<abstract> <p>High-fat diets have been associated with lower gut and fecal abundances of genus <italic>Bifidobacterium</italic>. Here, we investigated whether commonly consumed dietary free fatty acids have any detrimental effect on the growth of <italic>B. adolescentis</italic>, <italic>B. bifidum</italic>, and <italic>B. longum</italic>. We found that the presence of free fatty acids in the medium inhibits the growth of <italic>Bifidobacterium</italic> species to a varying degree, with capric (C10:0), oleic (C18:1), and linoleic (C18:2) acids displaying the largest effect. In comparison, free fatty acids did not affect the growth of <italic>Escherichia coli</italic>. When fats were added as a mixture of mono- and diacylglycerols, the inhibitory effect on <italic>Bifidobacterium</italic> growth was abolished.</p> </abstract>

Abstract (sommario):
Based on the first widely presented in Russia exhibition of works by the Russian emigrant artist Konstantin Ivanovich Gorbatov (1876–1945) organized in 2021 by the State Museum of History and Art ‘New Jerusalem’ with the participation of museum and private collections, the author analyzes a part of the master’s pictorial heritage dedicated to Italy. A graduate of the Imperial Academy of Arts and a follower of the Russian impressionist tradition, Gorbatov first came to Italy during his retirement trip in 1912. There he was able to feel the power of color and the influence of the light and aerial environment on the transformation of color, and forever remained faithful to the Italian landscape. Having left their homeland in 1922, Gorbatov and his wife settled in Berlin but made numerous journeys to Italy, where he always worked a lot. While staying in Capri, he would travel along the Italian coast, creating his chronicle of charming towns scattered on the coastal cliffs and poetic landscapes inspired by the emerald color of the sea and the bright sun. The subject of the research is the paintings by K.I. Gorbatov dedicated to Italy and his Notes on Art and Life that provide comments on the peculiarities of the master’s perception of nature, vision of color and balance of colors, and his work with color. The research aims to study K.I. Gorbatov’s works in the context of life and work of the Russian artistic emigration in Europe in the second quarter of the 20th century through the analysis of his Italian landscapes, and to analyze the master’s creative method using archival materials. This will contribute to raising understanding and awareness of his art among Russian viewers who were deprived of the opportunity to familiarize themselves with the artist’s work for a long time. The development of the declared research topic involves the application of the interdisciplinary approach, archival, empirical, and iconographic methods, and comparative, artistic and stylistic analyses.

CAPRI is a member of the GAP1 family of GTPase-activating proteins (GAPs) for small G proteins. It is known to function as an amplitude sensor for intracellular Ca2+ levels stimulated by extracellular signals and has a catalytic domain with dual RasGAP and RapGAP activities. Here, we have investigated the mechanism that switches CAPRI between its two GAP activities. We demonstrate that CAPRI forms homodimers in vitro and in vivo in a Ca2+-dependent manner. The site required for dimerization was pinpointed by deletion and point mutations to a helix motif that forms a hydrophobic face in the extreme C-terminal tail of the CAPRI protein. Deletion of this helix motif abolished dimer formation but did not affect translocation of CAPRI to the plasma membrane upon cell stimulation with histamine. We found that dimeric and monomeric CAPRI coexist in cells and that the ratio of dimeric to monomeric CAPRI increases upon cell stimulation with histamine. Free Ca2+ at physiologically relevant concentrations was both necessary and sufficient for dimer formation. Importantly, the monomeric and dimeric forms of CAPRI exhibited differential GAP activities in vivo; the wild-type form of CAPRI had stronger RapGAP activity than RasGAP activity, whereas a monomeric CAPRI mutant showed stronger RasGAP than RapGAP activity. These results demonstrate that CAPRI switches between its dual GAP roles by forming monomers or homodimers through a process regulated by Ca2+. We propose that Ca2+-dependent dimerization of CAPRI may serve to coordinate Ras and Rap1 signaling pathways.

The imposing and romantic remains of the Tiberian palace on the eastern heights of Capri are now visited by relatively few people, and it is often possible to wander among the ruins for an hour or more without meeting another soul. Yet for about a decade, from A.D. 27 to 37, this villa seems to have been the principal residence of the emperor of the Roman world, and Capri, together with Rome, the administrative centre of that world. As imperial residence, as administrative centre, and as a typical Campanian villa, this and the other imperial buildings on Capri must have been thronged with people, from the emperor's family and friends to the palace staff to administrative personnel. It is the purpose of this paper to consider Tiberius' stay on Capri in various contexts, especially that of the people who were on the island with him, in order to understand more clearly what his life there was like.

Some migratory birds have advanced their spring arrival to Northern Europe, possibly by increasing the speed of migration through Europe in response to increased temperature en route. In this paper we compare the phenology of spring arrival of seven trans-Saharan migrants along their migration route and test for patterns indicating that migration speed varied over the season using long-term data collected on the Italian island of Capri and at Ottenby Bird Observatory, Sweden. There was a linear relationship between median arrival dates on Capri and at Ottenby. The slope was not significantly different from one. On average, the seven species arrived 15 days later at Ottenby compared to Capri. There was a (non-significant) negative relationship between the species-specific arrival dates at Capri and the differences in median arrival dates between Capri and Ottenby, possibly indicating a tendency towards faster migration through Europe later in the season. To what extent different species are able to speed up their migration to benefit from the advancement of spring events is unknown.

The versatility of Ca2+ as a second messenger lies in the complex manner in which Ca2+ signals are generated. How information contained within the Ca2+ code is interpreted underlies cell function. Recently, we identified CAPRI and RASAL as related Ca2+-triggered Ras GTPase-activating proteins. RASAL tracks agonist-stimulated Ca2+ oscillations by repetitively associating with the plasma membrane, yet CAPRI displays a long-lasting Ca2+-triggered translocation that is refractory to cytosolic Ca2+ oscillations. CAPRI behavior is Ca2+- and C2 domain–dependent but sustained recruitment is predominantly Ca2+ independent, necessitating integration of Ca2+ by the C2 domains with agonist-evoked plasma membrane interaction sites for the pleckstrin homology domain. Using an assay to monitor Ras activity in real time, we correlate the spatial and temporal translocation of CAPRI with the deactivation of H-Ras. CAPRI seems to low-pass filter the Ca2+ signal, converting different intensities of stimulation into different durations of Ras activity in contrast to the preservation of Ca2+ frequency information by RASAL, suggesting sophisticated modes of Ca2+-regulated Ras deactivation.

Background: Manual data collection is still the gold standard for disease-specific patient registries. However, CAPRI-3 uses text mining (an artificial intelligence (AI) technology) for patient identification and data collection. The aim of this study is to demonstrate the reliability and efficiency of this AI-driven approach. Methods: CAPRI-3 is an observational retrospective multicenter cohort registry on metastatic prostate cancer. We tested the patient-identification algorithm and automated data extraction through manual validation of the same patients in two pilots in 2019 and 2022. Results: Pilot one identified 2030 patients and pilot two 9464 patients. The negative predictive value of the algorithm was maximized to prevent false exclusions and reached 94.8%. The completeness and accuracy of the automated data extraction were 92.3% or higher, except for date fields and inaccessible data (images/pdf) (10–88.9%). Additional manual quality control took over 3 h less time per patient than the original fully manual CAPRI registry (105 vs. 300 min). Conclusions: The CAPRI-3 patient-identification algorithm is a sound replacement for excluding ineligible candidates. The AI-driven data extraction is largely accurate and complete, but manual quality control is needed for less reliable and inaccessible data. Overall, the AI-driven approach of the CAPRI-3 registry is reliable and timesaving.
