Abstract (sommario):

A study on the “Phenetic analysis of Capsicum annuum L. and Capsicum frutescens L. “ has been conducted from June to August 2012. The aim of the research is to know the relationship amoung big chillis and small chillis based on morphology. The samples were taken from 5 villages in Sukamantri Sub-district, Ciamis. The method used in this research was explorative survey with purposive random sampling. The character data of big chili and small chili morphologies were analyzed descriptively to determine the relationship using UPGMA (Unweighted Pair Group Mean Arithmetic) methods. The result of this study showed that there were seven cultivars of big chilli, namely C. annuum „Hot Chili‟, Gada, Fantastik, Keriting TM 888, Tanjung 2, Keriting TM 999, Hot Beauty, and four cultivars of small chilli namely C. frutescens „Cakra Ungu‟, Cakra Hijau, Bendot, and Cakra Putih. The fenogram showed that there were five groups, two groups of C. annuum and three groups of C. frutescens. The first group consisted of C. annuum „Hot Chili‟, Keriting TM 888, Fantastik, and Tanjung 2. The second group consisted of C. annuum „Gada‟, Hot Beauty, and Keriting TM 999. The third group was C. frutescens „Bendot‟. The fourth group consisted of C. frutescens „Cakra Putih‟, and C. frutescens „Cakra Hijau‟. The fifth group was C. frutescens „Ungu‟. The closest relationship was between C. annuum „Keriting TM 999‟ and C. annuum „Hot Beauty‟ and the farthest relationship was between C. frutescens „Bendot‟ and C. frutescens „Ungu‟.

The influence of four arbuscular mycorrhizal fungi (AMF) (Glomus arenarium, G. caledonium, G. irregulare, and G. mosseae) and a root endophyte species (Piriformospora indica – Sebacinales) was investigated on four “day-neutral” strawberry (Fragaria×ananassa Duch.) cultivars (Albion, Charlotte, Mara des Bois, and Seascape) for their tolerance to salt stress. Fungal symbiosis tended to benefit strawberry plants in their tolerance to salinity, confirming the potential use of mycorrhizal biotechnology in horticulture in arid areas.

Penelitian ini bertujuan untuk mempelajari pengaruh lama blansir dan suhu pengeringan terhadap kualitas bubuk cabai merah varietas TM-999. Cabai merah merupakan komoditas hortikulturabernilai ekonomi tinggi yang digunakan luas dalam industri makanan dan farmasi. Cabai ini kayaakan nutrisi penting seperti karbohidrat, lemak, protein, kalsium, vitamin A, B1, dan C, serta memiliki potensi sebagai agen anti kanker berkat kandungan lasparaginase. Penelitian dilakukandi Laboratorium Teknologi Benih Jurusan Agroteknologi Fakultas Pertanian Universitas SyiahKuala, Banda Aceh, selama Februari hingga Mei 2023. Penggunaan berbagai peralatan laboratorium seperti oven, kipas angin, ayakan 60 mesh, dan lainnya, mendukung penelitian ini. Sementara itu, bahan yang digunakan meliputi 9 kilogram cabai merah varietas TM-999 yang dipanen setelah mencapai usia 90 hari, serta bahan kimia tertentu. Penelitian mengikuti desain Rancangan Acak Lengkap (RAL) dengan variasi lama blansir (0, 2, dan 3 menit) dan suhu pengeringan (50°C, 55°C, dan 60°C). Hasil penelitian menunjukkan bahwa lama blansir berpengaruh signifikan terhadap warna a dan aroma, dengan lama blansir 3 menit memberikan kualitas bubuk cabai merah optimal. Suhu pengeringan berpengaruh pada kadar vitamin C, dengan suhu 50°C menjadi yang terbaik. Selain itu, penelitian ini tidak menemukan interaksi yang signifikan antara lama blansir dan suhu pengeringan terhadap kualitas cabai merah. Hasil ini memberikan panduan berharga untuk meningkatkan proses pengeringan dan kualitas bubuk cabai merah dalam aplikasi industri pangan dan farmasi.

Penelitian ini bertujuan untuk mengetahui pengaruh lama fermentasi kulit pisang kepok (Musa paradisiaca L.) terhadap pertumbuhan dan kelangsungan hidup ikan bandeng (Chanos chanos). Penelitian ini dilaksanakan dalam kurun waktu 2 bulan di Tambak Bandeng Desa Bipolo. Lama waktu fermentasi yang diuji di Laboratorium Kimia Pakan, Fakultas Peternakan Kelautan dan Perikanan Universitas Nusa Cendana yaitu 6 hari, 8 hari, dan 10 hari. Penelitian ini menggunakan metode Rancangan Acak Lengkap (RAL). Dalam penelitian ini terdapat 3 perlakuan masing-masing 3 ulangan dan kontrol. Perlakuan yang dimaksud adalah Perlakuan A (Kulit pisang fermentasi 30 hari + pellet FF-999 30 hari berikutnya), Perlakuan B (Pellet FF-999 30 hari + kulit pisang fermentasi 30 hari berikutnya), Perlakuan C (Pemberian kulit pisang hasil fermentasi selama 60 hari) dan Kontrol (100% pemberian pellet FF-999 selama 60 hari). Hasil uji laboratorium menunjukkan bahwa lama fermentasi kulit pisang terbaik terdapat pada hari ke 10 dengan kandungan protein tertinggi yaitu sebesar 11,015%. Lama waktu fermentasi 10 hari digunakan untuk fermentasi kulit pisang sebagai pakan untuk menunjang pertumbuhan dan kelangsungan hidup ikan bandeng. Hasil penelitian menunjukkan bahwa adanya pengaruh lama fermentasi kulit pisang 10 hari terhadap pertumbuhan berat mutlak (p<0.05), namun tidak berpengaruh terhadap kelangsungan hidup ikan bandeng. Kata Kunci : Fermentasi, Kulit Pisang Kepok, Ikan Bandeng, Pertumbuhan, Kelangsungan Hidup

AbstractHomoclinic and heteroclinic solutions to a standard hepatitis C virus (HCV) evolution model described by T. C. Reluga, H. Dahari and A. S. Perelson, (SIAM J. Appl. Math., 69 (2009), pp. 999–1023) are considered in this paper. Inverse balancing and generalized differential techniques enable derivation of necessary and sufficient existence conditions for homoclinic/heteroclinic solutions in the considered system. It is shown that homoclinic/heteroclinic solutions do appear when the considered system describes biologically significant evolution. Furthermore, it is demonstrated that the hepatitis C virus evolution model is structurally stable in the topological sense and does maintain homoclinic/heteroclinic solutions as diffusive coupling coefficients tend to zero. Computational experiments are used to illustrate the dynamics of such solutions in the hepatitis C evolution model.

Mouse sperm-specific lactate dehydrogenase-C (LDH-C) cDNA was cloned and sequenced from lambda gt11 expression library. The LDH-C cDNA insert of 1236 bp consists of the protein-coding sequence (999 bp), the 5′ (54 bp) and 3′ (113 bp) non-coding regions, and the poly(A) tail (70 bp). The Northern blot analysis of poly(A)-containing RNAs from mouse testes and liver indicates that the LDH-C gene is expressed in testes but not in liver, and that its mRNA is approx. 1400 nucleotides in length. The nucleotide and amino acid sequences of the mouse LDH-C cDNA show 73% and 72% homologies, respectively, with those of the mouse LDH-A. The Southern blot analysis of genomic DNAs from mouse liver and human placenta indicates the presence of multiple LDH-C gene-related sequences.

Abstract (sommario):

The ring D-modified gibberellin [GA], 16,17-dihydro GA5, can retard stem growth in Lolium temulentum L. while promoting flowering (Evans et al., 1994, Planta193, 107–114). Using [1,2,3-3 H]GA20 to study the final biosynthetic step to GA1 (a known effector of shoot elongation in higher plants), it was shown that C-3b-hydroxylation of GA20 to GA1 is blocked by 16,17-dihydro GA5 but is little affected by GA5. Another late-stage biosynthetic inhibitor, the acylcyclohexanedione, LAB 198 999, also blocked GA1 formation. Furthermore, endogenous levels of GA20 built up after application of 16,17-dihydro GA5. Consequently, growth retardation by 16,17-dihydro GA5 and LAB 198 999 is likely to be the result of their inhibition of GA20 3b-hydroxylation to GA1. Another fate for GA20 in Lolium is its C-2b-hydroxylation to growth-inactive GA29. This conversion was also inhibited by 16,17-dihydro GA5 but less so by LAB 198 999. The analogous step involving 2b-hydroxylation of GA1 to GA8 appeared to be insensitive to either growth retardant. When [3H]GA20 was injected into the cavity within the young intact sheathing leaves, there was an appreciable metabolism of this GA20 to GA1 and thence to GA8 (ca 10% and 30% respectively within 5 h). For excised shoot tips, however, [3H]GA20 was converted rapidly and virtually completely to GA29 in 3–5 h. Interestingly, with these excised shoot tips, GA3 and GA5 as well as 16,17-dihydro GA5 when applied via the agar strongly inhibited 2b-hydroxylation of GA20 to GA29. In contrast, while 16,17-dihydro GA5 blocked GA20 metabolism to GA29 in intact sheath/stem tissue, this conversion was not inhibited by GA5. These differences in structural specificity for GAs which inhibit 2b-hydroxylation as opposed to 3b-hydroxylation are in accordance with these two Ring-A hydroxylation steps being catalysed by different enzymes. Finally, the differences in GA20 metabolism between intact versus excised tissue raise the possibility that tissue wounding with excision enhanced the activity of the GA20 2b-hydroxylase(s).

As illustrated by the work of Ralph White, traditional methods of electrochemical engineering are based on human intelligence: Mathematical models encoding physical hypotheses are proposed, tested against experimental data and refined by fitting adjustable parameters. Artificial intelligence is beginning to challenge this paradigm, since predictions can be made directly from data without the need for models, but such knowledge is often not transferrable to new situations. This talk will present a hybrid approach of solving PDE-constrained inverse problems to derive new multiscale models, in the context of Li-ion batteries. Examples include learning models of reaction kinetics and surface heterogeneities from x-ray adsorption images of phase separation in lithium iron phosphate nanoparticles [1], x-ray diffraction spectra for fictitious phase separation in nickel-rich oxide porous electrodes [2], and optical videos of lithium plating on graphite particles [3]. ABSTRACT Hepatitis C virus (HCV) NS5A has been reported to be important for the establishment of replication by adaptive mutations or localization, although its role in viral replication remains unclear. It was previously reported that NS5A interacts with NS5B via two regions of NS5A in the isolate JK-1 and modulates the activity of NS5B RdRp (Y. Shirota et al., J. Biol. Chem., 277:11149-11155, 2002), but the biological significance of this interaction has not been determined. In this study, we addressed the effect of this interaction on HCV RNA replication with an HCV replicon system derived from the isolate M1LE (H. Kishine et al., Biochem. Biophys. Res. Commun., 293:993-999, 2002). We constructed three internal deletion mutants, M1LE/5Adel-1 and M1LE/5Adel-2, each encoding NS5A which cannot bind NS5B, and M1LE/5Adel-3, encoding NS5A that can bind NS5B. After transfection into Huh-7 cells, M1LE/5Adel-3 was replication competent, but both M1LE/5Adel-1 and M1LE/5Adel-2 were not. Next we prepared 20 alanine-substituted clustered mutants within both NS5B-binding regions and examined the effect of these mutants on HCV RNA replication. Only 5 of the 20 mutants were replication competent. Subsequently, we introduced a point mutation, S225P, a deletion of S229, or S232I into NS5A and prepared cured Huh-7 cells that were cured of RNA replication by alpha interferon. Finally, with these point mutations and cured cells, we established a highly improved replicon system. In this system, only the same five mutants were replication competent. These results strongly suggest that the interaction between NS5A and NS5B is critical for HCV RNA replication in the HCV replicon system.

(a) More girls than boys were in the first top five class positions in Grades 1, 2, and 3. (b) No sex differences were found for Grades 4 and 5, and (c) more boys than girls were in the top five positions in Grades 6, 7, and 8. Is this a developmental phenomenon related to the differential growth of language and cognitive abilities?

Abstract (sommario):

Calcium silicate ceramics have been made using natural ingredients such as rice husk and snail shell with solid reaction techniques. The aim of this study to determine the characteristics of calcium silicate that have been sintered at a temperature of 900 °C, 1000 °C, and 1100 °C. Samples were characterized by X-Ray Diffraction (XRD), optical microscopy, and Fourier Transform Infra-Red (FTIR). The results of XRD characterization showed that CaSiO3 (calcium silicate) along with the increase in calcination temperature and quantity was increased due to the increase of temperature of calcination. FTIR analysis showed that the Ca-Si-O bond was formed at wave number ranging from 848 cm-1 and 999 – 1001 cm-1.

SummarySamples of UHT milk were contaminated at two levels (102 and 104 cfu/ml) with 52 bacterial strains associated with spoilage. Samples were assayed, using various tests, to determine efficiency in detecting non-sterility after an incubation period shorter than that advised in standard procedures. The tests performed were colony count, titratable acidity and pH modification, organoleptic assessment, stability towards ethanol (68, 80 and 88% v/v), resazurin and nitrate reduction. After 3 d incubation at 30 °C every case was detected by the resazurin test with a good statistical probability (P = 0·999). Reliability was not so high in all other tests, even after 14 d incubation.

Egg capsules and intracapsular embryos of Rapana venosa were studied to determine the reproductive pattern of this invader species in the south-eastern Black Sea. Egg deposition of R. venosa in the laboratory at 25°C was described. Egg laying began in mid-June, reached a peak in July and ended in late August in 1996. The total number of the egg capsules per whelk ranged between 197 and 999 (mean 575 ±41). The mean length of the egg capsules was 13.26 mm. Each capsule contained between 124 and 1090 embryos with a mean of 555 ±10. Mean larval production per rapa whelk was about 392,931 ±42,731. The mean size of eggs produced was 213.8 μm. Five different stages of intracapsular development were observed. Embryos developed without nurse eggs. Veliger larvae completed their intracapsular development and hatched in 22 days at 25°C.

Logbook data for the Taiwanese Pacific saury fishery and multi-sensor satellite images for 2006–2010 were used to characterize the habitat of Pacific saury (Cololabis saira) in the northwestern Pacific (NWP). An empirical cumulative distribution function (ECDF) approach identified that high cpue (catch per unit of effort) of Pacific saury occurred when sea surface temperature (SST) ranged from 14 to 16°C, chlorophyll-a concentration (Chl a) ranged from 0.4 to 0.6 mg m−3, and net primary production (NPP) ranged from 600 to 800 mg C m−2 d−1. A generalized additive model (GAM) and spatial non-stationary geographically weighted regression (GWR) were applied to predict the habitats of Pacific saury in the NWP. The spatial distributions of Pacific saury in the NWP estimated using the two approaches were similar and matched the nominal cpue distributions and those inferred from preferred habitat ranges based on ECDF. The density of Pacific saury is higher in coastal waters close to the island of Hokkaido and near the southern Kuril Islands than in the open sea. SST, Chl a, and NPP were substantially higher in the fishing grounds for Pacific saury during the main fishing season (September and October), corresponding to a high cpue for Pacific saury (23.1 t fishing day−1). The GAM explained more variability in spatial distribution (35.7%) than GWR (20.5%) VGPM (Vertically Generalized Production Model). Results derived from this study could improve our understanding of Pacific saury habitat distributions, which could be used to forecast fishing grounds and to develop fishery management advice based on oceanographic conditions that might be impacted by climate change.

Using a low temperature synthesis protocol, it was possible to obtain phase-pure synthetic aegirine (NaFeSi2O6) at temperatures as low as 130 °C, albeit only with rather long synthesis times of ~200 h; at 155 °C, a nano-crystallite shaped phase-pure material is formed after 24 h. These are, to the best of our knowledge, the lowest temperatures reported so far for phase-pure aegirine synthesis. Powder X-ray diffraction (PXRD) was used to characterize phase purity, structural state and microstructural properties (size and strain) of the as-synthesized (130–230 °C) and heat treated (300–900 °C) samples, via Rietveld analysis of powder patterns. Melting was observed at 999 °C. With increasing synthesis temperature, crystallite size linearly increased from 10 nm to 30 nm at 230 °C, while unit cell parameters decreased. The microstrain was very small. Additional heat treatment of as synthesized samples showed that the crystallite size remained rather unaffected up to 700 °C. The lattice parameters, however, already changed at low temperatures and successively became smaller, indicating increasing ordering towards more regular arrangements of building units. This was confirmed by 57Fe Mössbauer spectroscopy, where a distinct decrease of the quadrupole splitting with increasing synthesis temperature was found. Finally, Raman spectroscopy showed that some weakly-developed pre-ordering effects were present in the samples, which appeared to be amorphous in PXRD, while well-resolved spectra appeared as soon as the long-range ordered crystalline state could be found with X-ray diffraction.

Curcuma longa (turmeric) has traditionally been used in Ayurvedic, Unani and herbal drugs to cure numerous ailments. Due to the high demand, the quantitative standardization of herbal products is challenging to maintain their quality. We aim to develop a rapid, sensitive and validated high-performance thin-layer chromatography (HPTLC) method for the simultaneous determination and quantification of curcumin I, curcumin II and curcumin III in C. longa and herbal formulation. The three standards were separated using centrifugal preparative thin-layer chromatography (CPTLC) silica gel and identified by different spectroscopic methods. The developed HPTLC method was validated by following ICH guidelines (linearity; limit of detection, LOD; limit of quantitation; accuracy; precision; and robustness). The calibration curves of both the compounds were linear (50–500 ng/spot), with a correlation coefficient (r2) of >999. The developed HPTLC method was effectively applied to the concurrent detection and quantification of curcumins I–III in fresh, dry rhizomes and the herbal formulation of C. longa extracts was obtained by hot and cold extraction methods.

Aim: To evaluate the CANLPH score in in-hospital mortality after coronary artery bypass grafting. Materials & methods: The 999 patients were included in this retrospective cohort study. Neutrophil/lymphocyte ratio, C-reactive protein/albumin ratio and platelet/hemoglobin ratio were determined and the CANLPH score was calculated as the sum score of 0 or 1 by the cutoff in each ratio. Results: Twenty-five patients who reached the primary end point were defined as the mortality group and the remaining as the nonmortality group. The CANLPH score was noninferior to the European System for Cardiac Operative Risk Evaluation II in receiver-operating characteristic curve analysis with difference between AUC: 0.0162, standard error (SE): 0.0394, z statistics: 0.682 and p = 0.494. Conclusion: The CANLPH score may be more appropriate in assessing the risk of in-hospital mortality after coronary artery bypass grafting.

Wall inclination angles α considered include 10°, 15°, 20°, 25°, 30° and 90°. ABSTRACT Streptococcus suis infection is considered to be a major problem in the swine industry worldwide. Most virulent Canadian isolates of S. suis serotype 2 do not produce the known virulence markers for this pathogen. PCR-based subtraction hybridization was adapted to isolate unique DNA sequences which were specific to virulent strains of S. suis isolated in Canada. Analysis of some subtracted DNA clones revealed significant homology with bacteriophages of gram-positive bacteria. An inducible phage (named Ss1) was observed in S. suis following the incubation of the virulent strain 89-999 with mitomycin C. Phage Ss1 has a long noncontractile tail and a small isometric nucleocapsid and is a member of the Siphoviridae family. Ss1 phage DNA appears to be present in most Canadian S. suis strains tested in this study, which were isolated from diseased pigs or had proven virulence in mouse or pig models. To our knowledge, this is the first report of the isolation of a phage in S. suis.

Abstact:This article discusses the development of Islamic civilization in the east of Baghdad. The Abbasid empire once reached the peak era of Islamic civilization. This qualitative work was written by library method, the data collected by the writter of the history books. The study yileded results tht the Abbasid victory was short-lived, the dynamic polliticall situation, coupled with the power struggle between nations the Persian-Turkish and the Sunni-Shia conflict in the east region of Baghdad. The situation gave rise to a pollitical movement that resulted in new power from the Persians who founded power in the Thahirid Dynasty (820-897 c. e.), followed by The Shaffarid Dynasty (867-908 c. e), The Samanid Dynasty (819-999 c.e.), and The Buyid Dynasty (933-1055 c.e.). From Turk came the Ghaznavid Dynasty (977-1186 c.e.), The Seljuk Dynasty (1037-1157 c.e.) and The Khwarazmian Dynasty (1077-1231 c.e.). The Caliph is only symbol of the state of the Buyid-Seljuk era, and The Abbasid Empire are divided into small states led by governors known as amir or sulthan. It also gave rise the rival cities of Baghdad as the center of world civilization, such as Isfahan, Ghaznah, Shiraz and BukharaKeywords: Abbasid, Centra Asia, Islamic Civilization, East of Baghdad Abstrak:Artikel ini membahas perkembangan peradaban Islam di Timur Baghdad. Dinasti Abbaisyah pernah mencapai era puncak peradaban Islam. Artikel yang merupakan penelitan kualitatif ini ditulis dengan metode pustaka, data dikumpulkan oleh penulis dari buku-buku sejarah. Dari penelitian ini didapatkan hasil, bahwa kejayaan Abbasiyah tidak berlangsung lama, situasi politik yang berlangsung secara dinamis, diwarnai dengan perebutan pengaruh kekuasaan antara bangsa Persia-Turki dan konflik Sunni-Syiah di wilayah timur Baghdad. Situasi ini melahirkan gerakan politik yang membuat kekuasaan baru di mulai dari bangsa Persia yang mendirikan kekuatan di Dinasti Thahiriyah (820-897 M), kemudian dilanjutkan oleh Dinasti Shaffariyah (867-908 M), Dinasti Samaniyah (819-999 M), dan Dinasti Buwaihi (933-1055). Dari bangsa Turki, berdiri Dinasti Ghaznawiyah (977-1186 M), Dinasti Seljuk (1037-1157 M) dan Dinasti Khwarizm (1077-1231 M). Khalifah hanya sebagai simbol negara di era kekuasaan Buwaihi-Seljuk, dan negara Abbasiyah terpecah belah menjadi negara-negara kecil yang dipimpin oleh para gubernur yang dikenal dengan amir atau sulthan. Organic Rankine Cycle (ORC) systems are increasingly gaining relevance in the renewable and sustainable energy scenario. Recently our research group published a manuscript identifying a new type of thermodynamic cycle entitled Buoyancy Organic Rankine Cycle (BORC) [J. Schoenmaker, J.F.Q. Rey, K.R. Pirota, Renew. Energy 36, 999 (2011)]. In this work we present two main contributions. First, we propose a refined thermodynamic model for BORC systems accounting for the specific heat of the working fluid. Considering the refined model, the efficiencies for Pentane and Dichloromethane at temperatures up to 100 °C were estimated to be 17.2%. Second, we show a proof of concept BORC system using a 3 m tall, 0.062 m diameter polycarbonate tube as a column-fluid reservoir. We used water as a column fluid. The thermal stability and uniformity throughout the tube has been carefully simulated and verified experimentally. After the thermal parameters of the water column have been fully characterized, we developed a test body to allow an adequate assessment of the BORC-system's efficiency. We obtained 0.84% efficiency for 43.8 °C working temperature. This corresponds to 35% of the Carnot efficiency calculated for the same temperature difference. Limitations of the model and the apparatus are put into perspective, pointing directions for further developments of BORC systems.

Based on our results of genome-wide association analysis, we performed gene ontology (GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis; three candidate genes (ABCG2, CD44, SPP1) were screened in this study for SNPs association analysis with production traits in 999 Holstein cattle. In this research, flight mass spectrometry genotyping was used to detect the polymorphism of SNP seats. It was shown that four, four, and two single nucleotide polymorphisms (SNP) loci were detected for the ABCG2, CD44, and SPP1 genes, respectively, and the different genotypes of these 10 SNPs significantly affected the milk production performance of Chinese Holstein cattle in terms of milk yield, milk fat percentage, milk protein percentage, somatic cell score, and urea nitrogen content. Among them, ABCG2-G.80952G > T locus, ABCG2-G.120017G > A locus and CD44-G.2294G > C locus had significant effects on somatic cell score (p < 0.01). Cows with GG genotypes at ABCG2-G.80952G > T locus, AA and GG genotypes at ABCG2-G.120017G > A locus, and GG genotypes at CD44-G.2294G > C locus had lower somatic cell scores. The present study elucidated that ABCG2, CD44, and SPP1 could be selected for marker-assisted selection and will benefit for future precise molecular breeding.

Uno de los trastornos neurocognitivos más diagnosticado en España (5%) y reconocido por la LOMCE como Alumnado Con Neesidad Específica de Apoyo Educativo es el Trastorno por Déficit de Atención e Hiperactividad (TDAH). Escolares con problemas para concentrarse y en muchos casos con problemas de sueño e inquietud, que interfieren en su desarrollo social y escolar Este trabajo engloba dos proyectos piloto con diseño longitudinal experimental randomizado. Con el objetivo de conocer el efecto inmediato de 16min de “cooperative high-intensity interval training” (C-HIIT) sobre la concentración y evaluar el efecto de 10 semanas de C-HHIT en la calidad de sueño. En el proyecto 1 (n= 20) el Grupo Control visualizó un documental y el Grupo Experimental realizó Ejercicio Físico (4 minutos calentamiento + 16 minutos C-HIIT). En el proyecto 2 (n= 20) solo el Grupo Experimental asistió a las 10 semanas de actividad extraescolar C-HIIT (2 sesiones/semana, 30 minutos/sesión). Las herramientas empleadas fueron test d2 (concentración), test Pittsburg y pulseras Fitbit HR (calidad de sueño) y sistema Seego ® (intensidad de ejercicio). El análisis de medidas repetidas ANOVA 2 tiempos (pre-test vs. post-test) × 2 grupos (grupo control vs. grupo experimental) y test para medidas no para métricas, Willcoxon y U Mann-Whitney mostró que la realización del método C-HIIT tiene un efecto inmediato de mejora de la concentración [F(1.18)=20.50; p=.001; η 2=.532; 1–ß >.999] y mejora a largo plazo la calidad de sueño [F(1.18)=69.932; η 2=.795; 1–ß>.975; p<.001] en escolares con dificultades de aprendizaje.

Enzootic bovine hematuria (HEB) is a disease caused by exposure to, and consumption of pastures invaded by common fern (Pteridium aquilinum), also known as llashipa. The objective of the research was to identify the cattle areas of Cantón Chilla, establishing a relationship between their location and the incidence of enzootic bovine hematuria. Three zones were studied, each with four sectors, which were grouped according to their height - Zone A (300 - 999 masl), Zone B (1000 - 1999 masl) and Zone C (2000 - 3300 masl). The presence of llashipa in the surveyed areas corresponds to: 20% in Zone A, 60% in Zone B, and 50% in Zone C; data corresponding to recurrent invasive weeds in pastures. The urinary test strip techniques (TRU) and microscopic analysis of the urinary sediment (AMSU) were applied for the analysis of the samples. The results found were 36.1% (130/360) for AMSU and 5.28% (19/360) for TRU; whereupon the two methods demonstrate the existence of this disease in the canton of Chilla. Regarding the relation of the cellular elements variables (AMSU and TRU) and livestock areas (A, B and C), by means of the Chi square statistic test, it shows that the significance value p is less than 0.005, which indicates that there is a connection between the variables. It is concluded that the sectors grouped in the different livestock areas of Canton Chilla are endemic to enzootic bovine hematuria according to the height at which they are located.

This study provides data on the distribution and relationship of C. difficile PCR ribotypes in diarrhoeic calves in Germany. C. difficile was isolated from 176 of 999 (17.6 %) faecal samples or swabs of diarrhoeic calves from 603 farms collected between January 2010 and August 2012 by eight federal laboratories of six states. Strains were assigned to 17 PCR ribotypes. PCR ribotypes 033 (57 %), 078 (17 %) and 045/FLI01 (closest match to 045 in the WEBRIBO database; 9 %) were found the most frequently. Nine per cent of all culture-positive tested animals shed more than one multiple locus variable number tandem repeat analysis (MLVA) or PCR ribotype. Eight PCR ribotypes with related profiles (including 033, 078 and 045/FLI01) representing 92 % of all isolates were grouped into three clusters. Molecular relatedness was supported by the absence of the MLVA locus A6 Cd only in clustered strains and identical toxin gene profiles for strains within each cluster. Previously reported mulitilocus sequence typing analysis for PCR ribotypes that were also recovered in this study found identical sequence types and a tcdC deletion (Δ39 bp) for 033, 045, 078 and 126 (ST-11), confirming this clustering. A different geographical occurrence of PCR ribotypes was shown for cluster 033 (found more frequently in southern Germany) and 045 (found more frequently in northern Germany). This study showed that clusters of C. difficile PCR ribotypes related to 033, 078 and 045 are predominant in diarrhoeic calves in Germany. The high number of strains belonging to PCR ribotype 078 demonstrated that diarrhoeic calves are also potential reservoirs for human pathogenic C. difficile strains.

The impact of storage conditions on compound stability and compound solubility has been debated intensely over the past 5 years. At Novartis, the authors decided to opt for a storage concept that can be considered controversial because they are using a DMSO/water (90/10) mixture as standard solvent. To assess the effect of water in DMSO stocks on compound stability, the authors monitored the purity of a subset of 1404 compounds from ongoing medicinal chemistry projects over several months. The study demonstrated that 85% of the compounds were stable in wet DMSO over a 2-year period at 4 °C. This result validates the storage concept developed at Novartis as a pragmatic approach that takes advantage of the benefits of DMSO/water mixtures while mediating the disadvantages. In addition, the authors describe how purity data collected over the course of the chemical validation of high-throughput screening actives are used to improve the analytical quality of the Novartis screening deck. This study aimed to obtain a functional lipase (LipRM) from Ralstonia pickettii BK6 through a co-expression involving its foldase. The ORF of the LipRM was 999 bp while the gene encoding of lipase-specific foldase (LifRM) was 1,030 bp. LipRM and LifRM genes were cloned into a plasmid and were successfully co-expressed in Escherichia coli strains to produce functional LipRM. Enzyme activity from partially purified enzymes showed quite surprising results, LipRM activity in E. coli BL21 (DE3) was 25.84 U/ml, while the other strains (DH5α, HB101, S17-1λpir) were 628.98 U/ml, 761 U/ml, and 1206.46 U/ml, respectively. The highest relative activity of LipRM was found at 50-55°C and pH 7-8 with pNP-laurate (C12) as the preferred substrate specificity. LipRM activity was enhanced sharply in the presence of 30% organic solvents (methanol and ethanol) but decreased by more than 50% in the presence of detergents. This study was the first to report heterologous expression of Ralstonia pickettii lipase employing its native foldase resulting in functional lipase from subfamily I.2.

ABSTRACT A genomic library of Pseudomonas fluorescens DSM 50106 in a λRESIII phage vector was screened in Escherichia coli K-12 for esterase activity by using α-naphthyl acetate and Fast Blue RR. A 3.2-kb DNA fragment was subcloned from an esterase-positive clone and completely sequenced. Esterase EstF1 was encoded by a 999-bp open reading frame (ORF) and exhibited significant amino acid sequence identity with members of the serine hydrolase family. The deduced amino acid sequences of two other C-terminal truncated ORFs exhibited homology to a cyclohexanone monooxygenase and an alkane hydroxylase. However, esterase activity was not induced by growing of P. fluorescens DSM 50106 in the presence of several cyclic ketones. The esterase gene was fused to a His tag and expressed in E. coli. The gene product was purified by zinc ion affinity chromatography and characterized. Detergents had to be added for purification, indicating that the enzyme was membrane bound or membrane associated. The optimum pH of the purified enzyme was 7.5, and the optimum temperature was 43°C. The showed highest purified enzyme activities towards lactones. The activity increased from γ-butyrolactone (18.1 U/mg) to ɛ-caprolactone (21.8 U/mg) to δ-valerolactone (36.5 U/mg). The activities towards the aliphatic esters were significantly lower; the only exception was the activity toward ethyl caprylate, which was the preferred substrate.

BACKGROUND: Persistent headward fluid shift and mechanical unloading cause neuro-ocular, cardiovascular, and musculoskeletal deconditioning during long-term spaceflight. Lower body negative pressure (LBNP) reintroduces footward fluid shift and mechanical loading.METHODS: We designed, built, and tested a wearable, mobile, and flexible LBNP device (GravitySuit) consisting of pressurized trousers with built-in shoes to support ground reaction forces (GRF) and a thoracic vest to distribute load to the entire axial length of the body. In eight healthy subjects we recorded GRF under the feet and over the shoulders (Tekscan) while assessing cardiovascular response (Nexfin) and footward fluid shift from internal jugular venous cross-sectional area (IJVa) using ultrasound (Terason).RESULTS: Relative to normal bodyweight (BW) when standing upright, increments of 10 mmHg LBNP from 0 to 40 mmHg while supine induced axial loading corresponding to 0%, 13 ± 3%, 41 ± 5%, 75 ± 11%, and 125 ± 22% BW, respectively. Furthermore, LBNP reduced IJVa from 1.12 ± 0.3 cm2 to 0.67 ± 0.2, 0.50 ± 0.1, 0.35 ± 0.1, and 0.31 ± 0.1 cm2, respectively. LBNP of 30 and 40 mmHg reduced cardiac stroke volume and increased heart rate while cardiac output and mean arterial pressure were unaffected. During 2 h of supine rest at 20 mmHg LBNP, temperature and humidity inside the suit were unchanged (23 ± 1°C; 47 ± 3%, respectively).DISCUSSION: The flexible GravitySuit at 20 mmHg LBNP comfortably induced mechanical loading and desired fluid displacement while maintaining the mobility of hips and knee joints.
