Abstract (sommario):
Abstract Background: Proteasome inhibitors (PIs) are important agents against myeloma, but innate and acquired resistance limit their effectiveness. We previously found Tight junction protein (TJP)-1 to be a modulator and potential biomarker of PI sensitivity, via studies of bortezomib resistant models and clinically annotated gene expression profiling (GEP) databases of patient samples. Also, we showed that its expression reduced EGFR/JAK1/STAT3 pathway activation and proteasome capacity. However, the molecular mechanisms by which TJP1 expression suppressed EGFR signaling and enhanced PI sensitivity were not understood. Methods: We performed co-immunoprecipitation (co-IP) studies to understand interactions of TJP1 with EGFR, and evaluated the impact of various cytokines on proteasome capacity. Also, genetic and pharmacologic approaches were used to modulate EGFR/JAK/STAT activity to determine the effects on PI sensitivity. Finally, gene set enrichment analyses (GSEA) were performed to determine if EGFR activation signatures could be identified in primary samples, and if they correlated with outcomes. Results: Previous studies of TJP1 focused on myeloma models, but since bortezomib is also approved for mantle cell lymphoma, we sought to determine if TJP1 influenced sensitivity here as well. Suppression of TJP1 using shRNAs reduced the sensitivity of JeKo-1 and MINO cells to bortezomib. In addition, murine embryonic stem cells (mESCs) with TJP1 knockout were more resistant to PIs than was the case for wild-type mESCs. Moreover, the TJP1 knockout mESCs expressed higher levels of both activated EGFR and immunoproteasome subunits PSMB8 and 9, and had greater levels of proteasome chymotrypsin-like (ChT-L) activity. Also, re-introduction of TJP1 into the knockout mESCs ehanced their PI sensitivity, and reduced the ChT-L activity. Co-IP studies showed that IP of TJP1 also led to detection of EGFR, and vice versa. Phospho-specific antibodies showed that TJP1 precipitated predominantly the non-phosphorylated, inactive EGFR. Indeed, when cells were treated with EGF and extracts were subjected to IP for TJP1, reduced levels of EGFR were precipitated. Evaluating other receptor tyrosine kinases, we found that TJP1 precipitated IGF-1R but not IL-6R. Therefore, we treated myeloma cells with EGF, IL-6, or IGF-1; all reduced PI sensitivity, but only EGF enhanced proteasome ChT-L activity and capacity. Moreover, shRNA-mediated EGFR knockdown, or pharmacologic inhibition with erlotininb in myeloma cells reduced JAK1/STAT3 activity, levels of PSMB8 and 9, and ChT-L activity, and enhanced the activity of bortezomib. Since the PSMB8 and PSMB9 promoters have consensus STAT3 binding sites, we knocked down STAT3 and found that this also reduced PSMB8 and 9 and ChT-L activity, and enhanced the efficacy of bortezomib. In a mouse model of lytic bone destruction by xenografted RPMI 8226 cells, the ability of bortezomib to increase bone trabecular volume was blunted by TJP1 suppression, consistent with a lower anti-myeloma effect. Lastly, we used publically available EGF-stimulated and EGFR inhibitor gene signatures to examine the clinically annotated GEP data from myeloma patients. This showed that: 1) TJP1 expression in myeloma samples from patients on the University of Arkansas Total Therapy studies was positively correlated with known signatures of inhibitors of EGFR signaling; 2) in GSEA, signatures of EGF and EGFR signaling were significantly positively enriched when genes were ranked by correlation with TJP1; and 3) an EGFR-related gene set had significant negative correlation with survival duration in the Total Therapy 3 dataset. Conclusions: Taken together, our data support the role of TJP1 as a negative regulator of EGFR signaling through its ability to specifically bind, and possibly stabilize the non-phosphorylated form of EGFR. Furthermore, they demonstrate that TJP1 influences proteasome capacity through EGFR/JAK/STAT signaling, and that approaches to suppress this pathway hold promise to overcome PI resistance and achieve PI sensitization. Finally, they validate a heretofore underappreciated role for EGF/EGFR signaling in myeloma pathobiology and patient outcomes. Disclosures Mulligan: Millennium Pharmaceuticals: Employment. Abstract (sommario):
The 2020 election was a referendum on many pressing issues of the time. COVID-19, climate change, and civil unrest propelled Americans to the voting booth in unprecedented numbers, shattering previous turnout records for both previous Vice President Joe Biden and the incumbent Donald Trump. Biden and the Democratic Party field won, but narrowly. With the Democratic Party ostensibly holding only a slim majority in the House and the Vice Presidency as a tiebreaker in a dead-locked Senate, there have been large cries of a “stolen election” and “rampant voter fraud.” This has prompted several states, usually with Republican majorities, to pass sweeping restrictions on voting. Using both historical and contemporary lenses, this paper investigates the causes, effects, and motivations to curb voting accessibility and redistricting with an emphasis on red and swing states in the aftermath of the 2020 election, as well as potential solutions to combat the push to further restrict voting rights.

Abstract (sommario):
Nematic elastic bodies can develop a gradient of response to heat, light and other stimuli. They then bend and develop curvature in a complex manner. Using the results for a general weak response derived in the preceding paper, we solve for strong spontaneous distortion where bend in one direction causes stretch in another direction if that too is bending, and vice versa. Since stretch is elastically expensive, it can cause suppression of one of the bends (we determine which), thus eliminating Gaussian curvature. This is the spontaneous distortion equivalent of the classical Lamb calculation of the anti-clastic suppression when large distortions are imposed in classical elastica. In practice, spontaneously deforming nematic solids, e.g. in actuation, are in this strong bend limit.

Abstract (sommario):
In the decade following its founding in 1955, the men who led the foreign policy lobby the Committee of One Million Against the Admission of Communist China to the United Nations faced little concerted opposition to their attempts at preventing even the most minor alterations in the U.S. policy of both isolating and containing Communist China. But beginning with the Fulbright Hearings on China in March 1966, the trend of informed opinion moved sharply against them, as liberal Democrats became newly emboldened and moderates in both parties switched sides, inverting the bipartisan consensus against change the Committee relied upon. The 1968 election of former Vice President Richard M. Nixon, who had served alongside Committee hero John Foster Dulles, seemed to offer them newfound hope. But when the “New Nixon” proved unreceptive to the entreaties of his one-time allies, the Committee mounted a furious public relations campaign to rally belatedly the right-wing base and influence public opinion. Its failure illustrated both the limits of power of American conservatives over U.S. foreign policy while détente was ascendant, and the discontinuity in priorities between the Old Right from which the Committee emerged and the New Right that left it behind.

Abstract (sommario):
Abstract Background SIRT1 is a NAD+ dependent histone deacetylase, which has been shown to act as an important regulator of apoptosis, DNA-repair and is involved in the maintenance of genetic integrity under conditions of cellular stress. Beside deacetylation of histones H4K16, SIRT1 has numeral other substrates including KU70, FOXO1 or p53. SIRT1 deacetylates p53 at lysine 382 thereby reducing its transcriptional activity followed by loss of p53 dependent apoptosis in response to cell damage. The activity of SIRT1 is negatively regulated by DBC1 (Deleted in Breast Cancer 1) and involves protein–protein interaction (Kim et al., Nature 2008). Recent reports have demonstrated increased expression of SIRT1 in leukemic cells. Of note, SIRT1 seems to be specifically essential to maintain stem cell activity in CML leukemic stem cells (Li et al, Cancer Cell, 2012), however the role of SIRT1 in acute myeloid leukemia (AML) and in the context of other oncogenic tyrosine kinases is poorly understood. Aims To characterize the role of SIRT1 in acute myeloid leukemia in the context of defined genetic backgrounds. Methods To investigate the functional role of SIRT1 we performed expression analysis studies. Inhibition of SIRT1 was achieved by shRNA experiments and pharmacological targeting. Functional consequences were addressed by immunoblotting, co-immunoprecipitation, cell death assays, gamma-H2AX staining and xenotransplantion experiments. Results SIRT1 protein expression analysis in primary human AML samples (n=20) and cell lines (n=13) showed preferential high expression levels in cells harboring FLT3-ITD mutations or oncogenic KRAS/NRAS. Inhibition of FLT3 signaling using shRNA-mediated knockdown of FLT3 or upon treatment with tyrosine kinase inhibitors caused a time and dose dependent suppression of SIRT1 expression accompanied by an increase in p53 acetylation. Vice versa, overexpression of FLT3-ITD in BaF/3 cells or in several murine leukemia models (e.g. MLL/AF9 +/-FLT3-ITD; AML1/ETO +/-FLT3-ITD) induced significant upregulation of SIRT1. Targeting SIRT1 using the SIRT1/2 inhibitor Tenovin-6 (TV-6) or knockdown of SIRT1 resulted in a slight increase in apoptotic cell death in primary AML samples and cell lines. In contrast, inhibition of SIRT1 significantly sensitized to FLT3 inhibitor therapy. Of note, the combination of TV-6 and midostaurin inhibited colony growth and replating activity in MLL-AF9 or AML1-ETO cells harboring FLT3-ITD mutations. Further, knockdown of SIRT1 in the AML cell line MV4-11 expressing a doxycycline-regulated SIRT-shRNA resulted in prolonged survival in a xenotransplantation model. The observed effects prompted us to investigate whether mutated FLT3 is directly involved in the regulation of SIRT1. Inhibition of FLT3-ITD kinase activity caused an increased phosphorylation of the physiological SIRT1-inhibitor DBC1 followed by binding of SIRT1 as revealed by co-immunoprecipitation experiments. We also noticed a slight increase in ATM/ATR phosphorylation and an increase in ATM-DBC1 binding upon FLT3-ITD inhibition indicating SIRT1 inhibition via the ATM-DBC1-SIRT1 axis. These effects were substantially enhanced upon treatment with genotoxic agents (cytarabine; irradiation) in combination with midostaurin compared to either agent alone and were accompanied by increased p53 acetylation and an increased number of gamma-H2AX foci. Conclusion SIRT1 is overexpressed in AML cells and partially regulated by FLT3-ITD or oncogenic RAS. In FLT3-ITD mutated cells SIRT1 is constitutively activated, likely via the inhibition of the physiological inhibitor DBC1. Pharmacologic targeting of SIRT1 sensitizes AML cells to TKI therapy or genotoxic agents and partially restored the FLT3-ITD associated defective stress response pathway ATM-DBC1-SIRT1 resulting in activation of p53. Abstract (sommario):
On the occasion of continuation of The International Journal of Children’s Rights with two eminent new Editors-in-Chief, the founder of this Journal (Dr. Philip Veerman) interviewed a rising star of children’s rights: Dr. Benyam Dawit Mezmur (born in 1980 in Addis Ababa). Mezmur was elected at 30 years of age to the African Committee of Experts on the Rights and Welfare of the Child (acerwc Committee) of the African Union (au). Dr. Mezmur stands out not only by his great diplomatic skills, but also because he is a real builder. For instance, during his tenure as Chairperson of acerwc this Committee obtained greater visibility for its work and secured a separate budget from the African Union Commission. Dr. Mezmur’s vision and hard work contributed much to a new situation in which many colleagues in the children’s rights field recognise that the acerwc Committee is now doing significant and unique work of which the added value (next to the un Committee on the Rights of the Child, the crc Committee) has become clear. Dr. Mezmur was elected on 18 December 2012 (at 32 years of age) as a member of the crc Committee and in May 2013 he became the Vice-Chair of the crc Committee. From May 2015 until May 2017 he served with great dedication as Chairperson of the crc Committee. This unique combination of two different key posts led to interesting observations on developments in the children’s rights field. Through the interview we have a rare look behind the scenes of two important committees on children’s rights.

Abstract (sommario):
What are the driving forces behind the selection of individual Members of the European Parliament for committee leadership positions? To shed more light on this process, the article analyses an original dataset comprising biographical and behavioural information on all 751 Members of the European Parliament who assumed office in 2014. We show that the choice of committee chairs and vice-chairs is not determined by loyalty to party groups or national delegations. Instead, we demonstrate that Members of the European Parliament’ career ambitions and their ability to demonstrate relevant experience play a key role. These findings have important implications for our understanding of the power of national parties and European party groups and for the emerging literature on political career patterns at the European level.

Abstract (sommario):
1. Responses of 89 single olfactory bulb neurons from 43 channel catfish, Ictalurus punctatus, to amino acid odorants were recorded in vivo simultaneously with the electro-olfactogram (EOG). Recording time for individual neurons ranged from 16 to 344 min. The averaged spontaneous frequency ranged from x003C; 1 to 16 action potentials/s with a mean frequency of 5.2 +/- 3.6 (SD) action potentials/s. 2. Histological examinations of carbocyanine dye 1,1'diocadecyl-3,3,3',3'-tetramethylindocarbocyanine perchlorate (DiI)-labeled olfactory bulbs and electrical stimulation of the olfactory tracts in a subset of experiments suggested that the majority of the recorded olfactory bulb neurons in this study were mitral cells. 3. Olfactory bulb neurons responded to amino acids with either an excitation or suppression of the background neural activity. Of the 337 stimulus applications, 28% of the responses were excitatory, and 33% were suppressive. The approximately 1:1 ratio of excitatory to suppressive responses for all stimulus applications suggests that suppressive responses also play important roles in the coding of odorant information in the channel catfish. 4. Responses of single olfactory bulb neurons were highly reproducible over time (up to 5 h). Responses to any amino acid never changed from excitation to suppression, or vice versa over time. 5. Single olfactory bulb neurons responded with excitation or suppression to more than one amino acid previously indicated to bind to independent receptors. 6. Estimated threshold concentrations for activation of an olfactory bulb neuron ranged from 10(-7) to 10(-3) M and were different from neuron to neuron for a particular stimulus and from stimulus to stimulus for a particular neuron. Responses of single olfactory bulb neurons to a given amino acid did not change from excitation to suppression, or vice versa, across different suprathreshold concentrations.

Abstract (sommario):
AbstractRecurrent proposals to establish a constitutional supervisory committee have been pertinaciously rejected in spite of widespread recognition of the Chinese Constitution’s ineffectiveness. And yet, the Hong Kong Basic Law Committee has long epitomized in practice a prototypic form of constitutional supervision. Vested with quasi-judicial competences, the Committee seemed destined for a central role under the “One Country, Two Systems” arrangement. The tight secrecy imposed on its proceedings and the suppression of its potential to act consistently and with a distinct identity have fatally undermined the Committee’s ability to modulate constitutional tensions by way of coordinating expectations of the Basic Law’s proper meaning. The experience of the Basic Law Committee reveals the recalcitrance of the Party-state toward constitutional interpretation by any specialized body, even one whose powers are heavily circumscribed and whose membership is tightly controlled.

Abstract (sommario):
Several studies have shown that blind humans can gather spatial information through echolocation. However, when localizing sound sources, the precedence effect suppresses spatial information of echoes, and thereby conflicts with effective echolocation. This study investigates the interaction of echolocation and echo suppression in terms of discrimination suppression in virtual acoustic space. In the ‘Listening’ experiment, sighted subjects discriminated between positions of a single sound source, the leading or the lagging of two sources, respectively. In the ‘Echolocation’ experiment, the sources were replaced by reflectors. Here, the same subjects evaluated echoes generated in real time from self-produced vocalizations and thereby discriminated between positions of a single reflector, the leading or the lagging of two reflectors, respectively. Two key results were observed. First, sighted subjects can learn to discriminate positions of reflective surfaces echo-acoustically with accuracy comparable to sound source discrimination. Second, in the Listening experiment, the presence of the leading source affected discrimination of lagging sources much more than vice versa. In the Echolocation experiment, however, the presence of both the lead and the lag strongly affected discrimination. These data show that the classically described asymmetry in the perception of leading and lagging sounds is strongly diminished in an echolocation task. Additional control experiments showed that the effect is owing to both the direct sound of the vocalization that precedes the echoes and owing to the fact that the subjects actively vocalize in the echolocation task.

From a career in the aeronautical and electronics industry, rising from the shop floor and university to management, Kenneth Warren entered Parliament in 1970. During his time in Parliament he has been Chairman of the Conservative Back-Bench Aviation Committee and of the Western European Union's Science and Technology Committee. For four years he was Parliamentary Private Secretary to Sir Keith Joseph in the Department of Trade and Industry and then Education and Science. Currently he is Chairman of the British Russian Parliamentary Group as well as the Select Committee on Trade and Industry. Having visited some eighty countries, led parliamentary delegations overseas on a number of occasions and contributed papers at technical conferences in the United Kingdom, the United States, Holland, France, Japan and Hungary, he has a world-wide interest in the development of engineering and markets for technology. Communication between industry and politics and vice versa is not at the level of quality required in a major supplier/customer relationship. The author will develop a thesis on how this interface can be eroded. He will base his lecture on a background of international experience in politics and industry, including the Chairmanship of the Select Committee on Trade and Industry and as one of the four qualified engineers in the House of Commons.

Education management itself is seen as a process needed in the world of education to achieve the goals of education itself. The purpose of this paper is to explore school financial management in Vocational High Schools (SMK), as well as to find out the effectiveness of existing financial management in schools. The school used as the object was SMK N 2 Balikpapan with research informants namely the school principal, vice principal for finance, school treasurer, and committee. Based on the research, it was found that the main source of school finance came from BOS funds. The management process begins with deliberations and holding meetings of all teachers and employees with the process of preparing the RKAS and preparing the RAB, based on the plans needed by the school which will first be considered by the School Committee. Realization of school financial management sourced from BOS funds and school committee funds in Vocational High Schools for the 2021/2022 academic year has been carried out efficiently financial While the inhibiting factor is still a lack of BOS funds and school committee funds provided to support the process of school facilities and infrastructure or in other words not in accordance with the previous plan for its realization.

Abstract: The Maharaja Sayajirao University, Baroda's flagship programme is the Directorate of Students' Welfare (DSW). It was established by the then Vice Chancellor, Prof. Yogesh Singh, and higher authorities to provide financial aid to the University's economically disadvantaged students. The primary goal of DSW is to provide financial assistance to 'genuine but economically disadvantaged' future assets of the country who are pursuing their studies at the Maharaja Sayajirao University of Baroda. DSW was established in the academic year 2012-13 with the goal of putting the haves and have-nots on an equal footing. Students who apply for a scholarship must have a family income of less than 3.5 lakh rupees per year and have received 55 percent in their previous year's exam. The online application can be made on the Maharaja Sayajirao University website (https://msubaroda.ac.in/dsw) with all the supporting papers such as Aadhar card, Bank Account details, Electricity bills, and so on. The DSW collaborates with members of its Core Committee and Faculty Coordinators. The Faculty Committee, which consists of the Dean, three professors, the dean of students, and the dean of sports, conducts the initial examination. The core committee then verifies the qualifications and assigns Recommendation, Strongly Recommendation, or Not Recommended ratings based on the students' merits. Each academic year, the committee meets to debate the agenda, make plans, and act in accordance with the Vice Chancellor sir's instructions. Every year, needy and genuine students are discovered through a methodical process that culminates in a personal interview in which the students are asked for personal information and all documents are verified. If during the interview the committee feels the need to go for home visit for further verification. After they are shortlisted, the funds are directly transferred to their bank accounts. For educational year 2020-21 the method was completely online due to Covid situation. Total 2925 students had done registration out of that 2621 application was completed with necessary documents. A total Rs. 90, 40, 490 Lakh has been given to 2004 students this educational year. If we see the data we have helped 1662 Grant in Aid students and 342 Higher payment students as they have received the scholarship. Also gender wise distribution is seen it is 1181 boys and 823 girls were benefitted with DSW scholarship. Keyword: Directorate of Students’ Welfare, Scholarship, Higher Education
