Chern-Simons theory is formulated quantum field theoretically in terms of string operators, which are localized on finite non-selfintersecting paths in the zero-time plane R2. It is shown how the Weyl algebra approach to the abelian Chern-Simons theory leads to a bundle theoretic construction of these ‘Chern-Simons string operators’. To proceed to the non-abelian case topological Chern-Simons theory is considered as a specific model of a more general theory describing the quantum kinematics of coloured framed Plek ton s inR2. This theory allows the construction of string operators as pair-creation operators, mapping n-Plekton states into (n+2)-Plekton states. Link invariants are obtained as vacuum correlation functions of string operators and obey a natural version of reflection positivity. The vacuum sector of such a purely kinematical Plekton theory may be recovered from the link invariants by Osterwalder-Schrader reconstruction. To make the theory work one needs the structures of a ribbon graph category, for which the representations of a quasi-triangular quasi-Hopf algebra may serve as a specific realization.

Mathematical construction of quantum field theory is reviewed with emphasis on the conceptual structure of the construction and on the role of rigorous renormalization group analysis without technical details. After explaining a rigorous formulation of a renormalization group method in a weak coupling region, a new approach in a strong coupling region is proposed in the context of the hierarchical approximation. An idea of proving triviality in d≥4 dimensions utilizing this new proposal concludes this review.

We reconsider the construction of exponential fields in the quantized Liouville theory. It is based on a free-field construction of a continuous family or chiral vertex operators. We derive the fusion and braid relations of the chiral vertex operators. This allows us to simplify the verification of locality and crossing symmetry of the exponential fields considerably. The calculation of the matrix elements of the exponential fields leads to a constructive derivation of the formula proposed by Dorn/Otto and the brothers Zamolodchikov.

Commutator theory has its origins in constructive quantum field theory. It was initially developed by Glirnm and Jaffe [7] as a method to establish self-adjointness of quantum fields and model Hamiltonians. But it has subsequently proved useful for a variety of other problems in field theory [17] [15] [8] [3], quantum mechanics [5], and Lie group theory [6]. Despite all these detailed applications no attempt appears to have been made to systematically develop the theory although reviews have been given in [22] and [9]. The primary aim of the present paper is to partially correct this situation. The secondary aim is to apply the theory to the analysis of first and second order partial differential operators associated with a Lie group.

With a view to the more ultimate goal of providing fresh perspectives to address the perennial theological problem of the compatibility of divine sovereignty and human freedom, this thesis undertakes a critically constructive re-examination of fundamental orientations guiding Wolfhart Pannenberg’s thought, arguing that, when properly read through the far-reaching and decisive influences of Duns Scotus and Friedrich Schelling, influences hitherto only scantily acknowledged, Pannenberg’s overarching theology, from very early in his career to his latest writings, must be understood throughout as a ‘theology of history,’ and that when so understood, can also be seen as manifesting a previously unacknowledged unity and consistency. The critical demonstration of the deep influences of Scotus and Schelling, most notably for my purposes, on Pannenberg’s understanding of history and contingency crucially also makes possible a re-evaluation and a particular modification of Pannenberg’s use of field theory, which will contribute significantly to the goal of showing in new ways the compatibility of divine sovereignty and human freedom. The thesis begins with an examination of Pannenberg’s earliest works to establish the importance of contingency, understood in Scotist terms, and the influence of Schelling on Pannenberg’s view of history. Building upon a view of history as contingent and purposed, the thesis demonstrates that Pannenberg’s concept of history encompasses all of cosmic history and dismisses any distinction between histories, such as Historie and Geschichte, as artificial. The thesis shows that to affirm the sovereignty of God, Pannenberg maintains that the end of history is so assured as to have been seen proleptically in the historical resurrection of Christ. Likewise, in order to maintain the freedom of humanity, Pannenberg argues that, in one sense, God is not yet fully manifest in his sovereignty, yet he continues to interact within and create history as a manifestation of himself within which his creation moves contingently and freely. However, the thesis argues that the simultaneous affirmation of both positions is possible only when channelled through vital aspects of the influence of Scotus and Schelling and through a crucial reframing of Pannenberg’s use of field theory, which must be understood in temporal rather than material terms. By presenting himself as the defined end of history to the present moment, God is shown to give the temporal field into which creatures respond ‘ecstatically’ to such an end in a way that affirms the present and future contingency and freedom of human action, without denying the sovereignty of God.

The subject of the thesis is the study of singular stochastic partial differential equations (SPDEs), in connection with questions of mathematical physics and constructive field theory. The first part of the thesis is an introduction to constructive field theory, stochastic quantisation, and the resolution of singular SPDEs, topics in which the rest of the manuscript fits. The second part of the thesis, based on a paper written in collaboration with Ajay Chandra, is about the stochastic quantisation of non-local Euclidean field theories analogous to $$Phi^4_2$$ and $$Phi^4_3$$, called tensor field theories. The last part of the thesis deals with the construction of the $$Phi^4$$ measure on three-dimensional closed manifolds. This study, publishedin two works, was carried out in collaboration with Ismaël Bailleul, Viet Dang and Tat Dat Tô

The subject of this thesis in the study of nite extensions of p-adic fields, in different aspects. Via the study of the Galois module of p-th power classes L =(L )p of a general Galois extension L=K of degree p, it is possible to deduce and classify the extensions of degree p2 of a p-adic field. We exhibit formulae counting how many times a certain group appears as Galois group of the normal closure, generalizing previous results. In general degree we give a synthetic formula counting isomorphism classes of extensions of fixed degree. The formula is obtained via Krasner formula and a simple group-theoretic Lemma allowing to reduce the problem to counting cyclic extensions, which can be done easily via local class eld theory. When K is an unrami ed extension of Qp we study the problem of giving necessary and su cient conditions on the coe cients of an Eisenstein polynomial for it to have a prescribed group as Galois group of the splitting field. The techniques introduced allow to recover very easily Lbekkouri's result on cyclic extensions of degree p2, and to give a complete description of the Galois group, with its rami cation filtration, for splitting fields of Eisenstein polynomials of degree p2 which are a general p-extension. We then show how the same methods can be used to characterize Eisenstein polynomials defining a cyclic extension of degree p3. We then study Eisenstein polynomials in general, describing a family of special reduced polynomials which provide almost unique generators of totally rami ed extensions, and a reduction algorithm. The number of special polynomials generating a fixed extension L=K is always smaller than the number of conjugates of L over K, so that each Galois extension is generated by exactly one special polynomial. We give an algorithm to recover all special polynomials generating one extension, and a criterion that allows to detect when the extension generated by an Eisenstein polynomial is different from a fixed extension whose special generators are all given, the criterion does not only depend only on the usual distance on the set of Eisenstein polynomials defined by Krasner and others. An algorithm to construct the special polynomial generating an abelian class eld is given, provided a suitable description of a candidate norm subgroup of K x.

Esta tese trata a existência de Princpios de Grandes Desvios (PGD), no âmbito de sistemas fermiônicos em equilbrio. A motivação fsica detrás de nossos estudos são medidas experimentais de resistência elétrica de nanofios de silcio dopados com átomos de fósforo. Estas medidas mostram que efeitos quânticos no transporte de carga elétrica quase desaparecem para nanofios de comprimentos maiores que alguns nanômetros, mesmo para temperaturas muito baixas (4.2°K). A fim de provar matematicamente tal efeito, dividimos nosso trabalho em diversos passos: 1. No primeiro passo, para férmions não interagentes numa rede com desordem, mostramos que a incerteza quântica da densidade da corrente elétrica microscópica, em torno de seus valores macroscópicos(clássicos), é suprimida exponencialmente rápido em relação ao volume da região da rede onde um campo elétrico externo é aplicado. A desordem é modelada como um potencial elétrico aleatório juntamente com amplitudes aleatórias de saltos com valores complexos. O célebre modelo de Anderson de tight-binding é um exemplo particular do caso geral considerado aqui. Nossa análise matemática é baseada em estimativas de Combes-Thomas, o Teorema Ergódico de Akcoglu-Krengel e no formalismo de Grandes Desvios, em particular o Teorema de Gärtner-Ellis. 2. Em segundo lugar, provamos que, para férmions interagindo fracamente na rede, as funções geradoras J(s), s R de cumulantes de distribuições de probabilidades associadas com estados KMS pode ser escrito como o limite de logartmos de integrais gaussianas de Berezin. Mostramos que os determinantes das covariáncias associadas às integrais gaussianas são majorados uniformemente (via desigualdades de Hölder para normas Schatten). Tais covariâncias são também somáveis, em casos gerais de interesse, incluindo assim, sistemas que não são invariantes por translação. 3. No terceiro passo, analisamos expansões de logartmos de integrais gaussianas de Berezin, e assim combinando com métodos construtivos de teoria quântica de campos, mostramos a analiticidade de J(s) para s nas vizinhanças de 0. Finalmente, discutimos como combinar os passos 2-3, a fim de provar (matematicamente falando) os resultados experimentais mencionados acima para férmions interagindo em equilbrio. De fato, os resultados encontrados nesta tese, generalizam trabalhos prévios no âmbito do PGD usado para o estudo de sistemas quânticos.
This Thesis deals with the existence of Large Deviation Principles (LDP) in the scope of fermionic systems at equilibrium. The physical motivation beyond our studies are experimental measures of electric resistance of nanowires in silicon doped with phosphorus atoms. The latter demonstrate that quantum effects on charge transport almost disappear for nanowires of lengths larger than a few nanometers, even at very low temperature (4.2°K). In order to mathematically prove the latter, we divide our work in several steps: 1. In the first step, for noninteracting lattice fermions with disorder, we show that quantum uncertainty of microscopic electric current density around their (classical) macroscopic values is suppressed, exponentially fast with respect to the volume of the region of the lattice where an external electric field is applied. Disorder is modeled by a random external potential along with random, complex-valued, hopping amplitudes. The celebrated tight-binding Anderson model is one particular example of the general case considered here. Our mathematical analysis is based on Combes-Thomas estimates, the Akcoglu-Krengel ergodic theorem, and the large deviation formalism, in particular the Gärtner-Ellis theorem. 2. Secondly, we prove that for weakly interacting fermions on the lattice, the logarithm moment generating function J(s), s R of probability distributions associated with KMS states can be written as the limit of logarithms of Gaussian Berezin integrals. The covariances of the Gaussian integrals are shown to have a uniform determinant bound (via Hölder inequalities for Schatten norms) and to be summable in general cases of interest, including systems that are not translation invariant. 3. In the third step we analyze expansions of logarithms of Gaussian Berezin integrals, which combined with constructive methods of quantum field theory is useful to show the analyticity of J(s) for s in a neighborhood of 0. We finally discuss how to combine steps 2-3 in order to prove (mathematically speaking) for interacting fermions in equilibrium the experimental results above mentioned. In fact, the found results in this Thesis generalize previous works in the scope of LDP used to study quantum systems.

My dissertation traces the genesis and growth of the Chinese emergency management organizational field over the ten-year period since the 2003 Severe Acute Respiratory Syndrome (SARS) outbreak. I conducted my fieldwork in Beijing in 2012, using a multi-method approach that involved interviews, participant observation, and analysis of archival materials.

I found that governing elites and academic experts within the emerging field—what I call the "establishment"—relied upon insider cultural knowledge (e.g., myths) and party-state ideologies (e.g., communist folklore) to legitimize a new risk governance framework and create an emergency management policy domain separate from existing domains. The emergency management establishment enabled these legitimizing claims through its embedded position in government and academia. By using a strategy of curation, it carefully (re)assembled culturally-accepted accounts and ideas, both indigenous and adapted from afar, to create its claims. By becoming institutional evangelists, academic experts in particular entrenched the risk governance framework and propagated the ideas that helped form the field by advocating, advising, and training government officials through multiple field organizational entities.

By also incorporating Foucault's concept of governmentality, I found that the legitimacy project conducted by the establishment consisted of shaping the conduct of emergency management government officials according to its own risk governance framework. This expands current Foucauldian studies on governmentality by highlighting that not only the general population but also elites can be subjected to governmentalization. It also provides a more nuanced reading of the "hollowing of the state" thesis by highlighting how the Chinese party-state strengthened its role as the legitimate emergency manager by including the civil society and the private sector, albeit in lesser roles.

My findings draw attention to the foundational quality of cognitive legitimacy vis-à-vis pragmatic and moral legitimacy, an area which has received scant discussion in the neoinstitutional literature. It also informs the understudied topic of the interrelated influences of power relations, ideas, and experts on emerging organizational field formation within neoinstitutional theory, especially in a non-Western context. Finally, my research updates current understanding of national emergency management policies in the international arena, and especially in China.

As a consequence, we have ψ_nǁψ_nαǁǁψ_nβǁ < [(γ₁ γ₂)/(log ψ_n)] for all n, thus giving an effective proof of Littlewood's conjecture for the pair (α, β). Our proofs are elementary and use only standard results from algebraic number theory and the theory of continued fractions.

We passed a transversely cooled sodium atomic beam through an optical standing wave, which channeled the atoms into the intensity minima (maxima) for light tuned below (above) the sodium 32S1/2 → 32P3/2 resonance frequency. The channeled atoms retained their spatial distribution when they were deposited onto the silicon surface. The surface was thus covered by a permanent periodic sodium grating with an area of 1 x 0.1 cm2 and a period of d = 294.3 ±0.3 nm, which is half the wavelength of the standing wave field. The entire grating was completed in 10 min. We measured the period of the grating by backscattering weak probe light. The probe light had a wavelength λ, which we varied between 5580 and 5800 Å, and was incident at an angle ϕ relative to the normal. There is constructive interference when the angle θ with respect to the normal to the grating is such that λ = d(sin θ + sin ϕ). The dependence of θ on ϕ allowed us to measure d. Semiclassical theory suggests the width of the lines in the grating could be of the order of 100 Å.

The complexity of technical subjects related to the construction field, in Architecture Studies, is elevated in the first years of study due to the lack of fundamental knowledge of terms and properties in construction systems. Students enter their first year without previous fundamental training, in most cases, without having visited a construction site, generating pedagogical challenges. Given the complexity of the subject, traditional teaching causes mechanisation and memorisation, erroneous on many occasions, replacing reasoning and understanding of the usefulness of each system and constructive solution. Taking advantage of the entertaining and dynamic potential of Social Networks in students, this work analyses the design, development, and experimentation of a flipped classroom teaching model that combines explanations of main concepts and construction systems through recorded and explained videos by the students themselves, in search of an active, playful and motivating learning methodology. La complejidad de las asignaturas técnicas, correspondientes al área de Construcción del Grado en Estudios de Arquitectura, es incrementada aún más durante los primeros cursos por el desconocimiento de términos y propiedades fundamentales en los sistemas constructivos. Los alumnos acceden al primer curso sin una formación constructiva básica, en una mayoría de casos sin haber visitado una obra, lo cual genera un reto pedagógico, pues ante la complejidad de la materia, la enseñanza tradicional provoca la mecanización y memorización, en muchas ocasiones errónea, sustituyendo al razonamiento y compresión de la utilidad de cada sistema y solución constructiva. Aprovechando potencial lúdico y dinamizador de las Redes Sociales en los estudiantes, este trabajo analiza el diseño, desarrollo y experimentación desarrollada para integrar explicaciones de conceptos y sistemas constructivos mediante vídeos grabados y explicados por los propios estudiantes en la constitución de una metodología activa, lúdica y motivadora de aula invertida.

Construction inspection is a critical component of INDOT’s quality assurance (QA) program. Upon receiving an inspection notice/assignment, INDOT inspectors review the plans and specifications to identify the construction quality requirements and conduct their inspections accordingly. This manual approach to gathering inspection requirements from textual documents is time-consuming, subjective, and error-prone. This project addresses this critical issue by developing an inspection requirements database along with a set of tools to automatically gather the inspection requirements and provide field crews with customized construction checklists during the inspection with the specifics of what to check, when to check, and how to check, as well as the risks and the actions to take when noncompliance is encountered. This newly developed toolset eliminates the manual effort required to acquire construction requirements, which will enhance the efficiency of the construction inspection process at INDOT. It also enables the incorporation of field-collected data to automate future compliance checking and facilitate construction documentation.

A phased study of the dike fields within the Vicksburg and Memphis Districts of the US Army Corps of Engineers was conducted to document the channel morphology trends since dike construction on the Lower Mississippi River (LMR). This included the development of the hydrographic survey database and methodology utilized to identify changes in channel geometry in response to dike construction. A subsequent report will provide further refinements to the approach and results of the comprehensive assessment. Recent Mississippi River Geomorphology and Potamology program efforts have employed the database developed by Mr. Steve Cobb to assess the geomorphic changes in 21 dike systems along the LMR. Previous studies using this database have indicated that the dike fields have not caused a loss of channel capacity. Furthermore, these efforts suggested that the trends in the dike fields are closely related to the long-term geomorphic trends along the LMR. Previous efforts using the Cobb database provided considerable insight into the dike effects on the LMR, but they were limited spatially and temporally. In this study, a database and protocols were developed to allow for a more robust assessment of dike field impacts and to extend the spatial and temporal extents of the analysis.

A combined field and laboratory study was conducted to identify factors contributing to sliver spall of concrete pavements and recommend avenues for prevention. In this study, spall density maps of eight airfields were created, and cores were taken for petrographic analysis. A companion laboratory study evaluated nondestructive testing equipment for identifying concrete prone to sliver spalling. Concrete mix designs with good and poor performance were analyzed for trends in mixture proportioning and aggregate gradation. Spall density mapping indicated sliver spalling was more likely to occur on longitudinal joints and that the distress was not solely a material or mixture design–related issue. The laboratory study concluded that surface resistivity measurements were able to differentiate edge- finishing techniques (normal versus overworked, mortar-rich edge) after seven days of curing. An analysis of particle packing theory and mixture proportioning trends showed there was substantial overlap in the gradations for good and poor performing pavement. Thus, acceptable mixture designs can produce poor quality pavement if not constructed properly. The main contributors to early age sliver spalling of concrete airfield pavement occur during pavement construction.

Complex military operations often result in U.S. forces remaining at deployed locations for long periods. In such cases, more sustaina-ble facilities are required to better accommodate and protect forward-deployed forces. Current efforts to develop safer, more sustaina-ble operating facilities for contingency bases involve construction activities that require a redesign of the types and characteristics of the structures constructed, that reduce the resources required to build, and that decrease the resources needed to operate and maintain the completed facilities. The Automated Construction of Expeditionary Structures (ACES) project was undertaken to develop the capa-bility to “print” custom-designed expeditionary structures on demand, in the field, using locally available materials with the minimum number of personnel. This work investigated large-scale automated “additive construction” (i.e., 3D printing with concrete) for con-struction applications. This report, which documents ACES materials and testing, is one of four technical reports, each of which details a major area of the ACES research project, its research processes, and its associated results. There major areas include System Require-ments, Construction, and Performance; Energy and Modeling; Materials and Testing; Architectural and Structural Analysis.

The objective of this project was to develop guidelines pertaining to these issues and how they should be addressed in the field. A review of recent incidents involving girth weld failures in newly-constructed pipelines indicate that all incidents involved production welds in pipelines constructed using conventional techniques (i.e., using cellulosic-coated electrodes) � particularly welds at wall thickness transitions � and repair and tie-in welds made using cellulosic-coated electrodes in pipelines otherwise constructed using mechanized gas-metal arc welding. None of the recent failures involved mechanized gas-metal arc welds. Many of these failures were attributed, at least in part, to hydrogen-assisted cracking and high stresses. A review of the factors related to hydrogen cracking in modern pipeline girth welds indicates that managing weld hydrogen levels and the stresses acting on these welds is the most appropriate strategy for controlling the risk of hydrogen-assisted cracking, which became the primary focus of this project.

Poly-,Per-fluorinated alkyl substances (PFAS) are versatile chemicals that were incorporated in a wide range of products. One of their most important use was in aqueous film-forming foams for fighting liquid fuel fires. PFAS compounds have recently been identified as potential environmental contaminants. In the United States there are hundreds of potential military sites with PFAS contamination. The ERDC designed and constructed a mobile treatment system to address small sites (250,000 gallons or less) and as a platform to field test new adsorptive media. The PFAS Effluent Treatment System (PETS) has cartridge filters to remove sediments and a granular activated carbon (GAC) media filter to remove organic compounds that might compete with PFAS in the ion exchange process, although it may also remove PFAS too. The last process is an ion exchange resin specifically designed to remove PFAS to a target level of 70 ng/L or less (equivalent to the US Environmental Protection Agency (EPA) Drinking Water Health Advisory). The system was tested at Hurlburt Field, a US Air Force facility in Florida and at Naval Support Activity (NSA) Mid-South in Millington, TN.

During recent pipeline construction seasons, there have been reported difficulties in cold field bending different joints of line pipe produced by the same manufacturer during a particular production run. These difficulties were typically manifested as an inability to achieve bends of one to one-and-a-half degrees per diameter in the pipe without forming ripples. Some of these problems could be traced to poor bending practices, i.e., inappropriate bending machine set-up and operator inexperience. However, it was observed that even with proper machine set-up and experienced operators, some "bad pipe" exists that could not be bent as far as desired without introducing ripples. The objective of this program was to develop an acceptance criterion for an allowable ripple in a gas-transmission pipeline as a result of a cold field bend. Previous acceptance or rejection criteria for ripples in cold field bent pipe were quite subjective. The purpose of this study was to define quantitative limits for acceptable ripple heights that will not compromise the integrity of the pipe during its expected life. In a program discussed at the 9th PRCI/EPRG meeting, the work performed during a joint Australian Pipeline Industry Association/Line Pipe Research Supervisory Committee effort on the mechanics of cold field bending was presented. Based on theoretical and experimental studies, it was found that modern, high-strength, high D/t pipe forms ripples at very low bend angles during cold field bending. Although there was some circumstantial evidence that some amount of rippling can be tolerated in a pipe, there had been no systematic effort to quantify the influence that the ripples have on the long-term integrity of pipe.

Soils containing inorganic compounds are frequently encountered by transportation agencies during construction within the right-of-way, and they pose a threat to human health and the environment. As a result, construction activities may experience project delays and increased costs associated with management of inorganic compounds containing soils required to meet environmental regulations. Recalcitrance of metal-contaminated soils toward conventional treatment technologies is exacerbated in clay or organic content-rich fine-grained soils with low permeability and high sorption capacity because of increased treatment complexity, cost, and duration. The objective of this study was to develop an accelerated in situ electrochemical treatment approach to extract inorganic compounds from fine-grained soils, with the treatment time comparable to excavation and off-site disposal. Three reactor experiments were conducted on samples collected from two borehole locations from a field site in Illinois that contained arsenic (As)(~7.4 mg/kg) and manganese (Mn)(~700 mg/kg). A combination of hydrogen peroxide (H2O2) and/or citrate buffer solution was used to treat the soils. A low-intensity electrical field was applied to soil samples using a bench-scale reactor that resembles field-scale in situ electrochemical systems. For the treatment using 10% H2O2 and citrate buffer solution, average removal of 23% and 8% were achieved for Mn and As, respectively. With 4% H2O2 and citrate buffer, 39% and 24% removal were achieved for Mn and As; while using only citrate buffer as the electrolyte, 49% and 9% removal were achieved for Mn and As, respectively. All chemical regimes adopted in this study reduced the inorganic compound concentrations to below the maximum allowable concentration for Illinois as specified by the Illinois Environmental Protection Agency. The results from this work indicate that electrochemical systems that leverage low concentrations of hydrogen peroxide and citrate buffer can be effective for remediating soils containing manganese and arsenic.

The original objectives of the approved proposal were: 1. To construct a YFP fused Arabidopsis cDNA library in a mammalian expression vector. 2. To infect the library into a host fibroblast cell line and to screen for new cytoskeletal associated proteins using an automated microscope. 3. Isolate the new genes. 4. Characterize their role in plants. The project was approved as a feasibility study to allow proof of concept that would entail building the YFP library and picking up a couple of positive clones using the fluorescent screen. We report here on the construction of the YFP library, the development of the automatic microscope, the establishment of the screen and the isolation of positive clones that are plant cDNAs encoding cytoskeleton associated proteins. The rational underling a screen of plant library in fibroblasts is based on the high conservation of the cytoskeleton building blocks, actin and tubulin, between the two kingdoms (80-90% homology at the level of amino acids sequence). In addition, several publications demonstrated the recognition of mammalian cytoskeleton by plant cytoskeletal binding proteins and vice versa. The major achievements described here are: 1. The development of an automated microscope equipped with fast laser auto-focusing for high magnification and a software controlling 6 dimensions; X, Y position, auto focus, time, color, and the distribution and density of the fields acquired. This system is essential for the high throughput screen. 2. The construction of an extremely competent YFP library efficiently cloned (tens of thousands of clones collected, no empty vectors detected) with all inserts oriented 5't03'. These parameters render it well representative of the whole transcriptome and efficient in "in-frame" fusion to YFP. 3. The strategy developed for the screen allowing the isolation of individual positive cDNA clones following three rounds of microscopic scans. The major conclusion accomplished from the work described here is that the concept of using mammalian host cells for fishing new plant cytoskeletal proteins is feasible and that screening system developed is complete for addressing one of the major bottlenecks of the plant cytoskeleton field: the need for high throughput identification of functionally active cytoskeletal proteins. The new identified plant cytoskeletal proteins isolated in the pilot screen and additional new proteins which will be isolated in a comprehensive screen will shed light on cytoskeletal mediated processes playing a major role in cellular activities such as cell division, morphogenesis, and functioning such as chloroplast positioning, pollen tube and root hair elongation and the movement of guard cells. Therefore, in the long run the screen described here has clear agricultural implications.
