique de calcul en-ligne (nacel) decrit dans ce memoire permet d'implanter les operations arithmetiques usuelles telles que la multiplication, la division, l'extraction de racine carree et les fonctions elementaires trigonometriques et hyperboliques par une approximation polynomiale. Les architectures a flots de donnees sont insensibles aux difficultes sur lesquelles butent les concepteurs des ordinateurs modernes: temps d'acces a la memoire, latence de communication, occupation partielle du pipeline d'instructions. Je decris dans ce document une petite unite de calcul en-ligne (puce). Par une gestion adaptee des etiquettes inspiree pour le controle des donnees de celle utilisee par la manchester data flow machine. Puce reproduit le comportement complet d'une machine a flot de donnees. Elle comprend de plus les operations en-ligne de calcul scientifique. Nous presentons les resultats de simulations sur des calculs divers pour une ou plusieurs unites fonctionnelles

Barbara van den Broek (1932-2001) trained as an architect in Auckland, New Zealand before moving to Brisbane with her husband and fellow architect Joop, where they established an architectural practice. van den Broek went on to run an office as a sole practitioner and took on architecture and landscape architecture projects. Over the course of her career she completed post-graduate diplomas in Town and Country Planning, Landscape Architecture and Education, and a Master of Science – Environmental Studies, and collaborated on a number of key projects in Queensland and Papua New Guinea (PNG). Our paper will build an account of her career. In assessing the significance of her contribution to landscape architecture, planning and architecture in Australasia, it will bring a number of other spheres into the frame: conservation and Australia’s environment movement; landscape design and the bush garden; and van den Broek’s personal development that included artistic expression, single parenthood, teaching, and the navigation of male-dominated professional environments to develop a practice that contributed to town planning projects in cities across Australia, and made significant contributions to landscape projects in Queensland and PNG.

The Cambridge School of Architecture and Landscape Architecture, active from 1915 to 1942,1 was a groundbreaking institution in architectural education. It was the first U.S. school to grant master’s degrees to women and integrated the teaching of architecture and landscape architecture.2 Beginning and ending at the height of two world wars, the Cambridge School challenged prevailing academic and professional norms, offering a disruptive, barrier-free space that redefined architectural pedagogy and influenced the field.Largely forgotten to history, this paper uses the conceptual framework of the home front to reframe the school’s legacy as one of defiance to a system that tried to silence it, particularly its influence on Harvard’s Graduate School of Design (GSD). By collecting and uncovering previously disparate source materials from several archives, this paper connects the physical manifestations of the school to its pedagogical frameworks to elucidate the school’s complex and far-reaching influence during its active years and beyond.Recognizing the Cambridge School’s contributions provides a deeper understanding of the women’s role in architectural education and challenges historical dismissals of their contributions. This pioneering institution reshaped architectural pedagogy, promoted women’s agency, and continues to offer valuable lessons for a more inclusive and equitable architectural profession.

The enhancement process records an orientation that in recent years has found in the relevant digital tools one of the possible keys for setting up a data system and, in digital modeling, the medium for interesting developments also in relation to the use. Starting from the studies conducted in recent years on this specific topic, the research group is trying to identify the best strategies to be locally applied to enhance and make available on web different models at the various scales that describe Turin fortification's system and some portions of it.

Conservation and enhancement of ruined fortified systems in Tanaro Valley (Piedmont)The contribution reports on a research concerning the preparatory investigation of some fortified systems of Tanaro Valley (Piedmont) in view of their restoration. The research highlighted the strong connection between the castles and the memory of events and characters that were linked to these architectures; the relationship between landscape and ruins that, today, represents a potential resource for the territory; the willingness of local authorities to improve knowledge, protection and enhancement of this heritage; the need to investigate theoretical and methodological issues and define the proper procedures for the conservation, restoration, safeguard, maintenance and management of a widespread and vulnerable heritage. The study, taking advantage of interdisciplinary contributions, used innovative instruments for the survey and monitoring of this heritage, as well as graphic and digital restitution techniques to propose to the scientific community –and communicate to the native population– the interventions of restoration, conservation, enhancement and management of these fortified systems. The research demonstrates that particular attention has to be paid to these less-known historical architectures that can however contribute, as effectively as the most famous and eye-catching fortification systems, to the political and social development and to the historical identity whose recognition, preservation and valorization are strongly hoped for.

Vernacular architecture can be defined as a typology that considers the local or regional character, in which materials and resources from the environment where the building is inserted are used. Bioconstruction, in turn, in addition to considering parameters such as adequacy to local conditions, evaluates waste treatment and employs constructive techniques with low environmental impact in order to build sustainable buildings. Using applied, qualitative, descriptive and bibliographical research, we sought to unite both concepts, correlate them with obtaining sustainable seals and also investigate their contributions to encourage sustainability in architectural projects. It was concluded, therefore, that vernacular architecture and bioconstruction are already being resumed and put into practice in contemporany building, thus showing that they are capable of encouraging architecture for sustainable behavior.

The study of military architecture, a multidisciplinary task, prompts a reassessment of this cultural environment, often neglected or at risk. From prehistoric fortification traces to contemporary bunkers and military infrastructures, the discourse on documenting and preserving military heritage welcomes and encourages contributions from diverse fields, including architecture, engineering, archaeology, history, geography, and cultural heritage.FORTMED, the international conference on the Fortifications of the Mediterranean Coast, has evolved into a research-based platform that transcends borders and centuries, delving into the strategic, historical, and cultural significance of fortifications along the Mediterranean coast. The geophysical data sets available for the western Churchill Province have had a bearing on the understanding of its structure, evolution and metal endowment. New data were acquired and interpreted during the Geo-mapping for Energy and Minerals (GEM) Program (2008-2020). Regional, high-resolution aeromagnetic, and targeted gravity and magnetotelluric surveys were collected in GEM, in conjunction with geological mapping projects, in order to provide control on bedrock features beneath widespread glacial overburden and flat-lying sedimentary basins. Quantitative estimates of three-dimensional geometry were obtained in key areas through geophysical models integrating the geophysical characteristics with local rock property measurements. These geophysical data sets contributed to new knowledge and interpretations in three related research fields: 1) location and nature of Rae cratonmp;gt;'s boundaries within the western Churchill Province; 2) definition of internal Rae architecture; and 3) identification of reactivated structures controlling gold and uranium mineralization. The new data, models and emerging tectonic and metallogenic frameworks will serve as guides for future exploration in this remote, complex, challenging region.

Age dependent programs are responsible for the physiological and developmental differences of young and mature plants. These include a range of morphological characters such as leaf shape and leaf composition (waxes, lignin etc..) but also different in developmental potentials. Apical buds of juvenile plants are vegetative, while those of mature plants can be reproductive. Likewise, basal buds form in the axills of juvenile leaves have different fates than distal buds formed in the axils of mature leaves. The goal of our joint project is to understand and exploit theses age related programs for specific improvement of crop plants. To that end both the WIS group and the PGEC group are using mutants with age related defects as well as modified expression of miR156 to modify age related programs in crop plants- Tomato and potato in Israel and Maize, switchgrass and Brchipodium in the US. In the US, major effort were made to: Characterize the contribution of selected miR156 target genes to yield component traits of maize. Functional analysis of microRNAs and their targets in new crop plants. In Israel, the research progressed in several directions: Understanding the interplay between age dependent programs and the potential of tomato and potato meristems to produce tubers. Evaluation of the agronomic value of mutants that alter flowering regime in side shoots in general, and in the sympodial buds in particular Characterization of wild type axillary buds, comparing shoot ontogeny of gradually maturing apices from basal and distal positions along the main shoot of tomato.

We analyzed the main cloud services in the article. We also described the main contribution of mobile cloud technology to education. The article presents the author’s development from the field of mobile cloud education in higher mathematics. The design architecture of this application is described in detail: QR generator and scanner, authorization, sending tasks. Block diagrams and images are presented that clearly demonstrate the operation of the application. We showed an example of solving the integral from the section of integral calculus for higher mathematics and showed how to download the answer in the form of a QR code and find out whether it is correct or incorrect (this can be seen by the color on the smart phone screen). It is shown how this technology helps the teacher save time for checking assignments completed by students. This confirms its effectiveness. Such an application provides students and teachers with the ability to store and process data on a cloud computing platform.

In this project, we are determining the contribution of different types of variations, in gene function and in gene regulation, to altered shoot architecture first, and to field performance in the next stage. We are using tomato as a target, but also as a model for many other crops. Our focus is on two different components of yield associated traits - shoot architecture and organization of the inflorescence. Our focus was on two types of regulators; 1) genes involved in florigen - antiflogen balance and the way they impact the shoot, and 2) genes involved in inflorescence branching and it this way, the way they impact the number of flowers produced by the plant. For the first class, we described our thoughts on that matter in a joint review (Eshed and Lippman, 2019) where we argued that annualization of short-lived vine plants such as tomato and soybean was their major adaptation for intensive modern farming. This annualization was achieved by introduction of mutations in the anti florigen gene SELFPRUNING, mutations that were also used to domesticate/adapt cotton, roses, strawberries and more (Eshed and Lippman, 2019). Indeed, introduction of this mutation and additional one in another antifloraigen gene SP5G into a vine type tomatoes resulted in compact, early yielding plants suitable for urban agriculture (Kwon et al., 2020 - please note that Yossi Capua, a former graduate student from the Eshed group is a coauthor of this study). The other side of this project, customized generation of large branched inflorescences, relied on ongoing collaboration between the two labs on genes like S and genes of the SEP clade, J2 and EJ2. We first showed that highly branched j2 ej2 inflorescences can be quantitatively modified by introduction of either null or naturally weak alleles of the TM3/STM3 gene cluster (Alonge et al., 2020). We next showed that variation in inflorescence branching can be obtained by customized interreference in selected promoter motifs of S, a gene that is otherwise essential at the seed stage (Hendelamn et al., 2021). Overall, our joint project provided some prime examples for targeted use of genome editing for the formation of valuable alleles that can either improve modern crops or, significantly, open the door for rapid "domestication" of orphan crops

Abstract (sommario):
This study investigates the mechanisms of neighborhood relations and the challenges affecting neighborhood connectivity in urban communities. It examines factors contributing to declining neighborhood connections, such as lack of sincerity, absence of belonging, busy lifestyles, design issues, and the pervasive impact of mobile phones. The role of architects and urban planners in promoting connectivity through policies and design strategies is analyzed, alongside government initiatives like town meetings and community engagement. Emphasizing the importance of community leaders, neighborhood clubs, regular gatherings, and accessible spaces, the study also explores the potential of social media and technology in fostering community connections. Finally, it highlights the significance of architectural practices, such as diverse housing options, well-designed public spaces, pedestrian-friendly infrastructure, and sustainable design, in creating vibrant and connected neighborhoods.

PGPR bacteria include taxonomically diverse bacterial species that function for improving plant mineral nutrition, stress tolerance, and disease suppression. A number of PGPR are being developed and commercialized as soil and seed inoculants, but to date, their interactions with resident bacterial populations are still poorly understood, and-almost nothing is known about the effects of soil management practices on their population size and activities. To this end, the original objectives of this research project were: 1) To examine microbial community interactions with plant-growth-promoting rhizobacteria (PGPR) and their plant hosts. 2) To explore the factors that affect PGPR population size and activity on plant root surfaces. In our original proposal, we initially prqposed the use oflow-resolution methods mainly involving the use of PCR-DGGE and PLFA profiles of community structure. However, early in the project we recognized that the methods for studying soil microbial communities were undergoing an exponential leap forward to much more high resolution methods using high-throughput sequencing. The application of these methods for studies on rhizosphere ecology thus became a central theme in these research project. Other related research by the US team focused on identifying PGPR bacterial strains and examining their effective population si~es that are required to enhance plant growth and on developing a simulation model that examines the process of root colonization. As summarized in the following report, we characterized the rhizosphere microbiome of four host plant species to determine the impact of the host (host signature effect) on resident versus active communities. Results of our studies showed a distinct plant host specific signature among wheat, maize, tomato and cucumber, based on the following three parameters: (I) each plant promoted the activity of a unique suite of soil bacterial populations; (2) significant variations were observed in the number and the degree of dominance of active populations; and (3)the level of contribution of active (rRNA-based) populations to the resident (DNA-based) community profiles. In the rhizoplane of all four plants a significant reduction of diversity was observed, relative to the bulk soil. Moreover, an increase in DNA-RNA correspondence indicated higher representation of active bacterial populations in the residing rhizoplane community. This research demonstrates that the host plant determines the bacterial community composition in its immediate vicinity, especially with respect to the active populations. Based on the studies from the US team, we suggest that the effective population size PGPR should be maintained at approximately 105 cells per gram of rhizosphere soil in the zone of elongation to obtain plant growth promotion effects, but emphasize that it is critical to also consider differences in the activity based on DNA-RNA correspondence. The results ofthis research provide fundamental new insight into the composition ofthe bacterial communities associated with plant roots, and the factors that affect their abundance and activity on root surfaces. Virtually all PGPR are multifunctional and may be expected to have diverse levels of activity with respect to production of plant growth hormones (regulation of root growth and architecture), suppression of stress ethylene (increased tolerance to drought and salinity), production of siderophores and antibiotics (disease suppression), and solubilization of phosphorus. The application of transcriptome methods pioneered in our research will ultimately lead to better understanding of how management practices such as use of compost and soil inoculants can be used to improve plant yields, stress tolerance, and disease resistance. As we look to the future, the use of metagenomic techniques combined with quantitative methods including microarrays, and quantitative peR methods that target specific genes should allow us to better classify, monitor, and manage the plant rhizosphere to improve crop yields in agricultural ecosystems. In addition, expression of several genes in rhizospheres of both cucumber and whet roots were identified, including mostly housekeeping genes. Denitrification, chemotaxis and motility genes were preferentially expressed in wheat while in cucumber roots bacterial genes involved in catalase, a large set of polysaccharide degradation and assimilatory sulfate reduction genes were preferentially expressed.
